Abstract

Anticancer immunotherapies, such as immune checkpoint inhibitors, bispecific antibodies, and chimeric antigen receptor T cells, have improved outcomes for patients with a variety of malignancies. However, most patients either do not initially respond or do not exhibit durable responses due to primary or adaptive/acquired immune resistance mechanisms of the tumor microenvironment. These suppressive programs are myriad, different between patients with ostensibly the same cancer type, and can harness multiple cell types to reinforce their stability. Consequently, the overall benefit of monotherapies remains limited. Cutting-edge technologies now allow for extensive tumor profiling, which can be used to define tumor cell intrinsic and extrinsic pathways of primary and/or acquired immune resistance, herein referred to as features or feature sets of immune resistance to current therapies. We propose that cancers can be characterized by immune resistance archetypes, comprised of five feature sets encompassing known immune resistance mechanisms. Archetypes of resistance may inform new therapeutic strategies that concurrently address multiple cell axes and/or suppressive mechanisms, and clinicians may consequently be able to prioritize targeted therapy combinations for individual patients to improve overall efficacy and outcomes.

Abstract

Discussions regarding how to embed culturally relevant and responsive literacy learning towards more socially just and equitable classrooms have prompted the need for additional research. This sequential exploratory mixed-methods study explores (a) 288 literacy teachers' perceptions of their abilities to address issues of bias, equity, and diversity in their classrooms, (b) literacy teacher beliefs on their practices and material resources reflecting intentionality towards diversity and equity, and (c) 12 literacy coaches' reflections and reactions to teacher perspectives concerning their ability to address issues of diversity in their classrooms. Findings reflect strong correlations between those who can identify bias and bring issues of diversity into their instruction; number of years of teaching experience as well as advanced degrees to diversity being recognized as an asset to educational systems; how effective communication fosters pluralistic classroom spaces; and the need for ongoing professional development for all teachers.

Abstract

Post-acute sequelae of COVID-19 (PASC) represent an emerging global crisis. However, quantifiable risk factors for PASC and their biological associations are poorly resolved. We executed a deep multi-omic, longitudinal investigation of 309 COVID-19 patients from initial diagnosis to convalescence (2-3 months later), integrated with clinical data and patient-reported symptoms. We resolved four PASC-anticipating risk factors at the time of initial COVID-19 diagnosis: type 2 diabetes, SARS-CoV-2 RNAemia, Epstein-Barr virus viremia, and specific auto-antibodies. In patients with gastrointestinal PASC, SARS-CoV-2-specific and CMV-specific CD8+ T cells exhibited unique dynamics during recovery from COVID-19. Analysis of symptom-associated immunological signatures revealed coordinated immunity polarization into four endotypes, exhibiting divergent acute severity and PASC. We find that immunological associations between PASC factors diminish over time, leading to distinct convalescent immune states. Detectability of most PASC factors at COVID-19 diagnosis emphasizes the importance of early disease measurements for understanding emergent chronic conditions and suggests PASC treatment strategies.

Abstract

The spatially resolved star formation histories are studied for 32 normal star-forming galaxies drawn from the the Spitzer Extended Disk Galaxy Exploration Science survey. At surface brightness sensitivities fainter than 28 mag arcsec$^{-2}$, the new optical photometry is deep enough to complement archival ultraviolet and infrared imaging and to explore the properties of the emission well beyond the traditional optical extents of these nearby galaxies. Fits to the spectral energy distributions using a delayed star formation history model indicate a subtle but interesting average radial trend for the spiral galaxies: the inner stellar systems decrease in age with increasing radius, consistent with inside-out disk formation, but the trend reverses in the outermost regions with the stellar age nearly as old as the innermost stars. These results suggest an old stellar outer disk population formed through radial migration and/or the cumulative history of minor mergers and accretions of satellite dwarf galaxies. The subset of S0 galaxies studied here show the opposite trend compared to what is inferred for spirals: characteristic stellar ages that are increasingly older with radius for the inner portions of the galaxies, and increasingly younger stellar ages for the outer portions. This result suggests that either S0 galaxies are not well modeled by a delayed-$\tau$ model, and/or that S0 galaxies have a more complicated formation history than spiral galaxies., Comment: Accepted for publication in the Astronomical Journal. arXiv admin note: text overlap with arXiv:1511.03285

42. An Analysis of Second Graders' Reading Motivation: Investigating Intrinsic and Social Factors

Abstract

Reading motivation has been identified as a significant factor that contributes to academic achievement. However, research has demonstrated that reading motivation declines as students progress through the elementary years. Exploring younger students' reading motivation is warranted as it can identify practices that foster and sustain intrinsic reading motivation. The purpose of this mixed methods study was to investigate if distinct instructional contexts affect students' reading motivation and teachers' perceptions of instructional practices that impact reading motivation. This study is framed by self-determination theory and expectancy-value theory. Second grade teachers (n = 9) and their students (n = 74) from two Northeastern suburban school districts were the participants in the study. Data collection included administration of the Me and My Reading Profile (MMRP) and was followed by teacher focus group interviews. Survey data was analyzed quantitatively for mean differences and correlations while subsequent interview data was analyzed using the constant comparative method. Significant correlations identified relationships between motivation and three constructs measured: value of reading, self-concept, and literacy out loud. Teacher perceptions of motivational pedagogies revealed student autonomy, self-concept, teacher influence, and social learning as dominant themes. This study extends the extant research that supports the role of motivation in literacy development and the necessity to include affective components into literacy instruction. Limitations include the small sample size and the use of self-reported data. Opportunities for future research are discussed as well as implications on practice. Abstract

BackgroundIt has been observed that married cancer patients have lower mortality rates than unmarried patients, but data for different racial/ethnic groups are scarce. The authors examined the risk of overall mortality associated with marital status across racial/ethnic groups and sex in data from the California Cancer Registry.MethodsCalifornia Cancer Registry data for all first primary invasive cancers diagnosed from 2000 through 2009 for the 10 most common sites of cancer-related death for non-Hispanic whites (NHWs), blacks, Asians/Pacific Islanders (APIs), and Hispanics were used to estimate multivariable hazard ratios (HRs) and 95% confidence intervals (CIs) for marital status in relation to overall mortality by race/ethnicity and sex. The study cohort included 393,470 male and 389,697 female cancer patients and 204,007 and 182,600 deaths from all causes, respectively, through December 31, 2012.ResultsAll-cause mortality was higher in unmarried patients than in married patients, but there was significant variation by race/ethnicity. Adjusted HRs (95% CIs) ranged from 1.24 (95% CI, 1.23-1.26) in NHWs to 1.11 (95% CI, 1.07-1.15) in APIs among males and from 1.17 (95% CI, 1.15-1.18) in NHWs to 1.07 (95% CI, 1.04-1.11) in APIs among females. All-cause mortality associated with unmarried status compared with married status was higher in US-born API and Hispanic men and women relative to their foreign-born counterparts.ConclusionsFor patients who have the cancers that contribute most to mortality, being unmarried is associated with worse overall survival compared with being married, with up to 24% higher mortality among NHW males but only 6% higher mortality among foreign-born Hispanic and API females. Future research should pursue the identification of factors underlying these associations to inform targeted interventions for unmarried cancer patients.
