Man-made persistent pollutants (such as PCBs, pesticides and trace metals) reach aquatic organisms through the food chains. Pollutants are ingested and assimilated by smaller organisms, and their concentration in tissues increases from prey to predators. Being at the top of the food chains, marine mammals accumulate some of the highest environmental contaminant levels of all wildlife. They are good sentinel species for monitoring long-term environmental pollution. Exposure to contaminants may have large consequences, both on an individual and a population level. The prevalence and severity of diseases of aquatic wildlife has recently increased in many species. Scientists use new methods to understand how pollutants affect the immune system of marine mammals. Learning about contaminants may also contribute to our understanding of outbreaks of infectious diseases in marine mammals.

Bioacoustics combines the fields of biology and acoustics to answer questions about hearing, sound production and sound communication in animals. Marine mammals have specialised hearing abilities and use sounds in different ways underwater. How do whales and seals use sound for communication and to find prey? How are they affected by human-made sounds from ships, oil exploration and windfarms? To answer such questions, you need to study marine mammals, be well-trained in natural sciences and know about animal anatomy, physiology and behaviour. You also need a thorough understanding of the fundamentals of acoustics, maths and physics. Bioacoustics is a truly interdisciplinary research field involving biologists, physicists and engineers trying to understand the world of biological sound, how sounds are produced and used by animals. Additionally, underwater acoustic recordings can reveal which areas animals use during different seasons. Bioacoustics can also be used to improve wildlife protection by regulating damaging sound sources in marine mammal habitats. Using the exercises at the end of this chapter, students learn about frequencies, decibels and their own hearing abilities, as well as how to build their own underwater microphone.

There are various types of interactions between marine mammals and fisheries, and their presence in the same area inevitably leads to conflicts. Marine mammals may lose their lives as bycatch, which is the incidental catch of non-target species in fishing gear. On the other hand, some marine mammals specialise in feeding on fish caught in fishing gear, resulting in damaged and reduced catch as well as destroyed fishing gear. There are different methods to reduce bycatch and catch damage, such as reducing fishing efforts, using acoustic deterrent devices, employing temporal and zonal closure of fishing areas and using alternative fishing gear. Here we give an overview of whaling and sealing from historical and present perspectives, with examples for Baltic marine mammals. We present hands-on activities to familiarise students and teachers with investigations of prey remains, such as otoliths (fish ear stones) found in faeces, to learn about marine mammal diet. Additionally, we introduce a whaling role play designed as a debate between interest groups, to better understand whaling from different perspectives.

Many young students dream about a career working with marine mammals. Although marine mammal jobs are limited and highly competitive, there are several ways to obtaining them. But, there is no magic formula to pursue a career in marine mammal science. We describe various skill sets and experiences that can improve your chances. Not the least, it is important to actively work towards your career goals and believe in your strengths. We interview marine mammal researchers to inspire students to follow their passion and pursue a career in natural sciences, which may lead to work on marine mammals. In times of climate change, pollution, habitat and biodiversity loss, there is a huge need for students interested in science, technology, biology, engineering and mathematics, to provoke a general change for the better.

Simple Summary The progress of animal welfare in wildlife conservation and research calls for more non-invasive sampling techniques. In cetaceans, exhaled breath condensate (blow)—a mixture of cells, mucus and fluids expelled through the force of a whale’s exhale—is a unique sampling matrix for hormones, bacteria and genetic material, among others. Especially the detection of steroid hormones, such as cortisol, is being investigated as stress indicators in several species. As the only native cetacean in Germany, harbor porpoises (Phocoena phocoena) are of special conservation concern and research interest. So far, strandings and live captures have been the only method to obtain samples from free-ranging individuals, and novel, non-invasive monitoring methods are desirable for this small cetacean species. Hence, three different blow collection devices were tested on harbor porpoises. All samples were analyzed for cortisol using a commercially available immunosorbent assay. The most suitable protocol for sampling, storage and processing is using a sterile 50 mL centrifuge tube. This pilot study shows that cortisol can be detected in the exhale of harbor porpoises, thus paving the way for future studies and most likely successful non-invasive small cetacean health monitoring through blow. Abstract Over the last decades, exhaled breath sampling has been established for laboratory analysis in various cetacean species. Due to their small size, the usability of respiratory vapor for hormone assessments was questionable in harbor porpoises (Phocoena phocoena). This pilot study compared three different blow collection devices for their suitability in the field and during laboratory processing: a sterile petri dish covered by a Nitex membrane, as well as sterile 50 mL centrifuge tubes with or without manganese(II) chloride as a stabilizer. Collected exhales varied between three, five or ten, depending on feasibility. Hormones were extracted through an ether mix, followed by centrifugal evaporation and cortisol analysis using an immunoassay. Although close to the lower end of the assay’s dynamic range, the ELISA produced results (n = 110, 0.102–0.937 ng/mL). Hence, a simple 50 mL centrifuge tube was determined as the best suited blow collection device, while three consecutive exhales proved sufficient to yield results. These findings are promising regarding the suitability of exhaled breath as a matrix for future endocrine and immune system-related studies in harbor porpoises. If further advanced, blow sampling can become an important, non-invasive tool for studying and monitoring health, stress levels and diseases in harbor porpoises.

Tapeworms of the genus Mesocestoides (Cestoda: Cyclophyllidea: Mesocestoididae) are still enigmatic to scientists, due to their high morphological variability, low host specificity, and unknown details of their life cycle. They are found worldwide, with carnivorous mammals as the main definitive hosts, and the disease is potentially zoonotic. After ingestion by a definitive host, the tetrathyridium can occasionally migrate through the intestinal wall and reach the peritoneal cavity or abdominal organs causing peritoneal metacestodosis. Here, we report on a case of metacestodosis of a European wild cat (Felis silvestris silvestris) found dead in Croatia. At necropsy, a large number of white, rice-like structures were found free in the abdominal and thoracic cavities, as well as along the serous surfaces and in the lungs. DNA isolated from the nodules was genotyped and based on a 320-base pair long 12S fragment classified as Mesocestoides vogae. Although post-mortem changes were advanced, severe emaciation due to the severe parasitic infection and gastrointestinal bleeding was diagnosed as the likely cause of death. Intestinal cestodosis was previously reported in wild cats, but according to our knowledge, this is the first description of peritoneal and pleural metacestodosis caused by M. vogae tetrathyridia (metacestodes) in any wild carnivore species.

Acute-phase reactants indicate inflammation and are increasingly used in veterinary medicine to indicate and to monitor progression of disease. Hemostasis and inflammation have interconnected pathophysiologic pathways and influence each other on different levels. This study established observed normal ranges for acute-phase reactants and for coagulation and thromboelastographic (TEG) parameters in 49 dromedary camels ( Camelus dromedarius) and assessed the response to chronic and acute inflammation. Chronically infected animals suffering from lymph abscessation due to Corynebacterium spp. had significantly higher concentrations of the acute-phase reactants haptoglobin ( P0.005) and fibrinogen ( P0.013) and an increased clot strength characterized by an increase of the TEG parameters MA ( P0.039), representing the maximum amplitude of the clot strengths, and G, the global clot strength ( P0.022), compared to healthy animals. When the acute-phase and hemostatic responses of 10 males receiving a gonadotropin-releasing hormone vaccine and of 9 males that were surgically castrated over 7 days were studied, haptoglobin proved to be a minor positive acute-phase protein, with moderate levels in healthy animals. It increased significantly after both vaccination and castration and remained elevated 7 days postinsult. The negative reactant iron significantly decreased over the 7-day period after castration, whereas a similar decrease following vaccination lasted less than 3 days. Fibrinogen reacted as a positive, minor reactant, with a significant increase and a peak on days 3-5, with higher values seen after castration. Prothrombin time showed a slight shortening at days 5-7, and the TEG parameters MA and G showed significantly increased values, similar to fibrinogen. The acute-phase protein serum amyloid A showed poor repeatability, suggesting that the assay was not reliable.

Orca (Orcinus orca) strandings are rare and post-mortem examinations on fresh individuals are scarce. Thus, little is known about their parasitological fauna, prevalence of infections, associated pathology and the impact on their health. During post-mortem examinations of two male neonatal orcas stranded in Germany and Norway, lungworm infections were found within the bronchi of both individuals. The nematodes were identified as Halocercus sp. (Pseudaliidae), which have been described in the respiratory tract of multiple odontocete species, but not yet in orcas. The life cycle and transmission pathways of some pseudaliid nematodes are incompletely understood. Lungworm infections in neonatal cetaceans are an unusual finding and thus seem to be an indicator for direct mother-to-calf transmission (transplacental or transmammary) of Halocercus sp. nematodes in orcas.

A recent Science report predicted the global killer whale population to collapse due to PCB pollution. Here we present empirical evidence, which supports and extends the reports' statement. In 2016, a neonate male killer whale stranded on the German island of Sylt. Neonatal attributes indicated an age of at least 3 days. The stomach contained similar to 20 mL milk residue and no pathologies explaining the cause of death could be detected. Blubber samples presenting low lipid concentrations were analysed for persistent organic pollutants. Skin samples were collected for genotyping of the mitochondrial control region. The blubber PCB concentrations were very high [SPCBs, 225 mg/kg lipid weight (lw)], largely exceeding the PCB toxicity thresholds reported for the onset of immunosuppression [9 mg/kg lw Sigma PCB] and for severe reproductive impairment [41 mg/kg lw Sigma PCB] reported for marine mammals. Additionally, this individual showed equally high concentrations in p,p'-DDE [226 mg/kg lw], PBDEs [5 mg/kg lw] and liver mercury levels [1.1 mu g/g dry weight dw]. These results suggest a high placental transfer of pollutants from mother to foetus. Consequently, blubber and plasma PCB concentrations and calf mortality rates are both high in primiparous females. With such high pollutant levels, this neonate had poor prerequisites for survival. The neonate belonged to Ecotype I (generalist feeder) and carried the mitochondrial haplotype 35 present in about 16% of the North Atlantic killer whale from or close to the North Sea. The relevance of this data becomes apparent in the UK West Coast Community, the UK's only residentorca population, which is currently composed of only eight individuals (each four males and females) and no calves have been reported over the last 19 years.Despite worldwide regulations, PCBs persist in the environment and remain a severe concern for killer whale populations, placing calves at high risk due to the mother-offspring PCB-transfer resulting in a high toxicological burden of the neonates.

In case of apnea, arterial partial pressure of oxygen (pO2) decreases, while partial pressure of carbon dioxide (pCO2) increases. To avoid damage to hypoxia sensitive organs such as the brain, compensatory circulatory mechanisms help to maintain an adequate oxygen supply. This is mainly achieved by increased cerebral blood flow. Intermittent hypoxia is a commonly seen phenomenon in patients with obstructive sleep apnea. Acute airway obstruction can also result in hypoxia and hypercapnia. Until now, no adequate model has been established to simulate these dynamics in humans. Previous investigations focusing on human hypoxia used inhaled hypoxic gas mixtures. However, the resulting hypoxia was combined with hyperventilation and is therefore more representative of high altitude environments than of apnea. Furthermore, the transferability of previously performed animal experiments to humans is limited and the pathophysiological background of apnea induced physiological changes is poorly understood. In this study, healthy human apneic divers were utilized to mimic clinically relevant hypoxia and hypercapnia during apnea. Additionally, pulse-oximetry and Near Infrared Spectroscopy (NIRS) were used to evaluate changes in cerebral and peripheral oxygen saturation before, during, and after apnea.

A cold-stunned, sub-adult, female loggerhead sea turtle (Caretta caretta) was found stranded in December 2012 on Cape Cod, Massachusetts. The turtle was treated routinely; however, she did not gain weight as expected during rehabilitation. Eleven weeks after stranding, staff noticed stertorous upper airway sounds. Oral examination revealed a unilateral laryngeal paralysis-like condition resulting in partial airway obstruction. Magnetic resonance imaging and electromyography showed abnormalities of the abductor arytenoideae muscle. Muscle and hepatic biopsies were collected for histologic and toxicologic assessment to seek an underlying cause of the disorder. The turtle did not recover from anesthesia, and necropsy revealed a large amount of hemorrhage from the liver biopsy site. Unilateral myofiber atrophy of the abductor arytenoideae muscle was the predominant histological finding.
