The biobanks from dermal biopsies represent an interesting strategy for biodiversity conservation. Nevertheless, the morphological and cellular patterns of the dermis can be influenced by the age and sex of the individual. Therefore, evaluating these factors is interesting for forming biobanks of Antillean manatees. These animals, representatives of marine fauna, have had their population reduced, and biobanks are essential for their conservation. Then, we evaluated the effects of age (3.5 years vs. 3.6–16 years vs. 23.6 years) and sex (males vs. females) on morphological and cellular parameters using histological and in vitro culture techniques. Regardless of age, no differences were observed for dermal thickness, collagen fibres, tissue proliferative activity and viable cell recovery. Nonetheless, fibroblast reduction was observed in groups aged 23.6 years compared to other animals (p < 0.05). Additionally, cells from animals aged 3.6–16 years showed more significant mitochondrial damage than the other groups (p < 0.05). Regardless of sex, no differences were observed for dermal thickness, collagen fibres, tissue proliferative activity and viable cell recovery; however, females had fewer fibroblasts than males (p < 0.05). Cells from females showed lower mitochondrial damage when compared to cells from males. In summary, although age and sex do not influence dermal thickness and cell recovery, variations in the number of fibroblasts and mitochondrial characteristics were observed among the groups. Context: Several assisted reproduction techniques have been proposed for collared peccaries due to the interest in its conservation. However, there is little information about the requirements for sperm capacitation, an initial step in in vitro fertilisation. Aims: We aimed to determine the optimal conditions for collared peccary sperm capacitation by comparing Tyrode's albumin lactate pyruvate (TALP) or the same media plus heparin, caffeine, or their combination at different exposure times (1, 3, and 6 h). Methods: The samples were evaluated for kinetic parameters, membrane functionality and integrity, mitochondrial activity, morphology, DNA integrity, reactive oxygen species (ROS) expression levels, capacitation, and sperm binding ability using swine oocytes. Key results: Samples incubated with caffeine or heparin had a higher percentage of capacitated spermatozoa. The maximum percentage of capacitation was achieved after 3 h of incubation with either agent. Moreover, spermatozoa subjected to heparin capacitation showed better motility than when subjected to caffeine, and lower ROS expression levels after 1 h. No differences were observed among incubation times for the binding ability. Conclusions: In summary, collared peccary spermatozoa can be capacitated with caffeine or heparin; however, heparin better maintains sperm motility and ROS expression levels. The co-incubation of gametes in a medium with heparin for 3 h could be efficient for in vitro fertilisation in collared peccaries. Implications: This result will contribute to the development of assisted reproduction techniques for conservation and productivity of collared peccaries. In vitro fertilisation can contribute to the conservation and knowledge of the reproductive characteristics of collared peccary. Simple Summary: The collared peccary is a wild ungulate important for the maintenance of ecosystems in the Americas. The in vitro fertilization (IVF) technique can contribute to the conservation of and knowledge about the reproductive characteristics of this species. The collared peccary's spermatozoa are sensitive to stress conditions. Therefore, the steps of seminal plasma (SP) removal and sperm selection for fertilization need to be determined. Consequently, the most used methods in domestic ungulate species, namely, swim-up, Percoll
® gradient (PG), and washing by centrifugation (WC), were evaluated. Initially, it was observed that regardless of the sample preparation for the swim-up (with SP or without SP), this method was not efficient in isolating motile sperm. Different PG concentrations were compared, revealing PG 45–90% recovered spermatozoa with better motility patterns compared to PG 35–70% and non-selected sperm. Then, PG 45–90% was compared to WC for sperm motility and heterologous IVF with swine oocytes. PG 45–90% showed better motility patterns, but the rate of fertilization and development of embryos were similar compared to WC. Therefore, PG 45–90% and WC are recommended for SP removal and sperm selection in the collared peccary. Methods for seminal plasma (SP) removal and the selection of collared peccary sperm for fertilization were compared. The experiments evaluated the following: the (I) impact of centrifugation for SP removal before swim-up for sperm selection and (II) a comparison of different Percoll® gradient densities (PG 45–90% and PG 35–70%). Non-selected sperm served as the control. Sperm quality was assessed based on motility patterns, morphology, membrane functional integrity, viability, reactive oxygen species (ROS), glutathione (GSH), and DNA integrity. Subsequently, the most successful group in the previous experiment and washing by centrifugation (WC) were compared for motility patterns and fertilization using pig oocytes. Swim-up decreased motility and enhanced ROS compared to the control. Centrifugation before swim-up harmed integrity and viability compared to the control. PG 45–90% (96.8 vs. 69.7 vs. 40.7 µm/s) allowed for a better velocity average pathway (VAP), a better velocity straight line, and better linearity (LIN) than those of the control and PG 35–70% (88.4 vs. 56.0 vs. 27.3 µm/s). Thus, PG 45–90% was used for fertilization. PG 45–90% obtained a higher VAP, a higher amplitude of the lateral head, straightness, and higher LIN than those of the control and WC. Cleavage (25.2–26.3%) and morula (8.1–10.5%) rates did not differ between the groups. The water resource is one of the main limiting factors for crop productivity. Poor distribution of rainfall is a serious problem in rainfed agriculture. The occurrence of periods of water deficit in the soil in the most sensitive phases of the crop can result in significant losses in productivity and in the use of nutrients. The use of Superabsorbent Polymers can be a strategy to minimize the risks of planting under water deficit conditions. Thus, the objective was to determine physiological responses and production components of common bean (Phaseolus vulgaris) in response to phosphate fertilization combined with doses of polymers in the presence or absence of water deficiency in the soil. The experiment was set up in a greenhouse in 150 dm3 pots with 17 bean plants in each pot. The treatments consisted of three factors that were combined in a factorial scheme: two water regimes (with and without water deficit), two doses of P (ideal and 1/3 ideal) and five doses of polymers (0, 2, 4, 8, 16% equivalent of the mass of the phosphate fertilizer for that of 120 kg ha-1 of P2O5). Dry matter was increased by 0.9% at the lowest dose of P mixed with 16% of polymer without water deficit and by 0,5% at the highest dose of P mixed with 5% of polymer without water deficit. P accumulation was increased by 16 and 19% at the highest and lowest dose of P mixed with 8,6 and 16% non-water deficit polymer, respectively. The photosynthetic rate and stomatal conductance were lower when the plants were subjected to water deficit. The concentration of proline in the leaf was higher at the lowest dose of P mixed with 8% polymer with and without water deficit, respectively. The number of pods per plant increased by 21 and 6% at the highest and lowest doses of P mixed with 8,9 and 5.46% of polymer without water deficit, respectively. The weight of one hundred grains showed a difference of 0,5% in the highest and lowest doses of P mixed with 2,41 and 16% of polymer without water deficit. Grain yield per pot was increased by 22 and 24% at the highest and lowest dose of P mixed with 13,7 and 16% of polymers without water deficit, respectively. The EUA increased by 21 and 24% at the highest and lowest dose of P mixed with 5,89 and 6,81% of polymer without water deficit, respectively. El recurso hídrico es uno de los principales factores limitantes para la productividad de los cultivos. La mala distribución de las precipitaciones es un problema grave en la agricultura de secano. La ocurrencia de períodos de déficit hídrico en el suelo en las fases más sensibles del cultivo puede resultar en pérdidas significativas en la productividad y en el aprovechamiento de los nutrientes. El uso de Polímeros Superabsorbentes puede ser una estrategia para minimizar los riesgos de plantar en condiciones de déficit hídrico. Así, el objetivo fue determinar las respuestas fisiológicas y componentes productivos del frijol común (Phaseolus vulgaris) en respuesta a la fertilización fosfatada combinada con dosis de polímeros en presencia o ausencia de deficiencia hídrica en el suelo. El experimento se montó en un invernadero en macetas de 150 dm3 con 17 plantas de frijol en cada maceta. Los tratamientos consistieron en tres factores que se combinaron en un esquema factorial: dos regímenes hídricos (con y sin déficit hídrico), dos dosis de P (ideal y 1/3 ideal) y cinco dosis de polímeros (0, 2, 4, 8, 16% equivalente de la masa del fertilizante fosfatado por el de 120 kg ha-1 de P2O5). La materia seca se incrementó en 0,9% a la dosis más baja de P mezclado con 16% de polímero sin déficit de agua y en 0,5% a la dosis más alta de P mezclado con 5% de polímero sin déficit de agua. La acumulación de P aumentó en un 16 y 19 % con la dosis más alta y más baja de P mezclado con 8,6 y 16 % de polímero sin déficit de agua, respectivamente. La tasa fotosintética y la conductancia estomática fueron menores cuando las plantas fueron sometidas a déficit hídrico. Seeds with high vigor result in better plant establishment and can therefore improve the use of nutrients, especially phosphorus (P). Thus, the objective was to evaluate the growth and P absorption by the common bean as a function of seed vigor in soils with contracting textures. The treatments were set up in a 2x5 factorial scheme, combining high and low seed vigor with five doses of P that were 0, 35, 70, 140 and 280 mg dm-3 in medium texture soil and 0, 50, 100, 200, 400 mg dm-3 in clayey soil. Four plants of the cultivar Dama were grown in pots containing 5 dm-3 of soil and evaluated until flowering began. High vigor seeds resulted in an increase in plant dry matter (shoot + root) of 11 and 15% in medium textured and clayey soils, respectively. Photosynthesis increased in plants originating from high vigor seeds by 26.04% in medium textured soil, but it was similar between high and low vigor seed plants in clayey soil. The dose of P necessary to reach at least 95% of maximum photosynthesis was 46% higher in plants originated from seeds with low vigor. The accumulation of P in plants of high vigor seeds cultivated in medium-textured soil was 37% greater than that of low vigor seeds. In clayey soil there was no such difference, however, the dose of P in the soil to reach the maximum accumulation was lower when the seeds were of high vigor. The P recovery rate was higher in plants originated from seeds with high vigor in both soils. It is concluded that plants from seeds with high vigor present greater growth and accumulation of P, which highlights the importance of using seeds with high vigor for a better use of phosphate fertilizers in common bean. Las semillas con alto vigor dan como resultado un mejor establecimiento de la planta y, por lo tanto, pueden mejorar el uso de nutrientes, especialmente fósforo (P). Así, el objetivo fue evaluar el crecimiento y absorción de P por el frijol común en función del vigor de la semilla en suelos con texturas contraídas. Los tratamientos se configuraron en un esquema factorial 2x5, combinando alto y bajo vigor de semilla con cinco dosis de P que fueron 0, 35, 70, 140 y 280 mg dm-3 en suelo de textura media y 0, 50, 100, 200, 400 mg dm-3 en suelo arcilloso. Se cultivaron cuatro plantas del cultivar Dama en macetas que contenían 5 dm3 de suelo y se evaluaron hasta que comenzó la floración. Las semillas de alto vigor dieron como resultado un aumento de la materia seca de la planta (brote + raíz) de 11 y 15% en suelos de textura media y arcillosos, respectivamente. La fotosíntesis aumentó en plantas originadas de semillas de alto vigor en un 26.04% en suelos de textura media, pero fue similar entre plantas de semillas de alto y bajo vigor en suelos arcillosos. La dosis de P necesaria para alcanzar al menos el 95% de la fotosíntesis máxima fue un 46% mayor en plantas originadas a partir de semillas de bajo vigor. La acumulación de P en plantas de semillas de alto vigor cultivadas en suelo de textura media fue 37% mayor que la de semillas de bajo vigor. En suelo arcilloso no hubo tal diferencia, sin embargo, la dosis de P en el suelo para alcanzar la acumulación máxima fue menor cuando las semillas fueron de alto vigor. La tasa de recuperación de P fue mayor en plantas provenientes de semillas de alto vigor en ambos suelos. Se concluye que las plantas provenientes de semillas con alto vigor presentan mayor crecimiento y acumulación de P, lo que resalta la importancia de utilizar semillas con alto vigor para un mejor aprovechamiento de fertilizantes fosfatados en frijol común. Desta forma, objetivou-se avaliar o crescimento e absorção de P pelo feijoeiro em função do vigor das sementes em solos de texturas contratantes. Os tratamentos foram montados em esquema fatorial 2x5, combinando alto e baixo vigor de semente com cinco doses de P que foram 0, 35, 70, 140 e 280 mg dm-3 em solo de textura média e de 0, 50, 100, 200, 400 mg dm-3 em solo argiloso. Foram cultivadas quatro plantas do cultivar Dama em vasos contendo 5 dm3 de solo e avaliadas até início de floração. Sementes de alto vigor resultaram em incremento da matéria seca da planta (parte aérea + raiz) de 11 e 15% nos solos de textura media e argiloso, respectivamente. A fotossíntese aumentou nas plantas originadas de sementes de alto vigor em 26,04% no solo de textura media, mas foi similar entre as plantas de sementes de alto e baixo vigor no solo argiloso. A dose de P necessária para alcance de pelo menos 95% da máxima fotossíntese foi 46% maior nas plantas originadas de sementes de baixo vigor. O acúmulo de P das plantas de sementes de alto vigor cultivadas em solo de textura média foi 37% maior do que as de baixo vigor. No solo argiloso não houve esta diferença, no entanto, a dose de P no solo para o alcance do máximo acúmulo foi menor quando as sementes foram de alto vigor. A taxa de recuperação do P foi maior em plantas originadas de sementes de alto vigor nos dois solos. Conclui-se que as plantas oriundas de sementes de alto vigor apresentam maior crescimento e acúmulo do P o que evidência a importância da utilização de sementes de alto vigor para o maior aproveitamento das adubações fosfatadas no feijoeiro. more...

The aim of this study was to evaluate the quality of table beet (Beta vulgaris) as a function of potassium source. The experiment was conducted at the Federal University of Viçosa - Campus Rio Paranaíba - MG. Two experiments were carried out in the summer and winter seasons and with the same experimental matrix. The sources studied were potassium chloride, potassium sulfate, and potassium nitrate. The doses of K were 60, 120, and 240 mg/dm³ of K. The additional treatment was the control without K application. The factors under study were combined in a factorial scheme (3 x 3) + 1 and distributed in blocks to the random with six repetitions. The quality was positively altered by fertilization with K, regardless of the applied sources. Fertilization with K resulting in higher acidity, better firmness, higher L* A and B color intensity, and a linear increase in sucrose. After storage, these variables were reduced in roots without K fertilization and kept when there was fertilization. The quality of roots (soluble solids, texture, and sucrose) of plants grown in winter was higher than in summer crops, with no significant influence of the K source. After being stored, the roots of plants grown in winter showed increased concentration of sucrose, color intensity (L*), acidity, and soluble solids as a function of doses. Thus, it is concluded that K sources do not have significant effects on quality at harvest and after storage in table beets. These variables are increased by potassium fertilization independently of the applied source. El objetivo de este estudio fue evaluar la calidad de la remolacha de mesa (Beta vulgaris) en función a fuentes de potasio. El experimento se realizó en la Universidad Federal de Viçosa - Campus Rio Paranaíba - MG. Se realizaron dos experimentos en las temporadas de verano e invierno y con la misma matriz experimental. Las fuentes estudiadas fueron cloruro de potasio, sulfato de potasio y nitrato de potasio. Las dosis de K fueron 60, 120 y 240 mg / dm³ de K. El tratamiento adicional fue el control sin aplicación de K. Los factores en estudio se combinaron en un esquema factorial (3 x 3) + 1 y se distribuyeron en bloques completos al azar con seis repeticiones. La calidad fue alterada positivamente con la fertilización con K, independientemente de las fuentes aplicadas. La fertilización con K resultó en mayor acidez, mejor firmeza, mayor intensidad de cromática L * A y B y aumento lineal de sacarosa. Después del almacenamiento, estas variables se redujeron en las raíces sin fertilización con K y se mantuvieron cuando hubo fertilización. La calidad de las raíces (sólidos solubles, textura y sacarosa) de las plantas cultivadas en invierno fue mayor que en los cultivos de verano, sin influencia significativa de la fuente de K. Después de ser almacenadas, las raíces de las plantas cultivadas en invierno mostraron una mayor concentración de sacarosa, intensidad de color (L *), acidez y sólidos solubles en función a las dosis. Por tanto, se concluye que las fuentes de K no tienen efectos significativos sobre la calidad en la cosecha y en el almacenamiento en la remolacha de mesa. Estas variables aumentan con la fertilización potásica independientemente de la fuente aplicada. Objetivou-se avaliar a qualidade da beterraba de mesa (Beta vulgaris) em função da fonte de potássio. O experimento foi conduzido na Universidade Federal de Viçosa - Campus Rio Paranaíba - MG. Foram conduzidos dois experimentos nas épocas de verão e de inverno e com a mesma matriz experimental. As fontes estudadas foram o cloreto de potássio, o sulfato de potássio e o nitrato de potássio. As doses de K foram de 60, 120 e de 240 mg/dm³ de K. O tratamento adicional foi a testemunha sem aplicação de K. Os fatores em estudo foram combinados em esquema fatorial (3 x 3) + 1 e distribuídos em blocos ao acaso com seis repetições. A qualidade foi positivamente alterada pela adubação com K, independente das fontes aplicadas. A adubação com K resultou em maior acidez, melhor firmeza, maior intensidade da cor L* A e B e aumento linear de sacarose. Após armazenamento essas variáveis foram reduzidas em raízes sem adubação com K e mantidas quando houve adubação. A qualidade das raízes (sólidos solúveis, textura e sacarose) das plantas cultivadas em inverno foi maior do que as de cultivo de verão, sem influência expressiva da fonte de K. Após serem armazenadas, as raízes das plantas cultivadas no inverno apresentaram aumento da concentração de sacarose, intensidade da cor (L*), acidez e sólidos solúveis em função de doses. Assim, conclui-se que as fontes de K não apresentam efeitos significativos na qualidade na colheita e após o armazenamento na beterraba. Faced with the perspective of developing narratives in digital didactic materials from gamebooks aimed at the educational context, the objective is to describe how the narratives produced by higher education students when building gamebooks using the Twine software show the way they think teaching and learning school content. The research is qualitatively based on a Case Study. Three gamebooks developed by students from a Public Higher Education Institution (PHEI) are analyzed. Data collection occurs from two instruments: preparation script and finalized gamebook. Through methodological triangulation, the data analysis compared the information obtained in the two instruments, plus the theoretical framework, supported by the Discursive Textual Analysis. It´s perceptive that in all the narrative structures, the school contents were not presented in an expository way; non-linear narratives generated activities that were close to a constructionist teaching proposal., Dada la perspectiva de desarrollar narrativas en materiales didácticos digitales a partir de libros juegos dirigidos al contexto educativo, el objetivo es describir cómo las narrativas desarrolladas por estudiantes de educación superior al construir libros juegos usando el software Twine muestran la forma en que piensan enseñar y aprender contenidos escolares. La investigación es cualitativa basada en un estudio de caso. Se analizan tres libros juegos desarrollados por estudiantes de una Institución Pública de Educación Superior (IPES). La recopilación de datos se produce a partir de dos instrumentos: guion de preparación y libro juego finalizado. El análisis de datos, mediante triangulación metodológica al comparar la información obtenida en los dos instrumentos, más el marco teórico, sustentado en el Análisis Textual Discursivo.
