9. STING-Activating Polymer–Drug Conjugates for Cancer Immunotherapy

Abstract

The stimulator of interferon genes (STING) pathway links innate and adaptive antitumor immunity and therefore plays an important role in cancer immune surveillance. This has prompted widespread development of STING agonists for cancer immunotherapy, but pharmacological barriers continue to limit the clinical impact of STING agonists and motivate the development of drug delivery systems to improve their efficacy and/or safety. We developed SAPCon, a STING-activating polymer–drug conjugate platform based on strain-promoted azide–alkyne cycloaddition of a novel dimeric amidobenzimidazole (diABZI) STING prodrug to hydrophilic poly(dimethylacrylamide-co-azido-ethylmethacrylate) polymer chains through a cathepsin B-responsive linker to increase circulation time and enable passive tumor accumulation. We found that intravenously administered SAPCon accumulated at tumor sites, where it was endocytosed by tumor-associated myeloid cells, resulting in increased STING activation in the tumor tissue. Consequently, SAPCon promoted an immunogenic tumor microenvironment characterized by increased frequency of activated macrophages and dendritic cells and improved infiltration of CD8+T cells, resulting in inhibition of tumor growth, prolonged survival, and enhanced response to anti-PD-1 immune checkpoint blockade in orthotopic breast cancer models. Collectively, these studies position SAPCon as a modular and programmable platform for improving the efficacy of systemically administered STING agonists for cancer immunotherapy.

Abstract

In natural products (NPs) research, methods for the efficient prioritization of natural extracts (NEs) are key for discovering novel bioactive NPs. In this study a biodiverse collection of 1,600 NEs, previously analyzed by UHPLC-HRMS² metabolite profiling was screened for Wnt pathway regulation. The results of the biological screening drove the selection of a subset of 30 non-toxic NEs with an inhibitory IC50 ≤ 5 µg/mL. To increase the chance of finding structurally novel bioactive NPs, Inventa, a computational tool for automated scoring of NEs based on structural novelty was used to mine the HRMS² analysis and dereplication results. After this, four out of the 30 bioactive NEs were shortlisted by this approach. The most promising sample was the ethyl acetate extract of the leaves of Hymenocardia punctata (Phyllanthaceae). Further phytochemical investigations of this species resulted in the isolation of three known prenylated flavones (3, 5, 7) and ten novel bicyclo[3.3.1]non-3-ene-2,9-diones (1, 2, 4, 6, 8-13), named Hymenotamayonins. Assessment of the Wnt inhibitory activity of these compounds revealed that two prenylated flavones and three novel bicyclic compounds showed interesting activity without apparent cytotoxicity. This study highlights the potential of combining Inventa's structural novelty scores with biological screening results to effectively discover novel bioactive NPs in large NE collections. Abstract

The combination of various Intelligent Transportation System (ITS) strategies generally has manifold effects, much greater than when strategies are implemented individually. This research introduces a novel dynamic control strategy, which includes the dynamic use of a Model Predictive Control (MPC) based Variable Speed Limit (VSL) integrated with the dynamic use of Hard Shoulder Running (HSR). Both VSL and HSR are proactively triggered to allow for necessary control measures to be taken to delay and possibly avoid the formation of a bottleneck. For traffic prediction, a modified METANET model is developed which takes into consideration the complex nature of driver's behavior along with driver's compliance, capacity drop and posted speed limits. The modified METANET model is shown to be more efficient than conventional macroscopic prediction models in detecting traffic congestions. This MPC based strategy was tested on a section of Deerfoot Trail, Calgary, Alberta using an exclusively developed integrated VISSIM-COM-MATLAB interface. The results from this study suggested that the integrated VSL and HSR control strategy results in a 21.09% increase in average speed and 33.44%. in vehicle-throughput. Furthermore, there was a noticeable reduction in the average travel time by 39.98% and in the total number of stops, by 32.43%. Importantly, the safety analysis performed using Surrogate Safety Assessment Model (SSAM) revealed a notable reduction in collisions, by 29.73%. [ABSTRACT FROM AUTHOR]

Abstract

When the river Yamuna leaves the National Capital Territory of Delhi, its situation further deteriorates. Despite accounting for only 1% of the river's overall catchment area, this region is responsible for more than half of the pollutants discovered in the Yamuna. The river Yamuna, on the other hand, is Delhi's only natural resource for maintaining all forms of life. The Yamuna River is currently experiencing a significant level of pollution problem, and in order to control pollution in the Yamuna River, continual analysis is essential. The Yamuna River is contaminated by the discharge of untreated municipal sewage and industrial effluent through seven major drains: Najafgarh, Yamunapur, Sen Nursing Home, Barathpula, Maharani Bagh, Kalkaji, and Tuglakabad. In terms of people and chemicals, continuous sampling takes time and money. The primary objective of this study is to analyse the wastewater samples collected by sub-drains and STP's to predict the pollutant transportation in river Yamuna from Najafgarh Drain. The study focusses on the only pollutant, i.e., Biochemical Oxygen Demand from the starting point to after the confluence of Najafgarh Drain into river Yamuna. The prediction is to be done by using MATLAB software. This study would help to identify the main sources of sub-drains which are polluting Najafgarh Drain and eventually the river Yamuna. This shows how MATLAB may be used to calculate the pollution load caused by organic waste in the Yamuna River as it flows through Delhi, India's National Capital Territory. The model numerically solves a series of differential equations to simulate the dissolved oxygen and biochemical oxygen demand parameters in two dimensions. MATLAB is an interactive programming language that may be used to develop algorithms, graphics, and user interfaces in other computer languages. MATLAB helps estimate future water quality using present data, which saves time, labour, and other costs associated with the continuous study. There are various software programmes available in the market for predicting river water quality, however, MATLAB GUI provides an accessible and convenient user interface (Graphical User Interface). The applied techniques involve chemical modification, bioconjugation to specific ligands, and carrier-mediated strategies. Nanotechnology-based systems like liposomes, niosomes, solid lipid nanoparticles (SLNs), dendrimers, and polymeric nanoparticles (PNs) are also discussed.Expert opinionAlthough the mechanism of siRNA as a gene silencer is well-established, only a few products are available as therapeutics. There is a great need to develop and establish siRNA delivery systems that protects siRNAs and delivers them efficiently to the desired sitesare efficient and capable of targeted delivery. Several diseases are reported to be controlled by siRNA at their early stages. However, their targeted delivery is a daunting challenge.
