Environmental pollutants are claimed to be major factors involved in the progressive decline of the fertility rate worldwide. Exposure to the heavy metal Cadmium (Cd) has been associated with reproductive toxicity due to its ionic mimicry. However, the possible direct accumulation of Cd in human sperm cells has been poorly investigated. In this study, we aimed to clarify the possible direct effect of Cd exposure on sperm function through the analysis of its cell accumulation. Semen samples from 30 male subjects residing in high environmental impact areas and adhering to the “Exposoma e Plurifocalità nella Prevenzione Oncologica” campaign for testis cancer prevention were compared with semen samples from 15 males residing in low exposure areas. Semen levels and cell Cd content were quantified by inductively coupled plasma (ICP) spectroscopy. Cell Cd distribution was assessed by scanning electron microscopy coupled with energy dispersive spectroscopy (SEM-EDS). The impact of Cd on sperm function was evaluated by the in vitro exposure to the heavy metal, whilst possible scavenging approaches/agents were assessed. In addition to higher values of semen Cd, exposed subjects showed a reduction in total motile sperm fraction compared to not-exposed controls (59.6% ± 13.6% vs. 66.3% ± 7.3%, p = 0.037). Semen Cd levels were also significantly correlated with SEM-EDS signals of Cd detected on the head and neck of sperm (respectively p = 0.738, p < 0.001 and ρ = 0.465, p < 0.001). A total of 2 h of in vitro exposure to 0.5 μM Cd was associated with a significant reduction of sperm progressive motility. Scavenging approaches with either hypo-osmotic swelling or 10 μM reduced glutathione were ineffective in blunting cell Cd and restoring motility. The reduction of exposure levels appears to be the main approach to reducing the reproductive issues associated with Cd.

In the framework of sustainability, water shortages and water pollution are two important aspects to be considered. Proposing efficient and low-impact technologies is of paramount importance to promote circular economies associated with the use of water in the industrial context, especially in the textile industry. In this work, the application of a set of magnetic nanostructured adsorbents (MNAs) to cleanse metal ions from textile wastewaters was studied and analyzed. MNAs were generated with a low-cost process, involving iron (II/III) salts (e.g., chlorides), sodium or ammonium hydroxide solutions, and graphene oxide, obtained from graphite by a modified Hummers’ method at room temperature. The shape and the size were studied with transmission electron microscopy. Adsorbents were tested with different metal ions (e.g., copper, chromium (III), and nickel). Metal ion concentrations were analyzed by means of inductively coupled plasma optical emission spectroscopy (ICP-OES), and adsorption isotherms were characterized. From the results, the MNAs exhibited the capability of removing metal ions up to a yield of 99% for Cr3+, 94.7% for Cu2+, and 91.4% for Ni2+, along with adsorption loads up to 4.56 mg/g of MNAs.

In recent decades the definition of standard test methodologies suitable for use in assessing the biological stability of solid waste has become increasingly imperative. To meet this requirement, the BOD5/COD ratio, measured on waste eluate, has been proposed by Cossu et al. (2012) as a more appropriate parameter than the traditional respirometric indices and biogas production measured directly on solid samples. However, to ensure reproducibility, the parameter should undergo standardization of operational conditions. Previous studies have demonstrated that: the testing mode (static or dynamic) does not influence test representativeness; the long testing time (>6 h) does not influence BOD5/COD ratio; COD measured on unfiltered or filtered samples is consistent and significant in both cases. The main aim of this study was to further promote the standardization of this parameter. A series of static leaching tests on representative samples of five types of waste was carried out under different operative conditions: contact time, liquid to solid ratio and pretreatment. The results obtained demonstrate: the apparent adequacy of a short contact time (2 h), which is highly preferable and would speed up the procedure; a low liquid to solid ratio (5 l/kgTS) which is recommended as a water saving strategy; the applicability of centrifugation of the eluate prior to analysis which is faster that filtration method.

Bio-hydrogen, obtained by fermentation of organic residues, is considered a promising source of renewable energy. However, the industrial scale H 2 production from organic waste is far to be realized as technical and economical limitations have still to be solved. Low H 2 yields and lack of industrially robust microbes are the major limiting factors. To look for bacteria with both interesting hydrogen fermentative traits and proper robustness, granular sludge from a brewery full scale Upflow Anaerobic Sludge Blanket (UASB) digester was selected as trove of microbes processing complex substrates. One hundred and twenty bacterial strains, previously isolated from heat-treated granular sludge and genetically identified by 16S rDNA sequencing, were screened for extracellular hydrolytic enzymes on cellulose, hemicellulose, starch, pectin, lipids, protein. The most interesting hydrolytic strains were assessed for their H 2 production from glucose and soluble starch. Two Bacillus sp. strains, namely F2.5 and F2.8, exhibited high H 2 yields and were used as pure culture to convert Organic Fraction of Municipal Solid Waste (OFMSW) into hydrogen. The strains produced up to 61 mL of H 2 per grams of volatile solids and could be considered as good candidates towards the development of industrially relevant H 2 -producing inoculants. This is the first successful application of pure microbial cultures in bio-hydrogen production from OFMSW.

Aerobic and anaerobic indices, such as respirometric index (RI) and biomethane potential production (GB21), can be used in the estimation of the stability of solid waste samples. Previous studies showed different degrees of relationship between BOD₅/COD ratio compared with RI4. Aim of this study is to standardize the parameter BOD₅/COD ratio and to test the methodology under different operating conditions (dynamic or static leaching and leaching duration, 6 and 24-h) keeping constant temperature and liquid/solid ratio (L/S=10 l/kg(TS)), with the introduction of a COD fractioning method. The COD fractioning is based on the differentiation between the soluble fraction (COD(sol)) and the colloidal fraction (COD(coll)) using a flocculation method. The BOD₅/COD and the BOD₅/COD(sol) indices are both consistent and significant and can be used as stability indices. The BOD₅/COD ratio does not seem to be influenced, for the same test duration, by the type of test, static or dynamic. In the same way the longer test duration (24-h) does not influence significantly the values of BOD₅/COD ratio. As a consequence a leaching test duration of 6-h is preferable to avoid the beginning of the hydrolysis and oxidation processes.
