In this contribution, new data concerning the distribution of vascular flora alien to Italy are presented. It includes new records and status changes from casual to naturalized for Italy or for Italian administrative regions for taxa in the genera Callianthe, Chamaecyparis, Chamaeiris, Cotoneaster, Erigeron, Freesia, Hemerocallis, Juglans, Kalanchoë, Ludwigia, Nassella, Paulownia, Physocarpus, Pistia, Saccharum, Setaria, and Vachellia. 3. Assessing the Efficiency of Two Silvicultural Approaches for Soil Erosion Mitigation Using a Novel Monitoring Apparatus

Abstract

Soil erosion and hydrogeological risk are critical phenomena gaining increased recognition within the scientific community. Although these occurrences are naturally occurring, human activities can exacerbate their impacts. For example, deforestation consistently amplifies soil erosion. This study examines two distinct forest management strategies aimed at addressing soil erosion: the Banded Standards Method (BSM) and the Scattered Standards Method (SSM). We conducted a field experiment in two test areas located in central Italy, with one area employing the BSM and the other implementing the SSM. Two soil erosion plots were established, representing prototypes of a novel erosion monitoring apparatus called the Natural Erosion Trap (NET), or Diabrosimeter, specifically designed for forest environments. At regular intervals, particularly after significant storm events, sediment and leaf litter accumulated within the erosion plots were collected, dried, and weighed to quantify erosion rates and assess the efficacy of the silvicultural methods under investigation. The results revealed a 30.72% reduction in the eroded material with BSM compared to SSM, underscoring BSM’s ability to mitigate potential hazards and preserve environmental integrity.

Abstract

Orchidaceae is one of the largest and most diverse families of flowering plants in the world but also one of the most threatened. Climate change is a global driver of plant distribution and may be the cause of their disappearance in some regions. Forest orchids are associated with specific biotic and abiotic environmental factors, that influence their local presence/absence. Changes in these conditions can lead to significant differences in species distribution. We studied three forest orchids belonging to different genera (Cephalanthera, Epipactis and Limodorum) for their potential current and future distribution in a protected area (PA) of the Northern Apennines. A Habitat Suitability Model was constructed for each species based on presence-only data and the Maximum Entropy algorithm (MaxEnt) was used for the modelling. Climatic, edaphic, topographic, anthropogenic and land cover variables were used as environmental predictors and processed in the model. The aim is to identify the environmental factors that most influence the current species distribution and the areas that are likely to contain habitats suitable for providing refuge for forest orchids and ensuring their survival under future scenarios. This will allow PA authorities to decide whether to invest more resources in conserving areas that are potential refuges for threatened species.

Abstract

The European Union is targeting climate neutrality by 2050, with a focus on enhancing energy efficiency, expanding renewable energy sources, and reducing emissions. Within Italy’s electricity mix, bioenergy sources, namely biogas, solid biomass, and bioliquids, play a crucial territorial role. A comparative analysis was conducted through Life Cycle Assessment (LCA), utilizing national data from the ARCADIA project, to assess the environmental sustainability of the investigated bioenergy chains and identify the most convenient ones. The study revealed that, among the bioenergy sources, solid biomass emerges as the most environmentally friendly option since it does not rely on dedicated crops. Conversely, biogas shows the highest environmental impact, demonstrating less favorable performance across nine out of the sixteen evaluated impact categories. The LCA underscores that the cultivation of dedicated energy crops significantly contributes to environmental burdens associated with electricity generation, affecting both biogas and bioliquids performance. The cultivation process needs water and chemical fertilizers, leading to adverse environmental effects. These findings highlight the importance of prioritizing residual biomass for energy generation over dedicated crops. Utilizing forestry and agro-industrial residues, municipal solid waste, and used cooking oils presents numerous advantages, including environmental preservation, resource conservation and recovery, as well as waste reduction.

Abstract

Acoustic radiation forced impulse (ARFI) is an integrated ultrasound method, measuring stiffness by point shear wave elastography. To evaluate the diagnostic performance of the ARFI of the liver and the spleen, combined with spleen dimension and platelet count, in predicting high-risk esophageal varices (HRVs) in cirrhotic patients, a prospective and cross-sectional study was conducted between February 2017 and February 2021. The following ratio scores were calculated based on ARFI measurements: ALSDP (ARFI Liver–Spleen Diameter-to-Platelet Ratio Score), ASSDP (ARFI Spleen–Spleen Diameter-to-Platelet Ratio Score), ASSAP (ARFI Spleen–Spleen Area-to-Platelet Ratio Score), and ALSAP (ARFI Liver–Spleen Area-to-Platelet Ratio Score). In 100 enrolled subjects, spleen ARFI, ASSDP, and ASSAP were significantly associated with HRVs in the prospective short- and long-term follow-ups and in the cross-sectional study (p < 0.05), while ALSDP and ALSAP were associated with HRVs only in the prospective long-term follow-up and cross-sectional study (p< 0.05). ASSAP was the best ARFI ratio score for HRVs at the long-term follow-up [value of area under curve (AUC) = 0.88], although all the ARFI ratio scores performed better than individual liver and spleen ARFI (AUC > 0.7). In our study, ARFI ratio scores can predict, in well-compensated cirrhotic patients, the risk of developing HVRs in short- and long-term periods.

17. Multi-environment ecogenomics analysis of the cosmopolitan phylum Gemmatimonadota

Abstract

ABSTRACT Gemmatimonadota is a diverse bacterial phylum commonly found in environments such as soils, rhizospheres, fresh waters, and sediments. So far, the phylum contains just six cultured species (five of them sequenced), which limits our understanding of their diversity and metabolism. Therefore, we analyzed over 400 metagenome-assembled genomes (MAGs) and 5 culture-derived genomes representing Gemmatimonadota from various aquatic environments, hydrothermal vents, sediments, soils, and host-associated (with marine sponges and coral) species. The principal coordinate analysis based on the presence/absence of genes in Gemmatimonadota genomes and phylogenomic analysis documented that marine and host-associated Gemmatimonadota were the most distant from freshwater and wastewater species. A smaller genome size and coding sequences (CDS) number reduction were observed in marine MAGs, pointing to an oligotrophic environmental adaptation. Several metabolic pathways are restricted to specific environments. For example, genes for anoxygenic phototrophy were found only in freshwater, wastewater, and soda lake sediment genomes. There were several genomes from soda lake sediments and wastewater containing type IC/ID ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO). Various genomes from wastewater harbored bacterial type II RuBisCO, whereas RuBisCO-like protein was found in genomes from fresh waters, soil, host-associated, and marine sediments. Gemmatimonadota does not contain nitrogen fixation genes; however, the nosZ gene, involved in the reduction of N2O, was present in genomes from most environments, missing only in marine water and host-associated Gemmatimonadota. The presented data suggest that Gemmatimonadota evolved as an organotrophic species relying on aerobic respiration and then remodeled its genome inventory when adapting to particular environments. IMPORTANCE Gemmatimonadota is a rarely studied bacterial phylum consisting of a handful of cultured species. Recent culture-independent studies documented that these organisms are distributed in many environments, including soil, marine, fresh, and waste waters. However, due to the lack of cultured species, information about their metabolic potential and environmental role is scarce. Therefore, we collected Gemmatimonadota metagenome-assembled genomes (MAGs) from different habitats and performed a systematic analysis of their genomic characteristics and metabolic potential. Our results show how Gemmatimonadota have adapted their genomes to different environments.

Abstract

Background: This study assessed the potential cost-effectiveness of high (80–100%) vs low (21–35%) fraction of inspired oxygen (FiO2) at preventing surgical site infections (SSIs) after abdominal surgery in Nigeria, India, and South Africa. Methods: Decision-analytic models were constructed using best available evidence sourced from unbundled data of an ongoing pilot trial assessing the effectiveness of high FiO2, published literature, and a cost survey in Nigeria, India, and South Africa. Effectiveness was measured as percentage of SSIs at 30 days after surgery, a healthcare perspective was adopted, and costs were reported in US dollars ($). Results: High FiO2 may be cost-effective (cheaper and effective). In Nigeria, the average cost for high FiO2 was $216 compared with $222 for low FiO2 leading to a −$6 (95% confidence interval [CI]: −$13 to −$1) difference in costs. In India, the average cost for high FiO2 was $184 compared with $195 for low FiO2 leading to a −$11 (95% CI: −$15 to −$6) difference in costs. In South Africa, the average cost for high FiO2 was $1164 compared with $1257 for low FiO2 leading to a −$93 (95% CI: −$132 to −$65) difference in costs. The high FiO2 arm had few SSIs, 7.33% compared with 8.38% for low FiO2, leading to a −1.05 (95% CI: −1.14 to −0.90) percentage point reduction in SSIs. Conclusion: High FiO2 could be cost-effective at preventing SSIs in the three countries but further data from large clinical trials are required to confirm this.

Abstract

Adaptive subtraction is a key element in predictive multiple-suppression methods. It minimizes misalignments and amplitude differences between modeled and actual multiples, and thus reduces multiple contamination in the dataset after subtraction. Due to the high cross-correlation between their waveform, the main challenge resides in attenuating multiples without distorting primaries. As they overlap on a wide frequency range, we split this wide-band problem into a set of more tractable narrow-band filter designs, using a 1D complex wavelet frame. This decomposition enables a single-pass adaptive subtraction via complex, single-sample (unary) Wiener filters, consistently estimated on overlapping windows in a complex wavelet transformed domain. Each unary filter compensates amplitude differences within its frequency support, and can correct small and large misalignment errors through phase and integer delay corrections. This approach greatly simplifies the matching filter estimation and, despite its simplicity, narrows the gap between 1D and standard adaptive 2D methods on field data., Comment: 18 pages, 10 color figures

Abstract

Abstract Background Cyanobacteria are the major prokaryotic primary producers occupying a range of aquatic habitats worldwide that differ in levels of salinity, making them a group of interest to study one of the major unresolved conundrums in aquatic microbiology which is what distinguishes a marine microbe from a freshwater one? We address this question using ecogenomics of a group of picocyanobacteria (cluster 5) that have recently evolved to inhabit geographically disparate salinity niches. Our analysis is made possible by the sequencing of 58 new genomes from freshwater representatives of this group that are presented here, representing a 6-fold increase in the available genomic data. Results Overall, freshwater strains had larger genomes (≈2.9 Mb) and %GC content (≈64%) compared to brackish (2.69 Mb and 64%) and marine (2.5 Mb and 58.5%) isolates. Genomic novelties/differences across the salinity divide highlighted acidic proteomes and specific salt adaptation pathways in marine isolates (e.g., osmolytes/compatible solutes - glycine betaine/ggp/gpg/gmg clusters and glycerolipids glpK/glpA), while freshwater strains possessed distinct ion/potassium channels, permeases (aquaporin Z), fatty acid desaturases, and more neutral/basic proteomes. Sulfur, nitrogen, phosphorus, carbon (photosynthesis), or stress tolerance metabolism while showing distinct genomic footprints between habitats, e.g., different types of transporters, did not obviously translate into major functionality differences between environments. Brackish microbes show a mixture of marine (salt adaptation pathways) and freshwater features, highlighting their transitional nature. Conclusions The plethora of freshwater isolates provided here, in terms of trophic status preference and genetic diversity, exemplifies their ability to colonize ecologically diverse waters across the globe. Moreover, a trend towards larger and more flexible/adaptive genomes in freshwater picocyanobacteria may hint at a wider number of ecological niches in this environment compared to the relatively homogeneous marine system.

21. Two Strategies for Attenuating Internal Multiples in Land Data

Abstract

Abstract Two new internal multiple attenuation methodologies are presented. The first approach uses a 3D wave equation based multiple modeling technique followed by 3D adaptive subtraction. The recorded data are back propagated/propagated through a reflectivity model (obtained from a preliminary migration) of the overburden. All possible multiple raypaths are modeled by using different combinations of two sub-windows in this reflectivity model. Each combination results in a specific multiple model. The generated multiple models are then simultaneously adapted and subtracted from the input data. The second approach is a 3D dip extraction and filtering technique, taking advantage of any dip discrimination between the primaries and multiples. It works on post stack and pre stack volumes. We show how the two methods are complementary and how they can be combined. The first approach can be applied to any seismic data plagued with strong internal multiples and is perfectly adapted to modern dense, wide azimuth surveys. The second approach works on any seismic volume in which multiples and primaries have different dips in a given domain. Some real data examples from South Oman 3D seismic datasets are shown. All are characterized by a heavy multiple contamination, generated by strong shallow reflectivity sequences in a flattish overburden, and overlying deeper, weaker and heavily structured primaries. By revealing structures previously invisible, these techniques add a lot of value to existing seismic data sets. The generation mechanism of internal multiples is often quite complex making them difficult to predict. The main benefits of the methods described here are that they work in absence of move out discrimination and without the need of a precise identification of the internal multiple generators. Introduction In some areas of Middle East, it is common to have shallow carbonate sequences with strong reflectivity, high velocity contrasts, overlying lower reflectivity clastic series with weak impedance contrasts and lower velocities. This geologic sequence yields a heavy contamination of the lower unit by energetic interbed multiples generated in the upper unit ("curtain of multiples"). These multiples often have the same, or higher apparent velocity, than the primaries, offering no opportunity to remove them by move-out discrimination. Similarly, prediction methods (gapped deconvolution) have proved unsatisfactory. And in addition, it is mostly impossible to identify clearly the generators, given the large number of possible candidates. Historically these unwanted multiples have been removed by surgical 3D FK filtering after flattening of the suspected multiple generators. This method has been used for many years but has several strong limitations, in addition to the well known FK artifacts. Hence new methodologies were required to overcome the failings of this 3D FK approach.

Abstract

Abstract Recent studies demonstrated reduced blood lysosomal acid lipase (LAL) activity in patients with nonalcoholic fatty liver disease (NAFLD). We aimed to verify hepatic LAL protein content and activity in in vitro and in vivo models of fat overload and in NAFLD patients. LAL protein content and activity were firstly evaluated in Huh7 cells exposed to high-glucose/high-lipid (HGHL) medium and in the liver of C57BL/6 mice fed with high-fat diet (HFD) for 4 and 8 months. LAL protein was also evaluated by immunohistochemistry in liver biopsies from 87 NAFLD patients and 10 controls, and correlated with hepatic histology. Huh7 cells treated with HGHL medium showed a significant reduction of LAL activity, which was consistent with reduced LAL protein levels by western blotting using an antibody towards the N-term of the enzyme. Conversely, antibodies towards the C-term of the enzyme evidenced LAL accumulation, suggesting a post-translational modification that masks the LAL N-term epitope and affects enzymatic activity. Indeed, we found a high rate of ubiquitination and extra-lysosomal localization of LAL protein in cells treated with HGHL medium. Consistent with these findings, inhibition of proteasome triggered dysfunctional LAL accumulation and affected LAL activity. Accumulation of ubiquitinated/dysfunctional LAL was also found in the liver of HFD fed mice. In NAFLD patients, hepatic levels of non-ubiquitinated/functional LAL were lower than in controls and inversely correlated with disease activity and some of the hallmarks of reduced LAL. Fat overload leads to LAL ubiquitination and impairs its function, possibly reducing hepatic fat disposal and promoting NAFLD activity.

Abstract

The role of competitive interactions based on resource utilisation was explored in a phototrophic microbial mat from Byers Peninsula (Maritime Antarctica). Shotgun metagenomic profiling of the mat showed a taxonomic and functionally diverse microbial community. The heterotrophic bacterial community was dominated by Proteobacteria, where genera typically found in polar habitats, such as Janthinobacterium, Pseudomonas, and Polaromonas, were highly prevalent. Cyanobacteria played the main role as primary producers, accompanied by diatoms and chlorophytes. To test the potential effects of the inorganic nutrient (N and P) availability on this community, a fully factorial nitrate and phosphorus addition experiment was conducted in situ. The mat exhibited a functional and structural response to the nutrient amendments. Compared to the undisturbed mat, phosphorus fertilisation favoured the growth of (non-heterocystous) cyanobacteria relative to that of diatoms, as indicated by changes in the carotenoid pigment biomarkers. Although no mat accretion was visible, fertilisation improved the phototrophic activity, and, mainly, when P was amended, the production of exopolymeric substances was favoured, whereas further changes in the vertical distribution of primary production activity were observed as well. Illumina amplicon sequencing of the 16S rRNA gene also demonstrated changes in the relative abundance of heterotrophic prokaryotes, which were detectable from the phylum to the genus level and mainly related to the amendment of nitrogen. Predictions made on the functional skills of these shifted prokaryotic communities indicated changes in abundance selecting taxa with a metabolic adaptation to the new nutrient scenarios. They mainly consisted of the enhancement of ecological strategies and metabolic regulatory mechanisms related to the uptake and metabolising of either nitrogen or phosphorus, regulated by its availability whether in a balanced way or not. This study is a pioneer in demonstrating how shifts in the regional dynamic of nutrients might alter the metabolic equilibrium of these initially considered homeostatic benthic communities. They can be accordingly considered as taxonomically diverse microbiomes with a functional repertoire still inclined to respond to the biogeochemical alteration of nutrient cycles, although occurring in a cold extreme environment where biological activity is partially restricted by environmental harshness.

Multiple attenuation is one of the greatest challenges in seismic processing. Due to the high cross-correlation between primaries and multiples, attenuating the latter without distorting the former is a complicated problem. We propose here a joint multiple model-based adaptive subtraction, using single-sample unary filters’ estimation in a complex wavelet transformed domain. The method offers more robustness to incoherent noise through redundant decomposition. It is first tested on synthetic data, then applied on real-field data, with a single-model adaptation and a combination of several multiple models.

Abstract

Abstract Background The Black Sea is the largest brackish water body in the world, although it is connected to the Mediterranean Sea and presents an upper water layer similar to some regions of the former, albeit with lower salinity and temperature. Despite its well-known hydrology and physicochemical features, this enormous water mass remains poorly studied at the microbial genomics level. Results We have sampled its different water masses and analyzed the microbiome by shotgun and genome-resolved metagenomics, generating a large number of metagenome-assembled genomes (MAGs) from them. We found various similarities with previously described Black Sea metagenomic datasets, that show remarkable stability in its microbiome. Our datasets are also comparable to other marine anoxic water columns like the Cariaco Basin. The oxic zone resembles to standard marine (e.g. Mediterranean) photic zones, with Cyanobacteria (Synechococcus but a conspicuously absent Prochlorococcus), and photoheterotrophs domination (largely again with marine relatives). The chemocline presents very different characteristics from the oxic surface with many examples of chemolithotrophic metabolism (Thioglobus) and facultatively anaerobic microbes. The euxinic anaerobic zone presents, as expected, features in common with the bottom of meromictic lakes with a massive dominance of sulfate reduction as energy-generating metabolism, a few (but detectable) methanogenesis marker genes, and a large number of “dark matter” streamlined genomes of largely unpredictable ecology. Conclusions The Black Sea oxic zone presents many similarities to the global ocean while the redoxcline and euxinic water masses have similarities to other similar aquatic environments of marine (Cariaco Basin or other Black Sea regions) or freshwater (meromictic monimolimnion strata) origin. The MAG collection represents very well the different types of metabolisms expected in this kind of environment. We are adding critical information about this unique and important ecosystem and its microbiome.

Summary Model-based surface related multiple modeling (3D SRMM) can be achieved by the use of pre-stack demigration algorithms, thus avoiding the constrains on the shot positions distribution required by the data-based methods. In the following, we show a new method for modeling internal multiples in 3D by using a model-based technique. This method follows a parallel flow with respect to those employed by the data-based multiple modeling techniques, and allows for the construction of internal multiple events produced between upper layers reflecting the energy downward, and lower layers reflecting the energy upward. The method has been applied to Wide Azimuth Towed Streamer data from the Gulf of Mexico.

The acquisition of n-shots, more or less simultaneously, increases acquisition efficiency and collects a wider range of information for imaging and reservoir characterisation. Its success relies critically on the ability to separate n-shots from one recording. Stefani et al (2007) showed that while some datasets may be easily separated, others are more difficult. Using the more difficult data example from Stefani et al (loc.cit.), we show that a PEF-based adaptive subtraction (Spitz, 2007) of the estimated wavefield due to a secondary source provides an effective separation of the sources.

International audience; Nonlinear elastic waveform inversion has advanced to the point where it is now possible to invert real multiple‐shot seismic data. The iterative gradient algorithm that we employ can readily accommodate robust minimization criteria which tend to handle many types of seismic noise (noise bursts, missing traces, etc.) better than the commonly used least‐squares minimization criteria. Although there are many robust criteria from which to choose, we have tested only a few. In particular, the Cauchy criterion and the hyperbolic secant criterion perform very well in both noise‐free and noise‐added inversions of numerical data. Although the real data set, which we invert using the sech criterion, is marine (pressure sources and receivers) and is very much dominated by unconverted P waves, we can, for the most part, resolve the short wavelengths of both P impedance and S impedance. The long wavelengths of velocity (the background) are assumed known. Because we are deriving nearly all impedance information from unconverted P waves in this inversion, data acquisition geometry must have sufficient multiplicity in subsurface coverage and a sufficient range of offsets, just as in amplitude‐versus‐offset (AVO) inversion. However, AVO analysis is implicitly contained in elastic waveform inversion algorithms as part of the elastic wave equation upon which the algorithms are based. Because the real‐data inversion is so large—over 230,000 unknowns (340,000 when density is included) and over 600,000 data values—most statistical analyses of parameter resolution are not feasible. We qualitatively verify the resolution of our results by inverting a numerical data set which has the same acquisition geometry and corresponding long wavelengths of velocity as the real data, but has semirandom perturbations in the short wavelengths of P and S impedance.

32. OBS illumination: primary problems and mirror solutions

Abstract The world's demand for energy is accelerating, while its reserves of energy are diminishing. Producers are compelled to explore and produce oil and gas in more challenging environments and to maximize recovery in existing reservoirs. New seismic exploration technology has always been a key to success. One such new technology is Ocean Bottom Station (OBS) nodes. Since the cost of OBS deployment is high, a practical and relatively economical geometry for OBS is a sparse grid of nodes and a dense grid of shots. However, the sparse node geometry provides poor illumination, especially of reflectors whose depth under the seabed is less than the node interval. Fortunately, there is a good solution for this problem: the mirror imaging method. The sea surface acts as an acoustic convex mirror reflecting the image of subsurface structure. This mirror image offers wider illumination than a conventional image derived from primary reflections. We present the mirror imaging method and compare conventional and mirror imaging results from OBS data recorded in the North Sea, the Norwegian Sea and offshore West Africa. We then discuss the reasons why mirror imaging is superior to the conventional image of the primaries. Introduction Ocean Bottom Station (OBS) nodes (Berg et al., 1994; Ronen et al., 2003; Amal et al., 2005; Docherty et al., 2005; Granger et al., 2005) are an emerging seismic exploration technology. Surface-towed streamers provide excellent seismic data for exploration, development and production monitoring. However, streamers have limitations and this motivates a quest for alternative technologies. When using streamers, obstacles such as production platforms require undershooting and the data lack near offsets and have anomalous azimuth distributions. OBS nodes are much less sensitive to obstacles and allow more complete coverage. When deployed by a Remotely-Operated Vehicle (ROV), they can be placed very accurately, right up to, or even under, production installations. Obstacles are not the only motivation for OBS technology. Even in the absence of obstacles, OBS nodes facilitate wide-azimuth geometries, important for imaging structures under complex overburdens such as salt environments. In particular, by moving the receivers to the seabed and recording a dense shooting grid on the sea surface, we can create a dataset that is suitable for wave-equation migration, is well-populated in azimuth and offset, and provides optimal subsurface illumination; conventional towed-streamer acquisition cannot provide this type of dataset because of the constraints imposed by the fixed source-receiver geometry. Indeed, while the Wide-Azimuth Towed-Streamer (WATS) method is becoming popular, and can also provide data suitable for wave-equation migration (Threadgold et al., 2006), it requires separate source vessels as well as repeated shot lines with different streamer locations. Such effort comes at a cost, and, for small areas (up to about 400 sq.km), OBS technology can be less expensive than WATS. Similarly, monitoring with time-lapse seismic surveys (4D) requires acquisition repeatability; OBS nodes deployed by ROVs provide better repeatability than streamers, which are subject to feathering due to currents. Indeed, although the limitations of streamers are reduced with streamer-steering, streamer-steering can only have a small effect of just a few degrees, while feathering is sometimes ten degrees or more. Eiken et al. (2003) report a natural streamer feathering within +/- 6o for 95% of Norwegian Sea operations, and a streamer steering capability of +/- 3o that is not sufficient to achieve a zero-feather survey in a cost effective manner. Streamer steering also acts as an additional source of noise in seismic recordings. Finally, nodes deployed on the seabed record not only P-waves but also S-waves; these are a useful complement to P-waves but do not travel in water, and therefore cannot be recorded by surface-towed streamers. OBS nodes can provide four-component (4C) seismic data from a hydrophone and a three-component geophone, thereby enabling elastic-wave analysis, as well as wavefield separation and multiple removal. Applications of elastic-wave analysis include imaging beneath gas clouds, imaging low P-impedance reservoirs and fracture characterization.

Abstract

As reported in the recent literature, Nickel has become an important part of our daily life since the last decades. We can find it in skincare products, occupational exposures and foods. Only recently, research has started to show a link between Nickel and many health disorders, including adverse reactions to food containing nickel. Nowadays, the relationship between nickel-containing foods and well-being is becoming a topic of growing interest in clinical practice and will play an even larger role in the future. The use of foods with a high nickel content, largely present in a gluten free diet, could explain the lack of clinical remission in celiac patients and dispel a diagnosis of refractory celiac disease.

Abstract

Background & Aims: Type 2 diabetes is a major driver of fatty liver disease and its long-term complications. The aim of this study was to investigate the individual contribution of inborn and acquired risk factors for severe liver disease in individuals with type 2 diabetes from the UK Biobank study. Methods: A total of 22,812 UK Biobank participants of European descent without clinical history of liver disease and liver cancer were prospectively followed for the development of severe liver disease, defined as a composite diagnosis of cirrhosis, decompensated liver disease, hepatocellular carcinoma, and/or liver transplantation from the National Health Service records. The contribution of inborn and acquired risk factors to the risk of incident severe liver disease was assessed by Cox proportional hazards models. Results: During a median follow-up of 8.9 years (IQR 8.1–9.6), there were 279 individuals with severe liver disease, including 255 with cirrhosis and/or decompensated liver disease, 47 with hepatocellular carcinoma, and 5 with liver transplantation; death from severe liver disease occurred in 83 individuals. Risk factors independently associated with increased risk of incident severe liver disease included abnormal aspartate aminotransferase (adjusted hazard ratio [aHR] 4.85, 95% CI 2.76–8.54), decrease in serum albumin (aHR 2.39, 95% CI 1.76–3.24) and platelet count (aHR 1.12, 95% CI 1.09–1.16), cardiovascular disease (aHR 1.86, 95% CI 1.23–2.79), microalbuminuria (aHR 1.55, 95% CI 1.04–2.30), PNPLA3 rs738409 (aHR 1.67, 95% CI 1.27–2.18) and TM6SF2 rs58542926 (aHR 1.63, 95% CI 1.12–2.39), while the net effect of male sex was protective (aHR 0.49, 95% CI 0.26–0.94). Conclusions: These findings may help in clinical care to identify individuals with type 2 diabetes at risk of severe liver disease, in turn leading to personalised risk prediction and prevention strategies. Lay summary: Type 2 diabetes is a key driver of severe liver disease, namely cirrhosis, hepatocellular carcinoma, and liver-related mortality. In Europeans with type 2 diabetes from the prospective UK Biobank study, abnormal liver function, cardiovascular disease, microalbuminuria, and genetic variants in PNPLA3 and TM6SF2 genes are the major independent risk factors for severe liver disease. These findings may contribute in clinical care to identify and closely monitor individuals with type 2 diabetes at risk of developing severe liver disease, requiring more intensive follow-up strategies.

Abstract

Although in theory geophysicists should not apply multiple removal before surface related multiple attenuation, modern marine processing sequences include predictive deconvolution before or after SRME / SRMM. In practice the two methods complement each other well, predictive deconvolution tackles very short period multiples in very shallow water environments while SRME, although supposed to handle any multiples in the data, will thrive in slightly longer multiple period. The reason for the failure of the SRME / SRMM techniques in shallow water environments is that the convolution process between the primary response and the total response is jeopardized because the primary response (the water bottom) is incompletely recorded. Indeed, due to missing near offsets (near offsets are in the order of 150m), most of the primary water bottom reflection is not recorded, and in addition might interfere with the direct arrival wave (Verschuur, 2006). At least it is not sufficiently accurately recorded for the SRME convolution to successfully predict multiples for small offsets. Predictive deconvolution, on the other hand, is known to be very effective as it relies only on the periodicity of the multiples wave field and not on the water bottom reflection itself. In the first part of this case study we look at the relative effectiveness of 2D SRME / 3D SRMM compared with predictive devonvolution.

Abstract

Background & aims: Obeticholic acid (OCA) is the second-line treatment approved for patients with primary biliary cholangitis (PBC) and an inadequate response or intolerance to ursodeoxycholic acid. We aimed to evaluate the effectiveness and safety of OCA under real-world conditions. Methods: Patients were recruited into the Italian PBC Registry, a multicentre, observational cohort study that monitors patients with PBC at national level. The primary endpoint was the biochemical response according to Poise criteria; the secondary endpoint was the biochemical response according to normal range criteria, defined as normal levels of bilirubin, alkaline phosphatase (ALP), and alanine aminotransferase (ALT) at 12 months. Safety and tolerability were also assessed. Results: We analysed 191 patients until at least 12 months of follow-up. Median age was 57 years, 94% female, 61 (32%) had cirrhosis, 28 (15%) had histologically proven overlap with autoimmune hepatitis (PBC-AIH). At 12 months, significant median reductions of ALP (-32.3%), ALT (-31.4%), and bilirubin (-11.2%) were observed. Response rates were 42.9% according to Poise criteria, and 11% by normal range criteria. Patients with cirrhosis had lower response than patients without cirrhosis (29.5% vs. 49.2%, p = 0.01), owing to a higher rate of OCA discontinuation (30% vs. 12%, p = 0.004), although with similar ALP reduction (29.4% vs. 34%, p = 0.53). Overlap PBC-AIH had a similar response to pure PBC (46.4% vs. 42.3%, p = 0.68), with higher ALT reduction at 6 months (-38% vs. -29%, p = 0.04). Thirty-three patients (17%) prematurely discontinued OCA because of adverse events, of whom 11 experienced serious adverse events. Treatment-induced pruritus was the leading cause of OCA discontinuation (67%). Conclusions: Effectiveness and safety of OCA under real-world conditions mirror those in the Poise trial. Patients with cirrhosis had lower tolerability. Overlap PBC-AIH showed higher ALT reduction at 6 months compared with patients with pure PBC. Lay summary: Obeticholic acid (OCA) was shown to be effective in more than one-third of patients not responding to ursodeoxycholic acid in a real-world context in Italy. Patients with cirrhosis had more side effects with OCA, and this led to suspension of the drug in one-third of patients. OCA was also effective in patients who had overlap between autoimmune hepatitis and primary biliary cholangitis.

This atlas aims to provide an overview of the picocyanobacteria cultures hold by the CNR-IRSA collection. These are monoclonal strains mostly of the genera Synechococcus and Cyanobium. These strains were mainly isolated from lakes of different trophic status around the globe and, so far, they are not available in other collections of algae and (pico)cyanobacteria. Here we summarise the main characteristics of these strains, including some previously published data, and offer most of the strains as material available for research to every scientists upon request.

Abstract

Introduction Surgical site infection (SSI) is one of the most common healthcare-associated infections (HAIs). However, there is a lack of data available about SSI in children worldwide, especially from low-income and middle-income countries. This study aimed to estimate the incidence of SSI in children and associations between SSI and morbidity across human development settings.Methods A multicentre, international, prospective, validated cohort study of children aged under 16 years undergoing clean-contaminated, contaminated or dirty gastrointestinal surgery. Any hospital in the world providing paediatric surgery was eligible to contribute data between January and July 2016. The primary outcome was the incidence of SSI by 30 days. Relationships between explanatory variables and SSI were examined using multilevel logistic regression. Countries were stratified into high development, middle development and low development groups using the United Nations Human Development Index (HDI).Results Of 1159 children across 181 hospitals in 51 countries, 523 (45·1%) children were from high HDI, 397 (34·2%) from middle HDI and 239 (20·6%) from low HDI countries. The 30-day SSI rate was 6.3% (33/523) in high HDI, 12·8% (51/397) in middle HDI and 24·7% (59/239) in low HDI countries. SSI was associated with higher incidence of 30-day mortality, intervention, organ-space infection and other HAIs, with the highest rates seen in low HDI countries. Median length of stay in patients who had an SSI was longer (7.0 days), compared with 3.0 days in patients who did not have an SSI. Use of laparoscopy was associated with significantly lower SSI rates, even after accounting for HDI.Conclusion The odds of SSI in children is nearly four times greater in low HDI compared with high HDI countries. Policies to reduce SSI should be prioritised as part of the wider global agenda.

Abstract

Cancer immunotherapy has become a stronghold in modern oncology. Immune checkpoint inhibitors, in particular anti-PD-1 and anti-PD-L1 antibodies, are approved for the treatment of several solid cancers. In the near future, an increasing number of patients will be eligible for immunotherapy. Therefore, the management of immune-related adverse events is a daily challenge in clinical practice, among which hepatic immune-related toxicity has been described as a rare adverse event. We report the case of a patient treated with nivolumab (an anti-PD-L1 antibody) for a stage IV resected melanoma who developed recurrence of steroid-refractory liver toxicity that was later discovered to be associated with acute exacerbation of chronic undiagnosed hepatitis B. The patient significantly benefited from antiviral treatment. We conclude that serological viral screening is strongly recommended before starting immune checkpoint inhibitor treatment.

Abstract

Non-alcoholic fatty liver disease (NAFLD) is the most common cause of chronic liver disease in Western countries and is associated with aging and features of metabolic syndrome. Lipotoxicity and oxidative stress are consequent to dysregulation of lipid metabolism and lipid accumulation, leading to hepatocyte injury and inflammation. Lipophagy consists in selective degradation of intracellular lipid droplets by lysosome and mounting evidence suggests that lipophagy is dysregulated in NAFLD. Here we demonstrate lipophagy impairment in experimental models of NAFLD and in a NAFLD patient cohort by histomorphological and molecular analysis. High fat diet-fed C57BL/6J male mice and high-fat/high-glucose cultured Huh7 cells showed accumulation of both p62/SQSTM1 and LC3-II protein. In 59 NAFLD patients, lipid droplet-loaded lysosomes/lipolysosomes and p62/SQSTM1 clusters correlated with NAFLD activity score (NAS) and with NAS and fibrosis stage, respectively, and levels of expression of lysosomal genes, as well as autophagy-related genes, correlated with NAS and fibrosis stage. An increased amount of lipid droplets, lipolysosomes and autophagosomes was found in subjects with NAFLD compared to healthy subjects at ultrastructural level. In conclusion, here we observed that NAFLD is characterized by histological, ultrastructural and molecular features of altered autophagy that is associated with an impaired lipid degradation. Impaired autophagy is associated with features of advanced disease. Lipopolysosomes, as individuated with light microscopy, should be further assessed as markers of disease severity in NAFLD patients.

Abstract

Deltaic wetlands are highly productive ecosystems, which characteristically can act as C-sinks. However, they are among the most threatened ecosystems, being very vulnerable to global change, and require special attention towards its conservation. Knowing their climate change mitigating potential, conservation measures should also be oriented with a climatic approach, to strengthen their regulatory services. In this work we studied the carbon biogeochemistry and the specific relevance of certain microbial guilds on carbon metabolisms of the three main types of deltaic wetlands located in the Ebro Delta, north-eastern Spain, as well as how they deal with human pressures and climate change effects. We estimated the metabolic rates of the main carbon-related metabolisms (primary production and respiration) and the resulting carbon and global warming potential balances in sites with a different salinity range and trophic status. With the results obtained, we tried to define the influence of possible changes in salinity and trophic level linked to the main impacts currently threatening deltaic wetlands, on the C-metabolisms and GHG emissions, for a better understanding of the mitigating capacity and their possible enhancement when applying specific management actions. Metabolic rates showed a pattern highly influenced by the salinity range and nutrients inputs. Freshwater and brackish wetlands, with higher nutrient inputs from agricultural runoff, showed higher C-capture capacity (around 220-250 g C m-2 y-1), but also higher rates of degradative metabolisms (aerobic respiration and CH4 emissions). Contrastingly, the rates of C-related metabolisms and C-retention of Salicornia-type coastal salt marshes were lower (42 g C m-2 y-1). The study of the microbial metacommunity composition by the16S RNA gene sequencing revealed a significant higher presence of methanogens in the salt marsh, and also higher metabolic potential, where there was significantly more organic matter content in sediment. Salinity inhibition, however, explained the lower respiration rates, both aerobic and anaerobic, and prevented higher rates of methanogenesis despite the major presence of methanogens. Conservation measures for these wetlands would require, overall, maintaining the sediment contributions of the river basin intending to overcome the regression of the Delta and its salt marshes in a climate change scenario. Particularly, for reducing degradative metabolisms, and favour C-retention, nutrient inputs should be controlled in freshwater and brackish wetlands in order to reduce eutrophication. In salt marshes, the reduction of salinity should be avoided to control increases in methanogenesis and CH4 emissions.
