Insects can serve as a novel high-quality protein source for pet foods. However, there is an absence of research investigating the use of insects in pet food. The study objective was to evaluate the apparent total tract digestibility and possible health effects of diets containing graded levels of cricket (Gryllodes sigillatus) meal fed to healthy adult dogs. Thirty-two adult Beagles were randomly assigned to one of four dietary treatments: 0%, 8%, 16%, or 24% cricket meal. Dogs were fed their respective diet for a total of 29 d with a 6-d collection phase. Fecal samples were collected daily during the collection phase to measure total fecal output as well as apparent total tract digestibility for dry matter (DM), organic matter, crude protein, fat, total dietary fiber, and gross energy. Blood samples were taken prior to the study and on day 29 for hematology and chemistry profiles. Data were analyzed in a mixed model including the fixed effects of diet and sex. Total fecal output increased on both an as-is (P = 0.030) and DM basis (P = 0.024). The apparent total tract digestibility of each nutrient decreased (P < 0.001) with the increasing level of cricket meal inclusion. All blood values remained within desired reference intervals indicating healthy dogs. Slight fluctuations in blood urea nitrogen (P = 0.037) and hemoglobin (P = 0.044) levels were observed but were not considered of biological significance. Even with the decrease in digestibility with the inclusion of cricket meal, diets remained highly digestible at greater than 80% total apparent digestibility. In conclusion, crickets were demonstrated to be an acceptable ingredient for dog diets.

The gut microbiome plays an important role in the health of dogs. Both beneficial microbes and overall diversity can be modulated by diet. Fermentable sources of fiber in particular often increase the abundance of beneficial microbes. House crickets (Acheta domesticus) contain the fermentable polysaccharides chitin and chitosan. In addition, crickets are an environmentally sustainable protein source. Considering crickets as a potential source of both novel protein and novel fiber for dogs, 4 diets ranging from 0% to 24% cricket content were fed to determine their effects on healthy dogs’ (n = 32) gut microbiomes. Fecal samples were collected serially at 0, 14, and 29 days, and processed using high-throughput sequencing of 16S rRNA gene PCR amplicons. Microbiomes were generally very similar across all diets at both the phylum and genus level, and alpha and beta diversities did not differ between the various diets at 29 days. A total of 12 ASVs (amplicon sequence variants) from nine genera significantly changed in abundance following the addition of cricket, often in a dose-response fashion with increasing amounts of cricket. A net increase was observed in Catenibacterium, Lachnospiraceae [Ruminococcus], and Faecalitalea, whereas Bacteroides, Faecalibacterium, Lachnospiracaeae NK4A136 group and others decreased in abundance. The changes in Catenibacterium and Bacteroides are predicted to be beneficial to gut health. However, the total magnitude of all changes was small and only a few specific taxa changed in abundance. Overall, we found that diets containing cricket supported the same level of gut microbiome diversity as a standard healthy balanced diet. These results support crickets as a potential healthy, novel food ingredient for dogs.

Eco-labeling (or environmental certification) is often promoted as a regulatory instrument capable of incentivizing sustainable resource use, even in the absence of stringent government environmental regulations. Despite slow uptake in developing countries and high producer costs, a growing body of case study evidence suggests that producers benefit in varied ways from certification. A qualitative meta-synthesis approach is applied to this body of evidence in order to assess the type and extent of producer benefits reported in case studies of Forest Stewardship Council (FSC) and Marine Stewardship Council (MSC) certification, in developing countries. While benefits from price premiums and market access appear to be limited, less tangible benefits were more common, including learning, governance, community empowerment, and reputational benefits. These benefits may justify the cost of certification.

The Internet has created new communication alternatives and challenges in public relations. Despite the increase in social media and multimedia in college sports public relations, no research has been conducted into which tools are utilized most, where those tools may be found on athletic websites, or how PR offices utilize such tools. This study intends to fill that gap. Grounded in a co-orientation model of communication, it utilized a content analysis of college athletic websites (N = 120) and a telephone survey with 16 college sports public relations professionals. Twitter, Facebook and RSS feeds are the social media tools utilized most often, while live video and audio and archived video are the most frequently used multimedia tools. Most tools are identified by a graphic in the right-hand column on departments' home pages. Tools are used primarily for disseminating information. Usually the director maintains the site alone without a published policy.

This article examines the history of fire protection in London from the Great Fire of 1666 to the present and argues that many of the popular perceptions of market failure in the provision of fire protection that are repeated in standard economics textbooks are in fact myths. In reality, fire protection began in the private sector and only transferred to the public sector when insurance companies faced the prospect of collecting escalating premiums from their clients. To this day, voluntary and private provision in parts of the USA is a model of how fire protection can be provided more cheaply and more effectively outside the public sector.

Tyrosine phosphorylation has emerged as a mechanism to control cellular events in the nucleus. The c-Fes protein-tyrosine kinase is an important regulator of cell growth and differentiation in several cell types, and is found in the nucleus of hematopoietic cells. In this study, we showed nuclear localization of c-Fes in both hematopoietic (K562, TF-1, HEL, U937, and HL-60) and nonhematopoietic cell lines (293T, CaOv3, TfxH, MG-63, HeLa, DU-145) by immunofluorescence and confocal microscopy. c-Fes showed striking changes in subcellular localization at specific stages of mitosis. In interphase cells, the intranuclear distribution of c-Fes was diffuse with occasional bright foci. Some c-Fes was present in the cytosol after breakdown of the nuclear membrane, in prometaphase. At prometaphase and metaphase c-Fes was also associated with the chromosomes, in a punctate pattern that partially overlapped with the centromere. Further comparison with proteins that are known components of the kinetochore suggested that some c-Fes protein was located at the centromeric alpha-satellite DNA, between the kinetochores. At anaphase and telophase, c-Fes was entirely cytoplasmic and no protein was found associated with the chromosomes. The timing of c-Fes' appearance at the centromere coincides with the period of kinetochore assembly. These data suggest that c-Fes is recruited to the kinetochore during mitosis.

The main objectives of the advanced undergraduate applied hydrogeochemistry course at Eastern Connecticut State University include (1) thinking scientifically, (2) doing science, and (3) writing scientifically, all in the context of hydrogeochemistry. To these ends, students read and give presentations on scientific-journal articles, work collaboratively on a semester-long field-research project, and write a final formal report. The class is involved in every step of the research project from project design, sample collection and analysis, and data interpretation, to final report preparation. The course is process-oriented rather than task-oriented in an effort to concentrate on developing critical-thinking skills as opposed to learning “cookbook” techniques. Students who complete the course successfully take with them basic hydrogeochemical knowledge, collaborative research experience, and a report that they can show to employers or graduate schools as evidence of their writing ability. Cellular retinaldehyde-binding protein (CRALBP) is abundant in the retinal pigment epithelium (RPE) and Müller cells of the retina where it is thought to function in retinoid metabolism and visual pigment regeneration. The protein carries 11-cis-retinal and/or 11-cis-retinol as endogenous ligands in the RPE and retina and mutations in human CRALBP that destroy retinoid binding functionality have been linked to autosomal recessive retinitis pigmentosa. CRALBP is also present in brain without endogenous retinoids, suggesting other ligands and physiological roles exist for the protein. Human recombinant cellular retinaldehyde-binding protein (rCRALBP) has been over expressed as non-fusion and fusion proteins in Escherichia coli from pET3a and pET19b vectors, respectively. The recombinant proteins typically constitute 15-20% of the soluble bacterial lysate protein and after purification, yield about 3-8 mg per liter of bacterial culture. Liquid chromatography electrospray mass spectrometry, amino acid analysis, and Edman degradation were used to demonstrate that rCRALBP exhibits the correct primary structure and mass. Circular dichroism, retinoid HPLC, UV-visible absorption spectroscopy, and solution state 19F-NMR were used to characterize the secondary structure and retinoid binding properties of rCRALBP. Human rCRALBP appears virtually identical to bovine retinal CRALBP in terms of secondary structure, thermal stability, and stereoselective retinoid-binding properties. Ligand-dependent conformational changes appear to influence a newly detected difference in the bathochromic shift exhibited by bovine and human CRALBP when complexed with 9-cis-retinal. These recombinant preparations provide valid models for human CRALBP structure-function studies.

Purpose: The authors determine if specific features of the clinical presentation of acute postoperative endophthalmitis correlate with the microbiologic culture results. Methods: A total of 420 patients who had clinical evidence of endophthalmitis within 6 weeks after cataract surgery or secondary intraocular lens implantation were evaluated as part of a randomized clinical trial. Results of cultures performed on aqueous and vitreous specimens obtained at presentation were categorized as follows: gram-positive coagulase-negative micrococci, "other" gram-positive, gram-negative, and equivocal/no growth. Results: Eleven features of the initial clinical presentation were associated with significant differences in the microbiologic spectrum ( P Conclusions: The presenting characteristics of acute endophthalmitis after cataract surgery may be helpful in predicting the most likely culture results. Such predictions do not appear sufficiently strong to guide the initial empiric choice of intravitreal antibiotics.

The cellular fes gene encodes a 93-kilodalton protein-tyrosine kinase (p93) that is expressed in both normal and neoplastic myeloid cells. Increased c-Fes expression is associated with differentiation in normal myeloid cells and cell lines. Our hypothesis was that primary leukemia cells would show a similar pattern of increased expression in more differentiated cells. Therefore, we compared c-Fes expression in cells with an undifferentiated, blast phenotype (acute myelogenous leukemia--AML) to cells with a differentiated phenotype (chronic myelogenous leukemia--CML). Instead of differences in p93 expression levels, we found complex patterns of c-Fes immunoreactive proteins that corresponded with differentiation in normal and leukemic myeloid cells. The "blast" pattern consisted of c-Fes immunoreactive proteins p93, p74, and p70; the "differentiated" pattern showed two additional c-Fes immunoreactive proteins, p67 and p62. Using mRNA from mouse and human cell lines, we found deletion of one or more exons in the c-fes mRNA. Those deletions predicted truncation of conserved domains (CDC15/FCH and SH2) involved in protein-protein interactions. No deletions were found, however, within the kinase domain. We infer that alternative splicing generates a family of c-Fes proteins. This may be a mechanism to direct the c-Fes kinase domain to different subcellular locations and/or substrates at specific stages of myeloid cell differentiation.

Members of the Notch family of transmembrane receptors are found on primitive hematopoietic precursors, and Notch ligand expression has been demonstrated on the surface of stromal cells, suggesting a role for Notch signaling in mammalian blood cell development. The current report examines the expression of Notch receptors and their ligands in murine hematopoietic tissues to determine: A) which blood cell lineages in the adult are influenced by Notch activity, and B) whether fetal hematopoiesis in the embryo involves the Notch pathway. In the adult mouse, a combination of flow cytometry, immunohistochemistry and Northern analysis was used to examine Notch receptor or ligand expression in bone marrow and spleen. In the embryo, Northern analysis and in situ hybridization were used to characterize Notch receptor and ligand expression in fetal liver on embryonic day 12 (E12) through E17, an active period encompassing both erythropoiesis and granulopoeisis. Flow cytometry demonstrated the presence of Notch1 and Notch2 receptors on bone marrow-derived myeloid cells but not on erythroid cells positive for the marker, Ter-119. In situ hybridization of E12 through E17 fetal liver demonstrated widespread expression of Jagged1 and Delta1 in a pattern similar to but less abundant than that of the erythropoietin receptor. Taken together with earlier functional results, the current expression data suggest a role for Notch activity in establishing definitive hematopoiesis in fetal liver, as well as a selective use of Notch signaling in adult erythropoiesis and granulopoiesis. Notch receptors in the adult are most likely utilized by early erythroid precursors and intermediate-stage granulocytes, but not by terminally differentiating cells of either subset.
