Abstract

The most common primary intraocular malignant tumour is choroidal melanoma. Reported incidence of intraocular melanoma is less than 1 per 100,000. Liver is the most frequent site of metastasis, and involvement of the other sites generally occurs in association with liver metastasis. The present case had an unusual presentation of, rapidly refilling massive ascites and right pleural effusion, following exenteration of the right eye 4 years earlier for primary choroid melanoma. To the best of our knowledge this is the first case of intra abdominal melanotic rapidly refilling ascites following primary choroidal melanoma masquerading as an ovarian tumour. We have discussed the atypical presentation, difficulties in arriving at a diagnosis preoperatively, intraoperative dilemmas and the post operative management.

Abstract

Replicative DNA polymerases are blocked by nearly all types of DNA damage. The resulting DNA replication stress threatens genome stability. DNA replication stress is also caused by depletion of nucleotide pools, DNA polymerase inhibitors, and DNA sequences or structures that are difficult to replicate. Replication stress triggers complex cellular responses that include cell cycle arrest, replication fork collapse to one-ended DNA double-strand breaks, induction of DNA repair, and programmed cell death after excessive damage. Replication stress caused by specific structures (e.g., G-rich sequences that form G-quadruplexes) is localized but occurs during the S phase of every cell division. This review focuses on cellular responses to widespread stress such as that caused by random DNA damage, DNA polymerase inhibition/nucleotide pool depletion, and R-loops. Another form of global replication stress is seen in cancer cells and is termed oncogenic stress, reflecting dysregulated replication origin firing and/or replication fork progression. Replication stress responses are often dysregulated in cancer cells, and this too contributes to ongoing genome instability that can drive cancer progression. Nucleases play critical roles in replication stress responses, including MUS81, EEPD1, Metnase, CtIP, MRE11, EXO1, DNA2-BLM, SLX1-SLX4, XPF-ERCC1-SLX4, Artemis, XPG, FEN1, and TATDN2. Several of these nucleases cleave branched DNA structures at stressed replication forks to promote repair and restart of these forks. We recently defined roles for EEPD1 in restarting stressed replication forks after oxidative DNA damage, and for TATDN2 in mitigating replication stress caused by R-loop accumulation in BRCA1-defective cells. We also discuss how insights into biological responses to genome-wide replication stress can inform novel cancer treatment strategies that exploit synthetic lethal relationships among replication stress response factors.

Abstract

A more extensive attack surface for cyber incursions has resulted from the fast expansion of Internet of Things (IoT) devices, calling for more stringent security protocols. This research introduces a new method for protecting Internet of Things (IoT) networks against intrusion assaults by combining Game Theory with Ant Colony Optimization (ACO). Various cyber dangers are becoming more common as a result of the networked nature and frequently inadequate security measures of IoT devices. Because these threats are ever-changing and intricate, traditional security measures can't keep up. An effective optimization method for allocating resources and pathfinding is provided by ACO, which takes its cues from the foraging behavior of ants, while Game Theory provides a strategic framework for modeling the interactions between attackers and defenders. Attackers and defenders in the proposed system are modeled as players in a game where the objective is to maximize their payout. Minimizing damage by anticipating and minimizing assaults is the defender's task. The monitoring pathways are optimized and resources are allocated effectively with the help of ACO. In response to changes in network conditions, the system dynamically modifies defensive tactics by updating the game model in real time. The results of the simulation show that the suggested method successfully increases the security of the Internet of Things. Compared to 87.4% using conventional approaches, the detection accuracy increased to 95.8%. From 10.5 seconds down to 7.3 seconds, the average reaction time to identified incursions was cut in half. Furthermore, there was a 20% improvement in resource utilization efficiency, guaranteeing that defensive and monitoring resources were allocated optimally. Internet of Things (IoT) network security is greatly improved by combining Game Theory with Ant Colony Optimization. In addition to enhancing detection accuracy and reaction times, this combination method guarantees resource efficiency. Abstract

It takes time, money, and effort to keep a watch on your home. Numerous catastrophes, such as burglaries and vandalism, had a place in homes when the owners were negligent or absent. While employing staff is not thought to be a cost-effective alternative, some residential areas employ guards to watch over their homes. A mobile application-based Internet of Things (IoT) system called Home Intruder Detection System (HIDS) assists homeowners in keeping an eye on their houses by remotely alerting users to any potential dangers. The main goals of HIDS are to develop a trustworthy home security system using IoT, to apply the object detection algorithm to identify human presence, and to develop an intelligent mobile application that will allow users to monitor their homes from anywhere in the world and receive alerts if any threats are found. To identify intruders using a camera attached to the system, HIDS is used tested and compared the Region-based Convolution Neural Network (R-CNN), Single-Shot Multibox Detection (SSD) and You Only Look Once (YOLO) detection object detection method in the NVIDIA Jetson Nano. The system is capable of delivering detection video to the server and capturing video at frame rate (FPS) of 44.25 for YOLO, 37.81 for SSD and 27.21 for faster R-CNN respectively. Faster R-CNN, SSD, and YOLO algorithms' average precisions will be 94.08%, 89.75%, and 81.92% respectively.
