33. Optimizing Primary Recovery and Refolding of Human Interferon-b from Escherichia coli Inclusion Bodies

Abstract

Background: The refolding of proteins from inclusion bodies is affected by several factors, including solubilization of inclusion bodies by denaturants, removal of the denaturant, and assistance of refolding by small molecule additives. Objectives: The purpose of this study was optimization of recombinant human interferon-β purification in order to achieve higher efficiency, yield, and a product with a better and more suitable biological activity. Materials and Methods: Triton X-100 and sodium deoxycholate were used to wash the recombinant human interferon-β inclusion bodies prior to solubilization. The inclusion bodies solubilization process was performed by denaturants and reducing agents; guanidine-hydrochloride, urea, β-mercaptoethanol and dithiothritol. Results: The best recovery was obtained in the presence of 0.5% TritonX-100 (v/v). Low concentrations of urea only gave a marginal improvement on the refolding of recombinant human interferon-β. Successful refolding was achieved by gradient elution (decreasing the guanidine-hydrochloride concentration) in the presence of L-arginine. Partial purification was also achieved continuously, and recombinant human interferon-β was recovered with 93.5% purity. The interferon prepared in this project was biologically active and inhibited the replication of vesicular stomatitis virus in Hela cells, when compared to the standard interferon. Conclusions: In this research, the best recovery of inclusion bodies was found at a concentration 0.5 M of Triton X-100, the maximum efficiency of solubility was found in pH 10.5 and the maximum efficiency of refolding was achieved by final buffer containing 2M urea and 0.6 M L-Arg.

Abstract

Antimicrobial agents, such as nisin, are used extensively in the food industry. Here, we investigated various approaches to load nisin onto mesoporous silica nanoparticles (MSNs, 92 ± 10 nm in diameter), to enhance its stability and sustained release. The morphology, size, and surface charge of the as-prepared nanoparticles were analyzed using scanning transmission electron microscopy, dynamic light scattering, and ζ potential measurement. Nisin was either physically adsorbed or covalently attached to the variously functionalized MSNs, with high loading capacities (>600 mg of nisin g-1 of nanoparticles). The results of antibacterial activity analysis of nisin against Staphylococcus aureus showed that, despite the very low antibacterial activity of nisin covalently conjugated onto MSNs, the physical adsorption of nisin onto the unfunctionalized nanoparticles enhances its antimicrobial activities under various conditions, with no significant cytotoxicity effects on mouse fibroblast L929 cells. In conclusion, MSNs can be recommended as suitable carriers for nisin under various conditions.

Abstract

We compared the viability, proliferation, and differentiation of human Mesenchymal Stromal Cells (MSC) after culture on poly(lactic-co-glycolic acid) (PLGA) and PLGA/chitosan (PLGA/CH) hybrid scaffolds. We applied conventional and emulsion electrospinning techniques, respectively, for the fabrication of the PLGA and PLGA/ CH scaffolds. Electrospinning under optimum conditions resulted in an average fiber diameter of 166 ± 33 nm for the PLGA/CH and 680 ± 175 nm for the PLGA scaffold. The difference between the tensile strength of the PLGA and PLGA/CH nanofibers was not significant, but PLGA/CH showed a significantly lower tensile modulus and elongation at break. However, it should be noted that the extensibility of the PLGA/CH was higher than that of the nanofibrous scaffolds of pure chitosan. As expected, a higher degree of hydrophilicity was seen with PLGA/ CH, as compared to PLGA alone. The biocompatibility of the PLGA and PLGA/CH scaffolds was compared using MTS assay as well as analysis by scanning electron microscopy and confocal microscopy. The results showed that both scaffold types supported the viability and proliferation of human MSC, with significantly higher rates on PLGA/ CH nanofibers. Nonetheless, an analysis of gene expression of MSC grown on either PLGA or PLGA/CH showed a similar differentiation pattern towards bone, nerve and adipose tissues.

Abstract

Development of tissue engineering scaffolds with native-like biology and microarchitectures is a prerequisite for stem cell mediated generation of off-the-shelf-tissues. So far, the field of tissue engineering has not full-filled its grand potential of engineering such combinatorial scaffolds for engineering functional tissues. This is primarily due to the many challenges associated with finding the right microarchitectures and ECM compositions for optimal tissue regeneration. Here, we have developed a new microgel array to address this grand challenge through robotic printing of complex stem cell-laden microgel arrays. The developed microgel array platform consisted of various microgel environments that where composed of native-like cellular microarchitectures resembling vascularized and bone marrow tissue architectures. The feasibility of our array system was demonstrated through localized cell spreading and osteogenic differentiation of human mesenchymal stem cells (hMSCs) into complex tissue-like structures. In summary, we have developed a tissue-like microgel array for evaluating stem cell differentiation within complex and heterogeneous cell microenvironments. We anticipate that the developed platform will be used for high-throughput identification of combinatorial and native-like scaffolds for tissue engineering of functional organs.

Abstract

This study reports the importance of physicochemical characteristics of the core-shell structured polymer-coated Fe 3 O 4 cluster@SiO 2 nanocomposite particles for designing the biocatalysts used for transesterification of soybean oil. To demonstrate this issue, two different types of polymer molecules such as polyethylenimine (PEI) and polyacrylic acid (PAA) each with two different molecular weights are employed. The highest fatty acid methyl esters (FAMEs) synthesis yield values are achieved by the lipases immobilized onto the low- and high-molecular-weight PEI-coated particles as compared to those of the low- and high-molecular-weight PAA-coated particles ( i.e., ∼64.8% and ∼73.1% versus ∼51.5% and ∼46.4%, respectively). The lipases immobilized onto the PEI-coated particles also exhibit excellent stabilities over five repeated uses. The solvent content assays of the particles immersed in water, methanol and soybean oil reveal that the PEI-coated particles have the highest tendency to absorb the oil and the least ones to absorb water and methanol. Thus, those particles increase the substrate molecules accessibility to the immobilized lipases in the reaction medium, leading to the superior transesterification efficiency. This study provides insight into the highly effective enzymatic production of biodiesel with considering the physicochemical properties of support particles used for enzymes immobilization.
