Abstract

The present study assesses different hormonal treatments for the induction of gonadal maturation and spawning in female wild European eels Anguilla anguilla L., 1758. One of the groups received weekly injections of salmon pituitary extract and human chorionic gonadotropin, while a second group received the same treatment together with pymozide. Although previous studies had reported spontaneous release of oocytes, in the present study, during the final phases of oocyte development, a dose of 17.20β-dihydroxy-4-pregnen-3-one (17.20βP) was used to induce the first controlled spawning in A. anguilla. After 12-14 weeks of treatment, 5 spawns were obtained. Treatments also caused morphological changes, such as progressively darker coloration and increase in eye diameter. El objetivo principal del presente estudio fue la evaluación de distintos tratamientos hormonales para la inducción de la maduración gonadal y la puesta en hembras salvajes de anguila europea Anguilla anguilla L., 1758.Un grupo recibió inyecciones semanales de extracto de hipófisis de salmón y gonadotropina coriónica humana, mientras que un segundo grupo recibió además pymozide. Aunque estudios previos habían logrado liberación espontánea de oocitos, en el presente estudio se empleó una dosis de 17,20 β-dihidroxi-4-pregnen-3-ona (17,20 βP) durante las fases finales del desarrollo oocitario para inducir de forma controlada, por primera vez en la anguila europea, la ovulación y puesta. Después de 12-14 semanas de tratamiento se obtuvieron 5 puestas. Los tratamientos también causaron cambios morfológicos, como el oscurecimiento de la coloración o el aumento del diámetro ocular. Abstract

Abstract: The first goal of this study was the development of a short-term storage method for pufferfish (Takifugu niphobles) sperm. In this respect, the best results were obtained by diluting the sperm in a seminal-like solution and keeping it in a Petri dish in chilled storage (4°C). This method was successful in preserving sperm quality parameters without resulting in differences in fresh sperm for a relatively long-term period (7days), for use in aquaculture matters. The addition of bovine serum albumin (BSA) to the medium did not improve the results. On the other hand, both the osmolality and the ion composition of the media are essential factors which can modulate the sperm motility parameters. The osmolality of the activating medium was the most important factor in triggering pufferfish sperm motility, and osmolalities of 750–825mOsm/kg were necessary to initiate this process, demonstrating that it appears to be a dose-independent mechanism. Regarding the ion composition of the activation media, this study has shown that despite the spermatozoa being able to initiate movement without any ion in the activation medium, the absence of ions can negatively affect the kinetic parameters of the spermatozoa. Finally, in natural conditions (seawater), the activation of sperm motility generates intracellular increases in Ca2+ and K+, suggesting that these ions play an essential role in the activation mechanism of pufferfish sperm. Abstract

Abstract: Although it is widely accepted that osmolality and ion fluxes are the main factors triggering sperm motility in fish, a complex universal mechanism for sperm motility activation does not exist in fish, and studies of marine fish species are even more scarce. Therefore, the main goal of this study was to estimate the intracellular variations in the main ions involved in sperm activation for the first time in European eel, in order to provide additional new data about this little-known process. It was observed that levels of intracellular Ca2+ and K+ sperm ions increased significantly 30s after the hyperosmotic shock compared to baseline levels, and remained at this level until 120s post-activation. In contrast, the intracellular pH remained constant during the first 30s, and decreased gradually at 60 and 120s post-activation. Our data agree with the current main theory for explaining motility activation in marine fish, in which internal fluctuations of Ca2+ and K+ seem to participate in sperm activation. In addition, fluorescent images showed that both Ca2+ and K+ were concentrated in the apical area of the sperm head, which corresponds to the location of the eel sperm mitochondria, suggesting this organelle plays an important role in sperm motility activation.
