Historical wheat landraces are rich sources of genetic diversity offering untapped reservoirs for broadening the genetic base of modern varieties. Using a 20K SNP array, we investigated the accessible genetic diversity in a Central European bread wheat landrace collection with great drought, heat stress tolerance and higher tillering capacity. We discovered distinct differences in the number of average polymorphisms between Central and Western European collections, and identified a set of novel rare alleles present at low frequencies in the historical collection. The detected polymorphisms were unevenly distributed along the wheat genome, and polymorphic markers co-localized with genes of great agronomic importance. The efficiency of the highly diverse population for Genome-Wide Association study was confirmed and two significant marker trait associations with seed hardness were identified on the 5DS chromosome arm. The geographical distribution of the inferred Bayesian clustering revealed six genetically homogenous ancestral groups among the collection, where the Central European core bared an admixed background originating from four ancestral groups. We evaluated the effective population sizes (Ne) of the Central European collection and assessed changes in diversity over time, which revealed a dramatic ~97% genetic erosion between 1955 and 2015.

Chromatin–chromatin interactions and three-dimensional (3D) spatial structures are involved in transcriptional regulation and have a decisive role in DNA replication and repair. To understand how individual genes and their regulatory elements function within the larger genomic context, and how the genome reacts to environmental stimuli, the linear sequence information needs to be interpreted in three-dimensional space, which is still a challenging task. Here, we propose a novel, heuristic approach to represent Hi-C datasets by a whole-genomic pseudo-structure in 3D space. The baseline of our approach is the construction of a multigraph from genomic-sequence data and Hi-C interaction data, then applying a modified force-directed layout algorithm. The resulting layout is a pseudo-structure. While pseudo-structures are not based on direct observation and their details are inherent to settings, surprisingly, they demonstrate interesting, overall similarities of known genome structures of both barley and rice, namely, the Rabl and Rosette-like conformation. It has an exciting potential to be extended by additional omics data (RNA-seq, Chip-seq, etc.), allowing to visualize the dynamics of the pseudo-structures across various tissues or developmental stages. Furthermore, this novel method would make it possible to revisit most Hi-C data accumulated in the public domain in the last decade.

Knowledge of the degree of genetic diversity can provide fundamental information to breeders for use in various breeding programmes, for instance for the selection of lines with better adaptability. The genetic diversity analysis of 188 winter wheat genotypes demonstrated that this group of cultivars could be divided into four clusters based primarily on geographical origin. The first group contained mostly American and Asian cultivars, while cluster 2 consisted of Central European cultivars, cluster 3 of Hungarian and South European cultivars and cluster 4 mainly of cultivars from Western Europe. Cultivars used in breeding programmes in Central and South East European breeding programmes were found in all four clusters. Wheat genotypes originating from this region of Europe proved to have greater genetic variability than lines from Western and Northern Europe. Among the four clusters, there were also differencies in the frequencies of winter–spring alleles in Vrn-A1, Vrn-B1, Vrn-D1 vernalisation response genes and in the frequencies of sensitive–insensitive alleles in Ppd-B1 and Ppd-D1 photoperiod response genes, which explained the differences in heading date of the four clusters as well.

The reciprocal exchange of genetic information between homologous chromosomes during meiotic recombination is essential to secure balanced chromosome segregation and to promote genetic diversity. The chromosomal position and frequency of reciprocal genetic exchange shapes the efficiency of breeding programmes and influences crop improvement under a changing climate. In large genome cereals, such as wheat and barley, crossovers are consistently restricted to subtelomeric chromosomal regions, thus preventing favourable allele combinations being formed within a considerable proportion of the genome, including interstitial and pericentromeric chromatin. Understanding the key elements driving crossover designation is therefore essential to broaden the regions available for crossovers. Here, we followed early meiotic chromatin dynamism in cereals through the visualisation of a homologous barley chromosome arm pair stably transferred into the wheat genetic background. By capturing the dynamics of a single chromosome arm at the same time as detecting the undergoing events of meiotic recombination and synapsis, we showed that subtelomeric chromatin of homologues synchronously transitions to an open chromatin structure during recombination initiation. By contrast, pericentromeric and interstitial regions preserved their closed chromatin organisation and become unpackaged only later, concomitant with initiation of recombinatorial repair and the initial assembly of the synaptonemal complex. Our results raise the possibility that the closed pericentromeric chromatin structure in cereals may influence the fate decision during recombination initiation, as well as the spatial development of synapsis, and may also explain the suppression of crossover events in the proximity of the centromeres.

With a possible reference to heat priming and to characterize the extent and variation in the heat stress responses in wheat, the effects of single vs. repeated heat stresses were examined by measuring the changes in morphological and grain yield-related traits and photosynthetic parameters. To achieve these objectives, 51 winter wheat cultivars of various geographic origins were included in two independent experiments covering different phenological stages. In Experiment I, a single heat stress event was applied at stem elongation (SE) and booting (B), and the repeated heat stress was applied at both of these stages (SE+B). In Experiment II, the single heat stress was applied at stem elongation (SE) and full heading (CH), while the repeated heat stress was applied at both stages (SE+CH). While genotype was a more important factor for determining the morphological and yield-related traits, it was the treatment effect that mostly influenced the photosynthetic parameters, with the exception of the chlorophyll content. The heading stage was more sensitive to heat stress than the booting stage, which was primarily due to the larger decrease in the average seed number. The importance of biomass in contributing to grain yield intensified with the heat stress treatments. There was a large variation between the wheat cultivars not only in yielding abilities under control conditions but also in sensitivities to the various heat stresses, based on which 7 distinct groups with specific response profiles could be identified at a highly significant level. The 7 wheat groups were also characterized by their reaction patterns of different magnitudes and directions in their responses to single vs. repeated heat stresses, which depended on the phenological phases during the second cycle of heat stress. The possible association between these findings and heat priming is discussed.

Cytogenetic analysis and array-based SNP genotyping of wheat– Th. intermedium introgression lines allowed identification of 634 chromosome-specific SNP markers across all twenty-one chromosomes of Th. intermedium (StJ r J vs , 2 n = 6 x = 42). Thinopyrum intermedium (2n = 6x = 42, StJrJvs) is one of the most promising reservoirs of useful genes including tolerance to abiotic stresses, perenniality and disease resistance not available in the cultivated bread wheat. The transfer of genetic diversity from wild species to wheat offers valuable responses to the effects of climate change. The new array-based single-nucleotide polymorphism (SNP) marker technology provides cheap and easy-to-use molecular markers for marker-assisted selection (MAS) in wheat breeding programmes. Here, we focus on the generation of a new chromosome-specific SNP marker set that can be used to characterize and identify the Th. intermedium chromosomes or chromosome segments transferred into wheat. A progressive investigation of marker development was conducted using 187 various newly developed wheat–Th. intermedium introgression lines and the Axiom® Wheat-Relative Genotyping array. We employed molecular cytogenetic techniques to clarify the genome constitution of the Th. intermedium parental lines and validated 634 chromosome-specific SNPs. Our data confirmed the allohexaploid nature of Th. intermedium and demonstrated that the St genome-specific GISH signal and markers are present at the centromeric regions of chromosomes 1Jvs, 2Jvs, 3Jvs and 7Jvs. The SNP markers presented here will be introduced into current wheat improvement programmes, offering a significant speed-up in wheat breeding and making it possible to deal with the transfer of the full genetic potential of Th. intermedium into wheat.

Interspecific hybridization between bread wheat (Triticum aestivum, 2n = 42) and related species allows the transfer of agronomic and quality traits, whereby subsequent generations comprise an improved genetic background and can be directly applied in wheat breeding programmes. While wild relatives are frequently used as sources of agronomically favourable traits, cultivated species can also improve wheat quality and stress resistance. A salt-tolerant ‘Asakaze’/‘Manas’ 7H disomic addition line (2n = 44) with elevated β-glucan content, but with low fertility and an unstable genetic background was developed in an earlier wheat-barley prebreeding programme. The aim of the present study was to take this hybridization programme further and transfer the favourable barley traits into a more stable genetic background. Taking advantage of the breakage-fusion mechanism of univalent chromosomes, the ‘Rannaya’ winter wheat 7B monosomic line was used as female partner to the 7H addition line male, leading to the development of a compensating wheat/barley Robertsonian translocation line (7BS.7HL centric fusion, 2n = 42) exhibiting higher salt tolerance and elevated grain β-glucan content. Throughout the crossing programme, comprising the F1-F4 generations, genomic in situ hybridization, fluorescence in situ hybridization and chromosome-specific molecular markers were used to trace and identify the wheat and barley chromatin. Investigations on salt tolerance during germination and on the (1,3;1,4)-β-D-glucan (mixed-linkage glucan [MLG]) content of the seeds confirmed the salt tolerance and elevated grain MLG content of the translocation line, which can be directly applied in current wheat breeding programmes.

Chromosomes 5A (u) , 5S and 5D can be isolated from wild progenitors, providing a chromosome-based approach to develop tools for breeding and to study the genome evolution of wheat. The three subgenomes of hexaploid bread wheat originated from Triticum urartu (A(u)A(u)), from a species similar to Aegilops speltoides (SS) (progenitor of the B genome), and from Ae. tauschii (DD). Earlier studies indicated the potential of chromosome genomics to assist gene transfer from wild relatives of wheat and discover novel genes for wheat improvement. This study evaluates the potential of flow cytometric chromosome sorting in the diploid progenitors of bread wheat. Flow karyotypes obtained by analysing DAPI-stained chromosomes were characterized and the contents of the chromosome peaks were determined. FISH analysis with repetitive DNA probes proved that chromosomes 5A(u), 5S and 5D could be sorted with purities of 78-90 %, while the remaining chromosomes could be sorted in groups of three. Twenty-five conserved orthologous set (COS) markers covering wheat homoeologous chromosome groups 1-7 were used for PCR with DNA amplified from flow-sorted chromosomes and genomic DNA. These assays validated the cytomolecular results as follows: peak I on flow karyotypes contained chromosome groups 1, 4 and 6, peak II represented homoeologous group 5, while peak III consisted of groups 2, 3 and 7. The isolation of individual chromosomes of wild progenitors provides an attractive opportunity to investigate the structure and evolution of the polyploid genome and to deliver tools for wheat improvement.

3MbTriticum aestivum L. (Mv9kr1) – Aegilops biuncialis Vis. (MvGB642) addition lines were crossed with the Chinese Spring ph1b mutant genotype (CSph1b) to produce 3Mb–wheat chromosome rearrangements. In the F3generation, 3Mb(4B) substitution lines and 3Mb.4BS centric fusions were identified with in situ hybridization using repetitive and genomic DNA probes, and with SSR markers. Grain micronutrient analysis showed that the investigated Ae. biuncialis accession MvGB382 and the parental line MvGB642 are suitable gene sources for improving the grain micronutrient content of wheat, as they have higher K, Zn, Fe, and Mn contents. The results suggested that the Ae. biuncialis chromosome 3Mbcarries genes determining the grain micronutrient content, as the 3Mb.4BS centric fusion had significantly higher Zn and Mn contents compared with the recipient wheat cultivar. As yield-related traits, such as the number of tillers, the length of main spike, and spikelets per main spike, were similar in the 3Mb.4BS centric fusion and the parental wheat genotype, it can be concluded that this line could be used in pre-breeding programs aimed at enriching elite wheat cultivars with essential micronutrients.

CORE IDEAS We identified 1247 polymorphic single nucleotide polymorphisms between Triticum monococcum and wheat. We identified 191 markers validated across all seven chromosomes of T. monococcum. Detected a T. monococcum introgression in leaf-rust-resistant lines. Cultivated einkorn wheat (Triticum monococcum L. subsp. monococcum, 2n = 2x = 14, Am Am ) and its wild relative T. monococcum subsp. aegilopoides are important sources of economically useful genes that can be exploited for wheat (Triticum aestivum L.) breeding. Einkorn has excellent resistance to fungal diseases and gene transfer is relatively simple via standard breeding methods. To fulfill the growing demand by modern prebreeding programs for a cost-effective high-throughput procedure for accurately detecting introgressed chromosomes or chromosome segments from T. monococcum into wheat, we used the Axiom Wheat-Relative Genotyping Array and developed a set of Am genome-specific exome-based single nucleotide polymorphism (SNP) markers suitable for rapid identification of T. monococcum chromatin in a wheat background. We identified 1247 polymorphic SNPs between T. monococcum and wheat. We identified 191 markers across all seven chromosomes of T. monococcum that are also present on an existing Triticum urartu Thum. ex Gandil. genetic map and potentially ordered them on the basis of the high macrocollinearity and conservation of marker order between T. monococcum and T. urartu. The marker set has been tested on leaf-rust-resistant BC3 F4 progenies of wheat-T. monococcum hybrids. Two markers (AX-94492165, AX-95073542) placed on the distal end of the chromosome arm 7AL detected a T. monococcum introgression into wheat. The SNP marker set thus proved highly effective in the identification of T. monococcum chromatin in a wheat background, offering a reliable method for screening and selecting wheat-T. monococcum introgression lines, a procedure that could significantly speed up prebreeding programs.

as ´ ar, Hungary Abstract The dietary fibre (1,3;1,4)-β-D-glucan (β-glucan), is a major quality parameter of cereals. Grain β-glucan content is the most important factor from the aspect of human health maintenance. The grain of barley (Hordeum vulgare )i s one of the most important β-glucan sources having a β-glucan content 10 times higher than that of wheat (Triticum aestivum). Winter wheat/winter barley 'Mv9kr1'/'Igri' 1HS ditelosomic and 'Mv9kr1'/'Igri' 7H disomic addition lines carrying the HvCslF9 and HvCslF6 barley genes, respectively, were used to investigate the additive effect of barley cellulose synthase-like genes on the wheat β-glucan content. A significantly higher β-glucan level was detected in the leaves and grains of the wheat/barley 1HS and 7H addition lines compared to the control wheat line. The expression of the HvCslF9 and HvCslF6 genes in the genetic background of wheat was also determined by quantitative RT- PCR. The expression pattern of the HvCslF9 gene transcript showed a gradual increase throughout grain development, while the HvCslF6 gene was normally transcribed at relatively high levels. In leaves, the transcript of the HvCslF9 gene could not be detected at the end of tillering, while the HvCslF6 gene was still strongly expressed at this time in the 7H addition line. This study mapped the HvCslF9 gene to the short arm of the 1H chromosome. The HvGlb1 barley gene, encoding (1,3;1,4)-β-D-glucan endohydrolase isoenzyme EI, is possibly involved in the regulation of the β-glucan level during grain development. Previously this was also mapped to the barley 1H chromosome, and this study suggested that it was located on the 1HL chromosome arm. It was also concluded that the independent expression of the HvCslF9 gene in the wheat background resulted in a slight increase in the β-glucan content. The results provide new insights into the expression and regulation of the HvCslF genes in the genetic background of wheat and indicate that cisgenesis can be used to increase the leaf and grain β-glucan content in wheat.

The 4H(4D) wheat/barley substitution line was crossed with the ‘Chinese Spring’ ph1b mutant genotype in order to induce wheat-barley homoeologous recombinations. F3 and F4 seeds of the 4H(4D) × ‘Chinese Spring’ ph1b mutant cross were analysed using genomic in situ hybridization, and a Robertsonian translocation was detected in monosomic form. Disomic centric fusions were selected among the self-fertilized progenies. The presence of the long arm of 4H was confirmed with SSR markers. The long arm of the 5D wheat chromosome in the Robertsonian translocation was identified using fluorescent in situ hybridization with the help of three DNA probes: pSc119.2, Afa family and pTa71. The wheat/barley centric fusion was identified as a 4HL.5DL translocation. This line exhibited supernumerary spikelet character, but the number of seeds/plant did not increase. The 4HL.5DL centric fusion line is suitable genetic material to study the expression of genes located on 4HL in a wheat genetic background.

Goat grasses (Aegilops spp.) contributed to the evolution of bread wheat and are important sources of genes and alleles for modern wheat improvement. However, their use in alien introgression breeding is hindered by poor knowledge of their genome structure and a lack of molecular tools. The analysis of large and complex genomes may be simplified by dissecting them into single chromosomes via flow cytometric sorting. In some species this is not possible due to similarities in relative DNA content among chromosomes within a karyotype. This work describes the distribution of GAA and ACG microsatellite repeats on chromosomes of the U, M, S and C genomes of Aegilops, and the use of microsatellite probes to label the chromosomes in suspension by fluorescence in situ hybridization (FISHIS). Bivariate flow cytometric analysis of chromosome DAPI fluorescence and fluorescence of FITC-labelled microsatellites made it possible to discriminate all chromosomes and sort them with negligible contamination by other chromosomes. DNA of purified chromosomes was used as a template for polymerase chain reation (PCR) using Conserved Orthologous Set (COS) markers with known positions on wheat A, B and D genomes. Wheat-Aegilops macrosyntenic comparisons using COS markers revealed significant rearrangements in the U and C genomes, while the M and S genomes exhibited structure similar to wheat. Purified chromosome fractions provided an attractive resource to investigate the structure and evolution of the Aegilops genomes, and the COS markers assigned to Aegilops chromosomes will facilitate alien gene introgression into wheat.

In this article I give a brief overview about e-government and its’ opportunities, then I give a short description of a few important governmental and specialized administration on-line services which are provided to the farmers. According to many experts using of informatics may cause essential change in the operation of the administration because of the fact that the electronic administration may evolve. Generally speaking, today the egovernment services are already an indispensable device to the reformation of the public administration, the modification of services which suits to people's demands and the creation of the more adaptable, clearer public administration. The electronicdocuments are free from formal errors which is either important for clients or offices. The cases can be arranged even from home in 24 hours of the day. The spread and success of the use of electronic services depend not only on the quality of services, but the IT readiness of the target audience is also very important factor. Unfortunately the affect of e-gap is bigger in case of the farmers.

To increase the allelic variation in wheat–barley introgressions, new wheat–barley disomic addition lines were developed containing the 2H, 3H, 4H, 6H, and 7H chromosomes of the six-rowed Ukrainian winter barley ‘Manas’. This cultivar is agronomically much better adapted to Central European environmental conditions than the two-rowed spring barley ‘Betzes’ previously used. A single ‘Asakaze’ × ‘Manas’ wheat × barley hybrid plant was multiplied in vitro and one backcross plant was obtained after pollinating 354 regenerant hybrids with wheat. The addition lines were selected from the self-fertilized seeds of the 16 BC2plants using genomic in situ hybridization. The addition lines were identified by fluorescence in situ hybridization using repetitive DNA probes (HvT01, GAA, pTa71, and Afa family), followed by confirmation with barley SSR markers. The addition lines were grown in the phytotron and in the field, and morphological parameters (plant height, fertility, tillering, and spike characteristics) were measured. The production of the disomic additions will make it possible to incorporate the DNA of six-rowed winter barley into the wheat genome. Addition lines are useful for genetic studies on the traits of six-rowed winter barley and for producing new barley dissection lines.

Bidirectional allele-specific PCR (Bi-PASA) was used to detect a point mutation causing triazine resistance in common ragweed (Ambrosia artemisiifolia). The external primers amplified a 278 bp standard DNA fragment in all genotypes. In the susceptible S264S genotypes, a 208 bp fragment was expected while in resistant S264G common ragweed genotypes a 109 bp band was expected. In resistant plants, both the wild and mutant type fragments were detected, indicating that the original triazine sensitive cpDNA is maintained in a heteroplasmic state in the resistant S264G genotypes. Additionally, in silico analysis confirmed the potential applicability of our diagnostic assay for other plant species. In 24 out of 74 taxa (32%), the assay could be used without any change, while in the others some of the primers should be redesigned before use.

The subgenus Archaesolanum is a group composed of eight species with a characteristic chromosome number based on n = x = 23 and an area restricted to the South Pacific. This subgenus is an isolated group of Solanum for which extensive information about phylogenetic relationships based on molecular genetic methods is lacking. This study represents an approach to analyze genetic relationships within this group. In this context, seven species were examined using random amplified polymorphic DNA (RAPD) markers. In further analysis, the amplification products of two chloroplast regions (trnS-trnG and rbcL) were studied with polymerase chain reaction (PCR) restriction fragment length polymorphism (RFLP) method. Screening for the presence of unique mitochondrial rearrangements was also carried out using universal mitochondrial primers for the detection of fragment length polymorphisms. We identified two major groups within the subgenus; one was composed of the members of ser. Avicularia and Laciniata, while the other was formed by species belonging to ser. Similia. It is suggested that the taxonomic status of series within the Archaesolanum clade should be revised. The hybrid origin of S. laciniatum was also tested, and two hypotheses regarding its phylogeny are assumed.

The main objective of the present work was to develop a wheat genotype containing both the recessive crossability alleles (kr1kr1kr2kr2), allowing high crossability between 6x wheat and diploid rye, and the 1BL.1RS wheat/rye translocation chromosome. This wheat genotype could be used as a recipient partner in wheat-rye crosses for the efficient introduction of new allelic variation into 1RS in translocation wheats. After crossing the wheat cultivars 'Mv Magdaléna' and 'Mv Béres', which carry the 1BL.1RS translocation involving the 1RS chromosome arm from 'Petkus', with the line 'Mv9 kr1', 117 F(2) plants were analysed for crossability, ten of which had higher than 50% seed set with rye and thus presumably carried the kr1kr1kr2kr2 alleles. Four of the ten plants contained the 1BL.1RS translocation in the disomic condition as detected by genomic in situ hybridization (GISH). The wheat x rye F(1) hybrids produced between these lines and the rye cultivar 'Kriszta' were analysed in meiosis using GISH. 1BL.1RS/1R chromosome pairing was detected in 62.4% of the pollen mother cells. The use of fluorescent in situ hybridization (FISH) with the repetitive DNA probes pSc119.2, Afa family and pTa71 allowed the 1R and 1BL.1RS chromosomes to be identified. The presence of the 1RS arm from 'Kriszta' besides that of 'Petkus' was demonstrated in the F(1) hybrids using the rye SSR markers RMS13 and SCM9. In four of the 22 BC(1) progenies analysed, only 'Kriszta'-specific bands were observed with these markers, though the presence of the 1BL.1RS translocation was detected using GISH. It can be concluded that recombination occurred between the 'Petkus' and 'Kriszta' 1RS chromosome arms in the translocated chromosome in these plants.

In Hungary the Agricultural and Rural Development Agency (ARDA) started to introduce electronic filling and submission of application forms for the two-hundred-thousand farmers in 2008. For submission it is recommended to be registered using by client gate. The producers' benefits have more results from electronic filling. Filling in and submitting the applications electronically ensure data will be correct and the application technically flawless and this can be the basis on the payment of former financial support and avoidance of sanctions or subtractions due to technical faults. The farmers may ensure the money more quickly. Based on survey is analysed the users of system conceive of the electronic submission programme.

Hybrid plants and a high frequency of maternal haploids were obtained using an efficient wheat–barley hybridization system (with new genotype combinations) and confirmed by several cytological and molecular tools. An efficient hybridization system between wheat (Triticum aestivum L.) and barley (Hordeum vulgare L.) is presented on the basis of three new genotype combinations. A particularly high, 14 % frequency of plant regeneration per florets was achieved in the wheat–barley genotype combination of ‘Sichuan’ × ‘Morex’. The genome composition in 42 of the 95 plants regenerated by embryo rescue was determined using ploidy analysis, genomic in situ hybridization and the application of chromosome arm-specific molecular markers (SSR and STS). A high overall frequency (76 %) of maternal (wheat) haploids was observed in all the tests for all three cross combinations. A major implication of this observation is that this new hybridization system represents a useful tool to study the mechanism of uniparental chromosome elimination in cereals.

Public genomic databases have provided new directions for molecular marker development and initiated a shift in the types of PCR-based techniques commonly used in plant science. Alongside commonly used arbitrarily amplified DNA markers, other methods have been developed. Targeted fingerprinting marker techniques are based on the well-established practices of arbitrarily amplified DNA methods, but employ novel methodological innovations such as the incorporation of gene or promoter elements in the primers. These markers provide good reproducibility and increased resolution by the concurrent incidence of dominant and co-dominant bands. Despite their promising features, these semi-random markers suffer from possible problems of collision and non-homology analogous to those found with randomly generated fingerprints. Transposable elements, present in abundance in plant genomes, may also be used to generate fingerprints. These markers provide increased genomic coverage by utilizing specific targeted sites and produce bands that mostly seem to be homologous. The biggest drawback with most of these techniques is that prior genomic information about retrotransposons is needed for primer design, prohibiting universal applications. Another class of recently developed methods exploits length polymorphism present in arrays of multi-copy gene families such as cytochrome P450 and β-tubulin genes to provide cross-species amplification and transferability. A specific class of marker makes use of common features of plant resistance genes to generate bands linked to a given phenotype, or to reveal genetic diversity. Conserved DNA-based strategies have limited genome coverage and may fail to reveal genetic diversity, while resistance genes may be under specific evolutionary selection. Markers may also be generated from functional and/or transcribed regions of the genome using different gene-targeting approaches coupled with the use of RNA information. Such techniques have the potential to generate phenotypically linked functional markers, especially when fingerprints are generated from the transcribed or expressed region of the genome. It is to be expected that these recently developed techniques will generate larger datasets, but their shortcomings should also be acknowledged and carefully investigated.

Ebb?l a szempontból a KKV-k helyzete nehéz, hiszen ezen technológiák alkalmazása nem éri el a kívánatos szintet sem Magyarországon, sem az Európai unióban. A tanulmányban egy, a 2010 nyarán készült felmérés eredményeit mutatjuk be, amely az Észak-Alföldi régió elektronikus üzletviteli aktivitását méri. Ezt követ?en a KKV-k helyzetét és gazdasági szerepét ismertetjük a kiválasztott régióban, majd az adaptációs képességét vizsgáljuk meg. A felmérés menetét ismertetjük általánosan. Az összegy?jtött adatok alapján végrehajtott klaszterelemzés segítségével pedig arra a kérdésre keressük a választ, hogy a megkérdezett célcsoport milyen üzleti akció során milyen elektronikus üzletmenetet mennyire tart hasznosnak. A mez?gazdasági szektorban a kommunikációs csatornákra, a kapcsolódó partner és az üzleti folyamat típusokra alapozott klaszterelemzés alapján létrejött klaszterek segítik a vállalkozást a javasolt kommunikációs fejlesztések meghatározásában. Megítélésünk szerint ezzel segítséget nyújtunk a döntéshozóknak, hiszen a sz?kös er?forrásokkal rendelkez? vállalkozások célzottabban tudnak fejleszteni.

Az információ, mint új termelési tényez? jelent meg az agrárgazdaságban is. A megfelel? információnak a megfelel? id?ben való megléte a gazdálkodás eredményességét jelent?sen meghatározza. Napjainkban az interneten keresztül juthatunk leggyorsabban információhoz, ezért a vidéki területek szélessávú fejlesztése egyre jobban el?térbe kerül. Az agrárvállalkozások számára szintén fontos a szélessávú internet hozzáférés többek között az e-közigazgatás növekv? jelent?sége miatt, hiszen lehet?séget nyújt a törvényi szabályozások, rendeletek gyorsabb eléréséhez, tájékozódhatnak a beszerzési és értékesítési lehet?ségekr?l. A cikk el? részében az internet hozzáférés jelent?ségét és a szélessávú infrastruktúra vidéki jellemz?it ismertetjük. Ezek után kérd?íves felmérés eredményei alapján bemutatjuk, hogy a gazdák milyen módon és milyen célokra használják az internet szolgáltatásokat. A vizsgálataink alapján az infrastrukturális ellátottságban és az e-szolgáltatások használatában egyaránt elmaradás tapasztalható a nálunk fejlettebb európai országokhoz képest. Az információs társadalmi technológiák segíthetnek, hogy a kisebb agrárvállalkozások az elmaradottabb térségekben is életképesek, illetve versenyképesek lehessenek.

The effects of temperature and pH on the growth of 45 Hungarian Macrophomina phaseolina isolates from different locations and hosts were compared on the basis of their genetic diversity. One Spanish and two Serbian isolates were also included in the experiment. The most favourable temperature regimes for the development of the isolates ranged between 25 and 35 °C. The optimal pH for the pathogen varied between 4.0 and 6.0, but growth was observed on potato dextrose agar even at pH values of 3.0, 7.0 and 8.0. RAPD analysis with 13 different primer pairs generated 148 unambiguous bands. RFLP analysis involving 8 different restriction endonucleases was performed on a 1550 bp fragment of the rDNA region containing internal transcribed spacers (ITS1, ITS2), the 5.8S rDNA and part of the 25S rDNA. The greatest genetic distance values were obtained for three isolates, two from Hungary and one from Spain, which had similar values, but were quite distinct from all the others. A strong positive correlation was observed between the genetic distances and the growth parameters measured at various temperatures, and between the geographical data and the growth data sets at different pH values, but the correlation was less strong in the latter case. While Hungarian M. phaseolina populations are thought to reproduce clonally, the present results indicate the coexistence of different haplotypes in this area, and besides the geographical dominance of a given haplotype it was found that a closer genetic relationship might exist between spatially distinct haplotypes.

Common ragweed (Ambrosia artemisiifolia L.) is the most frequent weed in the Carpathian Basin and is spreading fast in other parts of Europe. In recent years, besides the wild type, a mutant genotype resistant to atrazine herbicides has evolved and is now widespread in many areas. The present study demonstrates that the atrazine resistance of ragweed is maternally inherited, and is caused by a point mutation inthe psb A chloroplast gene. The promoter 5′-untranslated region and the open reading frame regions of the gene were analysed, and a homology search was performed. Both the atrazine-resistant and susceptible types of cpDNA were present in atrazine-resistant plants, while the mixed presence of both genotypes in the same plant, known as heteroplasmy, was not unequivocally detectable in susceptible plants.

Diploid Aegilops umbellulata and Ae. comosa and their natural allotetraploid hybrids Ae. biuncialis and Ae. geniculata are important wild gene sources for wheat. With the aim of assisting in alien gene transfer, this study provides gene-based conserved orthologous set (COS) markers for the U and M genome chromosomes. Out of the 140 markers tested on a series of wheat-Aegilops chromosome introgression lines and flow-sorted subgenomic chromosome fractions, 100 were assigned to Aegilops chromosomes and six and seven duplications were identified in the U and M genomes, respectively. The marker-specific EST sequences were BLAST-ed to Brachypodium and rice genomic sequences to investigate macrosyntenic relationships between the U and M genomes of Aegilops, wheat and the model species. Five syntenic regions of Brachypodium identified genome rearrangements differentiating the U genome from the M genome and from the D genome of wheat. All of them seem to have evolved at the diploid level and to have been modified differentially in the polyploid species Ae. biuncialis and Ae. geniculata. A certain level of wheat-Aegilops homology was detected for group 1, 2, 3 and 5 chromosomes, while a clearly rearranged structure was showed for the group 4, 6 and 7 Aegilops chromosomes relative to wheat. The conserved orthologous set markers assigned to Aegilops chromosomes promise to accelerate gene introgression by facilitating the identification of alien chromatin. The syntenic relationships between the Aegilops species, wheat and model species will facilitate the targeted development of new markers specific for U and M genomic regions and will contribute to the understanding of molecular processes related to allopolyploidization.

� Background and Aims Aegilops markgrafii (CC) and its natural hybrids Ae. triuncialis (U t U t C t C t )a ndAe. cylindrica (D c D c C c C c ) represent a rich reservoir of useful genes for improvement of bread wheat (Triticum aestivum), but the limited information available on their genome structure and the shortage of molecular (cyto-) genetic tools hamper the utilization of the extant genetic diversity. This study provides the complete karyotypes in the three species obtained after fluorescent in situ hybridization (FISH) with repetitive DNA probes, and evaluates the potential of flow cytometric chromosome sorting. � Methods The flow karyotypes obtained after the analysis of 4’,6-diamidino-2-phenylindole (DAPI)-stained chromosomes were characterized and the chromosome content of the peaks on the flow karyotypes was determined by FISH. Twenty-nine conserved orthologous set (COS) markers covering all seven wheat homoeologous chromosome groups were used for PCR with DNA amplified from flow-sorted chromosomes and genomic DNA. � Key Results FISH with repetitive DNA probes revealed that chromosomes 4C, 5C, 7C t ,T 6U t S.6U t L-5C t L, 1C c and 5D c could be sorted with purities ranging from 66 to 91 %, while the remaining chromosomes could be sorted in groups of 2‐5. This identified a partial wheat‐C-genome homology for group 4 and 5 chromosomes. In addition, 1C chromosomes were homologous with group 1 of wheat; a small segment from group 2 indicated 1C‐2C rearrangement. An extensively rearranged structure of chromosome 7C relative to wheat was also detected. � Conclusions The possibility of purifying Aegilops chromosomes provides an attractive opportunity to investigate the structure and evolution of the Aegilops C genome and to develop molecular tools to facilitate the identification of alien chromatin and support alien introgression breeding in bread wheat.

A spontaneous interspecific Robertsonian translocation was revealed by genomic in situ hybridization (GISH) in the progenies of a monosomic 7H addition line originating from a new wheat ‘Asakaze komugi’ × barley ‘Manas’ hybrid. Fluorescence in situ hybridization (FISH) with repetitive DNA sequences (Afa family, pSc119.2, and pTa71) allowed identification of all wheat chromosomes, including wheat chromosome arm 4BS involved in the translocation. FISH using barley telomere- and centromere-specific repetitive DNA probes (HvT01 and (AGGGAG)n) confirmed that one of the arms of barley chromosome 7H was involved in the translocation. Simple sequence repeat (SSR) markers specific to the long (L) and short (S) arms of barley chromosome 7H identified the translocated chromosome segment as 7HL. Further analysis of the translocation chromosome clarified the physical position of genetically mapped SSRs within 7H, with a special focus on its centromeric region. The presence of the HvCslF6 gene, responsible for (1,3;1,4)-β-d-glucan production, was revealed in the centromeric region of 7HL. An increased (1,3;1,4)-β-d-glucan level was also detected in the translocation line, demonstrating that the HvCslF6 gene is of potential relevance for the manipulation of wheat (1,3;1,4)-β-d-glucan levels.
