Aberrant use of antibiotics is associated with the emergence of antibiotics resistance and resistant superbugs. Keeping in view the current scenario, the present study was designed for the first time from Pakistan with a specific aim to estimate the prevalence of the carbapenem-resistant Klebsiella pneumoniae in veterinary settings and the waste in Pakistan.Methods: A total of 138 samples from various veterinary sources were collected by employing a nonprobability sampling technique. Isolation and phenotypic identification of carbapenem-resistant K. pneumoniae were performed according to the CLSI standard. Molecular detection of various antibiotic resistance genes (ARGs) was done through PCR by using specific primers against each ARG. According to the pasture scheme, the multilocus sequence typing (MLST) was performed to characterize the K. pneumoniae sequence types (STs).Results: According to the results of the study, overall 9.4% (13/138) isolates were confirmed carbapenem-resistant K. pneumoniae. Among various carbapenem ARGs particularly, the blaNDM-1 was found in 92.3% (12/13) isolates followed by blaOXA-48 84.6% (11/13). MLST results revealed that overall 3 STs were found in the study which includes ST29, ST11, and ST258. Taking together, this is the first study to our best knowledge which demonstrated the prevalence of carbapenem-resistant K. pneumoniae and its various STs prevalent in veterinary settings and the waste of Pakistan.Conclusion: Based on the above-mentioned facts, we suggested that veterinary settings and waste are the potential source and reservoir of carbapenem-resistant K. pneumoniae, which may be disseminated to the environment and ultimately can affect the public and companion livestock health.Keywords: veterinary settings, carbapenem resistance, Klebsiella pneumoniae, Pakistan

The crisis of antimicrobial resistance has been ascribed to the misuse of these agents and due to unavailability of newer drugs attributable to exigent regulatory requirements and reduced financial inducements. Comprehensive efforts are needed to minimize the pace of resistance by studying emergent microorganisms, resistance mechanisms, and antimicrobial agents. Multidisciplinary approaches are required across health care settings as well as environment and agriculture sectors. Progressive alternate approaches including probiotics, antibodies, and vaccines have shown promising results in trials that suggest the role of these alternatives as preventive or adjunct therapies in future. The investigation of exact solitary wave solutions to the nonlinear partial differential equation plays an important role to understand any physical phenomena in diverse applied fields. The current work is related to the most prominent nonlinear model named as the van der Waals normal form that appeared naturally and also industrially for the granular materials. In oceanography, the sea ice, sand and snow are some examples of aforesaid matter among others. We employ two novel integration approaches named as the simplest equation method and the expa function method to explore the above mentioned van der Waals model. As a backlash, many new solitary waves and other exact solutions are retrieved. The obtained results depict that the used approaches are simple and effective to deal with nonlinear models. Also, the numerical simulation of some solutions via two and three dimension graphical configurations are presented for certainty and exactness.

Introduction: Intractable hemifacial pain or twitching can be an incapacitating disorder caused by trigeminal neuralgia. The aetiologies for trigeminal neuralgia are numerous and the most frequent cause is a neurovascular compression. However, there are other less common aetiologies which need to be assessed. Magnetic Resonance Imaging (MRI) with dedicated cranial nerve sequences remains the modality of choice in diagnosing and establishing a cause for this condition. Aim: This study aims in showing that all trigeminal neuralgias are not secondary to neurovascular compression and other unusual causes needs to be looked for and assessed during an MRI examination. Materials and Methods: This was a cross-sectional observational study conducted at the Saveetha Medical College and Hospital, Chennai, India (from September 2015 to August 2017). This study included 70 patients presenting with clinical symptoms of unilateral trigeminal neuralgia. The MRI was done using a 1.5 Tesla, Philips multiva system. A high resolution 3D T2 DRIVE or 3D bFFE cranial nerve sequences were performed in addition to the routine Magnetic Resonance (MR) sequences. Patients with known dental issues which could cause the pain, and patients with intracranial tumours were excluded from the study. Results: Out of the 70 patients, we found that the majority (53) had a neurovascular compression, which ranged from a simple indentation by a tortuous Superior Cerebellar Artery (SCA) to full blown vertebrobasilar dolichoectasia. Out of the remaining cases, six were found to be secondary to benign intracranial hypertension with prominent Meckel’s caves. There were two cases who had an unusual finding of “Trigeminal pontine sign”, which were post infective (postherpetic being the most probable cause). Conclusion: By tailoring the MRI protocol in present study, and based on the clinical presentation, we can conclude that in most cases of clinically diagnosed trigeminal neuralgia, it could be possible to demonstrate a probable cause and consequently aid the clinician for appropriate management, and to select only the deserving cases for microvascular decompression.

Abstract Hepatitis C virus is a major cause of chronic liver diseases which can lead to permanent liver damage, hepatocellular carcinoma and death. The presently available treatment with interferon plus ribavirin, has limited benefits due to adverse side effects such as anemia, depression and "flu-like" symptoms. Needless to mention, the effectiveness of interferon therapy is predominantly, if not exclusively, limited to virus type 3a and 3b whereas in Europe and North America the majority of viral type is 1a and 2a. Due to the limited efficiency of current therapy, RNA interference (RNAi) a novel regulatory and powerful silencing approach for molecular therapeutics through a sequence-specific RNA degradation process represents an alternative option. Several reports have indicated the efficiency and specificity of synthetic and vector based siRNAs inhibiting HCV replication. In the present review, we focused that combination of siRNAs against virus and host genes will be a better option to treat HCV

Differentiation of antigen-activated B cells into pro-proliferative germinal center (GC) B cells depends on the activity of the transcription factors MYC and BCL6, and the epigenetic writers DOT1L and EZH2. GCB-like Diffuse Large B Cell Lymphomas (GCB-DLBCLs) arise from GCB cells and closely resemble their cell of origin. Given the dependency of GCB cells on DOT1L and EZH2, we investigated the role of these epigenetic regulators in GCB-DLBCLs and observed that GCB-DLBCLs synergistically depend on the combined activity of DOT1L and EZH2. Mechanistically, inhibiting both enzymes led to enhanced derepression of PRC2 target genes compared to EZH2 single treatment, along with the upregulation of BCL6 target genes and suppression of MYC target genes. The sum of all these alterations results in a 'cell identity crisis', wherein GCB-DLBCLs lose their pro-proliferative GC identity and partially undergo PC differentiation, a state associated with poor survival. In support of this model, combined epi-drugging of DOT1L and EZH2 prohibited the outgrowth of human GCB-DLBCL xenografts in vivo. We conclude that the malignant behavior of GCB-DLBCLs strongly depends on DOT1L and EZH2 and that combined targeting of both epigenetic writers may provide an alternative differentiation-based treatment modality for GCB-DLBCL., (Copyright © 2025 American Society of Hematology.)

This study investigates the metabolic disruptions caused by nicotine (NIC) exposure, with a particular focus on amino acid and lipid metabolism, and evaluates resveratrol (RSV) as a potential protective agent. Mice were divided into four groups: control (CON), NIC-exposed, NIC + RSV-treated, and RSV-only. NIC exposure resulted in significant weight loss, elevated glucose levels, altered lipid profiles, and organ damage, particularly in the liver and kidneys. Increased inflammation was evidenced by elevated levels of IL-6 and CRP. In contrast, RSV treatment mitigated these effects by improving lipid profiles, glycemic indices, and reducing inflammatory markers. Histopathological analysis confirmed reduced tissue damage in the NIC + RSV group compared to the NIC-alone group. Metabolomics analysis using LC-MS/MS revealed significant dysregulation in lipid, amino acid, and nucleotide metabolism in NIC-exposed mice. Fold-change analysis identified altered metabolites, including sphingomyelin 36:1;02 (p < 0.001), valine (p < 0.001), triacylglycerol 4:0-18:1 (p < 0.001), and ceramide 32:1;02 (p < 0.001). Amino acids such as arginine, phenylalanine, glutamic acid, tyrosine, and lysine, as well as NIC metabolites like nornicotine and cotinine, were identified, underscoring molecular fragmentation analysis findings. RSV treatment partially restored metabolic balance, highlighting its role as a metabolic modulator. This study underscores the therapeutic potential of RSV in alleviating NIC-induced metabolic dysfunctions by restoring lipid homeostasis and reducing inflammation. Purpose: Specific genetic factors might serve as markers for risk stratification of AMD progression, but their association with key features of AMD has not been fully elucidated. Thus, we investigated the association between overall and pathway-specific genetic risk scores (GRS) and lead loci ( ARMS2, CFH ) with AMD stages and features of high-risk nonlate AMD, including reticular pseudodrusen (RPD) and large drusen area (LDA)., Methods: We performed a cross-sectional analysis of data from the Rhineland Study, a population-based study in Bonn, Germany. We included 4016 individuals aged 50 years and older of European descent. GRS and pathway-specific subscores were constructed based on a large genome-wide association study of AMD. Subscores were generated based on gene-pathways associations (complement, extracellular matrix remodeling (ECM) and lipid metabolism). Associations were assessed using logistic and multinomial regression., Results: The mean age of participants was 63.36 years and 1813 (45.1%) were men. The GRS was positive in 48.1% of individuals and increased, but did not fully overlap, across AMD stages. Pathway-specific subscores increased across AMD stages except for the ECM subscore, which only showed a trend for increasing in late AMD. Increasing overall GRS was associated with RPD and LDA (OR [95%CI] for RPD: 1.70 [1.33-2.15], for LDA: 1.64 [1.29-2.07]) among individuals with AMD. Similarly, higher complement and ECM subscores was associated with RPD, while for LDA, only an association with complement subscore was observed., Conclusions: In a population-based setting, we confirmed higher genetic risk to be associated with more severe AMD and identified associations with high-risk features of intermediate AMD. Conjoint analyses suggested that high-risk features and late AMD might be differentially associated with genetic architecture in AMD, such as ECM remodeling. Incorporation of genetic information such as GRSs might improve AMD risk prediction strategies.

AMP-activated protein kinase (AMPK) is the master regulator of cellular energy which gets activated during energy stress and restores tissue homeostasis. AMPK is widely expressed in the pancreas and is involved in protein synthesis. In cerulein-induced acute pancreatitis (AP), diminished AMPK activity in the pancreatic tissue may be associated with pancreatic inflammation and oxidative stress. Our results demonstrated that berberine (BR) treatment produced significant decrease in plasma amylase and lipase levels and improved histopathological features in AP mice model. Myeloperoxidase (MPO) activity indicated that BR suppressed the infiltration of neutrophils in pancreas. BR treatment markedly decreased the levels of proinflammatory cytokines including interleukins (IL)-6, IL-1β, and tumor necrosis factor-α (TNF-α) via inhibition of nicotinamide adenine dinucleotide phosphate oxidase 2 (NOX2) expression. In addition, BR activates the nuclear factor erythroid 2-related factor 2 (Nrf2) signaling and inhibits cerulein-induced oxidative-nitrosative stress. Mechanistically, we found inhibition of AMPK activity in cerulein-induced AP, while BR-treated animals showed marked increase in the AMPK expression. Together, our study indicated that BR-mediated AMPK activation in pancreatic tissues demonstrated attenuation of cerulein-induced oxidative stress and inflammation. The continuous use of fossil fuels has a huge impact on climate change because they release CO 2 , which is a major greenhouse gas that causes 70-75% of global warming. Shale reserves could be used to store CO 2 to lower greenhouse gas emissions. This could happen mostly through adsorbed gas, which can make up about 85% of all shale gas. It is important to fully understand the CO 2 adsorption processes in shale, especially when using isothermal models, to get accurate estimates of storage capacity and predictions of how shale will behave. This work examines the application of several isothermal models, including Langmuir, Freundlich, Brunauer-Emmett-Teller, Dubinin-Radushkevich, Dubinin-Astakhov, Sips, Toth, and Ono-Kondo lattice models, to explore the adsorption of CO 2 on shale formations. The aim of this research work is to assess the efficiency of these models in forecasting CO 2 adsorption in different shale samples with specific mineral compositions, total organic content (TOC), surface areas, and pore geometry at 298 K and up to 2 MPa. This review provides a state-of-the-art knowledge on the constraints of existing models and proposes adaptations, such as integrating density-dependent correction factors and hybrid modeling techniques, to enhance precision during numerical simulation work. Furthermore, the possible incorporation of molecular dynamic (MD) simulations with experimental data is suggested to improve the understanding of the CO 2 adsorption in the geological rock at the molecular scale. The spectrum of infections caused by methicillin-resistant Staphylococcus aureus (MRSA) ranges from minor conditions to potentially life-threatening diseases. The rising antibiotic resistance in MRSA often leads to treatment failures, underscoring the urgent need for novel eradication strategies. This study focuses on isolating MRSA from burn patients, determining its antibiogram profile, and isolating and characterizing bacteriophages from sewage water that target MRSA, alongside conducting genomic analysis of the phages. A total of 70 samples were collected from burn patients, with MRSA identification and characterization performed using a combination of biochemical and molecular techniques, as well as antibiotic sensitivity testing. Based on host range analysis, a specific phage (phage-3) was selected for detailed characterization, including proteomic analysis, genetic mapping, phylogenetic studies, and analysis of open reading frames (ORFs) and motifs. The prevalence of MRSA in the samples was found to be 28.6%. Antibiotic susceptibility tests indicated that 94% of the MRSA isolates were sensitive to tobramycin and gentamicin, while vancomycin exhibited the lowest sensitivity, with only 2% effectiveness. Using the soft agar overlay method, three bacteriophages (phage-1, phage-2, and phage-3) were successfully isolated from sewage water. Among these, phage-3 exhibited the broadest host range. Further analysis showed that phage-3 demonstrated optimal activity at pH levels between 6 and 8, and within a temperature range of 20-40 °C. Phage-3 also displayed a rapid adsorption phase within the first 0-5 min, and its one-step growth curve revealed a latent period lasting up to 30 min, followed by a significant increase in titer from 30 to 50 min. Proteomic analysis of phage-3 identified the presence of 33 kDa and 65 kDa proteins. Phylogenetic analysis showed that phage-3 shares 96.6% similarity with Mammallicoccus phage vB&#95;MscM-PMS3. Herein, we investigated the stereochemical effects of 4'-methyl substitution on A 3 adenosine receptor (A 3 AR) ligands by synthesizing and evaluating a series of truncated 4'-thioadenosine derivatives featuring 4'-α-methyl, 4'-β-methyl, and 4',4'-dimethyl substitutions. We successfully synthesized these derivatives, using the stereoselective addition of an organometallic reagent, KSAc-mediated sulfur cyclization, and Vorbrüggen condensation. Binding assays demonstrated that the 4'-β-methyl substitution conferred the highest affinity for A 3 AR, with compound 1 h exhibiting a K i  = 3.5 nM, followed by the 4',4'-dimethyl and 4'-α-methyl substitutions. Notably, despite the absence of the 5'-OH group, compound 1 h unexpectedly displayed partial agonism. Computational docking studies indicated that compound 1 h, the β-methyl derivative, adopted a South conformation and maintained strong interactions within the receptor, including a critical interaction with Thr94, a residue known to be notable for agonistic effects. Conversely, compound 2 h, the α-methyl derivative, also adopted a South conformation but resulted in a flattened structure that hindered interactions with Thr94 and Asn250. The dimethyl derivative 3 h exhibited steric clashes with Thr94, contributing to a reduction in binding affinity. However, the docking results for 3 h indicated a North conformation, suggesting that the change in sugar conformation due to the additional 4'-methyl group altered the angle between the α-methyl group and the sugar plane, enabling binding despite the increased steric bulk. These findings suggest that not only do the substituents and their stereochemistry influence receptor-ligand interactions, but the conformation and the resulting spatial orientation of the substituents also play a crucial role in modulating receptor-ligand interaction. The skin is at risk for injury to external factors since it serves as the body's first line of defense against the external environment. Hydrogels have drawn much interest due to their intrinsic extracellular matrix (ECM) properties and their biomimetic, structural, and durable mechanical characteristics. Hydrogels have enormous potential use in skin wound healing due to their ability to deliver bioactive substances easily. In this study, composite hydrogels were developed by blending guar gum (GG), polyvinyl alcohol (PVA), and carboxymethyl cellulose (CMC) with crosslinker TEOS for skin wound treatment. The structural, surface morphology, surface roughness, and stability features of the composite hydrogels were characterized by several techniques, such as FTIR, SEM-EDX, AFM, and DSC. The increasing ZiF-8 causes more surface roughness, with decreased swelling in different media (Aqueous > PBS > NaCl). The increasing ZiF-8 amount causes less hydrophilic behavior and biodegradation with increasing gel fraction. The cytocompatibility of Zinc imidazolate framework-8 (ZiF-8) based composites was evaluated against fibroblast cell lines by cell viability, proliferation, and cell morphology. The increasing ZiF-8 caused more cell viability and proliferation with proper cell morphology. Hence, the results show that synthesized composite hydrogels may be a potential candidate for numerous wound repair applications.

Here, we report for the first time, green-synthesized selenium nanoparticles (SeNPs) using pharmacologically potent herb of Polygonum bistorta Linn. for multiple biomedical applications. In the study, a facile and an eco-friendly approach is utilized for synthesis of SeNPs using an aqueous roots extract of P. bistorta Linn. followed by extensive characterization via Fourier transform infrared spectroscopy, X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and Energy Dispersive X-Ray (EDX) analysis. The XRD and FTIR data determine the phase composition and successful capping of plant extract onto the surface of NPs while SEM and TEM micrographic examination reveals the elliptical and spherical morphology of the particles with a mean size of 69 ± 23 nm. After comprehensive characterization, the NPs are investigated for antifungal, antibacterial, antileishmanial, antioxidant, and biocompatibility properties. The study reveals that Polygonum bistorta Linn. synthesized SeNPs exhibit significant antibacterial and antifungal activities with Staphylococcus aureus and Fusarium oxysporum inducing the highest zone of inhibition of 14 ± 1.0 mm and 20 ± 1.2 mm, respectively at the concentration of 40 mg/mL. The NPs are also found to have antiparasitic potential against promastigote and amastigote forms of Leishmania tropica. Furthermore, the NPs are discovered to have excellent potential in neutralizing harmful free radicals thus exhibiting considerable antioxidant potential. Most importantly, Polygonum bistorta Linn. synthesized SeNPs showed substantial compatibility against blood cells in vitro studies, which signifies the nontoxic nature of the NPs. The study thus concludes that medicinally important Polygonum bistorta Linn. roots can be utilized as an eco-friendly, sustainable, and green source for the synthesis of pharmacologically potent selenium nanoparticles., Competing Interests: Declarations. Cerebral small vessel disease (cSVD) is a leading cause of stroke and dementia. Genetic risk loci for white matter hyperintensities (WMH), the most common MRI-marker of cSVD in older age, were recently shown to be significantly associated with white matter (WM) microstructure on diffusion tensor imaging (signal-based) in young adults. To provide new insights into these early changes in WM microstructure and their relation with cSVD, we sought to explore the genetic underpinnings of cutting-edge tissue-based diffusion imaging markers across the adult lifespan. We conducted a genome-wide association study of neurite orientation dispersion and density imaging (NODDI) markers in young adults (i-Share study: N = 1 758, (mean[range]) 22.1[18-35] years), with follow-up in young middle-aged (Rhineland Study: N = 714, 35.2[30-40] years) and late middle-aged to older individuals (UK Biobank: N = 33 224, 64.3[45-82] years). We identified 21 loci associated with NODDI markers across brain regions in young adults. The most robust association, replicated in both follow-up cohorts, was with Neurite Density Index (NDI) at chr5q14.3, a known WMH locus in VCAN. Two additional loci were replicated in UK Biobank, at chr17q21.2 with NDI, and chr19q13.12 with Orientation Dispersion Index (ODI). Transcriptome-wide association studies showed associations of STAT3 expression in arterial and adipose tissue (chr17q21.2) with NDI, and of several genes at chr19q13.12 with ODI. Genetic susceptibility to larger WMH volume, but not to vascular risk factors, was significantly associated with decreased NDI in young adults, especially in regions known to harbor WMH in older age. Individually, seven of 25 known WMH risk loci were associated with NDI in young adults. In conclusion, we identified multiple novel genetic risk loci associated with NODDI markers, particularly NDI, in early adulthood. These point to possible early-life mechanisms underlying cSVD and to processes involving remyelination, neurodevelopment and neurodegeneration, with a potential for novel approaches to prevention., (© 2024. This study introduces a novel Blind Image Quality Assessment (BIQA) approach leveraging a multi-stream spatial and channel attention model. Our method addresses challenges posed by diverse image content and distortions by integrating feature maps from two distinct backbones. Through spatial and channel attention mechanisms, our algorithm prioritizes regions of interest, enhancing its ability to capture crucial image details. Extensive evaluations on four benchmark datasets demonstrate superior performance compared to existing methods, closely aligning with human perceptual assessment. Objectives: Undergoing women of pregnancy termination (PT) significantly faces the problem of physical and mental health. This study aims to assess the prevalence and socioeconomic disparity factors of PT in Bangladesh. This study analyzed data from the 2017-18 Bangladesh Demographic and Health Survey. Researchers employed chi-square tests to identify relationships between categorical variables and logistic regression to pinpoint factors associated with PT. To assess the socioeconomic variation of PT, the analysis utilized concentration curves, concentration indices, and decomposition techniques., Results: The study found that 21.0% of reproductive-aged women in Bangladesh had ever terminated a pregnancy. Our study revealed that women from Chittagong and Sylhet regions, with wealthier backgrounds, aged 30 or older, employed in business, taking short birth intervals, and whose husband/partner was 35 or older, were more likely to have had a pregnancy termination with statistical significance (p < 0.05). Besides, concentration curves showed a higher prevalence of PT among wealthier women (CCI = 0.029, p < 0.001). Decomposition of this inequality revealed that a woman's wealth status was the largest contributor (74.98%) to the observed disparities, followed by exposure to mass media (41.82%), place of residence (34.35%), occupation (24.81%), and preceding birth interval (6.53%). Ulnar dimelia often presents with restricted elbow movement. To develop a treatment strategy to improve elbow mobility, a detailed understanding of the skeletal deformity is needed. We created 3-D models of the cartilage and bone in the elbows of four children with ulnar dimelia by segmentation of MRI scans. We analysed the anatomy and performed a kinematic analysis. In all four children, the distal humerus was triangular in shape and consisted of a medial epicondyle, an enlarged lateral epicondyle and an anterior epicondyle. By comparing the 3-D images with conventional radiographs, we found that the projection of the enlarged lateral epicondyle created two longitudinal lines, which is a radiographic hallmark feature of the triangular-shaped distal humerus. In addition, during elbow flexion, the radial forearm bone slides along the lateral trochlea rather than rotates. This knowledge can aid surgeons to plan treatment for improving elbow mobility in ulnar dimelia. Introduction: Blood-derived microRNAs (miRNAs) are potential candidates for detecting and preventing subclinical cognitive dysfunction. However, replication of previous findings and identification of novel miRNAs associated with cognitive domains, including their relation to brain structure and the pathways they regulate, are still lacking., Methods: We examined blood-derived miRNAs and miRNA co-expression clusters in relation to cognitive domains, structural magnetic resonance imaging measures, target gene expression, and genetic variants in 2869 participants of a population-based cohort., Results: Five previously identified and 14 novel miRNAs were associated with cognitive domains. Protein-based wound dressings have garnered increasing interest in recent years owing to their distinct physical, chemical, and biological characteristics. The intricate molecular composition of proteins gives rise to unique characteristics, such as exceptional biocompatibility, biodegradability, and responsiveness, which contribute to the promotion of wound healing. Wound healing is an intricate and ongoing process influenced by multiple causes, and it consists of four distinct phases. Various treatments have been developed to repair different types of skin wounds, thanks to advancements in medical technology and the recognition of the diverse nature of wounds. This review has literature reviewed within the last 3-5 years-the recent progress and development of protein in wound dressings and the fundamental properties of an ideal wound dressing. Herein, the recent strides in protein-based state-of-the-art wound dressing emphasize the significant challenges and summarize future perspectives for wound healing applications.

For centuries, cannabinoids have been utilized for their medicinal properties, particularly in Asian and South-Asian countries. Cannabis plants, known for their psychoactive and non-psychoactive potential, were historically used for spiritual and remedial healing. However, as cannabis became predominantly a recreational drug, it faced prohibition. Recently, the therapeutic potential of cannabinoids has sparked renewed research interest, extending their use to various medical conditions, including cancer. This review aims to highlight current data on the involvement of cannabinoids in cancer signaling pathways, emphasizing their potential in cancer therapy and the need for further investigation into the underlying mechanisms. A comprehensive literature review was conducted using databases such as PubMed/MedLine, Google Scholar, Web of Science, Scopus, and Embase. The search focused on peer-reviewed articles, review articles, and clinical trials discussing the anticancer properties of cannabinoids. Inclusion criteria included studies in English on the mechanisms of action and clinical efficacy of cannabinoids in cancer. Cannabinoids, including Δ9-THC, CBD, and CBG, exhibit significant anticancer activities such as apoptosis induction, autophagy stimulation, cell cycle arrest, anti-proliferation, anti-angiogenesis, and metastasis inhibition. Clinical trials have demonstrated cannabinoids' efficacy in tumor regression and health improvement in palliative care. However, challenges such as variability in cannabinoid composition, psychoactive effects, regulatory barriers, and lack of standardized dosing remain. Cannabinoids show promising potential as anticancer agents through various mechanisms. Further large-scale, randomized controlled trials are essential to validate these findings and establish standardized therapeutic protocols. Neurofilament light chain (NfL) levels in circulation have been established as a sensitive biomarker of neuro-axonal damage across a range of neurodegenerative disorders. Elucidation of the genetic architecture of blood NfL levels could provide new insights into molecular mechanisms underlying neurodegenerative disorders. In this meta-analysis of genome-wide association studies (GWAS) of blood NfL levels from eleven cohorts of European ancestry, we identify two genome-wide significant loci at 16p12 (UMOD) and 17q24 (SLC39A11). We observe association of three loci at 1q43 (FMN2), 12q14, and 12q21 with blood NfL levels in the meta-analysis of African-American ancestry. In the trans-ethnic meta-analysis, we identify three additional genome-wide significant loci at 1p32 (FGGY), 6q14 (TBX18), and 4q21. In the post-GWAS analyses, we observe the association of higher NfL polygenic risk score with increased plasma levels of total-tau, Aβ-40, Aβ-42, and higher incidence of Alzheimer's disease in the Rotterdam Study. Furthermore, Mendelian randomization analysis results suggest that a lower kidney function could cause higher blood NfL levels. The global demand for an enhanced quality of life and extended lifespan has driven significant advancements in tissue engineering and regenerative medicine. These fields utilize a range of interdisciplinary theories and techniques to repair structurally impaired or damaged tissues and organs, as well as restore their normal functions. Nevertheless, the clinical efficacy of medications, materials, and potent cells used at the laboratory level is always constrained by technological limitations. A novel platform known as adaptable microneedles has been developed to address the abovementioned issues. These microneedles offer a solution for the localized distribution of various cargos while minimizing invasiveness. Microneedles provide favorable patient compliance in clinical settings due to their effective administration and ability to provide a painless and convenient process. In this review article, we summarized the most recent development of microneedles, and we started by classifying various microneedle systems, advantages, and fundamental properties. Subsequently, it provides a comprehensive overview of different types of microneedles, the material used to fabricate microneedles, the fundamental properties of ideal microneedles, and their applications in tissue engineering and regenerative medicine, primarily focusing on preserving and restoring impaired tissues and organs. In view of their exceptional approach, excellent inherent biocompatibility and biodegradability properties, and interaction with the local extracellular matrix, protein-based polymers have received attention in bone tissue engineering, which is a multidisciplinary field that repairs and regenerates fractured bones. Bone is a multihierarchical complex structure, and it performs several essential biofunctions, including maintaining mineral balance and structural support and protecting soft organs. Protein-based polymers have gained interest in developing ideal scaffolds as emerging biomaterials for bone fractured healing and regeneration, and it is challenging to design ideal bone substitutes as perfect biomaterials. Several protein-based polymers, including collagen, keratin, gelatin, serum albumin, etc., are potential materials due to their inherent cytocompatibility, controlled biodegradability, high biofunctionalization, and tunable mechanical characteristics. While numerous studies have indicated the encouraging possibilities of proteins in BTE, there are still major challenges concerning their biodegradability, stability in physiological conditions, and continuous release of growth factors and bioactive molecules. Robust scaffolds derived from proteins can be used to replace broken or diseased bone with a biocompatible substitute; proteins, being biopolymers, provide excellent scaffolds for bone tissue engineering. Herein, recent developments in protein polymers for cutting-edge bone tissue engineering are addressed in this review within 3-5 years, with a focus on the significant challenges and future perspectives. The first section discusses the structural fundamentals of bone anatomy and ideal scaffolds, and the second section describes the fabrication techniques of scaffolds. The third section highlights the importance of proteins and their applications in BTE. Hence, the recent development of protein polymers for state-of-the-art bone tissue engineering has been discussed, highlighting the significant challenges and future perspectives.

Bone is a dynamic tissue that can always regenerate itself through remodeling to maintain biofunctionality. This tissue performs several vital physiological functions. However, bone scaffolds are required for critical-size damages and fractures, and these can be addressed by bone tissue engineering. Bone tissue engineering (BTE) has the potential to develop scaffolds for repairing critical-size damaged bone. BTE is a multidisciplinary engineered scaffold with the desired properties for repairing damaged bone tissue. Herein, we have provided an overview of the common carbohydrate polymers, fundamental structural, physicochemical, and biological properties, and fabrication techniques for bone tissue engineering. We also discussed advanced biofabrication strategies and provided the limitations and prospects by highlighting significant issues in bone tissue engineering. Importance: Olfactory dysfunction is among the earliest signs of many age-related neurodegenerative diseases and has been associated with increased mortality in older adults; however, its genetic basis remains largely unknown., Objective: To identify the genetic loci associated with olfactory dysfunction in the general population., Design Setting and Partiicipants: This genome-wide association study meta-analysis (GWMA) included participants of European ancestry (N = 22,730) enrolled in four different large population-based studies, followed by a multi-ancestry GWMA including participants of African ancestry (N = 1,030). The data analysis was performed from March 2023 through June 2024., Exposures: Genome-wide single nucleotide polymorphisms., Main Outcomes and Measures: Olfactory dysfunction was the outcome and assessed using a 12-item smell identification test., Results: GWMA revealed a novel genome-wide significant locus (tagged by rs11228623 at 11q12) associated with olfactory dysfunction. Gene-based analysis revealed a high enrichment for olfactory receptor genes in this region. Phenome-wide association studies demonstrated associations between genetic variants related to olfactory dysfunction and blood cell counts, kidney function, skeletal muscle mass, cholesterol levels and cardiovascular disease. Using individual-level data, we also confirmed and quantified the strength of these associations on a phenotypic level. Mosquitoes serve as vectors for various diseases like malaria, dengue fever, yellow fever, and lymphatic filarial diseases causing significant global health problems, highlighting the importance of vector control. The study was conducted to assess the effectiveness of nanoformulated clothianidin and chlorfenapyr insecticides treated with ATSB in controlling three mosquito strains. The development of a natural thiolated polymer-coated ATSB nano formulation involved incorporating nano-carriers to deliver insecticides. Field- collected mosquito strains were subjected to laboratory-based bioassays using 1 % and 1.5 % concentrations of each conventionally used and nanoformulated insecticide with ATSB solution. Adult mosquitoes were left overnight to contact with N-ATSB and efficacy was recorded after 36 and 72 h. The results showed that nanoformulated chlorfenapyr was significantly more effective as compared to clothianidin against An. funestus and Cx. quinquefasciatus but the results were not significantly different against An. coluzzii (100 %). An. coluzzii was found to be the most susceptible strain followed by An. funestus and showed 100 % and ∼ 98 % mortality against nanoformulated chlorfenapyr (1.5 %). Nanoformulated clothianidin induced more than 92 % and ∼ 100 % mortality against An. funestus and An. coluzzii respectively. However, Cx. quinquefasciatus significantly showed less mortality against nanoformulated clothianidin (88 %) and chlorfenapyr (>95 %) as compared to Anopheline strains. Background Gamma-glutamyl transferase (GGT) mediates intracellular uptake of glutathione which is a known antioxidant. GGT levels are found to be elevated in conditions of oxidative stress. Ischemic stroke results in anoxic injury, which liberates free radicals, causing glutathione to rise, which may be accompanied by a rise in serum GGT levels. This study aimed to compare serum GGT levels in acute ischemic stroke patients with normal controls and to ascertain the relation of serum GGT levels with National Institute of Health Stroke Scale (NIHSS) scores. Materials and methods This cross-sectional study was carried out in a tertiary care hospital in South India from August 2023 to February 2024. The study included 57 patients who presented with acute ischemic stroke within 24 hours of onset and 57 age- and sex-matched controls. The serum GGT levels of the cases were compared with age- and sex-matched controls using an independent t-test. Mean serum GGT levels were compared among groups with varying NIHSS scores and different locations of infarction using the ANOVA test. Serum GGT levels were also compared based on age, gender, and various comorbidities. Results The mean serum GGT levels were significantly increased (p < 0.0001) in acute ischemic stroke patients, 43.96 ± 28.02 (mean ± SD), when compared to controls, 26.14 ± 5.93 (mean ± SD). The difference in serum GGT levels with NIHSS scores of 5-15 (moderate strokes) with 34.17 ± 18.39 (mean ± SD), 16-20 (moderate-severe strokes) with 46.64 ± 21.95 (mean ± SD), and >21 (severe stroke) with 84.62 ± 39.35 (mean ± SD) was significant (p < 0.00001). Serum GGT levels were not significant while comparing age, gender, location of infarction, type 2 diabetes mellitus, and hypertension. Conclusion Serum GGT levels were significantly elevated in acute ischemic stroke patients within 24 hours of presentation. Serum GGT levels were significantly elevated with increasing severity of stroke as calculated by NIHSS scores at the time of presentation. Pakistan is facing energy crises due to localized shortages, market manipulation, infrastructure disruption, rising demand, governance issues, climate and geopolitical events. In this situation Demand Side Management (DSM) is a promising solution to overcome the problem of energy crises. DSM strategy helps to manage consumer demand through energy conservation rather than to addition of new power capacity. In this study, Low Emissions Analysis Platform (LEAP) develops an energy model for Pakistan for the period 2022-2050. Three scenarios has been to constructed namely Baseline (BAS), Green Energy Policy (GEP), and Energy Efficiency (ENE) to predict the future energy demand, production, carbon emissions and the investment cost which covers capital, operational and maintenance costs. The model results suggest that DSM targets should be achieved through the implementation of ENE scenario. Predicted energy production and consumption under the ENE scenario are substantially less than those under the BAS scenario. The country can meet its 635.83,000 GWh energy demand with its 747.15,000 GWh energy production. Non-renewable sources produce 171.27,000 GWh, whilst renewable sources produce 575.88,000 GWh. According to this scenario, by 2050, CO 2 emissions will be produced around 93.16 million metric tons, requiring an investment cost of $46.80 billion for building new power capacity. Wound healing is a critical but complex biological process of skin tissue repair and regeneration resulting from various systems working together at the cellular and molecular levels. Quick wound healing and the problems associated with traditional wound repair techniques are being overcome with multifunctional materials. Over time, this research area has drawn significant attention. Metal-organic frameworks (MOFs), owning to their peculiar physicochemical characteristics, are now considered a promising class of well-suited porous materials for wound healing in addition to their other biological applications. This detailed literature review provides an overview of the latest developments in MOFs for wound healing applications. We have discussed the synthesis, essential biomedical properties, wound-healing mechanism, MOF-based dressing materials, and their wound-healing applications. The possible major challenges and limitations of MOFs have been discussed, along with conclusions and future perspectives. This overview of the literature review addresses MOFs-based wound healing from several angles and covers the most current developments in the subject. Background: Agriculture is one of the most crucial assets of any country, as it brings prosperity by alleviating poverty, food shortages, unemployment, and economic instability. The entire process of agriculture comprises many sectors, such as crop cultivation, water irrigation, the supply chain, and many more. During the cultivation process, the plant is exposed to many challenges, among which pesticide attacks and disease in the plant are the main threats. Diseases affect yield production, which affects the country's economy. Over the past decade, there have been significant advancements in agriculture; nevertheless, a substantial portion of crop yields continues to be compromised by diseases and pests. Early detection and prevention are crucial for successful crop management., Methods: To address this, we propose a framework that utilizes state-of-the-art computer vision (CV) and artificial intelligence (AI) techniques, specifically deep learning (DL), for detecting healthy and unhealthy cotton plants. Our approach combines DL with feature extraction methods such as continuous wavelet transform (CWT) and fast Fourier transform (FFT). The detection process involved employing pre-trained models such as AlexNet, GoogLeNet, InceptionV3, and VGG-19. Implemented models performance was analysed based on metrics such as accuracy, precision, recall, F1-Score, and Confusion matrices. Moreover, the proposed framework employed ensemble learning framework which uses averaging method to fuse the classification score of individual DL model, thereby improving the overall classification accuracy., Results: During the training process, the framework achieved better performance when features extracted from CWT were used as inputs to the DL model compared to features extracted from FFT. Among the learning models, GoogleNet obtained a remarkable accuracy of 93.4% and a notable F1-score of 0.953 when trained on features extracted by CWT in comparison to FFT-extracted features. It was closely followed by AlexNet and InceptionV3 with an accuracy of 93.4% and 91.8% respectively. To further improve the classification accuracy, ensemble learning framework achieved 98.4% on the features extracted from CWT as compared to feature extracted from FFT., Conclusion: The results show that the features extracted as scalograms more accurately detect each plant condition using DL models, facilitating the early detection of diseases in cotton plants. The modified Benjamin-Bona-Mahony (mBBM) model is utilized in the optical illusion field to describe the propagation of long waves in a nonlinear dispersive medium during a visual illusion (Khater 2021). This article investigates the mBBM equation through the utilization of the rational [Formula: see text]-expansion technique to derive new analytical wave solutions. The analytical solutions we have obtained comprise hyperbolic, trigonometric, and rational functions. Some of these exact solutions closely align with previously published results in specific cases, affirming the validity of our other solutions. To provide insights into diverse wave propagation characteristics, we have conducted an in-depth analysis of these solutions using 2D, 3D, and density plots. We also investigated the effects of various parameters on the characteristics of the obtained wave solutions of the model. Moreover, we employed the techniques of linear stability to perform stability analysis of the considered model. Additionally, we have explored the stability of the associated dynamical system through the application of phase plane theory. This study presents a numerical study of a highly sensitive photonic crystal fiber (PCF) surface plasmon resonance (SPR) sensor capable of detecting five types of cancer and bacterial contamination in water. By precisely arranging only two air holes in a single channel of an elliptical-shaped PCF, the sensor maximizes interaction between core-guided modes and surface plasmon polaritons (SPP) along the fiber. Evaluation using COMSOL Multiphysics simulation software, based on finite element method (FEM), demonstrates outstanding sensor performance across a wide refractive index (RI) range (1.33 to 1.43). With a maximum wavelength sensitivity (WS) of 188,000 nm/RIU and amplitude sensitivity (AS) of -22,377.99 RIU -1 , the sensor achieveStructural Design and Methodologys a sensor resolution (SR) of 5.3191 × 10 -7 RIU and figure of merit (FOM) of 854.55 RIU -1 . Notably, it exhibits AS and WS values tailored for specific cancer cell types and water contamination. These results endorse the sensor's potential in diverse biological and molecular analyte RI detection applications within the visible to near-infrared (VNIR) range (0.55 to 4 µm), offering high sensitivity, affordability, wide sensing range, good linearity, low propagation loss, and simplicity in construction.

Sugarcane smut is the most damaging disease that is present almost across the globe, causing mild to severe yield losses depending upon the cultivar types, pathogen races and climatic conditions. Cultivation of smut-resistant cultivars is the most feasible and economical option to mitigate its damages. Previous investigations revealed that there is a scarcity of information on early detection and effective strategies to suppress etiological agents of smut disease due to the characteristics overlapping within species complexes. In this study, 104 sugarcane cultivars were screened by artificial inoculation with homogenate of all possible pathogen races of Sporisorium scitamineum during two consecutive growing seasons. The logistic smut growth pattern and the disease intrinsic rate were recorded by disease growth curve. Variable levels of disease incidence i.e., ranging from 0 to 54.10% were observed among these sugarcane cultivars. Besides, pathogen DNA in plant shoots of all the cultivars was successfully amplified by PCR method using smut-specific primers except 26 cultivars which showed an immune reaction in the field trial. Furthermore, the plant germination and tillering of susceptible sugarcane cultivars were greatly influenced by pathogen inoculation. In susceptible cultivars, S. scitamineum caused a significant reduction in setts germination, coupled with profuse tillering, resulting in fewer millable canes. Correlation analysis demonstrated that there was a positive relationship between reduction in setts germination and increase in the number of tillers. The present study would be helpful for the evaluation of smut resistance in a wide range of sugarcane germplasm, especially from the aspects of setts germination and tillers formation, and it also screened out several excellent germplasm for potential application in sugarcane breeding., (© 2024. Background Bone marrow examination (BME) is an indispensable diagnostic tool to evaluate various hematological and non-hematological disorders. Bone marrow aspirate cytology and bone marrow trephine biopsy, even though performed simultaneously, are assessed at different points in time due to different processing methods. Aims and objective This study aims to assess and compare the role of bone marrow aspiration and trephine biopsy to formulate an effective and rapid method for diagnosing a wide spectrum of various hematological and non-hematological disorders. Materials and methods The approach of our study was a hospital-based prospective study conducted on 200 patients over a period of 1 year. The role of bone marrow aspiration and a trephine biopsy is to formulate an effective and rapid method for diagnosing a wide spectrum of hematological and non-hematological disorders. Results In our study, a total of 200 cases were studied, of whom 119 patients were male and 81 were female. The most common finding was erythroid hyperplasia, comprising 40 (20%) cases, followed by hypoplastic marrow, comprising 28 (14%) cases. Subsequently, there were 19 (9.5%) cases of acute leukemia, while 15 (7.5%) cases of chronic myeloid leukemia (CML) in the chronic phase were found. In our study, bone marrow aspirate and bone marrow trephine biopsy were found to positively correlate in 137 (68.5%) of the cases. Conclusion Bone marrow aspiration alone is sufficient for the diagnosis of megaloblastic anemia and most of the hematological malignancies. Bone marrow trephine biopsy is more appropriate for the detection of disorders of focal marrow involvement such as lymphoproliferative disorders and staging of lymphomas, metastatic cancers, granulomatous lesions, and hypoplastic marrow. The Fanconi anemia (FA) repair pathway governs repair of highly genotoxic DNA interstrand crosslinks (ICLs) and relies on translesion synthesis (TLS). TLS is facilitated by REV1 or site-specific monoubiquitination of proliferating cell nuclear antigen (PCNA) (PCNA-Ub) at lysine 164 (K164). A Pcna K164R/K164R but not Rev1 -/- mutation renders mammals hypersensitive to ICLs. Besides the FA pathway, alternative pathways have been associated with ICL repair (1, 2), though the decision making between those remains elusive. To study the dependence and relevance of PCNA-Ub in FA repair, we intercrossed Pcna K164R/+ ; Fancg -/+ mice. A combined mutation ( Pcna K164R/K164R ; Fancg -/- ) was found embryonically lethal. RNA-seq of primary double-mutant (DM) mouse embryonic fibroblasts (MEFs) revealed elevated levels of replication stress-induced checkpoints. To exclude stress-induced confounders, we utilized a Trp53 knock-down to obtain a model to study ICL repair in depth. Regarding ICL-induced cell toxicity, cell cycle arrest, and replication fork progression, single-mutant and DM MEFs were found equally sensitive, establishing PCNA-Ub to be critical for FA-ICL repair. Immunoprecipitation and spectrometry-based analysis revealed an unknown role of PCNA-Ub in excluding mismatch recognition complex MSH2/MSH6 from being recruited to ICLs. In conclusion, our results uncovered a dual function of PCNA-Ub in ICL repair, i.e. exclude MSH2/MSH6 recruitment to channel the ICL toward canonical FA repair, in addition to its established role in coordinating TLS opposite the unhooked ICL., (© The Author(s) 2024. Contact resistance is a multifaceted challenge faced by the 2D materials community. Large Schottky barrier heights and gap-state pinning are active obstacles that require an integrated approach to achieve the development of high-performance electronic devices based on 2D materials. In this work, we present semiconducting PtSe 2 field effect transistors with all-van-der-Waals electrode and dielectric interfaces. We use graphite contacts, which enable high I ON / I OFF ratios up to 10 9 with currents above 100 μA μm -1 and mobilities of 50 cm 2  V -1  s -1 at room temperature and over 400 cm 2  V -1  s -1 at 10 K. The devices exhibit high stability with a maximum hysteresis width below 36 mV nm -1 . The contact resistance at the graphite-PtSe 2 interface is found to be below 700 Ω μm. Our results present PtSe 2 as a promising candidate for the realization of high-performance 2D circuits built solely with 2D materials.

Suicidal hanging is common in Pakistan and is considered a serious challenge globally. We conducted a content analysis of reports about suicidal hangings published in six English-language newspapers in Pakistan between 1 January 2021 and 31 December 2022, employing a web-searching technique.124 news reports were yielded; suicidal authors were males (68.5%), Muslims (69.4%), and unmarried (31.4%), aged 16- 30 years old. Ligatures employed in hanging suicides were ropes (28.2%), fabrics (20.9%), and electric wires (20.1%). The most frequent leverage-points selected in suicides were ceiling fans (37%) followed by trees (11.2%). Familial discords (19.2%) and poverty (9.6%) were the leading risk factors for hanging suicide. Most of the suicides (58%) occurred equally in the spring (29%) and autumn (29%) months. These evidences suggest that psychological interventions on families are crucial. Also, dealing with poverty and other social consequences in Pakistan as in the rest of the world may probably reduce the rate of yearly suicides.

Background: Typhoid fever is a fatal disease in humans that is caused by Salmonella typhi. S. typhi infections need immediate antibiotic therapy, and their extensive use has led to multidrug-resistant (MDR) pathogens. The use of bacteriophages is becoming a new way to treat these resistant bacteria. This research was directed to bacteriophage isolation against S. typhi and to determine phage-antibiotic synergism., Aims: To isolate bacteriophages targeting S. typhi, the causative agent of typhoid fever, and investigate their potential synergistic effects when combined with antibiotics., Study Design: A cross-sectional study., Methods: The Widal test was positive; twenty diarrheal stool samples were taken, and for confirmation of S. typhi, different biochemical tests were performed. The disc-diffusion technique was used to determine antimicrobial resistance, and the double agar overlay method was used for bacteriophage isolation from sewage water against S. typhi. To test antibiotic-phage synergism, the S. typhi bacteria was treated by phages together with varying antibiotic concentrations., Results: Eleven samples were positive for S. typhi with black colonies on SS-agar. These were catalase and MR positive with alkali butt on TSI. Clear plaques were observed after the agar overlay. Isolated phages were stable at various pH and temperature levels. Synergism was observed on agar plate. The zone was enlarged when phages were combined with bacterial lawn culture and ciprofloxacin disk. Bacterial growth inhibition had a significant p-value of 0.03 in titration plates, with the phage-ciprofloxacin combination being more effective than the phage and antibiotic alone., Conclusion: The study highlights the synergistic effects of isolated bacteriophages with antibiotics, which are not only effective against S. typhi infection but also decrease antibiotic resistance., (© 2023.
