African pangolins are intensively harvested to feed illegal trade networks. We focused on the conservation genetics of the most trafficked African species, the white-bellied pangolin (WBP;Phataginus tricuspis), through the genotyping of 562 individuals from reference populations and urban bushmeat markets in a vibrant trade hub from western Central Africa. Across Cameroon, Equatorial Guinea and northern Gabon, we observed a lack of genetic differentiation and a signature of isolation-by-distance possibly due to unsuspected dispersal capacities involving a Wahlund effect. Despite a higher level of genetic diversity compared to western Africa, we detected a 74-83% decline in the effective population size of WBP during the Middle Holocene. Private allele frequency tracing approach indicated up to 600 km sourcing distance by large urban markets from Cameroon, involving transnational trade activities. The 20 microsatellites markers used in this study provided the necessary power to distinguish among all WBP individuals and should be considered a valuable resource for future forensic applications. Because lineage admixture was detected in the study area, we recommend a multi- locus approach for tracing the WBP trade. The Yaoundé market was a major recruiter of genetic diversity in the region, and should receive urgent conservation action to mitigate the pangolin trade.

Background: The scimitar‐horned oryx (Oryx dammah) (SHO) is a large African antelope that became extinct in the wild just over two decades ago. Conservation of the species is of prime importance, but it might face pathogen stressors. Methods and principal findings: Brucella melitensis biovar 1 was previously confirmed in a high‐density captive population of SHO held in Abu‐Dhabi emirate. The infection reached 67.0 % (95 % CI: 64.0–70.0) individual seroprevalence (n = 959) during testing performed between January 2013 and January 2015. A model based on a multivariable logistic regression analysis showed that the seroprevalence ranged from 51.2 (95 % CI: 39.6–62.7) to 86.9 % (95 % CI: 82.4–91.4) between six different enclosures, and probability of being seropositive was 1.83 (95 % CI: 1.32–2.55) higher in females than in males, 3.09 (95 % CI: 1.66–5.91) and 9.35 (95 % CI: 4.66–19.44) higher in subadults and adults than in juveniles, respectively. The three serological tests used in this study (Rose Bengal Test, lateral flow assay and in‐house i‐ELISA) had a perfect or near‐perfect agreement (Cohen’s Kappa coefficient >=0.97). Recurrent high seroprevalence in time and congruence of results from three different serological tests point toward a persistent B. melitensis infection in a high‐density captive SHO population. Conclusion and significance Testing strategy (Bengal Test, lateral flow assay or in‐house i‐ELISA) has no effect on the estimation of the brucellosis seroprevalence in SHO permitting the selection of a practical test. We call for an evidence‐based control program, and Brucella vaccine efficacy and innocuity studies in this endangered species.

Kangaroos are considered to be an important reservoir of Q fever in Australia, although there is limited knowledge on the true prevalence and distribution of coxiellosis in Australian macropod populations. Serological tests serve as useful surveillance tools, but formal test validation is needed to be able to estimate true seroprevalence rates, and few tests have been validated to screen wildlife species for Q fever. In this study, we modified and optimized a phase-specific indirect immunofluorescence assay (IFA) for the detection of IgG antibodies against Coxiella burnetii in macropod sera. The assay was validated against the commercially available ID Screen Q fever indirect multispecies enzyme-linked immunosorbent assay (ELISA) kit (IDVet, Grabels, France) to estimate the diagnostic sensitivity and specificity of each assay, using Bayesian latent class analysis. A direct comparison of the two tests was performed by testing 303 serum samples from 10 macropod populations from the east coast of Australia and New Zealand. The analysis indicated that the IFA had relatively high diagnostic sensitivity (97.6% [95% credible interval [CrI], 88.0 to 99.9]) and diagnostic specificity (98.5% [95% CrI, 94.4 to 99.9]). In comparison, the ELISA had relatively poor diagnostic sensitivity (42.1% [95% CrI, 33.7 to 50.8]) and similar diagnostic specificity (99.2% [95% CrI, 96.4 to 100]) using the cutoff values recommended by the manufacturer. The estimated true seroprevalence of C. burnetii exposure in the macropod populations included in this study ranged from 0% in New Zealand and Victoria, Australia, up to 94.2% in one population from New South Wales, Australia.

Unvaccinated animal science and veterinary students are at risk of Q fever, a vaccine-preventable zoonotic disease transmitted from animals to humans. We investigated students’ perspectives on Q fever prevention using a One Health approach combining animal, human and environmental health. Animal science and veterinary students enrolled at the University of Adelaide in 2019 were invited to participate in an online survey to explore their perceptions about Q fever and prevention strategies. We undertook descriptive analysis and logistic regression. Overall, 46% of students reported little to no knowledge of Q fever. Over three-quarters of students reported transmission of Q fever via aerosol inhalation and animal culling, whilst other modes including sexual transmission between humans (7%) was poorly identified. The majority of students reported exposure to cattle, sheep and goats. Of those who reported vaccination status, 5% veterinary and 61% animal science students were unvaccinated for Q fever. Challenges concerning vaccination included cost, time and access to healthcare with strategies aimed at promoting awareness, improving healthcare access and subsidized and mass vaccination. Knowledge of Q fever among respondents, particularly about disease transmission was suboptimal. As a key principle to One Health approach, adherence to biosecurity guidelines during contact with animals could potentially reduce zoonoses including Q fever transmission. Improving access to Q fever vaccine for unvaccinated students is a priority. Findings underscore that university policy for Q fever vaccination needs to consider subsidized vaccination for both animal science and veterinary students.

Reverse transcription polymerase chain reaction (RT-PCR) is currently the standard diagnostic method to detect symptomatic and asymptomatic individuals infected with Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). However, RT-PCR results are not immediate and may falsely be negative before an infected individual sheds viral particles in the upper airways where swabs are collected. Infected individuals emit volatile organic compounds in their breath and sweat that are detectable by trained dogs. Here, we evaluate the diagnostic accuracy of dog detection against SARS-CoV-2 infection. Fifteen dogs previously trained at two centres in Australia were presented to axillary sweat specimens collected from known SARS-CoV-2 human cases (n = 100) and non-cases (n = 414). The true infection status of the cases and non-cases were confirmed based on RT-PCR results as well as clinical presentation. Across dogs, the overall diagnostic sensitivity (DSe) was 95.3% (95%CI: 93.1-97.6%) and diagnostic specificity (DSp) was 97.1% (95%CI: 90.7-100.0%). The DSp decreased significantly when non-case specimens were collected over 1 min rather than 20 min (p value = .004). The location of evaluation did not impact the detection performances. The accuracy of detection varied across dogs and experienced dogs revealed a marginally better DSp (p value = .016). The potential and limitations of this alternative detection tool are discussed.

In 2013, Brucella melitensis biovar 1 was recovered from the stomach contents of a scimitar-horned Oryx - SHO (Oryx dammah) aborted foetus, and from the articular fluid of a sand gazelle (Gazella marica) in a captive wildlife collection near Abu Dhabi, United Arab Emirates. Other evidence of exposure to the pathogen was collected through serological testing (Rose Bengal test) and B. melitensis-specific PCR of samples from captive wildlife kept in six different enclosures. A Multiple Locus Variable Number of Tandem Repeats (VNTR) Analysis (MLVA) using 15 markers showed that the two strains isolated in animals kept in enclosures, located 1300 m apart from each other, shared an identical genotype. The phylogenetic analysis of MLVA-15 profiles retrieved from the public database suggested that these strains belong to the African clade, clustering regionally in the UAE, Oman and Qatar. This is the first confirmed case of B. melitensis in a SHO, an African antelope extinct in the wild and warrants further investigation.

To explore animal science and veterinary students' and livestock farmers' perceptions concerning Q fever prevention.An online survey with an open-ended question seeking knowledge and perceptions about Q fever prevention was distributed among participants during March-September 2019. We applied thematic analysis to identify emerging themes.Animal science and veterinary students enrolled at the University of Adelaide and members of Livestock South Australia representing cattle, sheep and goat farmers in South Australia.A total of56 animal science and veterinary students and 154 livestock farmers responded to the open-ended question.Not applicable.Perceived challenges and opportunities for a coordinated Q fever prevention approach including human vaccination reported by the participants.Two major themes arose in each group. Students and farmers viewed Q fever vaccination as important. However, excessive cost for students was a barrier and for farmers, it was general practitioners' lack of knowledge of Q fever and access to an accredited immunisation provider. Similarly, both groups highlighted the need for education and increasing public and community awareness of Q fever.Our findings underscore that a sector-wide approach involving community awareness programmes, education and training for general practitioners, and subsidised vaccination as well as commitment from government and industry partners may contribute to reducing the burden of Q fever among at-risk populations.

Simple Summary Streptococcus iniae (S. iniae) is a significant aquatic bacterial pathogen that has caused devastating losses to wild and cultured fish populations worldwide and is an important zoonotic pathogen in humans. Previously reported in captive dolphins only, this case report describes S. iniae associated infection and mortality in a free-ranging short-beaked common dolphin (Delphinus delphis). Unreported previously in wild marine mammals, its detection highlights a need for further investigation into the epidemiology of S. iniae to better understand the risks for wild marine mammal populations and potential transmission cycles between marine mammals, humans and wild and intensive fish populations. Abstract Streptococcus iniae (S. iniae) is a significant aquatic pathogen of farmed fish species, important zoonotic pathogen, and reported cause of disease in captive Amazon River dolphins (Inia geoffrensis) and a bottlenose dolphin (Tursiops truncatus). Here we report S. iniae as the cause of subcutaneous abscesses, sepsis and mortality in a juvenile free-ranging short-beaked common dolphin (Delphinus delphis) found deceased on a metropolitan Australian beach. Body surfaces were covered by multifocal, depressed, deep, irregular cutaneous ulcerations, which microscopically were characterised by ruptured subcutaneous abscesses with intralesional cocci. Routine microbiological investigations revealed a heavy growth of beta-haemolytic Streptococcus sp. identified as Streptococcus iniae in skin lesions as well as from heart blood, the latter supportive of sepsis. Tissues were negative for cetacean morbillivirus and no other disease processes were identified. S. iniae has not been reported in free-ranging marine mammals, nor in Australian delphinids, previously. More notably a pathogen of captive animals, this case report identifies S. iniae as a pathogen of wild dolphins also. In addition to expanding the host reservoir of a significant zoonotic pathogen, determining the source of infection as well as possible consequences for other marine mammals and wild and intensive fish stocks warrants further investigations.

Background/Objective To identify the issues around the prevention of Q fever as understood by trainee animal health practitioners and livestock farmers and to compare their varying perceptions. Methods An online survey with an open question seeking participants’ suggestions about Q fever prevention was distributed among all animal science and veterinary students enrolled at the University of Adelaide and members of Livestock SA representing livestock farmers during March-September 2019. Students were recruited through their University course whilst livestock farmers were recruited by advertisements and direct emails. Thematic analysis of the data followed the Braun and Clarke framework. Results Two major themes arose in each group, whilst each group identified similar themes and sub themes, there was a differing emphasis on the importance of these. Both students and farmers viewed Q fever vaccination is important, but barriers included excessive cost for students while access to a vaccine provider and general practitioners’ inadequate knowledge were emphasized by farmers. Awareness was another major theme: students – generalized lack of awareness amongst themselves and the public; farmers – increasing public awareness is crucial. Conclusions Both groups recognized the importance of Q fever vaccination and viewed subsidy an enabler, but students’ perceptions were shaped by the high cost of the vaccine while farmers were most concerned with the impacts of the disease. Findings underscore that strengthening targeted and community awareness programs coupled with a subsidy could promote increased uptake of Q fever vaccination. Key messages Increasing awareness and subsidized vaccination could merit Q fever prevention approaches including vaccination.

To examine stakeholder perspectives on the factors of an effective approach to reduce Q fever risk including disease prevention, and the perceived potential benefits of a One Health framework.Semi-structured interviews were conducted with general practitioners (GPs), veterinarians, government authorities, researchers, and representatives from the farming industry. Transcripts were thematically analysed.Six major themes were identified as key factors underpinning an effective approach to Q fever: understanding Q fever burden; effective surveillance; the role of general practitioners and other stakeholders; barriers and enablers of vaccination; an integrated approach; and increased Q fever awareness. Most participants perceived GPs to play a central role in disease detection, notification, treatment and prevention through health promotion and vaccination, despite GPs acknowledging limited awareness of Q fever. Participants suggested leadership is required from the Department of Health (DoH) to foster inter-sectoral communication and collaboration.A One Health approach holds opportunities for zoonosis prevention. We recommend that medical curricula and professional development be enhanced, zoonosis working group networks strengthened, government-industry partnerships established, and relevant stakeholders included within an integrated program.Updating medical curricula, GP professional development programs and inter-sectoral collaboration led by health departments may reduce Q fever burden.

This report describes two cases of occupational exposure to Chlamydia psittaci following dissection of an infected Rosella (Platycercus elegans). The C. psittaci infections (with one of them resulting in diagnosed pneumonia and hospitalisation) were undiagnosed during routine medical investigations but later established due to epidemiological and clinical evidence, and molecular testing of the archived Rosella’ specimens. This case report stresses the importance of correct application and interpretation of diagnostic tests and the need to raise awareness about this zoonotic pathogen among medical practitioners and people exposed to potential animal carriers. Our findings suggest other infected individuals might be misdiagnosed and that C. psittaci (psittacosis) is likely to be underreported in Australia. This case highlights the need to operationalise the One Health concept. We call for improved communication between human and animal health service providers to allow accurate and rapid diagnosis of this zoonotic disease and raised awareness among medical practitioners. Further targeted surveys of wild birds (and other animals) should be conducted to improve assessment of risks to the general population and people working with or exposed to wild birds.

Little is known about the global bat souvenir trade despite previous research efforts into bat harvest for bushmeat. We screened eBay listings of bats in Australia, Canada, Italy, Switzerland, United Kingdom and USA to assess the nature and extent of the online offers. A total of 237 listings were retrieved in between the 11th and 25th of May 2020 with a median price per item of US$38.50 (range: US$8.50–2,500.00). Items on offer were mostly taxidermy (61.2%) or skull (21.1%) specimens. Overall, 32 different species of bat were advertised, most of which (n = 28) are listed as “Least Concern” on the International Union for Conservation of Nature (IUCN) Red List. One species (Nycteris javanica) is classified as “Vulnerable” and one (Eidolon helvum) as “Near Threatened.” Pteropus spp. specimens were the most expensive specimens on offer and the conservations status of these species may range from “Critically Endangered” to “Data Deficient” by IUCN and the entire genus is listed in the Appendix II by the Convention on the International Trade in Endangered Species of Wild Fauna and Flora (CITES). However, the exact species concerned, and their respective conservation status, could not be confirmed based on the listings' photos. The sourcing of bat was restricted to mostly South-East Asian countries (a third of items sourced from Indonesia) and to two African countries. Our survey revealed that the online offer of bat products is diverse, abundant, and facilitated by worldwide sellers although most offered bats species are from South-East Asia. With a few exceptions, the species on offer were of little present conservation concern, however, many unknowns remain on the potential animal welfare, biosecurity, legal implications, and most importantly public health risks associated with this dark trade.

Chlamydia psittaci is traditionally regarded as a globally distributed avian pathogen that can cause zoonotic spill-over. Molecular research has identified an extended global host range and significant genetic diversity. However, Australia has reported a reduced host range (avian, horse, and human) with a dominance of clonal strains, denoted ST24. To better understand the widespread of this strain type in Australia, multilocus sequence typing (MLST) and ompA genotyping were applied on samples from a range of hosts (avian, equine, marsupial, and bovine) from Australia. MLST confirms that clonal ST24 strains dominate infections of Australian psittacine and equine hosts (82/88, 93.18%). However, this study also found novel hosts (Australian white ibis, King parrots, racing pigeon, bovine, and a wallaby) and demonstrated that strain diversity does exist in Australia. The discovery of a C. psittaci novel strain (ST306) in a novel host, the Western brush wallaby, is the first detection in a marsupial. Analysis of the results of this study applied a multidisciplinary approach regarding Chlamydia infections, equine infectious disease, ecology, and One Health. Recommendations include an update for the descriptive framework of C. psittaci disease and cell biology work to inform pathogenicity and complement molecular epidemiology.

All species of hard ticks associated with reptiles as hosts throughout their life cycle, are currently assigned to genera including Amblyomma and Africaniella. Among these species, based on literature data, Africaniella transversale has never been investigated for the presence of tick-borne pathogens. In this study, seven DNA extracts (two from A. transversale and five from Amblyomma exornatum) were screened for the presence of important tick-borne protozoa (piroplasms) and bacteria (Anaplasmataceae and Rickettsiaceae) with conventional PCRs and sequencing. A new heat shock protein chaperonin (groEL) gene-specific PCR was also developed to identify Occidentia spp. in these samples. In A. transversale, Occidentia massiliensis (previously detected in rodent-associated soft ticks) and Rickettsia hoogstraalii were present. While the latter was molecularly identical with formerly reported sequences of this rickettsia, the genotype of O. massiliensis was new based on sequence and phylogenetic analyses of its groEL gene. In A. exornatum, a Rickettsia genotype closely related to R. tamurae and R. monacensis, was detected. The ompA sequence of this genotype was identical to that of Rickettsia sp. Ae-8 reported from A. exornatum in a reptile breeding facility in the USA. These results show that A. transversale might carry O. massiliensis which (unless having a symbiotic nature in ticks) may originate either from the reptile host of this hard tick species or the rodent prey of reptiles. This is also the first detection of the reptile tick-associated Rickettsia sp. Ae-8 (phylogenetically aligning with R. tamurae, R. monacensis) in Africa, i.e. within the original geographical range of A. exornatum.

To determine Phascolarctid gammaherpesviruses (PhaHV) infection in South Australian koala populations, 80 oropharyngeal swabs from wild-caught and 87 oropharyngeal spleen samples and swabs from euthanased koalas were tested using two specific PCR assays developed to detect PhaHV-1 and PhaHV-2. In wild-caught koalas, active shedding of PhaHV was determined by positive oropharyngeal samples in 72.5% (58/80) of animals, of which 44.8% (26/58) had PhaHV-1, 20.7% (12/58) PhaHV-2 and 34.5% (20/58) both viral subtypes. In the euthanased koalas, systemic infection was determined by positive PCR in spleen samples and found in 72.4% (63/87) of koalas. Active shedding was determined by positive oropharyngeal results and found in 54.0% (47/87) of koalas. Koalas infected and actively shedding PhaHV-1 alone, PhaHV-2 alone or shedding both viral subtypes were 48.9% (23/47), 14.9% (7/47) and 36.2% (17/47), respectively. Only 45.9% (40/87) were not actively shedding, of which 40.0% (16/40) of these had systemic infections. Both wild-caught and euthanased koalas actively shedding PhaHV-2 were significantly more likely to be actively shedding both viral subtypes. Active shedding of PhaHV-2 had a significant negative correlation with BCS in the euthanased cohort, and active shedding of PhaHV-1 had a significant positive relationship with age in both wild-caught and euthanased cohorts.

Background Coxiella burnetii causes Q fever, a zoonotic bacterial disease with a multi-host cycle and reservoirs in wild and domestic animal species. Q fever has a significant impact on the Australian public health and economy but its ecology and contributing reservoir species remain poorly understood. In Europe, rabbits (Oryctolagus cuniculus) were identified as a major reservoir of C. burnetii and it is possible that they play a similar role in Australia. In absence of commercial kit available for rabbit, the Thermo Fisher - PrioCHECK™ Ruminant Q fever Ab Plate Kit was adapted to successfully screen rabbits population in Europe. However, this assay is not accessible in Australia and we assessed the equivalency of two commercially available kits in Australia – IDEXX - CHEKIT Q Fever Antibody ELISA kit and IDVet - ID Screen® Q Fever Indirect Multi-species with the Thermo Fisher kit (reference kit). Results A total of 94 rabbit sera were screened by all three ELISA kits using the same confirmed positive and negative controls. While the IDEXX kit failed to agree the other two assays (concordance correlation coefficient, rb rb = 0.927). Conclusion IDvet kit provides the best alternative for Thermo Fisher in the detection of C. burnetii specific antibodies in rabbits in Australia. Further trials are required to confirm these preliminary results due to the low seroprevalence of Coxiella burnetii observed in the study sera.

To investigate how such content is perceived by the global community, sentiment analysis techniques were employed to explore variations in attitudes expressed through text and emoji usage in public comments associated with 346 popular YouTube® videos starring exotic wild cats or primates in captive or domestic situations. Although a negative trend in mean text sentiment was observed in 2015 for primates, an otherwise consistent positive mean sentiment text and emoji score through time was revealed in response to both exotic wild cat and primate videos, implying the societal normalisation and acceptance of exotic pets. These findings highlight the urgency for effective YouTube® policy changes and content management to promote public education and conservation awareness, whilst extinguishing false legitimisation and potential demand for the exotic pet trade.

International audience; Contagious caprine pleuropneumonia (CCPP), caused by Mycoplasma capricolum subsp. capripneumoniae, has long been considered a goat-specific disease. Since 2007 there has been growing evidence that this disease can affect wild ungulates either kept in captivity or in the wild. In 2013, a large collection of sand gazelles (Gazella marica) held in the United Arab Emirates suffered heavy losses due to a CCPP epizootic confirmed by PCR and isolation. Animals displayed typical lesions, with unilateral pneumonia and profuse pleurisy. An initial antibiotic treatment consisting of tylosin administered in drinking water did not improve the animals' condition and vaccination failed to stop the spread to contiguous pens. A treatment with tetracycline mixed in feed pellets finally succeeded to stop the evolution of the disease. A subsequent vaccine trial, performed on naive animals, showed that only a reference CCPP vaccine produced according to OIE standards induced a sero-conversion by CCPP competition ELISA, while the commercially available vaccines did not. A SEIRD compartment transmission model was developed to better understand the dynamics of the disease. The parameters were initially set as per expert opinion and then adjusted to fit the observed mortality data. The basic reproductive number R-0 was estimated to be between 2.3-2.7, while the final mortality rate reached up to 70% in some pens. Transmission of infectious droplets from an external source, through a distance of at least the 50 m separating the pens from the perimeter fence, remains the most plausible explanation for the contamination of this stock of gazelles.

Simple Summary The exotic pet trade is impacted by social media via greater accessibility to photos and videos including these species and the increasing popularity of online animal marketplaces. The social media presence of public figures owning exotic pets has a major influence on exotic species demand. This study aimed to investigate exotic pet popularity, featured species on social media in the Middle East, and public perception of the animals spotlighted by public figures. We discuss the impact of such on the exotic pet trade and possible solutions to this issue. Male public figures from the United Arab Emirates predominated in the collected data, with most posts sourced from Instagram®. Eighty-five percent of the species displayed on social media posts were Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) Appendix I- and II-listed, including big cats, birds of prey, and great apes. Through an investigation of social media posts of public figures in the Middle East, we found that there was an overall positive audience reception toward endangered exotic pets. Geographic region, social media platform, animal species, and animal age all influenced the results. We recommend improving public education and awareness of wildlife conservation and laws regarding exotic pet possession to combat the idolization of the exotic pet industry. Abstract The popularization of exotic pets on celebrity social media in the Middle East has led to questionable impacts on exotic pet demand and threats to species conservation. The objective of this study was to identify exotic animal species featured on Middle Eastern celebrity social media account posts, the public perception of those posts, and their potential impacts on exotic pet demand and conservation (for global-scale extrapolation). Public social media accounts of highly influential persons from oil-rich Middle Eastern regions were manually investigated to evaluate subject demographics, species features, and post information (likes, comments) between January 2017 and August 2018. Twenty-five subjects possessed active social media accounts, from which 418 social media posts were extracted based on their inclusion of a privately owned exotic animal. SPSS Version 25 was used for frequency and descriptive analyses of these posts, in addition to comment analyses to evaluate quantitative (emojis) and qualitative (text) audience perceptions from a total of 10 social media posts of CITES Appendix I- or II-listed species. A greater frequency of positive than negative comments was observed (n = 8017), demonstrating the higher likelihood of social media promotion rather than negation of the exotic pet trade. Public education on wildlife conservation and exotic animal trade risks is imperative for successful conservation and welfare protection.
