The female reproductive tract (female-RT) must decipher the repertoire of molecular cues received from the male during copulation in order to activate and coordinate tract functionality necessary for high fertility. In Drosophila, this modulation is partially driven by spermathecal secretory cells (SSC). The SSC are a layer of cuboidal secretory glandular cells surrounding the spermatheca capsule where sperm is stored. It is unclear, however, how the SSC regulate the system's activity. Here we show that mating activates the secretory machinery of the SSC. The SSC release a heterogeneous population of extracellular vesicles (EVs) which is involved in initiating and managing the increase in egg-laying, and possibly sperm storage. Moreover, sperm and male accessory gland proteins are essential for such mating-mediated SSC activity. Thus, mating regulates secretory/endocytic pathways required for trafficking of vesicles to SSC-female-RT target sites, which modulate and coordinate reproductive tract activity to achieve high fertility., (© 2022. Purpose: To determine if the timing of mitomycin C (MMC) application during pterygium surgery affects endothelial cell loss., Methods: A retrospective, nonrandomized, case-series design was used. The study group included 40 patients who underwent pterygium surgery with the bare sclera technique. MMC 0.02% was applied intraoperatively for 5 minutes, either before (16 patients) or after (24 patients) excision of the head of the pterygium. Endothelial images were acquired at the center of the cornea with a specular microscope before surgery and at 3 intervals during follow-up (average of 3 measurements each). Differences in cell loss from baseline were analyzed by paired t test., Results: In the group in which MMC was applied post excision, mean preoperative endothelial cell count was 2254 +/- 128 cells per square millimeter. Mean postoperative values were 1775 +/- 63 cells per square millimeter at 1 week (percentage cell loss, 21.25% +/- 2.8%), 1707 +/- 41 cells per square millimeter at 1 month (24.26% +/- 1.8%), and 1780 +/- 72 cells per square millimeter at 3 months (21.05% +/- 3.2%). The difference in cell count from the preoperative value was significant at all time points (P < 0.02). In the group in which MMC was applied before excision, mean preoperative endothelial cell count was 2352 +/- 118 cells per square millimeter, and mean postoperative values were 2345 +/- 120 cell per square millimeter at 1 week, 2260 +/- 162 cells per square millimeter at 1 month, and 2230 +/- 144 cells per square millimeter at 3 months. No endothelial cell loss was documented in this group., Conclusion: The endothelial cell loss associated with the use of MMC during pterygium surgery might be avoided by applying MMC before excision of the pterygium head, so that it does not come into contact with the corneal surface.

NRL, a bZIP transcription factor of the Maf subfamily, interacts with the homeodomain protein CRX and synergistically regulates rhodopsin expression. Here we report that six isoforms of NRL (29-35 kDa) are generated by phosphorylation and expressed specifically in the mammalian retina. The anti-NRL antibody also cross-reacts with a cytosolic 45-kDa protein, which is detected in neuronal tissues but is not encoded by the NRL gene. In both human retinal cell cultures and sections of fetal and adult human retina, NRL is present in the nuclei of developing and mature rods but not cones. We propose that NRL regulates rod photoreceptor-specific gene expression and is involved in rod differentiation.

Objective: To investigate the coordination among hand movement, ground forces, and muscle activity in standing stroke patients reaching forward and lifting an object from a table., Design: Survey., Setting: Research laboratory., Patients: Eight stroke patients and 8 persons serving as controls., Main Outcome Measure: Symmetry of percentage of body weight (BW) during initial standing, velocity and hand path trajectory, ankle muscle electromyography. Temporal and spatial parameters in percentage of movement time were recorded by using 2 forceplates, 3-dimensional kinematics, and surface electromyography. Motor function, sensory function, and functional performance were also assessed., Results: Weight distribution during initial standing was significantly higher (57.4% +/- 8.1% BW) on the nonparetic leg. All subjects had preserved the preparatory loading phase, and after onset of hand movement loading shifted to the contralateral leg. Ankle muscle activity onset (lateral gastrocnemius [LG]) occurred after loading. In stroke subjects, LG was consistently activated first in the nonparetic leg, regardless of which arm performed the task. During paretic hand task, the reaching phase was significantly longer and the lifting phase significantly shorter compared with that of the nonparetic hand task and with that of the controls. In the paretic task, the hand path velocity was not bell-shaped; the object lifting was., Conclusions: Stroke subjects preserve the coordination between ground forces and hand movement. The lack of spontaneous use of the paretic hand is primarily caused by difficulties of planning the hand trajectory in space, as reflected by temporal and spatial parameters during task performance.

Normal as well as neoplastic cells traverse extracellular matrix barriers by mobilizing proteolytic enzymes in response to epidermal growth factor (EGF)-EGF receptor (EGFR) or hepatocyte growth factor/scatter factor (SF)-c-Met interactions. The plasminogen activator-plasminogen axis has been proposed to play a key role during cell invasion, but the normal development of plasminogen activator- as well as that of plasminogen-deficient mice supports the existence of alternate proteolytic systems that permit cells to traverse extracellular matrix barriers. To characterize the role that matrix-degrading proteinases play in EGF- or SF-stimulated invasion, a human squamous carcinoma cell line (UM-SCC-1) was triggered atop the matrices of type I collagen or human dermal explants in a three-dimensional culture system. During EGF- or SF-induced invasion, UM-SCC-1 cells expressed urokinase-type plasminogen activator (uPA) and uPA receptor as well as the matrix metalloproteinases (MMPs), membrane-type MMP-1, collagenase 1, stromelysin 1, and gelatinase B. Despite the presence of a positive correlation between uPA receptor-uPA expression and growth factor-stimulated invasion, UM-SCC-1 invasion was not affected by inhibitors directed against the plasminogen activator-plasminogen axis. In contrast, both recombinant and synthetic MMP inhibitors completely suppressed invasion by either EGF- or SF-stimulated cells without affecting either proteinase expression or cell motility across collagen-coated surfaces. These data demonstrate that MMPs, but not the plasminogen activator-plasmin system, can directly regulate the ability of either EGF- or SF-stimulated tumor cells to invade interstitial matrix barriers.

During angiogenesis, endothelial cells penetrate fibrin barriers via undefined proteolytic mechanisms. We demonstrate that the fibrinolytic plasminogen activator (PA)-plasminogen system is not required for this process, since tissues isolated from PA- or plasminogen-deficient mice successfully neovascularize fibrin gels. By contrast, neovessel formation, in vitro and in vivo, is dependent on fibrinolytic, endothelial cell-derived matrix metalloproteinases (MMP). MMPs directly regulate this process as invasion-incompetent cells penetrate fibrin barriers when transfected with the most potent fibrinolytic metalloproteinase identified in endothelium, membrane type-1 MMP (MT1-MMP). Membrane display of MT1-MMP is required, as invasion-incompetent cells expressing a fibrinolytically active, transmembrane-deleted form of MT1-MMP remain noninvasive. These observations identify a PA-independent fibrinolytic pathway wherein tethered MMPs function as pericellular fibrinolysins during the neovascularization process.

Oncogenesis is a process resulting from genetic events which cause loss of growth control or inhibition of appropriate cell death. The Bcl-XL protein is a recently discovered member of the bcl-2 family which has been shown to protect cells from some forms of programmed cell death, but has not yet been implicated in the genesis of human carcinomas. In this report we explore the role of Bcl-XL overexpression in protecting cancer cells from p53-mediated apoptosis. Increased levels of Bcl-XL were found in a subset of primary human breast carcinomas, as well as in the breast cancer line, T47D. T47D cells were then transfected with a temperature-sensitive mutant of the tumor suppressor p53 (p53ts). Although many tumor cell lines undergo apoptosis when p53 is expressed, the T47D transfectants remained viable at temperatures permitting wild-type p53 phenotype. This suggested that endogenous Bcl-XL could protect cancer cells from p53-mediated apoptosis. To test this hypothesis, murine erythroleukemia cells were transfected with bcl-XL and p53ts. While cell lines expressing p53 alone rapidly died, those cells co-expressing Bcl-XL survived. These results demonstrate that Bcl-XL is capable of protecting cells from p53-mediated apoptosis, and suggest a possible mechanism by which tumors expressing Bcl-XL are able to partly overcome the tumor suppressor functions of p53.

When an object held by a precision gripis subjected to an abrupt vertical load perturbation, somatosensory input from the digits triggers an increase in grip force to restore an adequate safety margin, preventing frictional slips. In adults the response occurs after a latency of 60-80 ms. In the present study, children from 2 years old upward and adults grasped and lifted an object using a precision grip. Sudden, unpredicted increases in load force (tangential to the grip surfaces) were induced by the experimenter by dropping a small disc on to a receptacle attached to the object. The impact elicited a grip force response which in young children had a longer latency and a smaller amplitude than was seen in adults. The grip response latency gradually become shorter and its amplitude increased with increasing age, reaching adult values at 6-10 years. The muscle activity underlying the response could have several bursts. The adults showed one brisk response, appearing 40-50 ms after impact, in extrinsic and intrinsic hand muscles, while younger children also exhibited a short-latency burst, appearing about 20 ms after impact. It is suggested that the short-latency response was mediated via spinal pathways, and that these pathways are disengaged by supraspinal centers during development. In a predictable loading situation, when subjects dropped the disc themselves into the receptable using the contralateral hand, they changed strategy. Adults induced a well-timed anticipatory grip force increase prior to the impact that was scaled to the weight of the object. The youngest children did not time the force increase properly in relation to the impact. Yet, they could scale their anticipatory grip force increase with respect to the weight of the dropped disc. This suggests a well-developed capacity to use information about the weight of objects held by one hand to parameterize a programmed force output to the other hand.

The gene coxI, encoding subunit I of mitochondrial cytochrome c oxidase, has been characterized from the normal (fertile) and Ogura (male-sterile) cytoplasms of radish to determine if a previously identified mitochondrial DNA rearrangement, and its associated transcriptional differences, could play a role in Ogura cytoplasmic male sterility (CMS). The normal and Ogura loci are virtually identical for 2.8 kb, including a 527-codon open reading frame whose product is approximately 95% identical to other plant COXI polypeptides. A rearrangement 120 bp 5' to the coding region results in different 5' transcript termini for the two genes. A comparison of several crucifer mitochondrial DNAs indicates that this rearrangement also occurs in the normal radish cytoplasm and is, therefore, not involved in Ogura CMS. Sequences present at the coxI locus belong to at least two different dispersed repeat families, members of which are also associated with other rearranged genes in radish.

We have previously shown that the mitochondrial gene atpA, encoding the alpha subunit of F1 ATP synthase, is associated with DNA rearrangements and nuclear-specific transcript patterns in the male-sterile cytoplasm of Ogura radish. Here we present a detailed characterization of this gene from both the normal (fertile) and Ogura (male-sterile) cytoplasms of radish to determine if it is involved in Ogura cytoplasmic male sterility. The normal and Ogura radish atpA loci are virtually identical for 3.8 kb, including a 507 codon open reading frame whose product is approximately 92% identical to other plant ATPA polypeptides. Rearrangement breakpoints have been identified 613 bp 5' and 1663 bp 3' to the atpA coding region. The 5' rearrangement breakpoint is located within a repeated sequence that has been associated with other rearrangement events in radish mitochondria. The previously identified transcript difference results from transcription originating upstream of this rearrangement site. Although the presence of this transcript is affected by nuclear background, analyses in several different sterile and fertile nuclear backgrounds indicate that the presence of this transcript is not strictly correlated with male sterility. In addition, normal levels of ATPA polypeptide are present in sterile plants containing the Ogura cytoplasm.

This article is an introduction into the West German Hooligan Scene to learn more about the social, sociological, psychological, criminal backgrounds, characters and causes for a better understanding to solve this multifunctional problem: by police, society and policy.

Campylobacter pylori strains were isolated and serum samples were obtained from 63 patients. Immunoblots of 52 patients sera using their own isolates as antigen showed a 120 kDa band, which was missing in the other 11 isolates and the respective sera. This band was not detected in other Campylobacter species. Effects of trypsin treatment of bacteria and absorption of sera by live organisms suggest a C. pylori-specific surface protein.
