We discuss a novel concept of a polarized nuclear target for accelerator fixed-target scattering experiments, which is based on parahydrogen induced polarization (PHIP). One may be able to reach a 33% free-proton polarization in the ethane molecule. The potential advantages of such a target include operation at zero magnetic field, fast ($\sim$100 Hz) polarization reversal, and operation with large intensity of an electron beam., Comment: 16 pages, 2 figures

Congenital heart defects (CHDs) are the most common birth defect in human with an incidence of almost 1% of all live births. Most cases have a multifactorial origin with both genetics and the environment playing a role in its development and progression. Adding an epigenetic component to this aspect is exemplified by monozygotic twins which share the same genetic background but have a different disease status. As a result, the interplay between the genetic, epigenetic and the environmental conditions might contribute to the etiology and phenotype. To date, the underlying causes of the majority of CHDs remain poorly understood. In this study, we performed genome-wide high-throughput sequencing to examine the genetic, structural genomic and epigenetic differences of two identical twin pairs discordant for Tetralogy of Fallot (TOF), representing the most common cyanotic form of CHDs. Our results show the almost identical genetic and structural genomic identity of the twins. In contrast, several epigenetic alterations could be observed given by DNA methylation changes in regulatory regions of known cardiac-relevant genes. Overall, this study provides first insights into the impact of genetic and especially epigenetic factors underlying monozygotic twins discordant for CHD like TOF.

- Abstract
Light is necessary for vision; it enables us to sense and perceive our surroundings and in many direct and indirect ways, via eye and skin, affects our physiological and psychological health. The use of light in built environments has comfort, behavioural, economic and environmental consequences. Daylight has many particular benefits including excellent visual performance, permitting good eyesight, effective entrainment of the circadian system as well as a number of acute non-image forming effects and the important role of vitamin D production. Some human responses to daylight seem to be well defined whilst others require more research to be adequately understood. This paper presents an overview of current knowledge on how the characteristics of daylight play a role in fulfilling these and other functions often better than electric lighting as conventionally delivered.

MicroRNAs (miRNAs) play an important role in guiding development and maintaining function of the human heart. Dysregulation of miRNAs has been linked to various congenital heart diseases including Tetralogy of Fallot (TOF), which represents the most common cyanotic heart malformation in humans. Several studies have identified dysregulated miRNAs in right ventricular (RV) tissues of TOF patients. In this study, we profiled genome-wide the whole transcriptome and analyzed the relationship of miRNAs and mRNAs of RV tissues of a homogeneous group of 22 non-syndromic TOF patients. Observed profiles were compared to profiles obtained from right and left ventricular tissue of normal hearts. To reduce the commonly observed large list of predicted target genes of dysregulated miRNAs, we applied a stringent target prediction pipeline integrating probabilities for miRNA-mRNA interaction. The final list of disease-related miRNA-mRNA pairs comprises novel as well as known miRNAs including miR-1 and miR-133, which are essential to cardiac development and function by regulating KCNJ2, FBN2, SLC38A3 and TNNI1. Overall, our study provides additional insights into post-transcriptional gene regulation of malformed hearts of TOF patients.

BACKGROUND: In a randomized, controlled trial that compared liraglutide, a glucagon-like peptide 1 analogue, with placebo in patients with type 2 diabetes and high cardiovascular risk who were receiving usual care, we found that liraglutide resulted in lower risks of the primary end point (nonfatal myocardial infarction, nonfatal stroke, or death from cardiovascular causes) and death. However, the long-term effects of liraglutide on renal outcomes in patients with type 2 diabetes are unknown. METHODS: We report the prespecified secondary renal outcomes of that randomized, controlled trial in which patients were assigned to receive liraglutide or placebo. The secondary renal outcome was a composite of new-onset persistent macroalbuminuria, persistent doubling of the serum creatinine level, end-stage renal disease, or death due to renal disease. The risk of renal outcomes was determined with the use of time-to-event analyses with an intention-to-treat approach. Changes in the estimated glomerular filtration rate and albuminuria were also analyzed. RESULTS: A total of 9340 patients underwent randomization, and the median follow-up of the patients was 3.84 years. The renal outcome occurred in fewer participants in the liraglutide group than in the placebo group (268 of 4668 patients vs. 337 of 4672; hazard ratio, 0.78; 95% confidence interval [CI], 0.67 to 0.92; P=0.003). This result was driven primarily by the new onset of persistent macroalbuminuria, which occurred in fewer participants in the liraglutide group than in the placebo group (161 vs. 215 patients; hazard ratio, 0.74; 95% CI, 0.60 to 0.91; P=0.004). The rates of renal adverse events were similar in the liraglutide group and the placebo group (15.1 events and 16.5 events per 1000 patient-years), including the rate of acute kidney injury (7.1 and 6.2 events per 1000 patient-years, respectively). CONCLUSIONS: This prespecified secondary analysis shows that, when added to usual care, liraglutide resulted in lower rates of the development and progression of diabetic kidney disease than placebo. Light is necessary for vision; it enables us to sense and perceive our surroundings and in many direct and indirect ways, via eye and skin, affects our physiological and psychological health. The use of light in built environments has comfort, behavioural, economic and environmental consequences. Daylight has many particular benefits including excellent visual performance, permitting good eyesight, effective entrainment of the circadian system as well as a number of acute non-image forming effects and the important role of vitamin D production. Some human responses to daylight seem to be well defined whilst others require more research to be adequately understood. This paper presents an overview of current knowledge on how the characteristics of daylight play a role in fulfilling these and other functions often better than electric lighting as conventionally delivered. - Abstract
Hintergrund: Mehr als die Hälfte aller angeborenen non-syndromalen Schwerhörigkeiten (NSHL) haben genetische Ursachen mit einer Heterogenität von über 70 bekannten Genen. Aufgrund finanzieller und zeitlicher Beschränkungen wurde bisher in der Regel lediglich das Gen GJB2 (Connexin 26) untersucht, welches in etwa die Hälfte der genetisch bedingten NSHL Fälle erklären kann. Eine umfassende molekulargenetische Diagnostik bei den verbleibenden Fällen war nicht möglich. Mit der Hochdurchsatzsequenzierung (next-generation sequencing; NGS) kann jetzt gezielt und gleichzeitig eine Liste von bekannten Schwerhörigkeitsgenen analysiert werden (gene panel approach).Material und Methoden: 5 Kinder mit angeborener, non-syndromaler Schallempfindungsschwerhörigkeit ohne Mutationen im Gen GJB2 und GJB6 und deren Eltern wurden zunächst einer umfassenden Phänotypisierung unterzogen. Besonderes Augenmerk wurde auf die Abklärung exogener Risikofaktoren (Infektionen, Hypoxie, Medikamente) gerichtet. Durch spezifische Anreicherung (SureSelect) von 95 Schwerhörigkeitsgenen und NGS (Illumina) wurden Kandidatenmutationen für jedes Trio analysiert.Ergebnisse: Die gezielte und gleichzeitige Hochdurchsatzsequenzierung in Form eines Diagnostik-Panels von 95 Schwerhörigkeitsgenen wurde in die Routine-Diagnostik eingeführt. Bei allen untersuchten Patienten konnten seltene nichtsynonyme Genvarianten entdeckt werden. Ein monogenetischer Erbgang (z.B. aut.-rez.) oder ein sicher krankheitsverursachendes Gen konnte in keinem der Fälle nachgewiesen werden.Diskussion: Mittels simultaner Hochdurchsatzsequenzierung ist eine umfassende molekulargenetische Diagnostik bei non-syndromaler Schwerhörigkeit in der Routine möglich. Die Hauptaufgabe wird zukünftig die Klärung der Frage sein, ob eine gefundene genetische Auffälligkeit Krankheit oder Variation ohne Krankheitswert bedeutet. Dazu können u.a. eine exakte Phänotypisierung, umfangreiche Datenbanken mit Allele Frequenzen, Stammbäume mit vielen Betroffenen

We report the results of experiments leading to the production of an image of a polarized [sup 129]Xe sample prepared by spin exchange with Cs, optically pumped with a spectrally narrowed 894.3 nm diode laser. Representative images of the average electron spin polarization are shown. Appreciable cesium electron polarization values were achieved, and a nuclear polarization of about 2.5% was measured for [sup 129]Xe. The absolute nuclear polarization was measured by water-calibrated free induction decay of the nuclear magnetic resonance signal, and the polarized xenon imaged using a 2 T magnetic resonance imaging system. © 1998 American Institute of Physics. Abstract: In this article we report the longitudinal relaxation times (T 1) of various 6Li salts (6LiI, 6LiCl and 6LiNO3) in D2O and H2O, measured in low magnetic fields (B 0 =3.5mT). This investigation serves the purpose of clarifying the relaxation behavior of different 6Li solutions and different concentrations. The measurement were undertaken to establish a framework for future applications of hyperpolarized 6Li in medical imaging, biological studies and investigations of lithium ion batteries. Time will pass during the transport of hyperpolarized lithium ions to the sample, which leads to a polarization loss. In order to store polarization as long as possible, it is necessary to examine which 6Li salt solution has the longest relaxation time T 1. Longitudinal relaxation times of 6Li salts in D2O and H2O were investigated as a function of concentration and the most extended T 1 was found for 6LiI in D2O and H2O. In agreement with the theory the relaxation time T 1 of all 6Li salts increase with decreasing concentration. In the case of 6LiI in H2O an inverse behavior was observed. We assume that the prolonged T 1 times occur due to formation of 6LiOH upon the solution of 6LiI in H2O, which settles as a precipitate. By diluting the solution, the precipitate continuously dissolves and approaches T 1 of 6LiOH (T 1 ∼28s), leading to a shorter T 1 relaxation time. [Copyright &y& Elsevier]
