International audience; We report herein the synthesis of zwitterionic sulfobetaine (SB) and dimethylamine oxide (AO) detergents whose alkyl chain is made of either a perfluorohexyl (F6H3) or a perfluoropentyl (F5H5) group linked to a hydrogenated spacer arm. In aqueous solution, the critical micellar concentrations (CMCs) measured by surface tensiometry (SFT) and isothermal titration calorimetry (ITC) were found in the millimolar range (1.3-2.4 mM). The morphologies of the aggregates were evaluated by dynamic light scattering (DLS), analytical ultracentrifugation (AUC), small-angle X-ray scattering (SAXS), and transmission electron microscopy (TEM), demonstrating that the two perfluoropentyl derivatives formed small micelles less than 10 nm in diameter, whereas the perfluorohexyl derivatives formed larger and more heterogeneous micelles. The two SB detergents were able to solubilize synthetic lipid vesicles in a few hours; by contrast, the perfluoropentyl AO induced much faster solubilization, whereas the perfluorohexyl AO did not show any solubilization. All detergents were tested for their abilities to stabilize three membrane proteins, namely, bacteriorhodopsin (bR), the Bacillus subtilis ABC transporter BmrA, and the Streptococcus pneumoniae enzyme SpNOX. The SB detergents outperformed the AO derivatives as well as their hydrogenated analogs in stabilizing these proteins. Among the four new compounds, F5H5SB combines many desirable properties for membrane-protein study, as it is a powerful yet gentle detergent.

Myelin basic protein (MBP) is an intrinsically disordered protein and in the central nervous system (CNS) mainly responsible for connecting the cytoplasmic surfaces of the multilamellar, compact myelin. Increased posttranslational modification of MBP is linked to both, the natural development (from adolescent to adult brains) of myelin, and features of multiple sclerosis. Here, we study how a combination of this intrinsically disordered myelin protein with varying the natural cholesterol content may alter the characteristics of myelin-like membranes and interactions between these membranes. Large unilamellar vesicles (LUVs) with a composition mimicking the cytoplasmic leaflet of myelin were chosen as the model system, in which different parameters contributing to the interactions between the lipid membrane and MBP were investigated. While we use cryo-transmission electron microscopy (TEM) for imaging, dynamic light scattering (DLS) and electrophoretic measurements through continuously-monitored phase-analysis light scattering (cmPALS) were used for a more global overview of particle size and charge, and electron paramagnetic resonance (EPR) spectroscopy was utilized for local behavior of lipids in the vesicles’ membranes in aqueous solution. The cholesterol content was varied from 0¬¬─60% in these LUVs and measurements were performed in the presence and absence of MBP. We find that the composition of the lipid layers is relevant to the interaction with MBP. Not only the size, the shape and the aggregation behavior of the vesicles depend on the cholesterol content, but also within each membrane, cholesterol’s freedom of movement, its environmental polarity and its distribution were found to depend on the content using the EPR-active spin-labeled cholesterol (CSOSL). In addition, DLS and EPR measurements probing the transition temperatures of the lipid phases allow a correlation of specific behavior with the human body temperature of 37 °C. Overall, our results aid in understanding the importance of the native cholesterol content in the healthy myelin membrane, which serves as the basis for stable and optimum protein-bilayer interactions. Although studied in this specific myelin-like system, from a more general and materials science-oriented point of view, we could establish how membrane and vesicle properties depend on cholesterol and/or MBP content, which might be useful generally when specific membrane and vesicle characteristics are sought for.

Although incorporation of photo‐activatable lipids into membranes potentially opens up novel avenues for investigating interactions with proteins, the question of whether diazirine‐modified lipids are suitable for such studies, remains under debate. Focusing on the potential for studying lipid/peptide interactions by cross‐linking mass spectrometry (XL‐MS), we developed a diazirine‐modified lipid (DiazPC), and examined its behaviour in membranes incorporating the model α‐helical peptide LAVA20. We observed an unexpected backfolding of the diazirine‐containing stearoyl chain of the lipid. This surprising behaviour challenges the potential application of DiazPC for future XL‐MS studies of peptide and protein/lipid interactions. The observations made for DiazPC most likely represent a general phenomenon for any type of membrane lipids with a polar moiety incorporated into the alkyl chain. Our finding is therefore of importance for future protein/lipid interaction studies relying on modified lipid probes., Can diazirine‐modified lipids be used to locate the position of UV‐induced cross‐links between this lipid and a membrane peptide by using MS? Although DiazPC fulfils all prerequisites for a photo‐activatable membrane lipid to study protein/lipid interactions and the cross‐links can be exactly assigned, we observed a backfolding of the lipid's alkyl chain. This is a general phenomenon for any type of membrane lipids with a hydrophilic group in the alkyl chain.

New technologies for purifying membrane-bound protein complexes in combination with cryo-electron microscopy (EM) have recently allowed the exploration of such complexes under near-native conditions. In particular, polymer-encapsulated nanodiscs enable the study of membrane proteins at high resolution while retaining protein-protein and protein-lipid interactions within a lipid bilayer. However, this powerful technology has not been exploited to address the important question of how endogenous─as opposed to overexpressed─membrane proteins are organized within a lipid environment. Membrane proteins can be examined in near-native lipid-bilayer environments with the advent of polymer-encapsulated nanodiscs. These nanodiscs self-assemble directly from cellular membranes, allowing in vitro probing of membrane proteins with techniques that have previously been restricted to soluble or detergent-solubilized proteins. Often, however, the high charge densities of existing polymers obstruct bioanalytical and preparative techniques. Thus, the authors aim to fabricate electroneutral-yet water-soluble-polymer nanodiscs. By attaching a sulfobetaine group to the commercial polymers DIBMA and SMA(2:1), these polyanionic polymers are converted to the electroneutral maleimide derivatives, Sulfo-DIBMA and Sulfo-SMA(2:1). Sulfo-DIBMA and Sulfo-SMA(2:1) readily extract proteins and phospholipids from artificial and cellular membranes to form nanodiscs. Crucially, the electroneutral nanodiscs avert unspecific interactions, thereby enabling new insights into protein-lipid interactions through lab-on-a-chip detection and in vitro translation of membrane proteins. Finally, the authors create a library comprising thousands of human membrane proteins and use proteome profiling by mass spectrometry to show that protein complexes are preserved in electroneutral nanodiscs.

Neurodegenerative disorders are among the most common diseases in modern society. However, the molecular bases of diseases such as multiple sclerosis or Charcot-Marie-Tooth disease remain far from being fully understood. Research in this field is limited by the complex nature of native myelin and by difficulties in obtaining good in vitro model systems of myelin. Here, we introduce an easy-to-use model system of the myelin sheath that can be used to study myelin proteins in a native-like yet well-controlled environment. To this end, we present myelin-mimicking nanodiscs prepared through one of the amphiphilic copolymers styrene/maleic acid (SMA), diisobutylene/maleic acid (DIBMA), and styrene/maleimide sulfobetaine (SMA-SB). These nanodiscs were tested for their lipid composition using chromatographic (HPLC) and mass spectrometric (MS) methods and, utilizing spin probes within the nanodisc, their comparability with liposomes was studied. In addition, their binding behavior with bovine myelin basic protein (MBP) was scrutinized to ensure that the nanodiscs represent a suitable model system of myelin. Our results suggest that both SMA and SMA-SB are able to solubilize the myelin-like (cytoplasmic) liposomes without preferences for specific lipid headgroups or fatty acyl chains. In nanodiscs of both SMA and SMA-SB (called SMA(-SB)-lipid particles, short SMALPs or SMA-SBLPs, respectively), the polymers restrict the lipids' motion in the hydrophobic center of the bilayer. The headgroups of the lipids, however, are sterically less hindered in nanodiscs when compared with liposomes. Myelin-like SMALPs are able to bind bovine MBP, which can stack the lipid bilayers like in native myelin, showing the usability of these simple, well-controlled systems in further studies of protein-lipid interactions of native myelin.

The insertion of protein domains into membranes is an important step in many membrane remodeling processes, for example in vesicular transport. The membrane area taken up by the protein insertion influences the protein binding affinity as well as the mechanical stress induced in the membrane and thereby its curvature. Total area changes in lipid monolayers can be measured on a Langmuir film balance. Finding the area per inserted protein however proves challenging for two reasons: The number of inserted proteins must be determined without disturbing the binding equilibrium and the change in the film area can be very small. Here we address both issues using Fluorescence Correlation Spectroscopy (FCS): Firstly, by labeling a fraction of the protein molecules fluorescently and performing FCS experiments directly on the monolayer, the number of inserted proteins is determinedin situwithout having to rely on invasive techniques, such as collecting the monolayer by aspiration. Secondly, by using another FCS color channel and adding a small fraction of fluorescent lipids, the reduction in fluorescent lipid density accompanying protein insertion can be monitored to determine the total area increase. Here, we use this method to determine the insertion area per molecule of Sar1, a protein of the COPII complex, which is involved in transport vesicle formation, in a lipid monolayer. Sar1 has an N-terminal amphipathic helix, which is responsible for membrane binding and curvature generation. An insertion area of (3.4 ± 0.8) nm2was obtained for Sar1 in monolayers from a lipid mixture typically used in reconstitution, in good agreement with the expected insertion area of the Sar1 amphipathic helix. By using the two-color approach, determining insertion areas relies only on local fluorescence measurements. No macroscopic area measurements are needed, giving the method the potential to be applied also to laterally heterogeneous monolayers and bilayers.Statement of SignificanceWe show that two color Fluorescence Correlation Spectroscopy (FCS) measurements can be applied to the binding of a protein to a lipid monolayer on a Langmuir film balance in order to determine the protein insertion area. One labelling color was used to determine the number of bound proteins and the other one to monitor the area expansion of the lipid monolayer upon protein binding. A strategy for the FCS data analysis is provided, which includes focal area calibration by raster image correlation spectroscopy and a framework for applying z-scan FCS and including free protein in the aqueous subphase. This approach allows determining an area occupied by a protein in a quasi-planar model membrane from a local, non-invasive, optical measurement.

The high abundance of most viruses in infected host cells benefits their structural characterization; endogenous viruses are present in low copy numbers, however, and are therefore challenging to investigate. Here, we retrieve cell extracts enriched with an endogenous virus, the yeast L-A virus. The determined cryo-EM structure discloses capsid-stabilizing cation-π stacking and an interplay of non-covalent interactions from ten distinct capsomere interfaces. The capsid-embedded mRNA decapping active site trench is supported by a constricting movement of two opposite-facing loops. tRNA-loaded polysomes and other biomacromolecules, presumably mRNA, are found in virus proximity while stacked dsRNA bundles and the sub-stoichiometric polymerase localize underneath the capsid surface. Mature viruses participate in larger viral communities resembling their rare in-cell equivalents in terms of size, composition, and inter-virus distances. Our results collectively describe a 3D-architecture of a viral milieu, opening the door to cellextract-based high-resolution structural virology.

The N-BAR domain of the human Bin1 protein is indispensable for T-tubule biogenesis in skeletal muscles. It binds to lipid mono- and bilayers that mimic the sarcolemma membrane composition, and it transforms vesicles into uniform tubules by generating a decorating protein scaffold. Six single-chain, 1,32-alkyl-branched bis(phosphocholines) PC-C32(1,32Cm)-PC have been synthesized as model lipids for naturally occurring archaeal membrane lipids. The preparation of these bipolar amphiphiles bearing lateral alkyl chains of different lengths (C4-C15) was realized using a Cu-catalyzed Grignard bis-coupling reaction of various primary alkyl-branched bromides as side parts and a 1,22-dibromide as the centre part. The aggregation behaviour of these bolalipids in water was initially investigated by differential scanning calorimetry and transmission electron microscopy. As a main result, the types of aggregates found and their stability upon heating were strongly connected to the length of the lateral alkyl chain of the bolalipid: short and long lateral chains led to lamellar structures, whereas side chains of medium length led to fibrous aggregates. In future, these bolalipids could be used to produce tailored and stabilized liposomes for oral drug delivery.

Most membrane-solubilising amphiphilic copolymers such as diisobutylene/maleic acid (DIBMA) and styrene/maleic acid (SMA) carry high negative charge densities. Their polyanionic character results in strong Coulombic repulsion, both between polymer chains and lipid membranes during the solubilisation process as well as among polymer-encapsulated nanodiscs after solubilisation. Coulombic repulsion is attenuated by charge screening and, more efficiently, by counterion association, which is particularly strong for multivalent cations binding to polyanionic copolymers. Here, we investigated the effects of the two common alkaline earth metal ions Mg2+ and Ca2+ on the solubilisation properties of and the nanodiscs formed by DIBMA. By quantifying the kinetics and the equilibrium efficiency of lipid solubilisation by static and dynamic light scattering, respectively, we found that millimolar concentrations of Mg2+ or Ca2+ accelerated DIBMA-mediated lipid solubilisation several-fold and resulted in considerably smaller nanodiscs than without divalent cations. Time-resolved Forster resonance energy transfer spectroscopy revealed that collisional transfer of phospholipids among nanodiscs was up to ∼20 and ∼25 times faster in the presence of 10 mM Mg2+ or 7.5 mM Ca2+ than in the absence of divalent cations. These major effects of Mg2+ and Ca2+ contrasted with a moderate influence on the morphology and the thermotropic phase behaviour of the nanodiscs. Finally, we compared the yields of membrane-protein extraction from Escherichia coli membranes, which increased by up to two-fold upon addition of Mg2+ or Ca2+. None of these effects could be explained by Coulombic screening alone, as the change in ionic strength resulting from low millimolar concentrations of divalent cations was minor. Thus, we conclude that Mg2+ and Ca2+ specifically associated with DIBMA to neutralise part of the polymer's carboxylate groups.

Advances in electron microscopy have provided unprecedented access to the structural characterization of large, flexible and heterogeneous complexes. Until recently, cryo-electron microscopy (cryo-EM) has been applied to understand molecular organization in either highly purified, isolated biomolecules or in situ. An emerging field is developing, bridging the gap between the two approaches, and focuses on studying molecular organization in native cell extracts. This field has demonstrated its potential by resolving the structure of fungal fatty acid synthase (FAS) at 4.7 Å [Fourier shell correlation (FSC) = 0.143]; FAS was not only less than 50% enriched, but also retained higher-order binders, previously unknown. Although controversial in the sense that the lysis step might introduce artifacts, cell extracts preserve aspects of cellular function. In addition, cell extracts are accessible, besides cryo-EM, to modern proteomic methods, chemical cross-linking, network biology and biophysical modeling. We expect that automation in imaging cell extracts, along with the integration of molecular/cell biology approaches, will provide remarkable achievements in the study of closer-to-life biomolecular states of pronounced biotechnological and medical importance. Such steps will, eventually, bring us a step closer to the biophysical description of cellular processes in an integrative, holistic approach.

Although the incorporation of photo-activatable lipids into membranes potentially opens new avenues for studying interactions with peptides and proteins, the question of whether azide- or diazirine-modified lipids are suitable for such studies remains controversial. We have recently shown that diazirine-modified lipids can indeed form cross-links to membrane peptides after UV activation and that these cross-links can be precisely determined in their position by mass spectrometry (MS). However, we also observed an unexpected backfolding of the lipid's diazirine-containing stearoyl chain to the membrane interface challenging the potential application of this modified lipid for future cross-linking (XL)-MS studies of protein/lipid interactions. In this work, we compared an azide- (AzidoPC) and a diazirine-modified (DiazPC) membrane lipid regarding their self-assembly properties, their mixing behavior with saturated bilayer-forming phospholipids, and their reactivity upon UV activation using differential scanning calorimetry (DSC), dynamic light scattering (DLS), small-angle X-ray scattering (SAXS), transmission electron microscopy (TEM), and MS. Mixtures of both modified lipids with DMPC were further used for photo-chemically induced XL experiments with a transmembrane model peptide (KLAW23) to elucidate similarities and differences between the azide and the diazirine moiety. We showed that both photo-reactive lipids can be used to study lipid/peptide and lipid/protein interactions. The AzidoPC proved easier to handle, whereas the DiazPC had fewer degradation products and a higher cross-linking yield. However, the problem of backfolding occurs in both lipids; thus, it seems to be a general phenomenon.

When membrane proteins are removed from their natural environment, the quality of the membrane-solubilizing agent used is critical for preserving their native structures and functions. Nanodiscs that retain a lipid-bilayer core around membrane proteins have attracted great attention because they offer a much more native-like environment than detergent micelles. Here, two small-molecule amphiphiles with diglucose headgroups and either a hydrocarbon or a fluorocarbon hydrophobic chain are shown to directly assemble lipids and membrane proteins to form native nanodiscs rather than mixed micelles. Self-assembly of nanodiscs of increasing complexity from both defined, artificial vesicles as well as complex, cellular membranes is demonstrated. A detailed investigation of bilayer integrity and membrane-protein activity in these nanodiscs reveals gentle effects on the encapsulated bilayer core. The fluorinated amphiphile appears particularly promising because its lipophobicity results in gentle, non-perturbing interactions with the nanoscale lipid bilayer. A sequential model of nanodisc self-assembly is proposed that proceeds through perforation of the original membrane followed by saturation and complete solubilization of the bilayer. On this basis, pseudophase diagrams are established for mixtures of lipids and nanodisc-forming diglucoside amphiphiles, and the latter are used for the extraction of a broad range of membrane proteins from cellular membranes.

Lateral flow immunoassays (LFI) are valuable tools for point-of-care testing. However, their sensitivity is limited and can be further improved. Nanoparticles (NP) of conjugated polymers (CPNs), also known as Pdots, are reported to be highly sensitive fluorescent probes, but a direct comparison with conventional colloidal gold-based (Au-NP) LFI using the same antibody–antigen pair is missing to date. Furthermore, the influence of brightness and Stokes shift of CPs on the signal : background ratio (SBR) needs to be evaluated. In this study, we encapsulated two different CPs, poly-(9,9-di-n-octyl-fluorenyl-2,7-diyl) (PDOF) and poly-(2,5-di-hexyloxy-cyanoterephthalylidene) (CN-PPV) in silica shell-crosslinked Pluronic© micelles (Si-NP) and Pdots and investigated the NP brightness with respect to CP loading dose. The brightest formulation of each NP system was conjugated to rabbit IgG as a model antigen and the SBR was investigated in an ELISA-like microplate assay and LFI. Two reference particles, Au-NP and a polystyrene NP (PS-NP) loaded with a small-molecule fluorescent dye were conjugated to IgG and compared to the Si-NP and Pdots. The mass of Pdots required for detection in LFI was at least two orders of magnitude lower than that of Si-NP and the reference NP. The SBR of CN-PPV (moderate brightness, large Stokes shift) was two to three times higher than the SBR of PDOF (high brightness, small Stokes shift). To combine the favourable properties of both CPs, a polymer blend of PDOF and CN-PPV was encapsulated in Pdots, and resulted in further increase of SBR in the microplate assay and LFI. In summary, combining two CPs with different properties can lead to fluorescent signal-transducers for applications such as ELISA and LFIs, which can enhance the detection limit of the assay by 2–3 orders of magnitude.

Here, we for the first time establish nanodiscs with the challenging lipid composition of myelin of the peripheral or central nervous systems, respectively (PNS and CNS, both containing >40% cholesterol, which so far has been thought to be detrimental for nanodisc formation).Thus, we prove that more complex lipid model membrane systems are in general accessible through nanodiscs and can study protein-lipid interactions in myelin and factors driving myelin formation or degradation using combinations of myelin proteins in a highly controlled lipid environment resembling myelin’s cytoplasmic leaflet. For the functional studies, initial proof-of-principle experiments using myelin basic protein have been performed.

Amphiphilic copolymers that directly extract membrane proteins and lipids from cellular membranes to form nanodiscs combine the advantages of harsher membrane mimics with those of a native-like membrane environment. Among the few commercial polymers that are capable of forming nanodiscs, alternating diisobutylene/maleic acid (DIBMA) copolymers have gained considerable popularity as gentle and UV-transparent alternatives to aromatic polymers. However, their moderate hydrophobicities and high electric charge densities render all existing aliphatic copolymers rather inefficient under near-physiological conditions. Here, we introduce Glyco-DIBMA, a bioinspired glycopolymer that possesses increased hydrophobicity and reduced charge density but nevertheless retains excellent solubility in aqueous solutions. Glyco-DIBMA outperforms established aliphatic copolymers in that it solubilizes lipid vesicles of various compositions much more efficiently, thereby furnishing smaller, more narrowly distributed nanodiscs that preserve a bilayer architecture and exhibit rapid lipid exchange. We demonstrate the superior performance of Glyco-DIBMA in preparative and analytical applications by extracting a broad range of integral membrane proteins from cellular membranes and further by purifying a membrane-embedded voltage-gated K+ channel, which was fluorescently labeled and analyzed with the aid of microfluidic diffusional sizing (MDS) directly within native-like lipid-bilayer nano-discs.

The mixing behavior of the symmetric long-chain bolaamphiphile dotriacontane-1,1′-diyl-bis-[2-(trimethylammonio)ethylphosphate] (PC-C32-PC) with conventional phospholipids (DPPC, DMPC, and POPC) was investigated by differential scanning calorimetry and transmission electron microscopy. PC-C32-PC by itself forms nanofibers at room temperature, which convert to micellar aggregates at higher temperatures. The length of the bolaamphiphile corresponds roughly to the thickness of a lipid bilayer. However, a significant insertion of the bolaamphiphile into the phospholipid bilayer vesicles was not observed in any of the mixed systems. This is apparently due to packing problems caused by the larger space requirement of the phosphocholine headgroups of PC-C32-PC compared to the small cross-sectional area of the alkyl chain. Obviously, the insertion of these bolaamphiphiles into membrane structures with parallel oriented molecules leads to large void volumes because of this packing problem, so that the bolaamphiphiles prefer to self-assemble into fibers, where the chains can get in closer contact. On the other hand, up to a certain molar ratio, double-chain phospholipids can easily be inserted into the hydrophobic pockets of the bolaamphiphile fibers, where they stabilize the fiber structure.

In a biological membrane, proteins require specific lipids of distinctive length and chain saturation surrounding them. The active tuning of the membrane thickness therefore opens new possibilities in the study and manipulation of membrane proteins. Here, we introduce the concept of stapling phospholipids to different degrees of interdigitation depth by mixing 1,3-diamidophospholipids with single-chain bolalipids. The mixed membranes were studied by calorimetric assays, electron microscopy, X-ray, and infrared measurements to provide a complete biophysical characterization of membrane stapling. The matching between the diamidophospholipids and the bolalipids can be so strong as to completely induce a new phase that is more stable than the gel phase of the individual components.

The natural product emodin (EO) exhibits anti-inflammatory, antiangiogenesis and antineoplastic properties in vitro and in vivo. Due to its biological properties as well as its fluorescence, EO can be useful in pharmacology and pharmacokinetics. To enhance its selectivity to cancer cells, EO was loaded into non-fluorescent and novel fluorescent spherical mesoporous nanoparticles bearing N-methyl isatoic anhydride (SNM~M) or lissamine rhodamine B sulfonyl moieties (SNM~L). The propylamine functionalized mesoporous silica nanomaterial (SNM) were characterized by powder X-ray diffraction (XRD), nitrogen gas sorption, scanning electron microscopy (SEM), transmission electron microscopy (TEM), fluorescence spectroscopy, thermogravimetric analysis (TGA) and UV spectroscopy. The cytotoxicity of EO-loaded nanoparticles was tested against the human colon carcinoma cell line HT-29. Non-loaded SNM did not affect cell proliferation, whereas those loaded with EO were at least as efficient as EO alone. It could be shown by fluorescence microscopy that the uptake of silica nanomaterial by the tumor cells occurred within 2 h and the release of EO occurred within 48 h of treatment. Flow cytometry and Western blot analysis showed that SNM containing EO induced apoptosis in HT-29 cells.

Bolaamphiphiles are well-known naturally occurring structures that can increase the thermal and mechanical stability of the phospholipid membrane by incorporation in a transmembrane manner. Modifications of bolaamphiphiles to introduce particular structural elements such as a conjugated aromatic backbone and lateral side chains in the hydrophobic region lead to bolapolyphiles (BPs). We investigated the ability of BPs to form lyotropic phases in water. The BPs had an identical backbone and side chains, but different headgroup structures, leading to different abilities to act as hydrogen bond donors and acceptors. BPs with hydrophilic headgroups capable of acting as hydrogen bond donors as well as acceptors did not form lyotropic phases and were insoluble in water, independent of whether the polar groups were small or large. The extended lipophilic core structure and the multiple intermolecular hydrogen bonds between the headgroups prevented the formation of well-hydrated lyotropic aggregates. A BP with two large hydrophilic headgroups of several ethylene oxide moieties terminated by methyl groups formed sheet- and vesicle-like aggregates in water. These headgroups act only as hydrogen bond acceptors and cannot form hydrogen bonds in the absence of water. The miscibility of BPs with vesicles of 1,2-dipalmitoyl- sn-glycero-3-phosphocholine (DPPC) in water and the resulting aggregate structures were also investigated. For BPs with headgroups acting as donors and acceptors of hydrogen bonds, macroscopic phase separation occurred in the mixed membranes, and two different membrane domains, a DPPC-rich one containing only little polyphile and a BP-rich one containing varying amounts of lipid, were formed. For headgroups without the ability to act as hydrogen bond donors, small BP aggregates were formed that were homogeneously distributed over the membrane. The lateral organization of BPs in lipid membranes can thus be controlled by the nature of the BP headgroup.

The linear polyester poly(glycerol adipate) (PGA) with its free pendant hydroxyl groups was covalently grafted with indomethacin which yields polymeric prodrugs. It was possible to produce nanospheres with narrow particle size distribution of these polymer-drug conjugates with an optimized interfacial deposition method. Nanospheres were characterized by zeta potential measurements, dynamic light scattering, electron microscopy and nanoparticle tracking analysis. Moreover, cell viability studies and cytotoxicity tests in three different cell lines were carried out showing low toxicity for three different degrees of grafting. In addition, the nanospheres had (in contrast to the free drug) low hemolytic activity in vitro. Release studies of nanodispersions are challenging. The use of a specially developed setup with highly porous aluminum oxide membranes enabled us to overcome problems associated with other setups (e.g. dialysis membranes). A slow and controlled release profile without any burst was observed over 15 days. The results indicate that indomethacin-PGA conjugates can be formulated successfully as nanospheres with the desired characteristics of small size with narrow distribution, controlled drug release and low toxicity. The newly developed particles have the potential to improve the therapy of inflammation and associated diseases.

Three novel single-chain bis(phosphocholines) bearing two lateral alkyl chains of variable length next to the headgroup have been synthesized as model lipids for naturally occurring archaeal membrane lipids. The synthesis was realized using the Cu-catalyzed Grignard bis-coupling reaction of a primary bromide as a side part and a 1,ω-dibromide as a centre part. We could show that the aggregation behaviour of the resulting bolalipids strongly depends on the length of the lateral alkyl chain: the C3-branched bolalipid self-assembles into lamellar sheets, whereas the C6- and C9-analogues form nanofibres. The lamella-forming bolalipids could be used in the future to prepare stable and tailored liposomes for oral drug delivery.

Poly(glycerol adipate) (PGA) was synthesized by enzymatic polymerization and grafted with different amounts of oleoyl chloride. A series of graft copolymers PGA-g-Ox was obtained where O represents the oleate side chains and x is the degree of grafting (mol% of the pendant OH groups of PGA esterified) ranging from 11 to 90. Small angle X-ray scattering (SAXS) data revealed nanophase separation of the oleate side chains into nanodomains in bulk and wide angle X-ray scattering (WAXS) profiles showed a broad halo, reflecting the amorphous nature of the synthesized graft polymers. Glycerol monooleate (GMO), an amphiphilic lipid with one hydrophobic oleate tail and a glycerol hydrophilic head group, self-assembles into various liquid crystalline phases in the presence of water. Since oleate chains are integral part of both GMO and the graft copolymers, a certain degree of miscibility can be expected influencing the self-assembly behavior. Using SAXS for phase identification, systematic studies were carried out at room temperature on the ternary system of GMO/PGA-g-O22/water and the ternary phase diagram was constructed. The addition of polymer did influence the phase behavior of the GMO/water system. The water in the lamellar and cubic mesophases was partially replaced by PGA-g-O22. Furthermore, keeping the GMO/water ratio constant at 1:1, the addition of PGA-g-O22 lead to swelling of the water channels in the inverse bicontinuous Pn3m cubic phase. Preliminary investigations on the potential application of PGA-g-O22 as a stabilizer of structured nanoparticles in aqueous dispersions were carried out employing different transmission electron microscopy techniques.

Non-viral gene delivery in its current form is largely dependent upon the ability of a delivery vehicle to protect its cargo in the extracellular environment and release it efficiently inside the target cell. Also a simple delivery system is required to simplify a GMP conform production if a marketing authorization is striven for. This work addresses these problems. We have developed a synthetic polycationic peptide-mimicking amphiphile, namely DiTT4, which shows efficient transfection rates and good biocompatibility without the use of a co-lipid in the formulation. The lipid–nucleic acid complex (lipoplex) was characterized at the structural (electron microscopy), physical (laser Doppler velocimetry and atomic force microscopy) and molecular levels (X-ray scattering). Stability studies of the lipoplexes in the presence of serum and heparin indicated a stable formation capable of protecting the cargo against the extracellular milieu. Hemocompatibility studies (hemolysis, complement activation and erythrocyte aggregation) demonstrated the biocompatibility of the formulation for systemic administration. The transfection efficiency was assessed in vitro using the GFP assay and confocal laser scanning microscopy studies. With the chorioallantoic membrane model, an animal replacement model according to the 3R strategy (replacement, refinement, and reduction), initial in vivo experiments were performed which demonstrate fast and efficient transfection in complex tissues and excellent biocompatibility.

Here we present the structure of mouse H-chain apoferritin at 2.7 Å (FSC=0.143) solved by single particle cryogenic electron microscopy (cryo-EM) using a 200 kV device. Data were collected using a compact, two-lens illumination system with a constant power objective lens, without the use of energy filters or aberration correctors. Coulomb potential maps reveal clear densities for main chain carbonyl oxygens, residue side chains (including alternative conformations) and bound solvent molecules. We argue that the advantages offered by (a) the high electronic and mechanical stability of the microscope, (b) the high emission stability and low beam energy spread of the high brightness Field Emission Gun (x-FEG), (c) direct electron detection technology and (d) particle-based Contrast Transfer Function (CTF) refinement have contributed to achieving resolution close to the Rayleigh limit. Fibrillation of supramolecular building blocks represents an important model system for complex proteins and peptides, such as amyloidogenic proteins, displaying aggregation and subsequent collapse of their biological functions. Once removed from their natural environment, membrane proteins depend on membrane‐mimetic systems to retain their native structures and functions. To this end, lipid‐bilayer nanodiscs that are bounded by scaffold proteins or amphiphilic polymers such as styrene/maleic acid (SMA) copolymers have been introduced as alternatives to detergent micelles and liposomes for in vitro membrane‐protein research. Herein, we show that an alternating diisobutylene/maleic acid (DIBMA) copolymer shows equal performance to SMA in solubilizing phospholipids, stabilizes an integral membrane enzyme in functional bilayer nanodiscs, and extracts proteins of various sizes directly from cellular membranes. Unlike aromatic SMA, aliphatic DIBMA has only a mild effect on lipid acyl‐chain order, does not interfere with optical spectroscopy in the far‐UV range, and does not precipitate in the presence of low millimolar concentrations of divalent cations.

H-Bonded semifluorinated dendritic polymer networks are generated by association of macromolecules, driven by the weak diaminopyridine (DAP) and thymine (THY) couple. Solvent-dependent morphologies are observed: a continuous film structure when drop-cast from tetrahydrofuran (THF) solution or uniform nanoparticles with a diameter of ∼50 nm in toluene.

Phosphatidylserine (PS) and phosphatidylglycerol (PG) are naturally occurring phospholipids (PL) with intrinsic anti-inflammatory properties. The therapeutic potential of PS and PG has not been extensively explored and the main focus had been directed towards PS- and PG-liposomes. In order to increase the formulation options, we explored whether mixed micelles (MM) could be an alternative to liposomes. Potential advantages of MM are their thermodynamic stability, small size and ease of manufacture. DOPS- and DOPG-enriched MM were obtained via a co-precipitation technique and physicochemical characterization was performed. The MM, approximately 10 nm in diameter, showed no toxicity on fibroblast cell lines in vitro and virtually no hemolytic activity. The MM suppressed the TNFα-production of mIFNγ/LPS-stimulated mouse peritoneal macrophages (MPM) in vitro similar to DOPS- and DOPG-liposomes. Therefore, DOPS- and DOPG-loaded MM are promising new options for the treatment of inflammatory diseases.

The nanofiber formation in aqueous suspension of two classes of symmetric single-chain bolaamphiphiles with different polar headgroups and a diacetylene-modified alkyl chain with a length of 32, 34, and 36 C atoms was investigated by differential scanning calorimetry, transmission electron microscopy, and small-angle neutron scattering. As observed before for other bolalipids with phosphocholine (PC) and dimethyl-phosphoethanolamine (Me2PE) headgroups, the molecules form fibers when suspended in water at low temperatures but disassemble into micellar-like aggregates upon heating. The introduction of a diacetylene group in the middle of the long chain leads to a perturbation of chain packing so that this fiber-micelle transition occurs at lower temperature compared to the other bolalipids having unmodified alkyl chains. The aim of our project was the introduction of diacetylene groups into alkyl chains to be able to polymerize the fibers at low temperature. This should enhance the fiber stability and prevent the disassembly into micellar aggregates at higher temperature. Polymerization of aggregates containing diacetylene-modified bolaamphiphiles can be easily traced by UV/vis spectroscopy as colored products are formed. We found that polymerization of bolaamphiphiles with PC headgroups leads to a breakdown of most fibers into micellelike aggregates, and only some longer fibers segments are still detectable. In contrast, the use of Me2PE headgroups improves polymerizability and length of the polymerized fibers. The compound with 36 C atoms in the chain could be polymerized at low temperatures, and the fibers remained stable at least up to a temperature of 60 °C. This shows that the perturbation of the chain packing due to the diacetylene groups in the chains can be overcome by elongation of the chains, so that thermostable fibers with a diameter of the length of the bolalipid molecule can be successfully formed.

Most membrane-solubilising amphiphilic copolymers such as diisobutylene/maleic acid (DIBMA) and styrene/maleic acid (SMA) carry high negative charge densities. Their polyanionic character results in strong Coulombic repulsion, both between polymer chains and lipid membranes during the solubilisation process as well as among polymer-encapsulated nanodiscs after solubilisation. Coulombic repulsion is attenuated by charge screening and, more efficiently, by counterion association, which is particularly strong for multivalent cations binding to polyanionic copolymers. Polymeric ionic liquids (POILs) are important materials in the field of ionic liquid gating, requiring the precise synthesis of new POILs with tailored structural variability and defined nanoscaled structure. In the current contribution, using reversible addition–fragmentation chain-transfer polymerization (RAFT) technique, the homopolymerization of three imidazolium-based acrylates with different counterions is reported, namely 1-[2-acryloylethyl]-3-methylimidazolium bis(trifluoromethane)sulfonamide (APMIN(Tf)2), 1-[2-acryloylethyl]-3-methylimidazolium hexafluorophosphate (APMIPF6), and 1-[2-acryloylethyl]-3-methylimidazolium tetrafluoroborate (APMIBF4), to afford the respective poly(ionic liquid)s (POILs). All polymerizations display pseudo-first-order kinetics and a rapid growth of P(APMIN(Tf)2), P(APMIPF6), and P(APMIBF4), yielding homopolymers with controlled molar mass, revealing a strong influence of the counterions on the polymerization rate, increasing in the order of BF4Θ < PF6Θ < N(Tf)2Θ. As a direct determination of molecular weights via size exclusion chromatography (SEC) of the POIL homopolymers from RI and UV detectors was not successful, we developed an alternative strategy to generate accurate, “normal” SEC peaks of POILs using RAFT copolymerization technique: APMIN(Tf)2 was copolymerized with a semifluorinated monomer 2,2,2-trifluoroethyl acrylate (TFEA), allowing to study the influence of the comonomer feeding ratio on the resulting SEC signals of P(APMIN(Tf)2-co-TFEA) copolymers. We found that when the feeding molar ratio of TFEA is adjusted to 0.77, symmetric SEC peaks from the resulting P(APMIN(Tf)2-co-TFEA) copolymers are obtained. Furthermore, copolymerizations of TFEA with the other two IL monomers, APMIPF6 and APMIBF4, are also performed to afford P(APMIPF6-co-TFEA) and P(APMIBF4-co-TFEA) copolymers. Moreover, the propensity of the so-obtained POIL random copolymers P(APMIN(Tf)2-co-TFEA), P(APMIPF6-co-TFEA), and P(APMIBF4-co-TFEA) to grow a new block (polypentafluorostyrene, PPFS) is explored, intending to generate the fluorinated POIL triblock copolymers P(APMIN(Tf)2-co-TFEA)-b-PPFS-b-P(APMIN(Tf)2-co-TFEA), P(APMIPF6-co-TFEA)-b-PPFS-b-P(APMIPF6-co-TFEA), and P(APMIBF4-co-TFEA)-b-PPFS-b-P(APMIBF4-co-TFEA), respectively. The morphology and size of such semifluorinated POILs are investigated using transition electron microscopy (TEM), atomic force microscopy (AFM), and dynamic light scattering (DLS), revealing the aggregated nanoparticles from P(APMIN(Tf)2-co-TFEA) due to the mesoscale organization of the ionic “multiplets”. Significantly larger and crowded globular objects/aggregates are formed from the chain-extended POIL triblock P(APMIN(Tf)2-co-TFEA)-b-PPFS-b-P(APMIN(Tf)2-co-TFEA) copolymers under the same conditions.

Liposomes are a promising class of drug delivery vehicles. However, no liposomal formulation has been approved for an oral application so far, due to stability issues of the liposomes in the gastrointestinal tract. Herein, we investigate the miscibility of three novel single-chain alkyl-branched bolalipids PC-C32(1,32Cn)-PC (n = 3, 6, 9) with either saturated or unsaturated phosphatidylcholines by means of differential scanning calorimetry (DSC), transmission electron microscopy (TEM) of stained samples, vitrified specimens, or replica of freeze-fractured samples, and dynamic light scattering (DLS). The novel bolalipids contain lateral alkyl chains of different length in 1- and 32-position of the long membrane-spanning C32 alkyl chain. We will show for the first time that these single-chain alkyl-branched bolalipids show a miscibility with bilayer-forming phospholipids—by maintaining the vesicular aggregate structure—due to the lateral alkyl substituents located next to the phosphocholine headgroup of the bolalipid. We are convinced that these alkyl side chains are able to fill the void volume, which is created when unmodified single-chain bolalipids are inserted in a transmembrane fashion into a phospholipid bilayer. Consequently, the miscibility of our alkyl-chained bolalipids with bilayer-forming phospholipids rose with increasing lengths of the lateral alkyl chain of the bolalipid. Finally, we were successful in preparing liposomes from various bolalipid/phospholipid mixtures, which were stable in size upon storage for at least 21 days. These mixed liposomes (bolasomes) could be used as oral drug delivery systems in the near future.

Liposomes have received attention as a promising class of drug delivery vehicles. To date, many approaches have been tried developing liposomes for oral use. However, no liposomal formulation is on the market so far that is approved for oral application. In this study, we investigate the miscibility of two glycerol diether bolalipids with classical saturated and unsaturated phosphatidylcholines by means of differential scanning calorimetry (DSC), transmission electron microscopy (TEM), and dynamic light scattering (DLS). Our bolalipids contain a long C32 alkyl chain bound to glycerol in the sn-3 position and a short C16 in the sn-2 position, which further carries a racemic methyl branch. The sn-1 position of the glycerol as well as the end of the long C32 alkyl chain contain polar headgroups: either two phosphocholine headgroups (PC-Gly(2C16Me)C32-PC) or a phosphocholine and a phosphodimethylethanolamine headgroup (PC-Gly(2C16Me)C32-Me2PE). We demonstrate that glycerol diether bolalipids show better miscibility with unsaturated phosphatidylcholines than with saturated ones. Both bolalipids in mixture with the unsaturated 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) form liposomes, which are stable in size upon storage. These mixed bolalipid/phospholipid vesicles could be used as an oral liposomal formulation in the future.

In the present work, we describe the synthesis and the temperature-dependent aggregation behavior of a new class of asymmetrical glycerol diether bolalipids. These bolalipids are composed of a membrane-spanning alkyl chain with 32 carbon atoms (C32) in the sn-3 position, a methyl-branched C16 alkyl chain in the sn-2 position, and a zwitterionic phosphocholine headgroup in the sn-1 position of a glycerol moiety. The long C32 alkyl chain is terminated either by a second phosphocholine (PC-Gly(2C16Me)C32-PC) or by a phosphodimethylethanolamine headgroup (PC-Gly(2C16Me)C32-Me2PE). The temperature- and pH-dependent aggregation behavior of both lipids was studied using differential scanning calorimetry (DSC), Fourier transform infrared (FTIR) spectroscopy, small-angle X-ray scattering (SAXS), and small-angle neutron scattering (SANS) experiments. In this study, synthesis and dynamic properties of amphiphilic core–shell polymers are reported as monitored through their interaction with small amphiphilic molecules. Brush-like structures are formed with a hydrophobic core surrounded by a hydrophilic shell utilizing controlled radical addition–fragmentation chain transfer (RAFT) polymerization of macromonomers consisting of linear polyglycerol chains attached to alkylene methacrylate. Continuous wave electron paramagnetic resonance (CW EPR) spectroscopy is employed to study how the amphiphilic, paramagnetic spin probe 16-DSA (16-doxyl stearic acid) interacts with polymers of different alkylene chain lengths in their hydrophobic cores and different polyglycerol chain lengths in their hydrophilic shells. The spin probe exhibits dynamic hydrophobic attachment to the polymers and reveals an indirect, dynamics-based view of polymer effects such as temperature response, aggregation and ligand binding properties. Increasing the hydrophobic alkylene chain length in the polymers alters the physical properties of the core region significantly. A large set of controllable functional polymer properties can be adjusted by the degree of polymerization and alkylene spacer length. Partial aggregation of the polymers further modifies the binding properties. Applying dynamic light scattering (DLS), transmission electron microscopy (TEM) and molecular dynamic (MD) simulations, the complex dynamic behavior found with EPR spectroscopy was further complemented and verified.

The synthesis of a new class of cationic lipids, tris(2-aminoethyl)amine-based α-branched fatty acid amides, is described resulting in a series of lipids with specific variations in the lipophilic as well as the hydrophilic part of the lipids. In-vitro structure/transfection relationships were established by application of complexes of these lipids with plasmid DNA (pDNA) to different cell lines. The α-branched fatty acid amide bearing two tetradecyl chains and two lysine molecules (T14diLys) in mixture with the co-lipid 1,2-di-[(9Z)-octadec-9-enoyl]-sn-glycero-3-phosphoethanolamine (DOPE) (1/2, n/n) exhibits effective pDNA transfer in three different cell lines, namely Hep-G2, A549, and COS-7. The presence of 10% serum during lipoplex incubation of the cells did not affect the transfection efficiency. Based on that, detailed investigations of the complexation of pDNA with the lipid formulation T14diLys/DOPE 1/2 (n/n) were carried out with respect to particle size and charge using dynamic light scattering (DLS), ζ-potential measurements, and transmission electron microscopy (TEM). Additionally, the lipoplex uptake was investigated by confocal laser scanning microscopy (CLSM). Overall, lipoplexes prepared from T14diLys/DOPE 1/2 (n/n) offer large potential as lipid-based polynucleotide carriers and further justify advanced examinations.
