Abstract

International audience; The PPILOW project aims to co-construct innovations to improve Poultry and Pig Welfare in Low-input outdoor and Organic farming systems through a multi-actor approach. PPILOW implements a participatory approach for proposing and studying welfare-improvement levers. It will provide a combination of practical solutions that can be applied at a pan-European level with specific adjustments depending on citizen’s expectations and the target market. The multi-actor approach consists in involving end-users including farmers, breeding companies, feed producers, consumer associations, retailers, advisers, processors, and scientists in National Practitioner Groups (NPG) in six participating countries. PPILOW partners facilitate the groups by connecting NPG at European level, transferring scientific information, interacting with partners engaged in animal experiments, and co-creating innovations rising from NPG-specific demands. They co-build with PPILOW partners welfare self-assessment tools (development of the PIGLOW app for pigs and refinement of the EBENE® app for poultry), and innovative breeding, feeding, and rearing strategies and techniques to improve the welfare of animals. They co-design protocols, test innovations on farm, and disseminate the results. In turn, they receive insights on methods and scientific results, and inputs from other NPG reinforcing the value of the expected outcomes. Approaches focus on avoiding physical damage and the elimination of layer male chicks, on reducing boar taint of intact male pigs, promoting positive behaviours, animal health, and robustness through field studies with pigs and poultry. Multicriteria analyses of the most effective levers of welfare improvement will be performed to evaluate their economic, social, and environmental impacts based on the ‘One Welfare’ concept; economic and business models will also be developed. To ensure the rapid uptake of the project results by end-users, the close involvement of PPILOW’s NPG throughout the EU will ensure disseminationactivities and the facilitation of change. Abstract

Amino acids are known to be anabolic factors that affect protein metabolism, but the response of animals to daily amino acid changes is little understood. We aimed to test the effects of feeding birds with alternations of diets varying in lysine content on the expression of genes related to proteolysis in chicken muscle. Cyclic feeding programs with 2 diets, each given for 24 h during 48-h cycles, were carried out from 10 d of age. Three programs were used: 1) control treatment with continuous distribution of a complete diet containing standard medium lysine level (ML; 11.9 g/kg); 2) alternation of diets with high (HL) and low (LL) lysine levels; 3) alternation of ML and LL diets, where LL = 70%, ML = 100%, HL = 130% of standard lysine level. The Pectoralis major muscles were sampled after 2 wk of cyclic feeding. Measurements included the expression patterns of 6 genes involved in proteolysis, and mammalian target of rapamycin and Forkhead box-O transcription factor (FoxO) signaling. Cathepsin B, m-calpain, and E3 ubiquitin ligases Muscle Ring Finger-1 and Muscle Atrophy F box were significantly overexpressed in chickens transiently fed the LL diet, whereas the mRNA levels of 20S proteasome C2 subunit and ubiquitin remained unchanged. Modifications of E3 ubiquitin ligase expression can be partly explained by significant changes in FoxO phosphorylation with cyclic dietary treatments. Our results suggest timing-sensitive regulation of proteolysis in chicken muscle according to dietary treatment and a high metabolism capacity to compensate for changes in amino acid supply, which might be used for nutritional purposes.

Abstract

In chicken, the ultimate pH (pHu) of meat is largely determined by the muscle glycogen content at slaughter age.A divergent selection on the Pectoralis major pHu allowed the creation of the pHu+ and pHu- lines, whichrepresent a unique model for studying the genetic and physiological control of energy reserves and the meatquality. Several characteristics have been measured in the two lines at hatch then at 5 days of age: animal weight,muscle yields, muscular and hepatic glycogen content and several plasma parameters. At hatch, the pHu+ andpHu- lines present equivalent body weights and muscle yields. However, the pHu- line shows a higher glycemiathan the pHu+ line, a difference that will persist until the slaughter age. In addition, primary myoblasts culturesfrom both lines show differences in insulin response. After 5 days, the pHu+ and pHu- lines are alreadydivergent in terms of glycogen (and pHu) and muscle yield, for a similar body weight. Differences between bothlines observed at hatch and at 5 days may have been associated with regulation of signaling pathways involvedin protein synthesis and muscle glycogen turnover. In conclusion, pHu+ and pHu- chicks already have at hatchthe ability to respond differently to nutrients and hormones consistent with the fact that after only 5 days offeeding, significant differences in yield and muscular glycogen contents exist between the two lines.; Chez le poulet, le pH ultime (pHu) de la viande est en grande partie déterminé par la teneur en glycogène dumuscle à l'abattage. Une sélection divergente sur le pHu du filet a permis la création des lignées pHu+ et pHu-,qui représentent un modèle unique pour étudier le contrôle génétique et physiologique des réserves énergétiqueset de la qualité de la viande. Plusieurs caractéristiques ont été mesurées dans les deux lignées à l’éclosion puis à5 jours d’âge: le poids vif, les rendements en muscles, le glycogène musculaire et hépatique et plusieursparamètres plasmatiques. A l’éclosion, les lignées pHu+ et pHu- présentent des poids vifs et des rendementsmusculaires équivalents. Toutefois, la lignée pHu- présente une glycémie supérieure à celle de la lignée pHu+,différence qui va persister jusqu'à l'âge d’abattage. Par ailleurs, les cultures primaires des myoblastes issues desdeux lignées révèlent des différences de réponse à l'insuline. Après 5 jours, les lignées pHu+ et pHu- sont déjàdivergentes en terme de glycogène (et de pHu) et de rendement en muscle, pour un poids vif similaire. Lesdifférences entre lignées observées à l'éclosion et à 5 jours ont pu être associées à des régulations au niveau desvoies de signalisation impliquées dans la synthèse protéique et le turn-over du glycogène musculaire. Enconclusion, les poussins pHu+ et pHu- présentent dès l’éclosion des capacités de réponses différentes auxnutriments et aux hormones cohérentes avec le fait qu’après seulement 5 jours d’alimentation, des différencessignificatives de rendement et de teneur en glycogène musculaire existent entre les deux lignées.

Abstract

To mitigate effects of stressful factors during the post-natal period in broiler chicks, we have tested the chickability to self-medicate when essential oils (EO) are provided. Chicks were either immediately placed in theexperimental rearing facility after hatching (group T), or submitted to a delayed period before their placement(group MEE-R) mimicking transport delay in commercial conditions. In experiment 1, half of them had onlyaccess to water and the other half to water and one EO (cardamom, marjoram or verbena) during the earlyrearing phase (13 days). In experiment 2, half of the chicks had only access to water and the other half wereoffered water and the 3 EO simultaneously.In experiment 1, the intake of the EO in groups MEE-R was higher than in groups T from D1 to D6 whatever theEO. The reduction of growth induced by the MEE-R was not mitigated by EO intake. In experiment 2, from D1to D6, the marjoram intake was the highest compared to the other EO in the groups T and MEE-R. Between D6and D12, cardamom EO tended to be less drunk in MEE-R groups than in groups T. EO intake did not allow toovercome the reduced growth in groups MEE-R, but tended to overcome the reduction of the relative percentageof Pectoralis major muscles. The EO intake did not reduce the prevalence of health disorder.In conclusion, chicks are able to make spontaneous choices for EO according to their post-natal experience.These choices had a few beneficial effects on their performances, but their health was not improved in thepresent experimental conditions., la capacité des poussins à s’automédiquer en leur mettant à disposition des huiles essentielles (HE). Pour mimer le stress du transport du couvoir en bâtiment d’élevage, des poussins ont été mis en élevage, soit directement après l’éclosion (lot T), soit exposés à une mise en élevage retardée de 24h (MEE-R).Dans l’expérience 1, la moitié des poussins avait accès uniquement à l’eau et l’autre moitié à l’eau et une HE (cardamome ou marjolaine à coquille ou verveine) pendant la phase de démarrage (13j). Dans l’expérience 2, la moitié des poussins avait accès uniquement à l’eau et l’autre moitié à l’eau et simultanément aux trois HE distribuées dans 3 biberons différents. Dans l’expérience 1, la consommation de chacune des HE dans les lots MEE-R était supérieure aux lots T de J1 à J6, quelle que soit l’HE. La réduction de la croissance des poussins induite par la MEE-R n’a pas été améliorée par la consommation des HE. Dans l’expérience 2, l’HE de marjolaine était la plus consommée de J1 à J6, quel que soit le lot. Entre J6 et J12, l’HE de cardamome tend à être moins consommée dans les lots MEE-R que dans les lots T. La consommation des HE n’a pas permis de pallier la réduction de croissance des poussins des lots MEE-R, mais a tendance à pallier la réduction du pourcentage relatif des muscles Pectoralis major. Elle n’a pas eu d’effet bénéfique sur la prévalence des troubles de santé des animaux. En conclusion, les poussins sont capables de faire des choix spontanés de consommation d’HE en fonction de leur expérience post-natale. Ces choix ont eu quelques effets bénéfiques sur les performances des poussins, mais aucun effet significatif sur leur santé dans les conditions de nos expérimentations.

International audience; The impact of divergent selection based on the ultimate pH (pHu) of pectoralis major (P. major) muscle on the chemical, biochemical, and histological profiles of the muscle and sensorial quality of meat was investigated in broiler chickens. The protein, lipid, DM, glycogen and lactate content, glycolytic potential, proteolysis, lipid and protein oxidation index, muscle fiber cross-sectional area, capillary density, and collagen surface were determined on the breast P. major muscle of 6-wk-old broilers issued from the high-pHu (pHu+) and low-pHu (pHu-) lines. Sensory attributes were also evaluated on the breast (roasted or grilled) and thigh (roasted) meat of the 2 lines. Protein, lipid, and DM content of P. major muscle were not affected by selection (P > 0.05). However, the P. major muscle of the pHu+ line was characterized by lower residual glycogen (-16%; P 0.05) in both lines, fewer capillaries per fiber (-15%; P 0.05) between the 2 lines.
