Written during the SARS-CoV-2 pandemic, and in recognition of Andy Grieve, the polymath, this article looks at an eclectic mix of topics where statistical thinking and practices should transcend typical dividing lines-with a particular focus on the areas of drug development, public health and social science. The case is made for embedding an experimental (or quasi-experimental) framework within clinical practice for vaccines and treatments following their marketing authorisation. A similar case is made for public health interventions-facilitated by pre-specification of effect size and by the greater use of data standards. A number of recommendations are made whilst noting that progress is being made in some areas.

Experiments prompting people to engage in more prosocial behavior (e.g., acts of kindness) or simple social interactions (e.g., acting extraverted) have both shown promise in boosting well-being. However, little is known about how much the impact on well-being depends on the type of interpersonal interaction (i.e., social vs. prosocial) or on other proximal features (e.g., whether the interaction takes place online vs. in-person, the closeness of the relationship, or amount of social connection associated with a given interaction). We randomly assigned a sample of full-time employees recruited via a special employed mTurk sample (N = 754) to perform weekly acts of kindness online versus in-person, to engage in weekly social interactions online versus in-person, or to list their daily activities (control) over the course of 4 weeks. First, on average, all conditions reported improvements in well-being (i.e., increases in positive affect and life satisfaction, decreases in negative affect) across the 4-week intervention period. Second, relative to controls, the four experimental groups reported increases in general social connectedness over time. Finally, according to auxiliary analyses collapsed across the experimental condition, closer relationship with target and non-digital medium of delivery predicted episode-level social connection, which, in turn, was associated with general social connectedness and positive affect. We conclude that the "who" and the "how" of a behavior (i.e., its target, its delivery method, and the feelings of social connection generated) are important for well-being, but not the "what" (i.e., whether the behavior is social or prosocial). Environment sensing and recognition can improve the safety and autonomy of human-robot locomotion, especially during transitions between environmental states such as walking to and from stairs. However, accurate and real-time perception on edge devices with limited computational resources is an open problem. Here we present the development and mobile deployment ofStairNet- a vision-based automated stair recognition system powered by deep learning. Building onExoNet- the largest open-source dataset of egocentric images of real-world walking environments - we designed a new dataset specifically for stair recognition with over 515,000 images. We then developed a lightweight and efficient convolutional neural network for image classification, which accurately predicted complex stair environments with 98.4% accuracy. We also studied different model compression and optimization methods and deployed our system on several mobile devices running a custom-designed iOS application with onboard accelerators using CPU, GPU, and/or NPU backend computing. Of the designs that we tested, our highest performing system showed negligible reductions in classification accuracy due to the model conversion for mobile deployment and achieved an inference time of 2.75 ms on an iPhone 11. The high speed and accuracy of theStairNetsystem on edge devices opens new opportunities for autonomous control and planning of robotic prosthetic legs, exoskeletons, and other assistive technologies for human locomotion.

Abstract. Background: In Australia, most state-based suicide registers now publicly release suicide mortality data alongside those the Australian Bureau of Statistics (ABS) releases annually. Aims: This study compared ABS’s recorded suicides with three state-based suicide registers (Queensland, Victoria, and Tasmania). We compared their case definitions and coding approaches to assist users in choosing the most suitable data source and interpret differences between sources. Method: We collated the number of suicides by year of registration and occurrence (2006–2020). We compared the scope and coding of suicides between ABS and the registers using publicly available suicide reports and data releases. Results: The ABS’s annual suicide numbers are similar to (and in Tasmania exceed) the numbers reported by state-based registers. The ABS year of occurrence data diverges substantially from the Victoria and Queensland register data in 2020, perhaps attributable to ongoing ABS revision processes. Minimal overlap exists between the case definitions and coding practices of the ABS and registers. Limitations: This is not an individual-level concordance study. Conclusion: Despite different case definitions and coding practices, the two sources produced largely consistent data. They have complementary strengths: timeliness (suicide register data) and enabling cross-jurisdictional comparisons (ABS data).

Concerns have been expressed that, at their most extreme, these consequences could manifest as increased suicide rates. We aimed to assess the early effect of the COVID-19 pandemic on suicide rates around the world. Methods: We sourced real-time suicide data from countries or areas within countries through a systematic internet search and recourse to our networks and the published literature. Between Sept 1 and Nov 1, 2020, we searched the official websites of these countries’ ministries of health, police agencies, and government-run statistics agencies or equivalents, using the translated search terms “suicide” and “cause of death”, before broadening the search in an attempt to identify data through other public sources. Data were included from a given country or area if they came from an official government source and were available at a monthly level from at least Jan 1, 2019, to July 31, 2020. Our internet searches were restricted to countries with more than 3 million residents for pragmatic reasons, but we relaxed this rule for countries identified through the literature and our networks. Areas within countries could also be included with populations of less than 3 million. We used an interrupted time-series analysis to model the trend in monthly suicides before COVID-19 (from at least Jan 1, 2019, to March 31, 2020) in each country or area within a country, comparing the expected number of suicides derived from the model with the observed number of suicides in the early months of the pandemic (from April 1 to July 31, 2020, in the primary analysis). Findings: We sourced data from 21 countries (16 high-income and five upper-middle-income countries), including whole-country data in ten countries and data for various areas in 11 countries). Rate ratios (RRs) and 95% CIs based on the observed versus expected numbers of suicides showed no evidence of a significant increase in risk of suicide since the pandemic began in any country or area. There was statistical evidence of a decrease in suicide compared with the expected number in 12 countries or areas: New South Wales, Australia (RR 0·81 [95% CI 0·72–0·91]); Alberta, Canada (0·80 [0·68–0·93]); British Columbia, Canada (0·76 [0·66–0·87]); Chile (0·85 [0·78–0·94]); Leipzig, Germany (0·49 [0·32–0·74]); Japan (0·94 [0·91–0·96]); New Zealand (0·79 [0·68–0·91]); South Korea (0·94 [0·92–0·97]); California, USA (0·90 [0·85–0·95]); Illinois (Cook County), USA (0·79 [0·67–0·93]); Texas (four counties), USA (0·82 [0·68–0·98]); and Ecuador (0·74 [0·67–0·82]). Interpretation: This is the first study to examine suicides occurring in the context of the COVID-19 pandemic in multiple countries. In high-income and upper-middle-income countries, suicide numbers have remained largely unchanged or declined in the early months of the pandemic compared with the expected levels based on the pre-pandemic period. Pragmatic prospection is the ability to think deeply about the future in order to identify and to work productively toward goals. It involves imagining desirable future outcomes, setting sensible goals, making plans, and flexibly executing those plans. We conducted an exploratory survey of full-time working U.S. adults (N = 1541), measuring pragmatic prospection along with life- and job-related outcomes. All data from the present survey are publicly available. Pragmatic prospection correlated positively with positive outcomes (e.g., life satisfaction, work productivity), correlated negatively with negative outcomes (e.g., anxiety, depression), and tracked with other adaptive personality traits associated with achievement and psychological adjustment (e.g., high conscientiousness, low neuroticism). These results point to pragmatic prospection as an important component of flourishing, both in the workplace and in daily life.

The innovator’s bias is defined as the tendency for innovators to focus mainly on the positive potential impact of their inventions and to neglect, ignore, or downplay any potential negative impact. Such bias may help sustain the motivation needed for business success but may create problems by failing to acknowledge and prepare for problematic outcomes. We report three studies (total n = 1608) designed to demonstrate this bias — and to show how to overcome it (while ideally preserving the innovators’ enthusiastic affection for their product). Three studies used hypothetical innovations, all with potential downsides. Feelings of ownership were manipulated by having some participants role-play being marketing manager, including naming the product, devising advertising slogans, and identifying target demographics for potential purchasers. Owners then rated their product, while non-owner controls rated a different product. Study 1 (n = 495) demonstrated the innovator’s bias by showing that owners rated the likely consequences of their product more favorably than non-owners did. Owners also displayed more enthusiastic zeal for their product. Study 2 (n = 553) tested interventions aimed at reducing the bias while preserving the zeal. Of six interventions, the most successful was having owners imagine the worst-case scenario involving the most negative outcome that the invention could cause. Study 3 (n = 560) was a preregistered replication of the main findings from Study 2 (osf.io/ew9cq).

People spend a substantial portion of their lives engaged in conversation, and yet our scientific understanding of conversation is still in its infancy. In this report we advance an interdisciplinary science of conversation, with findings from a large, novel, multimodal corpus of 1,656 recorded conversations in spoken English. This 7+ million word, 850 hour corpus totals over 1TB of audio, video, and transcripts, with moment-to-moment measures of vocal, facial, and semantic expression, along with an extensive survey of speaker post conversation reflections. We leverage the considerable scope of the corpus to (1) extend key findings from the literature, such as the cooperativeness of human turn-taking; (2) define novel algorithmic procedures for the segmentation of speech into conversational turns; (3) apply machine learning insights across various textual, auditory, and visual features to analyze what makes conversations succeed or fail; and (4) explore how conversations are related to well-being across the lifespan. We also report (5) a comprehensive mixed-method report, based on quantitative analysis and qualitative review of each recording, that showcases how individuals from diverse backgrounds alter their communication patterns and find ways to connect. A rapidly approaching dark object evokes an evolutionarily conserved fear response in both vertebrates and invertebrates, young to old. A looming visual stimulus mimics an approaching object and triggers a similarly robust fear response in mice, resulting in freeze and flight. However, the retinal neural pathway responsible for this innate response has not been fully understood. We first explored a variety of visual stimuli that reliably induced these innate responses, and found that a looming stimulus with 2-d acclimation consistently evoked fear responses. Because the fear responses were triggered by the looming stimulus with moving edges, but not by a screen flipping from light to dark, we targeted the starburst amacrine cells (SACs), crucial neurons for retinal motion detection. We used intraocular injection of diphtheria toxin (DT) in mutant mice expressing diphtheria toxin receptors (DTR) in SACs. The looming-evoked fear responses disappeared in half of the DT-injected mice, and the other mice still exhibited the fear responses. The optomotor responses (OMRs) were reduced or eliminated, which occurred independent of the disappearance of the fear responses. A histologic examination revealed that ON SACs were reduced in both mouse groups preserved or absent fear responses. In contrast, the number of OFF SACs was different among two groups. The OFF SACs were relatively preserved in mice exhibiting continued fear responses, whereas they were ablated in mice lacking fear response to looming stimulation. These results indicate that OFF SACs and the direction-selective pathway in the retina play a role in looming-induced fear behaviors.

Aims: We aimed to determine whether there has been a change in the number of suicides occurring in three Australian states overall, and in age and sex subgroups, since the COVID-19 pandemic began, and to see if certain risk factors for suicide have become more prominent as likely underlying contributing factors for suicide.Method: Using real-time data from three state-based suicide registers, we ran multiple unadjusted and adjusted interrupted time series analyses to see if trends in monthly suicide counts changed after the pandemic began and whether there had been an increase in suicides where relationship breakdown, financial stressors, unemployment and homelessness were recorded.Results: Compared with the period before COVID-19, during the COVID-19 period there was no change in the number of suicides overall, or in any stratum-specific estimates except one. The exception was an increase in the number of young males who died by suicide in the COVID-19 period (adjusted RR 1.89 [95% CI 1.11–3.23]).The unadjusted analysis showed significant differences in suicide in the context of unemployment and relationship breakdown during the COVID-19 compared to the pre-COVID-19 period. Analysis showed an increase in the number of suicides occurring in the context of unemployment in the COVID-19 period (unadjusted RR 1.53 [95% CI 1.18–1.96]). In contrast, there was a decrease in the number of suicides occurring in the context of relationship breakdown in the COVID-19 period (unadjusted RR 0.82 [95% CI 0.67–0.99]). However, no significant changes were identified when the models were adjusted for possible over-dispersion, seasonality and non-linear trend.Conclusion: Although our analysis found no evidence of an overall increase in suicides after the pandemic began, the picture is complex. The identified increase in suicide in young men indicates that the impact of the pandemic is likely unevenly distributed across populations. The increase in suicides in the context of unemployment reinforces the vital need for mitigation measures during COVID-19, and for ongoing monitoring of suicide as the pandemic continues.

PurposeWhole-body three-dimensional scanning is a tool utilised for the collection of body girths, volume, and surface area measurements. Few studies have investigated the validity and repeatability of this technology. The aim of the present study was to investigate the test retest variability of the NX-16 body scanner (NX-16, TC2, Cary, North Carolina, USA).MethodsPhase one involved the measurement of a mannequin on 300 occasions (30 scans over 10 sessions). In phase two, 13 apparently healthy male participants were recruited; each participant was scanned a total of four times (two scans over two sessions). Stature, body mass, and body fat % were obtained. Fourteen girth measurements were obtained (chest, underbust, stomach, waist, seat, hip, R/L bicep, R/L thigh, R/L mid-thigh, and R/L calf). Coefficient of variation was calculated for measurements obtained.ResultsCoefficient of variation for phase one ranged from 0.0% for the R calf, to 3.3% for the L thigh measurement. For phase two, values were higher, ranging from 0.5% for calf and chest to 4.6% for thigh measurements.ConclusionsTest–retest variability of the measurements provided by the NX-16 body scanner varied according to body location. However, variability within measurements was low using a mannequin or human participant. The NX-16 body scanner (TC2, Cary, North Carolina, USA) may be a useful tool for tracking changes in body composition over time during large population studies.

Editorial on the Research Topic:\ud \ud \ud Heat Acclimation for Special Populations:\ud \ud \ud This heat acclimation for special population's Research Topic questions the "one size fits all" approach for heat adaptation and that it may not be appropriate for all populations. Therefore, to highlight these differences we endeavored to collect a set of studies on how heat acclimation may benefit a wide range of special populations who have specific needs.\ud \ud \ud We have published 12 articles in this Research Topic and defined four main areas of research. (a) an epidemiological approach and the aging process; (b) understanding physiological mechanisms and a novel heat acclimation method; (c) adaptation to the heat for special populations including males, females, military personnel and Paralympic athletes; and (d) the use of heat therapy for special populations. We have summarized the most noteworthy evidence of each study in these research areas.

Progression of technology and computational power have led to a new age in data where the number of variables, p, is greater than the number of observations, n. These types of data, commonly called High-Dimensional Low Sample Size (HDLSS) data, are becoming prominent in statistical applications. One such HDLSS application where small samples are unavoidable is in the development and pre-clinical assessment of new drugs, such as in repeated low-dose challenge (RLC) studies. In RLC experiments, animals are assigned to an active or placebo candidate vaccine and then are repeatedly challenged (exposed) with some target pathogen, either until infection or some maximum number of challenges is reached (Nolen et al., 2015). Many times, the number of animals n in an RLC study is small (e.g. ≤ 20) and number of features p is large (e.g. ≥ 100), due to the high cost of each animal and the high number of antibody and functional measure features of interest (Chaudhury et al., 2018; Choi et al., 2015). Penalized regression techniques, like the lasso, are sometimes used in RLC experiments where n is typically small and p is large. However, the performance of such methods is not well established for this experiment setting. The performance of the lasso, elastic net, and a newly proposed discrete survival time penalized regression model is assessed via a simulation study. These methods are also applied to a recent RLC study evaluating a candidate HIV vaccine. All three methods rarely selected true positives regardless of the effect size, number of predictors, or the number of non-zero coefficients, with many models containing only false positives. Thus, penalized regression models should be used cautiously in the RLC setting when n is small and p is large. To improve upon penalized regression in the RCL setting, a recently-developed high-dimensional test known as the direction-projection-permutation (DiProPerm) test is suggested and adapted to the RLC setting. The DiProPerm test was designed specifically for the HDLSS setting and has many alluring qualities. The DiProPerm test is applied to the RLC setting to test whether animals are more likely to become infected early (i.e., before the median infection time) as opposed to late, given a set of antibody and functional measurements. The DiProPerm test has never been implemented in RLC settings as a valid tool for inference until now. Simulation processes revealed the advantages of using the DiProPerm test on RLC data when n is small and p is large. An RLC study evaluating a candidate HIV vaccine is used to demonstrate the DiProPerm test on a real-world dataset. To help disseminate the DiProPerm test to researchers, an R package was created. The diproperm R package can be used to conduct a DiProPerm test, display corresponding plots of interest, and look at the loadings of the binary linear classifier. The functionality of the diproperm package is explained and demonstrated on a real-world data set. The R package is freely available on CRAN and GitHub (https://github.com/allmondrew/diproperm) for anyone to use.

Introduction Investigate the effectiveness of short-term heat acclimation (STHA), over 5-days (permissive dehydration), on an intermittent sprint exercise protocol (HST) with females. Controlling for menstrual cycle phase. Materials and Methods Ten, moderately trained, females (Mean [SD]; age 22.6 [2.7] y; stature 165.3 [6.2] cm; body mass 61.5 [8.7] kg; V.⁢O2⁢peak 43.9 [8.6] mL⋅kg–1⋅min–1) participated. The HST (31.0°C; 50%RH) was 9 × 5 min (45-min) of intermittent exercise, based on exercise intensities of female soccer players, using a motorized treadmill and Wattbike. Participants completed HST1 vs. HST2 as a control (C) trial. Followed by 90 min, STHA (no fluid intake), for five consecutive days in 39.5°C; 60%RH, using controlled-hyperthermia (∼rectal temperature [Tre] 38.5°C). The HST3 occurred within 1 week after STHA. The HST2 vs HST3 trials were in the luteal phase, using self-reported menstrual questionnaire and plasma 17β-estradiol. Results Pre (HST2) vs post (HST3) STHA there was a reduction at 45-min in Tre by 0.20°C (95%CI −0.30 to −0.10°C; d = 0.77); T¯s⁢k (−0.50; −0.90 to −0.10°C; d = 0.80); and T¯b (−0.25; −0.35 to −0.15°C; d = 0.92). Cardiac frequency reduced at 45-min (−8; −16 to −1 b⋅min–1; d = 1.11) and %PV increased (7.0; −0.4 to 14.5%: d = 1.27). Mean power output increased across all nine maximal sprints by 56W (−26 to 139W; d = 0.69; n = 9). There was limited difference (P > 0.05) for these measures in HST1 vs HST2 C trial. Discussion Short-term heat acclimation (5-days) using controlled-hyperthermia, leads to physiological adaptation during intermittent exercise in the heat, in moderately trained females when controlling for menstrual cycle phase.

This study assessed the contribution of relative age, anthropometry, maturation, and physical fitness characteristics on soccer playing position (goalkeeper [GK], central-defender [CD], lateral-defender [LD], central-midfield [CM], lateral-midfielder [LM], and forward [FWD]) for 465 elite-youth players (U13–U18’s). U13–14 CD were relatively older than LD and CM (likely small effects). CD and GK were generally taller and heavier (likely small to very-likely moderate effects) than other players at each developmental stage and were advanced maturers at U13–14 (very-likely small to likely moderate effects). GK had inferior agility (very-likely small to likely moderate effects), endurance (very-likely small to likely moderate effects), and sprint capacities (likely small-moderate effects) vs. outfield positions at U13–14, but deficits in anaerobic phenotypes were diminished in U15–16 and U17–18. Position specific fitness characteristics were distinguished at U15–16 (likely small) and U17–18 (likely moderate), where LM were faster than their central counterparts. In summary, relative age, maturation and anthropometric characteristics appear to bias the allocation of players into key defensive roles from an early development stage, whereas position-specific physical attributes do not become apparent until the latter stages of talent development in outfield players. Given the inter-individual trajectories of physical development according to biological maturation, playing position allocation might be considered ‘plastic’ by selectors, until complete-maturity is achieved.

Objectives The principle aim of the current study was to examine within-match patterns of locomotor efficiency in professional soccer, determined as the ratio between tri-axial accelerometer data (PlayerLoad™) and locomotor activities. Between match variability and determinants of PlayerLoad™ during match play were also assessed. Design A single cohort, observational study. Methods Tri-axial accelerometer data (PlayerLoad™) was recorded during 86 competitive soccer matches in 63 English championship players (574 match observations). Accelerometer data accumulated (PlayerLoad Vector Magnitude [PL VM ]) from the individual-component planes of PlayerLoad™ (anterior–posterior PlayerLoad™ [PL AP ], medial–lateral PlayerLoad™ [PL ML ] and vertical PlayerLoad™ [PL V ]), together with locomotor activity (Total Distance Covered [TDC]) were determined in 15-min segments. Locomotor efficiency was calculated using the ratio of PL VM and TDC (PlayerLoad™ per metre). The proportion of variance explaining the within-match trends in PL VM , PL AP , AP ML , AP v , and TDC was determined owing to matches, individual players, and positional role. Results PL VM , PL AP , AP ML , AP v and TDC reduced after the initial 15-min match period ( p = 0.001; η 2 = 0.22–0.43, large effects). PL:TDC increased in the last 15 min of each half ( p = 0.001; η 2 = 0.25, large effect). The variance in PL VM during soccer match-play was explained by individual players (63.9%; p = 0.001) and between-match variation (21.6%; p = 0.001), but not positional role (14.1%; p = 0.364). Conclusions Locomotor efficiency is lower during the latter stages of each half of competitive soccer match-play, a trend synonymous with observations of increased injury incidence and fatigue in these periods. Locomotor efficiency may be a valuable metric to identify fatigue and heightened injury risk during soccer training and match-play.

The fat-tailed dunnart (Sminthopsis crassicaudata) is a small (10–20 g) native marsupial endemic to the south west of Western Australia. Currently little is known about the auditory capabilities of the dunnart, and of marsupials in general. Consequently, this study sought to investigate several electrophysiological and anatomical properties of the dunnart auditory system. Auditory brainstem responses (ABR) were recorded to brief (5 ms) tone pips at a range of frequencies (4–47.5 kHz) and intensities to determine auditory brainstem thresholds. The dunnart ABR displayed multiple distinct peaks at all test frequencies, similar to other mammalian species. ABR showed the dunnart is most sensitive to higher frequencies increasing up to 47.5 kHz. Morphological observations (Nissl stain) revealed that the auditory structures thought to contribute to the first peaks of the ABR were all distinguishable in the dunnart. Structures identified include the dorsal and ventral subdivisions of the cochlear nucleus, including a cochlear nerve root nucleus as well as several distinct nuclei in the superior olivary complex, such as the medial nucleus of the trapezoid body, lateral superior olive and medial superior olive. This study is the first to show functional and anatomical aspects of the lower part of the auditory system in the Fat-tailed dunnart.

Modeling and simulation is poised to transform drug development across the entire life cycle from discovery to commercialization. For the biopharmaceutical industry, this transformation will enable knowledge-based decision making and foster new collaborative ways of working that will translate into more high-value treatments and increased development efficiencies. In the health care arena, where value for money is paramount, modeling and simulation will inform future health care planning and practice.

Objective This study aimed to explore the effects of ambient temperature and relative humidity on insulin pharmacodynamics in adults with type 1 diabetes. Research Design A 3‐way, cross‐over, randomised study was performed in adults with type 1 diabetes mellitus (n=10). The pharmacodynamics profile of a single dose of short‐acting insulin (insulin lispro) was investigated under three environmental conditions: i) temperature: 15°C and humidity: 10%, ii) temperature: 30°C and humidity: 10%, and iii) temperature: 30°C and humidity: 60%, controlled in an environmental chamber. Euglycaemic glucose clamp technique ensured a constant blood glucose of 100 mg/dl (5.5 mmol/l). The following pharmacodynamic endpoints were calculated: maximum glucose infusion rate (GIRmax), time to GIRmax (tGIRmax), total area under the curve (AUC) for GIR from 0‐6 hours (AUCGIR.0–6h), and partial AUCs (AUCGIR.0‐1h, AUCGIR.0‐2h and AUCGIR.2‐6h). Results Higher temperature (30oC) under 10% fixed humidity resulted in a greater GIRmax (p=0.04), a later tGIR.max (p=0.049) compared to lower temperature (15oC). Humidity did not affect any pharmacodynamic parameter. When the combined effects of temperature and humidity were explored, tGIR.max (p=0.008) occurred earlier with a lower late insulin pharmacodynamic effect (AUCGIR.2‐6h, p=0.017) at temperature 15oC and humidity 10% compared to temperature 30oC and humidity 60%. Conclusions High ambient temperature resulted in greater insulin peak effect compared to low ambient temperature, with the contribution of high relative humidity only apparent at high ambient temperature. This suggests that patients with type 1 diabetes mellitus entering higher environmental temperatures with or without high humidity could experience more hypoglycaemic events.

Low intensity repetitive magnetic stimulation of neural tissue modulates neuronal excitability and has promising therapeutic potential in the treatment of neurological disorders. However, the underpinning cellular and biochemical mechanisms remain poorly understood. This study investigates the behavioural effects of low intensity repetitive magnetic stimulation (LI-rMS) at a cellular and biochemical level. We delivered LI-rMS (10 mT) at 1 Hz and 10 Hz (n=5 wells per group) to B50 rat neuroblastoma cells in vitro for 10 minutes and measured levels of selected metabolites immediately after stimulation. LI-rMS at both frequencies depleted selected tricarboxylic acid (TCA) cycle metabolites without affecting the main energy supplies. Furthermore, LI-rMS effects were frequency-specific with 1 Hz stimulation having stronger effects than 10 Hz. The observed depletion of metabolites was consistent with an increase in GABA release as a result of higher spontaneous activity. Although the absence of organised neural circuits and other cellular contributors (e.g. excitatory neurons and glia) in the B50 cell line limits the degree to which our results can be extrapolated to the human brain, the changes we describe provide novel insights into how LI-rMS modulates neural tissue.

Purpose:To assess the acute alterations in triaxial accelerometry (PlayerLoad [PLVM]) and its individual axial planes (anteroposterior PlayerLoad [PLAP], mediolateral PlayerLoad [PLML], and vertical PlayerLoad [PLV]) during a standardized 90-min soccer match-play simulation (SAFT90). Secondary aims of the study were to assess the test–retest reliability and anatomical location of the devices.Methods:Semiprofessional (n = 5) and university (n = 15) soccer players completed 3 trials (1 familiarization, 2 experimental) of SAFT90. PlayerLoad and its individual planes were measured continuously using micromechanical-electrical systems (MEMS) positioned at the scapulae (SCAP) and near the center of mass (COM).Results:There were no between-halves differences in PLVM; however, within-half increases were recorded at the COM, but only during the 1st half at the SCAP. Greater contributions to PLVM were provided by PLV and PLML when derived from the SCAP and COM, respectively. PLVM (COM 1451 ± 168, SCAP 1029 ± 113), PLAP (COM 503 ± 99, SCAP 345 ± 61), PLML (COM 712 ± 124, SCAP 348 ± 61), and PLV (COM 797 ± 184, SCAP 688 ± 124) were significantly greater at the COM than at the SCAP. Moderate and high test–retest reliability was observed for PlayerLoad and its individual planes at both locations (ICC .80–.99).Conclusions:PlayerLoad and its individual planes are reliable measures during SAFT90 and detected within-match changes in movement strategy when the unit was placed at the COM, which may have implications for fatigue management. Inferring alterations in lower-limb movement strategies from MEMS units positioned at the SCAP should be undertaken with caution.

Aims: To determine if clotting, platelet and endothelial function were affected by simulated short haul commercial air flight conditions (SF) in participants with type 2 diabetes (T2DM) compared to controls. Methods: 10 participants with T2DM (7 females, 3 males) and 10 controls (3 females, 7 males) completed the study. Participants were randomized to either spend 2 hours in an environmental chamber at sea level conditions (temperature: 23oC, oxygen concentration 21%, humidity 45%), or subject to a simulated 2-hour simulated flight (SF: temperature: 23oC, oxygen concentration 15%, humidity 15%), and crossed over 7 days later. Main outcome measures: clot formation and clot lysis parameters, functional platelet activation markers and endothelial function measured by reactive hyperaemia index (RHI) by EndoPAT and serum microparticles. Results: Comparing baseline with simulated flight conditions, clot maximal absorption was increased in controls (0.375±0.05 v 0.39±0.05, p
