Hypothesis. Isoniazid is one of the primary drugs used in tuberculosis treatment. Isoniazid encapsulation in liposomal vesicles can improve drug therapeutic index and minimize toxic and side effects. In this work, we consider mixtures of hydrogenated soy phosphatidylcholine/phosphatidylglycerol (HSPC/DPPG) to get novel biocompatible liposomes for isoniazid pulmonary delivery. Our goal is to understand if the entrapped drug affects bilayer structure. Experiments. HSPC-DPPG unilamellar liposomes are prepared and characterized by dynamic light scattering, $\zeta$-potential, fluorescence anisotropy and Transmission Electron Microscopy. Isoniazid encapsulation is determined by UV and Laser Transmission Spectroscopy. Calorimetry, light scattering and Surface Pressure measurements are used to get insight on adsorption and thermodynamic properties of lipid bilayers in the presence of the drug. Findings. We find that INH-lipid interaction can increase the entrapment capability of the carrier due to isoniazid adsorption. The preferential INH-HSPC dipole-dipole interaction promotes modification of lipid packing and ordering and favors the condensation of a HSPC-richer phase in molar excess of DPPG. It is widely acknowledged that mammalian exosomes (or extracellular vesicles), have a key role in intercellular communication, owing to the presence of various bioactive molecules such as lipids, proteins, and microRNAs within their inner compartment. Most recently, the discovery of extracellular vesicles isolated from edible plants (such as vegetables and fruits) and their similarity in terms of size and content with exosomes has opened new perspectives on possible intercellular communication and regulation of important biological processes in which these vesicles are involved. It is also well-known that a balanced diet rich of fruits and vegetables (i.e., the Mediterranean diet) can contribute to maintain a “healthy gut” by preserving the intestinal epithelial barrier integrity and avoid that inflammatory stimuli that can alter homeostasis. In our study, we optimized a method to isolate extracellular vesicles from the orange juice (Citrus sinensis) (CS-EVs), and we characterized their morphology and behavior when in contact with the intestinal epithelium. We showed that CS-EVs are stable in a simulated gastrointestinal environment and are absorbed by intestinal cells without toxic effects, as expected. Furthermore, we demonstrated that CS-EVs can alter the gene expression of several genes involved in inflammation (i.e., ICAM1 and HMOX-1) and tight junctions (i.e., OCLN, CLDN1, and MLCK), contributing to limit inflammatory stimuli and restore a functional barrier by increasing the tight junction OCLN protein. Therefore, our study emphasizes the relevant role of fruit-derived extracellular vesicles in modulating important biological processes and maintaining a healthy intestinal epithelium, ultimately promoting human health and well-being.

Size and absolute concentration of suspensions of nanoparticles are important information for the study and development of new materials and products in different industrial applications spanning from biotechnology and pharmaceutics to food preparation and conservation. Laser Transmission Spectroscopy (LTS) is the only methodology able to measure nanoparticle size and concentration by performing a single measurement. In this paper we report on a new variable gain calibration procedure for LTS-based instruments allowing to decrease of an order of magnitude the experimental indetermination of the particle size respect to the conventional LTS based on the double ratio technique. The variable gain calibration procedure makes use of a specifically designed tunable-gain, dual-channel, dual-phase Lock-In Amplifier (LIA) whose input voltage signals are those ones generated by two Si photodiodes that measure the laser beam intensities passing through the sample containing the nanoparticles and a reference optical path. The LTS variable gain calibration procedure has been validated by firstly using a suspension of NIST standard polystyrene nanoparticles even 36 hours after the calibration procedure was accomplished. The paper reports in detail the LIA implementation describing the design methodologies and the electronic circuits. As a case example of the characterization of biological nanostructures, we demonstrate that a single LTS measurement allowed to determine size density distribution of a population of extracellular vesicles extracted from orange juice (25 nm in size) with the presence of their aggregates having a size of 340 nm and a concentration smaller than 3 orders of magnitude.

Exosome-like nanoparticles (ELNs) are attracting interest as important vehicles of intercellular communication, both in prokaryotes and eukaryotes. Recently, dietary nanoparticles similar to mammalian exosomes have attracted attention for these features. In particular they appear to be relevant in the modulation of several cellular processes as well as candidate carriers of bioactive molecules (proteins, lipids, and nucleic acids, including miRNAs) with therapeutic value. Herein, we investigated the cellular uptake of blueberry-derived ELNs (B-ELNs) by a human stabilized endothelial cell line (EA.hy926) and the ability of B-ELNs to modulate the expression of inflammatory genes as the response of tumor necrosis factor-&alpha, (TNF-&alpha, ). Our results indicate that 1) EA.hy926 cells internalize B-ELNs in a dose-dependent manner, 2) pretreatment with B-ELNs counters TNF-&alpha, induced reactive oxygen species (ROS) generation and loss of cell viability and modulates the differential expression of 29 genes (fold change &gt, 1.5) induced by TNF-&alpha, compared to control, 3) pathway analysis reveals their involvement in a total of 340 canonical pathways, 121 KEGG pathways, and 121 GO Biological processes, and 4) the intersection between differentially expressed (DE) genes and miRNAs contained in B-ELNs unveils a set of candidate target genes, such as prostaglandin I2 synthase (PTGIS), mitogen-activated protein kinase 14 (MAPK14), and phosphodiesterase 7A (PDE7A), for ELNs-contained cargo. In conclusion, our study indicates that B-ELNs can be considered candidate therapeutic carriers of bioactive compounds potentially able to protect vascular system against various stressors.

Dynamic Light Scattering (DLS), Small Angle X-ray Scattering (SAXS) and Transmission Electron Microscopy (TEM) are physical techniques widely employed to characterize the morphology and the structure of vesicles such as liposomes or human extracellular vesicles (exosomes). Bacterial extracellular vesicles are similar in size to human exosomes, although their function and membrane properties have not been elucidated in such detail as in the case of exosomes. Here, we applied the above cited techniques, in synergy with the thermotropic characterization of the vesicles lipid membrane using a turbidimetric technique to the study of vesicles produced by Gram-negative bacteria (Outer Membrane Vesicles, OMVs) grown at different temperatures. This study demonstrated that our combined approach is useful to discriminate vesicles of different origin or coming from bacteria cultured under different experimental conditions. We envisage that in a near future the techniques employed in our work will be further implemented to discriminate complex mixtures of bacterial vesicles, thus showing great promises for biomedical or diagnostic applications.

Deep Eutectic Solvents (DESs) are gathering growing attention as sustainable solvents in different fields ranging from synthesis to electrochemical devices. Albeit DES are widely employed, the fundamental knowledge on their interspecies relations is still partial. Understanding the chemistry of this class of green solvent is an essential step to further tune their properties. In this study, we report a detailed experimental and theoretical investigation of two choline-chloride glycerol mixtures using electrochemical, spectroscopic (Raman/Far Infrared), diffraction (X-ray) and molecular simulation methods. Remarkably different Far-infrared spectra have been surprisingly collected for the two mixtures. Differences are attributed to the deconstruction of the extended hydrogen bond network characteristic of pure glycerol and of the 1:2 mixture and absent in the glycerol-richer mixture 1:3. From the analysis of X-ray profiles, that are very well reproduced by molecular dynamics, it was found that the lack of the glycerol H-bond interactions in the glycerol-richer mixture (Ch:Gly 1:3) can be attributed to the establishment of a full coordination shell of polyalcohol molecules around the chloride anion. In the 1:2 composition the coordination is probably defective, as signaled by the FIR spectrum that significantly maintains the features observed for the precursor glycerol, and the chloride stabilization is ensured by interaction with both choline hydroxyl and electrostatic interactions.

In the last years, significant research efforts have been devoted to the reduction of the environmental impact of biocides and to the improvement of their effectiveness over time against degradation of stone artifacts. This work reports the spectroscopic characterization of two silica nanosystems, with different geometry and structure, loaded with a biologically-active compound, the 2-mercaptobenzothiazole (MBT), for controlled antifouling release on the surface of outdoor stone artefacts. We have combined FT-IR spectroscopy in ATR mode and Raman spectroscopy to investigate the biocide loading mechanism in both nanosystems from the physical and chemical point of view. These techniques demonstrate that the geometry of confinement influences the loading of the nanoparticles and their interaction with the confining medium, with possible consequences in the biocide release rate. Moreover, we have seen that the biocide interacts with the surfactant (cetrimonium bromide, CTAB) and tends to dimerize.

The Protic Ionic Liquid (PIL) formed by neutralization of the super-strong base 1,7-diazabicyclo[5.4.0]undec-7-ene (DBU) with the super-strong acid bis(trifluoromethanesulfonyl)-imide (TFSI), indicated as [DBUH][TFSI], has been investigated. Its chemical physical properties and structural features have been explored using a synergy of experimental and computational tools. Molecular Dynamics-rationalised X-ray diffraction patterns highlight the major role played by hydrogen bonding (HB) in affecting morphology in this PIL. A comparison between HB features in this and in related PILs has been presented, on the base of far-IR experiments and DFT analysis. Indications of a weaker HB interaction in [DBUH][TFSI] in comparison with [DBUH][TfO], consistently with their ΔpKa difference, have been observed and rationalised in terms of geometrical properties of the main conformers contributing to the experimental spectra. In the liquid phase of [DBUH][TFSI] a particularly large conformational disorder is observed and the corresponding large dispersion of the frequencies of the HB stretching modes leads to a broad absorption band without a well defined peak. On the contrary, well detectable HB related absorptions are observable in the solid phase of [DBUH][TFSI] and at all temperatures in [DBU][TfO], where less configurational disorder occurs.

The programmable assembly of DNA strands is a promising tool for building tailored bottom-up nanostructures. Here, we present a plasmonic nanosystem obtained by the base-pairing mediated aggregation of gold nanoparticles (NPs) which are separately functionalized with two different single-stranded DNA chains. Their controlled assembly is mediated by a complementary DNA “bridge” sequence. We monitor the formation of DNA assembled NP aggregates in solution, and we study their Surface Enhanced Raman Scattering (SERS) response by comparison with the single NP constituents. We interpret the revealed SERS signatures in terms of the molecular and NP organization at the nanoscale, demonstrating that the action of the DNA bridge molecule yields regular NP aggregates with controlled interparticle distance and reproducible SERS response. In perspective, this demonstrates the potential of the present system as a stable, biocompatible, and recyclable SERS sensor.

Responsive microgels based on poly(N-isopropylacrylamide) (PNIPAM) exhibit peculiar behaviours due to the competition between the hydrophilic and hydrophobic interactions of the constituent networks. The interpenetration of poly-acrilic acid (PAAc), a pH-sensitive polymer, within the PNIPAM network, to form Interpenetrated Polymer Network (IPN) microgels, affects this delicate balance and the typical Volume-Phase Transition (VPT) leading to complex behaviours whose molecular nature is still completely unexplored. Here we investigate the molecular mechanism driving the VPT and its influence on particle aggregation for PNIPAM/PAAc IPN microgels by the joint use of Dynamic Light Scattering and Raman Spectroscopy. Our results highlight that PNIPAM hydrophobicity is enhanced by the interpenetration of PAAc promoting interparticle interactions, a crossover concentration is found above which aggregation phenomena become relevant. In this paper, a new model of real social networks is introduced. The main characteristic of the model is an attachment mechanism based on the existence of network communities, a key feature of real social networks. Using numerical simulations, we show how this model shares with these networks several peculiarities (such as high levels of clustering, low values of the characteristic path length and node betweenness, division into communities and heterogeneous degree distribution), not captured in other network models.

Self-organizing systems arise in many different fields. In this work we analyze data from social and biological systems. A central question is to demonstrate the presence of the determinism in time-series extracted from such systems that appear apparently not correlated but that are two good benchmarks for the study of complexity in real systems. We will apply the Kaplan test and we will define an order parameter for the biological data to characterize the complexity of the system.

The aim of this study was to determine if heightened stress had an adverse affect on the postural control of 14 young right‑handed females during quiet standing in either the presence or the absence of visual input. The level of stress was evaluated by measuring the free cortisol response to awakening (cortisol awakening response; CAR) and by employing the perceived stress scale (PSS). Overall mood disturbance was measured using the profile of mood states (POMS). Postural control was evaluated using a force platform by measuring the 95% confidence ellipse area described by the center of pressure during 5 balance positions maintained for at least 52 sec, each with open and closed eyes. The results of this study revealed a significant positive correlation between CAR, PSS and POMS for each of the studied subjects. Furthermore, it was observed that whilst the level of stress was capable of influencing postural stability, this influence was particularly evident when no visual information was available. Additionally, it was determined that maintenance of posture is easier when the dominant foot is ahead, regardless of visual input.

In this paper, we aim to merge the concept of quantum computing with the paradigm of Cellular Nonlinear Network (CNN) in order to show if and when spatial dynamical complex behaviors can emerge from the interaction of spatially extended quantum cells constituting a quantum cellular nonlinear network (Q-CNN).

In this paper, a dynamical model based on Kuramoto-like oscillators is used to represent the Italian high-voltage power grid. Nodes of the network are generators/substations, while links are the physical connections between generators/substations. The normal operating regime of the power grid corresponds to the regime in which the oscillations of all the nodes are synchronized. We studied the conditions for synchronization and the effect of dynamical perturbations on the nodes. The analysis allows to define several dynamical parameters assessing the dynamical robustness of the network.
