Abstract

Burkholderia pseudomallei is the causative agent of melioidosis, a disease endemic to Southeast Asia and northern Australia. Mortality rates in these areas are high even with antimicrobial treatment, and there are few options for effective therapy. Therefore, there is a need to identify antibacterial targets for the development of novel treatments. Cyclophilins are a family of highly conserved enzymes important in multiple cellular processes. Cyclophilins catalyze the cis-trans isomerization of xaa-proline bonds, a rate-limiting step in protein folding which has been shown to be important for bacterial virulence. B. pseudomallei carries a putative cyclophilin B gene, ppiB, the role of which was investigated. A B. pseudomallei ΔppiB (BpsΔppiB) mutant strain demonstrates impaired biofilm formation and reduced motility. Macrophage invasion and survival assays showed that although the BpsΔppiB strain retained the ability to infect macrophages, it had reduced survival and lacked the ability to spread cell to cell, indicating ppiB is essential for B. pseudomallei virulence. This is reflected in the BALB/c mouse infection model, demonstrating the requirement of ppiB for in vivo disease dissemination and progression. Proteomic analysis demonstrates that the loss of PpiB leads to pleiotropic effects, supporting the role of PpiB in maintaining proteome homeostasis. The loss of PpiB leads to decreased abundance of multiple virulence determinants, including flagellar machinery and alterations in type VI secretion system proteins. In addition, the loss of ppiB leads to increased sensitivity toward multiple antibiotics, including meropenem and doxycycline, highlighting ppiB inhibition as a promising antivirulence target to both treat B. pseudomallei infections and increase antibiotic efficacy.

Abstract

Soil moisture is a very important component of the climate system, regulating both energy and water fluxes on local and regional scales. To obtain accurate climatological and meteorological forecasts we therefore need to obtain accurate predictions of soil moisture from land surface models. Actual in situ measurements are crucial for testing and developing these models. It is to this end that we have obtained a new soil moisture dataset from the Boissy-le-Châtel hydrological station in the Orgeval catchment in France. This dataset has a time span of over 5 years, from late 1996 to early 2003. It includes soil moisture measurements for 11 layers, as well as meteorological observations which can be used to force a land surface model. The data will be made available to the public via the Rutgers University Center for Environmental Prediction Global Soil Moisture Data Bank. We force the NOAH land surface model with the meteorology observations, and evaluate the resulting soil moisture, sensible and latent heat fluxes, and runoff using the Boissy-le-Châtel data. This experiment can serve as a prototype for other models to conduct their own evaluations, in the spirit of PILPS Phase 2.; L`état hydrique est un élément très important du système climatique, régulant aussi bien les flux d`eau et d`énergie aux échelles régionales que locales. Afin d`obtenir des prévisions climatologiques et météorologiques précises il est nécessaire d`avoir des prédictions fiables de l`état hydrique du sol à partir de modèles de surface. Des mesures in situ sont donc cruciales pour tester et développer ces modèles. C`est dans ce but que nous avons acquis une nouvelle base de données provenant de Boissy-le-Châtel une station hydrologique du bassin de l`Orgeval en France. Cette base de données couvre une période de 5 ans, allant de 1996 à début 2003. Elle comprend des mesures d`humidité pour onze couches de sol ainsi que des données météorologiques qui peuvent être utilisées pour forcer le modèle de surface. Les données sont mises à disposition via la base de données du centre pour l`environnement et la prédiction globale de l`humidité du sol de l`université de Rutgers. Nous forçons le modèle NOAH de surface avec des observations météorologiques et nous évaluons l`état hydrique résultant, les flux de chaleur sensibles et latents et les écoulements en utilisant les données de Boissy le Châtel. Cette expérience peut servir de prototype à d`autres modèles pour effectuer leur propres évaluations dans l`esprit du projet PILPS Phase 2.

Abstract

One of the tasks within the framework of the EU “RESQUE”- project was to determine the lowest acceptable insert length for reconstituted Charpy V impact (Cv) and pre-cracked Charpy V (PC Cv) specimens. In order to obtain an optimized insert length, Cv impact specimens with three insert lengths ranging from 10 to 20 mm and fracture toughness PC Cv specimens with two different insert lengths of a reference material were manufactured by stud-welding or electron-beam welding and tested under various test conditions. Cv tests on reconstituted specimens exhibited the normal scatter in Charpy energies and that there was no significant influence of the insert length on the transition temperatures. However, the 10 mm insert caused a reduction in the upper shelf energy of 10–15%. The fracture toughness tests carried out in the transition regime showed that the 10 mm insert resulted in a lower estimate (non-conservative) of T0 compared with the values measured on 12mm insert and baseline specimens. It is therefore recommended that at least 12 mm long inserts should be used for fracture toughness testing in the transition region. In the ductile region a trend to lower initiation toughness values at higher temperatures was observed. To avoid this, 20 mm long inserts should be used for J-Δa curves at higher temperatures.

Abstract

This paper summarizes the experience of the RESQUE project. The final objective of the project was the development of a set of 'Recommendations for Reconstitution of Non-Irradiated and Irradiated Charpy-size Specimens,' based on the systematic analysis of the experimental data from the program and on the collective reconstitution experience available in different laboratories within Europe. The paper reviews the general information from RESQUE and summarizes the conclusions. Specific details are given in other contributions to this Symposium.

Abstract

This paper continues the presentation of detailed results of the EU-sponsored RESQUE project. Within the RESQUE project it was planned to determine the dependence of Charpy-V test results on tolerance deviations and different end block materials in reconstituted specimens. The conventional Charpy-V parameters - the total impact energy, lateral expansion and shear fracture appearance - are not significantly affected by the effects of oversized dimensional variability. Disturbing load oscillations arise from some of the geometrical deviations. This influences the characteristic yield load, yield deflection and impact energy at load maximum. In comparison to the existing scatter there is no significant effect of the end block material on the main Charpy parameters and the characteristic loads.

Abstract

This paper continues reporting on the EU-sponsored research project RESQUE. First detailed checks with different ways to instrument reconstitution inserts were performed and thin silver soldered thermocouples were chosen to be used in the project. Temperature transients in welded inserts were measured in detail in order to compare the properties of six different reconstitution systems. The measured temperature transients were repeatable and relatively consistent. Within approximately 4 mm of the near weld seam volume a typical material irradiation temperature was exceeded for a short time. This value was nearly the same by all devices. Validation tests with original and reconstituted specimens were performed. No indication of material annealing in reconstitution, i.e. a trend towards lower transition temperatures, was noticed. As compared to the requirements of ASTM E1253-88, the applied insert lengths were too short. Still original data was repeated with reconstituted specimens, which is an indication that the standard is conservative.

Abstract

Certain environmental microorganisms can cause severe human infections, even in the absence of an obvious requirement for transition through an animal host for replication ("accidental virulence"). To understand this process, we compared eleven isolate genomes of Burkholderia pseudomallei (Bp), a tropical soil microbe and causative agent of the human and animal disease melioidosis. We found evidence for the existence of several new genes in the Bp reference genome, identifying 282 novel genes supported by at least two independent lines of supporting evidence (mRNA transcripts, database homologs, and presence of ribosomal binding sites) and 81 novel genes supported by all three lines. Within the Bp core genome, 211 genes exhibited significant levels of positive selection (4.5%), distributed across many cellular pathways including carbohydrate and secondary metabolism. Functional experiments revealed that certain positively selected genes might enhance mammalian virulence by interacting with host cellular pathways or utilizing host nutrients. Evolutionary modifications improving Bp environmental fitness may thus have indirectly facilitated the ability of Bp to colonize and survive in mammalian hosts. These findings improve our understanding of the pathogenesis of melioidosis, and establish Bp as a model system for studying the genetics of accidental virulence.
