Outbreaks of infectious diseases involving depopulation of animals require on-farm practices to stage carcasses when final disposal methods are unavailable. The current study assessed various materials and techniques for containing carcasses to minimize leachate and biological substances. The tested materials included tarps, soil, corn stover (CS), and lime, while the methods involved covers, chemical additives, barriers, and containment. Treatments included the following: 1) control, carcasses in a pile; 2) carcasses wrapped in tarp material; 3) carcasses covered with tarp material; 4) carcasses covered by soil; 5) carcasses placed on CS base with tarp cover; 6) carcasses on CS base with CS covering; and 7) carcasses on CS base with a lime covering. Each treatment was run in triplicate using three carcasses per replicate. Temperatures, headspace gas, and leachate from carcasses were collected over a 91-day holding period. Pairwise comparisons of means were made when treatments were significantly different. Carcasses lost significant amounts of their liquid contents in the first 27 days. Leachate contents were initially filled with organic material and potassium that significantly declined with time, while Fe, Zn, and Cu concentrations increased significantly over time. Covers did not reduce leachate volume, but soil covers significantly reduced substances in the leachate. Corn stover barriers significantly reduced both leachate volume and substances in the leachate. Containing carcasses in tarp material was the most effective method for holding leachate and preventing its loss to the environment. Rapidly renewable tissues adapt different strategies to cope with environmental insults. While tissue repair is associated with increased intestinal stem cell (ISC) proliferation and accelerated tissue turnover rates, reduced calorie intake triggers a homeostasis-breaking process causing adaptive resizing of the gut. Here we show that activins are key drivers of both adaptive and regenerative growth. Activin-β (Actβ) is produced by stem and progenitor cells in response to intestinal infections and stimulates ISC proliferation and turnover rates to promote tissue repair. Dawdle (Daw), a divergent Drosophila activin, signals through its receptor, Baboon, in progenitor cells to promote their maturation into enterocytes (ECs). Daw is dynamically regulated during starvation-refeeding cycles, where it couples nutrient intake with progenitor maturation and adaptive resizing of the gut. Our results highlight an activin-dependent mechanism coupling nutrient intake with progenitor-to-EC maturation to promote adaptive resizing of the gut and further establish activins as key regulators of adult tissue plasticity., (© 2024. Tumor necrosis factor (TNF)-α is a highly conserved proinflammatory cytokine with important functions in immunity, tissue repair, and cellular homeostasis. Due to the simplicity of the Drosophila TNF-TNF receptor (TNFR) system and a broad genetic toolbox, the fly has played a pivotal role in deciphering the mechanisms underlying TNF-mediated physiological and pathological functions. In this review, we summarize the recent advances in our understanding of how local and systemic sources of Egr/TNF contribute to its antitumor and tumor-promoting properties, and its emerging functions in adaptive growth responses, sleep regulation, and adult tissue homeostasis. The recent annotation of TNF as an adipokine and its indisputable contribution to obesity- and cancer-associated metabolic diseases have provoked a new area of research focusing on its dual function in regulating immunity and energy homeostasis. It is well established that tumor necrosis factor (TNF) plays an instrumental role in orchestrating the metabolic disorders associated with late stages of cancers. However, it is not clear whether TNF/TNF receptor (TNFR) signaling controls energy homeostasis in healthy individuals. Here, we show that the highly conserved Drosophila TNFR, Wengen (Wgn), is required in the enterocytes (ECs) of the adult gut to restrict lipid catabolism, suppress immune activity, and maintain tissue homeostasis. Wgn limits autophagy-dependent lipolysis by restricting cytoplasmic levels of the TNFR effector, TNFR-associated factor 3 (dTRAF3), while it suppresses immune processes through inhibition of the dTAK1/TAK1-Relish/NF-κB pathway in a dTRAF2-dependent manner. Knocking down dTRAF3 or overexpressing dTRAF2 is sufficient to suppress infection-induced lipid depletion and immune activation, respectively, showing that Wgn/TNFR functions as an intersection between metabolism and immunity allowing pathogen-induced metabolic reprogramming to fuel the energetically costly task of combatting an infection.

A growing global meat demand requires a decrease in the environmental impacts of meat production. Cultured meat (CM) can potentially address multiple challenges facing animal agriculture, including those related to animal welfare and environmental impacts, but existing cost analyses suggest it is hard for CM to match the relatively low costs of conventionally produced meat. This study analyzes literature reports to contextualize CM's protein and calorie use efficiencies, comparing CM to animal meat products' feed conversion ratios, areal productivities, and nitrogen management. Our analyses show that CM has greater protein and energy areal productivities than conventional meat products, and that waste nitrogen from spent media is critical to CM surpassing the nitrogen use efficiency of meat produced in swine and broiler land-applied manure systems. The CM nutrient management costs, arising from wastewater treatment and land application, are estimated to be more expensive than in conventional meat production. Antimicrobial resistance (AMR) can develop in deep-pit swine manure storage when bacteria are selectively pressured by unmetabolized antibiotics. Subsequent manure application on row crops is then a source of AMR into soil and downstream runoff water. Therefore, understanding the patterns of diverse antibiotic resistance genes (ARGs) in manure among different farms is important for both interpreting the results of the detection of these genes from previous studies and for the use of these genes as bioindicators of manure borne antibiotic resistance in the environment. Previous studies of manure-associated ARGs are based on limited samples of manures. To better understand the distribution of ARGs between manures, we characterized manures from 48 geographically independent swine farms across Iowa. The objectives of this study were to characterize the distribution of ARGs among these manures and to evaluate what factors in manure management may influence the presence of ARGs in manures. Our analysis included quantification of two commonly found ARGs in swine manure, ermB and tetM . Additionally, we characterized a broader suite of 31 ARGs which allowed for simultaneous assays of the presence or absence of multiple genes. We found the company integrator had a significant effect on both ermB ( P=0.0007 ) and tetM gene concentrations ( P=0.0425 ). Our broad analysis on ARG profiles found that the tet(36) gene was broadly present in swine manures, followed by the detection of tetT , tetM , erm(35) , ermF , ermB , str , aadD , and intl3 in samples from 14 farms. Finally, we provide a comparison of methods to detect ARGs in manures, specifically comparing conventional and high-throughput qPCR and discuss their role in ARG environmental monitoring efforts. The control of organ size mainly relies on precise autonomous growth programs. However, organ development is subject to random variations, called developmental noise, best revealed by the fluctuating asymmetry observed between bilateral organs. The developmental mechanisms ensuring bilateral symmetry in organ size are mostly unknown. In Drosophila, null mutations for the relaxin-like hormone Dilp8 increase wing fluctuating asymmetry, suggesting that Dilp8 plays a role in buffering developmental noise. Here we show that size adjustment of the wing primordia involves a peak of dilp8 expression that takes place sharply at the end of juvenile growth. Wing size adjustment relies on a cross-organ communication involving the epidermis as the source of Dilp8. We identify ecdysone signaling as both the trigger for epidermal dilp8 expression and its downstream target in the wing primordia, thereby establishing reciprocal hormonal feedback as a systemic mechanism, which controls organ size and bilateral symmetry in a narrow developmental time window., (© 2022. Swine growers seeking to lower costs and environmental impact have turned to alternative carbohydrate feed sources. A feeding trial was conducted to determine the effect carbohydrate sources have on manure composition and gas emissions. A total of 48 gilts averaging 138 kg BW were fed diets consisting of (a) low fiber (LF) grain, or (b) high fiber (HF) aro-industrial co-product (AICP). The LF diets included corn and soybean meal (CSBM) and barley soybean meal (BSBM). The HF AICP diets were CSBM based and supplemented with one of the following materials: beet pulp; corn distillers dried grains with solubles; soybean hulls; or wheat bran. Diets were fed for 42 d with an average daily feed intake of 2.71 kg d -1 . Feces and urine were collected twice daily and added to manure storage containers in which manure slurries were monitored for gas emissions and chemical properties. Manures of animals fed HF diets had significantly (P < 0.05) more excretion of solids, C, N, and organic N, but less total S compared to pigs fed the LF diets. Animals feed HF diets had significantly (P < 0.05) higher levels of ammonia, sulfide, volatile fatty acids, and phenols in manure compared to pigs fed the LF diets. Manure of animals fed HF diets had 30% (P < 0.05) lower NH 3 and 17% lower hydrogen sulfide emissions; however, fiber had no impact on odor emissions. Based on the partitioning of nutrients, animals fed HF fiber diets had increased manure retention for C and N but decreased levels of N gas emissions and manure S. Foam accumulation in swine manure deep-pits has been linked to explosions and flash fires that pose devastating threats to humans and livestock. It is clear that methane accumulation within these pits is the fuel for the fire; it is not understood what microbial drivers cause the accumulation and stabilization of methane. Here, we conducted a 13-month field study to survey the physical, chemical, and biological changes of pit-manure across 46 farms in Iowa. Our results showed that an increased methane production rate was associated with less digestible feed ingredients, suggesting that diet influences the storage pit's microbiome. Targeted sequencing of the bacterial 16S rRNA and archaeal mcrA genes was used to identify microbial communities' role and influence. We found that microbial communities in foaming and non-foaming manure were significantly different, and that the bacterial communities of foaming manure were more stable than those of non-foaming manure. Foaming manure methanogen communities were enriched with uncharacterized methanogens whose presence strongly correlated with high methane production rates. We also observed strong correlations between feed ration, manure characteristics, and the relative abundance of specific taxa, suggesting that manure foaming is linked to microbial community assemblage driven by efficient free long-chain fatty acid degradation by hydrogenotrophic methanogenesis., Competing Interests: The authors have declared that no competing interests exist.

The Drosophila tumour necrosis factor (TNF) ligand-receptor system consists of a unique ligand, Eiger (Egr), and two receptors, Grindelwald (Grnd) and Wengen (Wgn), and therefore provides a simple system for exploring the interplay between ligand and receptors, and the requirement for Grnd and Wgn in TNF/Egr-mediated processes. Here, we report the crystallographic structure of the extracellular domain (ECD) of Grnd in complex with Egr, a high-affinity hetero-hexameric assembly reminiscent of human TNF:TNFR complexes. We show that ectopic expression of Egr results in internalisation of Egr:Grnd complexes in vesicles, a step preceding and strictly required for Egr-induced apoptosis. We further demonstrate that Wgn binds Egr with much reduced affinity and is localised in intracellular vesicles that are distinct from those containing Egr:Grnd complexes. Altogether, our data provide insight into ligand-mediated activation of Grnd and suggest that distinct affinities of TNF ligands for their receptors promote different and non-redundant cellular functions.

Multicellular organisms have evolved organs and tissues with highly specialized tasks. For instance, nutrients are assimilated by the gut, sensed, processed, stored, and released by adipose tissues and liver to provide energy consumed by peripheral organ activities. The function of each organ is modified by local clues and systemic signals derived from other organs to ensure a coordinated response accommodating the physiological needs of the organism. The intestine, which represents one of the largest interfaces between the internal and external environment, plays a key role in sensing and relaying environmental inputs such as nutrients and microbial derivatives to other organs to produce systemic responses. In turn, gut physiology and immunity are regulated by multiple signals emanating from other organs including the brain and the adipose tissues. In this review, we highlight physiological processes where the gut serves as a key organ in coupling systemic signals or environmental cues with organism growth, metabolism, immune activity, aging, or behavior. Robust strategies involving intraorgan and interorgan signaling pathways have evolved to preserve gut size in homeostatic conditions and restrict growth during damage-induced regenerative phases. Here we review some of the mechanisms that maintain gut size homeostasis and point out known examples of homeostasis-breaking events that promote gut plasticity to accommodate changes in the external or internal environment. Nutrients excreted from animals affect the nutritive value of manure as a soil amendment as well as the composition of gases emitted from manure storage facilities. There is a dearth of information, however, on how diet type in combination with dietary particle size affects nutrients deposited into manure storage facilities and how this affects manure composition and gas emissions. To fill this knowledge gap, an animal feeding trial was performed to evaluate potential interactive effects between feed particle size and diet composition on manure characteristics and manure-derived gaseous emissions. Forty-eight finishing pigs housed in individual metabolism crates that allowed for daily collection of urine and feces were fed diets differing in fiber content and particle size. Urine and feces were collected and stored in 446-L stainless steel containers for 49 d. There were no interactive effects between diet composition and feed particle size on any manure or gas emission parameter measured. In general, diets higher in fiber content increased manure nitrogen (N), carbon (C), and total volatile fatty acid (VFA) concentrations and increased manure VFA emissions but decreased manure ammonia emissions. Decreasing the particle size of the diet lowered manure N, C, VFAs, phenolics, and indole concentrations and decreased manure emissions of total VFAs. Grain producers are challenged to maximize crop production while utilizing nutrients efficiently and minimizing negative impacts on water quality. There is a particular concern about nutrient export to the Gulf of Mexico via loss from subsurface drainage systems. The objective of this study was to investigate the effects of crop rotation, tillage, crop residue removal, swine manure applications, and cereal rye (Secale cereale L.) cover crops on nitrate-N (NO 3 -N) and total reactive phosphorus (TRP) loss via subsurface drainage. The study was evaluated from 2008 through 2015 using 36 0.4-ha plots outfitted with a subsurface drainage water quality monitoring system. Results showed that when swine manure was applied before both corn (Zea mays L.) and soybean [Glycine max (L.) Merr.], drainage water had significantly higher 8-yr-average flow-weighted NO 3 -N concentrations compared with swine manure applied before corn only in a corn-soybean rotation. The lowest NO 3 -N loss was 15.2 kg N ha -1  yr -1 from a no-till corn-soybean treatment with rye cover crop and spring application of urea-ammonium nitrate (UAN) to corn. The highest NO 3 -N loss was 29.5 kg N ha -1  yr -1 from swine manure applied to both corn and soybean. A rye cover crop reduced NO 3 -N loss, whereas tillage and residue management had little impact on NO 3 -N loss. Losses of TRP averaged <32 g P ha -1  yr -1 from all treatments. Corn yield was negatively affected by both no-till management and cereal rye cover crops. Introduction: Surgical telementoring connects expert mentors with trainees performing urgent care in austere environments. However, such environments impose unreliable network quality, with significant latency and low bandwidth. We have developed an augmented reality telementoring system that includes future step visualization of the medical procedure. Pregenerated video instructions of the procedure are dynamically overlaid onto the trainee's view of the operating field when the network connection with a mentor is unreliable., Methods: Our future step visualization uses a tablet suspended above the patient's body, through which the trainee views the operating field. Before trainee use, an expert records a "future library" of step-by-step video footage of the operation. Videos are displayed to the trainee as semitransparent graphical overlays. We conducted a study where participants completed a cricothyroidotomy under telementored guidance. Participants used one of two telementoring conditions: conventional telestrator or our system with future step visualization. During the operation, the connection between trainee and mentor was bandwidth throttled. Recorded metrics were idle time ratio, recall error, and task performance., Results: Participants in the future step visualization condition had 48% smaller idle time ratio (14.5% vs. 27.9%, P < 0.001), 26% less recall error (119 vs. 161, P = 0.042), and 10% higher task performance scores (rater 1 = 90.83 vs. 81.88, P = 0.008; rater 2 = 88.54 vs. 79.17, P = 0.042) than participants in the telestrator condition., Conclusions: Future step visualization in surgical telementoring is an important fallback mechanism when trainee/mentor network connection is poor, and it is a key step towards semiautonomous and then completely mentor-free medical assistance systems.

An experiment was conducted to determine the effect of feeding finishing pigs a corn-soybean (CSBM) diet or a CSBM diet supplemented with 30% dried distillers grains with solubles (DDGS), in combination with or without a growth-promoting ionophore (0 or 30 mg narasin/kg of diet), has on manure composition, microbial ecology, and gas emissions. Two separate groups of 24 gilts (initial BW = 145.1 kg, SD = 7.8 kg) were allotted to individual metabolism crates that allowed for total but separate collection of feces and urine during the 48-d collection period. After each of the twice-daily feedings, feces and urine from each crate was collected and added to its assigned enclosed manure storage tank. Each tank contained an individual fan system that pulled a constant stream of air over the manure surface for 2 wk prior to air (day 52) and manure sampling (day 53). After manure sampling, the manure in the tanks was dumped and the tanks cleaned for the second group of pigs. Except for total manure Ca and P output as a percent of intake and for manure methane product rate and biochemical methane potential (P ≤ 0.08), there were no interactions between diet composition and narasin supplementation. Narasin supplementation resulted in increased manure C (P = 0.05), increased manure DM, C, S, Ca, and phosphorus as a percent of animal intake (P ≤ 0.07), and increased manure volatile solids and foaming capacity (P ≤ 0.09). No effect of narasin supplementation was noted on manure VFA concentrations or any of the gas emission parameters measured (P ≥ 0.29). In contrast, feeding finishing pigs a diet containing DDGS dramatically affected manure composition as indicated by increased concentration of DM, C, ammonia, N, and total and volatile solids (P = 0.01), increased manure DM, N, and C as a percent of animal intake (P = 0.01), increased manure total VFA and phenols (P ≤ 0.05), decreased gas emissions of ammonia and volatile sulfur compounds (VSC; P = 0.01), increased emissions of phenols and indoles (P ≤ 0.06), decreased methane production rate (P = 0.01), and slight differences in microbial ecology (R ≤ 0.47). In conclusion, feeding a diet which contains an elevated level of indigestible fiber (i.e., DDGS) resulted in more fiber in the manure which therefore dramatically affected manure composition, gas emissions, and microbial ecology, while narasin supplementation to the diet did not exhibit a significant effect on any of these parameters in the resultant swine manure.

Three experiments were conducted to determine the effect of narasin on growth performance and on GE and nutrient digestibility in nursery, grower, and finishing pigs fed either a corn-soybean meal (CSBM) diet or a CSBM diet supplemented with distillers dried grains with solubles (DDGS), in combination with either 0 or 30 mg narasin/kg of diet. In Exp. 1 (64 gilts, initial BW = 9.0 kg, SD = 1.0 kg) and Exp. 2 (60 gilts. initial BW = 81.1 kg, SD = 6.1 kg), gilts were allotted into individual pens and fed their experimental diets for 24 and 21 d, respectively. On the last 2 d of each experiment, fecal samples were collected to assess apparent total tract digestibility (ATTD) of GE and various nutrients. In Exp. 3, 2 separate groups of 24 gilts (initial BW = 145.1 kg, SD = 7.8 kg) were allotted to individual metabolism crates and fed their experimental diets for 30 d prior to a time-based 6-d total fecal collection period to assess GE and nutrient digestibility. In Exp. 1, there was an interaction between diet type and narasin addition for G:F and for many of the ATTD coefficients measured. When narasin was supplemented to the CSBM diet, ATTD of GE, DM, C, S, phosphorus, NDF, and ADF was either not changed or reduced, while when narasin was supplemented to DDGS diets, these same ATTD parameters were increased (interaction, ≤ 0.05). Even though ADG and ADFI were not affected, G:F was improved in pigs fed the CSBM diet with supplemental narasin, but was reduced in pigs fed the DDGS diet with supplemental narasin (interaction, < 0.05). In Exp. 2, there was an interaction between diet type and narasin supplementation only for ATTD of Ca (interaction, < 0.01), in that narasin supplementation did not change the ATTD of Ca in pigs fed the CSBM diet, while narasin supplementation reduced the ATTD of Ca in pigs fed the DDGS containing diet. In Exp. 3, there was an interaction between diet and narasin only for ATTD of C (interaction, < 0.01) in that narasin supplementation resulted in an increased ATTD of C in pigs fed the CSBM diet, while narasin supplementation to the DDGS containing diet resulted in a reduced ATTD of Ca. In general, the data indicate that narasin interacted with and had its largest effect on pig performance and GE or nutrient digestibility in 9 to 23 kg pigs compared to pigs weighing greater than 80 kg. The data also indicate that the addition of DDGS reduced GE, DM, Ca, and N digestibility, regardless of BW.

Body size is an intrinsic property of living organisms that is intimately linked to the developmental program to produce fit individuals with proper proportions. Final size is the result of both genetic determinants and sophisticated mechanisms adapting size to available resources. Even though organs grow according to autonomous programs, some coordination mechanisms ensure that the different body parts adjust their growth with the rest of the body. In Drosophila, Dilp8, a hormone of the Insulin/Relaxin family is a key player in this inter-organs coordination and is required together with its receptor Lgr3 to limit developmental variability. Coordination of organ growth during development is required to generate fit individuals with fixed proportions. We recently identified Drosophila Dilp8 as a key hormone in coupling organ growth with animal maturation. In addition, dilp8 mutant flies exhibit elevated fluctuating asymmetry (FA) demonstrating a function for Dilp8 in ensuring developmental stability. The signals regulating Dilp8 activity during normal development are not yet known. Here, we show that the transcriptional co-activators of the Hippo (Hpo) pathway, Yorkie (Yki, YAP/TAZ) and its DNA-binding partner Scalloped (Sd), directly regulate dilp8 expression through a Hpo-responsive element (HRE) in the dilp8 promoter. We further demonstrate that mutation of the HRE by genome-editing results in animals with increased FA, thereby mimicking full dilp8 loss of function. Therefore, our results indicate that growth coordination of organs is connected to their growth status through a feedback loop involving Hpo and Dilp8 signalling pathways.

The livestock and poultry production industry, regulatory agencies, and researchers lack a current, science-based guide and data base for evaluation of air quality mitigation technologies. Data collected from science-based review of mitigation technologies using practical, stakeholders-oriented evaluation criteria to identify knowledge gaps/needs and focuses for future research efforts on technologies and areas with the greatest impact potential is presented in the Literature Database tab on the air management practices tool (AMPAT). The AMPAT is web-based (available at www.agronext.iastate.edu/ampat) and provides an objective overview of mitigation practices best suited to address odor, gaseous, and particulate matter (PM) emissions at livestock operations. The data was compiled into Excel spreadsheets from a literature review of 265 papers was performed to (1) evaluate mitigation technologies performance for emissions of odor, volatile organic compounds (VOCs), ammonia (NH3), hydrogen sulfide (H2S), particulate matter (PM), and greenhouse gases (GHGs) and to (2) inform future research needs.

Foam accumulation in deep-pit manure storage facilities is of concern for swine producers because of the logistical and safety-related problems it creates. A feeding trial was performed to evaluate the impact of feed grind size, fiber source, and manure inoculation on foaming characteristics. Animals were fed: (1) C-SBM (corn-soybean meal): (2) C-DDGS (corn-dried distiller grains with solubles); and (3) C-Soybean Hull (corn-soybean meal with soybean hulls) with each diet ground to either fine (374 μm) or coarse (631 μm) particle size. Two sets of 24 pigs were fed and their manure collected. Factors that decreased feed digestibility (larger grind size and increased fiber content) resulted in increased solids loading to the manure, greater foaming characteristics, more particles in the critical particle size range (2-25 μm), and a greater biological activity/potential., (Copyright © 2015 Elsevier Ltd. Early transplantation and grafting experiments suggest that body organs follow autonomous growth programs [1-3], therefore pointing to a need for coordination mechanisms to produce fit individuals with proper proportions. We recently identified Drosophila insulin-like peptide 8 (Dilp8) as a relaxin and insulin-like molecule secreted from growing tissues that plays a central role in coordinating growth between organs and coupling organ growth with animal maturation [4, 5]. Deciphering the function of Dilp8 in growth coordination relies on the identification of the receptor and tissues relaying Dilp8 signaling. We show here that the orphan receptor leucine-rich repeat-containing G protein-coupled receptor 3 (Lgr3), a member of the highly conserved family of relaxin family peptide receptors (RXFPs), mediates the checkpoint function of Dilp8 for entry into maturation. We functionally identify two Lgr3-positive neurons in each brain lobe that are required to induce a developmental delay upon overexpression of Dilp8. These neurons are located in the pars intercerebralis, an important neuroendocrine area in the brain, and make physical contacts with the PTTH neurons that ultimately control the production and release of the molting steroid ecdysone. Reducing Lgr3 levels in these neurons results in adult flies exhibiting increased fluctuating bilateral asymmetry, therefore recapitulating the phenotype of dilp8 mutants. Our work reveals a novel Dilp8/Lgr3 neuronal circuitry involved in a feedback mechanism that ensures coordination between organ growth and developmental transitions and prevents developmental variability., (Copyright © 2015 Elsevier Ltd. Methane emission is an important tool in the evaluation of manure management systems due to the potential impact it has on global climate change. Field procedures used for estimating methane emission rates require expensive equipment, are time consuming, and highly variable between farms. The purpose of this paper is to report a simple laboratory procedure for estimating methane emission from stored manure. The test developed was termed a methane production rate (MPR) assay as it provides a short-term biogas production measurement. The MPR assay incubation time is short (3d), requires no sample preparation in terms of inoculation or dilution of manure, is incubated at room temperature, and the manure is kept stationary. These conditions allow for high throughput of samples and were chosen to replicate the conditions within deep-pit manure storages. In brief, an unaltered aliquot of manure was incubated at room temperature for a three-days to assay the current rate of methane being generated by the manure. The results from this assay predict an average methane emission factor of 12.2 ± 8.1 kg CH4 head(-1) yr(-1) per year, or about 5.5 ± 3.7 kg CH4 per finished animal, both of which compare well to literature values of 5.5 ± 1.1 kg CH4 per finished pig for deep-pit systems (Liu et al., 2013). The average methane flux across all sites and months was estimated to be 22 ± 17 mg CH4 m(-2)-min(-1), which is within literature values for deep-pit systems ranging from 0.24 to 63 mg CH4 m(-2)-min(-1) (Park et al., 2006) and similar to the 15 mg CH4 m(-2)-min(-1) estimated by (Zahn et al., 2001)., (Copyright © 2015 Elsevier Ltd. Disruption of epithelial polarity is a key event in the acquisition of neoplastic growth. JNK signalling is known to play an important part in driving the malignant progression of many epithelial tumours, although the link between loss of polarity and JNK signalling remains elusive. In a Drosophila genome-wide genetic screen designed to identify molecules implicated in neoplastic growth, we identified grindelwald (grnd), a gene encoding a transmembrane protein with homology to members of the tumour necrosis factor receptor (TNFR) superfamily. Here we show that Grnd mediates the pro-apoptotic functions of Eiger (Egr), the unique Drosophila TNF, and that overexpression of an active form of Grnd lacking the extracellular domain is sufficient to activate JNK signalling in vivo. Grnd also promotes the invasiveness of Ras(V12)/scrib(-/-) tumours through Egr-dependent Matrix metalloprotease-1 (Mmp1) expression. Grnd localizes to the subapical membrane domain with the cell polarity determinant Crumbs (Crb) and couples Crb-induced loss of polarity with JNK activation and neoplastic growth through physical interaction with Veli (also known as Lin-7). Therefore, Grnd represents the first example of a TNFR that integrates signals from both Egr and apical polarity determinants to induce JNK-dependent cell death or tumour growth.

Beef feedlots of all sizes are looking for more cost-effective solutions for managing feedlot runoff. Vegetative treatment systems are one potential option, but require performance evaluation for use on concentrated animal feeding operations. The performance of six vegetative treatment systems on open beef feedlots throughout Iowa was monitored from 2006 through 2009. These feedlots had interim, National Pollution Discharge Elimination System permits that allowed the use of vegetative treatment systems to control and treat runoff from the open feedlots. This manuscript focuses on making within site comparisons, i.e., from year-to-year and component-to-component within a site, to evaluate how management changes and system modifications altered performance. The effectiveness, in terms of effluent concentration reductions, of each system was evaluated; nutrient concentration reductions typically ranged from 60 to 99% during treatment in the vegetative components of the vegetative treatment systems. Monitoring results showed a consistent improvement in system performance during the four years of study. Much of this improvement can be attributed to improved management techniques and system modifications that addressed key performance issues. Specifically, active control of the solid settling basin outlet improved solids retention and allowed the producers to match effluent application rates to the infiltration rate of the vegetative treatment area, reducing the occurrence of effluent release. Additional improvements resulted from system maturation, increased operator experience, and the addition of earthen flow spreaders within the vegetative treatment area to slow flow and provide increased effluent storage within the treatment area, and switching to active management of settling basin effluent release., (Copyright © 2013 Elsevier Ltd. In animal species undergoing determinate growth, the making of a full-size adult body requires a series of coordinated growth events culminating in the cessation of growth that precedes sexual maturation. The merger between physiology and genetics now coming to pass in the Drosophila model allows us to decipher these growth events with an unsurpassed level of sophistication. Here, we review several coordination mechanisms that represent fundamental aspects of growth control: adaptation of growth to environmental cues, interorgan coordination, and the coordination of growth with developmental transitions. The view is emerging of an integrated process where organ-autonomous growth is coordinated with both developmental and environmental cues to define final body size., (Copyright © 2013 Elsevier Ltd. Little is known about how organ growth is monitored and coordinated with the developmental timing in complex organisms. In insects, impairment of larval tissue growth delays growth and morphogenesis, revealing a coupling mechanism. We carried out a genetic screen in Drosophila to identify molecules expressed by growing tissues participating in this coupling and identified dilp8 as a gene whose silencing rescues the developmental delay induced by abnormally growing tissues. dilp8 is highly induced in conditions where growth impairment produces a developmental delay. dilp8 encodes a peptide for which expression and secretion are sufficient to delay metamorphosis without affecting tissue integrity. We propose that Dilp8 peptide is a secreted signal that coordinates the growth status of tissues with developmental timing.

Drosophila MCRS2 (dMCRS2; MCRS2/MSP58 and its splice variant MCRS1/p78 in humans) belongs to a family of forkhead-associated (FHA) domain proteins. Whereas human MCRS2 proteins have been associated with a variety of cellular processes, including RNA polymerase I transcription and cell cycle progression, dMCRS2 has been largely uncharacterized. Recent data show that MCRS2 is purified as part of a complex containing the histone acetyltransferase MOF (males absent on first) in both humans and flies. MOF mediates H4K16 acetylation and regulates the expression of a large number of genes, suggesting that MCRS2 could also have a function in transcription regulation. Here, we show that dMCRS2 copurifies with RNA polymerase II (RNAP II) complexes and localizes to the 5' ends of genes. Moreover, dMCRS2 is required for optimal recruitment of RNAP II to the promoter regions of cyclin genes. In agreement with this, dMCRS2 is required for normal levels of cyclin gene expression. We propose a model whereby dMCRS2 promotes gene transcription by facilitating the recruitment of RNAP II preinitiation complexes (PICs) to the promoter regions of target genes.

The mammalian spliceosome has mainly been studied using proteomics. The isolation and comparison of different splicing intermediates has revealed the dynamic association of more than 200 splicing factors with the spliceosome, relatively few of which have been studied in detail. Here, we report the characterization of the Drosophila homologue of microfibril-associated protein 1 (dMFAP1), a previously uncharacterized protein found in some human spliceosomal fractions ( Jurica, M. S., and Moore, M. J. (2003) Mol. Cell 12, 5-14 ). We show that dMFAP1 binds directly to the Drosophila homologue of Prp38p (dPrp38), a tri-small nuclear ribonucleoprotein component ( Xie, J., Beickman, K., Otte, E., and Rymond, B. C. (1998) EMBO J. 17, 2938-2946 ), and is required for pre-mRNA processing. dMFAP1, like dPrp38, is essential for viability, and our in vivo data show that cells with reduced levels of dMFAP1 or dPrp38 proliferate more slowly than normal cells and undergo apoptosis. Consistent with this, double-stranded RNA-mediated depletion of dPrp38 or dMFAP1 causes cells to arrest in G(2)/M, and this is paralleled by a reduction in mRNA levels of the mitotic phosphatase string/cdc25. Interestingly double-stranded RNA-mediated depletion of a wide range of core splicing factors elicits a similar phenotype, suggesting that the observed G(2)/M arrest might be a general consequence of interfering with spliceosome function.

The Drosophila gene, pixie, is an essential gene required for normal growth and translation. Pixie is the fly ortholog of human RLI, which was first identified as an RNase L inhibitor, and yeast Rli1p, which has recently been shown to play a role in translation initiation and ribosome biogenesis. These proteins are all soluble ATP-binding cassette proteins with two N-terminal iron-sulfur clusters. Here we demonstrate that Pixie can be isolated from cells in complex with eukaryotic translation initiation factor 3 and ribosomal proteins of the small subunit. In addition, our analysis of polysome profiles reveals that double-stranded RNA interference-mediated depletion of Pixie results in an increase in empty 80 S ribosomes and a corresponding decrease in polysomes. Thus Pixie is required for normal levels of translation initiation. We also find that Pixie associates with the 40 S subunit on sucrose density gradients in an ATP-dependent manner. Our observations are consistent with Pixie playing a catalytic role in the assembly of complexes required for translation initiation. Thus, the function of this soluble ATP-binding cassette domain protein family in translation initiation has been conserved from yeast through to higher eukaryotes.

Early results suggested that the amphotropic murine leukemia virus (A-MLV) does not enter cells via endocytosis through clathrin-coated pits and this gammaretrovirus has therefore been anticipated to fuse directly with the plasma membrane. However, here we present data implicating a caveola-mediated endocytic entry route for A-MLV via its receptor Pit2. Caveolae belong to the cholesterol-rich microdomains characterized by resistance to nonionic detergents such as Triton X-100. Extraction of murine fibroblastic NIH 3T3 cells in cold Triton X-100 showed the presence of the A-MLV receptor Pit2 in detergent-insoluble microdomains. Using coimmunoprecipitation of cell extracts, we were able to demonstrate direct association of Pit2 with caveolin-1, the structural protein of caveolae. Other investigations revealed that A-MLV infection in contrast to vesicular stomatitis virus infection is a slow process (t(1/2) approximately 5 h), which is dependent on plasma membrane cholesterol but independent of NH4Cl treatment of cells; NH4Cl impairs entry via clathrin-coated pits. Furthermore, expression of dominant-negative caveolin-1 decreased the susceptibility to infection via Pit2 by approximately 70%. These results show that A-MLV can enter cells via a caveola-dependent entry route. Moreover, increase in A-MLV infection by treatment with okadaic acid as well as entry of fusion-defective fluorescent A-MLV virions in NIH 3T3 cells further confirmed our findings and show that A-MLV can enter mouse fibroblasts via an endocytic entry route involving caveolae. Finally, we also found colocalization of fusion-defective fluorescent A-MLV virions with caveolin-1 in NIH 3T3 cells. This is the first time substantial evidence has been presented implicating the existence of a caveola-dependent endocytic entry pathway for a retrovirus.
