Hierarchically built-up multicompartment nanoaggregate systems are of interest for, e.g., novel materials and medicine. Here we present a versatile strategy to generate and unambiguously characterize complex coacervate-core micelles by exploiting four different dendrimeric subcomponents as core-units. The resulting mesoscale structures have a hydrodynamic diameter of 50 nm and a core size of 33 nm, and host about thirty 6th generation polyamidoamine (PAMAM) dendrimers. We have used FRET (efficiency of similar to 0.2) between fluorescein and rhodamine moieties immobilized on separate PAMAM dendrimers (G6-F and G6-R, respectively) to prove synchronous encapsulation in the micelle core. Tuning the proximity of the FRET pair molecules either by varying the G6-F : G6-R ratio, or by co-assembling non-functionalized dendrimer (G6-E) in the core, reveals the optimal FRET efficiency to occur at a minimum of 70% loading with G6-F and G6-R. Additional co-encapsulation of 6th generation gold dendrimer-encapsulated nanoparticles (G6-Au) in the micelle core shows a dramatic reduction of the FRET efficiency, which can be restored by chemical etching of the gold nanoparticles from within the micellar core with thiols, leaving the micelle itself intact. This study reveals the controlled co-assembly of up to four different types of subcomponents in one single micellar core and concomitantly shows the wide variety of structures that can be made with a well-defined basic set of subcomponents. It is straightforward to design related strategies, to incorporate inside one micellar core, e.g., even more than 4 different dendrimers, or other classes of (macro)molecules, with different functional groups, other FRET pairs or different encapsulated metal nanoparticles.

The Lycurgus cup is worldwide admired in particular because of its fascinating dichroic property. The cup presents a green colour when the observer and the light source are on the same side (reflection), and a deep red colour when the observer and the light source are at opposite sides (transmission). This peculiar effect, which has perplexed scientists for centuries, was discovered to be due to the presence of nanoparticles in the glass. In particular, to two different metallic nanoparticles: silver nanoparticles (AgNP) and gold nanoparticles (AuNP). Here we show how dichroic silver nanoparticles and gold nanoparticles can be embedded in a 3D printable material in order to reproduce the same dichroic effect of the original Lycurgus cup.

Coacervate-core micelles are considered promising materials for several applications, from catalysis to drug delivery. However, oxidant-responsive coacervate-core micelles, able to undergo structural changes upon specific oxidation stimuli, are not well reported. Here, we present a novel ferrocene–dipicolinic acid derivative as redox-responsive subcomponent to be incorporated in cyclodextrin-based coacervate core micelles, C4Ms, with tuneable core structure and responsiveness towards H2O2 treatment. The Fc-C4Ms are formed combining three orthogonal supramolecular interactions, namely (i) metal-to-ligand coordination between europium(III) ions and dipicolinic acid molecules, (ii) host-guest interaction between beta cyclodextrins and ferrocenes and (iii) electrostatic coacervation interaction. The micelle stability against oxidation can be controlled by varying three main parameters: (a) the core-unit structure, from monomeric metal complexes to supramolecular oligomers, (b) the H2O2 equivalents and c) the ratio between redox-responsive and non-redox-responsive bislinker. The H2O2-responsive ferrocene-based systems might have an interesting application, e.g. reactive oxygen species-mediated drug delivery.

Metallic nanoparticles are small particles, with dimensions of less than 100 nm, which have unique characteristics. For example, the color of a nanoparticle solution is given by the surface plasmon resonance, in contrast to only absorbance of a molecule. Metallic nanoparticles have been used in the antiquity for the production of colored glass. Many of the medieval cathedral red windows are in fact, stained with gold nanoparticles. However, presenting just a red solution to a class and explaining that the solution contains gold nanoparticles can be disappointing as the red color can be obtained in many different ways without the need of nanoparticles. Something peculiar, like dichroism, can, on the other hand, shine the interest of an observer to the “nano-world”. Dichroic nanoparticles show different colors when illuminated from the front (scattering) or from the back (transmission). This effect is unique for nanoparticles and nanostructures in general and can generate the “WOW” effect in class or during public demonstrations. In this article we present a simple synthesis of dichroic silver and gold nanoparticles. Solutions of such nanoparticles are a stunning visual media for demonstrations, outreach and engaging audience in the “nano”world.

Plasma microcontact patterning (P mu CP) and replica molding were combined to make PDMS/glass microfluidic devices with beta-cyclodextrin (beta-CD) patterns attached covalently on the glass surface inside microchannels. The supramolecular reactivity, reusability and association constant of beta-CD with Cy5-Ad2 was tested by analyzing signal-to-noise ratios of patterns vs. spacing with fluorescence microscopy.

Polymer-based micellar assemblies are gaining increasing attention in the smart materials field, yet the design of micelles that show redox-responsive disassembly and, e. g., cargo release is still a challenge. To form redox-responsive micelles, we developed cyclodextrin-based coacervate core micelles that form under interplay of four orthogonal interactions: multivalent electrostatic coacervation, metal-to-ligand coordination chemistry, supramolecular host-guest interactions, and a reversible covalent disulfide metal-complex crosslinker. The cleavage of this crosslinker by dithiotreithol results in the breaking of oligomeric europium(III) structures in the core and results in the disassembly of the 70 nm size micelles. Over hours, due to the oxidation of thiolates to disulfides, monomeric units can re-crosslink into oligomeric core-units, favoring micellar reassembly. The time required to reassemble can be controlled by varying the reducing agent concentration or the ratio between redox-responsive and non-redox-responsive crosslinkers. Controlled Methyl Red encapsulation and release indicate the potential of these micelles, for, e. g., controlled drug uptake and delivery.

By fabricating microfluidic devices via (covalent) plasma microcontact patterning (PµCP) and replica molding, we were able create β-CD patterns inside a microfluidic channel. Chemical reactivity and reusability of the devices were validated through host-guest interactions with diadamantane functionalized Cyanine 5 dye (Cy5-Ad2).

Species-specific isolation of microsized entities such as microplastics and resistant bacteria from waste streams is becoming a growing environmental challenge. By studying the on-flow immobilization of micron-sized polystyrene particles onto functionalized silica surfaces, we ascertain if supramolecular host guest chemistry in aqueous solutions can provide an alternative technology for water purification. Polystyrene particles were modified with different degrees of adamantane (guest) molecules, and silica surfaces were patterned with beta-cyclodextrin (beta-CD, host) through microcontact printing (mu CP). The latter was exposed to solutions of these particles flowing at different speeds, allowing us to study the effect of flow rate and multivalency on particle binding to the surface. The obtained binding profile was correlated with Comsol simulations. We also observed that particle binding is directly aligned with particle's ability to form host guest interactions with the beta-CD-patterned surface, as particle binding to the functionalized glass surface increased with higher adamantane load on the polystyrene particle surface. Because of the noncovalent character of these interactions, immobilization is reversible and modified beta-CD surfaces can be recycled, which provides a positive outlook for their incorporation in water purification systems.

[Image: see text] There is a need to develop diagnostic and analytical tools that allow noninvasive monitoring of bacterial growth and dissemination in vivo. For such cell-tracking studies to hold translational value to controlled human infections, in which volunteers are experimentally colonized, they should not require genetic modification, and they should allow tracking over a number of replication cycles. To gauge if an antimicrobial peptide tracer, (99m)Tc-UBI(29–41)-Cy5, which contains both a fluorescent and a radioactive moiety, could be used for such in vivo bacterial tracking, we performed longitudinal imaging of a thigh-muscle infection with (99m)Tc-UBI(29–41)-Cy5-labeled Staphylococcus aureus. Mice were imaged using SPECT and fluorescence-imaging modalities at various intervals during a 28 h period. Biodistribution analyses were performed to quantitate radioactivity in the abscess and other tissues. SPECT and fluorescence imaging in mice showed clear retention of the (99m)Tc-UBI(29–41)-Cy5-labeled bacteria following inoculation in the thigh muscle. Despite bacterial replication, the signal intensity in the abscess only modestly decreased within a 28 h period: 52% of the total injected radioactivity per gram of tissue (%ID/g) at 4 h postinfection (pi) versus 44%ID/g at 28 h pi (15% decrease). After inoculation, a portion of the bacteria disseminated from the abscess, and S. aureus cultures were obtained from radioactive urine samples. Bacterial staining with (99m)Tc-UBI(29–41)-Cy5 allowed noninvasive bacterial-cell tracking during a 28 h period. Given the versatility of the presented bacterial-tracking method, we believe that this concept could pave the way for precise imaging capabilities during controlled-human-infection studies.

Luminescent Ag nanoclusters (Ag-NC) provide the next generation in bionanoparticles, wherein the luminescence (650 nm) and large Stokes shift of these inorganic nanoclusters are favorable for biological imaging. By combining these characteristics with those of human serum albumin (HSA; a protein capable of binding multiple endo- and exogenous compounds), the Ag nanoclusters can be shielded from the environment and functionalized with (receptor) targeting moieties. Encapsulation of the 1.5 nm Ag nanoclusters by HSA resulted in a threefold increase in luminescence intensity and a twofold increase of the luminescence lifetime (1.7 vs. 3.6 µs). To exemplify the potential of this targeted concept, we functionalized HSA-Ag nanoparticles with chemokine receptor 4 (CXCR4) targeting peptides [Ac-TZ14011(CO2H)]. The resulting Ac-TZ14011-HSA-Ag nanoparticles demonstrated specific binding to CXCR4-overexpressing tumor cells. Upon exposure to (ambient) light, particle-functionalized tumor cells were killed. Combined, these experiments illustrate that HSA-Ag nanoparticles may have a potential in biological imaging and possibly even in targeted theranostic applications.

A generic site-specific conjugation method that generates a homogeneous product is of utmost importance in tracer development for molecular imaging and therapy. We explored the protein-ligation capacity of the enzyme Sortase A to label camelid single-domain antibody-fragments, also known as nanobodies. The versatility of the approach was demonstrated by conjugating independently three different imaging probes: the chelating agents CHX-A"-DTPA and NOTA for single-photon emission computed tomography (SPECT) with indium-111 and positron emission tomography (PET) with gallium-68, respectively, and the fluorescent dye Cy5 for fluorescence reflectance imaging (FRI). After a straightforward purification process, homogeneous single-conjugated tracer populations were obtained in high yield (30-50%). The enzymatic conjugation did not affect the affinity of the tracers, nor the radiolabeling efficiency or spectral characteristics. In vivo, the tracers enabled the visualization of human epidermal growth factor receptor 2 (HER2) expressing BT474M1-tumors with high contrast and specificity as soon as 1 h post injection in all three imaging modalities. These data demonstrate Sortase A-mediated conjugation as a valuable strategy for the development of site-specifically labeled camelid single-domain antibody-fragments for use in multiple molecular imaging modalities. Introduction: The skin stage of malaria is a vital and vulnerable migratory life stage of the parasite. It has been characterised in rodent models, but remains wholly uninvestigated for human malaria parasites. To enable in depth analysis of not genetically modified (non-GMO) Plasmodium falciparum (Pf) sporozoite behaviour in human skin, we devised a labelling technology (Cy5M2, targeting the sporozoite mitochondrion) that supports tracking of individual non-GMO sporozoites in human skin. Methods: Sporozoite labelling with Cy5M2 was performed in vitro as well as via the feed of infected Anopheles mosquitos. Labelling was validated using confocal microscopy and flow cytometry and the fitness of labelled sporozoites was determined by analysis of infectivity to human hepatocytes in vitro, and in vivo in a rodent infection model. Using confocal video microscopy and custom software, single-sporozoite tracking studies in human skin-explants were performed. Results: Both in vitro and in mosquito labelling strategies yielded brightly fluorescent sporozoites of three different Plasmodium species. Cy5M2 uptake colocalized with MitoTracker® green and could be blocked using the known Translocator protein (TSPO)-inhibitor PK11195. This method supported the visualization and subsequent quantitative analysis of the migration patterns of individual non-GMO Pf sporozoites in human skin and did not affect the fitness of sporozoites. Conclusions: The ability to label and image non-GMO Plasmodium sporozoites provides the basis for detailed studies on the human skin stage of malaria with potential for in vivo translation. As such, it is an important tool for development of vaccines based on attenuated sporozoites and their route of administration.

Background: Today, both radioactive SPECT and PET imaging radiopharmaceuticals are used for clinical diagnosis of bacterial infections. Due to the possible applications in image-guided surgery, fluorescent imaging of infections has gained interest. We here present the highlights and recent developments in the use of fluorescence imaging for bacterial infections. In this overview, we also include the latest developments in multimodal bacterial imaging strategies that combine radioactive and fluorescent imaging. Based on this literature, we present our future perspectives for the field including the translational potential. Methods: In the current review, we complement earlier reports with the most recent fluorescent and multimodal radiopharmaceuticals for bacterial infection imaging. Where possible, in this review, the chemical structure of the compounds and clinical images were shown. Results: A total of 35 out of 77 original articles on pre-clinical and clinical imaging of bacterial infections with fluorescent tracers and multimodality radiopharmaceuticals were included for reviewing. Conclusion: In our view, the highest translational potential lies with compounds that are based on targeting vectors that are specific for bacteria: e.g., fluorescently labelled UBI 29–41 , polymyxin B, vancomycin, ZnDPA and a M. tuberculosis-specific β-lactamase-cleavable linker CNIP800. Multimodal concepts using dually labelled UBI 29–41 , vancomycin, and ZnDPA help connect optical imaging to the more traditional use of radiopharmaceuticals in infectious diseases. Multimodal bacterial imaging is a promising strategy not only to diagnose bacterial infections but also to evaluate the effectivity of surgical treatment for infections.

Micropatterns of beta-cyclodextrin (beta-CD) monolayers on glass are obtained by using a plasma etching approach with polydimethylsiloxane (PDMS) stamps. This simple and versatile approach provides a promising alternative to current techniques for creating patterns of covalently bound molecules. It is also possible to fabricate sub-10 mu m sized features.

Background: Nanotechnology, even if unknowingly, has been used for millennia. The occurrence of shiny colors in pottery and glass made hundreds and thousand of years ago is due to the presence of nanoparticles in the fabrication of such ornaments. In the last decade, 3D printing has revolutionized fabrication and manufacturing processes, making it easier to produce, in a simple and fast way, 3D objects.Results: In this paper we show how to fabricate a 3D-printable nanocomposite composed of dichroic gold nanoparticles and a 3D-printable polymer. The minute amount of gold nanoparticles used for obtaining the dichroic effect does not influence the mechanical properties of the polymer nor its printability. Thus, the nanocomposite can be easily 3D-printed using a standard 3D printer and shows a purple color in transmission and a brownish color in reflection.Conclusion: This methodology can be used not only by artists, but also for studying the optical properties of nanoparticles or, for example, for the 3D fabrication of optical filters.

Micelles have been recognized as versatile platforms for different biomedical applications, from bioimaging to drug delivery. Complex coacervate core micelles present great advantages compared to traditional micelles, however controlling the number of charges per core-unit and the stability is still a challenge. We here present cyclodextrin-based complex coacervate core micelles where the charge per core-unit can be straightforwardly tuned by cyclodextrin host–guest interactions. By varying the ratio between two adamantane guest molecules, 1-adamantanecarboxylic acid and 1,3-adamantanediacetic acid, the charge of the monomeric core-units can be finely tuned from 6− to 9−. By adding an adamantane bislinker, monomeric core-units can be combined together in dimeric and polymeric structures, increasing the micelles’ stability. The orthogonal supramolecular host–guest and coordination-chemistry allows for well-controlled cyclodextrin-based complex coacervate core micelles that offer a versatile platform for designing future, e.g., responsive systems.

The incorporation of dichroic nanoparticles in 3D printable plastic results in a dichroic nanocomposite material that can be easily 3D printed. Gold nanoparticles that shows dichroic properties have been embedded in polyvinylalcohol, a standard 3D printable plastic. The nanocomposite shows dichroic properties and standard printability properties.

Radioguided surgery (RGS) utilizes radiolabeled tracers that accumulate in the lesion of interest following local or systemic administration. RGS is an expanding surgical guidance technology, but unfortunately most currently available radiotracers are unable to provide intraoperative optical information regarding the location of the lesion. In 2011, a new hybrid tracer (both radioactive and fluorescent) was introduced for sentinel lymph node detection. This hybrid tracer (indocyanine green-99mTc-nanocolloid) has helped to generate a new hybrid surgical guidance concept, thereby expanding the traditional RGS-based approach. Based on its radioactive signature a hybrid tracer is able to preserve the “traditional” role for RGS. Furthermore, as an additional tool in the operating room, a hybrid tracer provides optical information via fluorescence guidance. This review discusses the most common RGS approaches. Different clinically used radioactive tracers and their potential hybrid derivatives are also discussed. Furthermore, the various imaging devices designed for radioactivity-based detection are reviewed in the context of generating hybrid-imaging modalities.

Introduction Fluorescence guidance is an upcoming methodology to improve surgical accuracy. Challenging herein is the identification of the minimum dose at which the tracer can be detected with a clinical-grade fluorescence camera. Using a hybrid tracer such as indocyanine green (ICG)-99mTc-nanocolloid, it has become possible to determine the accumulation of tracer and correlate this to intraoperative fluorescence-based identification rates. In the current study, we determined the lower detection limit of tracer at which intraoperative fluorescence guidance was still feasible. Methods Size exclusion chromatography (SEC) provided a laboratory set-up to analyze the chemical content and to simulate the migratory behavior of ICG-nanocolloid in tissue. Tracer accumulation and intraoperative fluorescence detection findings were derived from a retrospective analysis of 20 head-and-neck melanoma patients, 40 penile and 20 prostate cancer patients scheduled for sentinel node (SN) biopsy using ICG-99mTc-nanocolloid. In these patients, following tracer injection, single photon emission computed tomography fused with computed tomography (SPECT/CT) was used to identify the SN(s). The percentage injected dose (% ID), the amount of ICG (in nmol), and the concentration of ICG in the SNs (in μM) was assessed for SNs detected on SPECT/CT and correlated with the intraoperative fluorescence imaging findings. Results SEC determined that in the hybrid tracer formulation, 41 % (standard deviation: 12 %) of ICG was present in nanocolloid-bound form. In the SNs detected using fluorescence guidance a median of 0.88 % ID was present, compared to a median of 0.25 % ID in the non-fluorescent SNs (p-value

Quantitative assessment of affinity and kinetics is a critical component in the development of (receptor-targeted) radiotracers. For fluorescent tracers, such an assessment is currently not yet applied, while (small) changes in chemical composition of the fluorescent component might have substantial influence on the overall properties of a fluorescent tracer. Hybrid imaging labels that contain both a radiolabel and a fluorescent dye can be used to evaluate both the affinity (fluorescent label) and the in vivo distribution (radiolabel) of a targeted tracer. We present a hybrid label oriented and matrix-based scoring approach that enabled quantitative assessment of the influence of (overall) charge and lipophilicity of the fluorescent label on the (in vivo) characteristics of αvβ3-integrin targeted tracers. Systematic chemical alterations in the fluorescent dye were shown to result in a clear difference in the in vivo distribution of the different hybrid tracers. The applied evaluation technique resulted in an optimized targeted tracer for αvβ3-integrin, which combined the highest T/M ratio with the lowest uptake in other organs. Obviously this selection concept would also be applicable during the development of other (receptor-targeted) imaging tracers.

Leaving a lasting impression: Molecularly imprinted polymers (MIPs) featuring neutral binding sites for phosphotyrosine (P) is also shown to have affinity for sulfotyrosine (S) and a sulfotyrosine-containing peptide. The MIPs were capable of selectively capturing both a phosphorylated and a sulfated peptide in a mixture and could release the peptides separately based on elution conditions.

Radioguided surgery (RGS) allows a surgeon to intraoperatively identify the lesions of interest. This technique relies on the accumulation of a radiotracer in the lesion(s) of interest. Such accumulation can occur via the local administration of the radiotracer, followed by local staining or passive drainage via the lymphatic system, or can occur via the systemic administration followed by retention or targeted accumulation of the radiotracer. The range of radiotracers applied in RGS varies from the radioactive isotope itself, to small molecules, peptides, antibodies, and colloids. The choice of the radionuclide depends on various factors, such as half-life, desired radiation type and energy, and (chemical) means to attach it to an active entity. An often overlooked factor is the radiation burden for the patient and the medical personnel. The introduction of optical imaging technologies, such as fluorescence and Cherenkov imaging, expands the utility of RGS.

1289 Objectives The use of dual SPECT/PET functional imaging allows researchers a unique means for simultaneously surveying multiple functional elements in complex disease models. The use of magnetic resonance imaging allows the researcher to evaluate the microstructure and anatomy of tissue or an organ to offer a more versatile perspective. The ability to combine these highly sophisticated modalities along with integrated optical imaging modalities will allow researchers a vast array of data while maintaining a non-invasive approach. The advent of preclinical integrated MRI/PET and MRI/SPECT systems may facilitate the design and practical execution of dual SPECT/PET imaging protocols. Here, we utilize a technique for cross-platform preclinical imaging of PET or SPECT with MRI. Methods Foxn1nu mice (Jackson Laboratories; Bar Harbor, ME) were imaged 2-3 weeks after grafting subcutaneous HCT 116-hNIS-NEO tumor cells. Each formed tumor was approximately 1-3mm diameter in size at the time of imaging. PET and SPECT acquisitions of ten and twenty minutes were acquired using the Albira PET/SECT/CT imaging system. Mice were dosed with approximately 200µCi of 18F-FDG and 1mCi 99mTc. Imaging was done one hour after injection of the isotope. MR imaging was performed using the Bruker ICON 1T MR system using a 3D MR T2-weighted RARE sequence. Animals were transferred from one instrument to the other using the Bruker Multimodal Animal Bed (MMAB). This bed allows for the animal to remain under anesthesia without any movement and is compatible with both instruments. Precise co-registration between modalities was ensured due to the fiducial markers lid. This lid is designed to provide multiple reference points for fiducial matching. A thin channel runs along the top of the lid without interfering with the space of the animal and can be filled with contrast agent that is detectable in PET, SPECT, and MRI imaging modalities. All image registration was performed using the PMOD Technologies Fuse It module. Manual co-registration is simplified using the FML lid and can verify that the data is in alignment. Results Initial studies that were conducted without the use of the fiducial marker lid did not ensure complete accuracy of registration and were found to be quite subjective. The ability to transfer the animal from one instrument to the next instrument was largely critical to the success of these studies as well. The integration between the platform of the Albira and ICON was simple and allowed for complete immobilization of the animal preventing further complications to the study and the preparation process. Using mice that have a distinctive tumor present allowed us to easily identify our volume of interest in each of the modalities. PET and SPECT imaging has its limitations because the accompanied CT acquisition does not always provide sufficient tumor contrast. MRI however can provide this desired tumor margin contrast and allows for reliable PET or SPECT registration. In length this imaging protocol provides a more accurate method in production of VOIs based on tumor margins identified in MR images and application of functional PET or SPECT images. Conclusions Using MRI/PET or MRI/SPECT both functional and structural imaging modalities can be combined in order to enhance analytical value. Animal stability and insurance of accurate co-registration is necessary to fully leverage the potential of cross-platform modalities. Additionally, he FML fill line in the lid of the bed could also allow the researcher to add any number of optical applications in to the mix. This method serves to be one that is simple and reliable, relative to alternative solutions that use makeshift beds and/or capillary tubes, to be used in imaging studies. The streamlined transition from one modality to the next in separate instruments provides less stress for the animal and the researcher as well.

In trauma and orthopedic surgery, infection of implants has a major impact on the outcome for patients. Infections may develop either during the initial implantation or during the lifetime of an implant. Both infections, as well as aseptic loosening of the implant, are reasons for revision of the implants. Therefore, discrimination between aseptic-mechanical-loosening and septic-bacterial-loosening of implants is critical during selection of a patient-tailored treatment policy. Specific detection and visualization of infections is a challenge because it is difficult to discriminate infections from inflammation. An imaging tracer that facilitates bacterial identification in a pre- and intraoperative setting may aid the workup for patients suspicious of bacterial infections. In this study we evaluated an antimicrobial peptide conjugated to a hybrid label, which contains both a radioisotope and a fluorescent dye. After synthesis of DTPA-Cy5-UBI29-41 and-when necessary-radiolabeling with (111)In (yield 96.3 ± 2.7%), in vitro binding to various bacterial strains was evaluated using radioactivity counting and confocal fluorescence microscopy. Intramuscular bacterial infections (S. aureus or K. pneumoniae) were also visualized in vivo using a combined nuclear and fluorescence imaging system. The indium-111 was chosen as label as it has a well-defined coordination chemistry, and in pilot studies labeling DTPA-Cy5-UBI29-41 with technetium-99m, we encountered damage to the Cy5 dye after the reduction with SnCl2. As a reference, we used the validated tracer (99m)Tc-UBI29-41. Fast renal excretion of (111)In-DTPA-Cy5-UBI29-41 was observed. Target to nontarget (T/NT) ratios were highest at 2 h post injection: radioactivity counting yielded T/NT ratios of 2.82 ± 0.32 for S. aureus and 2.37 ± 0.05 for K. pneumoniae. Comparable T/NT ratios with fluorescence imaging of 2.38 ± 0.09 for S. aureus and 3.55 ± 0.31 for K. pneumoniae were calculated. Ex vivo confocal microscopy of excised infected tissues showed specific binding of the tracer to bacteria. Using a combination of nuclear and fluorescence imaging techniques, the hybrid antimicrobial peptide conjugate DTPA-Cy5-UBI29-41 was shown to specifically accumulate in bacterial infections. This hybrid tracer may facilitate integration of noninvasive identification of infections and their extent as well as real-time fluorescence guidance during surgical resection of infected areas.

Treatment of neurodegenerative disorders such as Alzheimer's disease is hampered by the blood-brain barrier (BBB). This tight cerebral vascular endothelium regulates selective diffusion and active transport of endogenous molecules and xenobiotics into and out of the brain parenchyma. In this study, glutathione targeted PEGylated (GSH-PEG) liposomes were designed to deliver amyloid-targeting antibody fragments across the BBB into the brain. Two different formulations of GSH-PEG liposomes based on 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) and egg-yolk phosphatidylcholine (EYPC) were produced. Both formulations encapsulate 15kDa amyloid beta binding llama single domain antibody fragments (VHH-pa2H). To follow the biodistribution of VHH-pa2H rather than the liposome, the antibody fragment was labeled with the radioisotope indium-111. To prolong the shelf life of the construct beyond the limit of radioactive decay, an active-loading method was developed to efficiently radiolabel the antibody fragments after encapsulation into the liposomes, with radiolabeling efficiencies of up to 68% after purification. The radiolabeled liposomes were administered via a single intravenous bolus injection to APPswe/PS1dE9 double transgenic mice, a mouse model of Alzheimer's disease, and their wildtype littermates. Both GSH-PEG DMPC and GSH-PEG EYPC liposomes significantly increased the standard uptake values (SUV) of VHH-pa2H in the blood of the animals compared to free VHH-pa2H. Encapsulation in GSH-PEG EYPC liposomes resulted in the highest increase in SUV in the brains of transgenic animals. Overall, these data provide evidence that GSH-PEG liposomes may be suitable for specific delivery of single domain antibody fragments over the BBB into the brain.

Photodynamic treatment (PDT) is an emerging therapeutic procedure for the management of cancer, based on the use of photosensitizers, compounds that generate highly reactive oxygen species (ROS) on irradiation with visible light. The ROS generated may oxidize a variety of biomolecules within the cell, loaded with a photosensitizer. The high reactivity of these ROS restricts their radius of action to 5-20 nm from the site of their generation. We studied oxidation of intracellular proteins during PDT using the ROS-sensitive probe acetyl-tyramine-fluorescein (acetylTyr-Fluo). This probe labels cellular proteins, which become oxidized at tyrosine residues under the conditions of oxidative stress in a reaction similar to dityrosine formation. The fluorescein-labeled proteins can be visualized after gel electrophoresis and subsequent Western blotting using the antibody against fluorescein. We found that PDT of rat or human fibroblasts, loaded with the photosensitizer Hypocrellin A, resulted in labeling of a set of intracellular proteins that was different from that observed on treatment of the cells with H2O2. This difference in labeling patterns was confirmed by 2D electrophoresis, showing that a limited, yet distinctly different, set of proteins is oxidized under either condition of oxidative stress. By matching the Western blot with the silver-stained protein map, we infer that alpha-tubulin and beta-tubulin are targets of PDT-induced protein oxidation. H2O2 treatment resulted in labeling of endoplasmic reticulum proteins. Under conditions in which the extent of protein oxidation was comparable, PDT caused massive apoptosis, whereas H2O2 treatment had no effect on cell survival. This suggests that the oxidative stress generated by PDT with Hypocrellin A activates apoptotic pathways, which are insensitive to H2O2 treatment. We hypothesize that the pattern of protein oxidation observed with Hypocrellin A reflects the intracellular localization of the photosensitizer. The application of acetylTyr-Fluo may be useful for characterizing protein targets of oxidation by PDT with various photosensitizers.

To investigate if the degree of functionalization of ferritin could be controlled using a supramolecular self-assembly process, two photophysical separable batches of ferritin were created by functionalizing ferritin capsids with either Cy3- or Cy5-dye (loading rate of about 50 %). After dis-assembly, Cy3-, Cy5- as well as non-functionalized ferritin subunits were mixed in variable ratios. Photophysical measurements revealed that the ratio in which the subunits were mixed was indeed indicative for the ratios in which the functionalized subunits were observed in the re-assembled capsids. During re-assembly, however, a slight preference for the inclusion of non-functionalized subunits was observed, indicating the reactivity decreased following functionalization. The iron biomineralization properties of ferritin were retained by the multi-functionalized capsids as Fe II diffused rapidly inside making it visible by transmission electron microscopy (TEM). These combined data indicate that it is possible to functionalize ferritin in an orthogonal manner using the supramolecular interaction between ferritin subunits.

Background In vivo bioluminescence, fluorescence, and single-photon emission computed tomography (SPECT) imaging provide complementary information about biological processes. However, to date these signatures are evaluated separately on individual preclinical systems. In this paper, we introduce a fully integrated bioluminescence-fluorescence-SPECT platform. Next to an optimization in logistics and image fusion, this integration can help improve understanding of the optical imaging (OI) results. Methods An OI module was developed for a preclinical SPECT system (U-SPECT, MILabs, Utrecht, the Netherlands). The applicability of the module for bioluminescence and fluorescence imaging was evaluated in both a phantom and in an in vivo setting using mice implanted with a 4 T1-luc + tumor. A combination of a fluorescent dye and radioactive moiety was used to directly relate the optical images of the module to the SPECT findings. Bioluminescence imaging (BLI) was compared to the localization of the fluorescence signal in the tumors. Results Both the phantom and in vivo mouse studies showed that superficial fluorescence signals could be imaged accurately. The SPECT and bioluminescence images could be used to place the fluorescence findings in perspective, e.g. by showing tracer accumulation in non-target organs such as the liver and kidneys (SPECT) and giving a semi-quantitative read-out for tumor spread (bioluminescence). Conclusions We developed a fully integrated multimodal platform that provides complementary registered imaging of bioluminescent, fluorescent, and SPECT signatures in a single scanning session with a single dose of anesthesia. In our view, integration of these modalities helps to improve data interpretation of optical findings in relation to radionuclide images.

Replacement of the sulfate groups, present in vivo on the N-terminus of the C5a-receptor (C5aR), by phosphate groups is explored. Phosphorylated mimics of the C5a-receptor N-terminus are synthesized and their binding to Chemotaxis Inhibitory Protein of Staphylococcus aureus (CHIPS) is studied by ITC and NMR. The phosphorylated C5aR mimics showed comparable binding affinity and a similar binding mode towards CHIPS compared to their sulfated forms. The activities of the phosphorylated peptides in a biological assay, however, were significantly lower compared to their sulfated counterparts.

Bacterial infections have always been, and still are, a major global healthcare problem. For accurate treatment it is of upmost importance that the location(s), severity, type of bacteria, and therapeutic response can be accurately staged. Similar to the recent successes in oncology, tracers specific for molecular imaging of the disease may help advance patient management. Chemical design and bacterial targeting mechanisms are the basis for the specificity of such tracers. The aim of this review is to provide a comprehensive overview of the molecular imaging tracers developed for optical and nuclear identification of bacteria and bacterial infections. Hereby we envision that such tracers can be used to diagnose infections and aid their clinical management. From these compounds we have set out to identify promising targeting mechanisms and select the most promising candidates for further development.

UnlabelledScreening of biomarker expression levels in tumor biopsy samples not only provides an assessment of prognostic and predictive factors, but may also be used for selection of biomarker-specific imaging strategies. To assess the feasibility of using a biopsy specimen for a personalized selection of an imaging agent, the chemokine receptor 4 (CXCR4) was used as a reference biomarker.MethodsA hybrid CXCR4 targeting peptide (MSAP-Ac-TZ14011) containing a fluorescent dye and a chelate for radioactive labeling was used to directly compare initial flow cytometry-based target validation in fresh tumor tissue to in vivo single photon emission computed tomography (SPECT) imaging and in vivo and ex vivo fluorescence imaging.ResultsFlow cytometric analysis of mouse tumor derived cell suspensions enabled discrimination between 4T1 control tumor lesions (with low levels of CXCR4 expression) and CXCR4 positive early, intermediate and late stage MIN-O lesions based on their CXCR4 expression levels; CXCR4(basal), CXCR4(+) and CXCR4(++) cell populations could be accurately discriminated. Mean fluorescent intensity ratios between expression in MIN-O and 4T1 tissue found with flow cytometry were comparable to ratios obtained with in vivo SPECT/CT and fluorescence imaging, ex vivo fluorescence evaluation and standard immunohistochemistry.ConclusionThe hybrid nature of a targeting imaging agent like MSAP-Ac-TZ14011 enables integration of target selection, in vivo imaging and ex vivo validation using a single agent. The use of biopsy tissue for biomarker screening can readily be expanded to other targeting hybrid imaging agents and can possibly help increase the clinical applicability of tumor-specific imaging approaches.

Hybrid tracers that are both radioactive and fluorescent help extend the use of fluorescence-guided surgery to deeper structures. Such hybrid tracers facilitate preoperative surgical planning using (3D) scintigraphic images and enable synchronous intraoperative radio- and fluorescence guidance. Nevertheless, we previously found that improved orientation during laparoscopic surgery remains desirable. Here we illustrate how intraoperative navigation based on optical tracking of a fluorescence endoscope may help further improve the accuracy of hybrid surgical guidance. After feeding SPECT/CT images with an optical fiducial as a reference target to the navigation system, optical tracking could be used to position the tip of the fluorescence endoscope relative to the preoperative 3D imaging data. This hybrid navigation approach allowed us to accurately identify marker seeds in a phantom setup. The multispectral nature of the fluorescence endoscope enabled stepwise visualization of the two clinically approved fluorescent dyes, fluorescein and indocyanine green. In addition, the approach was used to navigate toward the prostate in a patient undergoing robot-assisted prostatectomy. Navigation of the tracked fluorescence endoscope toward the target identified on SPECT/CT resulted in real-time gradual visualization of the fluorescent signal in the prostate, thus providing an intraoperative confirmation of the navigation accuracy.

Human serum albumin (HSA) is a biological nanocarrier that forms non-covalent complexes with a number of synthetic and biomolecules. Previously we demonstrated radiolabeled HSA-based nanoparticles can form non-covalent complexes with fluorescent cyanine dyes yielding imaging agents for surgical guidance towards tumor draining lymph nodes. Here the self-assembly approach enabled rapid clinical translation. Based on this experience we reasoned it would be interesting to expand this non-covalent technology to a targeted approach. Therefore, the ability of HSA to form non-covalent self-assembled complexes with peptides via near-infrared (NIR) cyanine dyes was explored. Foster resonance energy transfer (FRET) quenching interactions between HSA-Cy5 and the non-covalently bound fluorescent molecules indocyanine green (ICG), IR783-CO(2)H and three IR783-labeled targeting peptides were used to monitor complex assembly and disassembly. The host-guest interactions between HSA and IR783-labeled peptides enabled the formation of (bio)nanoparticles that are coated with peptides, which may target α(v)β(3)-integrins, the chemokine receptor 4 (CXCR4), or somatostatin receptors. The potential of CXCR4-targeted (bio)nanoparticles in sentinel lymph node procedures is demonstrated in vivo. By non-covalently binding NIR-dye labeled peptides to an already clinically approved HSA-scaffold, we have readily formed targeted bionanoparticles.

Complement factor C5a is one of the most powerful pro-inflammatory agents involved in recruitment of leukocytes, activation of phagocytes and other inflammatory responses. C5a triggers inflammatory responses by binding to its G-protein-coupled C5a-receptor (C5aR). Excessive or erroneous activation of the C5aR has been implicated in numerous inflammatory diseases. The C5aR is therefore a key target in the development of specific anti-inflammatory compounds. A very potent natural inhibitor of the C5aR is the 121-residue chemotaxis inhibitory protein of Staphylococcus aureus (CHIPS). Although CHIPS effectively blocks C5aR activation by binding tightly to its extra-cellular N terminus, it is not suitable as a potential anti-inflammatory drug due to its immunogenic properties. As a first step in the development of an improved CHIPS mimic, we designed and synthesized a substantially shorter 50-residue adapted peptide, designated CHOPS. This peptide included all residues important for receptor binding as based on the recent structure of CHIPS in complex with the C5aR N terminus. Using isothermal titration calorimetry we demonstrate that CHOPS has micromolar affinity for a model peptide comprising residues 7–28 of the C5aR N terminus including two O-sulfated tyrosine residues at positions 11 and 14. CD and NMR spectroscopy showed that CHOPS is unstructured free in solution. Upon addition of the doubly sulfated model peptide, however, the NMR and CD spectra reveal the formation of structural elements in CHOPS reminiscent of native CHIPS. In this communication, a new site specific synthesis of highly functionalized and multiple sulfated peptides using convential Fmoc-tBu solid phase peptide synthesis is described.

Complement component C5a is a potent pro-inflammatory agent inducing chemotaxis of leukocytes toward sites of infection and injury. C5a mediates its effects via its G protein-coupled C5a receptor (C5aR). Although under normal conditions highly beneficial, excessive levels of C5a can be deleterious to the host and have been related to numerous inflammatory diseases. A natural inhibitor of the C5aR is chemotaxis inhibitory protein of Staphylococcus aureus (CHIPS). CHIPS is a 121-residue protein excreted by S. aureus. It binds the N terminus of the C5aR (residues 1-35) with nanomolar affinity and thereby potently inhibits C5a-mediated responses in human leukocytes. Therefore, CHIPS provides a starting point for the development of new anti-inflammatory agents. Two O-sulfated tyrosine residues located at positions 11 and 14 within the C5aR N terminus play a critical role in recognition of C5a, but their role in CHIPS binding has not been established so far. By isothermal titration calorimetry, using synthetic Tyr-11- and Tyr-14-sulfated and non-sulfated C5aR N-terminal peptides, we demonstrate that the sulfate groups are essential for tight binding between the C5aR and CHIPS. In addition, the NMR structure of the complex of CHIPS and a sulfated C5aR N-terminal peptide reveals the precise binding motif as well as the distinct roles of sulfated tyrosine residues sY11 and sY14. These results provide a molecular framework for the design of novel CHIPS-based C5aR inhibitors.

The synthesis and biological evaluation of a new class of selective irreversible cysteine protease inhibitors is described. A set of amino acid based chloromethyl sulfoxides was prepared and they were found to inhibit irreversibly the cysteine protease papain. They were selective for cysteine proteases since no inhibition was found for the serine protease chymotrypsin.
