- Abstract
ObjectivePulmonary hypertension (PH) is a cardiopulmonary disease manifesting in increased pulmonary arterial pressure and right ventricular dysfunction. PH pathogenesis involves extensive pulmonary vascular remodeling precipitated, at least in part, by endothelial reprogramming. Mounting evidence points to endothelial-to-mesenchymal transition (EndMT) as an important potentiator of endothelial reprogramming in PH, yet progress in dissecting these processes remains limited.Approach and ResultsLung samples from pulmonary arterial hypertension (PAH) patients and two rodent models of PH were used. Expression of the scaffolding protein ezrin-radixin-moesin-binding phosphoprotein 50 (EBP50, or NHERF1) was downregulated in PAH patient pulmonary arteries and isolated pulmonary arterial endothelial cells (PAECs), and in PH animal lung tissue and mouse isolated PAECs. In human PAECs in vitro, EBP50 was downregulated by PH-relevant stimuli, hypoxia and proinflammatory cytokine interleukin-1 beta (IL-1β). Phenocopy of EBP50 reduction in PAECs time-dependently increased expression and nuclear abundance of EndMT transcription factors Snail and Zeb1, and potentiated hypoxia-driven upregulation of Slug. Loss of EBP50 also drove expression of mesenchymal markers S100A4, fibronectin, N-cadherin, and transgelin (SM22), and inhibited cell proliferation and barrier function. In vivo studies on female EBP50+/- mice demonstrated that downregulation of EBP50 exacerbated the chronic hypoxia-induced rise in RV maximum pressure.ConclusionsThese data identify EBP50 as a key regulator of EndMT in PH whose expression is downregulated in PH patient pulmonary endothelium and whose partial deletion exacerbates PH disease manifestations in rodents, opening doors for future therapeutic strategies to target EBP50 restoration to reverse PH.

Description, A network dependency platform was generated to define a landscape of cancer drug mechanisms in pulmonary hypertension., Cancer therapies are being considered for treating rare noncancerous diseases like pulmonary hypertension (PH), but effective computational screening is lacking. Via transcriptomic differential dependency analyses leveraging parallels between cancer and PH, we mapped a landscape of cancer drug functions dependent upon rewiring of PH gene clusters. Bromodomain and extra-terminal motif (BET) protein inhibitors were predicted to rely upon several gene clusters inclusive of galectin-8 (LGALS8). Correspondingly, LGALS8 was found to mediate the BET inhibitor–dependent control of endothelial apoptosis, an essential role for PH in vivo. Separately, a piperlongumine analog’s actions were predicted to depend upon the iron-sulfur biogenesis gene ISCU. Correspondingly, the analog was found to inhibit ISCU glutathionylation, rescuing oxidative metabolism, decreasing endothelial apoptosis, and improving PH. Thus, we identified crucial drug-gene axes central to endothelial dysfunction and therapeutic priorities for PH. These results establish a wide-ranging, network dependency platform to redefine cancer drugs for use in noncancerous conditions.

Purpose This paper aims, first, to advance the current understanding about the impact of innovation in non-traditional exports, and, second, to provide insights about the structure of emergent economies often not regarded by traditional innovation and export theories. Design/methodology/approach A longitudinal analysis using panel data based on Box Jenkins’ theory was conducted, so to identify statistically significant variables on export performance, regarding expenditure on research, development and innovation (R&D + I) activities, ICT and specialized training and formation. Findings This study suggests the need to design public policies aimed at stimulating innovation in potential export sectors, as a mechanism for competitive development and growth in emergent economies such as Colombia. Originality/value The introduction of innovations in goods and services exports has become more important in economies, such as the Colombian ones, where globalization openness processes force to establish minimum competitiveness levels regarding the international standards.

This chapter analyzes the entrepreneurial strategies that Colombian coffee growers develop in order to deal with adverse social, economic, and environmental conditions. These entrepreneurs are part of a long and rich heritage dating as far back as the end of the 19th century, when coffee became an important economic resource in Colombia. Constant variations, including coffee price volatility, instability of exchange rates, or environmental factors, such as climatic change and crop disease, are common conditions for coffee farmers. In order to survive during turbulent environments, coffee growers have adopted strategies such as the introduction of new services and final products; improvements in the production chain; horizontal and vertical cooperation; creativity and flexibility in order to be resilient to the changing market conditions. More than 560,000 vulnerable families in Colombia depend on coffee production as a main source of income, so studies to help strengthen their business are highly relevant.

Human induced pluripotent stem cells (hiPSCs) are capable of unlimited proliferation and can differentiate in vitro to generate derivatives of the three primary germ layers. Genetic and epigenetic abnormalities have been reported by Wissing and colleagues to occur during hiPSC derivation, including mobilization of engineered LINE-1 (L1) retrotransposons. However, incidence and functional impact of endogenous retrotransposition in hiPSCs are yet to be established. Here we apply retrotransposon capture sequencing to eight hiPSC lines and three human embryonic stem cell (hESC) lines, revealing endogenous L1, Alu and SINE-VNTR-Alu (SVA) mobilization during reprogramming and pluripotent stem cell cultivation. Surprisingly, 4/7 de novo L1 insertions are full length and 6/11 retrotransposition events occurred in protein-coding genes expressed in pluripotent stem cells. We further demonstrate that an intronic L1 insertion in the CADPS2 gene is acquired during hiPSC cultivation and disrupts CADPS2 expression. These experiments elucidate endogenous retrotransposition, and its potential consequences, in hiPSCs and hESCs., Genetic and epigenetic abnormalities have been found to result from reprogramming of differentiated cells into human induced pluripotent stem cells (hiPSCs).
