The exertion of self-control is known to result in subsequent detrimental effects on prosocial behaviors. Moreover, certain studies have demonstrated that positive emotions could drive people to allocate more attentional resources for conducting prosocial behaviors. However, whether and how awe – one important type of positive incidental emotion – moderates the effect of exerting self-control on subsequent prosocial behaviors remains unclear yet. The anonymous economic dictator game is an effective index of prosocial behaviors. We examined the influence of exerting self-control on prosocial behavior and the moderating role of awe on the effect of exerting self-control on prosocial behaviors in two experiments (N = 280). We adopted the incongruent Stroop task to induce the exertion of self-control and participants were required to allocate money to others in the anonymous dictator game (Experiment 1). We used the narrative recall task paradigm to elicit the emotion of awe during the interval between Stroop tasks and the dictator game (Experiment 2). Results indicated that the exertion of self-control was detrimental to prosocial behaviors and awe weakened the detrimental effects of exerting self-control on prosocial behavior. We interpreted these results in terms of the protective inhibition of self-regulation and motivation (PRISM) model.

This ethnographic study examines how participation in a Christian church community shapes Chinese international undergraduate students’ social experiences in an American university. Our findings reveal that Chinese international undergraduate students identify the church and its fellowship as (1) a social support community and (2) an informal learning community, one which fills in the gap in counseling services and interpersonal activities that the university fails to offer. Recommendations are made for higher education institutions to provide stronger support for international students, regardless of their nationalities and religions.

A smooth and dense surface of the base paper is advantageous when the goal is to apply a liquid coating as a barrier layer. For such a base paper, non-wood fibers derived from wheat straw could be an alternative to wood fibers. In this research paper, wheat straw pulp was refined with different beating levels (up to 600 revolutions) followed by different calendering pressure loads (up to 50 N/mm) to test its influence on mechanical and surface properties. Alkyl ketene dimer (AKD) was used as sizing agent with concentrations up to 0.2 wt% followed by a mineral-based precoating to test its influence on the smoothness. Eucalyptus pulp was chosen as a benchmark. After beating, the initial Schopper-Riegler degrees of 28 °SR increased to 56 °SR. Beating also increased the tensile index from 24 to 49 Nm/g, the burst index from 1.2 to 2.8 kPa·m²/g, and the tear index decreased from 3.3 to 2.8 mN·m2/g. Calendering reduced the initial roughness of 370 mL/min to 30 mL/min. When precoated and calendered again, the value was lowered to 15 mL/min. In summary, wheat straw paper is a relevant alternative to wood-derived base paper to produce barrier papers. Compared to eucalyptus, wheat straw paper showed better smoothness and much lower air permeability indicating excellent suitability for barrier coating.

Abstract Precisely measuring pressure in microfluidic flows is essential for flow control, fluid characterization, and monitoring, but faces specific challenges such as achieving sufficient resolution, non-invasiveness, or ease of use. Here, we demonstrate a fully integrated multiplexed optofluidic pressure sensor, entirely decoupled from the flow path, that enables local pressure measurements along any microfluidic channel without altering its flow geometry. The sensor itself relies on the compression of a soft mechano-actuated hydrogel, changing color in response to a pressure change. The hydrogel is separated from the fluid circulating in the channel by a thin membrane, allowing for the unrestricted use of different types of fluids. Imaging the gel through the transparent PDMS with a color camera provides a direct, easy, and contact-free determination of the fluid pressure at the sensing location for pressures as small as 20mbar with a resolution of around 10mbar. The sensitivity and accessible pressure range can be tuned via the mechanical properties of the sensing unit. The photonic gel can also be used to acquire 2D pressure or deformation maps, taking advantage of the fast response time and fine spatial resolution.

This paper exposes the role of universities in creating silence around gender-based violence in higher education, drawing on narratives from 39 qualitative interviews with victims/survivors and bystanders about reporting incidents and experiences. In this paper, we extend concept of ‘network silence’ around sexual harassment to other forms of gender-based violence. Our research applies three components of the theoretical model of network silence, namely, self-silencing by victims/survivors, silencing, and not hearing by others, and analyses their contextual manifestations through the reporting experiences of victims/survivors and bystanders. This helps to identify the traits of the informal organisational structures and power dynamics, gendered attitudes, actors, and factors which facilitate silencing. The intersectional approach in our analysis of organisational contextual traits contributes to the research on inequality regimes in universities. The findings suggest that universities are making limited efforts to address silence around gender-based violence. We conclude that shared beliefs among the leadership about the reputation and prestige of the university facilitate the endurance of silence in universities. Our findings indicate reasons why universities fail to create spaces that are safe from gender-based violence.

Immunosorbent turnip vein clearing virus (TVCV) particles displaying the IgG-binding domains D and E of Staphylococcus aureus protein A (PA) on every coat protein (CP) subunit (TVCVPA) were purified from plants via optimized and new protocols. The latter used polyethylene glycol (PEG) raw precipitates, from which virions were selectively re-solubilized in reverse PEG concentration gradients. This procedure improved the integrity of both TVCVPA and the wild-type subgroup 3 tobamovirus. TVCVPA could be loaded with more than 500 IgGs per virion, which mediated the immunocapture of fluorescent dyes, GFP, and active enzymes. Bi-enzyme ensembles of cooperating glucose oxidase and horseradish peroxidase were tethered together on the TVCVPA carriers via a single antibody type, with one enzyme conjugated chemically to its Fc region, and the other one bound as a target, yielding synthetic multi-enzyme complexes. In microtiter plates, the TVCVPA-displayed sugar-sensing system possessed a considerably increased reusability upon repeated testing, compared to the IgG-bound enzyme pair in the absence of the virus. A high coverage of the viral adapters was also achieved on Ta2O5 sensor chip surfaces coated with a polyelectrolyte interlayer, as a prerequisite for durable TVCVPA-assisted electrochemical biosensing via modularly IgG-assembled sensor enzymes.

Abstract Background How intestinal epithelial cells interact with the microbiota and how this is regulated at the gene expression level are critical questions. Smarcad1 is a conserved chromatin remodeling factor with a poorly understood tissue function. As this factor is highly expressed in the stem and proliferative zones of the intestinal epithelium, we explore its role in this tissue. Results Specific deletion of Smarcad1 in the mouse intestinal epithelium leads to colitis resistance and substantial changes in gene expression, including a striking increase of expression of several genes linked to innate immunity. Absence of Smarcad1 leads to changes in chromatin accessibility and significant changes in histone H3K9me3 over many sites, including genes that are differentially regulated upon Smarcad1 deletion. We identify candidate members of the gut microbiome that elicit a Smarcad1-dependent colitis response, including members of the poorly understood TM7 phylum. Conclusions Our study sheds light onto the role of the chromatin remodeling machinery in intestinal epithelial cells in the colitis response and shows how a highly conserved chromatin remodeling factor has a distinct role in anti-microbial defense. This work highlights the importance of the intestinal epithelium in the colitis response and the potential of microbial species as pharmacological and probiotic targets in the context of inflammatory diseases.

Teacher expertise research uses different approaches, among them competence models. The model of mathematics-specific professional competence in this study differentiates action-related and reflective competence, which pertain to different types of professional demands according to dual processing theories and should positively impact teaching quality. The cross-sectional study with early childhood teachers investigates the structure of teacher competence and its relations to the quality of learning support in line with the notion of adaptive teaching when playing board games. The findings support a differentiation of competences according to the model (structural validity evidence). Expected relations between competences and the quality of the learning support are partially supported (predictive validity evidence). The study hence adds to research on teacher expertise by developing a specific understanding of competence in line with existing theories. Affordances of using competence models, as well as difficulties and open questions with relation to the emerging approach are discussed.

Bacteria can swim upstream by reorienting with respect to fluid flows. Here, the authors observe an oscillatory motion and describe the mechanisms behind these reorientation dynamics, which could help designing strategies for bacterial contamination prevention.

Background & Aims: Hepatocellular carcinoma (HCC) typically arises in fibrotic or cirrhotic livers, which are characterized by pathogenic angiogenesis. Myeloid immune cells, specifically tumor-associated macrophages (TAMs), may represent potential novel therapeutic targets in HCC, complementing current ablative or immune therapies. However, the detailed functions of TAM subsets in hepatocarcinogenesis have remained obscure. Methods: TAM subsets were analyzed in-depth in human HCC samples and a combined fibrosis–HCC mouse model, established by i.p. injection with diethylnitrosamine after birth and repetitive carbon tetrachloride (CCl4) treatment for 16 weeks. Based on comprehensively phenotyping TAM subsets (fluorescence-activated cell sorter, transcriptomics) in mice, the function of CCR2+ TAM was assessed by a pharmacologic chemokine inhibitor. Angiogenesis was evaluated by contrast-enhanced micro–computed tomography and histology. Results: We show that human CCR2+ TAM accumulate at the highly vascularized HCC border and express the inflammatory marker S100A9, whereas CD163+ immune-suppressive TAM accrue in the HCC center. In the fibrosis–cancer mouse model, we identified 3 major hepatic myeloid cell populations with distinct messenger RNA profiles, of which CCR2+ TAM particularly showed activated inflammatory and angiogenic pathways. Inhibiting CCR2+ TAM infiltration using a pharmacologic chemokine CCL2 antagonist in the fibrosis–HCC model significantly reduced pathogenic vascularization and hepatic blood volume, alongside attenuated tumor volume. Conclusions: The HCC microenvironment in human patients and mice is characterized by functionally distinct macrophage populations, of which the CCR2+ inflammatory TAM subset has pro-angiogenic properties. Understanding the functional differentiation of myeloid cell subsets in chronically inflamed liver may provide novel opportunities for modulating hepatic macrophages to inhibit tumor-promoting pathogenic angiogenesis. Objective To assess satisfaction with the effectiveness and tolerability of treatments in patients with rheumatoid arthritis (RA).Methods Patients from the RABBIT register, starting a biological (b) or targeted synthetic (ts) disease-modifying antirheumatic drug (DMARD), or a conventional synthetic (cs)DMARD treatment after ≥1 csDMARD failure, were included. Treatment satisfaction was measured after 1 year of treatment in four categories and binarised for analysis. Logistic regression models were performed to calculate ORs for factors associated with treatment satisfaction.Results Data of 10 646 patients (74% women, mean 58 years) were analysed. At baseline, 55% of the patients were satisfied with the efficacy and 68% with the tolerability of their previously given treatments. After 1 year, 85% of the patients were satisfied with treatment effectiveness and 90% with tolerability. Baseline satisfaction (OR 2.98, 95% CI 2.58 to 3.44), seropositivity (OR 1.36, 95% CI 1.17 to 1.57), reduction of DAS28 (OR 1.38, 95% CI 1.31 to 1.46) and pain (OR 1.26, 95% CI 1.22 to 1.31), and the improvement of physical capacity (OR 1.22, 95% CI 1.17 to 1.29) were positively associated with treatment satisfaction at follow-up while glucocorticoids (GCs) >5 mg/day, depression, fibromyalgia, obesity, prior bDMARDs and therapy changes were negatively associated. The impact of GC on satisfaction was dose-dependent, becoming strongest for GC >15 mg (OR 0.24, 95% CI 0.16 to 0.34). A 5 mg/day reduction within 12 months was positively associated with satisfaction regarding efficacy (OR 1.19, 95% CI 1.11 to 1.27) and tolerability (OR 1.11, 95% CI 1.03 to 1.21).Conclusion Most patients were satisfied with their treatment’s effectiveness and tolerability after 1 year of treatment. Tapering GCs was positively associated with the improvement of patients’ satisfaction.

Histone post-translational modifications are known key regulators of gene expression. Here, the authors characterize histone crotonylation at histone H3 lysine 18 in intestinal epithelia and find that it is a highly dynamic cell cycle regulated mark under the regulation of the HDAC deacetylases.

Unraveling bacterial strategies for spatial exploration is crucial for understanding the complexity in the organization of life. Bacterial motility determines the spatiotemporal structure of microbial and controls infection spreading and the microbiota organization in guts or in soils. Most theoretical approaches for modeling bacterial transport rely on their run-and-tumble motion. For Escherichia coli, the run-time distribution is reported to follow a Poisson process with a single characteristic time related to the rotational switching of the flagellar motors. However, direct measurements on flagellar motors show heavy-tailed distributions of rotation times stemming from the intrinsic noise in the chemotactic mechanism. Currently, there is no direct experimental evidence that the stochasticity in the chemotactic machinery affects the macroscopic motility of bacteria. In stark contrast with the accepted vision of run and tumble, here we report a large behavioral variability of wild-type E. coli, revealed in their three-dimensional trajectories. At short observation times, a large distribution of run times is measured on a population and attributed to the slow fluctuations of a signaling protein triggering the flagellar motor reversal. Over long times, individual bacteria undergo significant changes in motility. We demonstrate that such a large distribution of run times introduces measurement biases in most practical situations. Our results reconcile the notorious conundrum between run-time observations and motor-switching statistics. We finally propose that statistical modeling of transport properties, currently undertaken in the emerging framework of active matter studies, should be reconsidered under the scope of this large variability of motility features.

Being able to precisely characterize the mechanical properties of soft microparticles is essential for numerous situations, from the understanding of the flow of biological fluids to the development of soft micro-robots. Here, we present a simple measurement technique for determining Poisson’s ratio of soft micron-sized hydrogels in the presence of a surrounding liquid. This method relies on the measurement of the deformation, in two orthogonal directions, of a rectangular hydrogel slab compressed uni-axially inside a microfluidic channel. Due to the in situ character of the method, the sample does not need to be dried, allowing for the measurement of the mechanical properties of swollen hydrogels. Using this method, we determined Poisson’s ratio of hydrogel particles composed of polyethylene glycol (PEG) and varying solvents fabricated using a lithography technique. The results demonstrate, with high precision, the dependence of the hydrogel compressibility on the solvent fraction and character. The method is easy to implement and can be adapted for the measurement of a variety of soft and biological materials.

Critical illness and sepsis are characterized by drastic changes in the systemic innate immune response, particularly involving monocytes. The exact monocyte activation profile during sepsis, however, has remained obscure. Therefore, we prospectively analyzed the gene expression profile of circulating CD14+ monocytes from healthy volunteers (n = 54) and intensive care unit (ICU) patients (n = 76), of which n = 36 had sepsis. RNA sequencing of selected samples revealed that monocytes from septic ICU patients display a peculiar activation pattern, which resembles characteristic functional stages of monocyte-derived macrophages and is distinct from controls or non-sepsis ICU patients. Focusing on 55 highly variable genes selected for further investigation, arachidonate 5-lipoxygenase-activating protein (ALOX5AP) was highly upregulated in monocytes of ICU patients and only normalized during 7 days in the ICU in non-sepsis patients. Strikingly, low monocytic guanine nucleotide exchange factor 10-like protein (ARHGEF10L) mRNA expression was associated with the disease severity and mortality of ICU patients. Collectively, our comprehensive analysis of circulating monocytes in critically ill patients revealed a distinct activation pattern, particularly in ICU patients with sepsis. The association with disease severity, the longitudinal recovery or lack thereof during the ICU stay, and the association with prognosis indicate the clinical relevance of monocytic gene expression profiles during sepsis.

Background: The Constitution of South Africa stipulates that all children have the right to basic nutrition; however, a great number of South African children are underweight for age. It is important to address malnutrition as it is associated with more than 50% of all child deaths in developing countries and also increases the risk for infective diseases. Aim: To determine the prevalence of underweight in children aged 5 years and younger attending primary health care clinics in the Mangaung area, Free State, and determine the possible underlying causes thereof. Setting: Six preselected primary health care clinics in the Mangaung area. Methods: This was a cross-sectional study. Demographic and clinical information and anthropometric measurements were collected from the children’s Road-to-Health clinic cards,obtained from the children’s caregivers. Results: In total, 240 children were included, of which 51.7% were girls. The median age was 7.5 months. The weight-for-age graph revealed that 7.7% (95% confidence interval: 4.8%;11.9%) of children were underweight or severely underweight for age. Length-for-age and weight-for-height graphs were mostly incomplete. Underweight children differed from normal weight children regarding birth weight (low birth weight 70.6% vs. 12.4%) and history of malnutrition (60.0% vs. 7.1%). Conclusion: The prevalence of underweight in children aged 0–5 years attending primary health care clinics in Mangaung is 7.7% based on information available from Road-to-Health cards. This figure could be higher if these cards were filled in more accurately. A low birth weight and history of malnutrition are associated with underweight.

Recent evidence suggests that hepatic dendritic cells (HDCs) contribute to the evolution of chronic liver diseases. However, the HDC subsets involved and the mechanisms driving these responses are still poorly understood. In this study, we have investigated the role of the fractalkine receptor CX3CR1 in modulating monocyte-derived dendritic cell (moDC) differentiation during liver inflammation. The phenotype of HDC and functional relevance of CX3CR1 was assessed in mice following necro-inflammatory liver injury induced by the hepatotoxic agent carbon tetrachloride (CCl4) and in steatohepatitis caused by a methionine/choline-deficient (MCD) diet. In both the experimental models, hepatic inflammation was associated with a massive expansion of CD11c+/MHCIIhigh/CD11b+ myeloid HDCs. These cells also expressed the monocyte markers Ly6C, chemokine (C-C Motif) receptor 2 (CCR2), F4/80 and CD88, along with CX3CR1, allowing their tentative identification as moDCs. Mice defective in CX3CR1 showed a reduction in liver-moDC recruitment following CCl4 poisoning in parallel with a defective maturation of monocytes into moDCs. The lack of CX3CR1 also affected moDC differentiation from bone marrow myeloid cells induced by granulocyte-macrophage colony stimulating factor (GM-CSF) and interleukin-4 (IL-4) in vitro. In wild-type mice, treatment with the CX3CR1 antagonist CX3-AT (150 µg, i.p.) 24 h after CCl4 administration reduced liver moDCS and significantly ameliorated hepatic injury and inflammation. Altogether, these results highlight the possible involvement of moDCs in promoting hepatic inflammation following liver injury and indicated a novel role of CX3CL1/CX3CR1 dyad in driving the differentiation of hepatic moDCs.

BACKGROUND:Ecstasy use has been associated with short-term and long-term memory deficits on a standard Word Learning Task (WLT). The clinical relevance of this has been debated and is currently unknown. The present study aimed at evaluating the clinical relevance of verbal memory impairment in Ecstasy users. To that end, clinical memory impairment was defined as decrement in memory performance that exceeded the cut-off value of 1.5 times the standard deviation of the average score in the healthy control sample. The primary question was whether being an Ecstasy user (E-user) was predictive of having clinically deficient memory performance compared to a healthy control group. METHODS:WLT data were pooled from four experimental MDMA studies that compared memory performance during placebo and MDMA intoxication. Control data were taken from healthy volunteers with no drug use history who completed the WLT as part of a placebo-controlled clinical trial. This resulted in a sample size of 65 E-users and 65 age- and gender-matched healthy drug-naïve controls. All participants were recruited by similar means and were tested at the same testing facilities using identical standard operating procedures. Data were analyzed using linear mixed-effects models, Bayes factor, and logistic regressions. RESULTS:Findings were that verbal memory performance of placebo-treated E-users did not differ from that of controls, and there was substantial evidence in favor of the null hypothesis. History of use was not predictive of memory impairment. During MDMA intoxication of E-users, verbal memory was impaired. CONCLUSION:The combination of the acute and long-term findings demonstrates that, while clinically relevant memory impairment is present during intoxication, it is absent during abstinence. This suggests that use of Ecstasy/MDMA does not lead to clinically deficient memory performance in the long term. Additionally, it has to be investigated whether the current findings apply to more complex cognitive measures in diverse 'user categories' using a combination of genetics, imaging techniques and neuropsychological assessments.

Fibers are widely used in different industrial processes, for example in paper manufacturing or lost circulation problems in the oil industry. Recently, interest towards the use of fibers at the microscale has grown, driven by research in bio-medical applications or drug delivery systems. Microfluidic systems are not only directly relevant for lab-on-chip applications, but have also proven to be good model systems to tackle fundamental questions about the flow of fiber suspensions. It has therefore become necessary to provide fiber-like particles with an excellent control of their properties. We present here two complementary in situ methods to fabricate controlled micro-fibers allowing for an embedded fabrication and flow-on-a-chip platform. The first one, based on a photo-lithography principle, can be used to make isolated fibers and dilute fiber suspensions at specific locations of interest inside a microchannel. The self-assembly property of super-paramagnetic colloids is the principle of the second fabrication method, which enables the fabrication of concentrated suspensions of more flexible fibers. We propose a flow gallery with several examples of fiber flow illustrating the two methods’ capabilities and a range of recent laminar flow results.
