Physiological homeostasis becomes compromised during ageing, as a result of impairment of cellular processes, including transcription and RNA splicing1–4. However, the molecular mechanisms leading to the loss of transcriptional fidelity are so far elusive, as are ways of preventing it. Here we profiled and analysed genome-wide, ageing-related changes in transcriptional processes across different organisms: nematodes, fruitflies, mice, rats and humans. The average transcriptional elongation speed (RNA polymerase II speed) increased with age in all five species. Along with these changes in elongation speed, we observed changes in splicing, including a reduction of unspliced transcripts and the formation of more circular RNAs. Two lifespan-extending interventions, dietary restriction and lowered insulin–IGF signalling, both reversed most of these ageing-related changes. Genetic variants in RNA polymerase II that reduced its speed in worms5 and flies6 increased their lifespan. Similarly, reducing the speed of RNA polymerase II by overexpressing histone components, to counter age-associated changes in nucleosome positioning, also extended lifespan in flies and the division potential of human cells. Our findings uncover fundamental molecular mechanisms underlying animal ageing and lifespan-extending interventions, and point to possible preventive measures.

Coronavirus disease (COVID-19) outbreaks on board cruise ships early in the pandemic highlighted gaps worldwide in public health emergency contingency plans (PHECPs) for responding to unknown threats. To restart cruise operations in 2021 and respond to potential COVID-19 outbreaks, a major tourist-based Greek island port (Port A) developed a COVID-19 PHECP. We assessed plan effectiveness by reviewing epidemiological data and monitoring outcomes, followed by an intra-action review (IAR) analyzing three event responses. From May to December 2021, 118 calls from 23 cruise ships with 119,930 passengers were recorded, with 29 COVID-19 cases in 11 cruises on board 7 ships. No outbreak was recorded during the study period. Strengths of the introduced PHECP included commitment of senior management; a core multi-disciplinary team of local authorities/ship agents involved in design and execution; interoperability agreements for port and ships’ PHECPs; cruise industry commitment to compliance; and pre-existing scenarios considering capacity needs. Central government coordination for preparedness planning at local ports is essential for successful responses. Monitoring local and country level response capacities is critical to inform planning, risk assessment, and decision-making. Immediately recording ports’ response actions provides the basis to capture lessons and improve contingency plans. To facilitate communication and common response protocols between European and non-European ports, IARs should be conducted between countries.

The collection of video data for action recognition is very susceptible to measurement bias; the equipment used, camera angle and environmental conditions are all factors that majorly affect the distribution of the collected dataset. Inevitably, training a classifier that can successfully generalize to new data becomes a very hard problem, since it is impossible to gather general enough training sets. Recent approaches in the literature attempt to solve this problem by augmenting a given training set, with synthetic data, so as to better represent the global distribution of the covariates. However, these approaches are limited because they essentially involve hand-crafted data synthesizers, which are typically hard to implement and problem specific. In this work, we propose a different approach to tackling the above issues, which relies on the combination of two techniques: pose extraction, and domain adaptation as a means to improve the generalization capabilities of classifiers. We show that adapted skeletal representations can be retrieved automatically in a semi-supervised setting and these help to generalize classifiers to new forms of measurement bias. We empirically validate our approach for generalizing across different camera angles.

Chronic exposure of workers to powder containing crystalline silica (Silicon dioxide; SiO2) can lead to chronic lung diseases (lung cancer, silicosis, etc.). Aim of this study was to evaluate the exposure of Greek construction workers to SiO2 and describe their pulmonary function. The study involved 86 outdoor and underground workers. Medical and professional history was obtained, and breath samples were collected at morning hours through a mask for the determination of SiO2 levels. Pulmonary function tests, radiological examination and evaluation of radiographs were also performed. Pulmonary function examination showed that the majority of the workers were within normal range (61.4%) while the rest were diagnosed with mild (26.5%) and more severe impairment (7.24%). Working conditions (underground-outdoor) were statistically significantly related to the categorization of pulmonary function (P=0.038). During radiological examination, the type of working activity/ conditions (underground-outdoor) were statistically significantly related to the categorization of these findings (P=0.044). Of the 69 employees, 52 did not present findings (75.4%) and 5 were diagnosed with findings specific to occupational diseases (7.23%). Environmental exposure to respirable crystalline silica (RCS) was detected at 0.0125 mg/m3 in the workplace, which is not beyond the legal limits. Underground workers with more than 15 years of exposure to SiO2 are more likely to present chronic silicosis compared to the workers of outdoor activities.

In this paper we present preliminary results of an approach for understanding human actions, based on a novel 2D image representation for 3D skeletal data. More specifically, motion information for human skeletal joints is transformed to a pseudo-colored image. A Convolutional Neural Network is then used for classification. Our approach is evaluated for actions that may be used in an ambient assisted living scenario.

In this paper we present an approach for the recognition of human actions targeting at activities of daily living (ADLs). Skeletal information is used to create images capturing the motion of joints in the 3D space. These images are then transformed to the spectral domain using 4 well-known image transforms. A deep Convolutional Neural Network is trained on those images. Our approach is thoroughly evaluated using a well-known, publicly available challenging dataset and for a set of actions that resembles to common ADLs, covering both cross-view and cross-subject cases.

The RDF(S) data model has been proposed for encoding metadata about Web resources. As more and more Web resources are annotated using RDF(S), there is an urgent need for efficiently dealing with this large volume of data. In this paper, we present Atlas, a peer-to-peer system for storing, updating and querying RDF(S) data. The Atlas system has been built using the distributed hash table Bamboo. Atlas was developed in the context of project OntoGrid, where it was used as a distributed repository for RDF(S) metadata describing Grid services and resources. The development of Atlas continues in other projects that our group participates currently. This paper gives an overview of the most recent version of Atlas and discusses a representative application.

Although the differentiation and classification of acute leukemia are based upon cytochemical features as well as immunologic, cytogenetic, and molecular characteristics, in many cases the morphological distinction of normal lymphocytes from lymphoblasts of acute lymphoblastic leukemia (ALL) is difficult using light microscopy. In this study the distinction between normal lymphocytes and lymphoblasts of childhood ALL is proposed using their spectral characteristics. The method has been based upon the analysis and classification of optical absorption characteristics of the bone marrow cells. Spectral microscopy system is capable of capturing a great number of narrow-band images, in the wide spectral range of the optical spectrum. The analysis showed statistically significant difference (P < 0.0001) between normal lymphocytes and lymphoblasts as far as it concerns the detection, identification and mapping of their spectral absorption characteristics. Our results suggest the potential of spectral imaging as a new method for the distinction of lymphocytes from lymphoblasts in cases that with the light microscope, the morphologic differences are not visible in the bone marrow smears at diagnosis or the follow up of the children with ALL.

A new type of low cost optomechanical laser-beam shutter without any wearing parts, consisting of a resonantly oscillating piezoelectric element and an optical telescope, is described. In intracavity operation, the 2.5 μs optical switching time is suitable for Q-switching lasers having long buildup times.
