The Chaos Canyon landslide, which collapsed on the afternoon of 28 June 2022 in Rocky Mountain National Park, presents an opportunity to evaluate instabilities within alpine regions faced with a warming and dynamic climate. Video documentation of the landslide was captured by several eyewitnesses and motivated a rapid field campaign. Initial estimates put the failure area at 66 630 m2, with an average elevation of 3555 m above sea level. We undertook an investigation of previous movement of this landslide, measured the volume of material involved, evaluated the potential presence of interstitial ice and snow within the failed deposit, and examined potential climatological impacts on the collapse of the slope. Satellite radar and optical measurements were used to calculate deformation of the landslide in the 5 years leading up to collapse. From 2017 to 2019, the landslide moved ∼ 5 m yr−1, accelerating to 17 m yr−1 in 2019. Movement took place through both internal deformation and basal sliding. Climate analysis reveals that the collapse took place during peak snowmelt, and 2022 followed 10 years of higher than average positive degree day sums. We also made use of slope stability modeling to test what factors controlled the stability of the area. Models indicate that even a small increase in the water table reduces the factor of safety to < 1, leading to failure. We posit that a combination of permafrost thaw from increasing average temperatures, progressive weakening of the basal shear zone from several years of movement, and an increase in pore-fluid pressure from snowmelt led to the 28 June collapse. Material volumes were estimated using structure from motion (SfM) models incorporating photographs from two field expeditions on 8 July 2022 – 10 d after the slide. Detailed mapping and SfM models indicate that ∼ 1 258 000 ± 150 000 m3 of material was deposited at the slide toe and ∼ 1 340 000 ± 133 000 m3 of material was evacuated from the source area. The Chaos Canyon landslide may be representative of future dynamic alpine topography, wherein slope failures become more common in a warming climate.

Bone marrow niches (endosteal and perivascular) play important roles in both normal bone marrow function and pathological processes such as cancer cell dormancy. Unraveling the mechanisms underlying these events in humans has been severely limited by models that cannot dissect dynamic events at the niche level. Utilizing microfluidic and stem cell technologies, we present a 3D in vitro model of human bone marrow that contains both the perivascular and endosteal niches, complete with dynamic, perfusable vascular networks. We demonstrate that our model can replicate in vivo bone marrow function, including maintenance and differentiation of CD34+ hematopoietic stem/progenitor cells, egress of neutrophils (CD66b+), and niche-specific responses to doxorubicin and granulocyte-colony stimulating factor. Our platform provides opportunities to accelerate current understanding of human bone marrow function and drug response with high spatial and temporal resolution.

Staphylococcus aureus (S. aureus) infections, including methicillin resistant stains, are an enormous burden on the healthcare system. With incidence rates of S. aureus infection climbing annually, there is a demand for additional research in its pathogenicity. Animal models of infectious disease advance our understanding of the host-pathogen response and lead to the development of effective therapeutics. Neutrophils play a primary role in the innate immune response that controls S. aureus infections by forming an abscess to wall off the infection and facilitate bacterial clearance; the number of neutrophils that infiltrate an S. aureus skin infection often correlates with disease outcome. LysM-EGFP mice, which possess the enhanced green fluorescent protein (EGFP) inserted in the Lysozyme M (LysM) promoter region (expressed primarily by neutrophils), when used in conjunction with in vivo whole animal fluorescence imaging (FLI) provide a means of quantifying neutrophil emigration noninvasively and longitudinally into wounded skin. When combined with a bioluminescent S. aureus strain and sequential in vivo whole animal bioluminescent imaging (BLI), it is possible to longitudinally monitor both the neutrophil recruitment dynamics and in vivo bacterial burden at the site of infection in anesthetized mice from onset of infection to resolution or death. Mice are more resistant to a number of virulence factors produced by S. aureus that facilitate effective colonization and infection in humans. Immunodeficient mice provide a more sensitive animal model to examine persistent S. aureus infections and the ability of therapeutics to boost innate immune responses. Herein, we characterize responses in LysM-EGFP mice that have been bred to MyD88-deficient mice (LysM-EGFP×MyD88-/- mice) along with wild-type LysM-EGFP mice to investigate S. aureus skin wound infection. Multispectral simultaneous detection enabled study of neutrophil recruitment dynamics by using in vivo FLI, bacterial burden by using in vivo BLI, and wound healing longitudinally and noninvasively over time.

The acidification of coastal waters is distinguished from the open ocean because of much stronger synergistic effects between anthropogenic forcing and local biogeochemical processes. However, ocean acidification research is still rather limited in polar coastal oceans. Here, we present a 16 year (2002–2018) observational dataset in the Chukchi Sea during the rapid sea‐ice melting season to determine the long‐term changes in pH and aragonite saturation state (Ωarag). We found that pH and Ωarag significantly declined in the water column with average rates of −0.0095 ± 0.0027 years−1 and −0.0333 ± 0.0098 years−1, respectively, and are 4–6 times faster than those solely due to increasing atmospheric CO2. We attributed the rapid acidification to the increased dissolved inorganic carbon owing to a combination of ice melt‐induced increased atmospheric CO2 invasion and subsurface remineralization induced by a stronger surface biological production as a result of the increased inflow of the nutrient‐rich Pacific water. Plain Language Summary: Anthropogenic CO2 absorbed by the ocean leads to a lower pH and the calcium carbonate saturation state (Ω) and threatens the marine ecosystems state of healthiness via a process called ocean acidification (OA). The Arctic Ocean is particularly sensitive to OA because more CO2 can be dissolved in cold water. This study used the observations collected over 16 years from 2002 to 2018 to estimate long‐term trends of Ωarag and pH in the Chukchi Sea. The results show that rapid acidification occurred in the water column during the rapid sea‐ice melting season from 2002 to 2018, leading to or approaching aragonite undersaturation. The rapid acidification is attributed to the enhanced increasing concentration of dissolved inorganic carbon. While sea ice melt induced uptake of anthropogenic CO2 partly explains the long‐term acidification, the remainder is due to the increased nutrient‐rich Pacific inflow water which promotes the high biological CO2 utilization in the surface waters but leads to stronger subsurface acidification due to the regenerated CO2. We suggest that the acidity in Chukchi Arctic Shelf waters will increase in the future if the increased inflow of Pacific water continues. High-resolution topographic modeling has become more accessible due to the development of structure from motion (SfM)-image-matching algorithms in digital photogrammetry. Large archival databases of historical aerial photographs are available in university, public, and government libraries, commonly as paper copies. The photographs can be in poor condition (i.e., deformed by humidity, scratched, or annotated). In addition, the negatives, as well as metadata, may be missing. Processing such photographs using classic stereo-photogrammetry is difficult and, in many instances, impossible. SfM can be applied to these photosets to access the valuable archive of geomorphic changes over the past century. In this paper, we illustrate the utility of the SfM technique using 568 digitized vertical aerial photographs of Mount Meager volcano, located in southwestern British Columbia, Canada. We use the aerial photographs, which span the period from 1947 to 2006, to track glaciers and glacier-landslide interactions on the volcano. Over this period, glaciers have thinned and retreated, interrupted by minor advances in the 1960s and 1970s. Landslides are frequent on the volcano and contribute to debris cover on the glaciers affecting the ablation process. SfM processing of the aerial photographs allowed us to unlock geomorphic information and reconstruct landscape change that would otherwise have been impossible. The results from SfM provide a visually effective way of presenting landscape change to a broad public audience, as a form of virtual geoheritage. The approach can thus be broadly applied in scientific and professional practices for improving land planning and hazard management. La modelisation topographique de haute resolution est devenue plus accessible en raison du developpement d'algorithmes de jumelage d'images de type << structure from motion >> (SfM, structure acquise a partir d'un mouvement) en photogrammetrie numerique. D'imposantes bases de donnees d'archives de photographies aeriennes sont disponibles dans les bibliotheques universitaires, publiques et gouvernementales, generalement en version papier. Ces photographies peuvent etre en mauvais etat (c.-a-d., deformees par l'humidite, egratignees ou annotees). En outre, des negatifs, ainsi que des metadonnees, peuvent manquer a l'appel. Le traitement de telles photographies par stereophoto-grammetrie classique est difficile, voire impossible dans bien des cas. La SfM peut etre appliquee a ses ensembles de photos pour acceder a l'important registre de changements geomorphologiques durant le siecle passe qu'ils renferment. Nous illustrons l'utilisation de la methode SfM en utilisant 568 photographies aeriennes verticales numerisees du volcan du mont Meager, dans le sud-ouest de la Colombie-Britannique (Canada). Nous utilisons les photographies aeriennes, qui couvrent la periode de 1947 a 2006, pour suivre les glaciers et les interactions entre glaciers et glissements de terrain sur le volcan. Cette periode est marquee par l'amincissement et le retrait des glaciers, interrompus par des avancees mineures dans les annees 1960 et 1970. Les glissements de terrain sont frequents sur le volcan et participent a l'accumulation d'un couvert de debris sur les glaciers qui influence le processus d'ablation. Le traitement SfM des photographies aeriennes nous a permis d'extraire de l'information geomorphologique et de reconstituer les changements du paysage, ce qui n'aurait pas ete possible autrement. Les resultats de SfM offrent un moyen visuellement efficace de presenter des modifications du paysage au grand public, comme geopatrimoine virtuel. The immune response to Staphylococcus aureus infection in skin involves the recruitment of polymorphonuclear neutrophils (PMNs) from the bone marrow via the circulation and local granulopoiesis from hematopoietic stem and progenitor cells (HSPCs) that also traffic to infected skin wounds. We focus on regulation of PMN number and function and the role of pore-forming α-toxin (AT), a virulence factor that causes host cell lysis and elicits inflammasome-mediated IL-1β secretion in wounds. Infection with wild-type S. aureus enriched in AT reduced PMN recruitment and resulted in sustained bacterial burden and delayed wound healing. In contrast, PMN recruitment to wounds infected with an isogenic AT-deficient S. aureus strain was unimpeded, exhibiting efficient bacterial clearance and hastened wound resolution. HSPCs recruited to infected wounds were unaffected by AT production and were activated to expand PMN numbers in proportion to S. aureus abundance in a manner regulated by TLR2 and IL-1R signaling. Immunodeficient MyD88-knockout mice infected with S. aureus experienced lethal sepsis that was reversed by PMN expansion mediated by injection of wild-type HSPCs directly into wounds. We conclude that AT-induced IL-1β promotes local granulopoiesis and effective resolution of S. aureus-infected wounds, revealing a potential antibiotic-free strategy for tuning the innate immune response to treat methicillin-resistant S. aureus infection in immunodeficient patients.
