Abstract

Objectives ANA-associated rheumatic and musculoskeletal (MSK) diseases (RMDs) [SLE, primary SS (pSS), scleroderma, inflammatory myositis, MCTD and UCTD] make up a disease spectrum with overlapping clinical and immunological features. MSK inflammation is common and impactful across ANA-associated RMDs. The objectives of this study were to evaluate MSK inflammation (ANA-associated arthritis) prevalence in a multidisease ANA-associated RMD study, assess its clinical impact across ANA-associated RMD diagnoses, propose new basket groupings of patients, and evaluate immunological profiles in legacy and new basket contexts. Methods An observational study enrolled patients with ANA-associated RMDs. Demographic variables, comorbidities, therapies, disease activity instruments [BILAG, SLEDAI, the EULAR SS disease activity index (ESSDAI), physician visual analogue scale (VAS)], patient-reported outcomes [SF36, FACIT-Fatigue, EQ5D, ICECAP-A, Work Productivity and Activity impairment (WPAI), patient VAS] and the biomarker profile (six-gene expression scores, flow cytometry, autoantibody profile) were analysed. Reclustering utilized Gaussian mixture modelling (GMM). The clinical and immune features of new and legacy clusters were compared. Results Inflammatory MSK symptoms were prevalent across ANA-associated RMDs, in 213/294 patients. In ANA-associated arthritis patients, most variables did not differ between diagnoses, with the exception of the EQ5D-5L index and mobility domains (lower in MCTD/pSS, both P  < 0.05). FM and OA prevalence were similar across diagnoses. Therapy use differed significantly, the use of biologics being greatest in SLE (P  < 0.05). GMM yielded two multidisease clusters: High MSK disease activity (n  = 89) and low MSK disease activity (n  = 124). The high MSK disease activity cluster included all patients with active joint swelling, and they had significantly higher prednisolone usage, physician global assessment (PGA), Sm/RNP/SmRNP/chromatin positivity, Tetherin mean fluorescence intensity (MFI), and IFN Score-A activity, along with numerically lower FM and OA prevalence. Conclusion We defined ANA-associated arthritis, a more clinically and immunologically homogeneous population than existing RMD populations for trials, and a more prevalent population for therapies in the clinic. The human microbiome has been inextricably linked to multiple facets of human physiology. From an engineering standpoint, the ability to precisely control the composition and activity of the microbiome holds great promise for furthering our understanding of disease etiology and for new avenues of therapeutic and diagnostic agents. While the field of microbiome research is still in its infancy, growing engineering efforts are emerging to enable new studies in the microbiome and to rapidly translate these findings to microbiome‐based interventions. At the 3rd International Conference on Microbiome Engineering, leading experts in the field presented state‐of‐the‐art work in microbiome engineering, discussing probiotics, prebiotics, engineered microbes, microbially derived biomolecules, and bacteriophage. Abstract

JWG A3.32 with CIRED completed the Technical Brochure, which provides comprehensive information on Non-Intrusive Condition assessment of the T&D switchgear that can be applied on HV and MV circuit breakers, HV and MV switching equipment, MV reclosers and fault interrupters. Many emerging non-intrusive techniques were reported in the literature, most of them allowing in service evaluation. The JWG reviewed the current and future trends of non-intrusive, especially in service, diagnostic methods completing the work on Condition Assessment of Switchgears dealt in TB 167 and on Online Monitoring HV Substations available in TB 462. The brochure will provide useful information for everybody who is going to apply or improve Condition Assessment Methods to Switchgears., QC 20181129

Abstract

Photovoltaic electric-powered flight is receiving a great deal of attention in the context of the United States' Unmanned Aerial Vehicle (UAV) program. This paper addresses some of the enabling technical areas and their potential solutions. Of particular interest are the long-duration, high-altitude class of UAV's whose mission it is to achieve altitudes between 60,000 and 100,000 feet, and to remain at those altitudes for prolonged periods performing various mapping and surveillance activities. Addressed herein are studies which reveal the need for extremely light-weight and efficient solar cells, high-efficiency electric motor-driven propeller modules, and power management and distribution control elements. Since the potential payloads vary dramatically in their power consumption and duty cycles, a typical load profile has been selected to provide commonality for the propulsion power comparisons. Since missions vary widely with respect to ground coverage requirements, from repeated orbiting over a localized target to long-distance routes over irregular terrain, we have also averaged the power requirements for on-board guidance and control power, as well as ground control and communication link utilization. In the context of the national technology reinvestment program, wherever possible we modeled components and materials which have been qualified for space and defense applications, yet are compatible with civilian UAV activities. These include, but are not limited to, solar cell developments, electric storage technology for diurnal operation, local and ground communications, power management and distribution, and control servo design. And finally, the results of tests conducted by Wright Laboratory on ultralight, highly efficient MOCVD GaAs solar cells purchased from EPI Materials Ltd. (EML) of the UK are presented. These cells were also used for modeling the flight characteristics of UAV aircraft.

40. Dual-purpose self-deliverable lunar surface PV electrical power system

Abstract

A safe haven and work supported PV power systems on the lunar surface will likely be required by NASA in support of the manned outpost scheduled for the post-2000 lunar/Mars exploration and colonization initiative. Initial system modeling and computer analysis shows that the concept is workable and contains no major high risk technology issues which cannot be resolved in the circa 2000 to 2025 timeframe. A specific selection of the best suited type of electric thruster has not been done; the initial modeling was done using an ion thruster, but Rocketdyne must also evaluate arc and resisto-jets before a final design can be formulated. As a general observation, it appears that such a system can deliver itself to the Moon using many system elements that must be transported as dead payload mass in more conventional delivery modes. It further appears that a larger power system providing a much higher safe haven power level is feasible if this delivery system is implemented, perhaps even sufficient to permit resource prospecting and/or lab experimentation. The concept permits growth and can be expanded to include cargo transport such as habitat and working modules. In short, the combined payload could be manned soon after landing and checkout. NASA has expended substantial resources in the development of electric propulsion concepts and hardware that can be applied to a lunar transport system such as described herein. In short, the paper may represent a viable mission on which previous investments play an invaluable role. A more comprehensive technical paper which embodies second generation analysis and system size will be prepared for near-term presentation.

Abstract

The CCR5 receptor plays a role in several key physiological and pathological processes and is an important therapeutic target. Inhibition of the CCR5 axis by passive or active immunisation offers one very selective strategy for intervention. In this study we define a new linear epitope within the extracellular domain of CCR5 recognised by two independently produced monoclonal antibodies. A short peptide encoding the linear epitope can induce antibodies which recognise the intact receptor when administered colinear with a tetanus toxoid helper T cell epitope. The monoclonal antibody RoAb 13 is shown to bind to both cells and peptide with moderate to high affinity (6x10^8 and 1.2x107 M-1 respectively), and binding to the peptide is enhanced by sulfation of tyrosines at positions 10 and 14. RoAb13, which has previously been shown to block HIV infection, also blocks migration of monocytes in response to CCR5 binding chemokines and to inflammatory macrophage conditioned medium. A Fab fragment of RoAb13 has been crystallised and a structure of the antibody is reported to 2.1 angstrom resolution.

Abstract

The National Museum of the Pacific War presents an oral interview with Jack O. Arnold. Arnold was born in El Paso, Texas on 27 June, 1923. He enlisted in the Army in May 1942 while in his second year at the Texas College of Mines and Metallurgy. After basic training at Camp Maxey, Texas, he joined the 14th Armored Division. Circa 1944-1945, he joined the 503rd Parachute Infantry Regiment. He participated in the battle for the recapture of Corregidor. He discusses the Japanese defensive network of tunnels on the island. He was awarded the Purple Heart for a shrapnel injury while on Negros Island. Arnold describes the living conditions of the soldiers. He also describes the Japanese practice of binding themselves up with cloth for battle. He was discharged from the Army on 31 December, 1945. After he finished college, he enlisted in the Air Force in 1949 at the age of 26 and served during the Korean War. He recounts stories of flying in an airplane during a typhoon, a bird strike, engine trouble and landing gear failure. He was discharged from the Air Force in 1952. Arnold provides information about his parents and siblings as well as his children.
