The antioxidant properties of the leaves of the Mediterranean strawberry tree (Arbutus unedo L.) are mainly attributed to the main bioactive compound, the phenolic glycoside arbutin. In this study, the safety profile of strawberry tree aqueous leaf extract (STE) and arbutin at the DNA level was assessed in vitro using porcine PK-15 kidney cells and HepG2 cells derived from human hepatomas. To examine the effects on cell viability and DNA damage, cells were treated for 24 h with STE or arbutin at three concentrations presumed to be non-toxic (400, 200, and 11.4 µg/mL). Assessments were performed using the MTS viability assay, dual acridine orange/ethidium bromide fluorescent staining, and alkaline comet assay. Results showed that the highest concentration (400 µg/mL) of both tested compounds had no significant cytotoxic effects on either PK-15 or HepG2 cells. Apoptosis was the predominant type of cell death and the total amount of DNA damage in treated cells was within acceptable limits. These results on the in vitro cytocompatibility of arbutin and STE with PK-15 and HepG2 cells could serve to make more reliable judgements about safe levels of arbutin in cosmetic products and functional foods, given the increased popularity of the compound in recent years. This study evaluated the effects of strawberry tree water leaf extract (STE) and arbutin in the brain tissue of Lewis rats. STE or arbutin were administered per os to male and female rats at a dose of 200 mg/kg body weight/day for 14 or 28 days. Treatment outcomes were evaluated using biochemical markers (lipid peroxidation and the activities of the antioxidative enzymes catalase and superoxide dismutase). The effects of the tested substances on DNA integrity in brain cells were evaluated using the alkaline comet assay. The results suggest a high biocompatibility of both tested substances with rat brain tissue. No significant harmful disturbances were observed in the oxidative/antioxidative status or impairments of DNA integrity in the rat brain cells. Nearly all post-treatment values were within tolerable limits as compared to the matched control rats. Such encouraging findings support further research using other subtle biomarkers to clarify the safety aspects of arbutin and STE prior to the development of specific nutraceutical products. The influence of wheat starch (6%, 8% and 10%, w/w) and a 3D printing program (program 1 vs. program 2) on the content of bioactive compounds, antioxidant capacity, color parameters and rheological and sensory properties was investigated in 3D strawberry and strawberry tree fruit snacks. Increasing the starch content led to a decrease in the content of almost all the bioactive compounds, while it had no effect on the antioxidant capacity. The printing program had no significant effect on the bioactive compounds (except hydroxycinnamic acids), antioxidant capacity and color parameters. A higher starch content improved the strength of the sample but had no effect on the mechanical properties. Smaller particles with a higher starch content improved the stability of the sample. In contrast to the programs, varying the starch content had a significant effect on all the color parameters except the a* values. Eight different sweeteners in two different concentrations were used for the sensory evaluation of the 3D-printed snacks. The variations in sweetener content only affected the sweet and harmonious taste. In summary, this study confirms the great potential of fruit bases for the production of 3D-printed snacks with excellent biological and rheological properties, which can be a step toward personalized food with the addition of sweeteners. Novel treatments in gastrointestinal stromal tumors (GISTs) are essential due to imatinib resistance and the modest results obtained with multi-target tyrosine kinase inhibitors. We investigated the possibility that the hydroalcoholic extract from the leaves of Arbutus unedo L. (AUN) could harbor novel chemotherapeutics. The bio-guided fractionation of AUN led to a subfraction, FR2-A, that affected the viability of both imatinib-sensitive and -resistant GIST cells. Cells treated with FR2-A were positive for Annexin V staining, a marker of apoptosis. A rapid PARP-1 downregulation was observed, although without the traditional caspase-dependent cleavage. The fractionation of FR2-A produced nine further active subfractions (FRs), indicating that different molecules contributed to the effect promoted by FR2-A. NMR analysis revealed that pyrogallol-bearing compounds, such as gallic acid, gallic acid hexoside, gallocatechin, myricetin hexoside, and trigalloyl-glucose, are the main components of active FRs. Notably, FRs similarly impaired the viability of GIST cells and peripheral blood mononuclear cells (PBMCs), suggesting a non-specific mechanism of action. Nevertheless, despite the lack of specificity, the established FRs showed promising chemotherapeutic properties to broadly affect the viability of GIST cells, including those that are imatinib-resistant, encouraging further studies to investigate whether pyrogallol-bearing compounds could represent an alternative avenue in GISTs. Background: Paraquat (1,1'-dimethyl-4-4'-bipyridinium dichloride) exposure is well-established as a neurotoxic agent capable of causing neurological deficits in offspring. This study aimed to investigate therapeutic effects of Arbutus unedo L. aqueous extract (AU) against paraquat (PQ) exposure. Methods: For that the phytoconstituents of AU was determined by LC/MS, and then its antioxidant potential was assessed by DPPH and ABTS assays. The assessment included its impact on cell viability and mitochondrial metabolism using N27 dopaminergic cells. Additionally, we evaluated the effects of prenatal PQ exposure on motor coordination, dopamine levels, trace element levels, and total antioxidant capacity (TAC) in rat progeny. Results: The phytochemical profile of AU extract revealed the presence of 35 compounds, primarily phenolic and organic acids, and flavonoids. This accounted for its strong in vitro antioxidant activities against DPPH and ABTS radicals, surpassing the activities of vitamin C. Our findings demonstrated that AU effectively inhibited PQ-induced loss of N27 rat dopaminergic neural cells and significantly enhanced their mitochondrial respiration. Furthermore, daily post-treatment with AU during the 21 days of the rat's pregnancy alleviated PQ-induced motor deficits and akinesia in rat progeny. These effects inhibited dopamine depletion and reduced iron levels in the striatal tissues. The observed outcomes appeared to be mediated by the robust antioxidant activity of AU, effectively counteracting the PQ-induced decrease in TACin the blood plasma of rat progeny. These effects could be attributed to the bioactive compounds present in AU, including phenolic acids such as gallic acid and flavonoids such as quercetin, rutin, apigenin, glucuronide, and kaempferol, all known for their potent antioxidant capacity. Discussion: In conclusion, this preclinical study provided the first evidence of the therapeutic potential of AU extract against PQ-induced neurotoxicity. The aim of this study was to evaluate the genetic diversity among 36 wild strawberry tree (Arbutus unedo L.) genotypes belonging to different altitude and geographical origins, using 16 ISSR (Inter Simple Sequence Repeats) markers. Results revealed that ISSR primers produced a total of 344 bands, of which 94.1% were polymorphic, indicating a high level of genetic variation among the studied populations. The mean values of polymorphism information content (PIC), efective multiplex ratio (EMR), resolving power (Rp) and marker index (MI) were 0.34, 19.13, 23.46 and 6.45, respectively. The total gene diversity (Ht), gene diversity within populations (Hs) and total genetic differentiation coefficient among the populations (Gst) values were 0.33, 0.15 and 0.50, respectively. The genetic distance between all pairwise combinations of the genotypes ranged from 0.113 to 0.512. The cluster analysis revealed three main groups and which are subdivided into eight subgroups and a two independant branches. Therefore, the grouping of strawberry tree genotypes was independent of their geographical origins and altitude with exception for the genotypes belonging to Bin El-ouidane and Laanoucer populations. The results showed that ISSR markers are suitable tools for the evaluation of genetic diversity among strawberry tree genotypes. The fruits of Arbutus unedo L. have a crimson colour and are enriched with remarkable concentrations of bioactive compounds such as anthocyanins and polyphenols. These fruits are commonly used in the production of a Portuguese Protected Geographical Indication distillate called "Aguardente de Medronho". During this process, a solid pomace is generated and presently discarded without valuable applications. In this work, two strategies have been developed for the valorisation of A. unedo pomace. The first approach considers the extraction of polyphenols from this by-product through the optimization of an ultrasound-assisted method using a Box-Behnken design coupled with response surface methodology. The results indicate that the temperature and the percentage of methanol, along with their interaction, significantly influence the total concentration of polyphenols and the antioxidant activity of the extracts obtained. The optimal conditions identified consider the extraction of 0.5 g of sample with 20 mL of a solvent containing 74% MeOH (aq), at a pH of 4.8, maintained at 70 °C for 15 min. On the other hand, the second valorisation strategy considered the use of A. unedo pomace in the development of functional cookies. The incorporation of 15–20% pomace in the cookie formulation was well-received by consumers. This incorporation results in an intake of ca. 6.55 mg of polyphenols per gram of cookie consumed, accompanied by an antioxidant activity of 4.54 mg Trolox equivalents per gram of cookie consumed. Overall, these results encourage the employment of A. unedo pomace either as a reliable source of extracts enriched in polyphenols or as a nutraceutical active ingredient in functional cookies, thereby positively impacting human health. The valorisation and conservation of plant genetic resources (PGRs) and wild fruit PGRs are critical to ensure the maintenance of genetic and cultural heritage and to promote new perspectives on resource use. New strategies to characterize PGRs are needed, and the omics approach can provide information that is still largely unknown. The Strawberry tree (Arbutus unedo L.) is an underutilized, drought and fire-resistant species distributed in the Mediterranean area and its berries have large ethnobotanical use. Although their phenolic profile and antioxidant capacity are known, they are not well characterised, particularly from a proteomic perspective. The aim of this work is the characterisation of two ecotypes of A. unedo (Campania and Sicily) from a molecular viewpoint to valorise and encourage the preservation of this wild fruit. Samples were collected from two different geographical areas to assess whether different geographical conditions could influence the characteristics of leaves and fruits at the three stages of ripening (green, veraison, red). Proteomic analysis identified 904 proteins, of which 122 showed significance along the ripening. Some of these differentially abundant proteins, such as chalcone synthase, show a marked increase during ripening. The protein functional classes with the highest representation are involved in protein and amino acid metabolism, glycolysis and in secondary metabolism. From a proteomic perspective, there are no differences between the fruits from the two regions compared by the ripening stage. However, the pedoclimatic metabolic imprinting allowed the observation of good diversity in the metabolomic profiles between the two ecotypes, especially for anthocyanins, 4 times more abundant in the Sicilian veraisoned fruit than in the Campania one, and catechins, with double the abundance in the Campania ecotype compared to the Sicilian ecotype in the green phase, but more abundant (3x) in the Sicilian veraisoned fruit. Phenolic compounds show a 20% greater abundance in the Campania green arbutus fruit than in the Sicilian one, values that then equalise as ripening progresses. Multi-omic characterisation enhanced the knowledge on a wild fruit plant species which shows specific adaptations and responses to the environment to be considered when addressing the issue of local agrobiodiversity. This study aims to identify the individual phenolics and volatile compounds, as well as the organic acids of strawberry tree (Arbutus unedo L.) genotype fruits. The antioxidant activities were also assessed using three methods (DPPH, ABTS and βeta carotene bleaching assays) significant differences (p˂0.05) were observed among all the genotypes. Total phenols varied from 25.37 to 39.06 mg GAE/g dried weight (DW), total flavonoids ranged between 3.30 and 7.07 mg RE/g DW, and anthocyanins varied from 0.15 to 0.64 mg cya-3-glu/100 g DW. Moreover, the antioxidant activities were in the range of 3.33–21.08, 2.25–19.58, and 1.08–13 mg ascorbic acid equivalent/g DW for DPPH, ABTS and βeta carotene bleaching assays, respectively. Seventeen phenolics compounds were identified by HPLC in A. unedo fruits. Gallocatechol and catechin were the most abundant compounds. Among the volatile compounds identified, hexadecanoic acid was the most abundant in all the genotype fruits. The principal component analysis revealed that the first two components formed 66.47% of the total inertia.

The strawberry tree (Arbutus unedo L.), an evergreen bush to small tree of the Ericaceae family, is a main component of the natural flora of the Mediterranean basin that also grows profusely through the Iberian Peninsula, southwestern France, and Ireland. The small edible red fruits are usually used to produce preserves, jams, and liquors, as the Portuguese "aguardente de medronho". The leaves and fruits have been used for a long time in traditional medicine, and their bioactive compounds are presently the subject of intense research. A strawberry tree germplasm collection was recently established by the company Corte Velada (Odiáxere, Portugal). A set of 50 germplasm accessions was selected for a breeding program. A next-generation sequencing project was performed, resulting in the establishment of the first strawberry tree genome assembly and further identification of 500 SSR and 500 SNP loci. Individual molecular fingerprints for the unequivocal identification of the selected 50 accessions were established based on 71 markers alleles amplified by 4 SSR and 9 SNP markers. The same species-specific markers alleles combined with 61 random amplified markers amplified by 5 RAPD and 5 ISSR primers were used to assess the genetic variability and genetic relationships among the selected accessions. Solar drying is affordable, requiring low energy and an eco-friendly method. Thus, the present paper studies the efficiency and characteristics of the indirect solar convective drying in the fruits of Arbutus unedo L. as well as its effects on the fruit phenolic compounds. The fruit samples were dried at 60 °C, 70 °C, and 80 °C. Phenolic compounds were investigated using a Liquid Chromatography platform. Experimental results revealed that the effective moisture diffusivity determined by Fick's second law varied from 1.51 × 10−9 to 4.68 × 10−9 m2/s, and the activation energy recorded was 2203.62 kJ/kg. Both the total energy consumption and the specific electrical energy of the dried fruits decreased as temperature increased. The Midilli–Kucuk model was selected as the best-fitted model for drying Arbutus unedo L. Significant effect of temperature on phenolics was observed. The concentration of the phenolic compounds decreased by 15.54, 39, and 40.63% at 60, 70, and 80 °C, respectively. [ABSTRACT FROM AUTHOR]

The Polyphenol oxidase (PPO) leads to the enzymatic browning of fruits and vegetables and needs to be prevented in food browning and quality. The present study aimed to investigate the use of natural extracts and chemical inhibitors to prevent browning of the PPO of blackberries. Purification, characterization, and kinetics of PPO of blackberry parameters for five substrates, namely, pyrocatechol, 4-methylcatechol, Pyrogallol, Gallic acid, and tyrosine, were described. The results showed that the DEAESepharose and Superdex G-200 purification methods, which achieved electrophoretic purity, increased PPO activity by 556 fold. Purification with Sephadex GE-200 and SDS-PAGE reveals two PPO isoenzymes with an apparent molecular weight of 22 kD and 70 kD. The optimum pH and temperature values indicated were 6.6 and 25°C, respectively. The PPO showed variable affinity towards o-dihydroxy phenolic substrates with catecholase activity but without any activity observed with phenol, a monohydroxy substrate, and it was very effective towards pyrocatechol, pyrogallol, and 4-methyl catechol. The results revealed that inhibition of the PPO using both synthetic inhibitors and natural extracts was the most effective method. Quercetin and ascorbic acid showed higher inhibition with the lowest Ki values. Fresh onion (Allium cepa) and wild Arbutus unedo extract were able to inhibit the blackberry PPO activity up to 50% and 60%, respectively. Therefore, the use of natural extracts from Arbutus unedo L as anti-browning agents on the blackberry PPO may provide new insight to overcome the enzymatic browning. [ABSTRACT FROM AUTHOR]

The incorporation of 15–20% pomace in the cookie formulation was well-received by consumers. This incorporation results in an intake of ca. 6.55 mg of polyphenols per gram of cookie consumed, accompanied by an antioxidant activity of 4.54 mg Trolox equivalents per gram of cookie consumed. Overall, these results encourage the employment of A. unedo pomace either as a reliable source of extracts enriched in polyphenols or as a nutraceutical active ingredient in functional cookies, thereby positively impacting human health.

Thus, it has been used for years in folk medicine to relieve various health conditions such as urological and kidney problems, dermatological, cardiovascular and gastrointestinal diseases. The fruits are traditionally used for making jams, jellies, and strong alcoholic beverages, while the leaves are mostly used for preparing tea. Since the leaves were more researched, previous results indicated that they have important biological effects, so further research should focus on the fruits. Due to its chemical composition, rich polyphenolic profile and the biological potential derived from it, the plant has great prospects for the production of functional foods and nutraceuticals. This work focused on the extraction, quantification, and characterization of bioactive compounds of Arbutus unedo L. fruits, comparing the results obtained from the different ripening states. Extractions were performed by different methods (such as maceration extraction and ultrasonic extraction) and food grade solvents (aqueous and hydroalcoholic solvents) in each of the all ripening states (four states considered, associated with four different colors, i.e., green, yellow, orange, and red). The presence of (poly)phenols was quantified and characterized, and scavenging activity was determined by the Folin–Ciocâlteu reagent and the DPPH method, respectively. The content of bioactive compounds was characterized by LC-MS/MS, such as multiple reaction monitoring (MRM) mass spectrometry. The results showed that ultrasound-assisted extraction (UAE) performed better than maceration extraction; ethanol–water mixture extracts showed a more positive effect than the use of aqueous extracts regarding the content of total phenolic compounds. Overall, the total phenolic compounds in the EtOH:H2O mixture at a ratio of 7:3 (v:v) were higher than that of the other solvents for both extraction methods. Some bioactive molecules were characterized for the first time in the extracts of A. unedo. The chemical profile of the strawberry tree extracts depended on the degree of fruit ripeness. The results suggest that A. unedo fruits may be of great interest for food and nutraceutical applications.

The main objective of this study was to optimize the extraction process using a natural deep eutectic solvent (NADES) to obtain the highest polyphenol content and antioxidant activity of strawberry tree fruits as determined by different in vitro assays (DPPH, FRAP and ABTS). For this purpose, extraction with NADES was performed in three steps. Among twenty different NADES systems screened for extraction in the first step of solvent screening, two solvents that gave the best results (N6: Betaine–glycerine–water with a molar ratio 1:2:1 and N9: Choline chloride–glycerine with a molar ratio 1:2) were screened in the next step. For these two solvents, two separate Face-centered central composite designs with S/L ratios (10–20-30 g NADES/g sample), temperature (40–55-70 °C), and extraction time (60–120-180 min) were used by response surface methodology (RSM), with total phenolic content and antioxidant activity as the target responses. Based on the software multi-response optimization prediction, the optimal conditions for N6 and N9 were: 1:30 S/L ratio, 65 °C temperature and an extraction time of 150 min, and 1:20 S/L ratio, 56 °C and 180 min, respectively. The strawberry tree (Arbutus unedo L.), an evergreen bush to small tree of the Ericaceae family, is a main component of the natural flora of the Mediterranean basin that also grows profusely through the Iberian Peninsula, southwestern France, and Ireland. The small edible red fruits are usually used to produce preserves, jams, and liquors, as the Portuguese “aguardente de medronho”. The leaves and fruits have been used for a long time in traditional medicine, and their bioactive compounds are presently the subject of intense research. A strawberry tree germplasm collection was recently established by the company Corte Velada (Odiáxere, Portugal). A set of 50 germplasm accessions was selected for a breeding program. A next-generation sequencing project was performed, resulting in the establishment of the first strawberry tree genome assembly and further identification of 500 SSR and 500 SNP loci. Individual molecular fingerprints for the unequivocal identification of the selected 50 accessions were established based on 71 markers alleles amplified by 4 SSR and 9 SNP markers. The same species-specific markers alleles combined with 61 random amplified markers amplified by 5 RAPD and 5 ISSR primers were used to assess the genetic variability and genetic relationships among the selected accessions.

Qualitative and quantitative analyses of individual phenolic compounds by high-performance liquid chromatography (HPLC) were also carried out. Significant differences (p˂0.05) were observed across the five genotypes in total phenols (25.37–39.06 mg GAE/g DW), total flavonoids (3.30–7.07 mg RE/g DW), total anthocyanins (0.15–0.64 mg cya-3-glu/100 g DW), pH (2.44–3.92), titrable acidity (0.65–1.01 g malic acid/100 g fw), and soluble solids (14.83–18.53%). The antioxidant activity was evaluated by three assays. The values were 3.33–21.08, 2.25–19.58, and 1.08–13 mg AAE/g DW for DPPH scavenging test, ABTS, and β-carotene bleaching, respectively. Gallocatechol and catechin were the most abundant phenolic compounds. Principal component analysis showed that the first three components formed 90.25% of the total inertia. Chlorogenic acid, ellagic acid derivative I, ellagic acid, rutin, and cyanidin-3,5-diglucoside were the most involved variables in the total variance explained.

This study aims to identify the individual phenolics and volatile compounds, as well as the organic acids of strawberry tree (Arbutus unedo L.) genotype fruits. The antioxidant activities were also assessed using three methods (DPPH, ABTS and βeta carotene bleaching assays) significant differences (p˂0.05) were observed among all the genotypes. Total phenols varied from 25.37 to 39.06 mg GAE/g dried weight (DW), total flavonoids ranged between 3.30 and 7.07 mg RE/g DW, and anthocyanins varied from 0.15 to 0.64 mg cya-3-glu/100 g DW. Moreover, the antioxidant activities were in the range of 3.33–21.08, 2.25–19.58, and 1.08–13 mg ascorbic acid equivalent/g DW for DPPH, ABTS and βeta carotene bleaching assays, respectively. Seventeen phenolics compounds were identified by HPLC in A. unedo fruits. Gallocatechol and catechin were the most abundant compounds. Among the volatile compounds identified, hexadecanoic acid was the most abundant in all the genotype fruits. Solar drying is affordable, requiring low energy and an eco-friendly method. Thus, the present paper studies the efficiency and characteristics of the indirect solar convective drying in the fruits of Arbutus unedo L. as well as its effects on the fruit phenolic compounds. The fruit samples were dried at 60 °C, 70 °C, and 80 °C. Phenolic compounds were investigated using a Liquid Chromatography platform. Experimental results revealed that the effective moisture diffusivity determined by Fick’s second law varied from 1.51 × 10−9 to 4.68 × 10−9 m2/s, and the activation energy recorded was 2203.62 kJ/kg. Both the total energy consumption and the specific electrical energy of the dried fruits decreased as temperature increased. The Midilli–Kucuk model was selected as the best-fitted model for drying Arbutus unedo L. Significant effect of temperature on phenolics was observed. The concentration of the phenolic compounds decreased by 15.54, 39, and 40.63% at 60, 70, and 80 °C, respectively.

The strategies of two evergreen Mediterranean tree species (Arbutus unedo L. and Quercus ilex L.) to face annual temperature variability and extreme event responses has been monitored in continuum by leaf fluorescence and reflectance. The effect of chilling spells and the accumulation of cold days with chilling and freezing temperatures on winter photo-inhibition was discussed in terms of modulation capacity for fluorescence parameters to daily temperature and irradiance conditions. Throughout the winter Q. ilex resulted less photoinhibited than A. unedo, showing a more dynamic response in all fluorescence parameters and a higher non-photochemical quenching capacity. These characteristics effectively act to maintain a higher electron transport capacity in Q. ilex than in A. unedo.

Resumo Os medronhos são frutos com um alto teor de açúcares (40%), antioxidantes e vitaminas, tais como vitamina C, β-caroteno, niacina, tocoferóis e ácidos orgânicos. No entanto, o rendimento de extração da polpa pelos métodos convencionais é baixo, correspondendo aproximadamente a 42%. Este estudo teve como objetivo avaliar o potencial uso de duas carboidrases comerciais, Viscozyme® L e Pectinex® Ultra SP-L, na extração de polpa de medronho e a sua influência nas características físico-químicas e reológicas da polpa extraída. As carboidrases foram testadas sob diferentes temperaturas (20 ºC a 25 ºC e 45 ºC), concentrações (1, 2 e 4 μL/g de fruto macerado) e tempos de reação (30 e 60 min). As mesmas condições foram aplicadas a amostras-controle sem adição de enzima. Foram determinados os rendimentos de extração (g de polpa extraída/ 100 g de fruto macerado), viscosidade aparente (a 20,0 ± 0,1 ºC), cor, acidez titulável, pH, sólidos solúveis (°Brix) e sólidos totais das polpas obtidas. A polpa com menor viscosidade aparente (0,88 ± 0,38 Pa.s) foi obtida pela aplicação da Viscozyme® L a 4 μL/g, 45 ºC, após 30 minutos de incubação. A Pectinex® Ultra SP-L foi responsável pelo rendimento de extração de polpa mais elevado (54,9 ± 1,1%) após 30 min, quando aplicada a 2 μL/g e 45 ºC. Foi também responsável pelo maior teor de sólidos solúveis (27,3 ± 0,1 °Brix), à temperatura ambiente (20 ºC a 25 ºC) após 60 minutos para a concentração 4 μL/g. Não foram encontradas diferenças significativas (p > 0,05) para as demais características físico-químicas das polpas, entre as condições testadas. A metodologia de extração de polpa assistida por carboidrases forneceu resultados promissores para a diversificação de produtos de valor acrescentado derivados do medronho, com elevado potencial de utilização na indústria alimentar (bebidas de fruta, doces e geleias, gelados, entre outros).

Although strawberry tree (Arbutus unedo L.) leaves have long been used as a herbal remedy, insufficient information is available on their nephrotoxicity. We assessed the safety of strawberry tree water leaf extract and its key component arbutin, administered per os to Lewis rats of both genders at 200 mg/kg b.w./day for 14 and 28 days. The effects of the tested compounds on DNA integrity in renal cells was evaluated using alkaline comet assay, while kidney function was studied using serum creatinine and urea levels. Strawberry tree water leaf extract showed high biocompatibility with kidney tissue. It did not impair DNA integrity of renal cells and kidney function, either in male or female rats. However, exposure to single arbutin affected the levels of primary DNA damage in renal cells which could be related to metabolic conversion of arbutin into hydroquinone, whose nephrotoxicity has previously been proven. Due to their beneficial health effects, strawberry tree (Arbutus unedo L.) leaves have for decades been used as herbal remedy in countries of the Mediterranean region. This pilot study is the first to investigate the liver function and DNA integrity in rat hepatocytes evaluated after 14 and 28 day treatments with strawberry tree water leaf extract and arbutin, administered per os to Lewis rats of both genders at a daily dose 200 mg/kg b.w. We focused on two types of biomarkers: enzyme serum markers of liver function (AST, ALT, and LDH), and primary DNA damage in the liver cells, which was estimated using the alkaline comet assay. At the tested dose, strawberry tree water leaf extract showed acceptable biocompatibility with liver tissue both in male and female rats, especially after shorter exposure. Our results also suggest that oral administration of single arbutin to rats was not associated with significant impairments either in the liver function or DNA integrity in hepatocytes. Considering that prolonged exposure to the tested compounds revealed minor changes in the studied biomarkers, future in vivo studies have to further clarify the biological and physiological relevance of these findings. The strawberry tree (Arbutus unedo L.), grown throughout the Mediterranean, produces edible fruit; as it is easily bruised, the sweet, reddish fruit is used mostly to prepare jams, marmalades and alcoholic beverages. As the genus is paraphyletic, phytochemical analysis can assist in defining the fruit composition with the species Arbutus unedo L. (A. unedo). Here we report on the carbohydrate, total sugar, protein, fat, fiber, ash, and mineral content of wild fruit, harvested from 45 specimens from five locations. The dominant nutrients were carbohydrates (78.2–84.8 g/100 g), total sugars (52.1–67.2 g/100 g) and dietary fiber (11.0–20.1 g/100 g). Other important nutrients supplied by A. unedo fruit include P, K, and Fe. The fruit was observed to contain health-promoting components providing 42 and 36%, of recommended daily allowance (RDA) for fiber and zinc, respectively, as well as iron and manganese, at levels exceeding minimum RDA. The free-sugar profile revealed high glucose followed by fructose content with minor amounts of sucrose (14, 11, and 6 g/100 g, respectively). Significant differences both between regions and within individuals were observed for several traits. The richness of fruit nutrients in A. unedo confers nutritional value and as such, a promising alternative fruit source.

Abstract Ethnopharmacological relevance Arbutus unedo L., (Ericaceae) is one of the most traditional plants commonly used to treat diabetes in people living in Eastern Morocco region particularly in Taza and Beni Mellal. Aim of the study The aim of the study was to find if there is a scientific support to the ethnopharmacological relevance use of Arbutus unedo L., roots bark (AU) to treat diabetes. Materials and methods We studied the effects of crude aqueous extract of AU on intestinal glucose absorption using short-circuit current technique in vitro and oral glucose tolerance test in vivo. Results The aqueous extract of AU (10 µg/mL to 1 mg/mL) induced concentration-dependent inhibition of sodium-dependent glucose transport across isolated mouse jejunum. The maximal inhibition was obtained with 1 mg/mL, which exhibited more than 80% of the Phloridzin inhibition with an IC 50 close to 216 µg/mL. A 6-week AU ingestion (2 g/(kg day)), improved oral glucose tolerance as efficiently as metformin (300 mg/(kg day)). Arbutus unedo L. and metformin also reduced body weight. Conclusions Arbutus unedo L. roots bark aqueous extract directly inhibited the electrogenic intestinal absorption of glucose in vitro. In addition it improved oral glucose tolerance and lowered body weight in rats after chronic oral administration in vivo. Mediterranean trees and shrubs form intra-annual density fluctuations (IADFs) in tree rings as a sign of their plasticity in wood formation in response to intraseasonal variations of environmental conditions. Different species show a different aptitude to form IADFs, due to their diverse ability to cope with climate stressors, since the occurrence of IADFs may affect plant hydraulics. Dendroecology and quantitative wood anatomy were used to characterise IADFs in Pinus pinea and Arbutus unedo co-occurring at a Mediterranean site in Italy. The relations between climate parameters (i.e. temperature and precipitation) and intra-annual tree-ring traits (i. e. IADF frequency and conduit size) were analysed to highlight the main triggers for IADF formation and their functional role. Data showed that both species are characterised by a high plastic response to climate and formed a high frequency of L -IADFs (occurrence of earlywoodlike conduits in latewood). The two species, although forming the same type of IADFs, showed different sensitivity to environmental factors. Pinus pinea showed a high dependence of tracheid size on temperature, while Arbutus unedo was more sensitive to precipitation in spring and autumn. Arbutus unedo promptly developed more than one IADF per year in response to rainfall events following drought periods. The overall results were useful to compare the aptitude of the two species in forming IADFs and to highlight the factors priming their formation. This is useful to understand wood growth reactions to environmental drivers and to evaluate the adaptive capabilities in these two species, and thus to predict forest reactions after climate changes. The aqueous stability of the extract was monitored using the anthocyanins' content as response (determined by HPLC-DAD) in function of time, temperature and pH. Aided by mechanistic/phenomenological models, the conditions that favours the stabilization of the extract were provided, highlighting the suitability of the colorant for pastry/bakery products. As a case study, the extract was incorporated into wafers and the changes on the nutritional profile, free sugars, fatty acids and antioxidant properties were monitored during 6 days of storage. The results provide information for: i) potential application of the rich extract in anthocyanins for producing a natural colorant with bioactive properties; and ii) shelf-life predictions. In this study, the effects of strawberry tree fruit addition to physical, chemical and sensory properties of ice cream were investigated. Strawberry tree fruit ice creams were produced with two different ratios (15%, 25%) and a control group was produced. Study was conducted in two replications. Dry matter, ash, overrun, melting time, first dripping time, complete melting time, pH, titrable acidity, fat, protein, color, vitamin C (ascorbic acid), antioxidant activity, sugar, water activity, and sensory analysis were carried out and results were evaluated with statistically. When the amount of dry matter, ash, fat, pH, and total nitrogen were decreased due to addition of pulp in ice creams, amount of ascorbic acid, antioxidant activity, and titrable acidity increased. Depending on the increase in the percentage of pulp, resistance to melting was decreased; overrun was increased. The strawberry tree fruit and leaf are a rich source of minerals, easily digestible sugars, dietary fibers, vitamins (especially vitamin C) and many bioactive compounds of significant functional value. Due to their favorable chemical composition, fruits have recently become increasingly popular in consumption. The aim of this study was to determine the physical-chemical composition, content of bioactive compounds, and also the antioxidant capacity of the fruit and leaves of wild strawberry tree populations among the Adriatic coast in Croatia, as well as to investigate the influence of location on the content of specific bioactive compounds. According to the obtained results, both fruit and leaves are pronouncedly high in vitamin C content, the average value for fruits amounted to 224.21 mg/100 g FW, while that for leaves amounted to 138.08 mg/100 g FW. Additionally, significantly high values of total polyphenolic compounds were recorded both in fruits (average value of 637.94 mg GAE/100 g FW) and especially in leaves (average value of 2157.01 mg GAE/100 g FW). Several pigments from different categories were determined in the fruit depending on fruit maturity, including: total anthocyanins, β-carotene and lycopene; while in leaves chlorophylls and carotenoids. Given the high content of different bioactive compounds high values of antioxidant capacity were determined (the average value for fruits was 2269.96 µmol TE/kg and for leaves, 2237.16 µmol TE/kg). Location strongly influenced the physical-chemical composition and also the content of specialized metabolites; populations collected from southern areas (central and south Dalmatia) of the Adriatic coast tended to have higher amounts of vitamin C, total phenols, total anthocyanins and β-carotene.

There are not many exhaustive works emphasizing the amount of genetic diversity among the strawberry tree (Arbutus unedo L.) genotypes in Morocco. This work aims to assess the biochemical composition of strawberry tree fruits, as well as to establish the variation of this composition among them. In this study, total phenols (TP), total flavonoids (TF), condensed tannins (CT) and hydrolyzable tannins (HT), total anthocyanins (TA), and free radical scavenging activity through ABTS were investigated in strawberry tree fruits. Furthermore, qualitative and quantitative analyses of individual phenolic compounds by high-performance liquid chromatography (HPLC) were carried out. Color parameters such as lightness (L*), Chroma (c*), and hue angle (h°) were also investigated. All studied variables showed highly significant differences among all samples with the exception of hydrolyzable tannins and chromatic coordinates. TP varied from 22.63 ± 1.74 to 39.06 ± 2.44 mg GAE/g DW, TF varied from 3.30 ± 0.60 to 8.62 ± 1.10 mg RE/g DW, and TA ranged between 0.12 ± 0.06 and 0.66 ± 0.15 mg cya-3-glu/100 g DW. In addition, CT and HT amounts were in the range of 10.41 ± 1.07–16.08 ± 1.50 mg TAE/g DW and 4.08 ± 2.43–6.34 ± 3.47 TAE/g DW, respectively. Moreover, the IC50 value (ABTS) ranged between 1.75 and 19.58 mg AAE/g DW. 17 phenolic compounds were detected in strawberry tree fruits. Gallocatechol and catechin were the most abundant phenolic compounds. Matrix of correlations revealed significant positive and negative correlations among variables particularly c*, a*, and b*. Principal component analysis (PCA) showed that the first three components formed than 68% of the total inertia. The following variables gallic acid, protocatechuic, gallocatechin, gallic acid derivative, chlorogenic acid, syringic acid, ellagic acid derivative II, L*, and h* were the most involved in the total variance explained. Hierarchical clustering classified samples into one main cluster, with a single branch. The results highlight a high biochemical diversity within studied strawberry genotypes, which is probably more genetically related.

Phenolic compounds are well-known bioactive compounds in plants that can have a protective role against cancers, cardiovascular diseases and many other diseases. To promote local food development, a comprehensive overview of the phenolic compounds’ composition and their impact on human health from typical Mediterranean plants such as Punica granatum L., Ziziphus jujuba Mill., Arbutus unedo L., Celtis australis L., Ficus carica L., Cynara cardunculus var. Scolymus L. is provided. Moreover, the potential use of these data for authenticity determination is discussed. Some of the plants’ phenolic compounds and their impact to human health are very well determined, while for others, the data are scarce. However, in all cases, more data should be available about the content, profile and health impacts due to a high variation of phenolic compounds depending on genetic and environmental factors. Quantifying variation in phenolic compounds in plants relative to genetic and environmental factors could be a useful tool in food authentication control. More comprehensive studies should be conducted to better understand the importance of phenolic compounds on human health and their variation in certain plants.

The lipophilic composition of wild Arbutus unedo L. berries, collected from six locations in Penacova (center of Portugal), as well as some general chemical parameters, namely total soluble solids, pH, titratable acidity, total phenolic content and antioxidant activity was studied in detail to better understand its potential as a source of bioactive compounds. The chemical composition of the lipophilic extracts, focused on the fatty acids, triterpenoids, sterols, long chain aliphatic alcohols and tocopherols, was investigated by gas chromatography–mass spectrometry (GC–MS) analysis of the dichloromethane extracts. The lipophilic extractives of the ripe A. unedo berries ranged from 0.72% to 1.66% (w/w of dry weight), and consisted mainly of triterpenoids, fatty acids and sterols. Minor amounts of long chain aliphatic alcohols and tocopherols were also identified. Forty-one compounds were identified and among these, ursolic acid, lupeol, α-amyrin, linoleic and α-linolenic acids, and β-sitosterol were highlighted as the major components. To the best of our knowledge the current research study provides the most detailed phytochemical repository for the lipophilic composition of A. unedo, and offers valuable information for future valuation and exploitation of these berries.

Background: Paraquat (1,1'-dimethyl-4-4'-bipyridinium dichloride) exposure is well-established as a neurotoxic agent capable of causing neurological deficits in offspring. This study aimed to investigate therapeutic effects of Arbutus unedo L. aqueous extract (AU) against paraquat (PQ) exposure., Methods: For that the phytoconstituents of AU was determined by LC/MS, and then its antioxidant potential was assessed by DPPH and ABTS assays. The assessment included its impact on cell viability and mitochondrial metabolism using N27 dopaminergic cells. Additionally, we evaluated the effects of prenatal PQ exposure on motor coordination, dopamine levels, trace element levels, and total antioxidant capacity (TAC) in rat progeny., Results: The phytochemical profile of AU extract revealed the presence of 35 compounds, primarily phenolic and organic acids, and flavonoids. This accounted for its strong in vitro antioxidant activities against DPPH and ABTS radicals, surpassing the activities of vitamin C. Our findings demonstrated that AU effectively inhibited PQ-induced loss of N27 rat dopaminergic neural cells and significantly enhanced their mitochondrial respiration. Furthermore, daily post-treatment with AU during the 21 days of the rat's pregnancy alleviated PQ-induced motor deficits and akinesia in rat progeny. These effects inhibited dopamine depletion and reduced iron levels in the striatal tissues. The observed outcomes appeared to be mediated by the robust antioxidant activity of AU, effectively counteracting the PQ-induced decrease in TAC in the blood plasma of rat progeny. These effects could be attributed to the bioactive compounds present in AU, including phenolic acids such as gallic acid and flavonoids such as quercetin, rutin, apigenin, glucuronide, and kaempferol, all known for their potent antioxidant capacity., Discussion: In conclusion, this preclinical study provided the first evidence of the therapeutic potential of AU extract against PQ-induced neurotoxicity. Background: Diabetes mellitus (DM) is currently a major health problem and the most common chronic disease worldwide. Traditional medicinal plants remedies remain a potential adjunct therapy to maintain better glycemic control while also imparting few side-effects. Arbutus unedo L. has been traditionally used to manage several diseases including diabetes. This study was undertaken to contribute the validation of the traditional use of Arbutus unedoL. (Ericaceae) in the treatment of diabetes. Methods: In-vitro antidiabetic effect of the A. unedo roots aqueous extract was conducted using α-glucosidase and α-amylase assays. While in-vivo antidiabetic activity was conducted using streptozotocin-nicotinamide (STZ-NA) induced diabetic mice. Diabetic animals were orally administered the aqueous extract in 500 mg/kg of body weight to assess the antidiabetic effect. The blood glucose level and body weight of the experimental animals were monitored for 4 weeks. In addition, the histopathological examination of the treated mice pancreas was also conducted to observe the changes of β-cells during the treatment process. Results: The extract produced a significant decrease in blood glucose level in diabetic mice. This decrease was equivalent to that which observed in mice treated with a standard after 2–4 weeks. In addition, the plant extract exhibited a potent inhibitory effect on α-amylase and α-glucosidase activity with IC50 values of 730.15±0.25 μg/mL and 94.81±5.99 μg/mL, respectively. Moreover, the histopathologic examination of the pancreas showed a restoration of normal pancreatic islet cell architecture which observed in the diabetic mice treated with plant extract. Conclusions: The aqueous A. unedo roots extract has a significant in vitro and in vivo antidiabetic effects and improves metabolic alterations. Strawberry tree (Arbutus unedo L.) leaves have long been used in the traditional medicine of the Mediterranean region. One of their most bioactive constituents is the glycoside arbutin, whose presence makes A. unedo suitable as a potential substitute for bearberry [Arctostaphylos uva ursi (L.) Spreng] leaves, an herbal preparation widely used for treating urinary tract infections. The safety and biocompatibility of strawberry tree water leaf extract have not yet been documented well. This study estimated arbutin content in strawberry tree water leaf extract (STE) using high performance liquid chromatography. Furthermore, we performed an in vitro safety assessment of the 24 h exposure to three presumably non-toxic concentrations of standardized STE and arbutin in human peripheral blood lymphocytes using the apoptosis/necrosis assay, the alkaline comet assay, and the cytokinesis-block micronucleus cytome assay. The STE was also tested for total antioxidant capacity and lipid peroxidation. At a concentration corresponding to the maximum allowable daily intake of arbutin, the tested extract was not cytotoxic, had a negligible potential for causing primary DNA damage and even hindered micronuclei formation in lymphocytes. It also showed a valuable antioxidant capacity, and did not exert marked lipid peroxidation. These promising results represent a solid frame for further development of STE-based herbal preparations. Although arbutin generally had a low DNA damaging potential, the slowing down of lymphocyte proliferation observed after 24 h of exposure points to a cytostatic effect, which merits further research. Background and Aims Mediterranean trees have patterns of cambial activity with one or more pauses per year, leading to intra-annual density fluctuations (IADFs) in tree rings. We analysed xylogenesis (January 2015-January 2016) in Pinus pinea L. and Arbutus unedo L., co-occurring at a site on Mt. Vesuvius (southern Italy), to identify the cambial productivity and timing of IADF formation. Methods Dendrochronological methods and quantitative wood anatomy were applied and enabled IADF identification and classification. Key Results We showed that cambium in P. pinea was productive throughout the calendar year. From January to March 2015, post-cambial (enlarging) earlywood-like tracheids were observed, which were similar to transition tracheids. The beginning of the tree ring was therefore not marked by a sharp boundary between latewood of the previous year and the new xylem produced. True earlywood tracheids were formed in April. L-IADFs were formed in autumn, with earlywood-like cells in latewood. In A. unedo, a double pause in cell production was observed, in summer and winter, leading to L-IADFs in autumn as well. Moreover, the formation of more than one IADF was observed in A. unedo. Conclusions Despite having completely different wood formation models and different life strategies, the production of earlywood, latewood and IADF cells was strongly controlled by climatic factors in the two species. Such cambial production patterns need to be taken into account in dendroecological studies to interpret climatic signals in wood from Mediterranean trees. [ABSTRACT FROM AUTHOR]

Genetic analysis of plants relies on high yields of pure DNA. For the strawberry tree (Arbutus unedo) this represents a great challenge since leaves can accumulate large amounts of polysaccharides, polyphenols and secondary metabolites, which co-purify with DNA. For this specie, standard protocols do not produce efficient yields of high-quality amplifiable DNA. Here, we present for the first time an improved leaf-tissue protocol, based on the standard cetyl trimethyl ammonium bromide protocol, which yields large amounts of high-quality amplifiable DNA. Key steps in the optimized protocol are the addition of antioxidant compounds—namely polyvinyl pyrrolidone (PVP), 1,4-dithiothreitol (DTT) and 2-mercaptoethanol, in the extraction buffer; the increasing of CTAB (3%, w/v) and sodium chloride (2M) concentration; and an extraction with organic solvents (phenol and chloroform) with the incubation of samples on ice. Increasing the temperature for cell lyses to 70 °C also improved both DNA quality and yield. The yield of DNA extracted was 200.0 ± 78.0 µg/µL and the purity, evaluated by the ratio A260/A280, was 1.80 ± 0.021, indicative of minimal levels of contaminating metabolites. The quality of the DNA isolated was confirmed by random amplification polymorphism DNA and by inter-simple sequence repeat amplification, proving that the DNA can be amplified via PCR.

Phytopharmaceuticals play an essential role in medicine, since the need to investigate highly effective and safe drugs for the treatment of diabetes mellitus disease remains a significant challenge for modern medicine. Arbutus unedo L. root has various therapeutic properties, and has been used widely in the traditional medicine as an antidiabetic agent. The current study aimed to isolate the pharmacologically active compound from A. unedo roots using accelerated solvent extraction technology, to determine its chemical structure using different instrumental analytical methods, and also to evaluate the α-glucosidase inhibitory activity. The roots of A. unedo were exhaustively extracted by high-pressure static extraction using the Zippertex® technology (Dionex-ASE, Paris, France), and the extract was mixed with XAD-16 resin to reach quantifiable amounts of active compounds which were identified by high-pressure liquid chromatography (HPLC), 1H NMR (300 MHz), and 13C NMR. The antidiabetic activity of the isolated compound was evaluated using the α-glucosidase inhibitory assay. The active compound was isolated, and its structure was identified as catechin using instrumental analysis.The results revealed that the isolated compound has potential α-glucosidase inhibitory activity with an IC50 value of 87.55 ± 2.23 μg/mL greater than acarbose. This was used as a positive control, which has an IC50 value of 199.53 ± 1.12 μg/mL. According to the results achieved, the roots of A. unedo were considered the best source of catechin and the Zippertex® technology method of extraction is the best method for isolation of this therapeutic active compound. In addition, the α-glucosidase inhibitory activity results confirmed the traditional use of A. unedo roots as an antidiabetic agent. Future clinical trials and investigations of antidiabetic and other pharmacological effects such as anticancer are required.

Three independent experiments assessed CO2 assimilation and metals leaf deposition of seven evergreen shrub sspecies (Arbutus unedo L., Elaeagnus ebbingei L., Laurus nobilis L., Ligustrum japonicum Thunb., Photinia fraseri Dress., Viburnum tinus subsp. lucidum L., and Viburnum tinus subsp. tinus L.). CO2 assimilation and carbon allocation were determined in 2011 (Exp. 1) under optimal water availability and in 2012 (Exp. 2) under drought on potted plants. A third experiment (Exp. 3) measured seasonal leaf depositions of Cd, Cu, Ni, Pb, and Zn in 2011 on plants transplanted in proximity of a four-lane road. E. ebbingei showed the highest CO2 assimilation under optimal water availability but one of the lowest under drought (Exp. 1, 2). Conversely, P. fraseri had intermediate CO2 assimilation but it declined less during drought compared to the other species. In Experiment 3, E. ebbingei showed the highest metal deposition, mainly due to its greater leaf area. Greater rainfall and RH% decreased metal depositions, whilst greater wind velocity and air temperature increased leaf depositions. Species which drastically reduce CO2 assimilation under drought (V. tinus subsp. lucidum, L. japonicum, E. ebbingei) are not recommended in droughtprone environments, where drought-tolerant "mesic" species (P. fraseri), should be preferred. E. ebbingei could be used to optimize deposition of metals. The three experiments provide useful insights especially about CO2 assimilation (Exp. 1, 2) and air pollution mitigation (Exp. 3) of widely used shrubs for application in urban areas and planning of roadside greening in southern Europe.
