Rhus coriaria (sumac) naturally grows worldwide and conta-ins many phytochemicals. Tannic acid is frequently used for treating medical conditions. The aim of this study is to de-termine the synergistic effects of methanolic and acetonic extracts of Rhus coriaria’s from unriped fruits in combination with tannic acid on angiogenesis by using the Chick Chori-oallantoic Membrane (CAM) assay. The effects of methanol and acetone extracts of Sumac (MES and AES, respectively, at a final concentration of 5 mg/mL for both) and tannic acid (Tan 1.25 and 5 mg/mL) alone or in combination were exa-mined for their effect on angiogenesis by applying the CAM assay. CAM assay results showed that AES is more effective than MES on suppressing angiogenesis. Tannic acid started to show antiangiogenic effects at the concentration of 5 mg/mL. Background/Aim: [Pd(sac)(terpy)](sac)•4H2O (sac=saccharinate and terpy=2,2′:6′,2-terpyridine) is newlysynthesized palladium(II) (Pd) complex. We investigated the antiproliferative and apoptotic effects of this complex on Ehrlich ascites carcinoma (EAC). Materials and Methods: EAC cells were administered to 33 Balb/c mice. Mice were divided randomly into four groups: control, cisplatin, Pd(II) complex and paclitaxel. Meme kanseri, tüm dünyada en önemli ölüm nedenlerinden biridir. Son yıllarda kanser tedavisinde çeşitli bileşiklerin, kemoterapötik ajanlar ile kombine edilerek aktivelerinin arttırılması hedeflenmektedir. Bir polifenol olan Mangiferin'in, kanserde önemli bir role sahip olabileceği bildirilmiştir. Bu tez çalışmasında Ehrlich Ascites Carcinoma (EAC) fare meme tümöründe, Mangiferin'in ve Paklitaksel'in tek başına ve kombinasyonlarının, hücre proliferasyonu, apoptozis, otofaji ve sağkalım üzerine etkisinin in vitro ve in vivo olarak incelenmesi amaçlanmıştır. Ayrıca, anjiyogenez üzerine etkisi de HUVEC tüp formasyon yöntemi ve in vivo CAM modelinde değerlendirilmiştir. EAC ve HUVEC hücrelerinde Mangiferin (6,25-300 µM) ve Paklitaksel (0,25-15,9 µM) tek başına ve kombinasyonunun etkisi, 48. saat ATP sitotoksisite testi ile değerlendirildi. Mangiferin ve Paklitaksel kombinasyonunda apoptozis, otofaji ve sağkalım belirteçleri western blot yöntemi ile belirlendi. HUVEC tüp formasyon testinde Mangiferin (12,5-50 µM), Paklitaksel (1, 99-15,93 µM) ve kombinasyonları uygulanarak 0-16 saat aralığında değerlendirildi. CAM yönteminde ise 12,5-5 mg/ml, Paklitaksel 0,3-0,0375 mg/ml ve kombinasyonları uygulandı. In vivo çalışmada 78 adet solid tümörlü dişi Balb/c fareye 25-50-100 mg/kg Mangiferin gavajla, 12,5 mg/kg Paklitaksel subkutan uygulandı ve 14. günde alınan dokularda proliferasyon, apoptozis, otofaji ve sağkalım belirteçleri immünohistokimyasal olarak incelendi, sonuçlar istatistiksel olarak değerlendirildi. ATP testi sonuçlarında sitotoksik etkinin, kombinasyon grubunda doza ve hücre çeşidine bağlı olarak sinerjistik biçimde arttığı görüldü. Western blot sonuçları değerlendirildiğinde, EAC hücrelerinde LC3B'nin yükselmesi ve p62'nin azalması ile FAS ve kırılmış PARP'ın artması, dış yolak üzerinde apoptozisin otofaji varlığında gerçekleştiği ve sağkalım parametrelerinin de baskılandığı görülmüştür. İn vivo da in vitro sonuçlarına parallelik göstermiş ve tümör büyüklüğü, tümör ağırlığı, apoptozis, otofaji ve sağkalım üzerinde en etkili dozun tek başına Mangiferin 50 mg/kg ve kombinasyonu grubunda olduğu görülmüştür. Mangiferin ve kombinasyonlarının anjiyogenez üzerine etkisinin incelendiği in vitro tüp formasyon testinde, tek başına 50 µM dozunun etkili olduğu, CAM sonuçlarında ise kombinasyon yapılan gruplarda daha etkili antianjiyogenik etkiye sahip olduğu görülmüştür. Sonuç olarak, tüm bu bulguların doğrultusunda in vitro ve in vivoda Mangiferin'in doza bağlı olarak, Paklitaksel kombinasyonu ile EAC hücrelerini seçici bir biçimde, otofaji varlığında apoptozisi arttırdığı, proliferasyonu ve sağkalım parametrelerini azalttığı, güçlü bir antianjiyogenik etkiye de sahip olmasıyla tümör büyüklüğünü baskıladığı ilk defa bu tez çalışması ile gösterilmiştir. Mangiferin'in düşük dozları Paklitaksel ile kombine edildiğinde, tek başına olduğundan daha güçlü sinerjik etki göstermektedir. Mangiferin'in özellikle kanser tedavisinde gelecekteki yeni stratejilerin geliştirilmesi için umut vaad edici potansiyele sahip olduğu düşünülmektedir. Recent years, various compounds to treat cancer is aimed to increase in combination with chemotherapeutic agents activate. Mangiferin is a polyphenol that has been reported may have an important role in cancer. The aim of this study is to investigate in vitro and in vivo effects of Mangiferin and Paclitaxel alone and in combinations of cell proliferation, apoptosis, autophagy and survival pathway on Ehrlich Ascites Carcinoma (EAC) mouse breast tumor. In addition, the effect on angiogenesis was also evaluated in the HUVEC tube formation method and in vivo CAM model.The effect of Mangiferin (6.25-300 µM) and Paclitaxel (0.25-15.9 µM) alone and in combination on EAC and HUVEC cells were assessed at 48th hour by ATP cytotoxicity test. In combination with Mangiferin and Paclitaxel, apoptosis, autophagy, and survival markers were determined by western blot method. The effect of Mangiferin (12.5-50 µM) and Paclitaxel (1.99-15.9 µM) alone and in combination on HUVEC cells were evaluated at 0-16 h by tube formation test. Mangiferin (12.5-5 mg/ml) and Paclitaxel (0.3-0.0375 mg/ml µM) alone and in combination were treated in CAM assay. In an in vivo study, female Balb/c mice with 78 solid tumors were administered 12.5 mg/kg Paclitaxel by subcutaneously and 25-50-100 mg/kg Mangiferin by gavage, then proliferation, apoptosis, autophagy, and survival markers were examined immunohistochemically in the tumors taken on the 14th day and the results were evaluated statistically.The ATP test results showed that the cytotoxic effect increased synergistically in the combination group due to the dose and cell type. According to evaluation of western blot results, the increase of LC3B with the decrease of p62 and the increase of FAS with cleaved PARP was shown that apoptosis occurred through the external pathway in the presence of autophagy. Furthermore, the survival parameters were suppressed. The in vitro results were parallel to in vivo and the most effective dose on tumor size, tumor weight, apoptosis, autophagy, and survival were determined as Mangiferin 50 mg/kg alone and combination. In vitro tube formation test, in which the effect of Mangiferin and its combinations on angiogenesis was examined, it was observed that 50 µM dose was effective alone and it was more effective antiangiogenic effect in combination groups on the CAM assay. In conclusion, according to all these results, dose-dependent Mangiferin with Paclitaxel combination of EAC cells selectively increased apoptosis in the presence of autophagy, decreased proliferation and survival parameters, and suppressed tumor size with a strong antiangiogenic effect, were shown for the first time in this thesis. Low doses of Mangiferin combination with Paclitaxel shown a stronger synergistic effect than alone treatment.
