ZusammenfassungEine weltweite Finanz- und Wirtschaftskrise gegen Ende der 2000er Jahre, wenige Jahre später die Corona-Pandemie und aktuell kaum zu kalkulierende Auswirkungen eines Krieges auf dem europäischen Kontinent: drei markante Beispiele für externe Schocks, die auf Unternehmen und Wirtschaft einwirken und denen sie standhalten müssen. Da ist es hilfreich, einen Blick auf Theorien, Modelle und Operationalisierungen von Resilienz zu werfen – sowohl aus arbeits- und organisationssoziologischer als auch aus betriebswirtschaftlicher und innovationspolitischer Perspektive.

Background We benchmarked sequencing technology and assembly strategies for short-read, long-read, and hybrid assemblers in respect to correctness, contiguity, and completeness of assemblies in genomes of Francisella tularensis. Benchmarking allowed in-depth analyses of genomic structures of the Francisella pathogenicity islands and insertion sequences. Five major high-throughput sequencing technologies were applied, including next-generation “short-read” and third-generation “long-read” sequencing methods. Results We focused on short-read assemblers, hybrid assemblers, and analysis of the genomic structure with particular emphasis on insertion sequences and the Francisella pathogenicity island. The A5-miseq pipeline performed best for MiSeq data, Mira for Ion Torrent data, and ABySS for HiSeq data from eight short-read assembly methods. Two approaches were applied to benchmark long-read and hybrid assembly strategies: long-read-first assembly followed by correction with short reads (Canu/Pilon, Flye/Pilon) and short-read-first assembly along with scaffolding based on long reads (Unicyler, SPAdes). Hybrid assembly can resolve large repetitive regions best with a “long-read first” approach. Conclusions Genomic structures of the Francisella pathogenicity islands frequently showed misassembly. Insertion sequences (IS) could be used to perform an evolutionary conservation analysis. A phylogenetic structure of insertion sequences and the evolution within the clades elucidated the clade structure of the highly conservative F. tularensis.

We report the first draft genome sequence of a Salmonella strain with plasmid-mediated resistance to colistin encoded by mcr-1 gene in Vietnam. Salmonella enterica subsp. enterica serovar Rissen was isolated from a Vietnamese pig slaughterhouse in 2013. We can confirm that mcr-1 gene is identical to the first reported mcr-1 gene of the Escherichia coli strain SHP45, isolated in 2015 from a Chinese pig. The plasmid containing this gene in the strain 19CS0416 was highly related (96.86% identity) to the plasmid (pHNSHP45) contained in this Chinese strain. Moreover, this plasmid was determined to be 100% identical to a plasmid (p13P477T-7) belonging to an Escherichia coli (13P477T) found in Hong Kong during the same year in pigs. Our results will aid in understanding the dissemination of mcr-1 gene in East Asia, dating back to as early as 2013.

Here, we report the draft genome sequence of Klebsiella grimontii AT013-Mero-001, which was isolated from feces from a sepsis patient treated with meropenem. This isolate is an antibiotic-susceptible but persistent Enterobacteriaceae strain.

Corynebacterium silvaticum is a newly identified animal pathogen of forest animals such as roe deer and wild boars. The species is closely related to the emerging human pathogen Corynebacterium ulcerans and the widely distributed animal pathogen Corynebacterium pseudotuberculosis. In this study, Corynebacterium silvaticum strain W25 was characterized with respect to its interaction with human cell lines. Microscopy, measurement of transepithelial electric resistance and cytotoxicity assays revealed detrimental effects of C. silvaticum to different human epithelial cell lines and to an invertebrate animal model, Galleria mellonella larvae, comparable to diphtheria toxin-secreting C. ulcerans. Furthermore, the results obtained may indicate a considerable zoonotic potential of this newly identified species.

Foodborne campylobacteriosis is the most common cause of human bacterial enteritis in Germany. Campylobacter jejuni and Campylobacter coli are the main causative agents for enteric disease, but a number of other species are involved, including rare ones. These rare Campylobacter spp. are emerging zoonotic pathogens in humans due to increasing international movement of supplies, livestock and people. Campylobacter lanienae was first isolated from healthy abattoir workers in Switzerland and at first its pathogenic potential for humans was considered to be low. Recently, the first case of Campylobacter lanienae -associated human enteritis was reported in Canada. Here, we describe a case of mild Campylobacter lanienae -associated enteritis with subsequent asymptomatic excretion in a butcher. The isolate is available at the TLV strain collection (no. TP00333/18). This first reported case of human Campylobacter lanienae campylobacteriosis in Germany demonstrates the agent’s likely zoonotic pathogenicity.

Clostridium perfringens causes a plethora of devastating infections, with toxin production being the underlying mechanism of pathogenicity in various hosts. Genomic analyses of 206 public-available C. perfringens strains´ sequence data identified a substantial degree of genomic variability in respect to episome content, chromosome size and mobile elements. However, the position and order of the local collinear blocks on the chromosome showed a considerable degree of preservation. The strains were divided into five stable phylogroups (I–V). Phylogroup I contained human food poisoning strains with chromosomal enterotoxin (cpe) and a Darmbrand strain characterized by a high frequency of mobile elements, a relatively small genome size and a marked loss of chromosomal genes, including loss of genes encoding virulence traits. These features might correspond to the adaptation of these strains to a particular habitat, causing human foodborne illnesses. This contrasts strains that belong to phylogroup II where the genome size points to the acquisition of genetic material. Most strains of phylogroup II have been isolated from enteric lesions in horses and dogs. Phylogroups III, IV and V are heterogeneous groups containing a variety of different strains, with phylogroup III being the most abundant (65.5%). In conclusion, C. perfringens displays five stable phylogroups reflecting different disease involvements, prompting further studies on the evolution of this highly important pathogen.

We describe a rare isolate of Salmonella enterica subsp. enterica serovar Panama with an extended-spectrum β-lactamase (ESBL) profile from a German cattle-fattening farm. Applying two next-generation sequencing methods we generated sequences of the genome as well as the plasmids; assembled the draft genome sequence of Salmonella enterica subsp. enterica serovar Panama isolate 18PM0209. Antimicrobial resistance genes, virulence-associated genes and plasmids were analyzed using bioinformatics. Occurrence of multidrug-resistant Salmonella serovars at cattle-fattening farms indicate the need of enhanced surveillance to prevent further spread of these organisms.

Zusammenfassung Auf dem Arbeitsmarkt existieren nach wie vor bestimmte soziale Ungleichheiten zwischen Frauen und Männern, etwa im Hinblick auf die Verortung in unterschiedlichen Berufen und hierarchischen Positionen sowie Verdienste. Andockend an die bisherige Forschung, fragen wir nach der Rolle von Betrieben bei der Generierung geschlechtsspezifischer Arbeitsmarktchancen. Bei dieser Forschungsfrage betrachten wir zum einen „egalitäre“ betriebliche Strukturen (Maßnahmen zur Geschlechtergleichstellung und zur Vereinbarkeit von Familie und Beruf, eine paritätische Geschlechterdemografie im Betrieb und in dessen Leitungspositionen, betriebliche Formalisierung der Beschäftigungspolitik) und ihren Einfluss auf geschlechterbezogene soziale Ungleichheiten. Zum anderen wird, unter Bezugnahme auf den Neo-Institutionalismus, diskutiert, inwieweit sich jene „egalitären“ Strukturen in der Betriebskultur bzw. in der subjektiven Wahrnehmung der Strukturen niederschlagen. Ein besonderes Ziel liegt darin, mögliche Ambivalenzen zwischen positiven und negativen Auswirkungen „egalitärer“ Betriebsstrukturen aufzudecken. Aufbauend auf diese Überlegungen wird auf den weiteren Forschungsstand aufmerksam gemacht, mit dem es möglich ist, eine solche Verschränkung von „Struktur“ und „Kultur“ und ihre Auswirkungen auf geschlechterbezogene soziale Ungleichheiten auf dem Arbeitsmarkt zu untersuchen.

We here report a family from Libya with three siblings suffering from early onset achalasia born to healthy parents. We analyzed roughly 5000 disease-associated genes by a next-generation sequencing (NGS) approach. In the analyzed sibling we identified two heterozygous variants in CRLF1 (cytokine receptor-like factor 1). Mutations in CRLF1 have been associated with autosomal recessive Crisponi or cold-induced sweating syndrome type 1 (CS/CISS1), which among other symptoms also manifests with early onset feeding difficulties. Segregation analysis revealed compound heterozygosity for all affected siblings, while the unaffected mother carried the c.713dupC (p.Pro239Alafs*91) and the unaffected father carried the c.178T>A (p.Cys60Ser) variant. The c.713dupC variant has already been reported in affected CS/CISS1 patients, the pathogenicity of the c.178T>A variant was unclear. As reported previously for pathogenic CRLF1 variants, cytokine receptor-like factor 1 protein secretion from cells transfected with the c.178T>A variant was severely impaired. From these results we conclude that one should consider a CRLF1-related disorder in early onset achalasia even if other CS/CISS1 related symptoms are missing.

Zoonotic pathogens that can be transmitted via food to humans have a high potential for large-scale emergencies, comprising severe effects on public health, critical infrastructures, and the economy. In this context, the development of laboratory methods to rapidly detect zoonotic bacteria in the food supply chain, including high-resolution mass spectrometry proteotyping are needed. In this work, an optimized sample preparation method for liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteome profiling was established for Francisella isolates, and a cluster analysis, as well as a phylogenetic tree, was generated to shed light on evolutionary relationships. Furthermore, this method was applied to tissues of infected hare carcasses from Germany. Even though the non-informative data outnumbered by a manifold the information of the zoonotic pathogen in the resulting proteome profiles, the standardized evaluation of MS data within an established automated analysis pipeline identified Francisella (F.) tularensis and, thus, could be, in principle, an applicable method to monitor food supply chains.

Objectives Corynebacterium ulcerans can colonize a wide variety of animals and also humans are infected, typically by zoonotic transmission. Symptoms range from skin ulcers or systemic infections to diphtheria-like illness. In contrast, Corynebacterium pseudotuberculosis is widely distributed among herds of sheep, goats and other farm animals, where it causes high economic losses due to caseous lymphadenitis. Here we describe the genome sequence of an atypical C. ulcerans strain isolated from a wild boar with necrotizing lymphadenitis. This strain has similarities to C. pseudotuberculosis. Data description Genome sequence data of C. ulcerans isolate W25 were generated, analyzed and taxonomical relationship to other Corynebacterium species as well as growth properties of the isolate were characterized. The genome of C. ulcerans W25 comprises 2,550,924 bp with a G+C content of 54.41% and a total of 2376 genes.

Campylobacter spp. are considered the most common bacterial cause of foodborne gastroenteritis in the world. The family Campylobacteraceae includes the genus Arcobacter with the three species Arcobacter butzleri, Arcobacter cryaerophilus, and Arcobacter skirrowii as emergent enteropathogens and potential zoonotic agents. Here, we characterized genome sequences of Arcobacter that were isolated from water poultry on farms in Germany. Isolates were cultured, identified by MALDI-TOF MS and identification was verified with PCR assays. Antibiotic susceptibility testing of isolates was carried out with erythromycin, ciprofloxacin, doxycycline, tetracycline, gentamicin, and streptomycin using the gradient strip method (E-test). We also sequenced whole genomes and predicted antibiotic resistance determinants, virulence factors, performed a phylogenetic analysis to determine the genetic relatedness of these isolates and searched for plasmids.

Francisella (F.) tularensis is a highly virulent, Gram-negative bacterial pathogen and the causative agent of the zoonotic disease tularemia. Here, we generated, analyzed and characterized a high quality circular genome sequence of the F. tularensis subsp. holarctica strain 12T0050 that caused fatal tularemia in a hare. Besides the genomic structure, we focused on the analysis of oriC, unique to the Francisella genus and regulating replication in and outside hosts and the first report on genomic DNA methylation of a Francisella strain. The high quality genome was used to establish and evaluate a diagnostic whole genome sequencing pipeline. A genotyping strategy for F. tularensis was developed using various bioinformatics tools for genotyping. Additionally, whole genome sequences of F. tularensis subsp. holarctica isolates isolated in the years 2008–2015 in Germany were generated. A phylogenetic analysis allowed to determine the genetic relatedness of these isolates and confirmed the highly conserved nature of F. tularensis subsp. holarctica.

Der Beitrag untersucht fur Zweiverdiener-Paare in Deutschland, welche Rolle die Erwerbstatigkeit in typischen „Frauenberufen“ und „Mannerberufen“ fur die Hausarbeitsteilung spielt. Fur die Erklarung eines solchen Zusammenhangs kontrastieren wir Argumente aus ressourcenbasierten Ansatzen und der Geschlechterrollensozialisation mit Prozessen im Rahmen von „Doing Gender“ und der Kompensationshypothese. Die Analysen mit Daten des Sozio-oekonomischen Panels (1993–2011) zeigen, dass Frauen, die eine geschlechts-untypische Berufstatigkeit ausuben, eine solche Abweichung von traditionellen Geschlechterrollen offenbar durch eine verstarkte Ubernahme „weiblich konnotierter“ Haushaltstatigkeiten (wie Waschen, Kochen, Putzen) kompensieren. Manner kompensieren eine entsprechende Abweichung demgegenuber eher durch eine verstarkte Ubernahme „mannlich konnotierter“ Aufgaben im Haushalt (wie Reparaturarbeiten). Letztgenannte Hausarbeiten werden generell dann besonders selten ausgeubt, wenn einer der Partner in einem geschlechtlich ausbalancierten Beruf tatig ist. Abweichungen von traditionellen Geschlechternormen auf dem Arbeitsmarkt scheinen sich daher sowohl fur Frauen als auch fur Manner auszuwirken und werden vor allem anhand von Hausarbeiten ausgeglichen, die traditionell in ihren jeweiligen Aufgabenbereich fallen.

"Die Studie untersucht mit Daten des Sozio-oekonomischen Panels (SOEP) und der fur die Welle 2011 durchgefuhrten Betriebsbefragung (SOEP-LEE) die Wirkungen auf Chancengleichheit orientierter betrieblicher Strukturen fur die Verdienste von Frauen und Mannern. Dabei werden in dem Beitrag solche betrieblichen Masnahmen fokussiert, von denen angenommen werden kann, dass sie einen egalisierenden Effekt auf geschlechtsspezifische Verdienstunterschiede haben. Die Analyse differenziert zwischen verbindlichen und weniger verbindlichen Formalisierungsmasnahmen sowie Masnahmen zur Vereinbarkeit von Familie und Beruf. Die Ergebnisse deuten an, dass die durchschnittlichen Verdienste der Befragten mit verbindlichen Formalisierungsmasnahmen in den Betrieben steigen, wobei dies fur Arbeitnehmerinnen in starkerem Mase gilt. Im Hinblick auf weniger verbindliche Formalisierungsmasnahmen zeigt sich hingegen kontrolliert um Drittvariablen kein Effekt. Bezuglich der Masnahmen zur Vereinbarkeit von Familie und Beruf werden bei dem Vorliegen von Masnahmen zur Arbeits(zeit)flexibilisierung vergleichsweise grose Egalisierungschancen deutlich, wahrend die Ergebnisse andererseits auch uberraschende Befunde zutage fordern, z. B., dass Manner von betrieblichen Masnahmen der Kinderbetreuung in starkerem Mase als Frauen profitieren. Die Ergebnisse werden unter anderem vor dem Hintergrund zu Annahmen des „Flexibility Stigmas“ sowie zu neo-institutionalistischen Arbeiten diskutiert. "

In humans, stress can be contagiously transmitted via chemosignals on a subconscious level. This study investigates how pregnancy affects neural responses to anxiety chemosignals. Using cotton pads, 28 men donated axillary sweat immediately before an academic examination (anxiety sweat) and during ergometer training (control). Via a constant-flow olfactometer, samples were presented (oddball paradigm) to 12 non-pregnant (NP) women, 14 women in their first (T1), and 18 in their third (T3) trimester of pregnancy. Chemosensory event-related potentials and current source densities (CSD) were analysed (60 electrode setup). Compared to NP-women, pregnant women display diminished evaluative processing of the sweat samples (targets; P3-1/ P3-2 amplitudes) and delayed evaluative processing of the anxiety sweat (targets; P3-2 latency). T3-women show attenuated early processing (targets; N1 amplitude) compared to NP-women, and reduced evaluative processing compared to T1-women (standards; P3-2 amplitude). CSDs (P3-1/ P3-2 latency ranges) reveal that T1- and T3-women show an atypical activation distribution to anxiety sweat. Most participants were unable to detect the sweat samples (anxiety sweat: 79.5%, sport sweat 88.6%). The results demonstrate that the processing of anxiety chemosignals progressively vanishes during pregnancy. This effect is likely to occur without any cognitive control.

Die wirtschaftlichen Folgen einer Scheidung stellen eine gravierende Belastung fur die Betroffenen dar, wie bisherige Forschungsergebnisse gezeigt haben. Eine Scheidung kann haufig einen wirtschaftlichen Abstieg verursachen, der im Extremfall sogar zu Armut fuhrt. Gleichzeitig ist auf dem Arbeitsmarkt ein Wandel in Richtung einer zunehmenden Arbeitsmarktflexibilisierung und Atypik von Arbeitsverhaltnissen zu beobachten, der neben einer objektiv, materiellen Prekarisierung zunehmend auch zu einer subjektiv, gefuhlten Prekarisierung im Sinne von Abstiegsangsten vermeintlich gesicherter Erwerbstatiger fuhrt. Wir untersuchen mit Daten des SOEPs, inwiefern sich die wirtschaftliche Lage von Alleinerziehenden infolge einer Trennung verschlechtert, im Hinblick einer objektiven Verschlechterung (Armutsrisiko) einerseits und einer subjektiv wahrgenommenen Verschlechterung (Sorgen um die wirtschaftliche Situation) andererseits. Darauf aufbauend wird betrachtet, inwiefern eine solche Entwicklung (auch) darauf zuruckzufuhren ist, dass die Alleinerziehenden nach der Trennung haufig atypische oder prekare Tatigkeiten ausuben. Vergleiche zwischen Frauen und Mannern mit und ohne Kindern werden fur drei Zeitpunkte vor und nach der Trennung durchgefuhrt. Es zeigt sich, dass sowohl alleinerziehende Mutter, aber auch alleinerziehende Vater eine Risikogruppe darstellen. Besonderes bei alleinerziehenden Mannern nehmen die Sorgen um die eigene wirtschaftliche Situation zu. Die Erwerbswege von alleinerziehenden Vatern fuhren mit der Trennung vergleichsweise haufiger als bei alleinerziehenden Muttern in atypische Beschaftigungen, jedoch nicht parallel dazu in Beschaftigungen mit Niedriglohnerwerb. Multivariate Modelle zeigen, dass die zunehmende Armutsquote infolge einer Trennung bei Frauen auf die Ausubung atypischer und prekarer Beschaftigungen zuruckzufuhren ist, wahrend dies bei Mannern nicht der Fall ist. Offenbar sind die von den Mannern ausgeubten atypischen Beschaftigungen nicht zwangslaufig mit okonomischen Einbusen verbunden.

During a routine health check of 12 breeding dromedary bulls using microscopy two of them were found to harbour trichomonad flagellates in their preputial washings. A PCR conducted on one of the samples did confirm trichomonads but not Tritrichomonas foetus. DNA sequencing identified the organism as Tetratrichomonas species. After repeated careful washings of the prepuce with oxidised water resampling revealed negative results.

Zusammenfassung Der Beitrag vergleicht den Effekt der Geschlechterzusammensetzung im Beruf auf die Verdienstunterschiede von Frauen und Männern zwischen Führungspositionen und Nicht-Führungspositionen in der Privatwirtschaft auf der Basis von Daten des Sozio-oekonomischen Panels (SOEP). Anhand gängiger theoretischer Ansätze wird die Hypothese entwickelt, dass in Führungspositionen die Qualifikationsanforderungen und Arbeitsinhalte der (wenigen) Frauenberufe jenen in Männerberufen ähnlicher sind als unter den sonstigen Angestellten. Entsprechend niedriger ist die Wahrscheinlichkeit von Verdienstabschlägen typischer Frauenberufe in Führungspositionen. Die Verdienstschätzungen unter Anwendung linearer Fixed-Effects-Modelle mit Heckman-Korrektur verdeutlichen, dass nur bei weiblichen Nicht-Führungskräften ein Verdienstabschlag für Frauenberufe besteht. Eine Komponentenzerlegung der Verdienstlücke zwischen Frauen und Männern („gender pay gap“) mit verschiedenen Dekompositionsverfahren zeigt darüber hinaus, dass die Geschlechtstypik des ausgeübten Berufes für Nicht-Führungskräfte eine bedeutsame Erklärungsgröße für den „gender pay gap“ darstellt. Für Führungskräfte sind solche Effekte nicht nachweisbar.

Als bedeutsame Erklarungsgrose fur geschlechtsspezifische Verdienstunterschiede („gender pay gap“) gilt die berufliche Geschlechtersegregation. Die vorliegende Studie untersucht daher Faktoren, die einer geringeren Entlohnung typischer Frauenberufe in Deutschland zugrunde liegen. Ausgehend von soziologischen und sozialpsychologischen Statustheorien ist anzunehmen, dass Frauenberufe aufgrund dort vorherrschender weiblich konnotierter Arbeitsinhalte, die auf dem Arbeitsmarkt abgewertet sind, geringer entlohnt werden – wahrend typisch mannlich konnotierte Arbeitsinhalte, die in Mannerberufen dominieren, eine monetare Aufwertung erfahren. Die Hypothesen werden mit Daten des Sozio-oekonomischen Panels (SOEP) 2000–2010 unter Anwendung linearer Fixed-Effects-Modelle untersucht. Berufliche Charakteristika, etwa zu den Arbeitsinhalten, werden von der BIBB/BAuA-Erwerbstatigenbefragung 2005/2006 und dem Mikrozensus dem SOEP zugespielt. Die Analyse bestatigt den vermittelnden Effekt geschlechtlich konnotierter Arbeitsinhalte auf Verdienstabschlage in den Bruttostundenverdiensten von Erwerbstatigen in Frauenberufen – jedoch nur fur Manner. Dieser geschlechtsspezifische Effekt wird mit unterschiedlichen Kompetenz- und Leistungserwartungen, die Frauen und Mannern fur geschlechtlich konnotierte Arbeitsinhalte entgegengebracht werden, verdichtet. Bei Frauen weisen vor allem geringere berufliche Uberstundennormen in Frauenberufen einen Erklarungsbeitrag fur die dort geringere Entlohnung auf. Eine Oaxaca/Blinder-Dekomposition verdeutlicht schlieslich, dass die Arbeitsinhalte bedeutsam zur Erklarung des „gender pay gap“ beitragen.
