Abstract

The causative agent of human Q fever, Coxiella burnetii, is highly adapted to infect alveolar macrophages by inhibiting a range of host responses to infection. Despite the clinical and biological importance of this pathogen, the challenges related to genetic manipulation of both C. burnetii and macrophages have limited our knowledge of the mechanisms by which C. burnetii subverts macrophages functions. Here, we used the related bacterium Legionella pneumophila to perform a comprehensive screen of C. burnetii effectors that interfere with innate immune responses and host death using the greater wax moth Galleria mellonella and mouse bone marrow-derived macrophages. We identified MceF (Mitochondrial Coxiella effector protein F), a C. burnetii effector protein that localizes to mitochondria and contributes to host cell survival. MceF was shown to enhance mitochondrial function, delay membrane damage, and decrease mitochondrial ROS production induced by rotenone. Mechanistically, MceF recruits the host antioxidant protein Glutathione Peroxidase 4 (GPX4) to the mitochondria. The protective functions of MceF were absent in primary macrophages lacking GPX4, while overexpression of MceF in human cells protected against oxidative stress-induced cell death. C. burnetii lacking MceF was replication competent in mammalian cells but induced higher mortality in G. mellonella, indicating that MceF modulates the host response to infection. This study reveals an important C. burnetii strategy to subvert macrophage cell death and host immunity and demonstrates that modulation of the host antioxidant system is a viable strategy to promote the success of intracellular bacteria.

Abstract

Speech was human’s first great tool for communication. By developing sounds capable of creating meaning for a group, human beings were able to exchange important information, such as threats to the group. Writing has emerged only very recently in human history: about 5,500 years ago . The opposite was true in the history of the Internet: writing came first, then sound. The big leap only arrived in the 21st century, with Apple’s launch of Siri in 2011. From then on, these voice tools gained the definition of voice assistants, as they were able to listen to, respond and perform tasks through voice command. Since then, voice assistants have become popular and easily accessible to the global population. Currently five major voice assistants are available in Western markets. Abstract

The aim of this study was to evaluate the changes in osmoregulatory metabolism of cotton genotypes subjected to water deficit and recovery period. In a greenhouse, six cotton genotypes and two water managements were combined in a 6 x 2 factorial scheme, in a completely randomized design, evaluated after 14 and 22 days under water stress and recovery conditions, respectively. The water status, growth indicators and compatible solutes in the leaves were evaluated. The genotypes BRS Seridó, BRS Aroeira and BRS 7MH withstand better the stress condition because they had lower variations in their water potentials due to the osmotic adjustment and maintained their growth under water deficit conditions. The BRS 286 is the most sensitive genotype to the water deficit condition, since it showed greater variations in its water potential, which ultimately stopped its growth. Water replacement during the recovery period allowed growth resumption in most of the genotypes, mainly CNPA 5M and BRS 286. After the recovery period, variations in the concentrations of osmoregulators indicate the plasticity of cotton in regulating the concentrations of osmoregulators under favorable and unfavorable water conditions. Abstract

To report the incidence of preterm pre-eclampsia (PE) in women who are screen positive according to the criteria of the National Institute for Health and Care Excellence (NICE) and the American College of Obstetricians and Gynecologists (ACOG), and compare the incidence with that in those who are screen positive or screen negative by The Fetal Medicine Foundation (FMF) algorithm. This was a secondary analysis of data from the ASPRE study. The study population consisted of women with singleton pregnancy who underwent prospective screening for preterm PE by means of the FMF algorithm, which combines maternal factors and biomarkers at 11-13 weeks' gestation. The incidence of preterm PE in women fulfilling the NICE and ACOG criteria was estimated; in these patients the incidence of preterm PE was then calculated in those who were screen negative relative to those who were screen positive by the FMF algorithm. A total of 34 573 women with singleton pregnancy delivering at ≥ 24 weeks' gestation underwent prospective screening for preterm PE, of which 239 (0.7%) cases developed preterm PE. At least one of the ACOG criteria was fulfilled in 22 287 (64.5%) pregnancies and the incidence of preterm PE was 0.97% (95% CI, 0.85-1.11%); in the subgroup that was screen positive by the FMF algorithm the incidence of preterm PE was 4.80% (95% CI, 4.14-5.55%), and in those that were screen negative it was 0.25% (95% CI, 0.18-0.33%), with a relative incidence in FMF screen negative to FMF screen positive of 0.051 (95% CI, 0.037-0.071). In 1392 (4.0%) pregnancies, at least one of the NICE high-risk criteria was fulfilled, and in this group the incidence of preterm PE was 5.17% (95% CI, 4.13-6.46%); in the subgroups of screen positive and screen negative by the FMF algorithm, the incidence of preterm PE was 8.71% (95% CI, 6.93-10.89%) and 0.65% (95% CI, 0.25-1.67%), respectively, and the relative incidence was 0.075 (95% CI, 0.028-0.205). In 2360 (6.8%) pregnancies fulfilling at least two of the NICE moderate-risk criteria, the incidence of preterm PE was 1.74% (95% CI, 1.28-2.35%); in the subgroups of screen positive and screen negative by the FMF algorithm the incidence was 4.91% (95% CI, 3.54-6.79%) and 0.42% (95% CI, 0.20-0.86%), respectively, and the relative incidence was 0.085 (95% CI, 0.038-0.192). In women who are screen positive for preterm PE by the ACOG or NICE criteria but screen negative by the FMF algorithm, the risk of preterm PE is reduced to within or below background levels. Abstract

The generation of co-products continues at an accelerating pace, driven by population growth. Many of these co-products can be included in ruminant feed, which has the ability to transform them into good quality animal protein, and also help to reduce the disposal of undesirable residues to the environment. One of the co-products with potential use in animal feed, the citrus pulp, represents residues of the crushing of the fruit to remove the juice. In this review, these co-products are studied for their nutritional value, nutrient digestibility, and their influence on animal performance, in addition to ruminal characteristics. Due to its chemical composition, citrus pulp is widely used as an energetic component in the diet, replacing mainly maize, since these two have a similar energy value. In addition to increasing energy intake in the diet, citrus pulp has a greater capacity to assist in the maintenance of adequate ruminal performance compared to starchy cereals. Abstract

Objective: To investigate the relationship between fetal congenital heart defects (CHD) and placental perfusion assessed by uterine artery pulsatility index (UtA-PI), in relation to development of pre-eclampsia (PE).Methods: This was a prospective screening study of singleton pregnancies at 19-24 weeks' gestation. Transvaginal ultrasound was used to measure UtA-PI and the values were converted into multiples of the normal median (MoM). Median MoM values in pregnancies with a fetus with isolated major CHD were compared to those without CHD, in relation to development of PE.Results: The 91 407 singleton pregnancies fulfilling the entry criteria included 206 (0.23%) with isolated major fetal CHD and 91 201 without CHD. The prevalence of PE was 4.4% in pregnancies with fetal CHD and 2.7% in those without CHD (relative risk (RR), 1.6 (95% CI, 0.84-3.04); P = 0.150); the respective values for preterm PE with delivery at < 37 weeks' gestation were 2.4% and 0.7% (RR, 3.4 (95% CI, 1.42-8.09); P = 0.006). In the total population, median UtA-PI MoM was significantly higher in those that developed PE compared to those without PE (1.22 (interquartile range (IQR), 0.94-1.57) vs 1.00 (IQR, 0.84-1.19); P < 0.0001) and, in the PE group, the median UtA-PI MoM was inversely related to gestational age at delivery (r = -0.458; P < 0.0001). The same pattern of inverse relationship between UtA-PI MoM and gestational age at delivery with PE was observed in pregnancies with and those without CHD, but, in the CHD group, compared to those without CHD, UtA-PI was significantly higher both in pregnancies with and in those without PE.Conclusions: In pregnancies both with and without fetal CHD that develop PE, impedance to flow in the UtAs is increased and this increase is particularly marked in those with preterm PE. The prevalence of preterm PE is more than three times higher in pregnancies with than those without fetal major CHD, and the prevalence of major CHD in pregnancies with preterm PE is also more than three times higher than in those without PE. Abstract

Background Acute respiratory distress syndrome (ARDS) is associated with high in-hospital mortality. Alveolar recruitment followed by ventilation at optimal titrated PEEP may reduce ventilator-induced lung injury and improve oxygenation in patients with ARDS, but the effects on mortality and other clinical outcomes remain unknown. This article reports the rationale, study design, and analysis plan of the Alveolar Recruitment for ARDS Trial (ART). Methods/Design ART is a pragmatic, multicenter, randomized (concealed), controlled trial, which aims to determine if maximum stepwise alveolar recruitment associated with PEEP titration is able to increase 28-day survival in patients with ARDS compared to conventional treatment (ARDSNet strategy). We will enroll adult patients with ARDS of less than 72 h duration. The intervention group will receive an alveolar recruitment maneuver, with stepwise increases of PEEP achieving 45 cmH2O and peak pressure of 60 cmH2O, followed by ventilation with optimal PEEP titrated according to the static compliance of the respiratory system. In the control group, mechanical ventilation will follow a conventional protocol (ARDSNet). In both groups, we will use controlled volume mode with low tidal volumes (4 to 6 mL/kg of predicted body weight) and targeting plateau pressure ≤30 cmH2O. The primary outcome is 28-day survival, and the secondary outcomes are: length of ICU stay; length of hospital stay; pneumothorax requiring chest tube during first 7 days; barotrauma during first 7 days; mechanical ventilation-free days from days 1 to 28; ICU, in-hospital, and 6-month survival. ART is an event-guided trial planned to last until 520 events (deaths within 28 days) are observed. These events allow detection of a hazard ratio of 0.75, with 90% power and two-tailed type I error of 5%. All analysis will follow the intention-to-treat principle. Discussion If the ART strategy with maximum recruitment and PEEP titration improves 28-day survival, this will represent a notable advance to the care of ARDS patients. Conversely, if the ART strategy is similar or inferior to the current evidence-based strategy (ARDSNet), this should also change current practice as many institutions routinely employ recruitment maneuvers and set PEEP levels according to some titration method. Contains fulltext : 171632.PDF (publisher's version ) (Open Access), INTRODUCTION: Determining the optimal time to vaccinate is important for influenza vaccination programmes. Here, we assessed the temporal characteristics of influenza epidemics in the Northern and Southern hemispheres and in the tropics, and discuss their implications for vaccination programmes. METHODS: This was a retrospective analysis of surveillance data between 2000 and 2014 from the Global Influenza B Study database. The seasonal peak of influenza was defined as the week with the most reported cases (overall, A, and B) in the season. The duration of seasonal activity was assessed using the maximum proportion of influenza cases during three consecutive months and the minimum number of months with >/=80% of cases in the season. We also assessed whether co-circulation of A and B virus types affected the duration of influenza epidemics. RESULTS: 212 influenza seasons and 571,907 cases were included from 30 countries. In tropical countries, the seasonal influenza activity lasted longer and the peaks of influenza A and B coincided less frequently than in temperate countries. Temporal characteristics of influenza epidemics were heterogeneous in the tropics, with distinct seasonal epidemics observed only in some countries. Seasons with co-circulation of influenza A and B were longer than influenza A seasons, especially in the tropics. DISCUSSION: Our findings show that influenza seasonality is less well defined in the tropics than in temperate regions. This has important implications for vaccination programmes in these countries. High-quality influenza surveillance systems are needed in the tropics to enable decisions about when to vaccinate.

Abstract

The aim of this study was to evaluate ruminant and monogastric animal manure co-digested with 10 % of manipueira through process monitoring parameters and biogas production. In this study, eight semi-continuous digesters, with a capacity of 7.8 liters of substrate in fermentation, operated with 30 days of hydraulic retention time, were used. Monitoring analyses were performed in order to assess: pH, total ammonia nitrogen (TAN) concentration, partial alkalinity (PA), content and biogas yield (m3·kg·Volatile Solids (VS)added-1). There was no statistic difference for all the pH values, which reached values around 6.8 to 8.0. For PA, all the substrates reached higher than 1.200 mg·L-1 values as recommended. During the process, there was no risk of failure due to TAN concentration. The average biogas productions accumulated per week were 0.00676; 0.01167; 0.01515 and 0.01856 m3 for substrates composed by dairy cattle, sheep, poultry and swine manure co-digested with manipueira, respectively. The biogas yield for those respective substrates were 0.122; 0.275; 0.535 e 0.843 m3·kg·VSadded-1. The highest biogas yield was obtained for anaerobic co-digestion of swine manure with 10 % (volume/volume) of manipueira., Objetivou-se avaliar o uso de manipueira na co-digestão anaeróbia com dejetos de animais monogástricos e ruminantes, por meio de parâmetros de monitoramento do processo e da produção de biogás. Foram utilizados oito biodigestores semicontínuos com volume útil de 7,8 L de substrato em fermentação operados com tempo de retenção hidráulica de 30 dias. Realizou-se análises de pH, nitrogênio amoniacal, alcalinidade parcial (AP), sólidos totais (ST), sólidos voláteis (SV), e rendimento do biogás (m3 kg SVadicionados-1). Os valores de pH e concentrações de AP apresentaram-se na faixa ideal para a ocorrência do processo de digestão, de 6,0 a 8,0 e acima de 1.200 mg·L-1, respectivamente. Durante o processo não houve risco de falência por concentrações de nitrogênio amoniacal. As produções médias de biogás acumuladas semanais foram de 0,00676, 0,1167, 0,01515 e 0,01856 m3, respectivamente para substratos formados por dejetos de bovinos de leite, ovinos, aves e/ou suínos codigeridos com manipueira. Os rendimentos para os respectivos substratos foram 0,122, 0,275, 0,535 e 0,843 m3 kg SVadicionados-1. Maiores produções de biogás são obtidas na co-digestão anaeróbia de dejetos de suínos com 10 % (volume/volume) de manipueira.

Abstract

No Estado do Rio de Janeiro, Tem-se constatado, nos ultimos anos, uma diminuicao significativa da area cultivada com o arroz irrigado. Entretanto, com a utilizacao das cultivares indicadas pela pesquisa e das tecnologias indutivas de alta produtividade, esta elevou-se de 1.800 kh/ha para 3.474 kg/ha, mantendo estavel a producao de graos em torno de 100.000 toneladas/ano. Na busca de novas cultivares, vem sendo realizadas avaliacoes nas principais regioes orizicolas do Estado. No periodo de 1985 a 1993 foram conduzidos 23 ensaios de arroz nas regioes Norte (Campos dos Goitacazes, Noroeste (Itaperuna e Itaicara) e Baixada Litoranea (Macae), nos sistemas de semeadura direta e transplante de mudas. No conjunto desses locais, as medias de produtividade dos genotipos P 1377-1-15M-32-2-1 e CNA 5551 foram similares ao da testemunha INCA 4440, apresentando o mesmo porte de planta e precocidade e melhor qualidade de graos. Nos solos turfosos das baixadas litoraneas, alem de apresentarem maior potencial produtivo do que a testemunha, as novas cultivares mostraram-se tolerantes a brusone (Pyricularia grisea Cav.), doenca ja bastante disseminada na regiao. Com base nesses resultados, a partir de 1994/95, a Pesagro-Rio recomenda para o cultivo no Estado do Rio de Janeiro, as linhagens P 1377-1-15M-32-2-1 e CNA 5551, com as designacoes de PESAGRO 106 e PESAGRO 107, respectivamente. Abstract

This investigation reports the association between air pollution and paediatric respiratory emergency visits in São Paulo, Brazil, the largest city in South America. Daily records of emergency visits were obtained from the Children's Institute of the University of São Paulo for the period from May 1991 to April 1993. Visits were classified as respiratory and non-respiratory causes. Respiratory visits were further divided into three categories: upper respiratory illness, lower respiratory illness and wheezing. Daily records of SO2, CO, particulate matter (PM10), O3 and NO2 concentrations were obtained from the State Air Pollution Controlling Agency of São Paulo. Associations between respiratory emergency visits and air pollution were assessed by simple comparative statistics, simple correlation analysis and by estimating a variety of regression models. Significant associations between the increase of respiratory emergency visits and air pollution were observed. The most robust associations were observed with PM10, and to a lesser extent with O3. These associations were stable across different model specifications and several controlling variables. A significant increase in the counts of respiratory emergency visits--more than 20%--was observed on the most polluted days, indicating that air pollution is a substantial paediatric health concern in São Paulo.
