To ensure Europe increases its domestic production of high quality and responsibly produced raw materials, the development of innovative technologies for 3D geological modeling in mineral exploration is paramount. The Erzgebirge in Germany provides an excellent framework to showcase the application of artificial intelligence and in particular Artificial Neural Networks (ANN) for 3D mineral prospectivity mapping. The Erzgebirge belongs to the Variscan Belt, withholding 800 years of mining history and it is also famous for Ag, Sn, W, Fe, Cu, Li mineralizations among others. The Bockau deposit is located at the western section of the Erzgebirge. The target area is a Paleozoic metasediment body that was formed during the Variscan orogeny. The metasediment body consists primarily of alternating micaschist, phyllite and quartzite and dips mostly 25° to 240° SW. The metasediment is surrounded by Late Variscan plutons which partly led to contact metamorphic zones. In addition there is a large Quartzite body which was mined near to the surface in the 17th century for Sn, following a stratiform tin anomaly which can reach up to 4000 ppm Sn. Thanks to the long mining history, the Bockau deposit condenses a large amount of geological, geochemical, geophysical and mineral data. To increase mineralogical knowledge of the deposit and to help identify drilling targets, a hybrid approach for 3D mineral predictivity mapping is implemented. Potentially mineralisation-controlling factors are identified in knowledge-driven genetic exploration models, taking into account the borehole data, major faults, electromagnetic data, intrusive bodies, contact metamorphic zones and lithological borders, followed by data-driven weighted ANN predictive modelling implemented in the in-house developed advangeo® 3D Prediction Software. The predictive model is guided by structural variables such as the euclidean distance to fault planes, lithological surfaces and to metamorphic contact zones. The model is also constrained by geophysical data by a magnetic susceptibility model obtained from an airborne magnetic data inversion. Finally, Sn anomaly data from boreholes is implemented as training data for the prediction. The results show the probability distribution of Sn mineralisation occurrence in 3D over a voxel model formed by blocks of approximately 684 m3 13(x), 13.5(y) and 4(z), increasing the mineralogical knowledge of the deposit and guiding exploration efforts complementing the decision making process for drilling new targets. The results are validated by iteratively implementing the jackknife method, splitting the training data into validation and training subsets. The first prediction iteration is performed with a subset containing 77 % of the Sn content data from boreholes as training data, followed by 50 and 30 % subsets. Thus, allowing at each iteration to perform a quantitative evaluation of the prediction by comparing the validation subset with the Sn content of the borehole that was not used for the prediction. From the middle of the 17th to the beginning of the 19th century, tin mining was carried out near Bockau and Aue in the Westerzgebirge. The object of the mining was stratiform mineralization. Near Bockau, (underground) tin mining was first mentioned in documents in 1663 and was active, with interruptions, until the beginning of the 19th century. On an area of more than 2 km², hundreds of dumps and pits, numerous abandoned mines, and historically very remarkable underground objects are known. Mining was preferably carried out near the surface. Despite favourable morphological conditions, hardly any deep adits were built and depths of more than 20 m were rarely reached. In this context, Pingen are abandoned ore pits or prospecting sites, where ores and other mineral resources were mined. Geometrically Pingen resemble round depressions created by the collapse of a mine workings (shaft, adit, underground drift), collapsed due to its age, leaving this relic usually funnel-shaped (down-facing cone), often surrounded by an annular dump (0.5 to 3 m) caused by the lowered surface.The main aim of this application is to identify mining relics (Pingen) using a UAV equipped with LiDAR technology. The LiDAR technology allows to obtain a high-resolution Digital Elevation Model (DEM) and Point cloud of the surveyed area. The DEM is the digital representation of topographic and manmade features located on the surface of the earth.In this framework, a LiDAR survey was conducted in a flight area of about 1,6 km² within the “Bockau” area during August 2022. The surveyed features include the elevations, colorized Point Cloud (RGB values), transparency levels, reflectance values, and number of returns (significant for the penetration of the vegetation). This information was used to derive the final products: DEM and classified Point cloud. Various spatial analyses were conducted and tested on the DEM and Point Cloud to automatically identify the mining relics. Hydrogeological analysis showed to be the best approach for the automatic identification of Pingen. As a result, the ground depressions were identified and nested surfaces were delineated.The automatically identified features were validated by examination of randomly selected samples on the surveyed point cloud, comparison to identified features based on the National dataset of the LiDAR Database and field verification. The validation revealed, that around 90% of the Pingen in the study area were successfully identified with the developed workflow. Other features of interests couldn’t be identified due to the similarity of geometric properties with other topographical features, dense vegetation, erosion etc.The paper has been prepared in the frame of the Horizon 2020 co-funded project GOLDENEYE, which has received funds through the Grant Agreement 869398.

Self-organizing maps (SOM) are a useful tool to analyze and interpret gridded datasets like potential field or stream sediment geochemistry data. The data are transformed from geographic space to SOM space where they can be clustered according to overall similarity. By transforming the clusters back to geographic space, geological interpretation of the clusters is facilitated. We present the application of a multilayer perceptron (MLP) in SOM space to produce mineral predictive maps. The reduced number of grid cells in SOM space greatly enhances the performance of the MLP and the tolerance to noise in the input data, compared to an application of the MLP to the original data. The method is applied to tin skarn deposits in the German part of the Erzgebirge. The training and validation data required for the MLP are compiled from mining and exploration records. The input data for the SOM are reprocessed gravimetric, magnetic, stream sediment geochemistry, geologic and tectonic data sets. Potentially ore-controlling spatial relationships, such as the distance to different types of partly covered granite intrusions, are derived from a regional scale 3D geological model. The resulting mineral prediction map allows the definition of exploration zones for detailed studies.The paper has been compiled in the frame of "NEXT - New EXploration Technologies" project. Background Plateletpheresis using a leukocyte reduction system (LRS) traps donor WBCs in the LRS chamber, which may lead to lymphopenia, especially in frequent plateletpheresis donors. It seems plausible that this might cause adverse effects. However, current knowledge about potential confounders and donor health impacts is incomplete. Donors and methods Recent platelet donors and donations collected at University Hospital Regensburg from 2016 to 2019 using the Terumo BCT Trima Accel LRS system were retrospectively analyzed and compared with historical platelet donors and donations collected mainly with Fresenius Kabi Amicus non-LRS system from 2010 to 2013. Additionally, recent donors were prospectively surveyed using a health-related topics questionnaire. Results Analysis of 819 recent donors with 11,254 blood counts and 1464 questionnaires and 1011 historical donors with 12,848 blood counts revealed that increased annual platelet donation frequencies were associated with decreased lymphocyte counts in both groups. BACKGROUND Granulocyte apheresis requires a sedimentation agent. Usually, hydroxyethyl starch (HES) is administered to donors for this purpose and, as granulocyte concentrate (GC) ingredient, also to patients. Authorities recently recommended suspending market authorizations for starch-containing products due to side effects. Therefore, we tested the efficacy of modified fluid gelatin (MFG, Gelafundin 4%) versus hetastarch (Hespan) for GC apheresis. STUDY DESIGN AND METHODS This retrospective matched-pair analysis compared MFG- and hetastarch-derived GCs. Each group consisted of 15 unrelated male donors mobilized with dexamethasone and granulocyte–colony-stimulating factor for apheresis on 1 or 2 days with the COBE Spectra's PMN program. RESULTS In each group, 24 GCs were collected from 15 male donors and analyzed. None of the HES-derived products, but two of the MFG-derived products (8.3%), had aggregates and could not be used. The HES-derived products had significantly higher neutrophil counts on the first day (7.7 × 1010/unit vs. 4.0 × 1010/unit; p = 0.00005) as well as second day of apheresis (4.0 × 1010/unit vs. 1.1 × 1010/unit; p = 0.0002). Median white blood cell collection efficacies were lower with MFG than with HES on Day 1 (24% vs. 43%) and Day 2 (15% vs. 37%). Twenty-one percent of the MFG-derived products had less than 1 × 1010 granulocytes. CONCLUSION These results indicate that granulocyte apheresis is feasible with MFG as well as with hetastarch and that the latter is superior for GC production, if used in the same dosage. In addition, aggregates in GC from the COBE Spectra were observed in the MFG group but not in the hetastarch group.

BACKGROUND Extracorporeal photopheresis (ECP) is a therapeutic technique that combines leukapheresis and ultraviolet (UV)A irradiation of the leukapheresate after 8-methoxypsoralen treatment with subsequent retransfusion. It can be achieved with a single device (online) or by combining an apheresis machine with a separate UVA light source (offline). The comparability of both established methods is unknown. STUDY DESIGN AND METHODS In a prospective setting, four ECP systems were evaluated: one with integrated UVA irradiation for online ECP (Therakos) and three with external UVA irradiation for offline ECP (Amicus, Optia, and Cobe Spectra). Apheresis variables and cell counts were determined by methods including flow cytometry. RESULTS The duration of apheresis ranged from 120 minutes (Amicus, Optia) to 275 minutes (Therakos). Mononuclear cell (MNC) counts in the treatment bags were comparable between offline ECP methods (median, 57 × 108 – 66 × 108) and lower for online ECP (14 × 108). CD16+ monocytes were abundant in online ECP (82%) but rarer in offline ECP (median, 14% – 19%). Hematocrit ranged from 0.1% (Therakos) to 8% (Amicus). There were no side effects in any patients. DISCUSSION All offline ECP systems studied yielded comparable cellular compositions and highly enriched populations of MNCs. In contrast, white blood cells from online ECP displayed enrichment of nonclassical monocytes. The relevance of these findings is unknown as there is no established biomarker to predict the therapeutic efficacy of these procedures.

Introduction Allogeneic haematopoietic stem cell transplantation (HSCT) is currently the only curative treatment for sickle cell disease (SCD) offered to patients with a fully matched sibling (MSD) or matched unrelated donor (MUD). With a MSD/MUD donor availability of below 20% a haploidentical HSCT from a family donor either with post-transplant cyclophosphamide (post-cy) or with a TCRαß/CD19 depleted graft (T-haplo) is an increasingly successful alternative for the majority of patients. In contrast to post-cy where mostly bone marrow is used, T-haploHSCT requires the generation of a G-CSF stimulated peripheral stem cell grafts. Almost uniformly haploidentical relatives of SCD patients are heterozygous carriers of SC trait. For these donors with a genetically heterozygous sickle trait, G-SCF represents a major safety concern with regard to triggering a sickle cell crisis. Therefore, the use of sickle trait donors is prohibited or not recommended in several countries, and guidelines are currently not available due to lack of evidence. Methods Haploidentical related donors with heterozygousSCD (group S, n=13) as well as healthy donors (group O, n=9) were stimulated with G-CSF. In all donors a similar mobilizations schedule using G-CSF was applied. All donors were applied 10µg/kg body weight (Neupogen®or Granocyte®) sub cutaneous daily with first apheresis on day 5 and day 6 when necessary. The amount of mobilized CD34+ cells were assessed after mobilization. In addition, the harvested stem cell preparations were analysed for CD34 content after apheresis and patients monitored for potential serious adverse effects (AE) during and after the apheresis. Results Mobilization was tolerated well with only mild and typical AE related to apheresis procedure such as citrate reactions, joint pain, ostealgia and headache, observed equally in both groups. In most of the donors (n=13/13 and 8/9), we were able to collect a sufficient amount of CD34+peripheral stem cells (mean of 16.3 and 8.2 x 10^6/kg in group S and O, respectively). We observed slight increases in LDH and reticulocyte counts in few donors also without significant differences between both groups. In particular, there were no severe neurologic side effects and no problems like sickle cell crisis despite mean pre-apheresis hemoglobin concentrations of 14.1 (range 11.9-17.5) g/dL in group S. Conclusions Collectively, these data indicate that in the absence of a MSD or MUD donor, haploidentical family donors with heterozygoussickle cell trait provide a safe and effective alternative for collection of peripheral CD34+stem cells for a T-haplo HSCT. Although burrowing ability has been widespread in tetrapods for more than 300 million years, subsurface dwelling structures that indicate communal behavior are poorly evidenced from pre-Cenozoic strata. Here we present recently discovered tetrapod burrows from Middle Triassic red beds of the Argana Basin in central Morocco, whose complexity suggests an origin by gregarious animals. The well-preserved burrows occur in interbedded mudstones and sandstones interpreted as channel and overbank deposits of ephemeral, braided streams. All burrows originate from the top of thick-bedded sandstones and descend as moderately inclined (10°–30°), partially spiral tunnels to laterally extended, branched chambers in underlying mudstones. Tunnel segments are biconvex to planoconvex in cross section, up to 20 cm wide and 12 cm in maximum height and exhibit transverse scratch marks along the ceilings and sidewalls. Distinctive burrow characteristics include a laterally sinuous geometry (wavelength λ = 38–45 cm; amplitude A = 5–10 cm) of the tubelike passages and the presence of grouped alcoves in terminal chambers. We attribute the burrows to procolophonids or therapsids based on closely associated tetrapod tracks and the limited diameter of the excavations. Our findings represent the second oldest record of communal fossorial behavior by tetrapods and the oldest example from low-latitude areas. Beyond providing refuge from predators, these elaborate underground structures probably functioned as a buffer against diurnal or seasonal variations of air temperature and humidity in a semiarid habitat that was situated just north of the paleoequator.

Footprints of Early Mesozoic terrestrial tetrapods are inadequately known from NW Africa despite pervasive and well-exposed continental strata of corresponding age in that region. Here, we report on the first occurrence of a tetrapod ichnofauna from the middle part of the Triassic Timezgadiouine Formation in the Argana Basin, Central Morocco that is outstanding in terms of abundance, preservation, and diversity. Footprints of this assemblage are assigned to the ichnotaxa Chirotherium barthii, Isochirotherium coureli, Synaptichnium isp., Atreipus–Grallator, Rotodactylus isp., Rhynchosauroides isp., and Procolophonichnium isp., suggesting early archosaurian, dinosauromorph, lepidosauromorph and possible therapsid or procolophonoid trackmakers. Based on comparison with the abundant record of Early Mesozoic vertebrate ichnofossils from Europe and North America, the ichnofauna presented here indicates a Middle Triassic (Anisian–Ladinian) age for the track-bearing horizon in the Argana Basin. Its clearly Euramerican affinity sheds light on the migration and dispersal of early archosaur-dominated tetrapod faunas considering that several ichnotaxa of the assemblage are documented from Africa for the first time.

Most comparative studies on neutrophil (PMN) isolation techniques have shown either activation or functional impairment of the cells due to the different separation processes. We have established a preparation method for PMN from human whole blood employing iron tagged, magnetizable antibodies against the cell surface antigen CD15. The aim of our study was to test whether this magnetic separation (MACS) alters cellular functions of PMN in comparison to a conventional density gradient technique (Percoll). The purity, cell yield, and pre-activation of the cells were evaluated. The latter was assessed by quantifying the expression of the integrin CD11b using flow cytometry. Furthermore, as functional tests, cell morphology and the oxidative burst reaction were investigated. We have shown that the use of 'magnetic' antibodies leads to highly purified PMN (> 99% of isolated leukocytes), while there is still contamination by eosinophils (about 6%) after Percoll separation. Platelet contamination was about the same in both procedures (approx. one platelet per two PMN). The basal expression of CD11b and, hence, neutrophil activation, was significantly lower and the upregulation of CD11b in response to FMLP was more pronounced after magnetic separation, as compared to density gradient centrifugation. The MACS technique did not lead to polarisation of PMN, nor did it affect the oxidative burst. This study suggests that magnetic separation is a simple, time-saving technique, yielding highly purified and functionally intact PMN.
