Local, regional and global targets have been set to halt marine biodiversity loss. Europe has set its own policy targets to achieve Good Environmental Status (GES) of marine ecosystems by implementing the Marine Strategy Framework Directive (MSFD) across member states. We combined an extensive dataset across five Mediterranean ecoregions including 26 Marine Protected Areas (MPAs), their reference unprotected areas, and a no-trawl case study. Our aim was to assess if MPAs reach GES, if their effects are local or can be detected at ecoregion level or up to a Mediterranean scale, and which are the ecosystem components driving GES achievement. This was undertaken by using the analytical tool NEAT (Nested Environmental status Assessment Tool), which allows an integrated assessment of the status of marine systems. We adopted an ecosystem approach by integrating data from several ecosystem components: the seagrass Posidonia oceanica, macroalgae, sea urchins and fish. Thresholds to define the GES were set by dedicated workshops and literature review. In the Western Mediterranean, most MPAs are in good/high status, with P. oceanica and fish driving this result within MPAs. However, GES is achieved only at a local level, and the Mediterranean Sea, as a whole, results in a moderate environmental status. Macroalgal forests are overall in bad condition, confirming their status at risk. The results are significantly affected by the assumption that discrete observations over small spatial scales are representative of the total extension investigated. This calls for large-scale, dedicated assessments to realistically detect environmental status changes under different conditions. Understanding MPAs effectiveness in reaching GES is crucial to assess their role as sentinel observatories of marine systems. There is increasing concern that accelerating environmental change attributed to human-induced warming of the planet may substantially alter the patterns, distribution and intensity of Harmful Algal Blooms (HABs). Changes in temperature, ocean acidification, precipitation, nutrient stress or availability, and the physical structure of the water column all influence the productivity, composition, and global range of phytoplankton assemblages, but large uncertainty remains about how integration of these climate drivers might shape future HABs. Presented here are the collective deliberations from a symposium on HABs and climate change where the research challenges to understanding potential linkages between HABs and climate were considered, along with new research directions to better define these linkages. In addition to the likely effects of physical (temperature, salinity, stratification, light, changing storm intensity), chemical (nutrients, ocean acidification), and biological (grazer) drivers on microalgae (senso lato), symposium participants explored more broadly the subjects of cyanobacterial HABs, benthic HABs, HAB effects on fisheries, HAB modelling challenges, and the contributions that molecular approaches can bring to HAB studies. There was consensus that alongside traditional research, HAB scientists must set new courses of research and practices to deliver the conceptual and quantitative advances required to forecast future HAB trends. These different practices encompass laboratory and field studies, long-term observational programs, retrospectives, as well as the study of socioeconomic drivers and linkages with aquaculture and fisheries. In anticipation of growing HAB problems, research on potential mitigation strategies should be a priority. It is recommended that a substantial portion of HAB research among laboratories be directed collectively at a small sub-set of HAB species and questions in order to fast-track advances in our understanding. Toxic blooms of Ostreopsis spp. are greatly challenging to monitor due to the complexity and variability of cell repartition among benthic and pelagic compartments. This results in marked differences in employed methodologies for the survey of their dynamics and hampers the definition of the associated toxic risk. The present study aims at testing and improving common methodologies used for sampling, processing and counting of field samples. It contributes to the identification of the most suitable strategies for the monitoring and mitigation of Ostreopsis blooms in coastal waters. For a sampling based on the collection of macrophytes, the role of the fixative addition and of agitation steps in the efficiency of epiphytic cell collection was defined. For planktonic estimations, the influence of the volume used for concentrating water samples was characterized as a function of Ostreopsis cell abundance. The deployment of artificial substrates was tested and confirmed strong advantages of this new sampling methodology, including an integration of a part of the spatial and temporal variability of the cell distribution.

Marine protected areas (MPAs) are key tools to mitigate human impacts in coastal environments, promoting sustainable activities to conserve biodiversity. The designation of MPAs alone may not result in the lessening of some human threats, which is highly dependent on management goals and the related specific regulations that are adopted. Here, we develop and operationalize a local threat assessment framework. We develop indices to quantify the effectiveness of MPAs (or individual zones within MPAs in the case of multiple-use MPAs) in reducing anthropogenic extractive and non-extractive threats operating at local scale, focusing specifically on threats that can be managed through MPAs. We apply this framework in 15 Mediterranean MPAs to assess their threat reduction capacity. We show that fully protected areas effectively eliminate extractive activities, whereas the intensity of artisanal and recreational fishing within partially protected areas, paradoxically, is higher than that found outside MPAs, questioning their ability at reaching conservation targets. In addition, both fully and partially protected areas attract non-extractive activities that are potential threats. Overall, only three of the 15 MPAs had lower intensities for the entire set of eight threats considered, in respect to adjacent control unprotected areas. Understanding the intensity and occurrence of human threats operating at the local scale inside and around MPAs is important for assessing MPAs effectiveness in achieving the goals they have been designed for, informing management strategies, and prioritizing specific actions.

This study describes the adoption and validation of two innovative methods for the automated count of Ostreopsis spp. concentration in sea water: a molecular assay based on qPCR and an opto-electronic device allowing to automatically acquire images from the whole sample and recognize the target algae on the basis of a machine learning algorithm. The two approaches were tested on samples coming from different locations in 5 countries along the Mediterranean Sea, collected in the framework of the cross-border EU ENPI CBCMED Project M3-HABs, and compared with the standard visual counting method. The results demonstrate the effectiveness of both automatic approaches which provide a valuable tool, mostly cost and time effective, for the establishment of large-scale and effective monitoring of Ostreopsis spp. blooms. Moreover, the two automatic methods demonstrated the ability to discriminate for the presence of a different but similar species, O. fattorussoi, for which new species-specific qPCR primers were developed
