Neste trabalho realizaram-se provas de identificação qualitativa e quantitativa do nitrato de econazol (matéria-prima); ensaios de caracterização térmica do nitrato de econazol, excipientes e formulação farmacêutica (creme), assim como desenvolvimento de metodologia analítica específica e seletiva para a quantificação do nitrato de econazol na formulação farmacêutica de creme. A caracterização térmica dos excipientes foi realizada usando-se a Termogravimetria (TG). Para a caracterização térmica do nitrato de econazol e da formulação foram empregadas a Termogravimetria, a Termogravimetria Derivada (TG/DTG) e a Calorimetria Exploratória Diferencial (DSC). Na caracterização térmica da formulação foram identificados e quantificados dois tipos de água com interações diferentes com a matriz. No desenvolvimento da metodologia específica utilizou-se a eletroforese capilar em zona, método linear com concentrações entre 80 e 120 µg mL-1, com coeficiente de correlação de 0,9995; precisão calculada como desvio padrão relativo (DPR) menor de 2%; exatidão do método comprovada mediante teste de recuperação, obtendo-se valores de 100±2,5%; limites de detecção e quantificação 1,853 µg.mL-1 e 5,617 µg.mL-1, respectivamente, comprovando-se também que o método é robusto. No desenvolvimento da metodologia seletiva foram usadas a cromatografia líquida de alta eficiência com eluição em gradiente (CLAE) e a eletroforese capilar pelo método da cromatografia eletrocinética micelar (MEKC). Nestes métodos foram separados o nitrato de econazol, as impurezas relatadas na literatura do nitrato de econazol (4-álcool clorobenzílico, alfa-2,4-diclorofenil-1 H¬imidazol-1-etanol) e os conservantes presentes na formulação (metilparabeno e propilparabeno). O método seletivo desenvolvido por CLAE foi linear para todas as moléculas, com coeficientes de correlação maiores de 0,99. A precisão calculada para o nitrato de econazol como DPR foi menor de 2%; a exatidão para o nitrato de econazol foi comprovada mediante teste de recuperação, obtendo-se valores de 100±2%; comprovou-se também que o método é robusto e que poderia ser aplicado para a quantificação de cada um destes cinco compostos. No método seletivo desenvolvido por MEKC se obteve tempo menor em comparação ao método por CLAE. In this work, were performed qualitative and quantitative identitication econazole nitrate (raw material); thermal characterization of econazole nitrate, excipients and formulation (cream). There also were developed and validated specific and selective analytical methodology for quantitative determination of econazole nitrate in creams. The thermal characterization of drug product excipients was performed using the Thermogravimetry (TG). For the thermal characterization of econazole nitrate drug and their formulation were used the Thermogravimetry, Derivative Thermogravimetry (TG/DTG) and Differential Scanning Calorimetry (DSC). From cream formulation were identified and quantified two kinds of water which have different interactions with matrix. As regards lo developing of specific methodology was used the capillary zone electrophoresis (CZE). The method was linear, with a correlation coefficient of 0.9995 in the concentration range of 80 to 120 µg.mL-1 of econazole nitrate, precision calculated as relative standard deviation (RSD) was less than 2%, accuracy calculated percentage of recovery for commercial sample was of 100 ± 2.5%, detection and quantitation limits were 1.853 µg.mL-1 and 5.617 µg.mL-1, respectively. The CZE method showed to be robust. For developing of selective methodology were used high performance liquid chromatography (HPLC) with gradient elution, and micellar electrokinetic chromatography (MEKC) methods. Econazole nitrate, related impurities the drug (4-Chlorobenzyl alcohol, alpha-2,4-dichlorophenyl-1H - imidazole-1-ethanol) and preservatives (methylparaben, propylparaben) present in the drug product were separated. The HPLC method was linear for all analytes, the correlation coefficients were higher than 0.99. The precision for nitrate econazol in terms of RSD was less than 2%, the percentage of recovery for commercial sample of econazole nitrate was 100±2%, the method is robust and could be applied for the quantitation of the five substances cited above. With the MEKC method developed the five substances were separated less time comparated with HPLC method.

A reversed-phase high performance liquid chromatographic (RP-HPLC) method for determination of econazole nitrate, preservatives (methylparaben and propylparaben) and its main impurities (4-chlorobenzyl alcohol and alpha-(2,4-dichlorophenyl)-1 H -imidazole-1-ethanol) in cream formulations, has been developed and validated. Separation was achieved on a column Bondclone ® C18 (300 mm × 3.9 mm i.d., 10 μm) using a gradient method with mobile phase composed of methanol and water. The flow rate was 1.4 mL min −1 , temperature of the column was 25 °C and the detection was made at 220 nm. Miconazole nitrate was used as an internal standard. The total run time was less than 15 min. The analytical curves presented coefficient of correlation upper to 0.99 and detection and quantitation limits were calculated for all molecules. Excellent accuracy and precision were obtained for econazole nitrate. Recoveries varied from 97.9 to 102.3% and intra- and inter-day precisions, calculated as relative standard deviation (R.S.D.), were lower than 2.2%. Specificity, robustness and assay for econazole nitrate were also determined. The method allowed the quantitative determination of econazole nitrate, its impurities and preservatives and could be applied as a stability-indicating method for econazole nitrate in cream formulations.

A simple, rapid, economical and reliable high performance liquid chromatographic method has been developed and successfully applied in simultaneous determination of ethinyl estradiol and drospirenone in coated tablets. The HPLC method was performed on a LiChroCART® 100RP column (125x4 mm i.d., 5 µm) with acetonitrile:water 50:50 (v/v) as mobile phase, pumped at a flow rate of 1.0 mL.min-1. The fluorescence detection for ethinyl estradiol was made at λex= 280 nm and λem= 310 nm and a UV detection for drospirenone was made at 200 nm. The elution time for ethinyl estradiol and drospirenone were 4.0 and 5.7 min, respectively. The method was validated in accordance to USP 34 guidelines. The proposed HPLC method presented advantages over reported methods and is suitable for quality control assays of ethinyl estradiol and drospirenone in coated tablets. Um método simples, rápido, econômico e confiável foi desenvolvido empregando a cromatografia líquida de alta eficiência para a determinação simultânea de etinilestradiol e drospirenona em comprimidos revestidos. O método foi realizado utilizando coluna LiChroCART® 100RP (125 x 4 mm d.i., 5 µm), a fase móvel constituída de acetonitrila:água, 50:50 (v/v) com vazão de 1,0 mL.min-1. A detecção foi realizada empregando fluorescência em λex= 280 nm e λem= 310 nm para o etinilestradiol e na região de UV em 200 nm para a drospirenona. O etinilestradiol e a drospirenona tiveram tempo de retenção de 4,0 e 5,7 min, respectivamente. O método foi validado de acordo com as diretrizes da USP 34. O método proposto apresentou vantagens sobre os relatados na literatura e pode ser considerado adequado para o controle de qualidade do etinilestradiol e da drospirenona em comprimidos revestidos.

O diabetes mellitus (DM) é uma síndrome na qual o metabolismo de hidratos de carbono, gorduras e proteínas está alterado, por falta de secreção de insulina ou por diminuição da sensibilidade tissular a este hormônio. O DM pode ser do tipo I, também denominado diabetes mellitus insulinodependente (DMID), caracterizado pela falta de secreção da insulina, e do tipo II, também denominado diabetes mellitus não insulinodependente (DMNID), caracterizado pela menor sensibilidade dos tecidos efetores às ações metabólicas da insulina. Embora ainda não haja cura definitiva, há vários tratamentos disponíveis que proporcionam qualidade de vida para o paciente portador, como a administração de hipoglicemiantes orais. Nesta pesquisa, objetivou-se desenvolver métodos indicativos de estabilidade para a glibenclamida e o cloridrato de metformina. Assim, foram desenvolvidos métodos de rastreamento ortogonais utilizando a cromatografia líquida de alta eficiência (HPLC) e a eletroforese capilar (CE), metodologias que foram desafiadas com os estudos da degradação forçada. Realizou-se, também, a identificação do principal produto de degradação da glibenclamida utilizando a cromatografia líquida acoplada à espectrometria de massa (LC-MS). O método por HPLC teve o melhor desempenho no monitoramento dos produtos de degradação da glibenclamida, apresentando boa linearidade nas concentrações entre 0,210 e 0,360 mg/mL; com coeficiente de correlação maior de 0,99. A precisão calculada como desvio padrão relativo (DPR) foi menor de 3%, exatidão do método comprovada mediante teste de recuperação, obtendo-se valores de 100±3,0%. No teste de especificidade, foram detectados três potenciais produtos de degradação, com os seguintes tempos de retenção relativos (TRR) de 0,33; 0,46 e 0,83. O método CZE teve o melhor desempenho no monitoramento dos produtos de degradação do cloridrato de metformina, apresentando boa linearidade nas concentrações entre 0,210 e 0,360 mg/mL, com coeficiente de correlação maior de 0,99. A precisão, calculada como DPR, foi menor do que 5%, exatidão do método comprovada mediante o teste de recuperação, obtendo-se valores de 100±3,5%. No teste de especificidade, foram detectados cinco potenciais produtos de degradação com os seguintes TRRs de 0,90; 1,14; 1,35; 1,45 e 1,56. Diabetes mellitus (DM) is a syndrome which alters the metabolism of carbohydrates, fats and proteins by lack of insulin secretion or a decrease in tissue sensitivity to insulin. Type 1 DM is also known as insulin-dependent diabetes mellitus is characterized by lack of insulin secretion. The second type of diabetes known as Type II, also called non-insulin dependent, is characterized by the reduced sensitivity of target tissues to the metabolic actions of insulin. Although there is no definitive cure, there are several treatments available that provide quality of life for the patient how treatment with oral hypoglycemic agents. This study had as objective to developed stability-indicating methods for glibenclamide and metformin hydrochloride. Thus, the techniques used in this study involved high performance liquid chromatography (HPLC) and capillary electrophoresis (CE), and both were challenged with forced degradation studies. The identification of degradation products of glibenclamide was also performed, utilizing the mass spectrometry (MS) technique. However, the HPLC technique had the best performance for monitoring the degradation products of glibenclamide, which showed a good linearity in concentrations between 0.210 - 0.360 mg/ml, with a correlation coefficient greater than 0.99. The precision was calculated as a relative standard deviation (RSD) which was less than 3%; the accuracy of the method calculated as the percent recovery was obtained with the following values: 100 ± 3.0%. On the specificity were detected three potential products of degradation utilizing forced degradation, with the following relative retention times (RRT) 0.33, 0.46 and 0.83. The CZE method had the best performance to monitoring the degradation products of metformin hydrochloride, which showed good linearity of concentrations between 0.210 and 0.360 mg/ml, with a correlation coefficient greater than 0.99. The intra-day and inter-day precision calculated as DPR was less than 5%, and an accuracy of this method was achieved using the values of 100 ± 3.5%. In the specificity were five potential degradation products were detected with the following TRRS: 0.90, 1.14, 1.35, 1.45 and 1.56.

A simple, fast, inexpensive and reliable capillary zone electrophoresis (CZE) method for the determination of econazole nitrate in cream formulations has been developed and validated. Optimum conditions comprised a pH 2.5 phosphate buffer at 20 mmol L(-1) concentration, +30 kV applied voltage in a 31.5 cm x 50 microm I.D. capillary. Direct UV detection at 200 nm led to an adequate sensitivity without interference from sample excipients. A single extraction step of the cream sample in hydrochloric acid was performed prior to injection. Imidazole (100 microg mL(-1)) was used as internal standard. Econazole nitrate migrates in approximately 1.2 min. The analytical curve presented a coefficient of correlation of 0.9995. Detection and quantitation limits were 1.85 and 5.62 microg mL(-1), respectively. Excellent accuracy and precision were obtained. Recoveries varied from 98.1 to 102.5% and intra- and inter-day precisions, calculated as relative standard deviation (RSD), were better than 2.0%. The proposed CZE method presented advantageous performance characteristics and it can be considered suitable for the quality control of econazole nitrate cream formulations.
