Inflammasomes are cytosolic protein platforms whose assembly/activation, in order to resolve various types of threats, can be triggered by a multitude of microbial and host derived stimuli. Their discovery was the key breakthrough in the understanding of inflammation initiation. Although beneficial in clearing microbes and restoring physiological or tissue homeostasis, aberrant or excessive stimulation of inflammasomes leads to various pathologies. The activation of NLRP3 inflammasome is associated with the physio-pathogenesis or the potentiation of the severity of so many diseases including respiratory diseases, such as asthma, COPD, COVID-19, malaria, diabetes, atherosclerosis, etc. Considering the extent of the NLRP3 inflammasome in diseases, therapies targeting the inhibition of its activation appear as a promising approach for the management of a wide range of diseases. The objective of this research work was to investigate the potential of Rwandan medicinal plants in the inhibition of the NLRP3 inflammasome. To this end, we first carried out a field survey to collect ethnobotanical information on plants used for the treatment of asthma in Rwanda. The present thesis is divided into two main parts:- The ethnobotanical study and screening of Rwandan medicinal plants with inhibitory activity on NLRP3 inflammasome activation;- The isolation and structure characterization of the active ingredients responsible for NLRP3 inflammasome inhibition, and the investigation of antiplasmodial activity of selected plants.In the first part, 19 herbal recipes were reconstituted from 61 medicinal plants based on an ethnobotanical survey. Most of these recipes exhibited significant inhibition of the NLRP3 inflammasome activation. Human Onchocerciasis is a neglected tropical disease of debilitating consequences caused by the nodule dwelling nematode Onchocerca volvulus. Excretory Secretory Products (ESPs) help the parasite to establishing infection in an immunocompetent host and thus make O. volvulus ESPs potential sources of diagnostic, therapeutic or immunomodulatory markers. Here, a blend of bioinformatics, biochemical, analytical and immunologic methods were employed to identify one such ESP as a GM2 activator protein orthologue. The cDNA of OvGM2AP was detected in most parasite stages and the protein was detected in ESPs of adult males, adult females and L3 stage. Recombinant OvGM2AP was expressed in Sf21 insect cells and was found to be glycosylated at Asn 173 by mass spectrometry analysis. The recombinant OvGM2AP could significantly differentiate infected from non-infected individuals on total IgG ELISA. No improvement in diagnostic indices was observed when IgG sub-classes were used to detect OvGM2AP by ELISA. A drop in IgG titre was observed to correlate with increased rounds of ivermectin administration. However, as observed from alignment of OvGM2AP sequences with other nematode species, recombinant OvGM2AP was found to cross react with sera from patients with other nematode infections such as Loa loa, Mansonella perstans and Brugia malayi. GM2 degradation to GM3 could not be mediated by OvGM2AP in the presence of human β-hexosaminidase A. Regulatory T cells (Treg) are a suppressive subset of helper T cells that controls immune responses using a variety of mechanisms. Despite initially being thought of as a homogenous population, Treg cells were recently found to adapt their function to their environment and acquire specialized phenotypes depending on their tissue of residence. The molecular mechanisms underlying this functional and tissular adaptation remain to be fully elucidated.In the course of this work, we studied the role of transcription factor c-Maf in the differentiation and the function of Treg cells. For this, we used a murine model invalidated for c-Maf specifically in Treg cells. Transcription factor c-Maf is preferentially expressed by intestinal Treg cells, particularly among the RORγt+ Treg subset, which controls immune responses directed towards the gut microbiota. We have shown that the differentiation of RORγt+ Treg cells results from the integration of multiple environmental signals, highlighting the plasticity of Treg specialization. Furthermore, we have shown that c-Maf is required for the differentiation of RORγt+ Treg cells as well as the expression of the anti-inflammatory cytokine IL-10 by intestinal Treg cells, thus endowing Treg cells with the ability to control homeostatic Th17 responses in the intestine. Mice deficient for c-Maf in Treg cells exhibit an exacerbated Th17 response and spontaneously develop colitis. However, c-Maf-deficient mice develop fewer polyps in a model of colitis-associated colon cancer. This suggests that c-Maf expression in Treg cells plays a beneficial role on intestinal homeostasis at steady state, but is detrimental in a tumoral context.Our results allow a better understanding of the mechanisms involved in the specialization of intestinal Treg cells. Le nombre de lymphocytes T (LT) présents dans l’organisme est contrôlé de manière étroite et maintenu constant. En réponse à une chute de ce nombre, par exemple suite à une infection, un traitement de chimiothérapie ou lors d’une greffe de moelle osseuse, les LT résiduels ou transférés se divisent activement et repeuplent les compartiments lymphoïdes, rétablissant ainsi un nombre «normal» de cellules immunes périphériques. Ce processus, appelé prolifération homéostatique, est soutenu par l’IL-7 ainsi que les signaux TCR et mène à la génération de LT effecteurs/mémoire. Ces cellules effectrices/mémoire peuvent favoriser le développement d’une réponse anti-tumorale mais sont aussi impliquées dans le développement de maladies auto-immunes et inflammatoires.Des changements métaboliques sont étroitement liés à la différenciation et aux fonctions des LT. De plus, un nombre grandissant de données suggère qu’une modulation du métabolisme permet d’influencer le cours d’une réponse immune. L’AMP-activated protein kinase (AMPK) est un senseurmajeur du stress métabolique qui régule l’homéostasie des mitochondries, la glycolyse et l’équilibre rédox. La spécification de l’identité ventrale ou dorsale des progéniteurs au cours du développement du télencéphale est la première étape cruciale du développement du cortex cérébral. Les gènes doublesex and mab-3 related (Dmrt) Dmrt3 et Dmrt5 codent pour des facteurs de transcription à doigt de Zinc. Ces gènes sont coexprimés selon un gradient fort caudomédialement à plus faible rostrolatéralement dans le primordium du cortex cérébral. Nous avons d’abord démontré qu’ils étaient tous deux nécessaires pour la formation normale de l’hème corticale, l’hippocampe et le néocortex caudomédian. Nous avons plus récemment adressé le rôle de Dmrt3 et Dmrt5 dans le contrôle de la régionalisation dorsale/ventrale du télencéphale chez la souris, en comparant les phénotypes d’embryons simple knock out (KO) aux double KO (dKO), et par une expression ectopique de Dmrt5 dans le télencéphale ventral. Nous avons mis en évidence que DMRT3 et DMRT5 agissent comme des régulateurs critiques de l’identité dorsoventrale des cellules progénitrices en réprimant les régulateurs ventralisants. Les régulateurs transcriptionnels précoces de la destinée ventrale exprimés dans la partie dorsale de l’éminence ganglionnaire latérale tel que Gsx2 sont régulés positivement dans le télencéphale dorsal embryons dKO et régulés négativement lorsque les progéniteurs du télencéphale ventral expriment Dmrt5 de manière ectopique. La surexpression conditionnelle de Dmrt5 au sein du télencéphale entier génère un profil d’expression et des défauts très similaires à ceux observés lors d’une activité Gsx2 diminuée. De plus, les embryons Emx2 ;Dmrt5 double KO montrent un phénotype similaire à celui des embryons dKO. DMRT3, DMRT5 et le facteur de transcription à homéobox EMX2 peuvent se lier à un enhancer spécifique du télencéphale ventral dans le locus Gsx2. Beyond its well-admitted role in development and organogenesis, it is now clear that the transcription factor c-Maf has owned its place in the realm of immune-related transcription factors. Formerly introduced solely as a Th2 transcription factor, the role attributed to c-Maf has gradually broadened over the years and has extended to most, if not all, known immune cell types. The influence of c-Maf is particularly prominent among T cell subsets, where c-Maf regulates the differentiation as well as the function of multiple subsets of CD4 and CD8 T cells, lending it a crucial position in adaptive immunity and anti-tumoral responsiveness. Recent research has also revealed the role of c-Maf in controlling Th17 responses in the intestine, positioning it as an essential factor in intestinal homeostasis. La voie Wnt est une voie de signalisation importante pour l’embryogenèse, la morphogenèse et l’homéostasie des tissus au stade adulte. Des mutations dans cette voie de signalisation sont souvent associées à une létalité embryonnaire ou à des pathologies sévères.Une caractéristique singulière de la signalisation Wnt est sa grande complexité génétique. Chez les vertébrés, ce sont 19 ligands Wnt différents qui peuvent potentiellement lier les 10 membres de la famille des récepteurs Frizzled (Fz). De plus, la liaison d’un ligand Wnt à un récepteur Fz peut conduire à l’activation d’au moins trois voies de transduction distinctes. Pourtant, l’interaction Wnt/Fz est incompatible avec une reconnaissance monospécifique étant donné que Wnt et Fz interagissent via des résidus conservés dans les deux familles. Les patrons d’expression des différents Wnt et des différents Fz sont complexes et souvent chevauchant. Malgré cela, la délétion sélective d’un Wnt peut conduire à des phénotypes spécifiques non-observés lors de la délétion d’un autre ligand Wnt co-exprimé. C’est notamment le cas des ligands Wnt7a et Wnt7b. En cas de lymphopénie -diminution du nombre de lymphocytes T (LT) présents en périphérie-, les LT restants prolifèrent. Ce processus, dit de prolifération homéostatique, est régulé par la disponibilité de la cytokine IL-7 ainsi que par des interactions TcR/MHC et mène à la différenciation de LT effecteurs/mémoires. La prolifération homéostatique peut augmenter l’efficacité des immunothérapies anti-cancéreuses ou avoir des conséquences délétères pour l’organisme (réaction du greffon contre l’hôte, auto-immunité). Les LT naïfs, effecteurs et de mémoire produisent leur énergie via des mécanismes différents et la capacité des lymphocytes à enclencher le programme métabolique adéquat au bon moment joue un rôle essentiel dans leur différenciation. En outre, l’inhibition de la glycolyse ou de la respiration mitochondriale altère la prolifération homéostatique, suggérant que ces deux voies métaboliques sont importantes dans ce processus. Les mécanismes par lesquels les LT accroissent leur métabolisme énergétique lors de la prolifération homéostatique ne sont, à ce jour, pas encore élucidés. L’AMPK est un régulateur essentiel du métabolisme cellulaire et est la cible de différents composés déjà utilisés chez l’homme. L’objectif de notre travail a été de déterminer les conséquences de l’absence d’AMPK sur la capacité des LT à proliférer de manière homéostatique et à adapter leur métabolisme en réponse à l’IL-7. Notre hypothèse a été que l’AMPK, en permettant de réorienter le métabolisme des LT, jouerait un rôle important dans le processus de prolifération homéostatique. Follicular helper T cells (Tfh) support high-affinity Ab production by germinal center B cells through both membrane interactions and secretion of IL-4 and -21, two major cytokines implicated in B-cell survival and Ab class switch. Tfh-2 cells recently emerged in humans as a strong IL-4 producer Tfh cell subset implicated in both autoimmune and allergic diseases. Although the molecular mechanisms governing Tfh cell differentiation from naive T cells have been widely described, much less is known about the regulation of cytokine secretion by mouse Tfh-2 cells. The purpose of our study was to evaluate the role of dendritic cell-derived IL-6 in fine-tuning cytokine secretion by Tfh cells. Our results demonstrate that priming of Th cells by IL-6-deficient antigen-presenting dendritic cells preferentially leads to accumulation of a subset of Tfh cells characterized by high expression of GATA3 and IL-4, associated with reduced production of IL-21. STAT3-deficient Tfh cells also overexpress GATA3, suggesting that early IL-6/STAT3 signaling during Tfh cell development inhibits the expression of a set of genes associated with the Th2 differentiation program. Follicular helper T cells (Tfh) have been identified as the primary cell subpopulation regulating B cell responses in germinal centers, thus supporting high-affinity antibody production. Among the transcription factors orchestrating Tfh cell differentiation and function, the role played by the proto-oncogene c-Maf remains poorly characterized. We report herein that selective loss of c-Maf expression in the T cell compartment results in defective development of Tfh cells in response to both antigen/adjuvant vaccinations and commensal intestinal bacteria. Accordingly,c-Maf expression in T cells was essential for the development and high-affinity antibody secretion in vaccinated animals. c-Maf was expressed early, concomitantly to BCL6, in Tfh cell precursors and found to regulate Tfh fate in a cell-autonomous fashion. Les tumeurs sont, au moins en partie, des maladies de la régulation du cycle cellulaire. Le point de restriction (R) est un point fondamental du cycle cellulaire où les cascades de signalisation mitogéniques (y compris leurs perversions oncogènes) et l'état métabolique des cellules sont intégrés pour engager le cycle de division cellulaire. Les CDK4 et CDK6 sont les premières CDKs à être activées en réponse aux signaux de prolifération cellulaire.En initiant la phosphorylation inactivatrice de l'oncosuppresseur central pRb, les CDK4 / 6 liées aux cyclines D jouent un rôle essentiel dans le passage du point R et donc dans la décision de multiplication cellulaire, en particulier dans la plupart des cellules cancéreuses dans lesquelles l'activité de CDK4 est fortement dérégulée. Les médicaments inhibiteurs de CDK4 / 6 sont maintenant évalués dans la plupart des cancers dans de nombreux essais cliniques. Notre groupe a identifié la phosphorylation activatrice sur la Thr172 comme l'étape hautement régulée qui détermine l'activation de CDK4. La question de savoir si les phosphorylations de CDK4 et de CDK6 sont catalysées par la seule « CDK-activating kinase » (CAK, cycline H-CDK4-Mat1) reste incertaine. En utilisant des cellules de cancer colorectal (HCT116) exprimant une version de CDK7 spécifiquement inhibable (dite « analog sensitive »), nous avons participé à une étude qui démontre, d’une part, une implication cruciale de la CDK7 dans la phosphorylation / activation de CDK4 et CDK6 et, d’autre part, l'existence de kinase(s) activatrice(s) de la CDK4 différente(s) de la CDK7. De plus, cette étude a démontré que l'activité de la CDK7 était conditionnée, au moins en partie, par la liaison de p21 à la CDK4, laquelle augmentait en réponse à l'inhibition de CDK7. In analogue-sensitive CDK7 (as/as) mutant HCT116 cells in which CDK7 can be specifically inhibited, we participated to a study demonstrating a crucial CDK7 involvement in phosphorylation/activation of CDK4 and CDK6 and existence of non-CDK7 CDK4 activating kinase(s). Moreover, this study demonstrated that CDK7 activity was conditioned, at least in part, by p21 binding to CDK4, which increased in response to CDK7 inhibition. This coincided with disappearance of the most abundant phosphorylation of p21, which was localized (by 2D-gel electrophoresis analyses) at Ser130 and found to be catalyzed by both CDK4 and CDK2. Surprisingly, Ser130 p21 phosphorylation was not inhibited only by CDK7 inhibition, but also by Roscovitine and CR8 (CDK2 inhibitors) and Palbociclib. Hyaluronan is a major component of the extracellular matrix and glycocalyx. Its main somatic degrading enzymes are hyaluronidases 1 and 2, neither of which is active in the bloodstream. We generated hyaluronidase 2-deficient mice. These animals suffer from chronic, mild anemia and thrombocytopenia, in parallel with a 10-fold increase in plasma hyaluronan concentration. In this study we explored the mechanism of these hematologic anomalies. The decreased erythrocyte and platelet counts were attributed to peripheral consumption. The erythrocyte half-life was reduced from 25 to 8 days without signs of premature aging. Hyaluronidase 2-deficient platelets were functional. Major intrinsic defects in erythrocyte membrane or stability, as well as detrimental effects of high hyaluronan levels on erythrocytes, were ruled out in vitro. Normal erythrocytes transfused into hyaluronidase 2-deficient mice were quickly destroyed but neither splenectomy nor anti-C5 administration prevented chronic hemolysis. Schistocytes were present in blood smears from hyaluronidase 2-deficient mice at a level of 1% to 6%, while virtually absent in control mice. Hyaluronidase 2-deficient mice had increased markers of endothelial damage and microvascular fibrin deposition, without renal failure, accumulation of ultra-large multimers of von Willebrand factor, deficiency of A Disintegrin And Metalloproteinase with ThromboSpondin type 1 motifs, member 13 (ADAMTS13), or hypertension. There was no sign of structural damage in hepatic or splenic sinusoids, or in any other microvessels. We conclude that hyaluronidase 2 deficiency induces chronic thrombotic microangiopathy with hemolytic anemia in mice. Adenosine monophosphate-activated protein kinase (AMPK) is a serine/threonine kinase that is crucial for cellular energy metabolism homeostasis. AMPK monitors cellular energy status in response to nutritional variations and, once activated by low energy status, switches on ATP-producing catabolic pathways and switches off ATP-consuming anabolic pathways to restore cellular energy homeostasis. When T lymphocytes encounter foreign antigens, they initiate a program of differentiation leading to the rapid generation of effector and memory cells that clear the pathogen and prevent future infection, respectively. Differentiation of naïve T cells in effector or long term memory cells is tightly associated with changes in their energy metabolic activity and recent data have revealed that fine-tuning of metabolism could modulate T cell functions. L’aide fournie aux lymphocytes B par les lymphocytes T CD4+ est cruciale pour une réponse humorale de qualité. Les lymphocytes T helper folliculaires (Tfh) jouent un rôle majeur dans l’aide apportée aux lymphocytes B au sein des centres germinatifs et peu de choses sont connues sur la capacité d’aide des autres populations de cellules T CD4+. Le but de notre étude est d’évaluer la capacité d’aide aux lymphocytes B des lymphocytes T helper de type 2 (Th2), une population considérée, à l’origine, comme responsable de l’aide apportée aux lymphocytes B in vivo ainsi que d’analyser le rôle de l’axe IL6 / STAT3 dans la différenciation des lymphocytes Tfh et leur plasticité phénotypique. Nous montrons que les lymphocytes Th2 co-expriment des formes actives des facteurs de transcription STAT6 et STAT3 et que l’expression de STAT3 est requise pour la fonction d’aide aux lymphocytes B des lymphocytes Th2. We generated hyaluronidase 2-deficient mice. These animals suffer from chronic, mild anemia and thrombocytopenia, in parallel with a 10-fold increase in plasma hyaluronan concentration. In this study we explored the mechanism of these hematologic anomalies. The decreased erythrocyte and platelet counts were attributed to peripheral consumption. The erythrocyte half-life was reduced from 25 to 8 days without signs of premature aging. Hyaluronidase 2-deficient platelets were functional. Major intrinsic defects in erythrocyte membrane or stability, as well as detrimental effects of high hyaluronan levels on erythrocytes, were ruled out in vitro. Normal erythrocytes transfused into hyaluronidase 2-deficient mice were quickly destroyed but neither splenectomy nor anti-C5 administration prevented chronic hemolysis. Schistocytes were present in blood smears from hyaluronidase 2-deficient mice at a level of 1% to 6%, while virtually absent in control mice. Hyaluronidase 2-deficient mice had increased markers of endothelial damage and microvascular fibrin deposition, without renal failure, accumulation of ultra-large multimers of von Willebrand factor, deficiency of A Disintegrin And Metalloproteinase with ThromboSpondin type 1 motifs, member 13 (ADAMTS13), or hypertension. There was no sign of structural damage in hepatic or splenic sinusoids, or in any other microvessels. We conclude that hyaluronidase 2 deficiency induces chronic thrombotic microangiopathy with hemolytic anemia in mice. The identification of DC-derived signals orchestrating activation of Th1 and Th17 immune responses has advanced our understanding on how these inflammatory responses develop. However, whether specific signals delivered by DCs also participate in the regulation of Th2 immune responses remains largely unknown. In this study, we show that administration of antigen-loaded, IL-6-deficient DCs to naïve mice induced an exacerbated Th2 response, characterized by the differentiation of GATA-3-expressing T lymphocytes secreting high levels of IL-4, IL-5, and IL-13. Coinjection of wild type and IL-6-deficient bone marrow-derived dendritic cells (BMDCs) confirmed that IL-6 exerted a dominant, negative influence on Th2-cell development. This finding was confirmed in vitro, where exogenously added IL-6 was found to limit IL-4-induced Th2-cell differentiation. iNKT cells were required for optimal Th2-cell differentiation in vivo although their activation occurred independently of IL-6 secretion by the BMDCs. Regulation of gene expression occurs at several levels in a cell. While control of transcription is often viewed as the main source of regulation, it is now clear that post-transcriptional processes are essential to fine-tune protein availability. The presence of AU-rich elements (ARE) in the 3’ untranslated region (3’UTR) of many important mRNAs exemplifies one such process. AREs alter the mRNA translation or degradation status by recruiting ARE-binding proteins (ARE-BP). ARE-BPs of the TTP/TIS11 family bind to their cognate ARE-RNAs using their conserved tandem zinc-finger domain and induce rapid decay of their targets. This allows for proper regulation of cell proliferation, cell death and inflammation. In this regard, TTP/TIS11 are main regulators of gene expression, and as such are put under strict transcriptional, post-transcriptional as well as several layers of post-translational control.In this work, we aimed at elucidating the degradation mechanisms affecting TTP/TIS11. Using Drosophila as a model, we found that dTIS11 protein turnover is rapid due to continuous degradation by the proteasome. However, proteasomal recognition did not require ubiquitination of dTIS11 as non-ubiquitinable mutants were efficiently degraded by the proteasome. In addition, dTIS11 was digested by the 20S proteasome that lacks ubiquitin-recognition domains. Our results further indicate that intrinsically disordered regions (IDR) in dTIS11 may be responsible for proteasomal recognition. In fact, dTIS11 is predicted as disordered and possesses the main characteristics of intrinsically disordered proteins (IDP). We also identified dTIS11 N- and C-terminal domains as functional signals for degradation, potentially due to their destructuration. This ubiquitination-independent, disorder-dependent degradation process is conserved throughout evolution as dTIS11 mammalian counterpart, TTP, undergoes the same degradation by default pathway. L’objectif de notre travail a été de caractériser le rôle de l’IL-6 dans la différenciation des lymphocytes Tfh et des lymphocytes Th2. Les lymphocytes Tfh ont pour fonction d’aider les lymphocytes B à produire des anticorps indispensables pour nous protéger contre divers pathogènes. Les lymphocytes Th2, quant à eux, sont spécialisés dans l’élimination de parasites extracellulaires tels que les helminthes.Dans un premier temps, nous avons voulu identifier les gènes dont l’expression est induite par l’IL-6, avec comme objectif une meilleure compréhension des mécanismes permettant aux lymphocytes T de se différencier en cellules Tfh.Au cours de notre travail, nous avons identifié le facteur de transcription, MyoR (Myogenic Repressor) comme étant exprimé au sein des lymphocytes T helper et dont l’expression est induite par l’IL-6. Nos observations expérimentales ont démontré que le facteur MyoR n’est pas indispensable pour la différenciation des lymphocytes Tfh, ni pour leur fonction. Cependant, l’expression de l’ARNm codant pour MyoR pourrait être utilisée comme un biomarqueur des cellules Tfh in vitro ou in vivo.Nous avons ensuite caractérisé la réponse immune induite in vivo par des cellules présentatrices d’antigènes issues de souris déficientes pour l’IL-6. Cette approche nous a permis de mettre en évidence le rôle immunosuppresseur de l’IL-6 sur le développement des réponses de type Th2. En effet, nous avons montré que l’injection de BMDCs (Bone Marrow derived dendritic cells) IL-6-/- dans des souris receveuses de type sauvage induisent une réponse Th2 augmentée in vivo.Nos résultats suggèrent que l’inhibition de la réponse Th2 par l’IL-6 in vivo et in vitro pourrait impliquer la présence d’un ou de plusieurs miRNAs.Cette inhibition pourrait être un mécanisme de rétrocontrôle afin d’éviter une exacerbation de la réponse immune Th2., Doctorat en Sciences, info:eu-repo/semantics/nonPublished
