Abstract

Human inflammatory breast cancer ( IBC) and canine inflammatory mammary cancer ( CIMC) are the most aggressive forms of mammary cancer. Current research aims to identify new therapeutic targets. Here, we investigated gene expression levels of biomarkers associated with the inflammatory microenvironment. A total of 32 formalin-fixed paraffin-embedded samples of canine mammary carcinoma ( CIMC = 26; non- CIMC = 6) were used and their cDNA subjected to quantitative polymerase chain reaction ( qPCR) to establish gene expression levels for mediators commonly implicated in linking carcinogenesis with inflammation. Gene expression differences between CIMC and non- CIMC types were obtained for cyclooxygenase 2 ( COX-2) ( P = 0.004), synuclein gamma (SNCG) ( P = 0.006), tribbles 1 ( P = 0.025), vascular endothelial growth factor ( VEGF) ( P = 0.017) and CSF1R ( P = 0.045). Among these biomarkers correlations were found, particularly between SNCG and tribbles 1 ( r = 0.512, P = 0.001). The efficient metastasis of CIMC is intimately linked to components in the tumour microenvironment. This study suggests that upregulation and correlation of SNCG and tribbles 1 deserves to be further explored. [ABSTRACT FROM AUTHOR]

Abstract

To test the potential of a multigene DNA vaccine against lentivirus \ud infection, we generated a defective mutant provirus of feline immunodeficiency \ud virus (FIV) with an in-frame deletion in pol (FIV[Delta]RT). In a first \ud experiment, FIV[Delta]RT DNA was administered intramuscularly to 10 animals, \ud half of which also received feline gamma interferon (IFN-γ) DNA. The DNA was \ud administered in four 100-μg doses at 0, 10, and 23 weeks. Immunization with \ud FIV[Delta]RT elicited cytotoxic T-cell (CTL) responses to FIV Gag and Env in the \ud absence of a serological response. After challenge with homologous virus at week \ud 26, all 10 of the control animals became seropositive and viremic but 4 of the \ud 10 vaccinates remained seronegative and virus free. Furthermore, quantitative \ud virus isolation and quantitative PCR analysis of viral DNA in peripheral blood \ud mononuclear cells revealed significantly lower virus loads in the FIV[Delta]RT \ud vaccinates than in the controls. Immunization with FIV[Delta]RT in conjunction \ud with IFN-γ gave the highest proportion of protected cats, with only two of five \ud vaccinates showing evidence of infection following challenge. In a second \ud experiment involving two groups (FIV[Delta]RT plus IFN-γ and IFN-γ alone), the \ud immunization schedule was reduced to 0, 4, and 8 weeks. Once again, CTL \ud responses were seen prior to challenge in the absence of detectable antibodies. \ud Two of five cats receiving the proviral DNA vaccine were protected against \ud infection, with an overall reduction in virus load compared to the five infected \ud controls. These findings demonstrate that DNA vaccination can elicit protection \ud against lentivirus infection in the absence of a serological response and \ud suggest the need to reconsider efficacy criteria for lentivirus vaccines.
