17. Computational Fluid Dynamics for Determining the Interplay between Stirring Conditions and Crystal Size Distribution in Small Laboratory Devices

Abstract

A computational fluid dynamics method has been developed to describe the hydrodynamics of different stirring devices, which are commonly used in the development stage of protein crystallization. The experimental setup consisted of minicrystallizers in which the crystallization cocktail was stirred using a magnetic stirrer, mechanical agitator, shaker, or rotating mixer at different rotational frequencies. The hydrodynamics of the setups was determined with respect to local and average values of flow velocity, turbulent dissipation rate (ε), turbulent kinetic energy (k), and shear rate (γ̇). The calculations were performed by using the ANSYS Fluent software, in which the k-ε RNG turbulence model was implemented. The model was used to determine the probability density functions (PDFs) of the hydrodynamic parameters, which were correlated with the morphology of the protein crystalline phase obtained in the crystallization experiments. Lysozyme was used as a model protein, which was crystallized from sodium chloride solutions. For the shaker and rotating mixer, the dependence of the crystal size distribution on the k-PDF or γ̇-PDF distributions followed a similar pattern. For the mechanical agitator, a minor discrepancy was observed from that pattern at higher rotational frequencies. The correlation could not be validated for the magnetic stirrer, in which no crystalline phase was obtained under the crystallization conditions used. This was attributed to the pulverization of the solid phase caused by the magnetic bar.

37. Preferential precipitation of acidic variants from monoclonal antibody pools.

Abstract

Microheterogeneity of monoclonal antibodies (mAbs) can impact their activity and stability. Formation of charge variants is considered as the most important source of the microheterogeneity. In particular, controlling the content of the acidic species is often of major importance for the production process and regulatory approval of therapeutic proteins. In this study, the preferential precipitation process was developed for reducing the content of acidic variants in mAb downstream pools. The process design was preceded by the determination of phase behavior of mAb variants in the presence of different precipitants. It was shown that the presence of polyethylene glycol (PEG) in protein solutions favored precipitation of acidic variants of mAbs. Precipitation yield was influenced by the variant composition in the mAb feed solutions, the concentration of the precipitant and the protein, and the ionic strength of the solutions. To improve yield, multistage precipitation was employed, where the precipitate was recycled to the precipitation process. The final product was a mixture of supernatants pooled together from the recycling steps. Such an approach can be potentially used either instead or in a combination with chromatography for adjusting the acidic variant content of mAbs, which can benefit in improvement in throughput and reduction in manufacturing costs. 46. Influence of operating parameters on process behavior and product quality in fertilizer manufacturing using continuous hopper transfer pan granulation

Abstract

The present work investigates continuous pan granulation using dolomite primary particles and sprayed solution of sodium lignosulphonate binder. Individual changes of operating parameters were investigated with respect to their influences on process behavior and product quality. The latter is characterized in terms of the granules’ characteristic size, compressive strength, angle of repose, bulk density, and humidity. The experimental results show clear trends between operation parameters and product granules’ characteristic properties. It is shown that all investigated operation parameters have a significant effect on the particle size distribution and the average diameter of the granules: the average particle size increases with increasing binder flow rate to feed rate ratio, increasing binder concentration and inclination angle of the pan. For the angle of repose and the bulk density the ratio of binder flow rate to raw material flow rate and humidity is shown to be the most significant influence factor.
