Background Ameloblastoma (AMB) is a benign odontogenic tumour, with an aggressive local behaviour and a high rate of recurrence. Previous studies have demonstrated that hypoxia-induced factor alpha 1 (HIF-1α) and activated caspase-3 contribute to tumour invasiveness and cytogenesis in ameloblastoma. Hypoxia increases HIF-1α levels, which triggers a number of signalling pathways. This paper aimed to present data in the study of hypoxia-activated signalling pathways that modulate proapoptotic and antiapoptotic events in AMB. Methods Twenty cases of AMB and ten cases of dental follicle (DF) were used to analyse the immunoexpression of HIF-1α, p53, BNIP3, Bcl-2, IAP-2, GLUT1, and Bax. To contribute to the study, an analysis of expression and genetic interaction was performed using the cell line AME-1. Results AMB and DF expressed the studied proteins. These proteins showed significantly greater immunoexpression in AMB compared with the DF (p < 0.05). HIF-1α showed an important association with GLUT1, and a positive correlation was observed among p53, Bcl-2, and IAP-2. Transcriptomic analysis showed the significant expression of the studied proteins, and the network generated showed a direct association of HIF-1αF with GLUT1 (SLC2A1), TP53, and LDHA. Interestingly, GLUT1 also exhibited direct interaction with TP53 and LDHA. Conclusion In AMB tumorigenesis, hypoxia is possibly related to antiapoptotic events, which suggests an important role for HIF-1α, GLUT1, Bcl-2, IAP-2, and possibly p53.

Corynebacterium diphtheriae (Cd) is a gram-positive human pathogen responsible for diphtheria infection and once regarded for high mortalities worldwide. The fatality gradually decreased with improved living standards and further alleviated when many immunization programs were introduced. However, numerous drug-resistant strains emerged recently that consequently decreased the efficiency of current therapeutics and vaccines, thereby obliging the scientific community to start investigating new therapeutic targets in pathogenic microorganisms. In 2014, our research group introduced the word panmodelome for the first time in the scientific world (Hassan et al., 2014). Inspired by panmodelomics approach, in this study, our group aimed to contribute including the prediction of modelome of thirteen C. diphtheriae strains, using the MHOLline workflow. Considering the quality of the models and using in-house scripts, a set of 465 conserved proteins were selected by combining the results of pangenomics based on core genome and core-modelome analyses. Further, using subtractive proteomics and modelomics approaches for target identification, a set of 23 proteins were selected as essential proteins for bacteria. Considering human as a host, 8 of these proteins (glpX, nusB, rpsH, hisE, smpB, bioB, DIP1084 and DIP0983) were seen as essential and non-host homologs. These proteins were subjected to virtual screening using three different compound libraries (extracted from ZINC database, plant-derived natural compounds, and Di-terpenoid Iso-steviol derivatives). The proposed drug molecules showing favorable interactions, lowered energy values and high complementarity with the predicted targets have also been reported in the present study. Our proposed approach expedites the rapid and efficient selection of C. diphtheriae putative proteins for developing a broad spectrum of novel drugs and vaccines because some of these targets have already been identified and validated in other organisms. Furthermore, we adopted a different approach using same number of genomes of C. diphtheriae to identify drug/vaccine targets based on the druggable pocketome. As a result, we identify 10 targets in which interestingly 3 (hisE-phosphoribosyl-ATP pyrophosphatase, glpX-fructose 1,6-bisphosphatase II, and rpsH 30S ribosomal protein S8) targets were common with our first study. Further, we are working on characterization of hisE-phosphoribosyl-ATP pyrophosphatase, glpX-fructose 1,6-bisphosphatase II, and rpsH 30S ribosomal protein S8 in C. diphtheriae strain NCTC13129. The selection of these proteins (hisE, glpX and rpsH) were made on the bases of their identification through two different computational approaches. Our proposed approaches expedite the selection of C. diphtheriae putative proteins for broad-spectrum development of novel drugs and vaccines, owing to the fact that our study is computational and need experimental validation

Altos fluxos positivos de metano para a atmosfera foram detectados na região amazônica. O gás metano é o segundo mais importante gás de efeito estufa e micro-organismos pertencentes ao Domínio Archaea são responsáveis pela produção de aproximadamente 70% do metano emitido para a atmosfera anualmente. O objetivo deste trabalho foi caracterizar a diversidade de arqueias em sedimentos dos rios Floresta e Madeira através de técnicas moleculares e do cultivo de arqueias metanogênicas. A maior parte das sequências obtidas nas duas bibliotecas pertence ao domínio Crenarchaeota, algumas com similaridade menor que 97% às sequências depositadas nos bancos de dados, revelando a existência de grupos ainda não descritos na literatura. Nos enriquecimentos do sedimento do rio Madeira detectaram-se células pertencentes às famílias Methanosarcinaceae e Methanobacteriaceae pelo emprego de sondas fluorescentes de RNA. Culturas dos gêneros Methanosarcina e Methanobacterium foram estabelecidas em laboratório. A grande diversidade de arqueias não cultivadas encontrada vem reforçar a necessidade de se estudar esse grupo, especialmente sua fisiologia e, consequentemente, seu papel ecológico nos diversos ambientes em que são encontrados. High positive fluxes of methane to the atmosphere have been detected in the Amazonian region. Methane is the second most important greenhouse gas and microorganisms belonging to the Archaea domain are responsible for approximately 70% of the total methane emitted to the atmosphere annually. The objective of this work was to characterize the Archaea diversity in two sites at Madeira and Floresta rivers sediments using molecular techniques and the culturing of methanogenic archaea. Most sequences obtained in the libraries from both rivers belonged to the Crenarchaeota domain, and around half of them presented less than 97% of similarity to sequences available in databases, revealing the existence of new archaea groups yet to be described in the literature. Cells belonging to the Methanosarcinaceae and Methanobacteriaceae families were detected in the enrichment cultures from Madeira River through the use of RNA fluorescent probes. Strains of Methanosarcina sp. and Methanobacterium sp. are being maintained under laboratory conditions. The great diversity of uncultured Archaea found emphasizes the need to study this group, mainly its physiology and, consequently, its role in the diverse environments they occupy.

Nostoc é um gênero cianobacteriano com distribuição ubíqua que tem importância em diversos ecossistemas. Contudo, poucos genomas estão atualmente disponíveis para esse gênero. Enquanto Nostoc spp. são as cianobactérias mais comumente relatadas em relações simbióticas com fungos, animais, plantas e outros organismos, associações com outros micro organismos não receberam atenção similar. Como consequência das fortes interações entre cianobactérias e heterótrofos, culturas não axênicas são geralmente obtidas no isolamento dessas bactérias, o que proporciona uma oportunidade interessante para o desenvolvimento tanto de estudos genômicos quanto metagenômicos. Este trabalho teve como objetivo investigar as características genômicas e funcionais da linhagem Nostoc sp. CENA67, isolada de terra preta antropogênica, bem como estudar sua comunidade associada. Para esse fim, células de uma cultura não axênica de Nostoc sp. CENA67 foram sequenciadas com as plataformas MiSeq e Ion PGM e analisados com ferramentas genômicas e metagenômicas. A linhagem CENA67 de fato pertence à família Nostocaceae e possui algumas características em comum com cianobactérias do gênero Nostoc, porém diverge em certos aspectos morfológicos e filogenéticos do grupo típico de Nostoc, sugerindo que seja representante de um novo táxon. Além disso, seu genoma apresenta diferenças em relação aos genomas atualmente disponíveis para cianobactérias relacionadas ao gênero. A mineração desse genoma revelou 31 agrupamentos gênicos hipoteticamente relacionados à síntese de metabólitos secundários, a maioria dos quais não mostrou similaridade significativa com agrupamentos conhecidos. A análise de um agrupamento gênico de microviridina desvendou uma maior diversidade de genes para precursores dessa molécula do que se acreditava anteriormente, sugerindo que um número considerável de variantes ainda está a ser descoberta. A análise taxonômica da comunidade associada confirmou a dominância de cianobactérias na cultura, mas também revelou a presença de grande número de gêneros microbianos que normalmente são capazes de fixar nitrogênio atmosférico e estabelecer simbiose com plantas, incluindo Mesorhizobium, Sinorhizobium e Starkeya, entre outros. Rascunhos genômicos foram obtidos para Bradyrhizobium diazoefficiens, Bradyrhizobium japonicum, Burkholderia lata e Hyphomicrobium nitrativorans. Todavia, genes para fixação de nitrogênio não foram detectados nesses genomas, apesar de serem encontrados no genoma da cianobactéria e no metagenoma da comunidade, o que sugere que algumas populações podem estar sob pressão de seleção para a perda da capacidade de fixação de nitrogênio, provavelmente devido a este nutriente estar sendo fornecido pelo organismo mais abundante nesta comunidade, a cianobactéria. A análise funcional indicou vias exclusivas tanto à cianobactéria quanto à comunidade associada, e sugeriu a complementariedade de certos metabolismos. Os resultados possibilitam o aumento do conhecimento sobre a diversidade molecular e química do filo Cyanobacteria e levantam possíveis interações com micro organismos simbiontes Nostoc is a cyanobacterial genus with ubiquitous distribution that is important in several ecosystems. However, few genomes are currently available for this genus. While Nostoc spp. are the most commonly reported cyanobacteria in symbiotic relationship with fungi, animals, plants, and other organisms, associations with other microorganisms have not received similar attention. As a consequence of tight interactions between cyanobacteria and heterotrophs, non-axenic cultures are usually achieved in the isolation of these bacteria, which provides an interesting opportunity for carrying out both genomic as metagenomic studies. This work aimed to investigate the genomic and functional characteristics of the strain Nostoc sp. CENA67, isolated from anthropogenic dark earth, and to study its associated community. For this purpose, cells from a non-axenic culture of Nostoc sp. CENA67 were sequenced with the platforms MiSeq and Ion PGM and analyzed with genomic and metagenomic tools. The strain CENA67 indeed belongs to the family Nostocaceae and presents some characteristics in common with cyanobacteria of the genus Nostoc, but diverges in certain morphological and phylogenetic aspects of the typical Nostoc group, suggesting that it is a representative of a new taxon. In addition, its genome presents differences in relation to the genomes currently available for cyanobacteria related to this genus. Genome mining revealed 31 gene clusters hypothetically related to the synthesis of secondary metabolites, most of which did not show significant similarity to known clusters. The analysis of a microviridin gene cluster unveiled a larger diversity of precursor genes for this molecule than was previously believed, suggesting that a considerable number of variants is still to be found. The taxonomic analysis of the associated community confirmed the dominance of cyanobacteria in the culture, but also revealed the presence of a great number of microbial genera that are usually capable of fixing atmospheric nitrogen and establishing symbiosis with plants, including Mesorhizobium, Sinorhizobium, and Starkeya, among others. Genomic drafts were obtained for Bradyrhizobium diazoefficiens, Bradyrhizobium japonicum, Burkholderia lata, and Hyphomicrobium nitrativorans. Nevertheless, genes for nitrogen fixation were not detected in these genomes, despite being found in the cyanobacterial genome and the community metagenome, suggesting that some populations might be under selection pressure for the loss of the ability to fix nitrogen, probably due to this nutrient being provided for the most abundant organism in this culture, the cyanobacterium. Functional analysis indicated pathways exclusive both to the cyanobacterium as to the associated community, and suggested the complementarity of certain metabolisms. The results allow the increase of the knowledge about the molecular and chemical diversity of the phylum Cyanobacteria and raise possible interactions with symbiotic microorganisms

Corynebacterium pseudotuberculosis é uma bactéria Gram-positiva, causadora da linfadenite caseosa em ovinos e caprinos. Para causar uma infecção bem sucedida, este patógeno intracelular facultativo precisa responder de modo eficiente às alterações do ambiente extracelular, sendo capaz de sobreviver às condições adversas encontradas no organismo hospedeiro. Um dos principais mecanismos utilizados por bactérias patogênicas para alcançar este objetivo consiste na ativação transitória de genes específicos de resposta aos estresses ambientais extracelulares por fatores sigma alternativos da RNA polimerase. Com o seqüenciamento do genoma de C. pseudotuberculosis pela Rede Genoma de Minas Gerais e Rede Paraense de Genômica e Proteômica, foi possível identificar sete fatores sigma alternativos nesta bactéria: os fatores sigma B, C, D, E, H, K e M. Dentre estes, destaca-se o fator sigma C, o qual tem sido associado à virulência em Mycobacterium tuberculosis, e cujo gene se encontra em uma ilha de patogenicidade em C. pseudotuberculosis. Neste contexto, nosso grupo de pesquisa tem se dedicado ao estudo do papel dos fatores sigma alternativos de C. pseudotuberculosis na resposta a diferentes condições de estresse in vitro e na virulência da bactéria. Neste trabalho, foi construída uma linhagem de C. pseudotuberculosis deficiente para o fator sigma C, utilizando um plasmídeo suicida, e foi avaliada a resistência desta linhagem aos estresses oxidativo, ácido, osmótico e térmico in vitro, através da comparação das diferenças no crescimento relativo e na viabilidade relativa das bactérias expostas às condições de estresse em relação às bactérias crescidas sob condições normais, entre os tempos sucessivos de exposição das linhagens 1002 selvagem e mutante para o fator sigma C a cada agente gerador de estresse. Não foi observada uma diferença significativa na susceptibilidade da linhagem mutante para o fator sigma C ao estresse ácido em relação à linhagem 1002 selvagem. Contudo, foi verificado que esta linhagem é significativamente mais susceptível aos estresses oxidativo, osmótico e térmico do que a linhagem 1002 selvagem, indicando que o fator sigma C deve desempenhar um papel importante para a resposta da bactéria a estas condições de estresse, e possivelmente, para a sua sobrevivência no meio ambiente e no interior do hospedeiro e para a sua virulência. Corynebacterium pseudotuberculosis is a Gram-positive bacterium, which causes caseous lymphadenitis in sheep and goats. To cause a successful infection, this facultative intracellular pathogen needs to respond efficiently to the changes in the extracellular environment, being able to survive faced with the harsh conditions found in the host organism. One of the main mechanisms used by pathogenic bacteria to achieve this goal is the transient activation of specific genes in response to extracellular environmental stresses by alternative sigma factors of RNA polimerase. After the sequencing of the genome of C. pseudotuberculosis by Rede Genoma de Minas Gerais e Rede Paraense de Gen?ica e Prote?ica, it was identified seven alternative sigma factors in this bacterium, the sigma factors B, C, D, E, H, K and M. Between these, the sigma factor C stands out, which has been linked to virulence in Mycobacterium tuberculosis, and whose gene is located in a pathogenicity island in C. pseudotuberculosis. In this context, our research group has been studying the role of the alternative sigma factors of C. pseudotuberculosis in response to different stress conditions in vitro and in virulence of the bacterium. In this work, we constructed a strain of C. pseudotuberculosis deficient for the sigma factor C, using a suicide plasmid, and evaluated the resistance of this strain to oxidative, acid, osmotic and heat stresses in vitro, by comparing the differences in relative growth and relative viability of bacteria exposed to stress conditions in relation to bacteria grown under normal conditions, between the successive times of exposure of 1002 wild strain and the strain mutant for the sigma factor C for each stress generator agent. There was no significant difference in the susceptibility of the strain mutant for the sigma factor C for the acid stress in relation to the 1002 wild strain. However, it was found that this strain is significantly more susceptible to oxidative, osmotic and heat stresses than the 1002 wild strain, showing that the sigma factor C should play an important role for the bacteriums response to this stress conditions, and possibly, for its survival in the environment and in the host and for its virulence.

O presente estudo teve como objetivo avaliar a comunidade envolvida na desnitrificação autotrófica pela oxidação de sulfetos, aplicada ao pós-tratamento de efluentes anaeróbios. O enriquecimento da comunidade bacteriana e da comunidade desnitrificante autotrófica foi realizado a partir de amostras da biomassa coletadas de três reatores verticais de leito fixo operados em condições distintas, sendo, redução autotrófica de nitrato, redução autotrófica de nitrito e redução autotrófica de nitrato com excesso de sulfeto. Após a determinação da melhor condição de enriquecimento, a cultura foi purificada, identificada por meio de ferramentas da biologia molecular e caracterizada quanto às melhores condições de crescimento. O enriquecimento foi bem sucedido com a biomassa dos três reatores. No entanto, a condição de redução de nitrato com relação \'N\'/\'S\' igual a 0,8 foi a que apresentou maior concentração de microrganismos desnitrificantes autotróficos. A bactéria isolada foi identificada como Pseudomonas stutzeri. A velocidade específica máxima de crescimento da cultura (\'mü\'máx) foi de 0,037/h, com tempo de duplicação de 18,7 horas. O rendimento celular (Y) do composto nitrogenado foi de 0,15 gSSV.g/\'N\' e a velocidade de desnitrificação foi de aproximadamente 0,24 g\'N\'/gSSV.h. Os dados obtidos indicaram a viabilidade da aplicação da espécie isolada no processo de desnitrificação autotrófica utilizando sulfeto como doador de elétrons. The aim of this study was to evaluate the community involved on autotrophic denitrification by sulfide oxidation applied to the post-treatment of anaerobic effluents. The enrichment of the bacterial community and autotrophic denitrifier community was accessed in three immobilized bed reactors operated at the conditions of autotrophic reduction of nitrate, autotrophic reduction of nitrite and autotrophic reduction of nitrate under excess of sulfide. Following the determination of the best enrichment conditions, the culture was purified, identified by molecular biology tools and the best growth conditions were characterized. Enriched cultures were obtained for the three operational conditions, but the best condition for the growth of autotrophic denitrifiers microorganisms was at \'N\'/\'S\' ratio of 0,8. The isolated microorganism was identified as Pseudomonas stutzeri. The maximum specific growth rate (\'mü\'máx) was 0.037/h, with a doubling time of 18.7 hours. The growth yield (Y) of nitrogen compound was 0.15 gSSV/g\'N\' and the specific rate of nitrogen utilization was approximately 0.24 g\'N\'/gSSV.h. The results indicated the viability of application of this microorganism for autotrophic denitrification using sulfide as electron donor.

A degradação da metilamina foi investigada por meio da avaliação da velocidade específica máxima de produção de metano (VEM CH4) e da comunidade microbiana relacionada aos Domínios Bacteria e Archaea. Para isso, foram realizados dois ensaios com reatores anaeróbios em batelada inoculados com lodo granulado oriundo de reator UASB usado no tratamento de água residuária de abatedouro de aves. Em todos os ensaios, os reatores controle, que não receberam adição de metilamina, apresentaram VEM CH4 de 0,04 mmol/L g STV dia. O primeiro ensaio avaliou a degradação da metilamina em diferentes concentrações de inóculo (2,5, 5,0 e 10,0 g STV/L) e substrato (1.550 e 3.100 mg metilamina/L). A concentração de \'CH IND.4\' esperada estequiometricamente foi atingida em todos os reatores (37,50 e 75,00 mmol \'CH IND.4\'/L, para concentrações de 1.550 e 3.100 mg metilamina/L, respectivamente), exceto para aquele com 2,5 g STV/L e 3.100 mg metilamina/L, que produziu somente 2,04 mmol \'CH IND.4\'/L. A maior velocidade específica máxima de produção de \'CH IND.4\' foi 4,42 mmol/L g STV dia, obtida nos reatores com 2,5 g STV/L e 1.550 mg metilamina/L. Os reatores inoculados com 5,0 g STV/L tiveram VEM CH4 de 2,31 e 2,34 para 1.550 e 3.100 mg metilamina/L, respectivamente. Os reatores com 1.550 mg metilamina/L e 10,0 g STV/L apresentaram VEM CH4 de 1,28 mmol/L g STV dia. A concentração de \'N\'-\'NH IND.4\'POT.+\' excedeu em 12,9%, 0,7% e 18,3% o valor esperado (698 mg/L) para os reatores com 1.550 mg metilamina/L e 2,5, 5,0 e 10,0 g STV/L, respectivamente. Nos reatores com 3.100 mg metilamina/L, as concentrações finais de \'N\'-\'NH IND.4\'POT.+\' foram 122 e 1.726 mg/L para concentrações de inóculo de 2,5 e 5,0 g STV/L, respectivamente. O segundo ensaio comparou diferentes relações metilamina/sulfato (0,71, 1,26 e 2,18) em reatores inoculados com 5,0 g STV/L contendo 1.550 mg metilamina/L. As concentrações de \'CH IND.4\' esperadas estequiometricamente foram atingidas em todos os reatores. As velocidades específicas máximas de formação de \'CH IND.4\' foram de 2,54, 2,31 e 3,14 mmol/L g STV dia para as relações metilamina/sulfato 0,71, 1,26 e 2,18, respectivamente. Em todos os reatores, a concentração de \'N\'-\'NH IND.4\'POT.+\' atingiu média final de 1200 mg/L. Os reatores controle consumiram 71,9% do sulfato adicionado. Os reatores com relação metilamina/sulfato 0,71, 1,26 e 2,18 consumiram 49,6%, 61,6% e 83,2% de todo o sulfato adicionado, respectivamente. Nos dois ensaios, os exames microscópicos revelaram a presença de cocos, bacilos, filamentos, cocos e sarcinas fluorescentes. Nos reatores alimentados apenas com metilamina, o seqüenciamento de fragmentos da região 16S do RNAr detectou cinco Filos do Domínio Bacteria (Acidobacteria 4%, Firmicutes 11%, Proteobacteria 14%, Spirochaetes 13% e Synergistes 47%). Nos reatores com metilamina e sulfato, sete Filos foram detectados (Firmicutes 45%, Proteobacteria 7%, Spirochaetes 2%, Synergistes 16%, Chloroflexi 4%, Thermotogae 8% e Planctomycetes 1%). Nos dois ensaios, o Domínio Archaea foi predominantemente representado pelas Famílias Methanomicrobiaceae, Methanosaetaceae e Methanosarcinaceae, com presença de Methanomethylovorans hollandica, uma espécie de arquéia metanogênica com metabolismo especializado na degradação de metilamina. The degradation of methylamine was investigated assessing the maximum specific methane production rate (MSR CH4) and the microbial community related to Bacteria e Archaea Domains. For this, two tests were performed in anaerobic batch reactors inoculated with granular sludge from an UASB reactor used in the treatment of poultry wastes. In all experiments, the control reactors, without methylamine addition, showed MSR CH4 of 0.04 mmol/L g TVS day. The first experiment evaluated the degradation of methylamine at different inoculum concentrations (2.5, 5.0 and 10.0 g TVS/L) and substrate concentrations (1,550 and 3,100 mg methylamine/L). The stoichiometrically expected \'CH IND.4\' concentration was reached in all reactors (37.50 and 75.00 mmol \'CH IND.4\'/L, for methylamine concentrations of 1,550 and 3,100 mg methylamine/L, respectively), except for the reactor with 2.5 g TVS/L and 3,100 mg methylamine/L, that produced only 2.04 mmol \'CH IND.4\'/L. The highest maximum specific methane production rate was 4.42 mmol/L g TVS day, reached in the reactors with 2.5 g TVS/L and 1,550 mg methylamine/L. The reactors inoculated with 5.0 g TVS/L had MSR CH4 of 2.31 and 2.34 for 1,550 and 3,100 mg methylamine/L, respectively. The reactors with 1,550 mg methylamine/L and 10.0 g TVS/L had MSR CH4 of 1.28 mmol/L g TVS day. The \'N\'-\'NH IND.4\'POT.-\' concentrations exceeded 12.9%, 0.7% and 18.3% the expected value (698 mg/L) for the reactors with 1,550 mg methylamine/L and 2.5, 5.0 and 10.0 g TVS/L, respectively. In the reactors with 3,100 mg methylamine/L, the final concentrations of \'NH IND.4\'POT.+\'-\'N\' were 122 and 1,726 mg/L for inoculum concentrations of 2.5 and 5.0 g TVS/L, respectively. The second experiment compared different methylamine/sulfate ratios (0.71, 1.26 and 2.18) on reactors inoculated with 5.0 g TVS/L containing 1,550 mg methylamine/L. The stoichiometrically expected \'CH IND.4\' concentration was reached in all reactors. The maximum specific methane production rates were 2.54, 2.31 and 3.14 mmol/L g TVS day for methylamine/sulfate ratios of 0.71, 1.26 and 2.18, respectively. In all reactors, the average \'NH IND.4\'POT.+\'-\'N\' final concentration was 1,200 mg/L. The control reactors consumed 71.9% of the added substrate. The reactors with methylamine/sulfate ratios of 0.71, 1.26 and 2.18 consumed 49.6%, 61.6% and 83.2% of all the added sulfate, respectively. In both experiments, the microscopic analysis revealed cocci, rods, filaments, fluorescent cocci and sarcinas. In the reactors fed with methylamine only, the sequencing of 16S rRNA fragments detected five Phyla of the Bacteria Domain (Acidobacteria 4%, Firmicutes 11%, Proteobacteria 14%, Spirochaetes 13% and Synergistes 47%). In the reactors with methylamine and sulfate, seven Phyla were detected (Firmicutes 45%, Proteobacteria 7%, Spirochaetes 2%, Synergistes 16%, Chloroflexi 4%, Thermotogae 8% e Planctomycetes 1%). In both experiments, Archaea Domain was mainly represented by Methanomicrobiaceae, Methanosaetaceae and Methanosarcinaceae Families, with the presence of Methanomethylovorans hollandica, a methanogenic archaea specie with specific metabolism for methylamine degradation.

CNPq - Conselho Nacional de Desenvolvimento Científico e Tecnológico Nos últimos 50 anos, o Brasil desenvolveu habilidades acadêmicas competitivas e avançou significativamente na formação de recursos humanos qualificados. O país está atualmente na 13ª posição na lista de países que mais publicam artigos científicos no mundo: 2,12% da produção global. Em fevereiro de 2007, o presidente da república decretou a Política de Desenvolvimento da Biotecnologia que prevê, entre outras ações, a promoção da cultura da inovação e o uso estratégico da propriedade intelectual a fim de assegurar maior competitividade à biotecnologia nacional. Neste sentido, faz-se necessário diagnosticar o estado atual e a evolução da proteção intelectual em biotecnologia no Brasil de forma a subsidiar ações de desenvolvimento para este setor. Este estudo consistiu no levantamento e análise dos depósitos de patentes no INPI (Instituto Nacional de Propriedade Industrial) entre os anos de 1996 e 2007 através de duas bases de dados online (INPI e Espacenet) e dos depósitos por residentes via PCT (Tratado de Cooperação em Matéria de Patentes) através do sistema PatentScope. Para identificação das patentes biotecnológicas foram utilizados os códigos da Classificação Internacional de Patentes (CIP) estabelecidos pela Organização para a Cooperação e Desenvolvimento Econômico (OCDE) como representativos da biotecnologia. A subclasse C12N foi adotada como mais representativa da biotecnologia moderna, englobando, principalmente, a engenharia genética e manipulação de microorganismos. Os parâmetros analisados foram: número de depósitos por ano, número de depósitos por país de origem e perfil dos principais depositantes. Foram depositadas cerca de 20.500 patentes no período analisado, mantendo-se uma média de cerca de 600 patentes por ano. Os brasileiros detêm apenas 3,1% das patentes nacionais em biotecnologia, indicando forte controle do conhecimento tecnológico por estrangeiros. Entre os países que mais depositam patentes biotecnológicas no Brasil, o predomínio é dos Estados Unidos, com 43,3% dos depósitos, seguido pela Alemanha (10,0%), Japão (5,6%), França (5,1%) e Suíça (5,1%). Em relação aos depositantes nacionais, as universidades e institutos de pesquisa foram responsáveis pelo maior número de depósitos, 47,9% das patentes, enquanto 16,5% correspondem a pessoas físicas, um patamar alto para um setor tecnológico. Apenas 15,3% dos depósitos estão em nome de empresas privadas. Este padrão de distribuição difere enormemente do de países como Estados Unidos e Alemanha, no qual as empresas privadas são as principais detentoras de patentes biotecnológicas. São várias as implicações deste trabalho para o desenvolvimento do setor brasileiro de biotecnologia, no entanto, dois pontos devem ser enfatizados: (1) o baixo registro de patentes biotecnológicas domésticas frente à forte produção científica do país neste setor; (2) o confinamento do conhecimento biotecnológico nas universidades e centros de pesquisa, o que indica a necessidade de desenvolvimento de mecanismos eficientes de transferência tecnológica e cooperação público/privada. Brazil has developed, over the last 50 years, competitive academic capabilities and has advanced consistently in the training of human resources. The country currently ranks 13th in the world classification of scientific publishing representing 2,12% of global production. In 2007, President da Silva signed a decree outlining the National Policy for Biotechnology Development which promotes, among other actions, intellectual property protection among biotech entrepreneurs and scientists, fostering innovation and competitiveness. This study investigates, thereby, the levels and patterns of biotechnology patenting in Brazil by examining patent applications in the Brazilian Patent Office (Instituto Nacional de Propriedade Industrial, INPI) as well as PCT applications from 1996 to 2007. Biotechnology-related patents were determined according to a list of biotechnology International Patent Classification (IPC) codes provided by the OECD. Subclass C12N was considered the most representative of modern biotechnology, encompassing mainly genetic engineering and manipulation of microorganisms. We analyzed total number of patents and how they evolved over time in terms of ownership structure (domestic or foreign-own) and country of origin. To determine which domestic sector owns the most patents we classified national assignees into six categories, as follows: universities and research institutes; state-owned firms; funding agencies; individuals; government and private firms. 20.500 patent applications were identified from 1996 to 2007, an average of 600 applications per year. Almost 97% of applicants were foreigners. The top-patenting countries at the Brazilian Patent Office were United States (43,7%), Germany (10,0%), Japan (5,6%), France (5,1%) and Switzerland (5,1%). Biotechnology patents in Brazil are predominantly owned by universities and research institutions (47,9%). Individuals come in second, with 16,5% of patent applications, a high number for a science-based sector. Only 15,3% of patents were assigned to private firms. This distribution differs considerably from other countries such as United States and Germany, in which private firms are the main patenters. This study demonstrated (1) the low domestic patenting activity compared to the national scientific production in this area; (2) the confinement of biotechnological knowledge inside universities and research centers, indicating that a well managed public-private cooperation will be key to the commercialization of biotech innovations in the country.
