We study the dynamics of multimode polariton lasing in organic microcavities by using a second-order cumulant equation approach. By inspecting the time evolution of the photon mode occupations, we show that if multiple lasing peaks are observed in time-integrated mode occupations, the reason can be either bi-modal lasing or temporal switching between several modes. The former takes place within a narrow range of parameters while the latter occurs more widely. We find that the origin of the temporal switching is different in the weak- and strong-coupling regimes. At weak coupling slope efficiency is the determining factor, while for strong coupling it is changes in the eigenmodes and gain spectrum upon pumping. This difference is revealed by investigating the photoluminescence and momentum-resolved gain spectra. Our results underscore the importance of understanding the time evolution of the populations when characterizing the lasing behaviour of a multimode polariton system, and show how these features differ between weak and strong coupling., Comment: 15 pages, 13 figures

Bosonic condensates offer exciting prospects for studies of non-equilibrium quantum dynamics. Understanding the dynamics is particularly challenging in the sub-picosecond timescales typical for room temperature luminous driven-dissipative condensates. Here we combine a lattice of plasmonic nanoparticles with dye molecule solution at the strong coupling regime, and pump the molecules optically. The emitted light reveals three distinct regimes: one-dimensional lasing, incomplete stimulated thermalization, and two-dimensional multimode condensation. The condensate is achieved by matching the thermalization rate with the lattice size and occurs only for pump pulse durations below a critical value. Protein maturation in the endoplasmic reticulum (ER) depends on a fine balance between oxidative protein folding and quality control mechanisms, which together ensure high-capacity export of properly folded proteins from the ER. Oxidative protein folding needs to be regulated to avoid hyperoxidation. The folding capacity of the ER is regulated by the unfolded protein response (UPR) and ER-associated degradation (ERAD). The UPR is triggered by unfolded protein stress and leads to up-regulation of cellular components such as chaperones and folding catalysts. These components relieve stress by increasing folding capacity and up-regulating ERAD components that remove non-native proteins. Although oxidative protein folding and the UPR/ERAD pathways each are well-understood, very little is known about any direct cross-talk between them. In this study, we carried out comprehensive in vitro activity and binding assays, indicating that the oxidative protein folding relay formed by ER oxidoreductin 1 (Ero1), and protein disulfide-isomerase can be inactivated by a feedback inhibition mechanism involving unfolded proteins and folding intermediates when their levels exceed the folding capacity of the system. This mechanism allows client proteins to remain mainly in the reduced state and thereby minimizes potential futile oxidation–reduction cycles and may also enhance ERAD, which requires reduced protein substrates. Relief from excess levels of non-native proteins by increasing the levels of folding factors removed the feedback inhibition. These results reveal regulatory cross-talk between the oxidative protein folding and UPR and ERAD pathways.

openaire: EC/H2020/745115/EU//OPLD Organic light-emitting diodes (OLEDs) have been established as versatile light sources that allow for easy integration in large-area surfaces and flexible substrates. In addition, the low fabrication cost of OLEDs renders them particularly attractive as general lighting sources. Current methods for the fabrication of white-light OLEDs rely on the combination of multiple organic emitters and/or the incorporation of multiple cavity modes in a thick active medium. These architectures introduce formidable challenges in both device design and performance improvements, namely, the decrease of efficiency with increasing brightness (efficiency roll-off) and short operational lifetime. Here we demonstrate, for the first time, white-light generation in an OLED consisting of a sub-100 nm thick blue single-emissive layer coupled to the photonic Bragg modes of a dielectric distributed Bragg reflector (DBR). We show that the Bragg modes, although primarily located inside the DBR stack, can significantly overlap with the emissive layer, thus efficiently enhancing emission and outcoupling of photons at selected wavelengths across the entire visible light spectrum. Moreover, we show that color temperature can be tuned by the DBR parameters, offering great versatility in the optimization of white-light emission spectra.

The major cat allergen Fel d 1 is one of the most common and potent causes of animal related allergy. Medical treatment of cat allergy has relied on immunotherapy carried out with cat dander extract. This approach has been problematic, mainly due to inconsistent levels of the major allergen in the produced extracts. Recombinant DNA technology has been proposed as an alternative method to produce more consistent pharmaceuticals for immunotherapy and diagnostics of allergy. Current approaches to produce recombinant Fel d 1 (recFel d 1) in the cytoplasm of Escherichia coli have however resulted in protein folding deficiencies and insoluble inclusion body formation, requiring elaborate in vitro processing to acquire folded material. In this study, we introduce an efficient method for cytoplasmic production of recFel d 1 that utilizes eukaryotic folding factors to aid recFel d 1 to fold and be produced in the soluble fraction of E. coli. The solubly expressed recFel d 1 is shown by biophysical in vitro experiments to contain structural disulfides, is extremely stable, and has a sensitivity for methionine sulfoxidation. The latter is discussed in the context of functional relevance.

We report on reversible all-optical emission control and lasing in plasmonic nanoparticle lattices. By incorporating photochromic molecules into the liquid gain medium composed of organic fluorescent molecules, we realize all-optical control over gain and absorption, the two key parameters associated with both conventional and nanoscale lasing. We demonstrate reversible photoswitching between two distinct modes of operation: (1) spontaneous emission to the lattice mode, characterized by broad emission line width, low emission intensity, and large angular distribution; and (2) lasing action, characterized by very narrow (sub-nm) line widths due to the emergence of increased gain and temporal coherence in the system, approximately 3 orders of magnitude increase in emission intensity, and narrow 0.7° angular divergence of the beam. A rate-equation model is employed to describe the operation of the switchable plasmonic laser. Our results provide the first demonstration of optically tunable losses in plasmonic lattice lasers, which is an important milestone for the development of active plasmonics and paves the way for ultrafast all-optical switching of plasmonic nanolasers. acceptedVersion

The folding of disulfide bond containing proteins in the endoplasmic reticulum (ER) is a complex process that requires protein folding factors, some of which are protein-specific. The ER resident saposin-like protein pERp1 (MZB1, CNPY5) is crucial for the correct folding of IgA, IgM and integrins. pERp1 also plays a role in ER calcium homeostasis and plasma cell mobility. As an important factor for proper IgM maturation and hence immune function, pERp1 is upregulated in many auto-immune diseases. This makes it a potential therapeutic target. pERp1 belongs to the CNPY family of ER resident saposin-like proteins. To date, five of these proteins have been identified. All are implicated in protein folding and all contain a saposin-like domain. All previously structurally characterized saposins are involved in lipid binding. However, there are no reports of CNPY family members interacting with lipids, suggesting a novel function for the saposin fold. However, the molecular mechanisms of their function remain elusive. To date, no structure of any CNPY protein has been reported. Here, we present the high-resolution (1.4 A) crystal structure of human pERp1 and confirm that it has a saposin-fold with unique structural elements not present in other saposin-fold structures. The implications for the role of CNPY proteins in protein folding in the ER are discussed.

We present a beyond-mean-field approach to predict the nature of organic polariton lasing, accounting for all relevant photon modes in a planar microcavity. Starting from a microscopic picture, we show how lasing can switch between polaritonic states resonant with the maximal gain, and those at the bottom of the polariton dispersion. We show how the population of non-lasing modes can be found, and by using two-time correlations, we show how the photoluminescence spectrum (of both lasing and non-lasing modes) evolves with pumping and coupling strength, confirming recent experimental work on the origin of blueshift for polariton lasing., 6 pages, 4 figures, plus supplemental material

Polarization textures of light may reflect fundamental phenomena such as topological defects, and can be utilized in engineering light beams. Three main routes are applied during their creation: spontaneous appearance in phase transitions, steering by an excitation beam, or structural engineering of the medium. We present an approach that uses all three in a platform offering advantages that are not simultaneously provided in any previous system: advanced structural engineering, strong-coupling condensate with effective photonic interactions, as well as room temperature and sub-picosecond operation. We demonstrate domain wall polarization textures in a plasmonic lattice Bose-Einstein condensate, by combining the dipole structure of the lattice with a non-trivial condensate phase revealed by phase retrieval. Candida antarctica lipase B (CalB) is a very efficient catalyst and is used in a wide range of industries from food flavour to pharmaceutical, and biodiesel manufacturing. It has a high degree of enantioselective and regioselective substrate specificity and is stable over a wide range of biophysical conditions including pH, temperature and solvent conditions. High-level expression of biologically active wild-type CalB has been problematic, partly due to folding events. Consequently, focus has been on modified CalB, which has allowed orders of magnitude increase in yields of protein. However, these modifications alter the quaternary structure of the protein. Here we produce soluble wild-type CalB in high yields in the cytoplasm of E.coli using a catalyzed system for cytoplasmic disulfide bond formation both in shake flasks and in fermentation in chemically defined media. The CalB produced had the expected stereospecific activity and had a higher activity than CalB from a commercial source.

openaire: EC/H2020/745115/EU//OPLD Bose–Einstein condensation is a remarkable manifestation of quantum statistics and macroscopic quantum coherence. Superconductivity and superfluidity have their origin in Bose–Einstein condensation. Ultracold quantum gases have provided condensates close to the original ideas of Bose and Einstein, while condensation of polaritons and magnons has introduced novel concepts of non-equilibrium condensation. Here, we demonstrate a Bose–Einstein condensate of surface plasmon polaritons in lattice modes of a metal nanoparticle array. Interaction of the nanoscale-confined surface plasmons with a room-temperature bath of dye molecules enables thermalization and condensation in picoseconds. The ultrafast thermalization and condensation dynamics are revealed by an experiment that exploits thermalization under propagation and the open-cavity character of the system. A crossover from a Bose–Einstein condensate to usual lasing is realized by tailoring the band structure. This new condensate of surface plasmon lattice excitations has promise for future technologies due to its ultrafast, room-temperature and on-chip nature.

I review our recent findings on lasing / condensation in plasmonic nanoparticle lattices1-5. The system properties can be tailored with high precision, including the lasing / condensation energies, linewidths, as well as the dimensionality of the feedback. For a 2-dimensional (2-D) square lattice, we identify lasing in the bright and the dark mode of the system1. By reducing the dimensionality to 1-D we observe the dark mode lasing2. In broken symmetry 2-dimensional rectangular lattices, we observe multimode lasing3. In honeycomb lattices with hexagonal symmetry, we observe 6 beams with specific off-normal angles and polarization properties corresponding to six-fold symmetry of such a lattice4. Finally, I review our recent studies in plasmonic Bose-Einstein condensation in plasmonic lattices5.

Display Omitted Automatic analysis of human spontaneous behavior has attracted increasing attention in recent years from researchers in computer vision. This paper proposes an approach for multi-modal video-induced emotion recognition, based on facial expression and electroencephalogram (EEG) technologies. Spontaneous facial expression is utilized as an external channel. A new feature, formed by percentage of nine facial expressions, is proposed for analyzing the valence and arousal classes. Furthermore, EEG is used as an internal channel supplementing facial expressions for more reliable emotion recognition. Discriminative spectral power and spectral power difference features are exploited for EEG analysis. Finally, these two channels are fused on feature-level and decision-level for multi-modal emotion recognition. Experiments are conducted on MAHNOB-HCI database, including 522 spontaneous facial expression videos and EEG signals from 27 participants. Moreover, human perception in emotion recognition compared to the proposed approach is tested with 10 volunteers. The experimental results and comparisons with the average human performance show the effectiveness of the proposed multi-modal approach.

Strong coupling between quantum emitters and surface plasmon polariton modes in metal nanostructures has been extensively studied in recent years. A natural direction of research and a prerequisite for many applications is the possibility of external, in situ manipulation of the strength of the coupling. We review research on active control of surface plasmon–emitter strong coupling phenomena. Active control has been demonstrated for a variety of systems, such as metal nanohole arrays, nanoparticles, rods and dimers, combined with photochromic molecules, J-aggregates, and monolayers of MoS2, WS2, and WSe2. We discuss work on optical switching realized by changing photochromic molecules by UV and visible light between forms that couple strongly or weakly with the electromagnetic field mode of the plasmonic nanostructure. Other forms of optical control such as use of polarization and phase are briefly covered. We review research on electrical, thermal, and chemical active control of light–matter coupling in plasmonic systems.

Lasing at the nanometre scale promises strong light-matter interactions and ultrafast operation. Plasmonic resonances supported by metallic nanoparticles have extremely small mode volumes and high field enhancements, making them an ideal platform for studying nanoscale lasing. At visible frequencies, however, the applicability of plasmon resonances is limited due to strong ohmic and radiative losses. Intriguingly, plasmonic nanoparticle arrays support non-radiative dark modes that offer longer life-times but are inaccessible to far-field radiation. Here, we show lasing both in dark and bright modes of an array of silver nanoparticles combined with optically pumped dye molecules. Linewidths of 0.2 nanometers at visible wavelengths and room temperature are observed. Access to the dark modes is provided by a coherent out-coupling mechanism based on the finite size of the array. The results open a route to utilize all modes of plasmonic lattices, also the high-Q ones, for studies of strong light-matter interactions, condensation and photon fluids., Comment: Accepted for publication in Nature Communications

Micro-expressions (MEs) are rapid, involuntary facial expressions which reveal emotions that people do not intend to show. Studying MEs is valuable as recognizing them has many important applications, particularly in forensic science and psychotherapy. However, analyzing spontaneous MEs is very challenging due to their short duration and low intensity. Automatic ME analysis includes two tasks: ME spotting and ME recognition. For ME spotting, previous studies have focused on posed rather than spontaneous videos. For ME recognition, the performance of previous studies is low. To address these challenges, we make the following contributions: (i) We propose the first method for spotting spontaneous MEs in long videos (by exploiting feature difference contrast). This method is training free and works on arbitrary unseen videos. (ii) We present an advanced ME recognition framework, which outperforms previous work by a large margin on two challenging spontaneous ME databases (SMIC and CASMEII). (iii) We propose the first automatic ME analysis system (MESR), which can spot and recognize MEs from spontaneous video data. Finally, we show our method outperforms humans in the ME recognition task by a large margin, and achieves comparable performance to humans at the very challenging task of spotting and then recognizing spontaneous MEs.

This study reveals novel regulatory mechanisms for human Ero1α and Ero1β, including cooperativity for oxygen binding and complex formation with PDI via a novel mixed disulfide linkage., Oxidative protein folding in the ER is driven mainly by oxidases of the endoplasmic reticulum oxidoreductin 1 (Ero1) family. Their action is regulated to avoid cell stress, including hyperoxidation. Previously published regulatory mechanisms are based on the rearrangement of active site and regulatory disulfides. In this study, we identify two novel regulatory mechanisms. First, both human Ero1 isoforms exist in a dynamic mixed disulfide complex with protein disulfide isomerase, which involves cysteines (Cys166 in Ero1α and Cys165 in Ero1β) that have previously been regarded as being nonfunctional. Second, our kinetic studies reveal that Ero1 not only has a high affinity for molecular oxygen as the terminal acceptor of electrons but also that there is a high cooperativity of binding (Hill coefficient >3). This allows Ero1 to maintain high activity under hypoxic conditions, without compromising cellular viability under hyper-hypoxic conditions. These data, together with novel mechanistic details of differences in activation between the two human Ero1 isoforms, provide important new insights into the catalytic cycle of human Ero1 and how they have been fine-tuned to operate at low oxygen concentrations.

We study the response of a finite-sized nanoparticle array to an incident field in the vicinity of the Γ-point of the lattice. Using the coupled dipole approximation, we find that the dipole distributions can be strongly inhomogeneous and that strong modulations appear as the energy is above the Γ-point. We highlight how this is reflected in real-space extinction efficiencies as well as in radiation patterns from the finite-sized array.
