Taisho Sanshoku is a variant of Koi fish (Cyprinus carpio) that has high demand due to its high economic value and relatively expensive price. This study aimed to determine the sex of the Taisho Sanshoku Koi fish by molecular sexing using the PCR method to amplify the ArS.9-15 gene. This study was initiated by rearing a 4–6 month-old of 10 Taisho Sanshoku Koi fish in a fish tank with a filter and oxygen aeration. The fish were fed with fish pellets for 1–3 days. The Koi fishes were then anesthetized using Koi anesthesia containing β-hydroxyethyl phenyl ether. Each fish's peripheral blood was collected as much as 0.5 mL per fish and then stored in tubes containing Ca-EDTA anticoagulant. The genomic DNA was extracted from peripheral blood samples and used as a template DNA for PCR amplification targeting ArS.9-15 gene. Agarose with 1.5% concentration and CybrSafe staining was used in electrophoresis for visualization of the PCR results then visualized in a dark chamber using a UV transilluminator. The Taisho Sanshoku Koi fish's sex was determined using descriptive analysis based on the electrophoresis results. According to the PCR results, the female Taisho Sanshoku Koi fish only produced one 800 bp DNA band, whereas the male fish produced two 800 bp and 1,100 bp DNA bands. The outcome of molecular fish sexing of the 10 Taisho Sanshoku Koi fish reported that 60% were male and 40% were female.

Background: Eonycteris spelaea is a sizable nectar-feeding bat that falls within the taxonomic classification of Order Chiroptera and Family Pteropododae. The form and structure of the tongue play a crucial role for bats in the intake and digestion of food in their mouth. Each papilla's morphology, dimensions, spatial arrangement, and physiological role exhibit variations among different animal species, contingent upon their respective lifestyles. Aim: This research attempts to examine the morphology and microstructure of the Eonycteris spelaea's lingual papillae, collected from Timor Island, East Nusa Tenggara, Indonesia. Methods: This study will achieved a scanning electron microscope and a light microscope in the presence of haematoxylin eosin staining and employed a sample of 6 sexually indiscriminate adult Eonyteris spelaea bats. Results: The tongue of Eonycteris spelaea separated into three distinct parts: the apex, corpus, and radix. The structure's apex contains filiform papillae, which come in many varieties, such as scale-like filiform papilla, enormous trifid papilla, and little crown-like papilla. Additionally, there is a cluster of fungiform papillae on the outside edge of the highest point and transitional papillae connecting the large trifid papillae with the smaller crown-shaped papillae. The corpus section comprises two papilla types: filiform papilla (leaf-shaped filiform papilla and big crown-shaped papilla) and fungiform papilla. The radix comprises the elongated conical papilla, rosette-shaped filiform papilla, short conical papilla, transitional papilla, and three circumvallate papillae at the back of the tongue. Conclusion: Lingual papillae E. spelaea comprise wide variety of mechanic papillae and also sensory papillae which specific dietary regimen on their living habitat. Salmonellosis is an infectious disease that is zoonotic and includes food borne disease. Salmonella spp. can also be found in pets so that it can be a source of transmission of Salmonellosis to humans. The purpose of this study was to determine the infection level of Salmonella spp. in dogs and cats in clinics/animal hospitals in the Special Region of Yogyakarta. A total of 250 rectal swabs from 83 dogs and 167 cats were taken in this study. Isolation and identification of Salmonella spp. using Xylose Lysine Deoxycholate (XLD), Triple Sugar Iron Agar (TSIA), and Lysine Iron Agar (LIA) media. The isolates were confirmed by Polymerase Chain Reaction (PCR)using a primer Forward primer (5'- GCT AAG TAT GAC ATT CCG GT -3') and reverse (5'- CCA AAG ACT ATC TGC GGA AT -3') eith targeting the STM2773 (IroB) gene. Information on the patient's medical history was obtained based on the anamnesis and questionnaire to the animal owner. Data analysis was descriptive statistics. A total of 32 isolates of Salmomella spp. obtained through conventional methods and as many as 30 samples (12%) confirmed Salmonella spp. using PCR. A total of 11 (13.25%) of 83 dogs and 19 (11.27%) of 167 cats were identified as positive for Salmonella spp. The results of this study indicated that 12% of pet animals in the Special Region of Yogyakarta were infected with Salmonella spp.

The Koi fish (Cyprinus carpio var. koi) is an ornamental fish with a high selling value because of its attractive colors, color patterns, body shape, and swimming motion. Koi fish is extensively traded in the international fish market because of their popularity among hobbyists from numerous countries worldwide. This review discusses various aspects of Koi fish cultivation, including genetic involvement, selective breeding strategies, and management systems. By examining crucial factors such as water parameters, technological innovations, and evolving cultivation methods, this review explored their influence on the quality of Koi fish. Breakthrough technologies, such as ornamental fish warehousing and recirculation aquaculture systems, enhance breeding efficiency and profitability. Molecular sexing, feed optimization, and color enhancement strategies are central to pursuing superior Koi fish. Reproduction management, disease prevention, and risk reduction during transport underscore ongoing efforts to ensure their survival. Despite notable progress, several challenges remain, including limited genetic studies, gaps in disease research, and unexplored herbal alternatives. The active involvement of hobbyists and breeders in research initiatives is a pivotal force in unlocking the untapped potential. The holistic approaches to enhance production efficiency and improve care standards require further exploration, paving the way for a sustainable future in the evolving management of Koi fish cultivation.

Penentuan jenis kelamin pada beberapa spesies burung cukup sulit dilakukan dikarenakan jantan dan betina memiliki ciri morfologi yang sama (monomorfik), salah satunya famili Columbidae. Teknik penentuan jenis kelamin burung secara molekuler yang populer adalah metode Polymerase Chain Reaction (PCR) dengan gen target Chromo Helicase DNA-binding (CHD), namun keberhasilan amplifikasi gen target pada PCR tersebut dipengaruhi salah satunya kesesuaian DNA template dengan primer yang digunakan. Penelitian ini bertujuan untuk mengevaluasi 5 jenis primer PCR P2/P8, 2550F/2718R, CHD1F/CHD1R, 1237L/1272H dan CHD1LF/CHD1LR untuk menentukan jenis kelamin famili Columbidae. Penelitian ini dilakukan dengan menguji 5 jenis primer tersebut di atas pada sampel DNA tiap pasang jantan dan betina dari burung merpati, balam jambi, punai, derkuku, dan perkutut. Hasil penelitian diketahui primer CHD1LF/CHD1LR menunjukkan hasil terbaik dan direkomendasikan untuk menentukan jenis kelamin famili Columbidae.

Determination sex of birds by the Polymerase Chain Reaction (PCR) method is increasingly popular among researchers and the public. The PCR method is known to provide accurate test results in determining the sex of birds, but one of its successes is greatly influenced by the compatibility between the primers used and the template DNA. This study was conducted to evaluate the use of primers 2550F/2718R, 1237L/1272H, P2/P8, and CHD1F/CHD1R in birds from the families Phasianidae, Estrildidae, Passeridae, and Psittacidae. PCR sexing was performed on DNA samples from the 4 bird families above, using primers 2550F/2718R, 1237L/1272H, P2/P8, and CHD1F/CHD1R. PCR results were evaluated by comparing the size of the PCR band with a reference. The results showed that the CHD1F/CHD1R primer could be used in 4 families tested. PCR products for male birds showed 1 band measuring approximately 500 bp, while for female birds 2 bands were 500 and 320 bp. Primer 2550F/2718R could only be used in the families Phasianidae and Psittacidae, while 1237L/1272H and P2/P8 could not be used in the samples tested. CHD1F/CHD1R primers can be used to determine the sex of birds from the families Phasianidae, Estrildidae, Passeridae, and Psittacidae.

Background and Aim: Enrofloxacin and tylosin can be combined into an antibiotic formulation which is expected to have a broader range of antibacterial activity against various infections in broilers. Validation method analysis of the levels of these two active compounds needs to be done for future use in pharmacokinetic or residual studies. The present study aims to determine a suitable validation method of isocratic high-performance liquid chromatography (HPLC) to measure the concentration of antibiotic combinations in the broiler liver, kidney, and muscles. Materials and Methods: The combination of enrofloxacin and tylosin in the liver, kidney, and muscle was validated by HPLC method to find the procedures, processes, equipment, and systems used, consistently provides the appropriate results. The chromatography system consisted of an Octadecyl-silica column of 5 μm in diameter and 150 mm in length with a mobile phase of a mixture of 0.05 M monobasic sodium phosphate (pH 2.5) and acetonitrile (65:35 v/v). The solution was detected at a wavelength of 280 nm, 30°C, a flow rate of 1 mL/min, and an injection volume of 20 μL. The combination antibiotics powder was produced from PT Tekad Mandiri Citra, Bandung, Indonesia, and broiler tissues obtained from day-old chick broilers maintained for 30 days with free antibiotic feed. Results: Validation of a combination solution of enrofloxacin and tylosin shows the linearity values of enrofloxacin and tylosin in the liver, kidney, and muscles as r2=0.9988, r2=0.9999, r2=0.9997, r2=0.9989, r2=0.9978, and r2=0.9962. The accuracy and precision values of enrofloxacin in the liver, kidney, and muscles were 5.53, 6.23, and 6.93, respectively. The values of accuracy and precision of tylosin in the liver, kidney, and muscles were 10.43, 4.63, and 7.16%, respectively. The retention times for enrofloxacin and tylosin were 1.945-2.000 min and 4.175-4.342 min. The limit of detection (LOD) and limit of quantity (LOQ) values for enrofloxacin were 3.03 and 10.1 μg/g, respectively. In contrast, the LOD and LOQ values for tylosin were 9.05 and 30.17 μg/g, respectively. Conclusion: The value of linearity, accuracy, precision, specificity, and sensitivity of the combined solution of enrofloxacin and tylosin showed promising results.

The yellow-crested cockatoo (Cacatua sulphurea, Gmelin 1788) is an endemic bird in eastern part of Indonesia with monomorphic characteristics and included in the list of endangered birds. A method of sex determine in monomorphic birds is by molecular sexing which is based on the PCR amplification of the chromodomain helicase DNA-binding 1 (CHD-1) gene of the bird sex chromosome. This study was aimed to sex determine of the C. sulphurea by amplifying the CHD-1 gene on the Z and W chromosomes and comparing the PCR amplification results from peripheral blood and plucked feathers samples. The samples used were four birds of C. sulphurea from the Wildlife Rescue Centre (WRC), Yogyakarta. The feathers obtained from the ventral wings of each bird were plucked. Through the cutting of the birds' nails, the peripheral blood samples were collected into microhematocrit tubes which contained Heparin. The amplification of the CHD-1 gene used the PCR method with specific primers, such as NP, P2, and MP. Moreover, the PCR results were visualized on 1.5% agarose gel using UV-Transilluminator, at a wavelength of 280 nm. The PCR products (amplicons) were in 297 bp and 392 bp DNA bands, depending on the sex of the bird being tested. It was also observed that the male C. sulphurea produced single 392 bp DNA fragment of the Z chromosome. However, the female birds produced two DNA fragments of the Z and W chromosomes, with a length of 297 bp and 392 bp. The results showed that samples obtained from peripheral blood produced clearer DNA bands compared to plucked feather. It concludes that the extracted DNA from peripheral blood samples have a better quality compared to samples from plucked feathers. Amplification of the CHD-1 gene in male C. sulphurea generated only a single DNA fragment in size of 392 bp, so the four C. sulphurea were male birds.

Sex identification of endangered and protected birds in captivity is very important for conservation programs. Half of the world’s bird species are monomorphic, where male and female are difficult to distinguished morphologically, including cockatoos. Sex identification using molecular bird sexing is more accurate and applicable because it directly targets the sex chromosomes. The purpose of this study was to determine the sex of Sulphur‐crested cockatoo (Cacatua galerita) by detecting differences in the intron size of the chromodomain helicase DNA‐binding 1 (CHD1) gene on the Z and W chromosomes by polymerase chain reaction (PCR) method and to compare of plucked feathers and blood samples as DNA sources for molecular bird sexing. DNA was extracted from feather and blood samples from four C. galerita. Extracted DNA was amplified on the CHD1 gene by PCR method with P2, MP, and NP primers, which were visualized using agarose gel 1.5% under UV transilluminator with a wavelength of 280 nm. The resulting PCR product was detected at 392 bp for the CHD1 Z gene segment and 297 bp for CHD1 W gene segments, where males showed a single DNA band (ZZ) and females showed a double DNA band (ZW). Four C. galerita were 100% successfully determined, consisting of one female and three males. Electrophoresis results showed DNA bands from blood samples were thicker and brighter than DNA bands from feather samples.

The aim of this work was the in vitro expression of the recombinant fusion (F) protein of Newcastle disease virus (NDV). The pBT7-N-His-Fusion-NDV expression plasmid which carries the recombinant F protein encoding gene from local Indonesian isolates, was prepared and transformed into E. coli BL21 (DE3). To detect bacterial colonies carrying the recombinant plasmid, a restriction endonuclease analysis was performed using the EcoRI restriction endonuclease. These results showed that the pBT-N-His-Fusion-NDV plasmid was successfully isolated with a size of 4.601 bp, and three recombinant plasmids carrying the gene coding for the recombinant F protein of NDV were obtained. Selected recombinant plasmids were then in vitro by using a cell-free protein expression system followed by visualization of the recombinant F protein on a 12% SDS-PAGE gel both by Coomassie Brilliant Blue staining and Western blotting. Recombinant F protein was successfully in vitro expressed by using a cell-free protein expression system as indicated by a specific single protein band with a molecular mass of 25.6 kDa.

This study aimed to determine the sex of Tanimbar Cockatoo (Cacatua goffiniana) birds by amplifying Chromodomain Helicase DNA-binding-1 (CHD-1) gene on Z and W sex chromosomes as well as to compare the quality of DNA extraction and PCR amplification products from samples derived from peripheral blood and plucked feathers. This work used five C. goffiniana birds which were collected from the Wildlife Rescue Center (WRC) in Pengasih, Kulon Progo, Yogyakarta. From each C. goffiniana, feather samples were collected by plucking feathers on the ventral wing and peripheral blood samples were taken by cutting their nails and collecting the blood into microhematocrit tubes containing heparin. The next stage was DNA extraction and DNA amplification on the CHD-1 gene using the PCR method by NP, P2, and MP primer pairs. Then, products of DNA extraction and PCR amplification were electrophoresed on 1.5% agarose gel and visualized under a UV light transilluminator with a wavelength of 260 nm. The visualization showed that samples from peripheral blood generated clearer DNA fragments compared to plucked feathers. Two of the five samples were male C. goffiniana and the other three samples were females. In the male Tanimbar Cockatoo was amplified a single DNA fragment of the Z sex chromosome in size of 300 bp, whereas in the female C. goffiniana was amplified double DNA fragments of Z and W sex chromosomes in size of 300 bp and 400 bp respectively. The DNA quality showed that the DNA quality from peripheral blood samples were better in quality than the DNA collected from plucked feather samples.

Background and Aim: The feasibility assessment of food products on the market becomes one of the milestones of food safety. The quality of food safety of animal origin especially pork need to get attention and more real action from the parties related and concerned. Since pork is also a source of transmission for the contagion of foodborne disease so that the study of the existence of several agents in the pork and its products become the benchmark of safety level. This study aimed to isolate, identify, and detect the Shiga toxin 2a (stx2a) gene from Escherichia coli O157:H7 in pork, pig feces, and clean water in the Jagalan slaughterhouse. Materials and Methods: A total of 70 samples consisting of 32 pork samples, 32 pig fecal samples, and 6 clean water samples were used to isolate and identify E. coli O157:H7 and the stx2a gene. Isolation and identification of E. coli O157:H7 were performed using culture on eosin methylene blue agar and Sorbitol-MacConkey agar media and confirmed molecularly with polymerase chain reaction to amplify the target genes rfbE (317 bp) and fliC (381 bp). The isolates, which were identified as E. coli O157:H7, were investigated for the stx2a gene (553 bp). Results: The results of this study show that of the total collected samples, E. coli O157:H7 was 28.6% in Jagalan slaughterhouse and consisted of 25% of pork samples, 31.25% of pig fecal samples, and 33.3% of clean water samples. The isolates that were identified to be E. coli O157:H7 mostly contained the stx2a gene, which was equal to 75%, and consisted of seven isolates from pork samples, seven isolates from fecal samples, and one isolate from clean water samples. Conclusion: E. coli O157:H7 was found in 28.6% of pork, pig feces, and clean water in Jagalan slaughterhouse and 75% of identified E. coli O157:H7 contained the stx2a gene.

Background and Aim: Many avian species are considered sexually monomorphic. In monomorphic bird species, especially in young birds, sex is difficult to identify based on an analysis of their external morphology. Accurate sex identification is essential for avian captive breeding and evolutionary studies. Methods with varying degrees of invasiveness such as vent sexing, laparoscopic surgery, steroid sexing, and chromosome inspection (karyotyping) are used for sex identification in monomorphic birds. This study aimed to assess the utility of a non-invasive molecular marker for gender identification in a variety of captive monomorphic birds, as a strategy for conservation. Materials and Methods: DNA was isolated from feather samples from 52 individuals representing 16 species of 11 families indigenous to both Indonesia and elsewhere. We amplified the chromodomain helicase DNA-binding (CHD) gene using polymerase chain reaction with MP, NP, and PF primers to amplify introns with lengths that differ between the CHD-W and the CHD-Z genes, allowing sex discrimination because the W chromosome is exclusively present in females. Results: Molecular bird sexing confirmed 33 females and 19 males with 100% accuracy. We used sequencing followed by alignment on one protected bird species (Probosciger aterrimus). Conclusion: Sex identification may be accomplished noninvasively in birds, because males only have Z sex chromosomes, whereas females have both Z and W chromosomes. Consequently, the presence of a W-unique DNA sequence identifies an individual as female. Sexing of birds is vital for scientific research, and to increase the success rate of conservation breeding programs.

Anaplasma platys is a tick-borne, Gram-negative bacterium that causes anaplasmosis, a companion vector-borne disease impacting dogs. Information on this disease remains limited in Indonesia. Its symptoms are not specific, so molecular analysis is required for a rapid and accurate diagnosis. GroEL is an essential gene commonly used for classification and species identification of many groups of bacteria, including Anaplasma spp. In this study, a molecular diagnosis of anaplasmosis based on the groEL gene sequence was conducted using PCR. In addition, the genetic diversity of Anaplasma platys in infected dogs was determined. Blood samples were collected from 51 dogs suspected of anaplasmosis from Prof. Dr. Soeparwi Animal Hospital, animal clinics, and pet shops in the Yogyakarta area, Indonesia, based on anamnesis, histories of tick infestations, and clinical symptom examinations. DNA extraction and PCR targeting the groEL gene were performed, followed by sequencing. Phylogenetic tree analysis and construction were carried out using the BLAST and MEGA programs. Positive PCR sample results (amplicon length of 624 bp) were found in 6 of 51 dogs. Samples A1 (KHJ/C2), A2 (KHJ/A2), A3 (KSK/L), A4 (KHJ/L), and A5 (KNP/M2) had close ties to Anaplasma platys (AF478129.1) from GenBank. Phylogenetic analysis showed a very high homology value (100%) and bootstrap value of 100%. It can be concluded that there was no genetic diversity in the Anaplasma platys found in infected dogs in the Yogyakarta area.

Aim: This study aimed to analyze the genetic variation and phylogenetic reconstruction of Indonesian indigenous catfish using mitochondrial cytochrome oxidase subunit III sequences. Materials and Methods: A total of 19 samples of catfish were collected from seven rivers (Elo [EM], Progo [PM], Kampar [KR], Musi [MP], Mahakam [MS], Kapuas [KS], and Bengawan Solo [BSBJ]) in five different geographical locations in Indonesia. The genome was isolated from the tissue. Mitochondrial DNA cytochrome oxidase subunit III was amplified using polymerase chain reaction (PCR) with CO3F and CO3R primers. The PCR products were sequenced and continued to analyze genetic variation and phylogenetic relationship using MEGA version 7.0 software. Results: Cytochrome c oxidase (COX)-III gene sequencing obtained 784 nucleotides encoding 261 amino acids. Sequenced COX-III gene fragments were aligned along with other catfish from Genbank using ClustalW program and genetic diversity among species was analyzed using the MEGA Version 7.0 software. Among all samples, there were substitution mutations at 78 nucleotide sites, and there were 14 variations in amino acids. Catfish from PM, KR, MP, and KS had the same amino acids as Hemibagrus nemurus (KJ573466.1), while EM catfish had eight different amino acids and catfish BSBJhad 12 different amino acids. Conclusion: Indonesian catfish divided into four clades. BBSJ Catfish were grouped with Pangasianodon gigas, EM catfish were grouped with Mystus rhegma, and KS catfish were grouped with Hemibagrus spilopterus, while catfish MS, KR, PM, and MP were grouped with H. nemurus.

Aim: This study aimed to assess the changes in neuronal density in CA1 and CA3 regions in the hippocampus of young adulthood and middle age rat model after feeding by Ocimum sanctum ethanolic extract. Materials and Methods: In this research, 30 male Wistar rats consist of young to middle-aged rats were divided into three groups (3, 6, and 9 months old) and treated with a different dosage of O. sanctum ethanolic extract (0, 50, and 100 mg/kg b.w.) for 45 days. Furthermore, cresyl violet staining was performed to analyze hippocampus formation mainly in CA1 and CA3 area. The concentrations of acetylcholine (Ach) in brain tissues were analyzed by enzyme-linked immunosorbent assay. Results: In our in vivo models using rat model, we found that the administration of O. sanctum ethanolic extract with a dosage of 100 mg/kg b.w. for 45 days induced the density of pyramidal cells significantly in CA1 and CA3 of the hippocampus. These results were supported by an increase of Ach concentrations on the brain tissue. Conclusion: The administration of O. sanctum ethanolic extract may promote the density of the pyramidal cells in the CA1 and CA3 mediated by the up-regulated concentration of Ach.

Aim: This research was conducted to identify the expression of choline acetyltransferase (ChAT) in human cerebral microvascular endothelial cells (HCMECs) and to clarify the capability of Ocimum sanctum Linn. ethanolic extract to stimulate the presence of ChAT in the aging HCMECs. Materials and Methods: In this study, we perform an in vitro analysis some in the presence of an ethanolic extract of O. sanctum Linn. as a stimulator for the ChAT expression. HCMECs are divided become two groups, the first is in low passage cells as a model of young aged and the second is in a high passage as a model of aging. Furthermore to analysis the expression of ChAT without and with extract treatments, immunocytochemistry and flow cytometry analysis were performed. In addition, ChAT sandwich enzyme-linked immunosorbent assay is developed to detect the increasing activity of the ChAT under normal, and aging HCMECs on the condition treated and untreated cells. Results: In our in vitro models using HCMECs, we found that ChAT is expressed throughout intracytoplasmic areas. On the status of aging, the ethanolic extract from O. sanctum Linn. is capable to stimulate and restore the expression of ChAT. The increasing of ChAT expression is in line with the increasing activity of this enzyme on the aging treated HCMECs. Conclusions: Our observation indicates that HCMECs is one of the noncholinergic cells which is produced ChAT. The administrated of O. sanctum Linn. ethanolic extract may stimulate and restore the expression of ChAT on the deteriorating cells of HCMECs, thus its may give nerve protection and help the production of acetylcholine.

Aim: Various studies have shown that secreted factors alone in culture medium without stem cell are capable of repairing tissues by itself in various conditions involving damaged tissue/organ. Therefore, this study was aimed to investigate the role of human umbilical cord mesenchymal stem cell-derived conditioned medium (CM) on the recovery of pancreatic β-cells in Wistar rats (Rattus norvegicus) with type 1 diabetes mellitus. Materials and Methods: The 0.05 ml CM induction was applied to the diabetic group of rats in weeks 1, 2, 3, and 4. 1 week after each CM induction, insulin concentration was analyzed using ELISA. The pancreas was divided into 3 regions, processed by paraffin method, stained with hematoxylin-eosin, and immunohistochemical method for insulin. Results: This study indicated the decrease in the total number of islets and insulin concentration after the injection of single dose of alloxan. The exocrine acini were also damaged. Microscopic observation detected the presence of small islets in the diabetic group 1 week after the first 0.05 ml CM induction. The number and size of the islets increased in line with the CM doses and time of inductions. Immunohistochemically, the presence of low intensity of insulin-positive cells could be recognized at the splenic and duodenal regions of the pancreas, but not gastric region, 1 week after the first and second 0.05 ml CM induction. The intensity of staining and the number of insulin-positive cells increased dramatically in 1 week after the third and fourth 0.05 ml of CM induction in all regions of the pancreas. The data of insulin blood concentration showed clear differences between the second and the fourth induction of 0.05 ml CM induction. Conclusions: This study showed very strong evidence on the role of human umbilical cord mesenchymal stem cell-derived CM in recovering the pancreatic β-cells damage in Wistar rats (R. norvegicus) with type 1 diabetes mellitus, structurally and functionally.

The purpose of this research was to identify avian influenza (AI) virus using serological and molecular methods on poultry which suspected as AI infected in Aceh province. This study used 37 samples of tracheal and cloacal swabs and organs from various species of poultry that were collected from several districts/cities in Aceh. Samples were collected and put into transport media and stored at 4° C before sending to the laboratory. Samples were inoculated in specific pathogen-free of embryonated chicken egg with the age of 9-11 days for further serological and molecular examination. From 37 samples which infected to embryonated chicken egg then followed by hemagglutinin agglutination test/hemagglutinin inhibition revealed that 7 samples were positively infected with AI virus. The amplification result of specific matrix gene primer was followed by electrophoresis on 2% agarose gel which were obtained in the form of a deoxyribonucleic acid (DNA) band at 276 bp for matrix gene and 1.725 bp for H5 gene for all isolates test. In conclusion, the virus which caused the death of various types of poultry in Aceh province is avian influenza A virus subtype H5. Aim: Avian encephalomyelitis (AE) is a viral disease which can infect various types of poultry, especially chicken. In Indonesia, the incidence of AE infection in chicken has been reported since 2009, the AE incidence tends to increase from year to year. The objective of this study was to analyze viral protein 2 (VP-2) encoding gene of AE virus (AEV) from various species of birds in field specimen by reverse transcription polymerase chain reaction (RT-PCR) amplification using specific nucleotides primer for confirmation of AE diagnosis. Materials and Methods: A total of 13 AEV samples are isolated from various species of poultry which are serologically diagnosed infected by AEV from some areas in central Java, Indonesia. Research stage consists of virus samples collection from field specimens, extraction of AEV RNA, amplification of VP-2 protein encoding gene by RT-PCR, separation of RT-PCR product by agarose gel electrophoresis, DNA sequencing and data analysis. Results: Amplification products of the VP-2 encoding gene of AEV by RT-PCR methods of various types of poultry from field specimens showed a positive results on sample code 499/4/12 which generated DNA fragment in the size of 619 bp. Sensitivity test of RT-PCR amplification showed that the minimum concentration of RNA template is 127.75 ng/μl. The multiple alignments of DNA sequencing product indicated that positive sample with code 499/4/12 has 92% nucleotide homology compared with AEV with accession number AV1775/07 and 85% nucleotide homology with accession number ZCHP2/0912695 from Genbank database. Analysis of VP-2 gene sequence showed that it found 46 nucleotides difference between isolate 499/4/12 compared with accession number AV1775/07 and 93 nucleotides different with accession number ZCHP2/0912695. Conclusions: Analyses of the VP-2 encoding gene of AEV with RT-PCR method from 13 samples from field specimen generated the DNA fragment in the size of 619 bp from one sample with sample code 499/4/12. The sensitivity rate of RT-PCR is to amplify the VP-2 gene of AEV until 127.75 ng/μl of RNA template. Compared to Genbank databases, isolate 499/4/12 has 85% and 92% nucleotide homology.

Penelitian ini dirancang untuk mendeteksi keberadaan virus AI pada beberapa titik kritis, yaitu unggas, kandang penampungan sementara, tempat pemotongan dan penjualan karkas, dan mengidentifikasi subtipe virus AI pada beberapa pasar tradisional di Kabupaten Aceh Besar dan Kota Banda Aceh, Propinsi Aceh. Sampel swab diambil secara acak (random) dari empat pasar di Kabupaten Aceh Besar dan Kota Banda Aceh, Propinsi Aceh, yaitu pasar Lambaro, Ketapang, Ulekareng dan Peunayong, dan dipoolkan. Satu pool sampel berisi 1 sampai 5 swab yang dikelompokkan berdasarkan pedagang, jenis unggas, dan titik kritis cemaran AI. Jumlah total sampel yang diambil adalah 314 swab, yang terdiri dari swab trakea unggas hidup, kandang penampungan, meja tempat pemotongan, dan karkas, kemudian dikelompokkan dalam 121 pool. Isolasi virus AI kedalam telur ayam bertunas (TAB), pemeriksaan secara serologis (uji HA/HI), dan identifikasi molekuler (metode RT-PCR) di lakukan di bagian Virologi Laboratorium Veteriner, dinas Kesehatan Hewan dan Peternakan Provinsi Aceh, dan Laboratorium Riset Terpadu Fakultas Kedokteran Hewan Universitas Syiah Kuala Banda Aceh. Analisis hasil pemeriksaan serologis dan identifikasi molekuler virus AI dilakukan dengan metode deskriptif. Hasil isolasi virus pada TAB menunjukkan bahwa embrio mengalami kematian pada 1 hari setelah di infeksikan dengan material virus dari sampel swab yang berasal dari pasar Lambaro Kabupaten Aceh Besar, dan titer antibodi terhadap virus AI hanya terdeteksi pada itik dan ayam pedaging yang berkisar antara 24 sampai dengan 27. Dari hasil serologis tersebut dilanjutkan dengan pemeriksaan RT-PCR. Berdasarkan hasil uji RT-PCR menggunakan primer spesifik terhadap gen M, H5, dan N1 diperoleh virus AI yang bersirkulasi pada unggas di pasar Lambaro tergolong subtipe H5N1. Hasil penelitian ini membuktikan bahwa diantara beberapa titik kritis cemaran virus AI di pasar tradisional, unggas hidup (itik dan ayam pedaging) yang berada dalam kandang penampungan sementara merupakan potensial penyebaran virus AI ke lingkungannya.

Hepatitis B virus (HBV) can be classified into 8 genotypes, genotype A to H. Genotype of HBV is important for clinical and etiological investigations. Research for HBV genotyping, HBV transmission study using nested PCR and HBV genotyping based on RFLP using restriction enzymes have been reported. However, both of those methods have been not applied for HBV genotyping study among hepatitis B patients in endemic area, like Indonesia yet. Molecular genotyping of HBV will describe epidemiology, pathogenesis and clinical implication of HBV. Combination of nested PCR and RFLP (nested PCR-RFLP) method to determine HBV genotype in Indonesia is still less information. The objectives of study were to develop a system for HBV genotyping by nested PCR combined with RFLP (nested PCR-RFLP) method based on nucleotide sequence of surface protein encodinggene (S gene) in HBV genome and to confirm HBV genotypes which predominantly found among hepatitis B patients in Daerah Istimewa Yogyakarta Province and surrounding area. Total of 149 sera from chronic hepatitis B patients from Daerah Istimewa Yogyakarta and surrounding areas were collected for in this work. Viral DNA were extracted from sera of hepatitis B patients and used as template for first round nested PCR amplification using outer primers set. Amplicons of first round PCR were used as template for second round amplification using inner primers set. Then, amplicons of second round nested PCR were restriction digested by Sty I and Bsr I enzymes. For HBV genotyping then the restriction products were analyzed by RFLP based on restriction pattern. Results showed that the first round nested PCR amplification generated DNA fragments of whole S gene in length 1.233 bp, and in second round nested PCR amplification using inner primer set generated DNA fragments 585 bp in length. Genotype analysis for all samples using nested PCR-RFLP methods by restriction digested of Sty I and Bsr I enzymes found only 2 HBV genotypes among hepatitis B patients, namely genotype B and C. Quantificationdata showed that most of hepatitis B patients found infected by HBV genotype B (92,8%), genotype C (3,6%) and unidentified genotype (3,6%). Nested PCR-RFLP methods for HBV genotyping is simple and inexpensive for clinical diagnostic in large scale.

Leptomycin B (LMB) was originally discovered as a potent anti-fungal antibiotic from Streptomyces species. The cellular target of LMB has been identified as the nuclear export receptor CRM-1 or exportin-1, which is involved in nuclear trafficking of cellular RNAs or proteins containing the nuclear export sequence (NES). CRM-1 is the main mediator of nuclear export in many cell types including hepatocyte cell lines. The ability of LMB to inhibit nuclear export has made it a useful tool in the study of the intracellular localization of manyregulatory proteins. In this study, we evaluated the effect of nuclear export inhibitor LMB treatment on the intracellular localization of HBV core protein into the hepatocyte cell lines, Huh-7 and HepG2 cells. We also reported the quantification of the distribution of EGFP-Core fusion protein with redundant core NLS as well as SV-40 NLS into cell compartments. Results shown that in Huh-7 cells treatment of LMB caused retention of EGFP-Core fusion protein into the nucleus, so increased the nuclear localization of EGFP-Core and all variants.In HepG2 cells, although not significantly, treatment of LMB increased a number of nuclear localization in all EGFP-Core constructions, even the nuclear localization in HepG2 cells is not so high as in Huh-7 cells. Bayer 41-4109 is heteroarylpyrimidine (HAP) which has been identified as potent of HBV capsid assemblyinhibitor. The present study was to study effect of Bayer 41-4109 treatment on the intracellular localization ofEGFP-Core fusion proteins into HepG2 cells. Three recombinant plasmids of pEGFP-Core with single, double andtriple NLS of HBV core (EGFP-Core 1C, 2C and 3C ) and two recombinant plasmids with single and triple NLS ofSV-40 (EGFP-Core 1 and 3 SV-40) were used in this work. After transient transfected into HepG2 cells and treatedwith Bayer 41-4109, the intracellular localization of expressed fusion proteins from all plasmid constructions weredetermined and quantified under confocal laser microscope. Results shown that Bayer 41-4109 treatment in HepG2cells inhibited the nuclear localization of EGFP-Core with single of triple HBV core NLS. As well as the constructionsof expressed fusion protein with single and triple SV-40 NLS (EGFP-Core 1 and 3 SV-40 NLS) showeddecreasing the nuclear localization after treated with Bayer 41-4109, even not as strong as EGFP-Core 1C and 3CNLS. Bayer 41-4109 has been identified as a potent inhibitors of HBV replication which has multiple effects on HBVcapsid assembly. Staurosporine was discovered to have biological activities ranging from anti-fungal toanti-hypertensive. The interest in these activities resulted in a large investigative effort in chemistry and biology andthe discovery of the potential for anti-cancer treatment. The main biological activity of Staurosporine is the inhibitionof protein kinases through the prevention of ATP binding to the kinase. In the present study, we have studied theintracellular localization of EGFP-Core fusion protein with triple HBV core and SV-40 nuclear localization signal atits carboxyl terminal in presence and absence of Staurosporine. We also to study the effect of Staurosporine treatmenton the intracellular localization of EGFP-Core fusion protein in the hepatocyte cells line of HepG2 cell. Resultsshowed that effect of Staurosporine is prevent the nuclear localization of EGFP-Core fusion protein into nucleusthrough an inhibition of the phosphorylation of core protein. Stauroporine also prevents cell division so that passivetrapping of core protein is inhibited.

Microneme protein (MIC) is one of proteins that belongs to excretory-secretory antigens (ESAs) of Toxoplasma gondii. Microneme 3 protein (MIC-3) is the protein that plays an important role in the invasion proccess during cell infection as a mediator attachment parasite to the host cell. The aim of this research is to clone mic3 (gene encoding for MIC-3) of T. gondii from local isolate using recombinant DNA technology by cloning mic3 in an expression vector. Deoxyribonucleic acid (DNA) from T. gondii tachyzoites was amplified by PuRe Taq RTG-PCR Beads using mic3 specific primers. Amplified DNA was double digested using EcoRV and HindIII restriction endonucleases and then purified using EZ-10 spin coloumn purification kit. The mic3 DNA was ligated into pET-32a(+) expression vector and transformated into Escherichia coli BL21. The results showed that recombinant mic3 gene 4.2 kDa has been successfully performed by cloning gene encoding for MIC-3 protein of T. gondii local isolate into pET-32a(+) and transformed to E. coli BL21.

world. We have evaluated the combination of one-step RT-PCR and multiplex nested PCR assays for detectingdengue viruses from clinical samples. Twelve patients were screened for the dengue virus, using a pair of primersthat conserve for several Flavivirus. The results showed that in 12 suspect patients, 100% were positive for Flavivirusand there are some genotypic variation among them, that indicated by several RT-PCR products higher than 511 bp,the expected product for RT-PCR. Further assay was performed to clarify the presence and serotypes of dengue virususing multiplex nested PCR. Serotyping results indicated that 83,3% of samples can be confirmed for dengue virus.Among the dengue virus positive 16,7 % are dengue-2, 16.7 % are dengue-3, and the most common 50% are dengue-4,whereas dengue-1 were not found among the patients. The combination of RT-PCR and multiplex nested PCR assaycan be used for rapid analysis dengue samples in early phase which is potentially useful for clinical, epidemiologyand also evolutionary studies.Key words: Flavivirus, dengue virus, serotype, RT-PCR, multiplex nested PCR

The application of fluorescent proteins as expression markers and protein fusion partners has provedimmensely valuable for resolving the organization of biological events in living cells. EGFP and DsRed2 arecommonly fluorescent marker protein which is used for biotechnology and cell biology research. The presentstudy was designed to identify the expression vector that suitable to ligate with DNA encoding HBV coreprotein for intracellular localization study in hepatocyte cell, which were expressed as fusion proteins. We alsocompared and quantified the expressed fluorescent protein which predominantly localized in the cellcompartment. The results indicated that DsRed2 shown as less than ideal for intracellular localization study ofthan EGFP, because of its tetrameric structure of the fluorescent protein and when fused to a protein of interest,the fusion protein often forms aggregates in the living cells. In contrast, EGFP fluorescent protein shown a muchhigher proportion of cytoplasmic localization, thus being more suitable for analysis of intracellular localizationthan DsRed2 fluorescent protein. EGFP fluorescent protein is also capable to produce a strong green fluorescencewhen excited by blue light, without any exogenously added substrate or cofactor, events inside living cell canthus be visualized in a non-invasive way. Isoform importin α molecules play a central role in the classical nuclear import pathway, that occurs throughthe nuclear pore complex (NPC) and typically requires a specific nuclear localization signal (NLS). In this study,it was investigated the role of isoforms importin α in the nuclear import of wild type recombinant hepatitis B viruscore protein (WT rHBc), phosphorylated recombinant HBV core (rHBc) and recombinant HBV core without NLSby co-immunoprecipitation. Four recombinant full-length isoforms importin α as 6x histidin-tagged fusion proteinwere expressed and analysed from expression plasmid vectors Rch1, pHM 1969, pHM 1967 and pHM 1965. Theresults indicated that importin α-1, importin α-3, importin α-4 and importin α-5 can be expressed and isolatedfrom E. coli transformed recombinant DNA plasmid as protein in size around 58-60 kDa. By the nuclear transportstudy shown that isoforms importin α are involved in the nuclear import of WT rHBc, phosphorylated rHBc andrHBc without NLS. Influenza A viruses which belong to the Family of Orthomyxoviridae are a group of viruses withsegmented ssRNA genome. The viruses can be subgroupped into many subtypes on the basis of theirsurface glycoproteins, hemagglutinin (HA) and neuraminidase (NA) proteins. Among the HA subtypes, H5and H7 have been found to be the most pathogenic. Conventional diagnosis of the viruses is usuallyperformed by isolation of the viruses in embryonated eggs, and hemagglutination (HA) and hemagglutinationinhibition test. Although those methods are sensitive and accurate, they are time consuming and requirelaboratory facilities with high biosafety level. Commercial methods such as emzyme-linked immonosorbentassay (ELISA) and immunoflurescense assay also provide a rapid result but less sensitive and specificthan conventional methods. Molecular diagnosis by amplification of M and H5 genes using one strepsimplex reverse transcriptase-polymerase chain eraction (RT-PCR) provices a rapid and accurate diagnosisfor the viruses. A study was therefore conducted to evaluate the accurate and rapidity of such the moleculartests for diagnosis of avian influenza A virus, subtype H5. As many as 10 sample of the virus isolateswhich were available at the Animal Desease investigation Center in Wates, Yogyakarta, were uses in thisstudy. The virus isolates were firstly propagated in specific antigen negative (SAN) chicken embryos andtested by HA/HI test. The viruses were then subjected for the RT-PCR test with varying annealingtemperatures of 500C and 520C. The result showed that all 10 isolates were type A influenza virus and 8out of 10 were influenza A subtype H5 influenza virus. RT-PCR used in this study appears to be moresensitive, rapid and accurate as compared to those by serological and isolation of the virus in embryonatedeggs.

Tujuan dari penelitian ini adalah mengidentifikasi keberadaan gen M, H5, dan N1 virus avian influenza (AI) melalui metode single step multiplex reverse transcriptase-polymerase chain reaction (RT-PCR), sebagai acuan untuk peneguhan diagnosis secara molekuler virus AI di Provinsi Aceh. Penelitian ini menggunakan 11 isolat virus AI asal Provinsi Aceh yang diperoleh dari Laboratorium Balai Penyidikan dan Pengujian Veteriner (BPPV) Regional I Medan di Sumatera Utara dari tahun 2006-2008. Penelitian dilakukan di Laboratorium Virologi BPPV Regional I di Medan, Sumatera Utara. Amplifikasi terhadap gen matriks (M) virus AI menggunakan metode simplex RT-PCR. Hasil simplex RT-PCR terhadap gen M diperoleh 10 isolat yang menunjukkan pita deoxyribonucleic acid (DNA) pada 276 bp dan satu isolat yang tidak muncul, kemudian dilanjutkan dengan metode single step multiplex RT-PCR menggunakan pasangan primer gen penyandi protein N1, H5, dan M. Produk PCR 131 bp (N1), 189 bp (H5), dan 276 bp (M) muncul sebagai hasil elektroforesis dari semua isolat virus AI. Semua virus AI yang mewabah dari tahun 2006-2008 di Provinsi Aceh termasuk ke dalam virus influenza A subtipe H5N1.

Tetelo or Newcastle Disease (ND) virus is belong to the family Paramyxoviridae, which has a singlestranded RNA (ss RNA) genome and it has viral envelope. The viral envelope consists of two majorproteins, namely Haemagglutinin/Neuraminidase (H/N) and Fusion (F) protein. Molecular diagnosticmethods OneStep RT-PCR is a commonly diagnostic tool that used to diagnose of Tetelo in poultry. In thisstudy the diagnosis of Tetelo is accomplished by amplification of F protein encoding gene of Tetelo virusdirectly from field specimens without inoculation and propagation of Tetelo virus into embryonated chickeneggs. The objective of this study is to conduct rapid diagnosis of Tetelo virus directly from the field specimensbased on the amplification of the F gene by a OneStep RT-PCR method, so that the results of this study canbe used to assist in the identification of Tetelo virus directly from the field specimens. The results showedthat from the 15 samples of virus which isolated from tracheal swabs of clinical poultry showing symptomsof Tetelo virus infection, a total of 12 samples from 15 tested sampels (80%) are positive tested for Tetelovirus infection. They were indicated by the amplification product of DNA fragments in size of 362 bp.OneStep RT-PCR is a method for rapid and effective diagnosis which can be used to diagnose of Tetelovirus directly from field specimens.

Penelitian ini bertujuan melakukan studi filogenetik virus AI tipe A subtipe H5N1 isolat asal Provinsi Aceh yang dapat memberikan informasi secara molekuler tentang kekerabatan antar isolat virus AI tipe A subtipe H5N1. Sebanyak 11 isolat virus AI asal Provinsi Aceh yang dikoleksi oleh Laboratorium Virologi Balai Penyidikan dan Pengujian Veteriner (BPPV) Regional I di Medan, Sumatera Utara selama kurun waktu 2007-2008 digunakan dalam penelitian ini. Metode penelitian meliputi preparasi sampel isolat virus, ekstraksi RNA, amplifikasi gen H, elektroforesis DNA hasil amplifikasi, sekuensing, dan analisis data hasil sekuensing dengan software MEGA 4.0. Hasil sekuensing diketahui bahwa daerah yang teramplifikasi sebesar 191 bp. Hasil allignment dari nukleotida 191 bp ditemukan urutan nukleotida yang menyandi 5 asam amino yang berbeda pada posisi asam amino ke-493, 498, 499, 504, dan 515 antara isolat virus AI subtipe H5N1 asal Provinsi Aceh selama tahun 2007-2008 dengan isolat dari pembanding dari gene bank. Kontruksi pohon filogenetik menunjukkan bahwa virus AI subtipe H5N1 asal Provinsi Aceh tahun 2007-2008 mengelompok secara terpisah dari isolat Indonesia yang lain, tetapi masih dalam klaster virus AI subtipe H5N1 Indonesia.

Virus avian influenza (AI) merupakan virus dengan materi genetik RNA single-stranded sense negatif, beramplop yang termasuk dalam famili Orthomyxoviridae. Onestep simplex reverse transcriptase-polymerase chain reaction (RT-PCR) merupakan salah satu metode diagnosis yang dapat diandalkan untuk mendeteksi virus AI. Tujuan dari penelitian ini adalah untuk menerapkan metode diagnosis cepat virus AI pada spesimen lapangan secara langsung dari pasar unggas berdasarkan amplifikasi RT-PCR gen M dan H5 dengan metode yang berbasis onestep simplex RT-PCR tanpa melalui proses inokulasi dan propagasi virus AI dalam telur ayam berembrio (TAB). Sebanyak 35 sampel spesimen lapangan dari swab trakea unggas yang berasal dari pasar unggas di Terban, Kotamadya Yogyakarta digunakan dalam penelitian ini. Amplifikasi DNA secara onestep simplex RT-PCR pada gen matriks (M) dilakukan terhadap seluruh sampel. Pada sampel-sampel yang menunjukkan hasil positif pada amplifikasi gen M kemudian dilakukan amplifikasi RT-PCR secara lebih lanjut untuk gen H5 virus AI. Produk hasil amplifikasi RT-PCR gen M dan H5 divisualisasikan menggunakan elektroforesis gel agarose konsentrasi 1% dengan pewarnaan SYBR® Safe DNA Gel Staining. Hasil amplifikasi RT-PCR gen M menunjukkan bahwa dari 35 sampel diperoleh 8 sampel positif terinfeksi virus AI tipe A yang ditunjukkan dengan adanya fragmen DNA sebesar 200 bp, sedangkan hasil amplifikasi gen H5 sebanyak 5 dari 8 sampel-sampel tersebut merupakan virus AI tipe A subtipe H5 yang ditunjukkan dengan adanya fragmen DNA sebesar 545 bp. Diagnosis cepat virus AI tipe A subtipe H5 secara langsung dari spesimen lapangan di pasar unggas dapat dilakukan dengan metode onestep simplex RT-PCR, namun metode diagnosis tersebut tidak dapat mendeteksi keberadaan virus AI dalam sampel yang virusnya terlalu sedikit.

The cellular target of LMB has been identified as the nuclear export receptor CRM-1 or exportin-1, which is involved in nuclear trafficking of cellular RNAs or proteins containing the nuclear export sequence (NES). CRM-1 is the main mediator of nuclear export in many cell types including hepatocyte cell lines. The ability of LMB to inhibit nuclear export has made it a useful tool in the study of the intracellular localization of many regulatory proteins. In this study, we evaluated the effect of nuclear export inhibitor LMB treatment on the intracellular localization of HBV core protein into the hepatocyte cell lines, Huh-7 and HepG2 cells. We also reported the quantification of the distribution of EGFP-Core fusion protein with redundant core NLS as well as SV-40 NLS into cell compartments. Results shown that in Huh-7 cells treatment of LMB caused retention of EGFP-Core fusion protein into the nucleus, so increased the nuclear localization of EGFP-Core and all variants. In HepG2 cells, although not significantly, treatment of LMB increased a number of nuclear localization in all EGFP-Core constructions, even the nuclear localization in HepG2 cells is not so high as in Huh-7 cells. Avian influenza (AI) virus is a segmented single stranded (ss) RNA virus with negative polarity andbelong to the Orthomyxoviridae family. Diagnose of AI virus can be performed using conventional methodsbut it has low sensitivity and specificity. The objective of the research was to apply rapid, precise, andaccurate diagnostic method for AI virus and also to determine its type and subtype based on the SingleStep Multiplex Reverse Transcriptase-Polymerase Chain Reaction targeting M, H5, and N1 genes. In thismethod M, H5 and NI genes were simultaneously amplified in one PCR tube. The steps of this researchconsist of collecting viral RNAs from 10 different AI samples originated from Maros Disease InvestigationCenter during 2007. DNA Amplification was conducted by Simplex RT-PCR using M primer set. Then, bysingle step multiplex RT-PCR were conducted simultaneously using M, H5 and N1 primers set. The RTPCRproducts were then separated on 1.5% agarose gel, stained by ethidum bromide and visualized underUV transilluminator. Results showed that 8 of 10 RNA virus samples could be amplified by Simplex RTPCRfor M gene which generating a DNA fragment of 276 bp. Amplification using multiplex RT-PCRmethod showed two of 10 samples were AI positive using multiplex RT-PCR, three DNA fragments weregenerated consisting of 276 bp for M gene, 189 bp for H5 gene, and 131 bp for N1. In this study, rapid andeffective diagnosis method for AI virus can be conducted by using simultaneous Single Step Multiplex RTPCR.By this technique type and subtype of AI virus, can also be determined, especially H5N1.

Hepatitis B virus (HBV) can be classified into 8 genotypes, genotype A to H. Genotype of HBV is important for clinical and etiological investigations. Research for HBV genotyping, HBV transmission study using nested PCR and HBV genotyping based on RFLP using restriction enzymes have been reported. However, both of those methods have been not applied for HBV genotyping study among hepatitis B patients in endemic area, like Indonesia yet. Molecular genotyping of HBV will describe epidemiology, pathogenesis and clinical implication of HBV. Combination of nested PCR and RFLP (nested PCR-RFLP) method to determine HBV genotype in Indonesia is still less information. The objectives of study were to develop a system for HBV genotyping by nested PCR combined with RFLP (nested PCR-RFLP) method based on nucleotide sequence of surface protein encodinggene (S gene) in HBV genome and to confirm HBV genotypes which predominantly found among hepatitis B patients in Daerah Istimewa Yogyakarta Province and surrounding area. Total of 149 sera from chronic hepatitis B patients from Daerah Istimewa Yogyakarta and surrounding areas were collected for in this work. Viral DNA were extracted from sera of hepatitis B patients and used as template for first round nested PCR amplification using outer primers set. Amplicons of first round PCR were used as template for second round amplification using inner primers set. Then, amplicons of second round nested PCR were restriction digested by Sty I and Bsr I enzymes. For HBV genotyping then the restriction products were analyzed by RFLP based on restriction pattern. Results showed that the first round nested PCR amplification generated DNA fragments of whole S gene in length 1.233 bp, and in second round nested PCR amplification using inner primer set generated DNA fragments 585 bp in length. Genotype analysis for all samples using nested PCR-RFLP methods by restriction digested of Sty I and Bsr I enzymes found only 2 HBV genotypes among hepatitis B patients, namely genotype B and C. Quantificationdata showed that most of hepatitis B patients found infected by HBV genotype B (92,8%), genotype C (3,6%) and unidentified genotype (3,6%). Nested PCR-RFLP methods for HBV genotyping is simple and inexpensive for clinical diagnostic in large scale.

Bayer 41-4109 is heteroarylpyrimidine (HAP) which has been identified as potent of HBV capsid assembly inhibitor. The present study was to study effect of Bayer 41-4109 treatment on the intracellular localization of EGFP-Core fusion proteins into HepG2 cells. Three recombinant plasmids of pEGFP-Core with single, double and triple NLS of HBV core (EGFP-Core 1C, 2C and 3C ) and two recombinant plasmids with single and triple NLS of SV-40 (EGFP-Core 1 and 3 SV-40) were used in this work. After transient transfected into HepG2 cells and treated with Bayer 41-4109, the intracellular localization of expressed fusion proteins from all plasmid constructions were determined and quantified under confocal laser microscope. Results shown that Bayer 41-4109 treatment in HepG2 cells inhibited the nuclear localization of EGFP-Core with single of triple HBV core NLS. As well as the constructions of expressed fusion protein with single and triple SV-40 NLS (EGFP-Core 1 and 3 SV-40 NLS) showed decreasing the nuclear localization after treated with Bayer 41-4109, even not as strong as EGFP-Core 1C and 3C NLS. Bayer 41-4109 has been identified as a potent inhibitors of HBV replication which has multiple effects on HBV capsid assembly. It may inhibit virus replication by inducing assembly inappropriately and by misdirecting assembly decreasing the stability of normal capsids. Staurosporine was discovered to have biological activities ranging from anti-fungal to anti-hypertensive. The interest in these activities resulted in a large investigative effort in chemistry and biology and the discovery of the potential for anti-cancer treatment. The main biological activity of Staurosporine is the inhibition of protein kinases through the prevention of ATP binding to the kinase. In the present study, we have studied the intracellular localization of EGFP-Core fusion protein with triple HBV core and SV-40 nuclear localization signal at its carboxyl terminal in presence and absence of Staurosporine. We also to study the effect of Staurosporine treatment on the intracellular localization of EGFP-Core fusion protein in the hepatocyte cells line of HepG2 cell. Results showed that effect of Staurosporine is prevent the nuclear localization of EGFP-Core fusion protein into nucleus through an inhibition of the phosphorylation of core protein. Stauroporine also prevents cell division so that passive trapping of core protein is inhibited.

world. We have evaluated the combination of one-step RT-PCR and multiplex nested PCR assays for detecting dengue viruses from clinical samples. Twelve patients were screened for the dengue virus, using a pair of primers that conserve for several Flavivirus. The results showed that in 12 suspect patients, 100% were positive for Flavivirus and there are some genotypic variation among them, that indicated by several RT-PCR products higher than 511 bp, the expected product for RT-PCR. Further assay was performed to clarify the presence and serotypes of dengue virus using multiplex nested PCR. Serotyping results indicated that 83,3% of samples can be confirmed for dengue virus. Among the dengue virus positive 16,7 % are dengue-2, 16.7 % are dengue-3, and the most common 50% are dengue-4, whereas dengue-1 were not found among the patients. The combination of RT-PCR and multiplex nested PCR assay can be used for rapid analysis dengue samples in early phase which is potentially useful for clinical, epidemiology and also evolutionary studies. The application of fluorescent proteins as expression markers and protein fusion partners has proved immensely valuable for resolving the organization of biological events in living cells. EGFP and DsRed2 are commonly fluorescent marker protein which is used for biotechnology and cell biology research. The present study was designed to identify the expression vector that suitable to ligate with DNA encoding HBV core protein for intracellular localization study in hepatocyte cell, which were expressed as fusion proteins. We also compared and quantified the expressed fluorescent protein which predominantly localized in the cell compartment. The results indicated that DsRed2 shown as less than ideal for intracellular localization study of than EGFP, because of its tetrameric structure of the fluorescent protein and when fused to a protein of interest, the fusion protein often forms aggregates in the living cells. In contrast, EGFP fluorescent protein shown a much higher proportion of cytoplasmic localization, thus being more suitable for analysis of intracellular localization than DsRed2 fluorescent protein. EGFP fluorescent protein is also capable to produce a strong green fluorescence when excited by blue light, without any exogenously added substrate or cofactor, events inside living cell can thus be visualized in a non-invasive way. Based on our present quantitative data and some reasons above shown that EGFP is more suitable than DsRed2 as a fluorescent marker protein for intracellular localization study into HuH-7 cell. Isoform importin α molecules play a central role in the classical nuclear import pathway, that occurs through the nuclear pore complex (NPC) and typically requires a specific nuclear localization signal (NLS). In this study, it was investigated the role of isoforms importin α in the nuclear import of wild type recombinant hepatitis B virus core protein (WT rHBc), phosphorylated recombinant HBV core (rHBc) and recombinant HBV core without NLS by co-immunoprecipitation. Four recombinant full-length isoforms importin α as 6x histidin-tagged fusion protein were expressed and analysed from expression plasmid vectors Rch1, pHM 1969, pHM 1967 and pHM 1965. The results indicated that importin α-1, importin α-3, importin α-4 and importin α-5 can be expressed and isolated from E. coli transformed recombinant DNA plasmid as protein in size around 58-60 kDa. By the nuclear transport study shown that isoforms importin α are involved in the nuclear import of WT rHBc, phosphorylated rHBc and rHBc without NLS. It also indicated that they have an important role for nuclear transport of from cytoplasm into the nucleus.
