Abstract

Over the past few decades, neuroimaging has become a ubiquitous tool in basic research and clinical studies of the human brain. However, no reference standards currently exist to quantify individual diferences in neuroimaging metrics over time, in contrast to growth charts for anthropometric traits such as height and weight1 . Here we assemble an interactive open resource to benchmark brain morphology derived from any current or future sample of MRI data (http://www.brainchart.io/). With the goal of basing these reference charts on the largest and most inclusive dataset available, acknowledging limitations due to known biases of MRI studies relative to the diversity of the global population, we aggregated 123,984 MRI scans, across more than 100 primary studies, from 101,457 human participants between 115 days post-conception to 100 years of age. MRI metrics were quantifed by centile scores, relative to non-linear trajectories2 of brain structural changes, and rates of change, over the lifespan. Brain charts identifed previously unreported neurodevelo pmental milestones3 , showed high stability of individuals across longitudinal assessments, and demonstrated robustness to technical and methodological diferences between primary studies. Centile scores showed increased heritability compared with non-centiled MRI phenotypes, and provided a standardized measure of atypical brain structure that revealed patterns of neuroanatomical variation across neurological and psychiatric disorders. In summary, brain charts are an essential step towards robust quantifcation of individual variation benchmarked to normative trajectories in multiple, commonly used neuroimaging phenotypes.

Abstract

In this study, the optical properties and dc resistivity of a series of FeSiAl(N) films reactively sputtered with different partial pressures of N were investigated. Spectroscopic ellipsometry was used to measure the real and imaginary parts of the complex dielectric functions. There is a distinct micro/nanostructural transition from single-phase columnar body-centered-cubic (bcc) grains for partial pressure (pp) of nitrogen in sputtering gas ⩽4% to a two-phase nanocomposite of equiaxed bcc nanograins in an amorphous matrix for films deposited with ⩾5% pp N. To assess the effect of surface oxidation on the optical properties, optical measurements were repeated on the 2 and 5% pp N films (representative of the two different types of films with different structures) after they were sputter etched in situ while performing depth profiling of the chemical composition using x-ray photoelectron spectroscopy. The low-nitrogen films (⩽4% pp N) showed a dielectric function typical of a metal whose charge carrier contribution can be described by a classical free electron Drude model. The nanostructured films (⩾5% pp N) showed a positive real part of the dielectric function ∊[SUB1] and no evidence of free-carrier plasmon excitation. The optical conductivity decreased and the dc resistivity increased by about a factor of 2.5 as the film structure changed from a single-phase columnar structure to the two-phase material that consisted of nanograins in an amorphous matrix. 26. Echocardiographic evidence for the existence of a distinct diabetic cardiomyopathy (the Framingham Heart Study)

Abstract

Although several reports have described early changes of cardiac structure and function in diabetic patients, controversy persists regarding the existence of a clinically distinct diabetic cardiomyopathy. To this end, sex-specific linear regression analyses were used to examine the contribution of diabetes mellitus and glucose intolerance to age-adjusted echocardiographic parameters in 1,986 men (mean age 48 years) and 2,529 women (mean age 50 years) from the original Framingham Study cohort and the Framingham Offspring Study. Subjects with evidence of cardiovascular disease at the time of echocardiogram were excluded. Diabetics had higher heart rates than nondiabetics (67.9 vs 64.0 beats/min (p = 0.002) in men, and 73.1 vs 68.3 beats/min (p = 0.004) in women). Diabetic women had increased left ventricular (LV) wall thickness (18.7 vs 17.1 mm, p less than 0.001), relative wall thickness (0.403 vs 0.377, p = 0.008), LV end-diastolic dimension (46.9 vs 45.7 mm, p = 0.03) and LV mass corrected for height (100.4 vs 82.2 g/m, p less than 0.001). Women with glucose intolerance showed similar, less significant trends (p = 0.007 for wall thickness, p less than 0.01 for LV mass). In diabetic men, fractional shortening was slightly reduced (0.355 vs 0.360, p less than 0.05). In a multivariate model that included potentially confounding factors, diabetes remained an independent contributor to LV mass (p = 0.004) and wall thickness (p = 0.008) in women. In a separate linear regression model, which assessed the association of age with LV mass, the age-coefficient for diabetic women was much higher than that for nondiabetics (13.6 vs 6.6 g/m per 10-year increment in age).(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract

Several inhibitors of the arachidonic acid-metabolizing enzyme, 5-lipoxygenase reduce proliferation of hematopoietic and non-hematopoietic cells and cell lines and some cells undergo limited differentiation. Cells were cultured from patients with chronic myelogenous leukemia in "blast" crisis with the selective inhibitor of 5-lipoxygenase,SC41661A[3-{3,5-bis(1,1-dimethyl)-4-hydroxyphenyl}hiol]-N-methyl-N-[2-(2-phridinyl-propanamide)]. Cells cultured for 3 to 5 days with 40 μM SC41661A exhibited reduced cellular numbers along with ultrastructural changes and DNA laddering characteristic of apoptosis. Similar culture conditions reduced proliferation of U937 monoblastoid cells. In U937 cells, the ultrastructural features of apoptosis were not observed at 72 hours, when DNA laddering was present and cell numbers were reduced, but was present after 144 hours of culture. Dissociation between certain morphologic and biochemical sequelae of apoptosis has been described in other systems. These observations are of interest since the induction of apoptosis in dividing chronic myelogenous leukemia (CML) cells by a noncytotoxic agent suggests paradigmatically new sites for therapeutic intervention.
