1. Response of rubber tree clones to black crust.

Abstract

The occurrence of black crust, a disease previously considered secondary, has become a cause for concern due to early leaf fall in rubber trees (Hevea brasiliensis). This study aimed to identify sources of resistance to black crust in 21 clones of rubber trees in a clonal garden. To achieve this, the disease severity was evaluated through eight assessments conducted over a 2‐year period. In each evaluation, 15 leaflets from three different parts of the plant (bottom, middle third and crown) were randomly collected, resulting in a total of 45 leaflets per plant. These leaflets were then taken to the laboratory, and the software Leaf Doctor was used to measure the percentage of leaf area exhibiting symptoms of black crust. The results were analysed using the Scott‐Knott test at a 5% probability level. The clones with the highest infected leaf area observed during the evaluations were IAC300, IAC418, IAC503, PB311 and RRIM600. Clone IAC511 did not show any symptoms of the disease in any of the evaluations. Clones IAC301, IAC411, IAC501, IAC507 and IRCA111 exhibited low percentages of affected leaf areas. Additionally, this study revealed the progression of the disease over the evaluation period. The infection initially starts on the youngest leaves (crown) in March and subsequently intensifies in the middle third of the plant, corresponding to the youngest leaf previously infected in the crown. Furthermore, environmental data collected in the experimental area over the 2‐year evaluation period indicated that the disease manifests with greater severity when the average temperatures fall below 25°C after periods of heavy rainfall, followed by dry periods with low relative humidity. Abstract

The objective of this work was to evaluate the development of Davidiella sp. and its asexual form, Cladosporium sp., under different environmental conditions in the rubber tree (Hevea brasiliensis). Rubber tree leaves were inoculated with a spore suspension and kept in a humid chamber under different temperatures and wetness periods. The behavior of the fungi was evaluated using a scanning electron microscope (SEM) and an ultraviolet light microscope (UV). In the images obtained in SEM, four hours after inoculation of the fungus, it was possible to verify the germination and penetration of conidia at temperatures of 10 to 20 °C. The formation of conidiophores was verified from six hours after inoculation, indicating that it is in the reproductive period. In the sexual phase, in SEM, from four hours after inoculation, it was possible to verify the formation of small protuberances at temperatures between 10 and 20 °C. These black dots evolve into circular, protruding black spots, like the symptoms of black crust, with apparent spore formation on them. The data obtained from the UV analyses corroborate those from SEM, showing that the fungus has good development in its two phases between temperatures of 20 and 25 °C and that the period of wetness on the leaf can contribute to the initial development of the pathogen. Apesar de o país não figurar entre os maiores produtores de látex no planeta, a heveicultura possui características positivas no aspecto socioambiental como a geração de empregos, cultivo em pequenas e médias propriedades e mitigação da emissão de gases do efeito estufa. Atualmente o Estado de São Paulo é o maior produtor de borracha natural do país, sendo que a área ocupada pelos seringais apresenta crescimento nos últimos anos, porém alguns fatores limitantes como os problemas fitossanitários, como a antracnose, limitam uma expansão ainda maior da cultura. A antracnose é uma doença fúngica causada por diferentes espécies do gênero Colletotrichum que tem como principais sintomas manchas foliares e danos em pecíolos sendo que, em casos extremos, pode provocar queda de folhas, fato que leva a redução da produção de látex. Devido a alta variabilidade genética do patógeno e as dificuldades encontradas no controle da doença, este trabalho teve como objetivo caracterizar 17 isolados de Colletotrichum obtidos a partir lesões características de antracnose em folíolos de seringueira coletados em localidades distintas, e testar a resistência de diferentes clones de seringueira a doença. A caracterização dos isolados, realizada através de sequenciamento de parte do ITS, fatores morfo-culturais e patogênicos, indicou a presença de isolados causadores da doença pertencentes aos complexos C. acutatum e C. gloeosporioides, sendo verificada diferença na agressividade entre isolados. O teste de resistência clonal indicou graus de susceptibilidade a doença distintos entre os materiais estudados, assim como variação na resistência de cada clones aos 3 diferentes isolados de Colletotrichum utilizados. Os resultados obtidos confirmam a importância da identificação correta dos isolados em áreas com presença da doença e/ou expansão da cultura, e da implementação do clone adequado em relação aos patógenos presentes na área, além de servir como base para estudos futuros para o desenvolvimento de materiais genéticos resistentes a doença. The culture of the rubber tree (Hevea brasiliensis) is of great importance within Brazilian agribusiness. Although the country is not among the largest producers of latex on the planet, rubber farming has positive characteristics in the socio-environmental aspect, such as job creation, cultivation in small and medium-sized properties and mitigation of greenhouse gas emissions. Currently, the State of São Paulo is the largest producer of natural rubber in the country, and the area occupied by rubber trees has grown in recent years, but some limiting factors such as phytosanitary problems, such as anthracnose, limit an even greater expansion of the crop. Anthracnose is a fungal disease caused by different species of the genus Colletotrichum whose main symptoms are leaf spots and damage to petioles and, in extreme cases, can cause leaf fall, a fact that leads to reduced latex production. Due to the high genetic variability of the pathogen and the difficulties encountered in controlling the disease, this study aimed to characterize 17 Colletotrichum isolates obtained from characteristic anthracnose lesions in rubber tree leaflets collected in different locations, and test the resistance of different rubber tree clones to disease. The characterization of the isolates, carried out by sequencing the ITS region, morphocultural and pathogenic factors, indicated the presence of disease-causing isolates belonging to the C. acutatum and C. gloeosporioides complexes, with a difference in aggressiveness among isolates. The clonal resistance test indicated different degrees of susceptibility to disease among the studied materials, as well as variation in the resistance of each clone to the 3 different Colletotrichum isolates used. The results obtained confirm the importance of the correct identification of the isolates in areas with the presence of the disease and / or expansion of the culture, and of the implementation of the appropriate clone in relation to the pathogens present in the area, in addition to serving as a basis for future studies for the development of genetic materials resistant to disease. In initial development, cotyledon herbivory makes the majority of species more susceptible to the environment. This study aimed to reproduce heibivory situations occurring in the field, caused by insect pests, through manual partial and total removal of the cotyledons, in order to evaluate the deleterious effects on the initial development of soybean [Glycine max (L.) Merrill]. The experimental design was completely randomized and arranged in a 2 x 3 factorial scheme, with two soybean cultivars (BMX Potencia RR and Syngenta 1562 IPRO) and three situations involving removal of cotyledons (0%, 50% and 100%) in four replications, totalling 24 experimental plots. The partial or total removal of cotyledons caused a reduction in parameters related to the growth and initial development of soybean (stalk diameter, shoot length, root primary length, shoot dry mass and root dry mass), which is probably due to the nutritional and morphological changes documented in this study. In general, the Syngenta 1562 IPRO cultivar was more sensitive to cotyledon removal than the BMX Potencia RR cultivar. Abstract

RESUMO O fungo Ceratocystis cacaofunesta é um dos principais problemas na cultura do cacaueiro no Brasil. Para o cacaueiro há poucas alternativas para o controle da doença, sendo que a indução de resistência vem sendo testada como uma possibilidade de controle. Neste trabalho foram utilizados três indutores de resistências em três concentrações diferentes visando a indução de resistência do cacaueiro atacado por Ceratocystis sendo elas: glicose (0,1M, 0,5M e 1M), sacarose (0,1M, 0,5M, 1M) e acibenzolar-s-metílico (1ppm, 100ppm e 1000ppm). Além disso, amostras das plantas tratadas foram coletadas e analisadas para verificar a presença de grãos de amido. O único tratamento que não apresentou nenhuma morte de planta foi o acibenzolar-s-metílico a 1 ppm. Foi observado grãos de amido em todas as plantas tratadas, porém em quantidades diferentes. São necessários estudos mais detalhados para confirmar a efetividade do produto na indução de resistência de plantas de cacaueiro ao fungo Ceratocistys. ABSTRACT The fungus Ceratocystis cacaofunesta is one of the main problems in cacao culture in Brazil. For cocoa, there are few alternatives to control the disease, and induction of resistance has been tested as a possibility of control. In this study, three resistance inducers were used at three different concentrations with the aim of inducing resistance in the cacao tree attacked by Ceratocystis: glucose (0.1M, 0.5M and 1M), sucrose (0.1M, 0.5M, 1M) and acibenzolar-Î2-methyl (1ppm, 100ppm and 1000ppm). In addition, samples were collected from the treated plants and analyzed for the presence of starch grains. The only treatment that did not show any plant death was acibenzolar-s-methyl at a concentration of 1 ppm. Starch grains were observed in all treated plants, but in different amounts. More detailed studies are needed to confirm the effectiveness of the product in inducing resistance of cacao plants to the fungus Ceratocistys.
