Purpose: Cardiovascular disease (CVD) is more prevalent in men than women, but the mechanisms responsible for this are not fully understood. We aimed to evaluate differences in trimethylamine (TMA), a microbial metabolite and its oxidized form, trimethylamine N-oxide (TMAO), which is thought to promote atherosclerosis, between men and women with coronary heart disease (CHD), using as a reference a non-CVD population. Materials and Methods: This study was carried out within the framework of the CORDIOPREV study (NCT00924937; June 19, 2009), a clinical trial which included 827 men and 175 women with CHD, with a non-CVD population of 375 individuals (270 men and 105 women) as a reference group. Plasma TMA and TMAO were measured by HPLC-MS/MS. The carotid study was ultrasonically assessed bilaterally by the quantification of intima-media thickness of both common carotid arteries (IMT-CC). Results: We found higher TMAO levels and TMAO/TMA ratio in CHD men than CHD women (p=0.034 and p=0.026, respectively). No TMA sex differences were found in CHD patients. Abstract Background Increasing evidence supports the role of advanced glycation end products (AGEs) in atherosclerosis in both diabetic and non-diabetic patients, suggesting that therapeutic strategies targeting AGEs may offer potential benefits in this population. The Mediterranean diet is associated with improved biomarkers and anthropometric measurements related with atherosclerosis in addition to its ability to modulate AGE metabolism. Our aim was to determine whether the reduction in atherosclerosis progression (measured by changes in intima-media thickness of both common carotid arteries (IMT-CC)), observed after consumption of a Mediterranean diet compared to a low-fat diet, is associated with a modulation of circulating AGE levels in patients with coronary heart disease (CHD). Methods 1002 CHD patients were divided in: (1) Non-increased IMT-CC patients, whose IMT-CC was reduced or not changed after dietary intervention and (2) Increased IMT-CC patients, whose IMT-CC was increased after dietary intervention. Serum AGE levels (methylglyoxal–MG and Nε-Carboxymethyllysine-CML) and parameters related to AGE metabolism (AGER1 and GloxI mRNA and sRAGE levels) and reduced glutathione (GSH) levels were measured before and after 5-years of dietary intervention. La conciencia metacognitiva entre los estudiantes ha sido ampliamente investigada anteriormente. Sin embargo, la metacognición entre los docentes universitarios no se ha examinado tan exhaustivamente. En consecuencia, el propósito del presente estudio fue investigar la conciencia metacognitiva autoinformada, utilizando el Inventario de Conciencia Metacognitiva para Docentes (ICMD) entre una muestra de docentes universitarios colombianos (N =188) de diversas disciplinas y especializaciones. Las estadísticas descriptivas revelaron que los docentes universitarios tendieron a sobrestimar la conciencia metacognitiva en sus seis dimensiones (conocimiento metacognitivo: declarativo, procedimental y condicional; regulación metacognitiva: planificación, monitoreo y evaluación). Además, una comparación entre disciplinas reveló que los docentes de ciencias sociales y humanas informaron significativamente menos conocimiento declarativo, monitoreo y evaluación en comparación con otras disciplinas. Curiosamente, años de experiencia en la enseñanza predijeron negativamente el conocimiento condicional, la planificación, el monitoreo y la evaluación, de modo que se evidenció que una mayor experiencia condujo a una menor conciencia metacognitiva en estas dimensiones. Se discuten las implicaciones de estos hallazgos para la investigación y la práctica.

A long-term regional solar energy assessment improves decision-making processes for the selection of the best solar energy technology and helps understand how to define focused policies and investments. This study presents long-term solar resource monthly maps for Qatar. The availability of solar radiation throughout the country has been mapped here using ground-based measurements and satellite data. The regression-kriging algorithm and its variants are used to calibrate satellite data, through interpolation of ground solar radiation data. Digital surface model data is used as an auxiliary variable to further scale down to 30 m resolution. Long-term monthly maps of daily total solar radiation have been produced which consist of 12 monthly maps for global horizontal irradiance (GHI) and 12 monthly maps for direct normal irradiance (DNI) for each month of the year. A significant improvement of 9.15% and 9.32% in GHI and DNI estimation has been observed after calibration, which brings down the Normalized Root Mean Square Error (NRMSE) to 4% and 11.70% for GHI and DNI respectively as compared to the non-calibrated satellite data estimation.

The use of organoid models in biomedical research has grown substantially since their inception. As they gain popularity among scientists seeking more complex and biologically relevant systems, there is a direct need to expand and clarify potential uses of such systems in diverse experimental contexts. Herein we outline a high-content screening (HCS) platform that allows researchers to screen drugs or other compounds against three-dimensional (3D) cell culture systems in a multi-well format (384-well). Furthermore, we compare the quality of robotic liquid handling with manual pipetting and characterize and contrast the phenotypic effects detected by confocal imaging and biochemical assays in response to drug treatment. We show that robotic liquid handling is more consistent and amendable to high throughput experimental designs when compared to manual pipetting due to improved precision and automated randomization capabilities. We also show that image-based techniques are more sensitive to detecting phenotypic changes within organoid cultures than traditional biochemical assays that evaluate cell viability, supporting their integration into organoid screening workflows. Finally, we highlight the enhanced capabilities of confocal imaging in this organoid screening platform as they relate to discerning organoid drug responses in single-well co-cultures of organoids derived from primary human biopsies and patient-derived xenograft (PDX) models. Altogether, this platform enables automated, imaging-based HCS of 3D cellular models in a non-destructive manner, opening the path to complementary analysis through integrated downstream methods.

IntroductionParkinson’s disease (PD) is a neurodegenerative movement disorder causing severe disability and cognitive impairment as the disease progresses. It is necessary to develop biomarkers for cognitive decline in PD for earlier detection and prediction of disease progression.MethodsWe reviewed literature which used artificial intelligence-based techniques, which can be more sensitive than other analyses, to determine potential biomarkers for cognitive impairment in PD.ResultsWe found that combining biomarker types, including those from neuroimaging and biofluids, resulted in higher accuracy. Focused analysis on each biomarker type revealed that using structural and functional magnetic resonance imaging (MRI) resulted in accuracy and area under the curve (AUC) values above 80%/0.80, and that beta-amyloid-42 and tau were able to classify PD subjects by cognitive function with accuracy and AUC values above 90%/0.90.DiscussionWe can conclude that applying both blood-based and imaging-based biomarkers may improve diagnostic accuracy and prediction of cognitive impairment in PD.

Abstract Background Type 2 diabetes mellitus (T2DM) is recognized an independent risk factor for chronic kidney disease (CKD). The precise contribution and differential response to treatment strategies to reduce kidney dysfunction, depending on whether obesity is present alongside T2DM or not, remain to be fully clarified. Our objective was to improve our understanding of how obesity contributes to kidney function in patients with T2DM and coronary heart disease (CHD), who are highly predisposed to CKD, to assign the most effective dietary approach to preserve kidney function. Methods 1002 patients with CHD and estimated glomerular filtration rate (eGFR)≥30 ml/min/1.73m2, were randomized to consume a Mediterranean diet (35% fat, 22% MUFA, 55% carbohydrates). Patients were classified into four groups according to the presence of T2DM and/or obesity at baseline: Non-Obesity/Non-T2DM, Obesity/Non-T2DM, Non-Obesity/T2DM and Obesity/T2DM. Abstract Background Telomere Length (TL), a marker of cellular aging, holds promise as a biomarker to elucidate the molecular mechanism of diabetes. This study aimed to investigate whether shorter telomeres are associated with a higher risk of type 2 diabetes mellitus (T2DM) incidence in patients with coronary heart disease; and to determine whether the most suitable dietary patterns, particularly a Mediterranean diet or a low-fat diet, can mitigate the development of diabetes in these patients after a follow-up period of five years. Methods The CORonary Diet Intervention with Olive oil and cardiovascular PREVention study (CORDIOPREV study) was a single-centre, randomised clinical trial done at the Reina Sofia University Hospital in Córdoba, Spain. Patients with established coronary heart disease (aged 20–75 years) were randomly assigned in a 1:1 ratio by the Andalusian School of Public Health to receive two healthy diets. Clinical investigators were masked to treatment assignment; participants were not. Quantitative-PCR was used to assess TL measurements. Findings 1002 patients (59.5 ± 8.7 years and 82.5% men) were enrolled into Mediterranean diet (n = 502) or a low-fat diet (n = 500) groups. Abstract Clinical, cognitive, and atrophy characteristics depending on sex have been previously reported in Parkinson’s disease (PD). However, though sex differences in cortical gray matter measures in early drug naïve patients have been described, little is known about differences in cortical thickness (CTh) as the disease advances. Our multi-site sample comprised 211 non-demented PD patients (64.45% males; mean age 65.58 ± 8.44 years old; mean disease duration 6.42 ± 5.11 years) and 86 healthy controls (50% males; mean age 65.49 ± 9.33 years old) with available T1-weighted 3 T MRI data from four international research centers. Sex differences in regional mean CTh estimations were analyzed using generalized linear models. The relation of CTh in regions showing sex differences with age, disease duration, and age of onset was examined through multiple linear regression. PD males showed thinner cortex than PD females in six frontal (bilateral caudal middle frontal, bilateral superior frontal, left precentral and right pars orbitalis), three parietal (bilateral inferior parietal and left supramarginal), and one limbic region (right posterior cingulate). In PD males, lower CTh values in nine out of ten regions were associated with longer disease duration and older age, whereas in PD females, lower CTh was associated with older age but with longer disease duration only in one region. Overall, male patients show a more widespread pattern of reduced CTh compared with female patients. Disease duration seems more relevant to explain reduced CTh in male patients, suggesting worse prognostic over time. Further studies should explore sex-specific cortical atrophy trajectories using large longitudinal multi-site data.

Abstract Background Cardiovascular diseases (CVD), including coronary heart disease (CHD), display a higher prevalence in men than women. This study aims to evaluate the variations in the intestinal microbiota between men and women afflicted with CHD and delineate these against a non-CVD control group for each sex. Methods Our research was conducted in the framework of the CORDIOPREV study, a clinical trial which involved 837 men and 165 women with CHD. We contrasted our findings with a reference group of 375 individuals (270 men, 105 women) without CVD. The intestinal microbiota was examined through 16S metagenomics on the Illumina MiSeq platform and the data processed with Quiime2 software. Results Our results showed a sex-specific variation (beta diversity) in the intestinal microbiota, while alpha-biodiversity remained consistent across both sexes. Linear discriminant analysis effect size (LEfSe) analysis revealed sex-centric alterations in the intestinal microbiota linked to CVD. Moreover, using random forest (RF) methodology, we identified seven bacterial taxa—g_UBA1819 (Ruminococcaceae), g_Bilophila, g_Subdoligranulum, g_Phascolarctobacterium, f_Barnesiellaceae, g_Ruminococcus, and an unknown genus from the Ruminococcaceae family (Ruminococcaceae incertae sedis)—as key discriminators between men and women diagnosed with CHD. The same taxa also emerged as critical discriminators between CHD-afflicted and non-CVD individuals, when analyzed separately by sex. Conclusion Our findings suggest a sex-specific dysbiosis in the intestinal microbiota linked to CHD, potentially contributing to the sex disparity observed in CVD incidence. Currently, roughly one third of college students fit the federal government’s definition of first-generation college student status, meaning that neither parent has earned a baccalaureate degree (RTI International, 2023). This study utilized archival data at an access institution in the southeastern United States in a causal comparative study using binary logistic regression analysis to determine if first-generation college student status, gender, socioeconomic status, and academic preparedness are predictors for six-year graduation rates. Findings from this quantitative study determined that gender, socioeconomic status, and academic preparedness were significant predictors for graduation within six years of matriculation at the institution. A future qualitative study may provide context for the student experience and what factors influenced student success. These findings are intended to help administrators understand their student population and implement intervention strategies to increase graduation outcomes.

Abstract Infant mortality remains high and uneven in much of sub-Saharan Africa. Even low-cost, highly effective therapies can only save lives in proportion to how successfully they can be targeted to those children who, absent the treatment, would have died. This places great value on maximizing the accuracy of any targeting or means-testing algorithm. Yet, the interventions that countries deploy in hopes of reducing mortality are often targeted based on simple models of wealth or income or a few additional variables. Examining 22 countries in sub-Saharan Africa, we illustrate the use of flexible (machine learning) risk models employing up to 25 generally available pre-birth variables from the Demographic and Health Surveys. Using these models, we construct risk scores such that the 10 percent of the population at highest risk account for 15-30 percent of infant mortality, depending on the country. Successful targeting in these models turned on several variables other than wealth, while models that employ only wealth data perform little or no better than chance. Consequently, employing such data and models to predict high-risk births in the countries studied flexibly could substantially improve the targeting and thus the life-saving potential of existing interventions.

The increasing costs of traditional protein sources, such as soybean meal (SBM), have prompted interest in alternative feeds for ruminants. Non-protein nitrogen (NPN) sources, like urea, offer a cost-effective alternative by enabling rumen microorganisms to convert NPN into microbial protein, which is crucial for ruminant nutrition. However, the rapid hydrolysis of urea in the rumen can result in excessive ammonia (NH3) production and potential toxicity. Slow-release urea (SRU) has been developed to mitigate these issues by gradually releasing nitrogen, thereby improving nutrient utilization and reducing NH3 toxicity risks. This review explores SRU’s development, types, mechanisms, and benefits, highlighting its potential to enhance ruminal fermentation, microbial protein synthesis, and overall feed efficiency. SRU formulations include polymer-coated urea, lipid-coated urea, calcium-urea, starea, and zeolite-impregnated urea, each designed to control nitrogen release and minimize adverse effects. Studies have demonstrated that SRU can improve microbial nitrogen efficiency and reduce nitrogen losses, although results regarding feed intake, digestibility, and milk yield are mixed. These discrepancies indicate that factors such as SRU type, diet formulation, and animal breed may influence outcomes. Continued research is essential to optimize SRU applications, aiming to enhance ruminant production, economic viability, and environmental stewardship.

Abstract This work proposes a system complexity metric and its application to Intensive Care Unit (ICU) system. The methodology for applying said complexity metric comprises: (i) parameters sensitivity indices calculation, (ii) mapping connections dynamics between system components, and (iii) system’s complexity calculation. After simulating the ICU computer model and using the proposed methodology, we obtained results regarding: number of admissions, number of patients in the queue, length of stay, beds in use, ICU performance, and system complexity values (in regular or overloaded operation). As the number of patients in the queue increased, the ICU system complexity also increased, indicating a need for policies to promote system robustness.

Abstract Objective We aimed to identify a lipidic profile associated with type 2 diabetes mellitus (T2DM) development in coronary heart disease (CHD) patients, to provide a new, highly sensitive model which could be used in clinical practice to identify patients at T2DM risk. Methods This study considered the 462 patients of the CORDIOPREV study (CHD patients) who were not diabetic at the beginning of the intervention. In total, 107 of them developed T2DM after a median follow-up of 60 months. They were diagnosed using the American Diabetes Association criteria. A novel lipidomic methodology employing liquid chromatography (LC) separation followed by HESI, and detection by mass spectrometry (MS) was used to annotate the lipids at the isomer level. The patients were then classified into a Training and a Validation Set (60–40). Next, a Random Survival Forest (RSF) was carried out to detect the lipidic isomers with the lowest prediction error, these lipids were then used to build a Lipidomic Risk (LR) score which was evaluated through a Cox. Finally, a production model combining the clinical variables of interest, and the lipidic species was carried out. Results LC-tandem MS annotated 440 lipid species. From those, the RSF identified 15 lipid species with the lowest prediction error. These lipids were combined in an LR score which showed association with the development of T2DM. The LR hazard ratio per unit standard deviation was 2.87 and 1.43, in the Training and Validation Set respectively. Likewise, patients with higher LR Score values had lower insulin sensitivity (P = 0.006) and higher liver insulin resistance (P = 0.005). The receiver operating characteristic (ROC) curve obtained by combining clinical variables and the selected lipidic isomers using a generalised lineal model had an area under the curve (AUC) of 81.3%. Conclusion Our study showed the potential of comprehensive lipidomic analysis in identifying patients at risk of developing T2DM. In addition, the lipid species combined with clinical variables provided a new, highly sensitive model which can be used in clinical practice to identify patients at T2DM risk. Moreover, these results also indicate that we need to look closely at isomers to understand the role of this specific compound in T2DM development. Trials registration NCT00924937.

This study aimed to evaluate the sulfur hexafluoride (SF6) tracer technique for estimating methane (CH4) emissions in dual-flow continuous culture systems (DFCCS). In experiment 1 (Exp1), fermenters were filled with water, and known CH4 concentrations (0, 1.35, 2.93, or 4.43 g/d) were injected using permeation tubes with SF6 release rates (3.30 or 9.65 mg/d). Headspace gas was collected using canisters, and the SF6 technique estimated CH4 recovery. Experiment 2 (Exp2) involved a DFCCS fermentation trial with ruminal fluid from three Holstein cows, testing diets with soybean meal or its partial replacement (50%) by Chlorella or Spirulina. Headspace gas was collected at intervals post-feeding. Standard curves for SF6 and CH4 quantification were inadequate for DFCCS samples, with the CH4:SF6 ratio differing from standards, indicating the data needs further SF6 release rate evaluation. In Exp1, a high correlation (r = 0.97) was found between infused and calculated CH4, indicating good repeatability. Low and high SF6 rates performed similarly at low CH4 infusion, but high SF6 overestimated CH4 at high infusion. Exp2 showed CH4 emissions irrespective of SF6 rate and indicated reduced CH4 emissions and increased NDF degradation with algae-containing diets. Further evaluation of the SF6 tracer technique is warranted for DFCCS.

Colleges and universities across the United States face continual pressure to meet enrollment and retention goals, as budgets in this performance-based environment continue to become more important. On-campus student involvement, such as in undergraduate leadership development programs, has been shown to have a positive influence on both student retention and success. A survey was utilized to examine leadership self-efficacy and engagement of undergraduate students that participated in campus-based leadership development programs and explore some motivators (contributing factors) and barriers (detracting factors) to involvement in those programs. One emergent theme within contributing factors to participation was alignment with personal goals (74.7%), whereas, a theme for detracting factors was lack of time to invest in the leadership opportunity (51.1%). Exploration of which factors contributed to and detracted from leadership development participation showed that contributing factors were a positive and significant predictor of leadership self-efficacy. For every one unit increase in contributing factors, leadership self-efficacy score increased by β - = .38 standard deviations. This study encourages leadership educators to examine their own leadership development programs and build recruitment strategies to increase engagement among student demographics such as male students, non-White students, and first-generation college students. In the future, researchers could consider including students that did not participate in leadership programs to gain more valuable insights on the motivators and the barriers that students face to participation in these programs.

Abstract Elevated levels of ruminal lipopolysaccharides (LPS) have been linked to ruminal acidosis; however, they result in reduced endotoxicity compared to LPS derived from species like Escherichia coli. Additionally, there is a knowledge gap on the potential effect of LPS derived from ruminal microbiome on ruminal bacteria species whose abundance is associated with ruminal acidosis. The objective of this study was to evaluate the effects of LPS-free anaerobic water (CTRL), E. coli-LPS (E. COLI), ruminal-LPS (RUM), and a 1:1 mixture of E. coli and ruminal-LPS (MIX) on the growth characteristics and fermentation end products of lactate-producing bacteria (Streptococcus bovis JB1, Selenomonas ruminantium HD4) and lactate-utilizing bacterium (Megasphaera elsdenii T81). The growth characteristics were predicted based on the logistic growth model, the ammonia concentration was determined by the phenolic acid/hypochlorite method and organic acids were analyzed with high performance liquid chromatography. Results indicate that, compared to the CTRL, the maximum specific growth rate of S. bovis JB1 decreased by approximately 19% and 23% when RUM and MIX were dosed, respectively. In addition, acetate and lactate concentrations in Se. ruminantium HD4 were reduced by approximately 30% and 18%; respectively, in response to MIX dosing. Compared to CTRL, lactate concentration from S. bovis JB1 was reduced approximately by 31% and 22% in response to RUM and MIX dosing; respectively. In summary, RUM decreased the growth and lactate production of some lactate-producing bacteria, potentially mitigating the development of subacute ruminal acidosis by restricting lactate availability to some lactate-utilizing bacteria that metabolize lactate into VFAs thus further contributing to the development of acidosis. Also, RUM did not affect Megasphaera elsdenii T81 growth.

Abstract The accumulation of beta amyloid in the brain has a complex and poorly understood impact on the progression of Parkinson’s disease pathology and much controversy remains regarding its role, specifically in cognitive decline symptoms. Some studies have found increased beta amyloid burden is associated with worsening cognitive impairment in Parkinson’s disease, especially in cases where dementia occurs, while other studies failed to replicate this finding. To better understand this relationship, we examined a cohort of 25 idiopathic Parkinson’s disease patients and 30 healthy controls from the Parkinson’s Progression Marker Initiative database. These participants underwent [18F]Florbetaben positron emission tomography scans to quantify beta amyloid deposition in 20 cortical regions. We then analyzed this beta amyloid data alongside the longitudinal Montreal Cognitive Assessment scores across 3 years to see how participant’s baseline beta amyloid levels affected their cognitive scores prospectively. The first analysis we performed with these data was a hierarchical cluster analysis to help identify brain regions that shared similarity. We found that beta amyloid clusters differently in Parkinson’s disease patients compared to healthy controls. In the Parkinson’s disease group, increased beta amyloid burden in cluster 2 was associated with worse cognitive ability, compared to deposition in clusters 1 or 3. We also performed a stepwise linear regression where we found an adjusted R2 of 0.495 (49.5%) in a model explaining the Parkinson’s disease group’s Montreal Cognitive Assessment score 1-year post-scan, encompassing the left gyrus rectus, the left anterior cingulate cortex, and the right parietal cortex. Taken together, these results suggest regional beta amyloid deposition alone has a moderate effect on predicting future cognitive decline in Parkinson’s disease patients. The patchwork effect of beta amyloid deposition on cognitive ability may be part of what separates cognitive impairment from cognitive sparing in Parkinson’s disease. Thus, we suggest it would be more useful to measure beta amyloid burden in specific brain regions rather than using a whole-brain global beta amyloid composite score and use this information as a tool for determining which Parkinson’s disease patients are most at risk for future cognitive decline.

Background: Split-belt treadmill (SBTM) training has been proposed to improve gait symmetry and overall gait performance of patients with Parkinson's disease (PD). Objectives: To determine whether patient's baseline features affect gait adaptation to SBTM in PD with freezing of gait (FOG). Methods: Twenty participants with idiopathic PD and treatment-resistant FOG underwent several clinical assessments including the Toronto Cognitive Assessment (TorCA) prior to treadmill training. This study carries out snowball sampling technique to document supply chain map and determine its performance in terms of efficiency and responsiveness of the goat industry supply chain in Northwestern Leyte. Results revealed that there were three chains identified in the area, namely: individual farmers who market their produced goats to a terminal goat market, the two registered associations, Tabango Integrated Goat Entrepreneurs and Raisers (TIGER) and Villaba Association of Goat Entrepreneurs (VILLAGE). Detailed supply chain mapping shows that goats from the farmers’ point are brought to different places such as Cebu, Eastern Samar, Hinunangan Southern Leyte and Palompon, Leyte. Supply chain in the associations is generally simple, as it only constitutes direct seller-buyer market, while auction-market driven is a distinct configuration. Goat trading formally takes place once a week in the identified terminal market which were participated by processors, assembler-traders and end consumers. A simple supply and demand analysis revealed that there is a lack of the volume requirements by institutional and contract buyers, indicating an unmet demand and shortage of supply. This unmet demand opens a door of opportunity for goat production intensification in the area. Although profit margins were generated, logistical issues suggest production inefficiencies along the supply chain. Hence, this baseline information on goat supply chain provides empirical evidence on the need for improvement in the production to consumption in order to enhance the productivity of the goat industry in Northwestern Leyte, Philippines.

Abstract Background Recovery of hand function is crucial for the independence of people with spinal cord injury (SCI). Wearable devices based on soft robotics (SR) or functional electrical stimulation (FES) have been employed to assist the recovery of hand function both during activities of daily living (ADLs) and during therapy. However, the implementation of these wearable devices has not been compiled in a review focusing on the functional outcomes they can activate/elicit/stimulate/potentiate. This narrative review aims at providing a guide both for engineers to help in the development of new technologies and for clinicians to serve as clinical guidelines based on the available technology in order to assist and/or recover hand function in people with SCI. Methods A literature search was performed in Scopus, Pubmed and IEEE Xplore for articles involving SR devices or FES systems designed for hand therapy or assistance, published since 2010. Only studies that reported functional outcomes from individuals with SCI were selected. The final collections of both groups (SR and FES) were analysed based on the technical aspects and reported functional outcomes. Results A total of 37 out of 1101 articles were selected, 12 regarding SR and 25 involving FES devices. Most studies were limited to research prototypes, designed either for assistance or therapy. From an engineering perspective, technological improvements for home-based use such as portability, donning/doffing and the time spent with calibration were identified. From the clinician point of view, the most suitable technical features (e.g., user intent detection) and assessment tools should be determined according to the particular patient condition. A wide range of functional assessment tests were adopted, moreover, most studies used non-standardized tests. Conclusion SR and FES wearable devices are promising technologies to support hand function recovery in subjects with SCI. Technical improvements in aspects such as the user intent detection, portability or calibration as well as consistent assessment of functional outcomes were the main identified limitations. These limitations seem to be be preventing the translation into clinical practice of these technological devices created in the laboratory.

Artificial Intelligence (AI) brings enormous opportunities into learning, teaching, and assessment processes. Among them, it is convenient to explore its ability to channel students' creativity, which is described as a basic competence in the training of people with both the OECD and the recent Spanish LOMLOE law pointing to the need to foster it in educational settings. In this context, the objective of this research is to explore the creative potential of prospective elementary school teachers related to storytelling, via a project including the rational use of AI generative tools. A combination of qualitative and quantitative instruments was used to get insight on the implications of those AI tools in the creative process and to gain understanding on the concerns of prospective teachers about AI at both their training and future teaching practice. The results show the potential of AI from an educational point of view, specially in self-assessment and co-evaluation processes, since it allows confronting not only the result of the creative task, but also the process itself by reflecting on the asked questions. Finally, the importance of continuing research on the ability to ask questions (a creative skill in itself) in the new context of AI is discussed.

This study delves into the intricate challenges surrounding incorporating information and communication technologies (ICTs) in education, particularly in regions characterized by digital exclusion, such as rural areas. By recognizing the potential of ICTs to support and expand student learning opportunities, this research explores the necessity for effective implementation through tailored professional development aligned with teachers' contextual conditions and needs. In addressing these challenges, the study explores the concept of technological appropriation, underscoring the significance of individual and contextual elements in adapting ICTs to users' specific needs and environments. Through unstructured face-to-face interviews with 21 teachers immersed in a digitally vulnerable rural context, this research unveils insights into how educators appropriate technologies to enhance student learning. The findings of this study contribute to the development of educational practices tailored to rural contexts, focusing on providing a meaningful and effective learning experience for students. By shedding light on teachers' strategies for navigating the digital landscape in challenging environments, the research aims to inform policies and practices that bridge the technological gap, ultimately fostering equitable access and enhanced educational outcomes in rural settings.

Environments or learning spaces in school classrooms contribute to the improvement of educational processes. Specifically, in the Early Childhood Education, teachers can design learning spaces, which fosters an environment of safety and autonomy for the students of the Early Childhood Education. This study explores the teachers' narratives concerning the meanings they assign to the school space, the criteria by which they design and transform it, and its connection to model of educational. We have collected 34 stories of pre-primary education teachers through a semi-structured interview in which they had to reflect on this pedagogical design. As most relevant result we identify a model sustained under constructivist or cognitive criteria, rather than a dialogical one. We conclude that this epistemological perspective of the construction of the school environment should be oriented towards a more participatory and co-constructed model of the educational space.

In order to evaluate whether telomere maintenance is associated with type 2 diabetes remission, newly diagnosed type 2 diabetes patients without glucose-lowering treatment (183 out of 1002) from the CORDIOPREV study (NCT00924937) were randomized to consume a Mediterranean or low-fat diet. Patients were classified as Responders, those who reverted from type 2 diabetes during the 5 years of dietary intervention (n = 69), and Non-Responders, who did not achieve diabetes remission by the end of the follow-up period (n = 104). We found no differences in diabetes remission between the two diets, and we determined telomere length (TL) by measuring qPCR, telomerase activity using the TRAP assay, and direct redox balance based on the ratio of reduced glutathione (GSH) to oxidized glutathione (GSSH) via colorimetric assay. Responders exhibited higher baseline TL in comparison with Non-Responders (p = 0.040), and a higher TL at baseline significantly predicted a higher probability of type 2 diabetes remission (OR 2.13; 95% CI, 1.03 to 4.41). After the dietary intervention, Non-Responders showed significant telomere shortening (−0.19, 95% CI −0.32 to 0.57; p = 0.005). Telomere shortening was significantly pronounced in type 2 diabetes patients with a worse profile of insulin resistance and/or beta-cell functionality: high hepatic insulin resistance fasting, a high disposition index (−0.35; 95% CI, −0.54 to −0.16; p < 0.001), and a low disposition index (−0.25; 95% CI, −0.47 to −0.01; p = 0.037). In addition, changes in TL were correlated to the GSH/GSSG ratio. Responders also showed increased telomerase activity compared with baseline (p = 0.048), from 0.16 (95% CI, 0.08 to 0.23) to 0.28 (95% CI, 0.15 to 0.40), with a more marked increase after the dietary intervention compared with Non-Responders (+0.07; 95% CI, −0.06–0.20; p = 0.049). To conclude, telomere maintenance may play a key role in the molecular mechanisms underlying type 2 diabetes remission in newly diagnosed patients. However, further larger-scale prospective studies are necessary to corroborate our findings.

Abstract This study aimed to evaluate the effects of Saccharomyces cerevisiae and Megasphaera elsdenii as direct fed microbials (DFM) in beef cattle finishing diets to alleviate acute ruminal lactic acidosis in vitro. A dual-flow continuous culture system was used. Treatments were a Control, no DFM; YM1, S. cerevisiae and M. elsdenii strain 1; YM2, S. cerevisiae and M. elsdenii strain 2; and YMM, S. cerevisiae and half of the doses of M. elsdenii strain 1 and strain 2. Each DFM dose had a concentration of 1 × 108 CFU/mL. Four experimental periods lasted 11 days each. For the non-acidotic days (day 1–8), diet contained 50:50 forage to concentrate ratio. For the challenge days (day 9–11), diet contained 10:90 forage to concentrate ratio. Acute ruminal acidosis was successfully established. No differences in pH, d-, l-, or total lactate were observed among treatments. Propionic acid increased in treatments containing DFM. For N metabolism, the YMM treatment decreased protein degradation and microbial protein synthesis. No treatment effects were observed on NH3–N concentration; however, efficiency of N utilization by ruminal bacteria was greater than 80% during the challenge period and NH3–N concentration was reduced to approximately 2 mg/dL as the challenge progressed.

Abstract Transcription Factor 4 (TCF4) has been associated with autism, schizophrenia, and other neuropsychiatric disorders. However, how pathological TCF4 mutations affect the human neural tissue is poorly understood. Here, we derive neural progenitor cells, neurons, and brain organoids from skin fibroblasts obtained from children with Pitt-Hopkins Syndrome carrying clinically relevant mutations in TCF4. We show that neural progenitors bearing these mutations have reduced proliferation and impaired capacity to differentiate into neurons. We identify a mechanism through which TCF4 loss-of-function leads to decreased Wnt signaling and then to diminished expression of SOX genes, culminating in reduced progenitor proliferation in vitro. Moreover, we show reduced cortical neuron content and impaired electrical activity in the patient-derived organoids, phenotypes that were rescued after correction of TCF4 expression or by pharmacological modulation of Wnt signaling. This work delineates pathological mechanisms in neural cells harboring TCF4 mutations and provides a potential target for therapeutic strategies for genetic disorders associated with this gene.

Context: Transperineal prostate biopsy is associated with a significantly lower risk of infectious complications than the transrectal approach. In fact, the risk of infectious complications with transperineal prostate biopsy is so low that the utility of administering periprocedural antibiotics with this procedure has come under question. Objective: To perform a systematic review and meta-analysis to assess for differences in the rates of infectious complications (septic, nonseptic, and overall) after performing transperineal prostate biopsy with and without the administration of periprocedural antibiotic prophylaxis. Evidence acquisition: Three electronic databases (PubMed, Embase, and MEDLINE) were searched, and studies were included if they included patients who underwent transperineal prostate biopsy, were published after January 2000, included information on periprocedural antibiotic administration, and reported postbiopsy complications. Preferred Reporting Items for Systematic Reviews and Meta-analyses and Agency for Healthcare Research and Quality guidelines were utilized. Evidence synthesis: A total of 106 unique studies describing 112 cohorts of patients were identified, of which 98 (37 805 men) received antibiotic prophylaxis and 14 (4772 men) did not receive it. All patients were included in the analysis of septic complications. In total, there were 19/37 805 (0.05%) episodes of sepsis in the group of men who received antibiotics, which was similar to the no antibiotic group with 4/4772 (0.08%) episodes (p = 0.2). For overall infections (septic plus nonseptic), there were 403/29 880 (1.35%) versus 58/4772 (1.22%) events among men with evaluable data who received and did not receive antibiotic prophylaxis, respectively (p = 0.8). En el presente estudio se examinó la relación entre las medidas objetivas y subjetivas de metacognición, los rasgos de personalidad de los Cinco Grandes (extraversión, amabilidad, responsabilidad, neuroticismo y apertura) y el género. Una muestra de conveniencia de 352 estudiantes completó el Inventario de Conciencia Metacognitiva, el instrumento Adjetivos para Evaluar la Personalidad y completaron 3 pruebas de dominio específico (vocabulario, probabilidades, plegado de papel) junto con la confianza en los juicios de desempeño para cada reactivo en estas pruebas. A través de una combinación de estadísticas descriptivas, correlaciones bivariadas de orden cero, regresión múltiple simultánea y un análisis multivariante de covarianza, los hallazgos indican que las medidas objetivas y subjetivas de metacognición están débilmente relacionadas y que las medidas subjetivas de metacognición están más fuertemente relacionadas con los rasgos de personalidad. Además, la responsabilidad y la apertura fueron los únicos rasgos de personalidad que predijeron positivamente la metacognición. Finalmente, el género afectó las medidas subjetivas y objetivas de metacognición, incluso después de controlar por tipo de universidad (privada, pública) y percepciones del desempeño académico (alto, bajo) de tal manera que los hombres fueron más precisos y menos sesgados en su monitoreo que las mujeres solo en el razonamiento matemático y reportaron una mayor conciencia de su conocimiento y regulación de la cognición que sus contrapartes femeninas. Los hallazgos respaldan la necesidad de comprender mejor cómo los rasgos de personalidad y el género afectan las habilidades de aprendizaje autorregulado como la metacognición para mejorar la práctica educativa.

La metacognición se define como un proceso reflexivo que permite a las personas monitorear, controlar y autorregular su comportamiento. Sin embargo, la metacognición rara vez se ha estudiado con variables más allá del contexto educativo, como variables sociales, conductuales o clínicas. Así, el propósito del presente estudio fue explorar la relación entre las variables sociales y la conciencia metacognitiva subjetiva y si estas mismas variables sociales predicen componentes de la conciencia metacognitiva (conocimiento y regulación de la cognición). Con una muestra de estudiantes universitarios (N = 246), los resultados revelaron que no solo existen correlaciones significativas entre algunas variables sociales y ambos componentes de la conciencia metacognitiva, sino que esas mismas variables sociales predijeron la conciencia metacognitiva. Se discuten las implicaciones de estos resultados en contextos educativos y clínicos.
