Staphylococcus aureus is often the cause of various diseases and food poisoning in animals and humans. Avocado seeds (Persea americana, Mill) contain important compounds that have antimicrobial activity. This study aimed to observe the hematological dynamics of mice (Mus musculus domesticus) infected with S. aureus and induced by P. americana extract. BALB/c male mice aged 12 weeks were divided into 6 groups of 5 in each group, named K-1, K-2, K-3, K-4, K-5, and K-6. All groups were infected with S. aureus 1x108 CFU cell/mL intraperitoneally (IP) on day 0. After the blood sample was taken, then K-1 was given a single dose of tetracyclin 130 mg/kg BW, K-2 was given 130 mg/kb BW oferythromycin, K-3 was given aquabidest (placebo), K-4 was given P. americana extract 300 mg/kg BW, K-5 at a dose 600 mg/kg BW and K-6 at a dose 1200 mg/kg BW. The treatment was given for 7 consecutive days, then a second blood sample was taken and analyzed with the VetScan-HM5 analyzer and interpreted. It was concluded that S. aureus infection caused leukopenia and was potentially followed by lymphopenia, neutrophilia, and monocytosis. The occurrence of microcytosis can lead to anemia, which can be categorized as microcytic hypochromic or normocytic normochromic. Administration of P. americana extract at a dose 300 mg/kg BW (K-4) for 7 days after S. aureus infection was found to be effective in improving hematological values in mice back to normal.

Background: Hearing screening is a method for early identification of hearing loss. Hearing screening is widely used in hospitals as part of physical examination. However, some of them are difficult to operate outside the hospitals regarding portability and user-friendliness. Therefore, we developed a simple yet easy-to-use portable electronic whisper test (EWT) for field deployment, particularly on many subjects. The EWT validity is a hearing screening method compared to pure-tone audiometry., Methods: This study was a cross-sectional comparative with a prospective approach. The subjects were outpatients at the Audiology Clinic of Dr. Soetomo Academic Medical Center, Surabaya, Indonesia, who met the inclusion and exclusion criteria. Statistical analysis made sensitivity and specificity tables using a 2 × 2 table, positive and negative predictive values., Results: Samples of 50 participants (100 ears) obtained a sensitivity value of 83%, specificity of 78%, positive predictive value of 45%, and negative predictive value of 95%., Conclusions: The EWT is valid for use as a hearing screening method., Competing Interests: There are no conflicts of interest., (Copyright: © 2023 International Journal of Critical Illness and Injury Science.)

Purpose: This study aims to evaluate the efficiency of infrared LEDs with a magnetic solenoid field in lowering the quantity of gram-positive Staphylococcus aureus and gram-negative Escherichia coli bacteria, as well as the best exposure period and energy dose for inactivating these bacteria., Method: Research has been performed on a photodynamic therapy technique called photodynamic inactivation (PDI), which combines infrared LED light with a wavelength range of 951-952 nm and a solenoid magnetic field with a strength of 0-6 mT. The two, taken together, can potentially harm the target structure biologically. Infrared LED light and an AC-generated solenoid magnetic field are both applied to bacteria to measure the reduction in viability. Three different treatments infrared LED, solenoid magnetic field, and an amalgam of infrared LED and solenoid magnetic field, were used in this study. A factorial statistical ANOVA analysis was utilized in this investigation., Results: The maximum bacterial production was produced by irradiating a surface for 60 min at a dosage of 0.593 J/cm 2 , according to the data. The combined use of infrared LEDs and a magnetic field solenoid resulted in the highest percentage of fatalities for Staphylococcus aureus , which was 94.43 s. The highest percentage of inactivation for Escherichia coli occurred in the combination treatment of infrared LEDs and a magnetic field solenoid, namely, 72.47 ± 5.06%. In contrast, S. aureus occurred in the combined treatment of infrared LEDs and a magnetic field solenoid, 94.43 ± 6.63 percent., Conclusion: Staphylococcus aureus and Escherichia coli germs are inactivated using infrared illumination and the best solenoid magnetic fields. This is evidenced by the rise in the proportion of bacteria that died in treatment group III, which used a magnetic solenoid field and infrared LEDs to deliver a dosage of 0.593 J/cm 2 over 60 min. According to the research findings, the magnetic field of the solenoid and the infrared LED field significantly impact the gram-positive bacteria S. aureus and the gram-negative bacteria E. coli .

A good knowledge of the genome properties of the populations makes it possible to optimize breeding methods, in particular genomic selection (GS). In oil palm (Elaeis guineensis Jacq), the world's main source of vegetable oil, this would provide insight into the promising GS results obtained so far. The present study considered two complex breeding populations, Deli and La Mé, with 943 individuals and 7324 single-nucleotide polymorphisms (SNPs) from genotyping-by-sequencing. Linkage disequilibrium (LD), haplotype sharing, effective size (N e ), and fixation index (F st ) were investigated. A genetic linkage map spanning 1778.52 cM and with a recombination rate of 2.85 cM/Mbp was constructed. The LD at r 2 =0.3, considered the minimum to get reliable GS results, spanned over 1.05 cM/0.22 Mbp in Deli and 0.9 cM/0.21 Mbp in La Mé. The significant degree of differentiation existing between Deli and La Mé was confirmed by the high F st value (0.53), the pattern of correlation of SNP heterozygosity and allele frequency among populations, and the decrease of persistence of LD and of haplotype sharing among populations with increasing SNP distance. However, the level of resemblance between the two populations over short genomic distances (correlation of r values between populations >0.6 for SNPs separated by <0.5 cM/1 kbp and percentage of common haplotypes >40% for haplotypes <3600 bp/0.20 cM) likely explains the superiority of GS models ignoring the parental origin of marker alleles over models taking this information into account. Purpose: This study aims to examine the effects of red 649 nm 4 J/cm 2 and blue 403 nm 8 J/cm 2 diode laser treatment for post-extraction wounded healing in rats through histopathological and immunohistochemical analysis., Methods: Samples of 54 Wistar rats were divided into six groups: C- control group without treatment; C + wounded group without treatment; TB wound group with Povidone-iodine treatment; TD wounded group with doxycycline treatment; TLB wounded group with 403 nm diode laser treatment; and TLR wounded group with 649 nm diode laser treatment. Mandibular samples were observed for the number of lymphocytes and fibroblasts cells, new blood vessels formation, Interleukin 1β, and Collagen 1α expression level., Results: Based on the histopathological test results, red laser diode treatment significantly increased the number of lymphocyte, fibroblast cells and the formation of new blood vessels. Meanwhile, immunohistochemical tests showed an increase in the expression of the Colagen-1α protein which plays a role in the formation of collagen for new tissues formation after damage, as well as a decrease in Interleukin-1β expression level. Blue laser is also able to show a positive effect on wound healing even though its penetration level into the tissue is lower compared to red laser., Conclusion: The red diode laser 649 nm has been shown to accelerate the process of proliferation in wound healing post molar extraction based on histopathological and immunohistochemical test results., (© 2021. Background : The objective of this study was to test the validity of automated audiometry as a method of hearing examination in patients with multidrug-resistant tuberculosis. Method s : This was a cross-sectional comparative study with a retrospective approach, using patient medical records. Patients with multidrug-resistant tuberculosis (MDR-TB) were recruited based-on medical records that met the inclusion and exclusion criteria at the Pulmonology outpatient unit, then referred to the Otorhinolaryngology outpatient unit of the Dr. Soetomo Academic Medical Center. The subjects' hearing function was measured with two different devices (automated audiometer and conventional audiometer) before being given anti-tuberculosis drug therapy (aminoglycoside injection) as ototoxicity monitoring from July to December 2019 period. Sensitivity and specificity analysis was used to assess the validity of the test. Results : A total of 36 patients (72 ears) were included. The comparison test results using the Mann-Whitney test showed that there were significant differences between automated audiometry and conventional audiometry in both ears. Analysis values were: sensitivity 80-97%, specificity 37-96%, positive predictive value 74-98%, and negative predictive value 59-96%. Background: Periodontitis is a chronic inflammatory disease characterized by progressive damage on the structure of tooth-supporting tissues. The aim of the study is determining the combination photodynamic effect of diode laser 405 nm treatments and the administration of doxycycline 0.1% within 1, 3, 5, and 7 days on a Wistar rat model of periodontitis., Methods: Samples were induced with Porphyromonas gingivalis ATCC33277 to allow periodontitis development and were treated with combination of doxycycline and laser diode, then statistical analysis was carried out (One-Way ANOVA test and the post-hoc Duncan test; Kruskal-Wallis test and Mann-Whitney follow-up test for non-parametric data). Samples were divided into five groups, laser exposure used was 405-nm diode laser with energy density of 8 J/cm 2 . The expression level of histomorphometric was calculated by measuring the number of macrophages, lymphocytes, fibroblasts and the distance between the CEJ-AV., Results: The results showed that the combination treatment of doxycycline and laser exposure yielded immunomodulatory effects. The expression level of fibroblast and the distance between CEJ-AV bone showed that the combination of doxycycline and laser therapy exerted healing effect in rat models of periodontitis on day 5 and 7., Conclusion: The combination of doxycycline 0.1% and diode laser therapy provides a healing effect in rats models of periodontitis.

Several diagnostic tools are available for veterinarians and fish health professionals to evaluate fish health and their abnormalities. However, reference data regarding the character and size of fish blood cells are limited. The purpose of this study was to analyze the morphology and morphometry of normal blood cells in zebrafish ( Danio rerio ), common carp ( Cyprinus carpio carpio ), and tilapia ( Oreochromis niloticus ). To get a representative sample, we took blood from ten 6-mo old healthy fish from each species. Fish were purchased from an ornamental fish market and the local government fish breeding center in Yogyakarta, Indonesia. A total of 100 erythrocytes and a maximum of 30 leukocytes (neutrophils, eosinophils, basophils, lymphocytes and monocytes) were randomly sampled and analyzed qualitatively and quantitatively, including morphometric analysis of both the long axis (LA) and short axis (SA) of these cells. All data obtained were analyzed using ANOVA statistical tests to compare the blood cells in each species with SPSS software. The findings revealed distinct differences in both morphology and morphometry of the blood cells among the species. Basic knowledge obtained from this research will aid in the development of biomarkers and other ancillary diagnostic tools for further hematology research, conservation, and clinical diagnosis in these 3 fish species.

Biofilms are able to cause microorganisms to be 80% more resistant to antibiotics. The extracelullar polymeric substance (EPS) in biofilm functions to protect bacteria, making it difficult for antibiotics to penetrate the biofilm layer. This study aims to determine the effectiveness of photodynamic inactivation with blue diode laser to reduce Staphylococcus aureus biofilm at various ages of biofilms. The light source is a 403 nm blue diode laser with an energy power of about 27.65±0.01 mW. The study was designed with two groups: Group C was the untreated control group with variations in age of biofilms (0; 6; 11; 17; 24; 32; 40 and 48) hours; Group T was a laser treatment group with variations in age of biofilm and energy density (4.23; 8.46; 12.70; 16.93 and 21.16) J/cm 2 . Biofilm reduction measurement method using ELISA test was performed to calculate OD 595 value. The statistical analysis results of variance showed that there was an influence of biofilm age and irradiation energy density of laser on biofilm reduction. Optical density analysis showed the most optimum biofilm reduction happened when biofilm age is perfectly constructed (about 17 hours) and with 91% reduction. The longer biofilm age lived among those biofilms, the greater the reduction. The results of the Scanning Microscope Electron and fluorescent microscope measurement showed destruction site of the EPS biofilm and bacterial cell death. On- and off-frequency compression at the 4000- and 8000-Hz cochlear places were estimated using a new version of the additivity of forward masking (AFM) technique, that measures the effects of combining two non-overlapping forward maskers. Instead of measuring signal thresholds to estimate compression of the signal as in the original AFM technique, the decrease in masker threshold in the combined-masker condition compared to the individual-masker conditions is used to estimate compression of the masker at the signal place. By varying masker frequency it is possible to estimate off-frequency compression. The maskers were 500-Hz-wide bands of noise, and the signal was a brief pure tone. Compression at different levels was estimated using different overall signal levels, or different masker-signal intervals. It was shown that the new AFM technique and the original AFM technique produce consistent results. Considerable compression was observed for maskers well below the signal frequency, suggesting that the assumption of off-frequency linearity used in other techniques may not be valid. Reducing the duration of the first masker from 200 to 20 ms reduced the compression exponent in some cases, suggesting a possible influence of olivocochlear efferent activity.

Auditory compression was estimated at 250 and 4000 Hz by using the additivity of forward masking technique, which measures the effects on signal threshold of combining two temporally nonoverlapping forward maskers. The increase in threshold in the combined-masker condition compared to the individual-masker conditions can be used to estimate compression. The signal was a 250 or 4000 Hz tone burst and the maskers (M1 and M2) were bands of noise. Signal thresholds were measured in the presence of M1 and M2 alone and combined for a range of masker levels. The results were used to derive response functions at each frequency. The procedure was conducted with normal-hearing and hearing-impaired listeners. The results suggest that the response function in normal ears is similar at 250 and 4000 Hz with a mid level compression exponent of about 0.2. However, compression extends over a smaller range of levels at 250 Hz. The results confirm previous estimates of compression using temporal masking curves (TMCs) without assuming a linear off-frequency reference as in the TMC procedure. The impaired ears generally showed less compression. Importantly, some impaired ears showed a linear response at 250 Hz, providing a further indication that low-frequency compression originates in the cochlea.
