Sea urchin (class Echinoidea) gonads are a prized delicacy in Japan and many other world cultures. The complexity of its fatty acid (FA) profile, particularly minor FA, presents a formidable analytical challenge. We applied solvent mediated (SM) covalent adduct chemical ionization (CACI) tandem mass spectrometry to comprehensive de novo structural and quantitative characterization of the FA profile of Gulf of Mexico Atlantic sea urchin (Arbacia punctulata). >100 FA were detected including many with unusual double bond structure. Gulf sea urchin gonad lipids are rich in Δ5 monounsaturated FA 20:1(5Z) at 2.7% and the polymethylene-interrupted (PMI) diene 20:2(5Z,11Z) at 4.9%, as well as common omega-3 eicosapentaenoic acid (EPA; 5Z, 8Z, 11Z, 14Z, 17Z) at 9.8%±3.1% and arachidonic acid (AA; 5Z, 8Z, 11Z, 14Z) at 6.1%±2.1%. We propose plausible desaturation/elongation-based biochemical pathways for the endogenous production of unusual unsaturates. Unusual unsaturates may modify mammalian signaling and present novel bioactivities.

BACKGROUND AND PURPOSE Sperm from many species share the sperm-specific Ca2+ channel CatSper that controls the intracellular Ca2+ concentration and, thereby, the swimming behaviour. A growing body of evidence suggests that the mechanisms controlling the activity of CatSper and its role during fertilization differ among species. A lack of suitable pharmacological tools has hampered the elucidation of the function of CatSper. Known inhibitors of CatSper exhibit considerable side effects and also inhibit Slo3, the principal K+ channel of mammalian sperm. The compound RU1968 was reported to suppress Ca2+ signaling in human sperm by an unknown mechanism. Here, we examined the action of RU1968 on CatSper in sperm from humans, mice, and sea urchins. EXPERIMENTAL APPROACH We resynthesized RU1968 and studied its action on sperm from humans, mice, and the sea urchin Arbacia punctulata by Ca2+ fluorimetry, single-cell Ca2+ imaging, electrophysiology, opto-chemistry, and motility analysis. KEY RESULTS RU1968 inhibited CatSper in sperm from invertebrates and mammals. The compound lacked toxic side effects in human sperm, did not affect mouse Slo3, and inhibited human Slo3 with about 15-fold lower potency than CatSper. Moreover, in human sperm, RU1968 mimicked CatSper dysfunction and suppressed motility responses evoked by progesterone, an oviductal steroid known to activate CatSper. Finally, RU1968 abolished CatSper-mediated chemotactic navigation in sea urchin sperm. CONCLUSION AND IMPLICATIONS We propose RU1968 as a novel tool to elucidate the function of CatSper channels in sperm across species.

Natural phenomena act as external cues that stimulate the reproductive activity of many marine organisms. In this study, we report the gonadal maturation in relation to the lunar cycle of wild-caught Atlantic sea urchin, Arbacia punctulata (Echinodermata: Arbaciidae, an edible marine invertebrate), in the Gulf of Mexico. Weekly changes in gonadal development were observed histologically according to the lunar cycle during the summer months from May to July. Histological observations revealed that testes were developed synchronously around the new moon in June and July. Several testes collected in the same moon phase contained residual sperm (~7%) or testicular lobules/acini fully occupied by mature sperm (~46%). Cyclic ovarian development was also observed during the summer months, some ovaries collected around the new moon in June and July contained oocytes at maturation stage (ova: ~39-71%) or empty follicles, suggesting that the Atlantic sea urchin spawns according to the lunar cycle in the Gulf of Mexico. To the best of our knowledge, this is the first report on the lunar reproductive rhythm of Arbacia species in the Gulf of Mexico.

Cilia serve as cellular antennae that translate sensory information into physiological responses. In the sperm flagellum, a single chemoattractant molecule can trigger a Ca2+ rise that controls motility. The mechanisms underlying such ultra‐sensitivity are ill‐defined. Here, we determine by mass spectrometry the copy number of nineteen chemosensory signaling proteins in sperm flagella from the sea urchin Arbacia punctulata. Proteins are up to 1,000‐fold more abundant than the free cellular messengers cAMP, cGMP, H+, and Ca2+. Opto‐chemical techniques show that high protein concentrations kinetically compartmentalize the flagellum: Within milliseconds, cGMP is relayed from the receptor guanylate cyclase to a cGMP‐gated channel that serves as a perfect chemo‐electrical transducer. cGMP is rapidly hydrolyzed, possibly via “substrate channeling” from the channel to the phosphodiesterase PDE5. The channel/PDE5 tandem encodes cGMP turnover rates rather than concentrations. The rate‐detection mechanism allows continuous stimulus sampling over a wide dynamic range. The textbook notion of signal amplification—few enzyme molecules process many messenger molecules—does not hold for sperm flagella. Instead, high protein concentrations ascertain messenger detection. Similar mechanisms may occur in other small compartments like primary cilia or dendritic spines., Reversed enzyme‐messenger ratios in sperm cilia ensure high‐sensitivity, low noise messenger detection.

The effects of microplastic pollution on sea urchins has received little attention despite their ecological and economical importance. This is the first study to focus on adult sea urchins (Arbacia punctulata). These organisms were exposed to storm-like sediment resuspension of microplastic concentrations (9-μm polystyrene 25,000 spheres L−1) combined with salinity reductions (salinity 25 vs. 33) associated with high precipitation. Urchins were exposed to these parameters for 24 h before assessing righting times and for 48 h before assessing oxygen consumption rates. No significant impacts on urchin physiology were observed showing resilience to short-term exposures of storm-like induced microplastics and salinity. No microplastic particles blocked the madreporite pores indicating the active removal of particles by cilia and pedicellariae. Gut tissue samples indicated consumption of microplastics. Studies on more species are urgently required to determine their responses to plastic pollution to inform management decision-making processes.

The purple-spined sea urchin ( Arbacia punctulata ) is commonly found in shallow waters of the western Atlantic Ocean from the New England area of the United States to the Caribbean. Sea urchins play a major role in ocean ecology, echinoculture, and biomedical research. Additionally, sea urchins are commonly displayed in public aquaria. Baseline parameters were developed in unanesthetized urchins for righting reflex (time to regain oral recumbency) and spine response time to tactile stimulus. Tricaine methanesulfonate (MS-222) was used to sedate and anesthetize purple-spined sea urchins and assess sedation and anesthetic parameters, including adhesion to and release from a vertical surface, times to loss of response to tactile stimulus and recovery of righting reflex, and qualitative observations of induction of spawning and position of spines and pseudopodia. Sedation and anesthetic parameters were evaluated in 11 individuals in three circumstances: unaltered aquarium water for baseline behaviors, 0.4 g/L MS-222, and 0.8 g/L MS-222. Induction was defined as the release from a vertical surface with the loss of righting reflex, sedation as loss of righting reflex with retained tactile spine response, anesthesia as loss of righting reflex and loss of tactile spine response, and recovery as voluntary return to oral recumbency. MS-222 proved to be an effective sedative and anesthetic for the purple-spined sea urchin at 0.4 and 0.8 g/L, respectively. Sodium bicarbonate used to buffer MS-222 had no measurable sedative effects when used alone. Anesthesia was quickly reversed with transfer of each individual to anesthesia-free seawater, and no anesthetic-related mortality occurred. The parameters assessed in this study provide a baseline for sea urchin anesthesia and may provide helpful comparisons to similar species and populations that are in need of anesthesia for surgical procedures or research.

The majority of aquatic toxicity data for petroleum products has been limited to a few intensively studied crude oils and Corexit chemical dispersants, and acute toxicity testing in two standard estuarine test species: mysids (Americamysis bahia) and inland silversides (Menidia beryllina). This study compared the toxicity of two chemical dispersants commonly stock piled for spill response (Corexit EC9500A®, Finasol®OSR 52), three less studied agents (Accell Clean®DWD dispersant; CytoSol® surface washing agent; Gelco200® solidifier), and three crude oils differing in hydrocarbon composition (Dorado, Endicott, Alaska North Slope). Consistent with listings on the U.S. National Contingency Plan Product Schedule, general rank order toxicity was greatest for dispersants and lowest for the solidifier. The results indicate that freshwater species can have similar sensitivity as the conventionally tested mysids and silversides, and that the sea urchin (Arbacia punctulata) appears to be a reasonable addition to increase taxa diversity in standardized oil agent testing.

Gossypol (GP), a phenolic aldehyde isolated from cotton seed, exhibits various biological activities. Zn vitro effects include inhibition of sperm motility (1) inactivation of some parasites (2), and inhibition of the infectivity of HIV (3). GP is lipophilic and has a maximum attainable concentration in aqueous media of about 100 WM. Although an effective dose for the in vitro activities is around I10 pLM, 1 O-fold higher doses are required when 5% serum albumin is present in the medium. In some studies, high concentrations of GP are desired to establish dose-response relationships, and this proves difficult because of the limited aqueous solubility. This problem has hampered the development of potentially useful pharmaceutical preparations of GP, such as a protective vaginal cream (4). Water soluble gossypol (GP-S) was prepared at the Institute of Bioorganic Chemistry, Uzbek Academy of Science (Tashkent, USSR). The content of gossypol in GP-S was determined by optical absorption spectrophotometry. The spectrum was analyzed for its shape by log-normal curve fitting analysis, as described by Metzler et al. (5). GP-S contains 7.5% gossypol (w/ w), based upon its spectral analysis in 1 N NaOH and an apparent molar absorptivity of 22,000 cm-’ Mm’. We were able to prepare up to 6 mM GP-S solution in aqueous media (i.e., 0.1 M potassium phosphate buffer, pH 8.0). Spectra at different pHs (21 1) showed the identical shift, as observed with the GP standard. Arbacia and Spisula sperms were used, as described before (6), to examine the potency of GP-S to inhibit sperm motility. Figure 1 shows the effect of GP-S on the fertilizability of Arbaciu sperms. Fifty percent inhibition is observed at 10 PM of GP-S. Complete inhibition is observed at doses greater than 100 FLM. Identical results were obtained with Spisulu sperms (data not shown). The present results reveal that GP-S can inhibit sperm motility at concentrations equivalent to those of GP (7). Kanwar et al. (8) previously reported that gossypol (10 pA4) reduced the 45Ca++ uptake (25%) of isolated human spermatozoa1 plasma membrane vesicles. This effect of GP-S upon calcium uptake was further examined with whole sperms from Spisulu. Sperms were collected from clams, suspended, and washed in Ca”-free artificial seawater (MBL). Time-dependent incorporation of 45Ca++ into intact sperm was examined by: 1) incubating the sperm suspension with 45Ca+i (10 ji a steady state was then maintained during the subsequent 40 min, followed by a slight reduction. Study question Is Raman micro-spectroscopy a valid approach to assess the biochemical hallmarks of sperm regions (head, midpiece and tail) in four different species? Summary answer Non-invasive Raman micro-spectroscopy provides spectral patterns enabling the biochemical characterization of the three sperm regions in the four species, revealing however high similarities for each region among species. What is known already Raman micro-spectroscopy has been described as an innovative method to assess sperm features having the potential to be used as a non-invasive selection tool. However, except for nuclear DNA, the identification and assignment of spectral bands in Raman-profiles to the different sperm regions is scarce and controversial. Study design size, duration Raman spectra from head, midpiece and tail of four different species were obtained. Sperm samples were collected and smeared on microscope slides. Air dried samples were subjected to Raman analysis using previously standardized procedures. Participants/materials, setting, methods Sperm samples from (i) two donors attending the infertility clinic at the Centre of Reproductive Medicine and Andrology; (ii) two C57BL/6 -TgN (ACTbEGFP) 1Osb adult mice; (iii) two adult Cynomolgus monkeys (Macaca fascicularis) and (iv) two sea urchins (Arbacia punctulata) were used to characterize and compare their spectral profiles. Differences and similarities were confirmed by principal component analysis (PCA). Main results and the role of chance Several novel region-specific peaks were identified. The three regions could be differentiated by distinctive Raman patterns irrespective of the species. However, regardless of the specie, their main spectral pattern remains mostly unchanged. These results were corroborated by the PCA analysis and suggest that the basic constituents of spermatozoa are biochemically similar among species. Limitations reasons for caution Further research should be performed in live sperm to validate the detected spectral bands and their use as markers of distinctive regions. Wider implications of the findings Raman peaks that have never been described in the sperm cell were detected. Particularly important are those that are unique to the midpiece as they might be a reference to the identification of sperm mitochondria, whose function is highly correlated with that of sperm. Spermatozoa sea urchin swimming behaviour is regulated by small peptides from the egg outer envelope. Speract, such a peptide, after binding to its receptor in Strongylocentrotus purpuratus sperm flagella, triggers a signaling pathway that culminates with a train of intracellular calcium oscillations, correlated with changes in sperm swimming pattern. This pathway has been widely studied but not fully characterized. Recent work on Arbacia punctulata sea urchin spermatozoa has documented the presence of the Ca2+ CatSper channel in their flagella and its involvement in chemotaxis. However, if other calcium channels participate in chemotaxis remains unclear. Here, based on an experimentally-backed logical network model, we conclude that CatSper is fundamental in the S. purpuratus speract-activated sea urchin sperm signaling cascade, although other Ca2+ channels could still be relevant. We also present for the first time experimental corroboration of its active presence in S. purpuratus sperm flagella. We argue, prompted by in silico knock-out calculations, that CatSper is the main generator of calcium oscillations in the signaling pathway and that other calcium channels, if present, have a complementary role. The approach adopted here allows us to unveil processes, which are hard to detect exclusively by experimental procedures.

The isotope composition of boron (B) in marine biogenic carbonates has been predominantly studied as a proxy for monitoring past changes in seawater pH and carbonate chemistry. In order to derive seawater pH from boron isotope ratio data, a number of assumptions related to chemical kinetics and themodynamic isotope exchange reactions are necessary. Furthermore, the boron isotope composition (δ 11 B) of biogenic carbonates (δ 11 B CaCO3 ) is assumed to reflect the δ 11 B of dissolved borate (B(OH) 4 − ) in seawater. Here we report the development of methodology for measuring the δ 11 B in biogenic carbonate samples at the multi-collector inductively coupled mass spectrometry facility at Ifremer (Plouzane, France) and the evaluation of δ 11 B CaCO3 in a diverse range of marine calcifying organisms. We evaluated the δ 11 B CaCO3 of 6 species of marine calcifiers (a temperate coral, Oculina arbuscula ; a coralline red alga, Neogoniolithion sp.; a tropical urchin, Eucidaris tribuloides ; a temperate urchin, Arbacia punctulata ; a serpulid worm, Hydroides crucigera ; and an American oyster, Crassostrea virginica ) that were reared for 60 days in isothermal seawater (25 °C) equilibrated with an atmospheric p CO 2 of ca. 409 μatm. We observe large inter-species variability in δ 11 B CaCO3 (ca. 20 ‰) and significant discrepancies between measured δ 11 B CaCO3 and δ 11 B CaCO3 expected from established relationships between δ 11 B CaCO3 and seawater pH. We discuss these results in the context of various proposed mechanisms of biocalcification, including the potential dominant role that internal calcifying site pH plays in regulating CaCO 3 saturation state and borate δ 11 B at the site of calcification and, thus, the δ 11 B composition of calcifers’ shells and skeletons.

Sperm guidance is controlled by chemical and physical cues. In many species, Ca(2+) bursts in the flagellum govern navigation to the egg. In Arbacia punctulata, a model system of sperm chemotaxis, a cGMP signaling pathway controls these Ca(2+) bursts. The underlying Ca(2+) channel and its mechanisms of activation are unknown. Here, we identify CatSper Ca(2+) channels in the flagellum of A.punctulata sperm. We show that CatSper mediates the chemoattractant-evoked Ca(2+) influx and controls chemotactic steering; a concomitant alkalization serves as a highly cooperative mechanism that enables CatSper to transduce periodic voltage changes into Ca(2+) bursts. Our results reveal intriguing phylogenetic commonalities but also variations between marine invertebrates and mammals regarding the function and control of CatSper. The variations probably reflect functional and mechanistic adaptations that evolved during the transition from external to internal fertilization.

Herbivores tend to increase feeding rate and fitness when consuming a mixed diet relative to a single diet. According to the detoxification limitation hypothesis (DLH), feeding choices and rates when confronted with chemically rich plants are determined by herbivore physiology, and specifically by the metabolic pathways that herbivores use to manipulate secondary metabolites. We tested two predictions of the DLH using two generalist herbivores, the urchin Arbacia punctulata and amphipod Ampithoe longimana. These herbivores have geographic ranges which overlap with brown seaweeds that produce diterpenes (Dictyota menstrualis, D. ciliolata) and a green seaweed that produces sesquiterpenes and diterpenes (Caulerpa sertularioides). As predicted by the DLH, herbivore consumption rates in no-choice feeding assays were limited by extract intake rates. This suggests an upper limit in the herbivores’ abilities to physiologically manipulate seaweed metabolites. Contrary to a second prediction of the DLH, urchins consumed equal amounts of foods coated with limiting concentrations of two seaweed extracts offered singly, as a mixture, or as a pairwise choice. This result suggests that secondary metabolites of these seaweeds are manipulated by a linked set of detoxification pathways. Improving our understanding of the mechanisms that underlie diet mixing depends on greater attention to the physiology of herbivore resistance to secondary metabolites.

Many terrestrial and most marine herbivores have generalist diets, yet the role that evolutionary history plays in their foraging behaviors is poorly documented. On tropical hard-bottom reefs, generalist fishes and sea urchins readily consume seaweeds that produce lipophilic secondary metabolites. In contrast, herbivores on temperate reefs less commonly encounter seaweeds with analogous metabolites. This biogeographic pattern suggests that tropical herbivores should evolve greater feeding resistance to lipophilic defenses relative to temperate herbivores, but tests of this biogeographic pattern are rare. We offered lipophilic extracts from nine subtropical seaweeds at two concentrations to sea urchins (four subtropical and three cold-temperate populations) and quantified urchin feeding resistance. Patterns of feeding resistance toward lipophilic defenses were more similar within genera than across genera of urchins, indicating a substantial role for phylogenetic history in the feeding ecology of these generalist herbivores. The biogeographic origin of urchins also influenced feeding resistance, as subtropical species displayed greater feeding resistance than did temperate species. Similarly, a subtropical population of Arbacia punctulata had greater feeding resistance for Dictyota and Stypopodium extracts relative to temperate A. punctulata. We conclude that evolutionary history plays a more central role in the foraging ecology of generalist herbivores than is currently appreciated.

Larval survival and growth of Arbacia punctulata (Echinodermata: Echinoidea) fed with five micro-algae at two salinities. Fertilized eggs from an spontaneously spawn of thirty sexually mature sea urchins (Arbacia punctulata) were incubated to complete embryonic development. The echinopluteus larvae (3 ind/ml) were distributed into 50 plastic containers (25 containers at 30 psu and 25 containers at 40 psu) and fed on Tetraselmis chuii, Nannochloropsis oculata, Isochrysis galbana, Chaetoceros gracilis and C. calcitrans under a natural photoperiod. The water of the containers was partially renewed (75%) everyday. Larval anatomic development aspects, daily survival and growth were determined. The growth was determined through postoral arms and body length measurement, and body diameter of twelve larvae during metamorphosis. During the planktonic larval phase, only the I. galbana diet produced similar results for both salinities. The relative growth of larvae was isometric (I) for larvae fed on I. galbana at two salinities and positive allometric for those fed on C. gracilis and C. calcitrans at both salinities. In this study A. punctulata started metamorphosis at day 14 and was completed 30 days after fecundation. The morphologically convergent larvae of the echinoderm classes Ophiuroidea and Echinoidea have been suggested to be functionally dissimilar when it comes to their capacities to feed, but little is known about whether these larvae are similar in terms of energetics. Here, we compare the energetics of early development of a tropical ophiuroid, Ophiocoma alexandri, and a temperate to tropical echinoid, Arbacia punctulata, two species with similarly sized eggs. Measurements of respiration and constituent analyses were performed on eggs and unfed larvae of both species. Members of both species showed an increase in oxygen consumption during morphogenesis followed by a lower, static rate once morphogenesis was complete (3 d for O. alexandri and 1.3 d for A. punctulata). Compared to the echinoid larvae, the ophiuroid larvae developed more slowly and had peak respiration rates that were 3.1× lower. Eggs of O. alexandri contained significantly more protein and significantly less triacylglycerol than eggs of A. punctulata. Energy utilization, as calculated via respiration measurements, closely matched decreases in energy content from the eggs to larvae as measured with biochemical constituent assays. Larvae of A. punctulata used 1.4× more energy to reach the pluteus stage than larvae of O. alexandri, and used 4× more energy during the first 9 d of larval life. These data suggest that echinoid larvae require more energy to develop to the feeding stage than ophiuroid larvae, and likewise have higher requirements for maintenance metabolism. Ophiuroid larvae may be more tolerant of low food levels due to their very low metabolic rates, but this advantage may be offset by their slower rate of development.

Life-history models for marine invertebrate animals assume a strong correlation between the size of the egg and the time from fertilization until metamorphosis. This assumption is supported by comparative data across a wide range of phyla. However, the exact form of the relationship between egg size and development time has a strong effect on the predicted outcomes of life-history models and has been the subject of much debate. Comparative data suggest that as egg size increases the effect of egg size reductions on development time will decrease and the effects of size reductions on juvenile size will increase. I used blastomere separations to test the effects of a 50% reduction in egg volume on development time and juvenile size in three species of echinoid echinoderms (Arbacia punctulata, Strongylocentrotus purpuratus, and Dendraster excentricus) that develop from eggs of a range of sizes (80–124 μm). I also manipulated the food level given to developing embryos and larvae to investigate the potential interaction between the effects of egg size reductions and food availability. Larvae from halved zygotes took significantly longer to develop to metamorphosis than their whole size counterparts in all three species. In only one species was I able to detect a significant reduction in juvenile size for offspring developing from halved zygotes. When compared with similar manipulations in species with larger eggs (140–387 μm), egg size reductions have a stronger effect on development time in species with small eggs. As predicted, development time does not change linearly with egg size but instead increases exponentially as egg size is reduced. The relationship between egg size and juvenile size remains unclear. Further investigations into the factors, including egg size, that influence juvenile performance are warranted.

Regular echinoid skeletons, or tests, comprise plate patterns and overall shapes that have proven challenging to analyse solely on the basis of any one approach or process. Herein, we present a computational model, Holotestoid, that emulates four macrostructural ontogenic processes involved in test growth (plate growth, plate addition, plate interaction, and plate gapping). We devise a geometric representation for analysing tests and describe how we use analogies (bubble interactions and close-packing) to emulate the processes. In the computational model, the emulated processes are used to determine the plate size and plate shape and combined to simulate a growth zone. We simulated growth zones for Arbacia punctulata and for Strongylocentrotus franciscanus by changing the value for one parameter, the ambulacral column angle. We quantitatively compared morphological features for simulated forms to those for real specimens to test the computational model. Additionally, we simulated growth zones for A. punctulata, S. franciscanus, Eucidaris thouarsii, and Mellita quinquiesperforata by changing three parameters, ambulacral column angle, peristome radius to apical system radius ratio, and apical system radius to column length ratio. Holotestoid can be used to explain morphological disparity among echinoid tests.

Sodium hypochlorite (NaOCl) or bleach, found in effluent from wastewater treatment plants, can act as an environmental toxin. The sea urchin Arbacia punctulata is a common subject of embryological toxicity tests due to its sensitivity to environmental pollutants. Using concentrations of NaOCl that mimic those found in treated wastewater (0.11 ppm, 0.06 ppm, and 0.03 ppm) we observed minimal affects on early larval development, though most larvae took longer to develop at higher NaOCl concentration. There was a significant difference in the percentage of non-normal plutei based on concentration (P = 0.038) and significant interaction between the percent of each morphology and NaOCl concentration (P = 0.0027). The most significant change in non-normal plutei was in the retarded (shortened skeletal rods) malformation which increased in frequency with NaOCl concentration (P = 0.001). There was a significant reduction in skeletal length in both normal and retarded plutei (P < 0.05) as NaOCl increased.

The role of echinoid parental nutrition in early-life stage toxicity is not well understood. Arbacia punctulata were fed either a fresh diet consisting of organic lettuce and carrots or a dry feed. Embryos from parents fed the dry feed exhibited lower sensitivity to copper, whereas the opposite occurred with 1,3,5-trinitrobenzene and sodium dodecyl sulfate (SDS). EC(50) values for the dry and fresh feed treatments, respectively, were 41.0 and 29.9 microg/L for copper, 0.5 and 1.8 mg/L for 1,3,5-trinitrobenzene, and 3.5 and 5.6 mg/L for SDS. The data suggests that nutritional standardization for sea urchins in ecotoxicological laboratories needs to be addressed and further investigated.

Gulf of Mexico blooms of the dinoflagellate Karenia brevis produce neurotoxic cyclic polyethers called brevetoxins. During and after a red tide bloom in southwestern Florida, K. brevis cells lyse and release brevetoxins, which then sink to the benthos and coat the surfaces of seagrasses and their epiphytes. We tested the possibility that these brevetoxin-laden foods alter the feeding behavior and fitness of a common benthic herbivore within Floridean seagrass beds, the amphipod Ampithoe longimana. We demonstrated that coating foods with K. brevis extracts that contain brevetoxins at post-bloom concentrations (1 microg g(-1) drymass) does not alter the feeding rates of Florida nor North Carolina populations of A. longimana, although a slight deterrent effect was found at eight and ten-fold greater concentrations. During a series of feeding choice assays, A. longimana tended not to be deterred by foods coated with K. brevis extracts nor with the purified brevetoxins PbTx-2 and PbTx-3. Florida juveniles isolated with either extract-coated or control foods for 10 days did not differ in survivorship nor growth. A similar lack of feeding response to brevetoxin-laden foods also was exhibited by two other generalist herbivores of the southeastern United States, the amphipod A. valida and the urchin Arbacia punctulata. Given that benthic mesograzers constitute a significant portion of the diet for the juvenile stage of many nearshore fishes, we hypothesize that the ability of some mesograzers to feed on and retain brevetoxins in their bodies indicates that mesograzers may represent an important route of vertical transmission of brevetoxins through higher trophic levels within Gulf of Mexico estuaries.

Quantitative structure-activity relationship models were obtained by applying the Molecular Descriptor Family approach to eight ordnance compounds with different toxicity on five marine species (arbacia punctulata, dinophilus gyrociliatus, sciaenops ocellatus, opossum shrimp, and ulva fasciata). The selection of the best among molecular descriptors generated and calculated from the ordnance compounds structures lead to accurate monovariate models. The resulting models obtained for six endpoints proved to be accurate in estimation (the squared correlation coefficient varied from 0.8186 to 0.9997) and prediction (the correlation coefficient obtained in leave-one-out analysis varied from 0.7263 to 0.9984).
