1. Natural abundance 195Pt-13C correlation NMR spectroscopy on surfaces enabled by fast MAS dynamic nuclear polarization

Abstract: Surface organometallic chemistry has developed as an effective strategy for the rational design and synthesis of well-defined, single-site Pt-based heterogeneous catalysts. Given its high sensitivity to changes in electronic structure, 195Pt solid-state NMR spectroscopy offers a unique approach to investigate the chemical structure and local environment of Pt surface sites, providing invaluable insights for establishing structure-activity relationships. However, this approach is typically hindered by severe sensitivity issues, due to the low loading of Pt sites and the often-encountered large 195Pt chemical shift anisotropies. To overcome this limitation, 195Pt NMR signature of surface metal centers can be indirectly detected through protons. Indirect detection on 13C spins, has also been demonstrated to be feasible by combining isotopic labeling with dynamic nuclear polarization (DNP). Here, we extend this methodology to a supported Pt complex at natural abundance. The material was prepared by grafting (COD)PtMeOSi(OtBu)3 (COD = 1,5-cyclooctadiene, Me = methyl and tBu = tert‑butyl) onto partially dehydroxylated silica. DNP enhanced two-dimensional through-bond 13C{195Pt} heteronuclear correlation experiments were successfully implemented at fast magic angle spinning. They enabled the detection of the 0.37 % NMR-responsive surface species, thereby showcasing the remarkable sensitivity of this approach and its broad applicability. Key bonding information was obtained by measuring the correlated 13C and 195Pt isotopic chemical shifts as well as 1J(13C-195Pt) coupling constants, confirming directly the coordination structure of the surface Pt sites.

Abstract: International audience; The atomic-scale characterization of surface active sites on γ-alumina still represents a great challenge for numerous catalytic applications. Here, we combine advanced density functional theory (DFT) calculations with one- and two-dimensional proton solid-state NMR experiments to identify the exact location and the spatial proximity of hydroxyl groups on γ-alumina crystallites. Our approach relies on revisited models for the (100), (111), basal (110)b, and lateral (110)l facets of γ-alumina, as well as for the edges at their intersections. Notably, we show that the ≃0 ppm AlTd-μ1-OH protons are predominantly located on edges, where these are free from the H-bond network. The proximities among the AlTd-μ1-OH as well as with μ2-OH groups are revealed by 1H–1H dipolar correlation experiments and analyzed in the light of the DFT calculations, which identify their location on the basal (110)b facet and on the (110)b/(100) and (110)b/(110)l edges. Using chlorine atoms to probe the presence of hydroxyls, we show that the chlorination occurs selectively by exchanging μ1-OH located on edges and on lateral (110)l facets. By contrast, the basal (110)b and lateral (111) facets are not probed by Cl. This exchange explains the disappearance of the ≃0 ppm peak and of the correlations involving AlTd-μ1-OH species. Moreover, after chlorination, a deshielding of the AlTd is observed on high-resolution 27Al NMR spectra. More subtle effects are visible on the proton correlation spectra, which are attributed to the disruption of the H-bond network in the course of chlorination.

4. In Vitro Pharmacological Profile of a New Small Molecule Bradykinin B2 Receptor Antagonist

Abstract: We here report the discovery and early characterization of Compound 3, a representative of a novel class of small molecule bradykinin (BK) B2 receptor antagonists, and its superior profile to the prior art B2 receptor antagonists Compound 1 and Compound 2. Compound 3, Compound 2, and Compound 1 are highly potent antagonists of the human recombinant B2 receptor (Kb values 0.24, 0.95, and 1.24 nM, respectively, calcium mobilization assay). Compound 3 is more potent than the prior art compounds and icatibant in this assay (Kb icatibant 2.81 nM). The compounds also potently inhibit BK-induced contraction of endogenous B2 receptors in a human isolated umbilical vein bioassay. The potencies of Compound 3, Compound 2, Compound 1, and icatibant are (pA2 values) 9.67, 9.02, 8.58, and 8.06 (i.e. 0.21, 0.95, 2.63, and 8.71 nM), respectively. Compound 3 and Compound 2 were further characterized. They inhibit BK-induced c-Fos signaling and internalization of recombinant human B2 receptors in HEK293 cells, and do not antagonize the venous effects mediated by other G protein-coupled receptors in the umbilical vein model, including the bradykinin B1 receptor. Antagonist potency of Compound 3 at cloned cynomolgus monkey, dog, rat, and mouse B2 receptors revealed species selectivity, with a high antagonist potency for human and monkey B2 receptors, but several hundred-fold lower potency for the other B2 receptors. The in vitro off-target profile of Compound 3 demonstrates a high degree of selectivity over a wide range of molecular targets, including the bradykinin B1 receptor. Compound 3 showed a lower intrinsic clearance in the microsomal stability assay than the prior art compounds. With an efflux ratio of 1.0 in the Caco-2 permeability assay Compound 3 is predicted to be not a substrate of efflux pumps. In conclusion, we discovered a novel chemical class of highly selective and very potent B2 receptor antagonists, as exemplified by Compound 3. The compound showed excellent absorption in the Caco-2 assay, predictive of good oral bioavailability, and favourable metabolic stability in liver microsomes. Compound 3 has provided a significant stepping stone towards the discovery of the orally bioavailable B2 antagonist PHA-022121, currently in phase 1 clinical development.

Abstract: Background: Hypertrophic pyloric stenosis in otherwise healthy neonates frequently requires urgent surgical procedure but anaesthesia care may result in respiratory complications, such as hypoxaemia, pulmonary aspiration of gastric contents, and postoperative apnoea. The primary aim was to study whether or not the incidence of difficult airway management and of hypoxaemia in neonates undergoing pyloric stenosis repair was higher than that in neonates undergoing other surgeries. Methods: Data on neonates and infants undergoing anaesthesia and surgery for pyloric stenosis were extracted from the NEonate and Children audiT of Anesthesia pRactice In Europe (NECTARINE) database, for secondary analysis. Results: We identified 310 infants who had anaesthesia for surgery for pyloric stenosis. Difficult airway management (more than two attempts at laryngoscopy) was higher in children with pyloric stenosis when compared with the entire NECTARINE cohort (7.9% [95% confidence interval {CI}, 5.22–11.53] vs 4.4% [95% CI, 1.99–6.58]; relative risk [RR]=1.81 [95% CI, 1.21–2.69]; P=0.004), whereas transient hypoxaemia with oxygen saturation

Abstract: Nuclear magnetic resonance suffers from an intrinsically low sensitivity, which can be overcome by dynamic nuclear polarization (DNP). Gd(III) complexes are attractive exogenous polarizing agents for magic angle spinning (MAS) DNP due to their high chemical stability in contrast to nitroxide-based radicals. However, even the state-of-the-art Gd(III) complexes have so far provided relatively low DNP signal enhancements of ca. 36 in comparison to standard DNP biradicals, which show enhancements of over 200. Here, we report a series of new Gd(III) complexes for DNP and show that the observed DNP enhancements of the new and existing Gd(III) complexes are inversely proportional to the square of the zero-field splitting (ZFS) parameter D, which is in turn determined by the ligand-type and the local coordination environment. The experimental DNP enhancements at 9.4 T and the ZFS parameters measured with pulsed electron paramagnetic resonance (EPR) spectroscopy agree with the above model, paving the way for the development of more efficient Gd(III) polarizing agents., The Journal of Physical Chemistry C, 126 (27), ISSN:1932-7455, ISSN:1932-7447

Abstract: International audience; The structural characterization of supported molecular catalysts is challenging due to the low concentration of surface sites and the presence of several organic/organometallic surface groups resulting from the often complex surface chemistry associated with support functionalization. Here, we provide a complete atomic-scale description of all surface sites in a silica-supported iridium-N-heterocyclic carbene (Ir-NHC) catalytic material, at all stages of its synthesis. By combining a suitable isotope labelling strategy with the implementation of multi-nuclear dipolar recoupling DNP enhanced NMR experiments, the 3D structure of the Ir-NHC sites, as well as that of the synthesis intermediates were determined. As a significant fraction of parent surface fragments does not react during the multi-step synthesis, site-selective experiments were implemented to specifically probe proximities between the organometallic groups and the solid support. The NMR-derived structure of the iridium sites points to a well-defined conformation. By interpreting extended x-ray absorption fine structure (EXAFS) spectroscopy and chemical analysis data augmented by computational studies, the presence of two coordination geometries is demonstrated: Ir-NHC fragments coordinated by a 1,5-cyclooctadiene and one Cl ligand, as well as, more surprisingly, a fragment coordinated by two NHC and two Cl ligands. This study demonstrates a unique methodology to disclose individual surface structures in complex, multi-site environments, a long-standing challenge in the field of heterogeneous/supported catalysts, while revealing new, unexpected structural features of metallo-NHC supported substrates. It also highlights the potentially large diversity of surface sites present in functional materials prepared by surface chemistry, an essential knowledge to design materials with improved performances.

Paramagnetic metal ions such as Gd 3+ have recently shown promising results as polarizing agents for 1 H, 13 C, and 15 N nuclear spins. We demonstrate that the widely available and inexpensive chemical agent Gd(NO 3) 3 achieves significant signal enhancements for the 13 C and 15 N nuclear sites of [2-13 C, 15 N]glycine at 9.4 T and ∼105 K. Analysis of the signal enhancement profiles at two magnetic fields, in conjunction with electron paramagnetic resonance data, reveals the solid effect to be the dominant signal enhancement mechanism. The signal amplification obtained paves the way for efficient dynamic nuclear polarization without the need for challenging synthesis of Gd 3+ polarizing agents.

Abstract: Magnetic resonance techniques are successfully utilized in a broad range of scientific disciplines and in a number of practical applications, with medical MRI being the most widely-known example. Currently, both fundamental and applied magnetic resonance are enjoying a major boost owing to the dramatic signal enhancement provided by the rapidly-developing field of spin hyperpolarization. Such techniques are able to enhance signal intensities in magnetic resonance by several orders of magnitude, and thus to largely overcome its major disadvantage of relatively low sensitivity compared to other analytical techniques. This provides new impetus for existing applications, and, even more importantly, this opens the gates to numerous novel and exciting possibilities in the broad fields of fundamental and applied magnetic resonance. There are many different techniques that fall under the umbrella term “hyperpolarization”. Existing reviews cover the various subfields, but they are mostly addressed separately, and are seldom perceived as integral parts of the same field. In this review we attempt to unify the many methods that are used to hyperpolarize nuclear spins into one picture. Specifically, before delving into the individual techniques, we provide a detailed analysis of the underlying principles of spin hyperpolarization. We attempt to uncover and classify the origins of hyperpolarization; to establish its sources and the specific mechanisms that enable the flow of polarization from a source to the target nuclear spins. After outlining the inner workings of hyperpolarization, we give a more detailed analysis of individual hyperpolarization techniques: the mechanisms by which they work, fundamental and technical requirements, characteristic applications, unresolved issues and possible future directions. While a substantial progress has been achieved in the field of spin hyperpolarization in recent years, the continuing growth of activity indicates that this is just the beginning. It is unlikely that fundamentally new sources of hyperpolarization will be uncovered in the near future, but we expect the field to flourish as new ways to improve and utilize current hyperpolarization techniques are identified and implemented. There is great scope for cross-fertilization between known methods, and developments in one area (e.g., prolonging polarization lifetimes, or creating more efficient excitation-detection schemes) can have a very broad impact across the entire field of hyperpolarization. We hope this review will facilitate this process, since advances in hyperpolarization will help to overcome existing challenges in magnetic resonance and enable novel applications.

Abstract: Dynamic nuclear polarization (DNP) at cryogenic temperatures has proved to be a valuable technique to enhance the sensitivity of solid-state NMR spectroscopy. Over the years, sample formulations have been optimized for experiments at cryogenic temperatures. At 9.4 T, the best performing polarizing agents are dinitroxides such as AMUPol and TEKPol that lead to enhancement factors of around 250 at 100 K. However, the performance of these radicals plummets at higher temperatures. Here we introduce trehalose-based DNP polarizing matrices, suitable to embed biomolecular assemblies. Several formulation protocols are investigated, in combination with various polarizing agents, including a new biradical structure chemically tethered to a trehalose molecule. The DNP efficiency of these new polarizing media is screened as a function of the radical concentration, the hydration level of the matrix and the protein content. Sizeable enhancement factors are reported at 100 K and 9.4 T. More importantly, we show that the DNP performance of these new polarizing media outperform the conventionally used water/glycerol mixture at temperatures above 180 K. This study establishes trehalose matrices as a promising DNP medium for experiments at temperatures >150 K where conventional water-based formulations soften and are no longer viable, thus opening new avenues for DNP enhanced solid-state NMR spectroscopy at temperatures close to ambient temperature.

Abstract: Pairing the spectral resolution provided by high magnetic fields at ambient temperature with the enhanced sensitivity offered by dynamic nuclear polarization (DNP) is a major goal of modern solid-state NMR spectroscopy, which will allow one to unlock ever-challenging applications. This study demonstrates that, by combining HyTEK2, a hybrid BDPA-nitroxide biradical polarizing agent, with ortho-terphenyl (OTP), a rigid DNP matrix, enhancement factors as high as 65 can be obtained at 230 K, 40 kHz magic angle spinning (MAS), and 18.8 T. The temperature dependence of the DNP enhancement and its behavior around the glass transition temperature (Tg) of the matrix is investigated by variable-temperature EPR measurements of the electron relaxation properties and numerical simulations. A correlation is suggested between the decrease in enhancement at the passage of the Tg and the concomitant drop of both transverse electron relaxation times in the biradical.

Abstract: Gallia–alumina (Ga,Al)2O3(x : y) spinel-type solid solution nanoparticle catalysts for propane dehydrogenation (PDH) were prepared with four nominal Ga : Al atomic ratios (1 : 6, 1 : 3, 3 : 1, 1 : 0) using a colloidal synthesis approach. The structure, coordination environment and distribution of Ga and Al sites in these materials were investigated by X-ray diffraction, X-ray absorption spectroscopy (Ga K-edge) as well as 27Al and 71Ga solid state nuclear magnetic resonance. The surface acidity (Lewis or Brønsted) was probed using infrared spectroscopy with pyridine and 2,6-dimethylpyridine probe molecules, complemented by element-specific insights (Ga or Al) from dynamic nuclear polarization surface enhanced cross-polarization magic angle spinning 15N{27Al} and 15N{71Ga} J coupling mediated heteronuclear multiple quantum correlation NMR experiments using 15N-labelled pyridine as a probe molecule. The latter approach provides unique insights into the nature and relative strength of the surface acid sites as it allows to distinguish contributions from Al and Ga sites to the overall surface acidity of mixed (Ga,Al)2O3 oxides. Notably, we demonstrate that (Ga,Al)2O3 catalysts with a high Al content show a greater relative abundance of four-coordinated Ga sites and a greater relative fraction of weak/medium Ga-based surface Lewis acid sites, which correlates with superior propene selectivity, Ga-based activity, and stability in PDH (due to lower coking). In contrast, (Ga,Al)2O3 catalysts with a lower Al content feature a higher fraction of six-coordinated Ga sites, as well as more abundant Ga-based strong surface Lewis acid sites, which deactivate through coking. Overall, the results show that the relative abundance and strength of Ga-based surface Lewis acid sites can be tuned by optimizing the bulk Ga : Al atomic ratio, thus providing an effective measure for a rational control of the catalyst performance., Coordination geometry and Lewis acidity of Ga and Al (bulk and surface) sites in mixed oxide gallia–alumina nanoparticles is correlated with the performance in propane dehydrogenation.

16. Unravelling the molecular structure and confinement of an or-ganometallic catalyst heterogenized within amorphous porous polymers

Abstract: The catalytic activity of multifunctional microporous materials is directly linked to the spatial arrangement of their struc-tural building blocks. Despite great achievements in the design and use of isolated catalytic sites within such materials, the precise determination of their atomic-level structure and their local environment still remains a fundamental chal-lenge, especially when they are hosted in non-crystalline solids. Here, we show that by combining NMR measurements with pair distribution function (PDF) analysis and computational chemistry, a very accurate picture of the organometallic Cp*Rh catalytic sites inside the cavity of porous organic polymers can be determined. Two microporous supports based on bipyridine and biphenyl motifs functionalized with NH2 or NO2 groups were considered. Making use of differential PDF, Dynamic Nuclear Polarization (DNP) enhanced solid-state NMR spectroscopy on 15N labelled–NH2 and –NO2 materi-als, and 129Xe NMR, the detailed structure of the heterogenized organometallic complex and its confinement within the amorphous porous organic polymers is revealed with a precision of 0.1 Å, fully matched by the computed models. While the same well-defined molecular structure is observed for the organometallic catalyst independently of the functionalisa-tion of the porous organic polymer, subtle changes are detected in the average ligand-pore wall distance and interactions in the two materials.

Abstract: Background: Neonates and infants are susceptible to hypoxaemia in the perioperative period. The aim of this study was to analyse interventions related to anaesthesia tracheal intubations in this European cohort and identify their clinical consequences. Methods: We performed a secondary analysis of tracheal intubations of the European multicentre observational trial (NEonate and Children audiT of Anaesthesia pRactice IN Europe [NECTARINE]) in neonates and small infants with difficult tracheal intubation. The primary endpoint was the incidence of difficult intubation and the related complications. The secondary endpoints were the risk factors for severe hypoxaemia attributed to difficult airway management, and 30 and 90 day outcomes. Results: Tracheal intubation was planned in 4683 procedures. Difficult tracheal intubation, defined as two failed attempts of direct laryngoscopy, occurred in 266 children (271 procedures) with an incidence (95% confidence interval [CI]) of 5.8% (95% CI, 5.1e6.5). Abstract: BACKGROUND Little is known about current clinical practice concerning peri-operative red blood cell transfusion in neonates and small infants. Guidelines suggest transfusions based on haemoglobin thresholds ranging from 8.5 to 12��g��dl-1, distinguishing between children from birth to day 7 (week 1), from day 8 to day 14 (week 2) or from day 15 (��week 3) onwards. OBJECTIVE To observe peri-operative red blood cell transfusion practice according to guidelines in relation to patient outcome. DESIGN A multicentre observational study. SETTING The NEonate-Children sTudy of Anaesthesia pRactice IN Europe (NECTARINE) trial recruited patients up to 60 weeks' postmenstrual age undergoing anaesthesia for surgical or diagnostic procedures from 165 centres in 31 European countries between March 2016 and January 2017. PATIENTS The data included 5609 patients undergoing 6542 procedures. Inclusion criteria was a peri-operative red blood cell transfusion. MAIN OUTCOME MEASURES The primary endpoint was the haemoglobin level triggering a transfusion for neonates in week 1, week 2 and week 3. Secondary endpoints were transfusion volumes, 'delta haemoglobin' (preprocedure - transfusion-triggering) and 30-day and 90-day morbidity and mortality. RESULTS Peri-operative red blood cell transfusions were recorded during 447 procedures (6.9%). The median haemoglobin levels triggering a transfusion were 9.6 [IQR 8.7 to 10.9] g��dl-1 for neonates in week 1, 9.6 [7.7 to 10.4] g��dl-1 in week 2 and 8.0 [7.3 to 9.0] g��dl-1 in week 3. The median transfusion volume was 17.1 [11.1 to 26.4] ml��kg-1 with a median delta haemoglobin of 1.8 [0.0 to 3.6] g��dl-1. Thirty-day morbidity was 47.8% with an overall mortality of 11.3%. CONCLUSIONS Results indicate lower transfusion-triggering haemoglobin thresholds in clinical practice than suggested by current guidelines. The high morbidity and mortality of this NECTARINE sub-cohort calls for investigative action and evidence-based guidelines addressing peri-operative red blood cell transfusions strategies. Abstract: International audience; Over the last decade, halogenated semiconducting polymers have attracted considerable interest due to their outstanding optoelectronic properties. Thus, most today's OPV benchmark organic semiconductors are halogenated materials, either electron donor polymers or non-fullerene acceptor (NFA) small molecules. However, the nature and position of the substituted halogen atoms in halogenated semiconducting polymers impact, through self-assembly modification, on their optoelectronic properties in a way which is 2 difficult to predict. Yet, the solid-state self-assembling of this materials has been shown to be a key parameter towards high charge transport properties and photovoltaic efficiencies. In this context, there is still a need to develop analytical methods that will enable an atomicscale structural characterization of these materials as function of the halogenation. In this manuscript, we explore the solid-state nuclear magnetic resonance (NMR) under magic angle spinning (MAS) as a tool to investigate the local structure and supramolecular organization of a series of conjugated polymers, specially designed for this study. Through a comprehensive study using complementary techniques including MAS-NMR, small and wideangle X-ray scattering (SWAXS) and molecular modelling investigations, we have definitely succeeded in determining the molecular conformation of these polymers in relation to their chemical composition.

Abstract: The molecular level characterization of heterogeneous catalysts is challenging due to the low concentration of surface sites and the lack of techniques that can selectively probe the surface of a heterogeneous material. Here, we report the joint application of room temperature proton-detected NMR spectroscopy under fast magic angle spinning (MAS) and dynamic nuclear polarization surface enhanced NMR spectroscopy (DNP-SENS), to obtain the 195Pt solid-state NMR spectra of a prototypical example of highly dispersed Pt sites (single site or single atom), here prepared via surface organometallic chemistry, by grafting [(COD)Pt(OSi(OtBu)3)2] (1, COD = 1,5-cyclooctadiene) on partially dehydroxylated silica (1@SiO2). Compound 1@SiO2 has a Pt loading of 3.7 wt %, a surface area of 200 m2/g, and a surface Pt density of around 0.6 Pt site/nm2. Fast MAS 1H{195Pt} dipolar-HMQC and S-REDOR experiments were implemented on both the molecular precursor 1 and on the surface complex 1@SiO2, providing access to 195Pt isotropic shifts and Pt–H distances, respectively. For 1@SiO2, the measured isotropic shift and width of the shift distribution constrain fits of the static wide-line DNP-enhanced 195Pt spectrum, allowing the 195Pt chemical shift tensor parameters to be determined. Overall the NMR data provide evidence for a well-defined, single-site structure of the isolated Pt sites.

Abstract: A silica-supported monomeric alkylaluminum co-catalyst was prepared via surface organometallic chemistry by contacting tris(neopentyl)aluminum and partially dehydroxylated silica. This system, fully characterized by solid-state 27 Al NMR spectroscopy augmented by computational studies, efficiently activates (n Bu3 P)2 NiCl2 towards dimerization of ethene, demonstrating comparable activity to previously reported dimeric diethylaluminum chloride supported on silica. Three types of aluminum surface species have been identified: monografted tetracoordinated Al species as well as two types of bisgrafted Al species-tetra- and pentacoordinated. Of them, only the monografted Al species is proposed to be able to activate the (n Bu3 P)2 NiCl2 complex and generate the active cationic species.

Abstract: Techniques that can characterize the molecular structures of dilute surface species are required to facilitate the rational synthesis and improvement of single-site and single-atom, such as the important class of Pt-based systems. In this context, 195Pt solid-state NMR spectroscopy could be an ideal tool for this task because 195Pt NMR spectra and sizeable chemical shift anisotropy (CSA) are highly sensitive probes of the local chemical environment and electronic structure. However, the broadening of 195Pt solid-state NMR spectra by CSA often results in low NMR sensitivity. Furthermore, characterization of Pt sites on surfaces is complicated by the typical low Pt loadings that are between 0.2 to 5 wt.%. Here, we introduce a set of solid-state NMR methods that exploit fast MAS and indirect detection of a sensitive spy nucleus (1H or 31P) to enable rapid acquisition of 195Pt MAS NMR spectra. We demonstrate that high-resolution wideline 195Pt MAS NMR spectra can be in minutes to a few hours for a series of molecular and single-site Pt species grafted on silica with Pt loading of only 3-5 wt.%. Low-power, long-duration, sideband-selective excitation and saturation pulses are incorporated into t1-noise eliminated (TONE) dipolar heteronuclear multiple quantum coherence (D-HMQC), perfect echo resonance echo saturation pulse double resonance (PE RESPDOR) or J-resolved pulse sequences. The complete 195Pt MAS NMR spectrum is then reconstructed by recording a series of 1D NMR spectra where the offset of the 195Pt pulses is varied. Analysis of the 195Pt MAS NMR spectra yields the 195Pt chemical shift tensor parameters. Analysis of the NMR signatures based on relativistic zeroth order approximation (ZORA) DFT calculations enables the rationalization of changes in the observed 195Pt CSA across the series of Pt compounds. Simple and predictive orbital models relate the measured spectral signatures to specific electronic environments and allows the identification of coordination environment by inspection of the CSA (isotropic chemical shift and measured spans). The methodology developed here paves the way for the detailed structural and electronic analysis of dilute platinum sites in single-atom and single-site heterogeneous catalysts.

Abstract: A new heteronuclear decoupling pulse sequence is introduced, dubbed ROtor-Synchronized Phase-Alternated Cycles (ROSPAC). It is based on a partial refocusing of the coherences (spin operator products or cross-terms) [Filip et al., J. Mag. Reson. 176, 2 (2005)] responsible for transverse spin-polarization dephasing, on the irradiation of a large pattern of radio-frequencies, and on a significant minimization of the cross-effects implying 1H chemical-shift anisotropy. Decoupling efficiency is analyzed by numerical simulations and experiments and compared to that of established decoupling sequences [swept-frequency two-pulse phase-modulated (TPPM), TPPM, small phase incremental alternation (SPINAL), refocused Continuous-wave (CWApa), and Rotor-Synchronized Hahn-Echo pulse train (RS-HEPT)]. It was found that ROSPAC offers good 1H offset robustness for a large range of chemical shifts and low radio-frequency (RF) powers, and performs very well in the ultra-fast magic-angle spinning (MAS) regime, where it is almost independent from RF power and permits it to avoid rotary-resonance recoupling conditions ( v1 = nv r, n = 1, 2). It has the advantage that only the pulse lengths require optimization and has a low duty cycle in the pulsed decoupling regime. The efficiency of the decoupling sequence is demonstrated on a model microcrystalline sample of the model protein domain GB1 at 100 kHz MAS at 18.8 T.

Abstract: Significance Amyloid-β (Aβ) is the subject of intense scrutiny because of its close association with Alzheimer’s disease (AD), which currently afflicts about 50 million people worldwide. The results reported in this manuscript focus on the new possibilities provided by ultrafast magic-angle spinning (MAS) 1H detection and fast-MAS dynamic nuclear polarization (DNP), which have ushered in a new era for NMR-based structural biology, but whose potential has not yet been fully exploited for the structural investigation of complex amyloid assemblies. This work demonstrates the expeditious structural analysis of amyloid fibrils, without requiring preparation of large sample amounts, and sets the stage for future studies of unlabeled AD peptides derived from tissue samples available in limited quantities., Several publications describing high-resolution structures of amyloid-β (Aβ) and other fibrils have demonstrated that magic-angle spinning (MAS) NMR spectroscopy is an ideal tool for studying amyloids at atomic resolution. Nonetheless, MAS NMR suffers from low sensitivity, requiring relatively large amounts of samples and extensive signal acquisition periods, which in turn limits the questions that can be addressed by atomic-level spectroscopic studies. Here, we show that these drawbacks are removed by utilizing two relatively recent additions to the repertoire of MAS NMR experiments—namely, 1H detection and dynamic nuclear polarization (DNP). We show resolved and sensitive two-dimensional (2D) and three-dimensional (3D) correlations obtained on 13C,15N-enriched, and fully protonated samples of M0Aβ1-42 fibrils by high-field 1H-detected NMR at 23.4 T and 18.8 T, and 13C-detected DNP MAS NMR at 18.8 T. These spectra enable nearly complete resonance assignment of the core of M0Aβ1-42 (K16-A42) using submilligram sample quantities, as well as the detection of numerous unambiguous internuclear proximities defining both the structure of the core and the arrangement of the different monomers. An estimate of the sensitivity of the two approaches indicates that the DNP experiments are currently ∼6.5 times more sensitive than 1H detection. These results suggest that 1H detection and DNP may be the spectroscopic approaches of choice for future studies of Aβ and other amyloid systems.

Abstract: International audience; Obtaining an atomic-scale description of the chemical interactions of phosphates with an oxide support, such as γ-Al2O3, is essential to get a rational understanding of the role of phosphate additives for a great number of heterogeneous catalysts, as well as to improve the use of this element. Combining cutting-edge Dynamic Nuclear Polarization Surface Enhanced NMR Spectroscopy (DNP SENS) techniques with Density Functional Theory (DFT) calculations, we provide an accurate molecular description of phosphate speciation on γ-Al2O3 surfaces for various P surface coverages after drying at 120 °C. Thanks to 31P double- and triple-quantum filtered NMR experiments as well as to 27Al–31P dipolar- and scalar-based correlation spectra, we demonstrate the presence of polyphosphates and of Al–O–P connectivities at the exposed facets of γ-Al2O3. DFT-based thermodynamics shows that phosphates (mono- or di-) are preferentially covalently bonded on the (1 1 0) γ-Al2O3 facet with high-dentation modes. These high-dentation modes are favored by entropy gain due to water desorption. We used the gauge-including projector-augmented wave (GIPAW) DFT method for 31P NMR chemical shifts calculations and propose a systematic identification of the various types of phosphates covalently or noncovalently bonded to the alumina surface. The calculations confirm the existence of polyphosphates as observed experimentally. Since the surface condensation into polyphosphates is endergonic, the presence of polyphosphates on the surface is likely to result from their direct adsorption in impregnation solution. The observed increasing concentration of polyphosphates with the coverage could be related to a less likely hydrolysis due to the reduced availability of sites to stabilize the fragmented oligomers. This understanding opens the way to a better control over the speciation of phosphate species that are known to be key in the preparation of supported catalysts over alumina.

Abstract: International audience; A novel heterobimetallic tantalum/iridium hydrido complex, [{Ta(CH2 t Bu)3}{IrH2(Cp*)}] 1, featuring a very short metal-metal bond, has been isolated through an original alkane elimination route from Ta(CH t Bu)(CH2 t Bu)3 and Cp*IrH4. This molecular precursor has been used to synthesize well-defined silica-supported low-coordinate heterobime-tallic hydrido species [≡SiOTa(CH2 t Bu)2{IrH2(Cp*)}] 5 and [≡SiOTa(CH2 t Bu)H{IrH2(Cp*)}] 6 using a surface organometallic chemistry approach (SOMC). The SOMC methodology prevents undesired dimerization as encountered in solution and leading to a tetranuclear species [{Ta(CH2 t Bu)2}(Cp*IrH)]2, 4. This approach therefore allows access to unique low-coordinate species not attainable in solution. These original supported Ta/Ir species exhibit drastically enhanced catalytic performances in H/D exchange reactions with respect to (i) monometallic analogues as well as (ii) homogeneous systems. In particular, material 6 promotes the H/D exchange between fluorobenzene and C6D6 or D2 as deuterium sources with excellent productivity (TON up to 1422; TOF up to 23.3 h-1) under mild conditions (25°C, subatmospheric D2 pressure) without any additives.

Abstract: Dynamic Nuclear Polarization (DNP) is an effective approach to alleviate the inherently low sensitivity of solid-state NMR (ssNMR) under magic angle spinning (MAS) towards large-sized multi-domain complexes and assemblies. DNP relies on a polarization transfer at cryogenic temperatures from unpaired electrons to adjacent nuclei upon continuous microwave irradiation. This is usually made possible via the addition in the sample of a polarizing agent. The first pioneering experiments on biomolecular assemblies were reported in the early 2000s on bacteriophages and membrane proteins. Since then, DNP has experienced tremendous advances, with the development of extremely efficient polarizing agents or with the introduction of new microwaves sources, suitable for NMR experiments at very high magnetic fields (currently up to 900 MHz). After a brief introduction, several experimental aspects of DNP enhanced NMR spectroscopy applied to biomolecular assemblies are discussed. Recent demonstration experiments of the method on viral capsids, the type III and IV bacterial secretion systems, ribosome and membrane proteins are then described.

Abstract: The adsorption and reaction properties of heterogeneous zeolite catalysts (e.g. for catalytic cracking of petroleum, partial oxidation of natural gas) depend strongly on the types and distributions of Al heteroatoms in the aluminosilicate frameworks. The origins of these properties have been challenging to discern, owing in part to the structural complexity of aluminosilicate zeolites. Herein, combined solid-state NMR and synchrotron X-ray powder diffraction analyses show the Al atoms locate preferentially in certain framework sites in the zeolite catalyst Al-SSZ-70. Through-covalent-bond 2D 27 Al{29 Si} J-correlation NMR spectra allow distinct framework Al sites to be identified and their relative occupancies quantified. The analyses show that 94 % of the Al atoms are located at the surfaces of the large-pore interlayer channels of Al-SSZ-70, while only 6 % are in the sub-nm intralayer channels. The selective siting of Al atoms accounts for the reaction properties of catalysts derived from SSZ-70.

Abstract: Background: Neonates and infants requiring anaesthesia are at risk of physiological instability and complications, but triggers for peri-anaesthetic interventions and associations with subsequent outcome are unknown. Methods: This prospective, observational study recruited patients up to 60 weeks' postmenstrual age undergoing anaesthesia for surgical or diagnostic procedures from 165 centres in 31 European countries between March 2016 and January 2017. The primary aim was to identify thresholds of pre-determined physiological variables that triggered a medical intervention. The secondary aims were to evaluate morbidities, mortality at 30 and 90 days, or both, and associations with critical events. Results: Infants (n=5609) born at mean (standard deviation [SD]) 36.2 (4.4) weeks postmenstrual age (35.7% preterm) underwent 6542 procedures within 63 (48) days of birth. Abstract: Neonatal and infant anaesthesia are associated with a high risk of perioperative complications. The aim of the current study was to describe those risks in France using the French data from the NECTARINE study.Data from the French centres that participated to the NECTARINE study were analysed. The primary goal of the study was the description of patients' characteristics, procedures and perioperative management and their comparison with the results of the European NECTARINE study. Secondary outcomes were the description of major perioperative complications and death.Overall, 926 procedures collected in 15 centres (all teaching hospitals) were analysed. Comparison between the French and European NECTARINE cohorts found few differences related to patients' characteristics and procedures. The rate of interventions for critical events (respiratory, haemodynamic, and metabolic) was similar between the two cohorts. Near-infrared spectroscopy monitoring was used in 12% of procedures. Nearly none of the thresholds for these interventions met the published standards. By day 30, complications (respiratory, haemodynamic, metabolic, renal, and liver failure) and death were observed in 14.4% [95% CI 11.6-16.4]% and 1.8% [95% CI 1.1-2.9] of cases, respectively.Although the health status of the patients in the French cohort was less severe, procedures, management and postoperative complications and mortality rates were similar to the European cohort. However, thresholds for interventions were often inadequate in both cohorts. Efforts should be undertaken to improve the knowledge and use of new monitoring devices in this population.

Abstract: Alumina and aluminosilicates, prepared under various synthesis conditions, play a central role in heterogeneous catalysis with a broad range of industrial applications. We report herein the atomic-scale structure of alumina layers obtained by atomic layer deposition (ALD) of trimethylaluminum onto partially dehydroxylated silica. Such a detailed insight into the atomic structure of the species formed with increasing Al content was gained using a variety of one- and two-dimensional solid-state nuclear magnetic resonance (NMR) experiments involving 27Al, 1H, and 29Si nuclei. Multicomponent fittings of the 1D and 2D experimental data sets allowed us to show that at 3.4 wt % of deposited Al, a submonolayer containing [4]Al(3Si), [4]Al(4Si), and [5]Al(2Si) species forms on the silica surface, with most of these sites carrying OH groups. The films obtained after additional ALD cycles (depositing 9.2 or 15.4 wt % Al) feature characteristics of an amorphous alumina phase with a high concentration of [5]Al species and abundant OH groups. The most probable species at the interface between silica and alumina are [4]Al(2Si), [4]Al(3Si), and [5]Al(2Si). 15N dynamic nuclear polarization surface-enhanced NMR spectroscopy (15N DNP SENS) and infrared spectroscopy using 15N-labeled pyridine as a probe molecule reveal that aluminum oxide layers on amorphous silica contain both strong Brønsted and strong Lewis acid sites, whereby the relative abundance and nature of these sites, and therefore the acidity of the surface, evolve with increasing thickness of the alumina films (controlled by the number of ALD cycles). This study provides the first in-depth atomic-scale description of (sub-)nanometer-scale aluminum oxide films prepared by ALD as a function of their growth on a partially dehydroxylated silica support, opening the way to molecular-level understanding of the catalytic activity of such heterogeneous catalysts with tailored acidity., Chemistry of Materials, 33 (9), ISSN:0897-4756

Abstract: Despite decades of extensive studies, the atomic-scale structure of the active sites in heterogeneous Ziegler-Natta (ZN) catalysts, one of the most important processes of the chemical industry, remains elusive and a matter of debate. In the present work, the structure of “active sites” of ZN catalysts in the absence of ethylene, referred to as “dormant active sites”, is elucidated from magnetic resonance experiments, carried out on samples reacted with increasing amounts of BCl3 so as to enhance the concentration of active sites and observe clear spectroscopic signatures. Using EPR and NMR spectroscopies, in particular 2D HYSCORE experiments complemented by DFT calculations, we show that the activated ZN catalysts contain bimetallic alkyl-Ti(III),Al species whose amount is directly linked to the polymerization activity of MgCl2-supported Ziegler-Natta catalysts. This connects those spectroscopic signatures to the active species formed in the presence of ethylene, and enables us propose an ethylene polymerization mechanism on the observed bimetallic alkyl-Ti(III),Al species based on DFT computations

Abstract: International audience; The nature and spectroscopic expression of external surface sites of zeolites, in particular ZSM-5, is a long-debated question. Herein, we use three cutting-edge techniques: Fourier-transform infrared spectroscopy (FTIR) with Fourier self-deconvolution (FSD), high magnetic field proton NMR spectroscopy under fast magic-angle spinning (MAS) and periodic boundary Density Functional Theory (DFT) calculations to study external surface models and analyze the effect of crystallite size. This provides an unequaled description of the various kinds of hydroxyl groups and of their proximities. The hydrogen-bond donor, acceptor or isolated nature of the hydroxyls results in distinct signals both in FTIR and NMR spectra, but the peak assignment is not the same from one technique to the other when the chemical nature of the hydroxyl changes. Bridging Si–(OH)–Al groups and Al–(H2O) lead to overlapping signals in one-dimensional 1H MAS NMR, whereas their contributions are strongly different in FTIR spectra. However, quantification and proximity assessment could only be obtained by 1H MAS NMR. With DFT, we confirm previous assignments for silanols and Si–(OH)–Al bridging OH groups. Other signals (between 3750 and 3600 cm–1, and between 1 and 4 ppm) are not only assigned to extra-framework species (which we confirm with dedicated models), but also enclose the signature of sites exposed at the external surface of ZSM-5. In particular, Al–(H2O) species (∼3665 cm–1; 3.8, 2.6 ppm) and silanol–Al (∼3740, 3720, 3665 cm–1; 2.6, 2.2 ppm) contribute to several features depending on their environment. μ1-Al–OH are also present at the external surface in low amount, with a 3780 cm–1 signal in IR, and weak signals in the 0–2 ppm interval in 1H MAS NMR.

In the present work, the structure of active sites of ZN catalysts in the absence of ethylene, referred to as dormant active sites, is elucidated from magnetic resonance experiments carried out on samples reacted with increasing amounts of BCl3 so as to enhance the concentration of active sites and observe clear spectroscopic signatures. Using electron paramagnetic resonance (EPR) and NMR spectroscopies, in particular 2D HYSCORE experiments complemented by density functional theory (DFT) calculations, we show that the activated ZN catalysts contain bimetallic alkyl-Ti(III),Al species whose amount is directly linked to the polymerization activity of MgCl2-supported Ziegler–Natta catalysts. This connects those spectroscopic signatures to the active species formed in the presence of ethylene and enables us to propose an ethylene polymerization mechanism on the observed bimetallic alkyl-Ti(III),Al species based on DFT computations.

Abstract: International audience; This paper aims at designing chromophores with high aggregation induced emission (AIE) properties for twophoton fluorescence microscopy (2PFM), which is one of the best suited types of microscopy for the imaging of living organism or thick biological tissues. Tetraphenylethylene (TPE) derivatives are common building blocks in the design chromophores with efficient AIE properties. Therefore, in this study, extended TPE AIEgens specifically optimized for twophoton absorption (2PA) are synthetized and the resulting (E/Z) isomers are separated using chromatography on chiral supports. Comparative characterization of the AIE properties is performed on the pure (Z) and (E) isomers and the mixture, allowing us, in combination with powder X-ray diffraction and solid-state NMR to document a profound impact of crystallinity on solid state fluorescence properties. In particular, we show that stereopure AIEgens form aggregates of superior crystallinity, which in turn exhibit a higher fluorescence quantum yield compared to diastereoisomers mixtures. Preparation of stereopure organic nanoparticles affords very bright fluorescent contrast agents, which interest for cellular and intravital two-photon microscopy is illustrated on human breast cancer cell line and on zebrafish embryos.

Abstract: International audience; A stepwise surface organometallic chemistry (SOMC) methodology consisting in using a silica-supported tantalum species, [(≡SiO)Ta(CH t Bu)(CH2 t Bu)2], as a reactive center to coordinate an iridium site, was developed to construct tantalum/iridium heterobimetallic edifices. The resulting material, MAT-2, exhibits enhanced catalytic performances-both in H/D isotopic exchange and alkane metathesis reactions-in comparison to MAT-1, which was prepared from the direct grafting on silica of a well-defined heterobimetallic Ta/Ir complex. We projected that the difference in catalytic activity was due to the presence of distinct active sites and we used a combination of advanced spectroscopic methods (IR, solid-state NMR and XAS spectroscopies) as well as modeling (computational studies and molecular models) to identify the structure of these surface species. These investigations point towards the presence of three types of active sites at the surface of MAT-2: the heterobimetallic surface species, [≡SiOTa(CH2 t Bu)2{IrH2(Cp*)}] 2-s, which is also found in MAT-1, as well as an unanticipated heterotrimetallic species, [≡SiOTa(CH2 t Bu)2{IrH2(Cp*)}], 3-s along with some unreacted monometallic Ta sites [(≡SiO)Ta(CH t Bu)(CH2 t Bu)2], 1-s. The well-defined trimetallic surface species 3-s was independently prepared and characterized to support this hypothesis. This study highlights the importance of the synthetic methodology used for the preparation of heterobimetallic species through SOMC, and the difficulty to obtain true single-sites surface species.

Abstract: Structure determination of adjuvant-coupled antigens is essential for rational vaccine development but has so far been hampered by the relatively low antigen content in vaccine formulations and by their heterogeneous composition. Here we show that magic-angle spinning (MAS) solid-state NMR can be used to assess the structure of the influenza virus hemagglutinin stalk long alpha helix antigen, both in its free, unformulated form and once chemically coupled to the surface of large virus-like particles (VLPs). The sensitivity boost provided by high-field dynamic nuclear polarization (DNP) and proton detection at fast MAS rates allows to overcome the penalty associated with the antigen dilution. Comparison of the MAS NMR fingerprints between the free and VLP-coupled forms of the antigen provides structural evidence of the conservation of its native fold upon bioconjugation. This work demonstrates that high-sensitivity MAS NMR is ripe to play a major role in vaccine design, formulation studies, and manufacturing process development.

Abstract: The development of magic-angle spinning dynamic nuclear polarization (MAS DNP) has allowed atomic-level characterization of materials for which conventional solid-state NMR is impractical due to the lack of sensitivity. The rapid progress of MAS DNP has been largely enabled through the understanding of rational design concepts for more efficient polarizing agents (PAs). Here, we identify a new design principle which has so far been overlooked. We find that the local geometry around the unpaired electron can change the DNP enhancement by an order of magnitude for two otherwise identical conformers. We present a set of 13 new stable mono- and dinitroxide PAs for MAS DNP NMR where this principle is demonstrated. The radicals are divided into two groups of isomers, named open (O-) and closed (C-), based on the ring conformations in the vicinity of the N-O bond. In all cases, the open conformers exhibit dramatically improved DNP performance as compared to the closed counterparts. In particular, a new urea-based biradical named HydrOPol and a mononitroxide O-MbPyTol yield enhancements of 330 ± 60 and 119 ± 25, respectively, at 9.4 T and 100 K, which are the highest enhancements reported so far in the aqueous solvents used here. We find that while the conformational changes do not significantly affect electron spin-spin distances, they do affect the distribution of the exchange couplings in these biradicals. Electron spin echo envelope modulation (ESEEM) experiments suggest that the improved performance of the open conformers is correlated with higher solvent accessibility.
