Critical family history expands the frame of a life story beyond the accumulation of facts and figures to an acknowledgement of context, a deeper understanding of structure, a reckoning of circumstance and response and a comparison across time and space. This article explores the complexity of family history in the context of colonial pasts in British India; the possibilities offered by group analysis of colonial actors; and the moral obligation of the family historian to address difficult pasts in all their complexity. Through the migratory careers and migration stories of colonial actors—the dislocated people, objects and memories that sustain identity—a longitudinal dimension is added to family history. Taken collectively, the family history of a domiciled British community in India reveals not just important blood ties, but critical associational links and shared characteristics that structure experience and enhance power. Colonial power must always be measured by its negative effects, but is also relational, situational, variable, commutable and resisted. The article further reflects on the ways in which critical research into settler-colonial migrations delivers our family histories to the doorstep of the present; their possibilities for informing truth-telling at individual and national levels; and the need for a pedagogy of historical contextualisation and ethical citizenship.

PurposeThis paper discusses how service design can be used to activate a transition of textile artisan communities towards a sustainable future.Design/methodology/approachTwo participatory case studies were undertaken with textile artisans in the UK and South Africa. These led to the development of an original methodological framework for “crafting situated services” – services designed to be meaningful to the local communities within which they are embedded. An evaluation study assessed the originality of the framework, its relevance for tackling real-world problems, its extensibility and the rigour of the research process.FindingsThe framework brings together a variety of roles, methods and tools that designers can adopt in order to enter communities, make sense of sustainable futures, facilitate the co-design of situated services and activate legacies within communities. Building on emerging anthropological approaches, the framework makes a bridge between service management and service design for social innovation, advancing the field towards design for social entrepreneurship.Originality/valueArguing against the idea of the designer “parachuting” into communities to create services regardless of the local context, the concept of “situated services” is proposed in this paper, alongside a process for “crafting” meaningful social innovations. This requires the service designer to adopt a more situated and embedded approach to designing with communities in order to align with their needs and aspirations, interweave places, time, people and practices within the process, and co-design contextually better services.

A civic engagement and data science design was used to develop a report-back intervention to address stakeholder concerns related to air emissions surrounding a coke oven factory near Buffalo, NY, United States. This factory had historically emitted high levels of benzene pollution and ceased operation in October 2018 due to violations of the United States Clean Air Act and United States Resource Conservation and Recovery Act. Using publicly-available air pollution and weather data, descriptive time series and windrose data visualizations were developed using open-source software as part of a 2-page report-back brief. Data from two air toxics monitoring sites in this direction suggest that industrial sources were likely the major contributor to the benzene in the air at these locations prior to May 2018, after which traffic emissions became the likely major contributor. Windrose visualizations demonstrated that the wind typically blew towards the northeast, which was qualitatively consistent with locations of stakeholder concerns. With the factory closed, collective efforts subsequently shifted to address traffic emission air pollution sources, factory site clean-up, ground and water pollution mitigation. Because this intervention utilized open-source software and publicly-available data, it can serve as a blueprint for future data-driven nursing interventions and community-led environmental justice efforts.

Adjusting to university represents a substantial challenge in the lives of emerging adults and has been the focus of significant educational and personality psychology research. To date, however, no study has examined university adjustment as a function of sensory processing sensitivity (SPS) - a personality trait characterised by strong emotional reactivity and a heightened ability to detect and deeply process environmental stimulation, but with a consequent risk for debilitating feelings of overwhelm. Using a multiethnic sample of 580 first year South African psychology students, we examined university adjustment differences between students scoring high and low on SPS. Furthermore, we tested whether the effects of SPS on adjustment were a) independent from the five-factor model and b) moderated by levels of early parental care. We found that students scoring high (vs low) on SPS reported significantly worse adjustment to university. Opposite to theoretical claims, we only found a moderating effect for parental care for students low on SPS. By deconstructing SPS into its component factors, we discovered that poor adjustment was driven by a propensity towards negative affect, but the ability of high SPS individuals to carefully and deeply process stimulation served to partially offset adjustment difficulties. Importantly, SPS appeared to capture variation in university adjustment independent of the five-factor model. We conclude that SPS may be a useful personality construct for identifying students who find university adjustment particularly overwhelming and could thus benefit from targeted support and intervention.

Cardiovascular diseases (CVDs) are a leading cause of mortality and morbidity in South Africa. Risk stratification is the preferred approach to disease prevention, but identifying patients at high risk for CVD remains challenging. Assessing genetic risk could improve stratification and inform a clinically relevant precision medicine (PM) approach. Clinicians are critical to PM adoption, thus, this study explores practicing clinicians’ perceptions of PM-based CVD risk stratification in South Africa’s public health setting. Practicing clinicians (n = 109) at four teaching hospitals in Johannesburg, South Africa, completed an electronic self-administered survey. The effect of demographic and professional characteristics on PM-based CVD risk stratification perceptions was assessed. Fewer than 25% of respondents used clinical genetic testing, and 14% had formal genetics training. 78% had a low mean knowledge score, with higher scores associated with genetic training (p < 0.0005) and research involvement (p < 0.05). Despite limited knowledge and resources, 84% perceived PM approaches positively. 57% felt confident in applying the PM-based approach, with those already undertaking CVD risk stratification more confident (p < 0.001). High cost and limited access to genetics services are key barriers. Integrating genetic information into established clinical tools will likely increase confidence in using PM approaches. Addressing the genetics training gap and investment into the country’s genomics capacity is needed to advance PM in South Africa.

Objective To evaluate a method of measuring the change in palatal length and shape following maxillary advancement using synchronous lateral videofluoroscopy and voice recording in order to understand how movement of the maxilla may affect VPI risk in patients with cleft lip and/or palate (CL/P). Design Retrospective cohort study of children with cleft lip and/or palate. Setting Single center, tertiary children's hospital. Participants Patients with cleft lip and/or palate who underwent maxillary advancement between 2016–21 inclusive. Interventions Maxillary advancement surgery, including those who underwent concurrent mandibular procedures. Main outcome measures The length of the soft palate and the genu angle were measured throughout palatal dynamic range. Pre- and post-operative measurements were compared using a one sided T-test, with subgroup analysis for patients with clinical VPI. Results Ten patients were examined. The mean distance of maxillary advancement was 10.5 mm. The average increase in pre-genu soft palate length was 2.8 mm in the resting position and 2.9 mm in the closed position. The genu angle decreased in the closed position by 16.3 degrees. Conclusions The soft palate showed limited ability to lengthen following maxillary advancement and this may explain the risk of VPI. There was partial compensation by the muscle sling of the palate as demonstrated by a more acute post-operative genu angle and this suggests one reason for the variability of VPI reported. Future research is required to investigate how length and shape changes measured using this method can predict VPI risk.

Sensory Processing Sensitivity (SPS) is a personality trait that describes highly neurosensitive individuals who, for better and for worse, are permeable to their environmental context. Recently, these individuals have been noted for their amenability to positive intervention efforts - an observation that may have important psychosocial value. SPS is currently assessed through the 27-item Highly Sensitive Person Scale (HSPS). However, this instrument has not been adequately scrutinised in cross-cultural samples, and has generated varied factor solutions that lack consistent support. We assessed the HSPS in South African university student samples which were ethno-culturally diverse, across four academic years (n = 750). The HSPS demonstrated strong reliability across samples (α > .84). Factor analysis revealed a novel five-factor solution (Negative Affect, Neural Sensitivity, Propensity to Overwhelm, Careful Processing and Aesthetic Sensitivity). As per previous reports, latent class analysis suggested a three class solution. We validated these findings in a general population sample that was part of the longitudinal Birth to Twenty Plus cohort (n = 1400). In conclusion, we found the HSPS to be reliable in culturally diverse samples. The instrument remains a robust tool for identifying sensitive individuals and may be an important addition to psychosocial studies in low-to-middle income countries.

A massive transfusion protocol (MTP) was implemented at a Level I trauma center in 2007 for patients with massive blood loss. A goal ratio of plasma to pheresed platelets to packed red blood cells (PRBCs) of 1:1:1 was established. From 2007 to 2014, trauma nurse clinicians (TNCs) administered the MTP during initial resuscitation and anesthesia personnel administered the MTP intraoperatively. In 2015, TNCs began administering the MTP intraoperatively. This study evaluates intraoperative blood product ratios and crystalloid volume administered by anesthesia personnel or TNCs. A retrospective review of trauma registry patients requiring MTP from 2007 to 2017 was performed. Patient data were stratified according to MTP administration by either anesthesia personnel (2007–2015) or TNCs (2015–2017). Ninety-seven patients were included with 54 anesthesia patients and 44 TNC patients. Patients undergoing resuscitation by MTP administered by TNCs received less median crystalloid (3000 mL vs 1500 mL, P < 0.001). The ratio of plasma:PRBC (0.75 vs 0.93, P = 0.027) and platelets:PRBC (0.75 vs 1.04, P = 0.003) was found to be significantly closer to 1:1 for TNC patients. MTP intraoperative blood product administration by TNCs reduced the amount of infused crystalloid and improved adherence to MTP in achieving a 1:1:1 ratio of blood products.

Refereed/Peer-reviewed Background: antimicrobial silver has had a role in wound antisepsis throughout history and, with the rise in acquired antibiotic resistance, silver dressings are once again commonly used. Issues with silver dressings include the important environmental consideration of nanoparticle manufacture, and the significant financial cost of these products. One solution to these problems may be to adopt an opened-but-unused model of wound care whereby dressing materials are used in piecemeal fashion and excess stored in between dressing changes. Due to a lack of literature on the topic, this project was designed with the aim of testing the antimicrobial efficacy of available silver dressings during storage after opening. Methods: four commonly used silver dressings were tested for antimicrobial activity using a zone of inhibition assay against clinically important pathogens. The assay was performed on opening of dressings and repeated over 3 months in storage at 4, 25 or 37°C. Analysis was performed using repeated measures ANOVA. Swab cultures were taken at each simulated dressing change to detect microbial contamination of the dressings during storage. Results: there was no effect of time or storage temperature on the zone of inhibition over the 12 week test period. No swabs taken returned culture consistent with microbial contamination of stored dressings. Conclusion: opened silver dressings maintain antimicrobial activity for at least 12 weeks in storage and are resistant to contamination. An opened-but-unused model for wound care is likely to improve cost-effectiveness while preserving effectiveness and safety.

Service Design focuses on designing services which are usable and desirable from the users’ perspective, and efficient and different from the organizations’ perspective. However, users are diverse and possess a variety of capabilities, needs, and desires, which presents specific challenges for Service Designers. Inclusive Design is an approach to designing products and services that addresses the needs of the widest possible audience, irrespective of age or ability. Draw upon these two approaches, this paper aims to present the concept of inclusive service design as an essential element in designing better services. The potential benefits of such an approach were explored through a series of qualitative studies of the bus service in Guadalajara, Mexico.Based upon the research, several advantages for service improvement were identified, drawn from the richness of data generated by younger and older customers, and the uniqueness of the information gained from them. Likewise, the value of the big picture data obtained from stakeholders. This information highlights areas where service designers can help reduce the gap between what service providers do and what a broad range of users expect or need.

Skin is an important barrier to pathogenic microorganisms and plays a critical role in a ctivation of innate immune responses. When the skin barrier is breached following wounding or burn injury, pathogens can invade and complicate healing with infection resulting in delayed healing and symptomatic scarring. Wound infection is a significant problem after burn injury and in patients with chronic wounds. Antimicrobial silver has had a significant role in wound antisepsis throughout history and, given the rise in community acquired antibiotic resistance, silver dressings are now commonly used to combat wound infection. The multi-modal mechanism of action, low potential for toxicity and formation of microbial resistance makes silver dressings suitable tools against a wide array of clinically important microbes. There are, however, a number of issues with silver dressings including a conflicting evidence base, the important environmental consideration of nanoparticle manufacture, and the significant cost of these products. One solution may be to adopt an ‘opened-but-unused’ means of wound care whereby bulk dressing materials are used piecemeal and stored in between dressing changes to increase the cost-effectiveness and reduced wastage. Measurements of cosmic microwave background (CMB) polarization may reveal the presence of a background of gravitational waves produced during cosmic inflation, providing thus a test of inflationary models. The Q&U Bolometric Interferometer for Cosmology (QUBIC) is an experiment designed to measure the CMB polarization. It is based on the novel concept of bolometric interferometry, which combines the sensitivity of bolometric detectors with the properties of beam synthesis and control of calibration offered by interferometers. To modulate and extract the input polarized signal of the CMB, QUBIC exploits Stokes polarimetry based on a rotating half-wave plate (HWP). In this work, we illustrate the design of the QUBIC instrument, focusing on the polarization modulation system, and we present preliminary results of beam calibrations and the performance of the HWP rotator at 300 K.

Significance Cas9 proteins have revolutionized biotechnology by enabling flexible and facile targeted cleavage of nucleic acids. Nearly all of these proteins are active only at moderate, near-physiological temperatures. Through mini-metagenomic sequencing of hot spring samples from Yellowstone National Park, we discovered and characterized a novel hyperthermophilic Cas9 protein from an unculturable Ignavibacterium. This Cas9 protein, IgnaviCas9, expands the temperature range at which targeted nucleic acid cleavage is possible, thus speeding the development of new biotechnological techniques. We demonstrated one such application by using IgnaviCas9 to deplete undesired amplicons during the amplification step of library preparation in sequencing workflows. This Cas9 protein underscores the exciting applications that can be made possible by exploring nature’s diversity., Clustered regularly interspaced short palindromic repeats (CRISPR)-associated 9 (Cas9) systems have been effectively harnessed to engineer the genomes of organisms from across the tree of life. Nearly all currently characterized Cas9 proteins are derived from mesophilic bacteria, and canonical Cas9 systems are challenged by applications requiring enhanced stability or elevated temperatures. We discovered IgnaviCas9, a Cas9 protein from a hyperthermophilic Ignavibacterium identified through mini-metagenomic sequencing of samples from a hot spring. IgnaviCas9 is active at temperatures up to 100 °C in vitro, which enables DNA cleavage beyond the 44 °C limit of Streptococcus pyogenes Cas9 (SpyCas9) and the 70 °C limit of both Geobacillus stearothermophilus Cas9 (GeoCas9) and Geobacillus thermodenitrificans T12 Cas9 (ThermoCas9). As a potential application of this enzyme, we demonstrate that IgnaviCas9 can be used in bacterial RNA-seq library preparation to remove unwanted cDNA from 16s ribosomal rRNA without increasing the number of steps, thus underscoring the benefits provided by its exceptional thermostability in improving molecular biology and genomic workflows. IgnaviCas9 is an exciting addition to the CRISPR-Cas9 toolbox and expands its temperature range.

This study, with 19 households, set out to analyse the expected benefits and anticipated challenges to the introduction of smart home technology (SHT), and to compare these to post-use experiences after living with the technology for 1 year. Contextual interviews were undertaken with householders during the installation of a range of SHT, and again approximately 1 year later, when householders had the option to keep the technology or have some or all of it removed. This identified whether initial expectations were met, whether initial concerns persisted and whether new issues, concerns, and benefits (that had not been initially anticipated) arose after an extended period of use. Initial expectations from all households were high—related to comfort, convenience, improved control, energy demand reduction, and integration of technologies. There were also many initial concerns, including reliability, usability, and how these systems would be ‘domesticated’ to fit existing routines. After living with the systems for 1 year, many expectations were met, but the added value (and hence the match between expectations and reality) was higher for households with varied routines and large, partially occupied houses. Some of the anticipated challenges—such as concerns over aesthetics—largely disappeared, issues to do with the awkward location of network components persisted and new issues arose relating to the usability of some components and the time and effort required to configure them. Recommendations are given to maximise the added value that SHT can provide to householders.

Closed-cycle single-shot 4 He sorption coolers are used to provide cooling below 1 K in a number of mm-wave astronomical receivers. Helium, condensed by a 4 K stage, fills a coldhead; the vapour above the liquid is then driven off by a charcoal sorption pump to reduce the pressure, and hence temperature, of the liquid until thermal equilibrium is reached with the heat load. The coldhead then maintains this equilibrium until the liquid has completely evaporated, at which point the cooler is recycled. Below the λ -transition temperature, the mass transport of superfluid 4 He via the thermomechanical effect from the cold head into the pumping tube(s) has been shown to significantly reduce the hold time and cooling power of these systems. We present modelling and experimental demonstration of a mechanically simple and highly effective superfluid film breaker developed for the 1 K sorption cooler currently being used in the QUBIC experiment.

Type I CRISPR–Cas systems are the most abundant adaptive immune systems in bacteria and archaea1,2. Target interference relies on a multi-subunit, RNA-guided complex called Cascade3,4, which recruits a trans-acting helicase-nuclease, Cas3, for target degradation5–7. Type I systems have rarely been used for eukaryotic genome engineering applications owing to the relative difficulty of heterologous expression of the multicomponent Cascade complex. Here, we fuse Cascade to the dimerization-dependent, non-specific FokI nuclease domain8–11 and achieve RNA-guided gene editing in multiple human cell lines with high specificity and efficiencies of up to ~50%. FokI–Cascade can be reconstituted via an optimized two-component expression system encoding the CRISPR-associated (Cas) proteins on a single polycistronic vector and the guide RNA (gRNA) on a separate plasmid. Expression of the full Cascade–Cas3 complex in human cells resulted in targeted deletions of up to ~200 kb in length. Our work demonstrates that highly abundant, previously untapped type I CRISPR–Cas systems can be harnessed for genome engineering applications in eukaryotic cells. A type I CRISPR–Cas system, rather than the conventional type II CRISPR systems, is adapted for gene editing by fusing Cascade to the FokI nuclease domain.

Even though co-design is a well-accepted approach for designing to meet user needs, what influence it has on design outcomes remains unclear. This article presents the co-design process of a prototype energy advice service. We evaluate the impact this process had on the outcome over time, demonstrate how co-design generated informative insights, and identify the benefits and challenges of employing a co-design process to design and develop meaningful content for future ‘information-intensive’ services. A theoretical framework, a “think aloud” approach, and systematic data coding, enabled us to uncover user perceptions of the evolving design qualities. This meaning-making co-design process enabled user needs to surface and be iteratively addressed. As the content of the reports became increasingly tailored, and the users’ familiarity with the topic increased, the process highlighted further evolving and underlying information needs. This confirms the value of adopting a content first approach when designing information intensive services and foregrounding meaning making within the complex energy demand reduction context. Harnessing high-frequency spin dynamics in three-dimensional (3D) nanostructures may lead to paradigm-shifting, next-generation devices including high density spintronics and neuromorphic systems. Despite remarkable progress in fabrication, the measurement and interpretation of spin dynamics in complex 3D structures remain exceptionally challenging. Here, we take a first step and measure coherent spin waves within a 3D artificial spin ice (ASI) structure using Brillouin light scattering. The 3D-ASI was fabricated by using a combination of two-photon lithography and thermal evaporation. Two spin-wave modes were observed in the experiment whose frequencies showed nearly monotonic variation with the applied field strength. Numerical simulations qualitatively reproduced the observed modes. The simulated mode profiles revealed the collective nature of the modes extending throughout the complex network of nanowires while showing spatial quantization with varying mode quantization numbers. The study shows a well-defined means to explore high-frequency spin dynamics in complex 3D spintronic and magnonic structures.

International audience; The Q and U Bolometric Interferometer for Cosmology (QUBIC) Technical Demonstrator (TD) aiming to shows the feasibility of the combination of interferometry and bolometric detection. The electronic readout system is based on an array of 128 NbSi Transition Edge Sensors cooled at 350mK readout with 128 SQUIDs at 1K controlled and amplified by an Application Specific Integrated Circuit at 40K. This readout design allows a 128:1 Time Domain Multiplexing. We report the design and the performance of the detection chain in this paper. The technological demonstrator unwent a campaign of test in the lab. Evaluation of the QUBIC bolometers and readout electronics includes the measurement of I-V curves, time constant and the Noise Equivalent Power. Introduction The internet has improved access to knowledge, with a reported 72% of internet users accessing health information online. YouTube is the most popular video-sharing website and is a frequently accessed source for health information. There are concerns, however, that patients and carers may be unable to identify and filter for trustworthy sources of information on these platforms. This study aimed to assess the quality of videos that are likely to be accessed when families search YouTube for transcranial procedures for the management of craniosynostosis. Methodology This was a cross-sectional, observational study that evaluated YouTube videos when searching for “fronto-orbital advancement” or “cranial vault remodeling.” The first 25 videos for each term were included. Videos were assessed for quality using the previously validated JAMA and DISCERN scoring tools. Results There was low interobserver variability for the scoring tools used. There was no statistically significant correlation between the video quality and any video characteristics assessed. Videos published by organizations had higher mean quality scores for both the JAMA and DISCERN tools when compared to surgeon, patient, or medical device company-produced videos. Conclusion This study showed that searching YouTube for fronto-orbital advancement and cranial vault remodeling yields videos of low quality and therefore patients and carers should be discouraged from “YouTube-ing” these procedures. Platforms such as YouTube may provide an opportunity for craniofacial units to create and disseminate high-quality educational videos to complement routine perioperative education.
