This paper aims to use non-linear physical analysis to calculate the displacement of beams and slabs in a cracked state. This study uses the commercial software SAP2000 to perform a numerical analysis using the finite element formulation, applying the multi-layer method. Initially, a parametric study was carried out to evaluate the vertical displacement for service loads of reinforced concrete beams and slabs using different spans, support conditions and geometry. In order to validate the finite element model, the study compared displacement values for linear analysis using Bares tables. Subsequently, simplified methods of displacement calculation in the long term are applied, using an abacus from Comité Euro-International du Béton (CEB). These values are then compared with the physically non-linear analysis in the long-term cracked state with SAP2000. Two structural codes were used in the numerical and analytical methods, Regulamento de Estruturas de Betão Armado e Pré-Esforçado (REBAP) and Eurocode 2 (EC2), to evaluate their differences in deformation control. Therefore, the main goal is to establish comparisons between the two methods of analysis in order to show that non-linear finite element modelling provides values that are lower than the analytical calculation, thus allowing greater economy in the design of structural reinforced concrete elements. In addition, it can be noted that EC2 has some inconsistencies in the design of simply supported slabs, requiring a greater thickness than a flat square slab and that indirect deformation for REBAP is underestimated for beams and, in some cases, for slabs.

Background: Reports have shown correlations between the immune response to vector saliva and Leishmaniasis outcome. We followed dogs in an endemic area for two years characterizing resistance or susceptibility to canine visceral leishmaniasis (CVL) according to Leishmania infantum diagnosis and clinical development criteria. Then, we aimed to identify a biosignature based on parasite load, serum biological mediators' interactions, and vector exposure intensity associated with CVL resistance and susceptibility., Methodology/principal Findings: A prospective two-year study was conducted in an area endemic for CVL. Dogs were evaluated at 6-month intervals to determine infection, clinical manifestations, immune profile, and sandfly exposure. CVL resistance or susceptibility was determined upon the conclusion of the study. After two years, 78% of the dogs were infected with L. infantum (53% susceptible and 47% resistant to CVL). Susceptible dogs presented higher splenic parasite load as well as persistence of the parasite during the follow-up, compared to resistant ones. Susceptible dogs also displayed a higher number of correlations among the investigated biological mediators, before and after infection diagnosis. At baseline, anti-saliva antibodies, indicative of exposure to the vector, were detected in 62% of the dogs, reaching 100% in one year. Higher sandfly exposure increased the risk of susceptibility to CVL by 1.6 times (CI: 1.11-2.41). We identified a discriminatory biosignature between the resistant and susceptible dogs assessing splenic parasite load, interaction of biological mediators, PGE2 serum levels and intensity of exposure to sandfly. All these parameters were elevated in susceptible dogs compared to resistant animals., Conclusions/significance: The biosignature identified in our study reinforces the idea that CVL is a complex multifactorial disease that is affected by a set of factors which are correlated and, for a better understanding of CVL, should not be evaluated in an isolated way., Competing Interests: The authors have declared that no competing interests exist.

Central nervous system (CNS) is the main site for encystment of Neospora caninum in different animal species. In this tissue, glial cells (astrocytes and microglia) modulate responses to aggression in order to preserve homeostasis and neuronal function. Previous data showed that when primary cultures of glial cells are infected with N. caninum, they develop gliosis and the immune response is characterized by the release of TNF and IL-10, followed by the control of parasite proliferation. In order to elucidate this control, three enzymatic systems involved in parasite-versus-host interactions were observed on a model of neuron/glia co/cultures obtained from rat brains. Indoleamine 2,3-dioxygenase (IDO), induced nitric oxide synthase (iNOS) responsible for the catabolism of tryptophan and arginine, respectively, and cycloxigenase (COX) were studied comparing their modulation by respective inhibitors with the number of tachyzoites or the immune response measured by the release of IL-10 and TNF. Cells were treated with the inhibitors of iNOS (1.5 mM L-NAME), IDO (1 mM 1-methyl tryptophan), COX-1 (1 μM indomethacin) and COX-2 (1 μM nimesulide) before infection with tachyzoites of N. caninum (1:1 cell: parasite). After 72 h of infection, immunocytochemistry showed astrogliosis and a significant increase in the number and length of neurites, compared with uninfected co-cultures, while an increase of IL-10 and TNF was verified. N. caninum did not change iNOS activity, but the inhibition of the basal levels of this enzyme stimulated parasite proliferation. Additionally, a significant increase of about 40% was verified in the IDO activity, whose inhibition caused 1.2-fold increase in parasitic growth. For COX-2 activity, infection of cultures stimulated a significant increase in release of PGE 2 and its inhibition by nimesulide allowed the parasitic growth. These data indicate that iNOS, IDO and COX-2 control the proliferation of N. caninum in this in vitro model. Sugarcane growing area in Brazil sharply expanded between 2000 and 2010 due to the increasing world demand for sugar and ethanol. Since this expansion of sugarcane is said to occur in areas covered by degraded pastures, it is likely not threatening the environment or food production. In order to verify this assumption, we investigate at farm and field levels which types of land use sugarcane cropping replaced between 2005 and 2010 and the reasons for farmers shifting or not shifting to sugarcane, as a case study in two counties in the state of Goiás. Within the studied period, sugarcane cropping expansion was related to large farms, lower risk perceived by farmers, and higher profitability compared with soybean and beef cattle-raising. For smallholders, particularly dairy farmers, the need to comply with the set-aside rules under Brazilian Forest Code (Código Florestal Brasileiro) made a shift to sugarcane less attractive, as it would have forced them to reduce farm cultivable area, with loss of incomes. From 30,408 ha under sugarcane surveyed, 45.7% had used to be pastures, 31% had previously been pastures rotated with soybean and maize, and 23.3% had been cropped exclusively with soybean or maize.

Hypoxia and necrosis are fundamental features of glioma, and their emergence is critical for the rapid biological progression of this fatal tumor. The presence of vaso-occlusive thrombus is higher in grade IV tumors [glioblastoma multiforme (GBM)] compared with lower grade tumors, suggesting that the procoagulant properties of the tumor contribute to its aggressive behavior, as well as the establishment of tumor hypoxia and necrosis. Tissue factor (TF), the primary cellular initiator of coagulation, is overexpressed in GBMs and likely favors a thrombotic microenvironment. Phosphatase and tensin homolog (PTEN) loss and hypoxia are two common alterations observed in glioma that may be responsible for TF upregulation. In the present study, ST1 and P7 rat glioma lines, with different levels of aggressiveness, were comparatively analyzed with the aim of identifying differences in procoagulant mechanisms. The results indicated that P7 cells display potent procoagulant activity compared with ST1 cells. Flow cytometric analysis showed less pronounced levels of TF in ST1 cells compared with P7 cells. Notably, P7 cells supported factor X (FX) activation via factor VIIa, whereas no significant FXa generation was observed in ST1 cells. Furthermore, the exposure of phosphatidylserine on the surface of P7 and ST1 cells was investigated. The results supported the assembly of prothrombinase complexes, accounting for the production of thrombin. Furthermore, reverse transcription-quantitative polymerase chain reaction showed that CoCl 2 (known to induce a hypoxic-like stress) led to an upregulation of TF levels in P7 and ST1 cells. Therefore, increased TF expression in P7 cells was accompanied by increased TF procoagulant activity. In addition, hypoxia increased the shedding of procoagulant TF-bearing microvesicles in both cell lines. Finally, hypoxic stress induced by treatment with CoCl 2 upregulated the expression of the PAR1 receptor in both P7 and ST1 cells. In addition to PAR1, P7, but not ST1 cells, expressed higher levels of PAR2 under hypoxic stress. Thus, modulating these molecular interactions may provide additional insights for the development of more efficient therapeutic strategies against aggressive glioma.
