Scanning Photo-Induced Impedance Microscopy (SPIM) is an impedance imaging technique that is based on photocurrent measurements at field-effect structures. The material under investigation is deposited onto a semiconductor-insulator substrate. A thin metal film or an electrolyte solution with an immersed electrode serves as the gate contact. A modulated light beam focused into the space charge region of the semiconductor produces a photocurrent, which is directly related to the local impedance of the material. The absolute impedance of a polymer film can be measured by calibrating photocurrents using a known impedance in series with the sample. Depending on the wavelength of light used, charge carriers are not only generated in the focus but also throughout the bulk of the semiconductor. This can have adverse effects on the lateral resolution. Two-photon experiments were carried out to confine charge carrier generation to the spacecharge layer. The lateral resolution of SPIM is also limited by the lateral diffusion of charge carriers in the semiconductor. This problem can be solved by using thin silicon layers as semiconductor substrates. A resolution of better than 1 mu m was achieved using silicon on sapphire (SOS) substrates with a I l.Lm thick silicon layer.

The reproducible degradation of thin polymer films in the presence of an analyte or a reaction product of an analyte has potential applications in the development of highly sensitive, disposable biosensors. In this study, a novel combination of quartz crystal microbalance (QCM) and electrochemical impedance spectroscopy (EIS) has been developed to monitor the degradation of thin polymer films. Unlike a conventional QCM, the instrument described here allows rapid in situ measurement of quartz crystal impedance spectra. Simultaneously, classical electrochemical impedance spectra are measured in situ, affording the polymer film capacitance and bulk resistance. The combination of QCM impedance and classical EIS provides a wealth of information about the process of degradation of thin polymer films such as mass variation, swelling, delamination, viscoelasticity, and pore formation. Three different systems have been analyzed with this experimental setup; in two of the systems, polymer degradation was promoted by hydrolytic enzymes, and in the third one by a pH change. The results obtained show that the degradation of these three systems follows very different mechanisms. It is also underlined how the complementary information obtained by the two techniques allows a detailed description of the dissolution process.

A biosensor based on the enzyme-catalysed dissolution of biodegradable polymer films has been developed. Three polymer-enzyme systems were investigated for use in the sensor: a poly(ester amide), which is degraded by the proteolytic enzyme alpha-chymotrypsin; a dextran hydrogel, which is degraded by dextranase; and poly(trimethylene) succinate, which is degraded by a lipase. Dissolution of the polymer films was monitored by Surface Plasmon Resonance (SPR). The rate of degradation was directly related to enzyme concentration for each polymer/enzyme couple. The poly(ester amide)/alpha-chymotrypsin couple proved to be the most sensitive over a concentration range from 4 x 10(-11) to 4 x 10(-7) mol l(-1) of enzyme. The rate of degradation was shown to be independent of the thickness of the poly(ester amide) films. The dextran hydrogel/dextranase couple was less sensitive than the poly(ester amide)/alpha-chymotrypsin couple but showed greater degradation rates at low enzyme concentrations. Enzyme concentrations as low as 2 x 10(-11) mol l(-1) were detected in less than 20 min. Potential fields of application of such a sensor system are the detection of enzyme concentrations and the construction of disposable enzyme based immunosensors, which employ the polymer-degrading enzyme as an enzyme label.

A novel transducer based on the dissolution of biodegradable polymer films as a direct result of enzymatic reaction has been developed. Three polymers were investigated for use in the transducer: a poly(ester amide), which is degraded by the proteolytic enzyme alpha-chymotrypsin; a dextran hydrogel, which is degraded by dextranase; and poly(trimethylene) succinate, which is degraded by a lipase. Degradation of the polymer films was monitored by surface plasmon resonance (SPR) and impedance measurements. SPR was shown to be suitable for a greater variety of materials, since it does not require the polymer film to be electrically insulating. Rate of degradation was shown to be directly related to enzyme concentration for each polymer/enzyme couple. The poly(ester amide)/alpha-chymotrypsin couple proved to be the most sensitive. Degradation of the films was complete in less than 20 min for enzyme concentrations greater than 9 x 10(-9) mol dm-3. Enzyme concentrations as low as 4 x 10(-11) mol dm-3 were detected in less than 30 min. The transducer has great potential for the detection of enzyme concentrations as well as for use in immunosensing where the enzyme degrading the polymer would be the enzyme label.

the effects of water and hydrophobic solvents on the stability of the open and closed forms of R. miehei lipase were evaluated with different solvent models. Desolvation of arginine 86 at the water-lipid interface plays a key role in the activation process, while the contribution from hydrophobic stabilization of the open form is less important.

DNA sequence information underpins genetic research, enabling discoveries of important biological or medical benefit. Sequencing projects have traditionally used long (400-800 base pair) reads, but the existence of reference sequences for the human and many other genomes makes it possible to develop new, fast approaches to re-sequencing, whereby shorter reads are compared to a reference to identify intraspecies genetic variation. Here we report an approach that generates several billion bases of accurate nucleotide sequence per experiment at low cost. Single molecules of DNA are attached to a flat surface, amplified in situ and used as templates for synthetic sequencing with fluorescent reversible terminator deoxyribonucleotides. Images of the surface are analysed to generate high-quality sequence. We demonstrate application of this approach to human genome sequencing on flow-sorted X chromosomes and then scale the approach to determine the genome sequence of a male Yoruba from Ibadan, Nigeria. We build an accurate consensus sequence from >30x average depth of paired 35-base reads. We characterize four million single-nucleotide polymorphisms and four hundred thousand structural variants, many of which were previously unknown. Our approach is effective for accurate, rapid and economical whole-genome re-sequencing and many other biomedical applications.
