1. Habitability in 4-D: Predicting the Climates of Earth Analogs across Rotation and Orbital Configurations

Abstract

Earth-like planets in the circumstellar habitable zone (HZ) may have dramatically different climate outcomes depending on their spin-orbit parameters, altering their habitability for life as we know it. We present a suite of 93 ROCKE-3D general circulation models (GCMs) for planets with the same surface conditions and average annual insolation as Earth, but with a wide range of rotation periods, obliquities, orbital eccentricities, and longitudes of periastra. Our habitability metric $f_\mathrm{HZ}$ is calculated based on the temperature and precipitation in each model across grid cells over land. Latin Hypercube Sampling (LHS) aids in sampling all 4 of the spin-orbit parameters with a computationally feasible number of GCM runs. Statistical emulation then allows us to model $f_\mathrm{HZ}$ as a smooth function with built-in estimates of statistical uncertainty. We fit our emulator to an initial set of 46 training runs, then test with an additional 46 runs at different spin-orbit values. Our emulator predicts the directly GCM-modeled habitability values for the test runs at the appropriate level of accuracy and precision. For orbital eccentricities up to 0.225, rotation period remains the primary driver of the fraction of land that remains above freezing and with precipitation above a threshold value. For rotation periods greater than $\sim 20$ days, habitability drops significantly (from $\sim 70$% to $\sim 20$%), driven primarily by cooler land temperatures. Obliquity is a significant secondary factor for rotation periods less than $\sim 20$ Earth days, with a factor of two impact on habitability that is maximized at intermediate obliquity., Comment: Accepted for publication in the Astrophysical Journal

4. Solute mixing, dynamic uncertainty and effective dispersion in two-dimensional heterogeneous porous media

Abstract

We study the mixing dynamics of a solute that is transported by advection and dispersion in a heterogeneous Darcy scale porous medium. We quantify mixing and dynamic uncertainty in terms of the mean squared solute concentration and the concentration variance. The latter measures the degree of mixing of the solute and at the same time the uncertainty around the mean concentration. Its evolution is controlled by the creation of concentration fluctuations due to solute spreading and its destruction by local dispersion. For moderate heterogeneity, this interplay can be quantified by using apparent and effective dispersion coefficients. For increasing heterogeneity we find deviations from the predicted behavior. In order to shed light on these behaviors and separate solute mixing and spreading, we decompose the solute plume into partial plumes, transport Green functions, and analyze their dynamics relative to those of the whole plume. This reveals that the variability in the dispersive scales of the Green functions in the plume and their interactions due to the strong focusing of preferential flow paths play an important role in highly heterogeneous porous media.

8. Case report: Selpercatinib in the treatment of RET fusion-positive advanced lung adenocarcinoma: a challenging clinical case

Abstract

BackgroundRearranged during transfection (RET) fusions represent a distinct molecular subset of non-small cell lung cancer (NSCLC) with targeted therapeutic potential. Selpercatinib, a highly selective RET inhibitor, has demonstrated efficacy in various solid tumors harboring RET alterations. Here, we present a case highlighting the use and clinical outcomes of selpercatinib in a patient diagnosed with advanced lung adenocarcinoma harboring a RET fusion.Case presentationA 59-year-old woman with a history of stage IV lung adenocarcinoma harboring a KIF5B-RET fusion presented with disease progression following first-line chemo-immunotherapy. Selpercatinib was initiated as a targeted therapy, leading to a notable radiographic response and clinical improvement. The patient experienced a significant reduction in tumor burden and reported improved symptom control, with no significant adverse effects during the 21-month follow-up period.ConclusionsThis case highlights the efficacy and tolerability of selpercatinib in treating advanced lung adenocarcinoma with a RET fusion. The observed clinical response supports the early use of selpercatinib as a targeted therapy for RET fusion-positive NSCLC, including in patients with compromised general and respiratory conditions, especially in cases refractory to conventional treatments. Long-term follow-up studies are warranted to validate these findings and assess the durability of responses.

11. The cultivation of Primula palinuri Petagna for ex situ conservation: lessons learned from the Royal Rotterdam Zoological and Botanical Gardens

Abstract

Primula palinuri Petagna is an endangered species endemic to a tiny coastal area in southern Italy. Investigating the possibility of growing and propagating this endangered species with ex situ actions should be encouraged, as this could provide a second line of security for rare plants by allowing specimens to be grown in the absence of natural environmental challenges. Currently, the Royal Rotterdam Zoological and Botanical Gardens (RRZBG) hold 31 accessions of P. palinuri, which represents an ex situ collection with potential value for conservation programmes. Over the years, horticulturists at RRZBG have created a solid protocol to produce viable seeds through hand-pollination techniques, thus allowing conservation programmes for this species to become more effective. In this article the cultivation of the species is described, including information on seed germination, general maintenance, cultivation to flowering stage and propagation from the resulting seeds.

Abstract

Abstract Background An international panel of obesity medicine experts from multiple professional organizations examined patterns of obesity care and current obesity treatment guidelines to identify areas requiring updating in response to emerging science and clinical evidence. Aims The panel focused on multiple medical health and societal issues influencing effective treatment of obesity and identified several unmet needs in the definition, assessment, and care of obesity. Methods The panel was held in Leesburg, Virginia in September 2019. Results The panelists recommended addressing these unmet needs in obesity medicine through research, education, evaluation of delivery and payment of care, and updating clinical practice guidelines (CPG) to better reflect obesity’s pathophysiological basis and heterogeneity, as well as the disease’s health, sociocultural, and economic complications; effects on quality of life; need for standards for quantitative comparison of treatment benefits, risks, and costs; and the need to more effectively integrate obesity treatment guidelines into routine clinical practice and to facilitate more direct clinician participation to improve public understanding of obesity as a disease with a pathophysiological basis. The panel also recommended that professional organizations working to improve the care of people with obesity collaborate via a working group to develop an updated, patient‐focused, comprehensive CPG establishing standards of care, addressing identified needs, and providing for routine, periodic review and updating. Conclusions Unmet needs in the definition, assessment and treatment of obesity were identified and a blueprint to address these needs developed via a clinical practice guideline that can be utilized worldwide to respond to the increasing prevalence of obesity.

Abstract

The obesity pandemic continues unabated despite a persistent public health campaign to decrease energy intake ("eat less") and increase energy expenditure ("move more"). One explanation for this failure is that the current approach, based on the notion of energy balance, has not been adequately embraced by the public. Another possibility is that this approach rests on an erroneous paradigm. A new formulation of the energy balance model (EBM), like prior versions, considers overeating (energy intake > expenditure) the primary cause of obesity, incorporating an emphasis on "complex endocrine, metabolic, and nervous system signals" that control food intake below conscious level. This model attributes rising obesity prevalence to inexpensive, convenient, energy-dense, "ultra-processed" foods high in fat and sugar. An alternative view, the carbohydrate-insulin model (CIM), proposes that hormonal responses to highly processed carbohydrates shift energy partitioning toward deposition in adipose tissue, leaving fewer calories available for the body's metabolic needs. Thus, increasing adiposity causes overeating to compensate for the sequestered calories. Here, we highlight robust contrasts in how the EBM and CIM view obesity pathophysiology and consider deficiencies in the EBM that impede paradigm testing and refinement. Rectifying these deficiencies should assume priority, as a constructive paradigm clash is needed to resolve long-standing scientific controversies and inform the design of new models to guide prevention and treatment. Nevertheless, public health action need not await resolution of this debate, as both models target processed carbohydrates as major drivers of obesity.

Abstract

Partially aromatic polyamides owing to their excellent thermal stability are widely used in high temperature applications, however, like their aliphatic counterparts, they are readily flammable and more challenging to process. In this work, several organophosphorus flame retardants were synthesized and compounded with partially aromatic polyamide and evaluated for their processability, thermal, and fire behaviour. The compounds containing a commercial flame retardant, Exolit® OP 1230 (EX), and two new flame retardants, namely 1,4-phenylenebis(diphenylphosphine oxide) (MP) and (1,1′-biphenyl]-4,4′-diylbis(diphenylphosphine oxide) (BP), showed self-extinguishing capability (i.e., UL94 V0 class) with 4 wt% phosphorus (P) loading, together with a substantial reduction in the pHRR (up to 47 %), with respect to the pristine PAP. Rheological measurements on extended timescales were used to assess the melt stability of partially aromatic polyamide compounds. The presence of MP and BP in the polymer matrix did not trigger any excessive degradation phenomena such as chain scission, branching, or crosslinking reactions, thus, allowing a stable processability similar to a pristine partially aromatic polyamide sample. Finally, analysis of evolved gases during thermal decomposition revealed that MP and BP mainly exert a flame inhibition effect quite early in the decomposition process.

Abstract

According to a commonly held view, the obesity pandemic is caused by overconsumption of modern, highly palatable, energy-dense processed foods, exacerbated by a sedentary lifestyle. However, obesity rates remain at historic highs, despite a persistent focus on eating less and moving more, as guided by the energy balance model (EBM). This public health failure may arise from a fundamental limitation of the EBM itself. Conceptualizing obesity as a disorder of energy balance restates a principle of physics without considering the biological mechanisms that promote weight gain. An alternative paradigm, the carbohydrate-insulin model (CIM), proposes a reversal of causal direction. According to the CIM, increasing fat deposition in the body-resulting from the hormonal responses to a high-glycemic-load diet-drives positive energy balance. The CIM provides a conceptual framework with testable hypotheses for how various modifiable factors influence energy balance and fat storage. Rigorous research is needed to compare the validity of these 2 models, which have substantially different implications for obesity management, and to generate new models that best encompass the evidence.

Abstract Long-term human space exploration missions require environmental control and closed Life Support Systems (LSS) capable of producing and recycling resources, thus fulfilling all the essential metabolic needs for human survival in harsh space environments, both during travel and on orbital/planetary stations. This will become increasingly necessary as missions reach farther away from Earth, thereby limiting the technical and economic feasibility of resupplying resources from Earth. Further incorporation of biological elements into state-of-the-art (mostly abiotic) LSS, leading to bioregenerative LSS (BLSS), is needed for additional resource recovery, food production, and waste treatment solutions, and to enable more self-sustainable missions to the Moon and Mars. There is a whole suite of functions crucial to sustain human presence in Low Earth Orbit (LEO) and successful settlement on Moon or Mars such as environmental control, air regeneration, waste management, water supply, food production, cabin/habitat pressurization, radiation protection, energy supply, and means for transportation, communication, and recreation. In this paper, we focus on air, water and food production, and waste management, and address some aspects of radiation protection and recreation. We briefly discuss existing knowledge, highlight open gaps, and propose possible future experiments in the short-, medium-, and long-term to achieve the targets of crewed space exploration also leading to possible benefits on Earth.

Abstract

Abstract Advancements in plant space biology are required for the realization of human space exploration missions, where the re-supply of resources from Earth is not feasible. Until a few decades ago, space life science was focused on the impact of the space environment on the human body. More recently, the interest in plant space biology has increased because plants are key organisms in Bioregenerative Life Support Systems (BLSS) for the regeneration of resources and fresh food production. Moreover, plants play an important role in psychological support for astronauts. The definition of cultivation requirements for the design, realization, and successful operation of BLSS must consider the effects of space factors on plants. Altered gravitational fields and radiation exposure are the main space factors inducing changes in gene expression, cell proliferation and differentiation, signalling and physiological processes with possible consequences on tissue organization and organogenesis, thus on the whole plant functioning. Interestingly, the changes at the cellular and molecular levels do not always result in organismic or developmental changes. This apparent paradox is a current research challenge. In this paper, the main findings of gravity- and radiation-related research on higher plants are summarized, highlighting the knowledge gaps that are still necessary to fill. Existing experimental facilities to simulate the effect of space factors, as well as requirements for future facilities for possible experiments to achieve fundamental biology goals are considered. Finally, the need for making synergies among disciplines and for establishing global standard operating procedures for analyses and data collection in space experiments is highlighted.

Abstract

This study examines the effectiveness of pre-laboratory presentations in an undergraduate general chemistry laboratory using two different modes of delivery: a traditional lecture versus an in-class video presentation. It was anticipated that implementation of videos could result in improved laboratory efficiency, safety, and necessary technical skills for the students. Thus, the purpose of this study was to examine this hypothesis by comparing laboratory quiz scores and completion times of students who received their pre-laboratory instruction in a traditional lecture versus in-class video format. The results of this study provide new insight into the benefits of using technology for delivery of prelaboratory instruction and offer suggestions for considerations when implementing technological tools (such as videos) into laboratory instruction.

Abstract

Nowadays, additive manufacturing technologies have impacted different engineering sectors. Three- and four-dimensional printing techniques are increasingly used in soft and flexible electronics thanks to the possibility of working contemporarily with several materials on various substrates. The materials portfolio is wide, as well as printing processes. Shape memory polymers, together with composites, have gained great success in the electronic field and are becoming increasingly popular for fabricating pH, temperature, humidity, and stress sensors that are integrated into wearable, stretchable, and flexible devices, as well as for the fabrication of communication devices, such as antennas. Here, we report an overview of the state of the art about the application of 4D printing technologies and smart materials in electronics.

Abstract

MEMS devices are more and more commonly used as sensors, actuators, and microfluidic devices in different fields like electronics, opto-electronics, and biomedical engineering. Traditional fabrication technologies cannot meet the growing demand for device miniaturisation and fabrication time reduction, especially when customised devices are required. That is why additive manufacturing technologies are increasingly applied to MEMS. In this review, attention is focused on the Italian scenario in regard to 3D-printed MEMS, studying the techniques and materials used for their fabrication. To this aim, research has been conducted as follows: first, the commonly applied 3D-printing technologies for MEMS manufacturing have been illustrated, then some examples of 3D-printed MEMS have been reported. After that, the typical materials for these technologies have been presented, and finally, some examples of their application in MEMS fabrication have been described. In conclusion, the application of 3D-printing techniques, instead of traditional processes, is a growing trend in Italy, where some exciting and promising results have already been obtained, due to these new selected technologies and the new materials involved.

Abstract

Quantifying the vertical distribution of atmospheric aerosols is crucial for estimating their impact on the Earth energy budget and climate, improving forecast of air pollution in cities, and reducing biases in the retrieval of greenhouse gases (GHGs) from space. However, to date, passive remote sensing measurements have provided limited information about aerosol extinction profiles. In this study, we propose the use of a spectral sorting approach to constrain the aerosol vertical structure using spectra of reflected sunlight absorption within the molecular oxygen (O2) A-band collected by the Orbiting Carbon Observatory-2 (OCO-2). The effectiveness of the approach is evaluated using spectra acquired over the western Sahara coast by comparing the aerosol profile retrievals with lidar measurements from the Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observation (CALIPSO) Cloud-Aerosol Lidar with Orthogonal Polarization (CALIOP). Using a radiative transfer model to simulate OCO-2 measurements, we found that high-resolution O2 A-band measurements have high sensitivity to aerosol optical depth (AOD) and aerosol layer height (ALH). Retrieved estimates of AOD and ALH based on a look up table technique show good agreement with CALIPSO measurements, with correlation coefficients of 0.65 and 0.53, respectively. The strength of the proposed spectral sorting technique lies in its ability to identify spectral channels with high sensitivity to AOD and ALH and extract the associated information from the observed radiance in a straightforward manner. IntroductionLong-duration missions in outer Space will require technologies to regenerate environmental resources such as air and water and to produce food while recycling consumables and waste. Plants are considered the most promising biological regenerators to accomplish these functions, due to their complementary relationship with humans. Plant cultivation for Space starts with small plant growth units to produce fresh food to supplement stowed food for astronauts’ onboard spacecrafts and orbital platforms. The choice of crops must be based on limiting factors such as time, energy, and volume. Consequently, small, fast-growing crops are needed to grow in microgravity and to provide astronauts with fresh food rich in functional compounds. Microgreens are functional food crops recently valued for their color and flavor enhancing properties, their rich phytonutrient content and short production cycle. Candidate species of microgreens to be harvested and eaten fresh by crew members, belong to the families Brassicaceae, Asteraceae, Chenopodiaceae, Lamiaceae, Apiaceae, Amarillydaceae, Amaranthaceae, and Cucurbitaceae.MethodsIn this study we developed and applied an algorithm to objectively compare numerous genotypes of microgreens intending to select those with the best productivity and phytonutrient profile for cultivation in Space. The selection process consisted of two subsequent phases. The first selection was based on literature data including 39 genotypes and 25 parameters related to growth, phytonutrients (e.g., tocopherol, phylloquinone, ascorbic acid, polyphenols, lutein, carotenoids, violaxanthin), and mineral elements. Parameters were implemented in a mathematical model with prioritization criteria to generate a ranking list of microgreens. The second phase was based on germination and cultivation tests specifically designed for this study and performed on the six top species resulting from the first ranking list. For the second selection, experimental data on phytonutrients were expressed as metabolite production per day per square meter.Results and discussionIn the final ranking list radish and savoy cabbage resulted with the highest scores based on their productivity and phytonutrient profile. Overall, the algorithm with prioritization criteria allowed us to objectively compare candidate species and obtain a ranking list based on the combination of numerous parameters measured in the different species. This method can be also adapted to new species, parameters, or re-prioritizing the parameters for specific selection purposes.
